Verfahren zur Herstellung von Gramicidin S
Gegenstand der vorliegenden Erfindung ist die Herstellung des Antibiotikums Gramicidin S, eines cyclischen Dekapeptides mit der zweimal auftretenden Aminosäuresequenz (L-Valin-L-ornithin-L-leucin D-phenylalanin-L-prolin).
Gemäss dem Hauptpatent Nr. 324532 werden Amide durch Umsatz von Carbonsäureestern, die in der Alkoholkomponente einen elektronenanziehenden Substituenten aufweisen, mit Aminen erhalten.
Im Schweiz. Zusatzpatent Nr. 363350 ist die Anwendung dieses Verfahrens zur Herstellung linearer oder cyclischer Polypeptide unter Schutz gestellt.
Unter anderem ist darin auch ein Verfahren zur Herstellung des Gramicidin S beschrieben. Es ist dadurch gekennzeichnet, dass man ein Salz eines dem Gramicidin entsprechenden linearen 10gliedrigen Prolinesters, dessen b-Aminogruppen der Ornithinreste geschützt sind und der in der Alkoholkomponente einen elektronenanziehenden Substituenten aufweist, z. B. das Trifluoracetat des entsprechenden Dekapeptidp-nitrophenylesters, mit basischen Mitteln behandelt und sodann die geschützten Aminogruppen in die freien Aminogruppen überführt. Dieses Verfahren ist insofern kompliziert, als das als Ausgangsprodukt verwendete Salz des Dekapeptidesters in mehreren Reaktionsstufen aus dem Pentapeptid hergestellt werden muss.
Es wurde nun gefunden, dass man in einfacher Weise das Gramicidin S erhält, wenn man ein Salz eines L-Valyl-L-ornithyl-L-leucyl-D-phenylalanyl-L- prolin-carbonsäureesters, dessen < 3-Aminogruppe des Ornithinrestes geschützt ist und der in der Alkoholbzw. Phenolkomponente einen elektronenanziehenden Substituenten aufweist, mit basischen Mitteln behandelt und sodann die geschützten b-Aminogruppen in die freien Aminogruppen überführt.
Es war überraschend, dass in diesem Fall nicht Selbstcyclisierung des Pentapeptidmoleküls eintritt, sondern zwei Peptidmoleküle direkt zum cyclischen Dekapeptid zusammengelagert werden. Eine solche Verdoppelung der Ringbausteine bei der Bildung eines cyclischen Polypeptids wurde bisher nur beim c-Hexaglycyl beobachtet, das aus dem linearen Triglycinpeptid z. B. nach dem beschriebenen Verfahren erhalten wird. Man hielt diese Reaktionsweise der Triglycinderivate bisher für eine Ausnahme, die vielleicht in einer besonderen Unstabilität des c-Triglycyls begründet sei (Helv. Chim. Acta 39, 879 [1956]). Andere Tripeptide, wie z. B. das Glycyl DL-phenylalanyl-glycin und das Glycyl-glycyl-DLphenylalanin, ergaben das cyclische Tripeptid und kein Hexapeptid. Auch das Tetraglycinpeptid verdoppelt sein Molekül beim Ringschluss nicht.
Bekannt ist ferner das c-Glycyl-L-leucyl-glycyl-L-leucyl- glycyl, das aus dem Thioester des Pentapeptidhydrobromids durch Selbstcyclisierung erhalten wird.
Schon erwähnt wurde das cyclische Dekapeptid des Gramicidins, das aus dem entsprechenden linearen Dekapeptid ebenfalls ohne Molekülverdoppelung gewonnen wird. Eine Verdoppelung des L-Valyl-L ornithyl - L - leucyl - DL- phenylalanyl-L-prolin-penta- peptides bei der Cyclisierung war daher nicht voraussehbar. Die neue Gramicidin-Synthese stellt wegen ihrer Einfachheit einen bedeutenden technischen Fortschritt dar.
Die Herstellung des als Ausgangsprodukt verwendeten L-Valyl-L-ornithyl-L-leucyl-DL-phenylalanyl- L-prolinesters erfolgt nach den im Hauptpatent Nr. 324532 und im Zusatzpatent Nr. 363350 beschriebenen Verfahren. Die 8-Aminogruppe des Ornithinrestes wird zweckmässigerweise durch Einführung eines Restes, der ohne Ringöffnung abspalt bar ist, wie z. B. der Tosyl-, Trityl-, Carbobenzoxyoder Trifluoracetylrest geschützt.
Elektronenanziehende Substituenten der Alkohol- oder Phenolkomponente der als Ausgangsmaterial verwendeten Ester sind z. B. die Cyano-, Nitro-, Sulfo- oder Methansulfonylgruppe, die vorzugsweise durch eine Kohlenstoffkette von 1-5 C-Atomen vom Estersauerstoff getrennt sind. Beispiele solcher Alkohol- bzw. Phenolkomponenten sind z. B. der Rest des Cyanmethylalkohols, des p-Nitrophenols oder des p-Methansulfonylphenols.
