Verfahren zur Herstellung neuer sekundärer Amine
Gegenstand der Erfindung ist ein Verfahren zur Herstellung von sekundären Aminen der Formel

worin R einen gesättigten niederen Kohlenwasser stoffrest und Rl einen niederen Alkylrest bedeuten.
Der gesättigte niedere Kohlenwasserstoffrest R soll vor allem aliphatischer Natur sein, z. B. ein niederer gerader oder verzweigter Alkylrest, z. B. Methyl, Äthyl, Propyl, Isopropyl, Butyl, sek.-Butyl, Amyl oder Hexyl. Als niedere Alkylreste Ri sind die eben erwähnten, vor allem aber Methyl anzuführen.
Die neuen Verbindungen und ihre Salze besitzen ausgesprochen sympathicolytische Wirkung mit einer bradykarden und sedativen Wirkungskomponente. Sie sollen dementsprechend als Medikamente verwendet werden.
Besonders wertvoll sind die Verbindungen der Formel I, worin Ri und R niedere Alkylreste mit insgesamt mindestens 3 Kohlenstoffatomen bedeuten, und ihre Salze, und besonders diejenigen der Formel

Das erfindungsgemässe Verfahren zur Herstellung der neuen Verbindungen ist dadurch gekennzeichnet, dass man eine Verbindung der Formel

worin X eine reaktionsfähige veresterte Hydroxylgruppe bedeutet, mit einem Amin der Formel H2N-R kondensiert.
Als Reste X kommen insbesondere mit starken anorganischen oder organischen Säuren, z. B. Halogenwasserstoffsäuren oder organischen Sulfonsäuren, wie p-Toluolsulfonsäure, veresterte Hydroxylgruppen in Frage.
Je nach der Arbeitsweise erhält man die neuen Verbindungen in Form ihrer Basen oder Salze. Aus den Salzen können in an sich bekannter Weise die freien Aminbasen gewonnen werden. Von letzteren wiederum lassen sich durch Umsetzung mit Säuren, die zur Bildung therapeutisch verwendbarer Salze geeignet sind, Salze gewinnen, wie z. B. der Halogenwasserstoffsäuren, Schwefelsäure, Salpetersäure, Phosphorsäure, Rhodanwasserstoffsäure, Essigsäure, Propionsäure, Oxalsäure, Malonsäure, Weinsäure, Bernsteinsäure, Apfelsäure, Methansulfonsäure, ¯thansulfonsÏure, Oxyäthansulfonsäure, Benzol-oder Toluolsulfonsäure oder von therapeutisch wirksamen Säuren.
Die Ausgangsstoffe können nach an sich bekannten Methoden gewonnen werden.
Die neuen Verbindungen können als Heilmittel, z. B. in Form pharmazeutischer Präparate, Verwendung finden, welche sie oder ihre Salze in Mischung mit einem für die enterale, parenterale oder topicale Applikation geeigneten pharmazeutischen organischen oder anorganischen, festen oder flüssigen Träger- material enthalten.
In den folgenden Beispielen sind die Temperaturen in Celsiusgraden angegeben.
Beispiel 1
11, 55 g (0, 05 Mol) o-Methoxy-phenoxyäthyl- bromid und 7, 3 g (0, 1 Mol) prim.-Isobutylamin werden in einem Rundkolben mit Luftkühler 90 Minuten lang auf 1009 erhitzt. Nach dem Abkühlen nimmt man das Reaktionsgemisch in Chloroform auf und schüttelt die Lösung zunächst mit verdünntem Ammoniak und dann mit Wasser aus. Die Chloroformlösung hinterlässt nach dem Trocknen mit wasserfreiem Natriumsulfat und anschliessendem Eindampfen N-(o-Methoxy-phenoxyäthyl)-isobutylamin der Formel

