Verfahren zur Herstellung von heterocyclischen Verbindungen Gegenstand der vorliegenden Erfindung st ein Verfahren zur Hersltellung von Pyrazolo [3, 4-d] pyrimidinen der Formel

worin Ar einen Arylrest und R4 eine freie oder substi- tuierte Aminogruppe und R. dasselbe wie R4 oder Wasserstoff oder einen gegebenenfalls substituierten Kohlenwasserstoffrest bedeuten. Als freie oder sub stituierte Aminogruppen kommen z. B. mono-oder disubstituierte Amino-, wie Dialkyl-oder Alkylenaminogruppen, Furylmethylaminogruppen oder Hy droxylamino-oder Hydrazinogruppen in Frage. Sind die Aminogruppen durch Kohlenwasserstoffreste, wie z. B.
Alkylreste, substituiert, so können diese auch durch Heteroatome, wie Sauerstoff oder Stickstoff, unterbrochen sein. So können diese Substituenten eine Oxa-oder eine Azaalkylaminogruppe darstellen, wie z. B. die Dimethylamino-, Diathylamino-, Piperidino- oder Morpholinbäthylaminogruppe. A1s Arylreste kommen vorzugswiese unsubstituierte oder durch Al- kyl oder Halogen substituierte Phenylreste und als Kohlenwasserstoffreste in 6-Stellung kommen besonders Alkylreste in Betracht.
Die genannten Verbindungen und ihre Salze sind neu und zeigen wertvolle pharmakologische Eigenschaften. Sie können auf den verschiedenen Gebieten der Medizin als Heilmittel dienen oder als Zwischen- produkte zu ihrer Herstellung Verwendung finden. Sie zeichnen sich aus durch eine besondere Wirkung gegen verschiedene tierische Parasiten, wie z. B. gegen über Bakterien, Fungi, Amoeben, Protozoen oder Trypanosomen. Zudem sind diese Verbindungen Purinantagonisten und zeigen eine hemmende Wirkung gegenuber Tumoren.
Das erfindungsgemässe Verfahren ist dadurch ge kennzeichnet, d'ass man Pyrazolo [3, 4-d]pyrimidine der Formel

worin Hal ein Halogenatom und R6' Wasserstoff, Halogen oder einen gegebenenfalls substituierten Kohlenwasserstoffrest darstellen, mit Verbindungen der Formel HR4 umsetzt. So setzt man die Halogen- verbindungen z. B. mit Ammoniak, Aminen, Hy droxylaminen oder Hydrazinen in üblicher Weise um.
Die neuen Verbindungen von basischem Charakter bilden Salze mit anorganischen oder organischen Säu- ren, wie z. B. mit Halogenwasserstoffsäuren, Schwe- felsäuren, Salpeftersäutre, Perchlorsäure, Phosphor- säuren, Ameisensäure, Essigsäure, Propionsäure, Milchsäure, Oxalsäure, Bernsteinsäure, Äpfelsäure, Weinsäure, Zitronensäure, Ascorbinsäufe, Methan- sulfonsäure, Äthlansulfonsäure, Oxyäthansulfonsäure, Benzoesäure, Salicylsäure, p-Aminosalicylsäure, Toluolsulfonsäure oder Naphthalinsulfonsäuren.
Die als Ausgangsstoffe verwendeten Pyrazolpyri- midine können nach dem Verfahren gemä# dem Schweizer Patent Nr. 363 985 her, gestellt werden.
Die erfindungsgemäss erhaltenen Pyrazolopyri- midine oder ihre Salze können wie gesagt als Arznei- mittel Verwendung finden. Diese enthalten die genannten Verbindungen gewöhnlich in Mischung mit einem für die enterale, parenterale oder topicale Applikation geeigneten pharmazeutischen organischen Trägermaterial. Für dasselbe kommen solche Stoffe in Frage, die mit den beschriebenen Verbindungen nicht reagieren, wie z. B. Gelatine, Milchzucker, Stärke, Magnesiumstearat, Talk, pflanzliche Ole, Benzylalkohole, Gummi, Polyalkylenglykole, Vaseline, Cholesterin oder andere bekannte Arzneimittelträger. Die pharmazeutischen Präparate können z.
B. als Tabletten, Dragees, Salben, Cremen oder in flüssiger Form als Lösungen, Suspensionen oder Emulsio- nen vorliegen. Gegebenenfalls sind sie sterilisiert und bzw. oder enthalten Hilfsstoffe, wie Konservierungs-, Stabilisierungs-, Netz-oder Emulgiermittel. Sie können auch noch andere therapeutisch wertvolle Stoffe enthalten.
In den nachfolgenden Beispielen ist die Beziehung zwischen Gewichtsteilen und Volumteilen dieselbe wie zwischen Gramm und Kubikzentimeter. Die Tempe- raturen werden in Celsiusgraden angegeben.
Beispiel 1
23 Gewichsteile 1-phenyl-4-chlor-pyrazolo [3, 4d] pyrimidin und 100 Volumteile flüssiger Ammoniak werden zusammen in einem verschlossenen Rohr während sechs Stunden auf 120 erhitzt. Nach Abdamp fen des flüssigen Ammoniaks bleibt ein weisses Produkt zurück, das aus Methylenchlorid umkristallisiert werden kann. I-Phenyl-4-amino-pyrazolo [3, 4-d] pyri- midin wird so in weissen Kristallen vom F. 205-206 erhalten.

In 200 Volumteile Benzol werden 15 Gewichtsteile Dimethylamin eingeleitet. Die Lösung wird mit 15 Gewichtsteilen I-Phenyl-4-chlor [3, 4-d] pyrazolopyrimidin versetzt und es wird dann während drei Stunden in verschlossenem Rohr auf 100 erhitzt.

2, 2 Gewichtsteile Natrium werden in 200 Volum- teile wasserfreien Alkohol gebracht. Sobald alles Natrium aufgelöst ist, werden 6, 9 Gewichtsteile Hy droxylamin-hydrochlorid zu der Lösung gegeben, und es wird dann eine Stunde bei Zimmertemperatur gerührt. Man fügt hernach 11, 5 GewichtsteNe des ge mäss Beispiel 2 erhältlichen 1-Phenyl-4-chlor-py°- azolo [3, 4-d] pyrimidins zu obiger Lösung, rührt wäh, rend zwei Stunden bei Zimmertemperatur und erhitzt während vier Stunden zum Sieden. Es wird heiss von dem ausgefallenen Kochsalz abgenutscht, und das Filtrat wird im Vakuum zur Trockne eingedampft.
Man kristallisiert den Rückstand aus Alkohol und erhält so 1-phenyl-4-hedroxylamino-pyrazolo[3, 4-d]pyrimidin der Formel

Beispiel 9
In eine Lösung von 12 Gewichtsbeilen Athylendiamin in 200 Volumteilen Alkohol bringt man 11, 5 Gewichtsteile des gemäss Beispiel 2 erhältlichen 1 Phenyl-4-chlor-pyrazolo[3,4-d]pyrimidins und erhitzt während sechs Stunden zum Sieden. Es wird dann von den ausgefallenen Kristallen abgenutscht. Das Filtrat wird im Vakuum zur Trockne eingedampft und der Rückstand in wenig Wasser aufgenommen, worauf ein weisser kristalliner Niederschlag ausfällt, von dem abgenutscht wird.
Man versetzt die beiden kristallinen Niederschläge mit alkoholischer Salzsäure und erhält so ein Hydrochlorid des l-Phenyl4-(ss- amino-äthylamino)-pyrazolo [3, 4-d] pyrimidins der Formel