Die als Ausgangsmaterial verwendeten Salze der Peptidcarbonsäureester, die in der Alkoholkomponente einen elektronenanziehenden Substituenten aufweisen, lassen sich beispielsweise durch Umsatz des N-Triphenylmethyl-Pentapeptids mit einem Halogenwasserstoffester des entsprechenden Alkohols in Gegenwart einer tertiären organischen Base und anschliessende Abspaltung des Triphenylmethylrestes durch Behandlung mit verdünnter Säure, oder durch katalytische Hydrierung eines entsprechenden Esters des N-Carbobenzoxy-Pentapeptids in Gegenwart einer Säure herstellen. Als zur erfindungsgemässen Umsetzung geeignete Salze solcher Peptidcarbonsäureester sind insbesondere diejenigen der Halogenwasserstoffsäuren, wie der Salzsäure, oder von halogenierten Fettsäuren, wie der Trifluoressigsäure, zu nennen.
Als alkalische Mittel für die erfindungsgemässe Umsetzung werden z. B. anorganische Basen, wie Alkalihydroxyde oder -carbonate, oder organische Basen, vorzugsweise tertiäre Amine, verwendet.
Das Verfahren wird vorteilhaft in Gegenwart von organischen Lösungsmitteln, wie Dimethylformamid, Acetonitril, Dioxan, Tetrahydrofuran oder deren Gemischen, gegebenenfalls auch in Anwesenheit von Wasser durchgeführt. Die Ausbeute kann durch Zusatz von sauren Katalysatoren, beispielsweise Eisessig oder Schwefelsäure, zum Salz des Peptidesters weitergesteigert werden.
Im folgenden Beispiel sind die Temperaturen in Celsiusgraden angegeben.
Beispiel
340 mg L-Valyl-N8-tosyl-L-ornithyl-L-leucyl-D- phenylalanyl - L - prolin-nitrophenylester-trifluoracetat werden in 6 ml Dimethylformamid gelöst, mit 6 Tropfen Eisessig versetzt und in 5 Stunden unter Rühren bei 55O in 70 ml Pyridin eingetropft. Anschliessend wird noch 2 Stunden bei dieser Temperatur gerührt, dann werden die Lösungsmittel im Vakuum vollständig verdampft. Der Rückstand wird in 60 ml Methanol-Isopropanol-Wasser 1:1:1 gelöst und durch je eine Säule mit stark saurem und basischem Jonenaustauscher filtriert. Dieser wird noch mit 200 ml des gleichen Gemisches ausgewaschen; die vereinigten Filtrate werden im Vakuum zur Trockne verdampft. Der Rückstand wird aus Methanol-Wasser kristallisiert, wobei 125 mg Di- < 3tosyl-gramicidin S in Form des Dihydrates erhalten werden.
Die Mutterlauge wird zur Trockne verdampft und der Rückstand an 2 g Aluminiumoxyd chromatographiert. Mit Chloroform und mit Essigester lassen sich 20 mg Rohprodukt eluieren; diese ergeben nach Umkristallisation aus Methanol-Wasser noch 10 mg reines Di-tosyl-gramicidin-S-dihydrat. Die Gesamt ausbeute beträgt demnach 135 mg = 31 /o d. Th.
Dieses Produkt wird wie folgt in das Gramicidin S umgewandelt:
60 mg des Dihydrates des Di-tosyl-gramicidin S werden in 20 ml flüssigem Ammoniak gelöst und mit 100 mg Natrium (in kleinen Stücken) versetzt. Nach vollständiger Lösung des Natriums bleibt die Lösung blau. Nach Zugabe von wenig Ammoniumchlorid wird der Ammoniak verdunstet und der Rückstand im Hochvakuum von entstandenem Merkaptan befreit. Das Reaktionsprodukt wird mit absolutem Äthanol, dem einige Tropfen einer Lösung von Salzsäure in Essigester beigefügt sind, aufgenommen. Das Lösungsmittel wird nach der Filtration verdampft.
Der Rückstand wird in wenigen Tropfen 65proz.
Äthanol gelöst und bei 50O so lange mit Wasser versetzt, bis eine deutliche Trübung entstanden ist.
Darauf kristallisiert das Dihydrochlorid des Gramicidin S in Form feiner Nadeln. Smp. 268-2700 unter Zersetzung (Einführung des Smp.-Röhrchens bei 2500).
Das als Ausgangsstoff verwendete trifluoressigsaure Salz des L-Valyl-Na-tosyl-L-ornithyl-L-leucyl- D-phenylalanyl-L-prolin-p-nitrophenylesters kann wie folgt erhalten werden:
500 mg N-Trityl-L-valyl-(N5-tosyl)-L-ornithyl-L- leucyl-D-phenylalanyl-L-prolin (dargestellt z. B. nach Zusatzpatent Nr. 363350) und 800 mg Di-(p-nitrophenyl)-sulfit werden in 1,5 ml abs. Pyridin 20 Stunden bei Zimmertemperatur stehengelassen. Das Reaktionsgemisch wird dann mit Essigester verdünnt, dreimal mit einer Citronensäurelösung, dann mit Wasser ausgeschüttelt, über Natriumsulfat getrocknet und im Vakuum zur Trockne verdampft. Das zurück bleibende Harz wird mit Äther-Petroläther 1 : 1 zu 1 ver- rieben, wobei der Nitrophenylester fest zurückbleibt.