Dieses wird in Essigester gelöst, mit einer 2n Lösung von Chlorwasserstoff in Essigester neutralisiert und die Lösung eingedampft.
Nach einmaligem Umkristallisieren aus Athanol-Essigester erhält man das Hydrochlorid als farblose Kristalle vom F. 147 bis 149 .
In analoger Weise kann man z. B. die folgende Verbindung herstellen : N-(o-Methoxy-phenoxyäthyl)-methylamin-hydro- chlorid, F. 134-136 (aus AthanollEssigester).
Das als Ausgangsmaterial verwendete o-Methoxyphenoxyäthylbromid kann wie folgt hergestellt werden :
Zu einer Lösung von 11, 5 g (0, 5 Mol) Natrium in 250 cm3 absolutem Äthanol lässt man 62 g (0, 5 Mol) in 100 ml absolutem Athanol gelöstes Gujakol zufliessen. Nachdem der Kolbeninhalt auf 0 abgekühlt worden ist, werden 282 g (1, 5 Mol) Athylenbromid zugegeben. Unter kräftigem Rühren kocht man das Reaktionsgemisch unter Rückfluss, bis es neutral reagiert (etwa 4 Stunden). Nach dem Abkühlen filtriert man das entstandene Natriumbromid ab und engt im Wasserstrahlvakuum ein. Der Rück- stand wird in 200 cm3 Chloroform aufgenommen, zweimal mit je 50 cm3 2n Natronlauge ausgezogen, dann mit Wasser neutralgewaschen und schliesslich über wasserfreiem Natriumsulfat getrocknet.
Nach dem Abdestillieren des Lösungsmittels kühlt man den Rückstand in Eis und filtriert das als Nebenprodukt entstandene 1, 2-Di- (o-Methoxy-phenoxy)-äthan (F. 148-150 ) ab. Das Filtrat wird am Vakuum destilliert. Das o-Methoxy-phenoxyäthylbromid ist eine farblose Flüssigkeit vom Kp 141-142", welche nach einiger Zeit kristallinisch erstarrt und dann einen Schmelzpunkt von 40-42 aufweist.
Beispiel 2
Auf gleiche Weise wie im Beispiel 1 angegeben, erhält man aus 11, 55 g (0, 05 Mol) o-Methoxyphenoxyäthylbromid und 7, 3 g (0, 1 Mol) n-Butylamin das N- (o-Methoxy-phenoxyäthyl)-n-butylamin der Formel

als gelbliches 01. Umsetzen mit methanolischer Salzsäure führt zum Hydrochlorid, das aus Essigester umkristallisiert bei 148-150 schmilzt.
Beispiel 3
Man erhitzt 10 g (0, 042 Mol) o-Methoxyphenoxyäthylbromid und 10 g (0, 22 Mol) Athylamin in 50 cm3 Athylalkohol im geschlossenen Rohr während 14 Stunden auf 100". Das Reaktionsgemisch wird eingedampft, in 100 cm3 2n Salzsäure aufgenommen, der Neutralteil durch Extraktion mit Ather entfernt und die wässrige Lösung mit Aktivkohle entfärbt. Man macht durch Zugabe von ver dünnter Natronlauge alkalisch und extrahiert die ausfallende Base mit Chloroform. Das N- (o-Methoxy phenoxyäthyl)-äthylamin der Formel

10 g (0, 042 Mol) o-Methoxy-phenoxyäthyl- bromid und 15 g (0, 25 Mol) n-Propylamin werden in 20 cm3 Athanol 15 Stunden unter Rückfluss erhitzt. Man dampft das Lösungsmittel und das über- schüssige Amin im Wasserstrahlvakuum ab und arbeitet wie im Beispiel 3 angegeben auf. Das N- (o- Methoxy-phenoxyäthyl)-n-propylamin der Formel

wird als viskoses, fast farbloses bl erhalten. Das Hydrochlorid, auf gleiche Weise wie im Beispiel 1 angegeben hergestellt, kristallisiert aus Essigester Athanol in farblosen Kristallen vom F. 140-142 .

8, 0 g (0, 035 Mol) o-Methoxy-ph, enoxyäthyl- bromid und 30 cm3 tert.-Butylamin werden in 50 cm3 Athanol über Nacht unter Rückfluss gekocht. Lösungsmittel und überschüssiges Amin dampft man im Wasserstrahlvakuum ab. Der Rückstand wird in 100 cm3 verdünnter Salzsäure aufgenommen. Durch Ausschütteln mit Ather entfernt man etwaige Neutralprodukte, filtriert dann die wässrige Phase durch Aktivkohle und macht das Filtrat durch Zugabe von verdünnter Natronlauge alkalisch. Extraktion mit Chloroform und anschliessende Destillation im Hochvakuum liefern das N- (o-Methoxy-phenoxyäthyl)- tert.-butylamin der Formel

Zu einer Lösung von 46 g (2, 0 Mol) Natrium in 1000 cm3 absolutem Athanol lässt man 276 g (2, 0 Mol) Guäthol (o-Athoxy-phenol) zufliessen. Man kühlt auf 10 , gibt 564 g (3, 0 Mol) Äthylenbromid zu und kocht anschliessend 5 Stunden unter Rückfluss. Die Aufarbeitung, wie im Beispiel 1 beschrieben durchgeführt, ergibt 130, 3 g (0, 53 Mol) o-, 2ithoxy-phenoxyäthylbromid vom Kp. il : 142 bis 145 .

Man setzt 12, 8 g (0, 052 Mol) o-Athoxy-phenoxy äthylbromid nach der im Beispiel 5 beschriebenen Methode mit 40 cm3 n-Butylamin um. Es wird das N- (o-Athoxy-phenoxyäthyl)-n-butylamin der Formel

The invention relates to a process for the preparation of secondary amines of the formula

The saturated lower hydrocarbon radical R should above all be aliphatic in nature, e.g. B. a lower straight or branched alkyl radical, e.g. B. methyl, ethyl, propyl, isopropyl, butyl, sec-butyl, amyl or hexyl. The lower alkyl radicals Ri are those just mentioned, but especially methyl.
The new compounds and their salts have a pronounced sympathicolytic effect with a bradycardic and sedative component. Accordingly, they should be used as drugs.

The radicals X come in particular with strong inorganic or organic acids, eg. B. hydrohalic acids or organic sulfonic acids such as p-toluenesulfonic acid, esterified hydroxyl groups in question.
Depending on the procedure, the new compounds are obtained in the form of their bases or salts. The free amine bases can be obtained from the salts in a manner known per se. From the latter, in turn, salts can be obtained by reaction with acids which are suitable for the formation of therapeutically useful salts, e.g. B. the hydrohalic acids, sulfuric acid, nitric acid, phosphoric acid, hydrofluoric acid, acetic acid, propionic acid, oxalic acid, malonic acid, tartaric acid, succinic acid, malic acid, methanesulfonic acid, ¯thanesulfonsÏure, oxyethanesulfonic acid, benzene or toluenesulfonic acids or of therapeutically active acids.
The starting materials can be obtained by methods known per se.
The new compounds can be used as remedies, e.g. B. in the form of pharmaceutical preparations, which contain them or their salts in a mixture with a pharmaceutical, organic or inorganic, solid or liquid carrier material suitable for enteral, parenteral or topical application.
In the following examples the temperatures are given in degrees Celsius.

11.55 g (0.05 mol) of o-methoxyphenoxyethyl bromide and 7.3 g (0.1 mol) of primary isobutylamine are heated to 1009 for 90 minutes in a round bottom flask with an air condenser. After cooling, the reaction mixture is taken up in chloroform and the solution is extracted first with dilute ammonia and then with water. After drying with anhydrous sodium sulfate and subsequent evaporation, the chloroform solution leaves behind N- (o-methoxyphenoxyethyl) isobutylamine of the formula

The o-methoxyphenoxyethyl bromide used as the starting material can be prepared as follows:
62 g (0.5 mol) of gujakol dissolved in 100 ml of absolute ethanol are allowed to flow into a solution of 11.5 g (0.5 mol) of sodium in 250 cm3 of absolute ethanol. After the contents of the flask have been cooled to 0, 282 g (1.5 mol) of ethylene bromide are added. The reaction mixture is refluxed with vigorous stirring until it reacts neutrally (about 4 hours). After cooling, the sodium bromide formed is filtered off and concentrated in a water jet vacuum. The residue is taken up in 200 cm3 of chloroform, extracted twice with 50 cm3 of 2N sodium hydroxide solution each time, then washed neutral with water and finally dried over anhydrous sodium sulfate.
After the solvent has been distilled off, the residue is cooled in ice and the 1,2-di (o-methoxyphenoxy) ethane (mp 148-150) formed as a by-product is filtered off. The filtrate is distilled in vacuo. The o-methoxyphenoxyethyl bromide is a colorless liquid with a boiling point of 141-142 "which solidifies in crystalline form after some time and then has a melting point of 40-42.
Example 2
In the same way as in Example 1, from 11.55 g (0.05 mol) of o-methoxyphenoxyethyl bromide and 7.3 g (0.1 mol) of n-butylamine, N- (o-methoxyphenoxyethyl) - n-butylamine of the formula

8.0 g (0.035 mol) of o-methoxy-ph, enoxyethyl bromide and 30 cm3 of tert-butylamine are refluxed in 50 cm3 of ethanol overnight. Solvent and excess amine are evaporated off in a water jet vacuum. The residue is taken up in 100 cm3 of dilute hydrochloric acid. Any neutral products are removed by shaking with ether, then the aqueous phase is filtered through activated charcoal and the filtrate is made alkaline by adding dilute sodium hydroxide solution. Extraction with chloroform and subsequent distillation in a high vacuum yield the N- (o-methoxyphenoxyethyl) tert-butylamine of the formula