Die Ausbeute beträgt 550 mg = 98/0 d. Th.
435 mg des so erhaltenen Nitrophenylesters werden in 10 ml Trifluoressigsäure gelöst und bei-5O langsam mit 10 ml Wasser versetzt. Es bildet sich eine Fällung von Triphenylcarbinol. Die Lösungsmittel werden im Hochvakuum bei 30O vollständig verdampft, dann der Rückstand mehrmals mit Ather Petroläther 1:1 und abs. Äther verrieben. Als feste Fällung werden 340 mg Trifluoracetat erhalten.
Process for the preparation of gramicidin S
The present invention relates to the production of the antibiotic gramicidin S, a cyclic decapeptide with the amino acid sequence occurring twice (L-valine-L-ornithine-L-leucine D-phenylalanine-L-proline).
According to the main patent No. 324532, amides are obtained by reacting carboxylic acid esters, which have an electron-withdrawing substituent in the alcohol component, with amines.
In Switzerland. Additional patent no. 363350 is the application of this process for the production of linear or cyclic polypeptides under protection.
Among other things, a method for producing gramicidin S is also described therein. It is characterized in that a salt of a linear 10-membered proline ester corresponding to gramicidin, whose b-amino groups of the ornithine residues are protected and which has an electron-withdrawing substituent in the alcohol component, e.g. B. the trifluoroacetate of the corresponding decapeptide p-nitrophenyl ester, treated with basic agents and then converted the protected amino groups into the free amino groups. This process is complicated in that the salt of the decapeptide ester used as the starting product has to be prepared from the pentapeptide in several reaction stages.
It has now been found that gramicidin S is obtained in a simple manner if a salt of an L-valyl-L-ornithyl-L-leucyl-D-phenylalanyl-L-proline carboxylic acid ester whose <3-amino group of the ornithine residue is protected and the one in the alcohol or Phenol component has an electron-withdrawing substituent, treated with basic agents and then converted the protected b-amino groups into the free amino groups.
It was surprising that in this case the pentapeptide molecule does not self-cyclize, but rather two peptide molecules are directly combined to form the cyclic decapeptide. Such a doubling of the ring building blocks in the formation of a cyclic polypeptide has so far only been observed in c-hexaglycyl, which is derived from the linear triglycine peptide z. B. is obtained by the method described. This mode of reaction of the triglycine derivatives has hitherto been regarded as an exception, perhaps due to a particular instability of the c-triglycyl (Helv. Chim. Acta 39, 879 [1956]). Other tripeptides such as B. the glycyl DL-phenylalanyl-glycine and the glycyl-glycyl-DLphenylalanine, gave the cyclic tripeptide and no hexapeptide. The tetraglycine peptide also does not double its molecule upon ring closure.
Also known is c-glycyl-L-leucyl-glycyl-L-leucyl-glycyl, which is obtained from the thioester of pentapeptide hydrobromide by self-cyclization.
The cyclic decapeptide of gramicidin, which is also obtained from the corresponding linear decapeptide without doubling the molecule, has already been mentioned. A doubling of the L-valyl-L ornithyl-L-leucyl-DL-phenylalanyl-L-proline-pentapeptide during the cyclization was therefore not foreseeable. The new gramicidin synthesis represents a significant technical advance because of its simplicity.
The L-valyl-L-ornithyl-L-leucyl-DL-phenylalanyl-L-proline ester used as the starting product is produced according to the processes described in the main patent no. 324532 and in the additional patent no. 363350. The 8-amino group of the ornithine residue is conveniently removed by introducing a residue that can be split off without ring opening, such as. B. the tosyl, trityl, carbobenzoxy or trifluoroacetyl radical.
Electron-attracting substituents of the alcohol or phenol component of the ester used as the starting material are, for. B. the cyano, nitro, sulfo or methanesulfonyl group, which are preferably separated from the ester oxygen by a carbon chain of 1-5 carbon atoms. Examples of such alcohol or phenol components are z. B. the remainder of the cyanomethyl alcohol, the p-nitrophenol or the p-methanesulfonylphenol.
The salts of the peptide carboxylic acid esters used as starting material, which have an electron-withdrawing substituent in the alcohol component, can be obtained, for example, by reacting the N-triphenylmethyl pentapeptide with a hydrogen halide ester of the corresponding alcohol in the presence of a tertiary organic base and then splitting off the triphenylmethyl radical by treatment with dilute acid , or by catalytic hydrogenation of a corresponding ester of N-carbobenzoxy-pentapeptide in the presence of an acid. Salts of such peptide carboxylic acid esters suitable for the reaction according to the invention are in particular those of hydrohalic acids, such as hydrochloric acid, or of halogenated fatty acids, such as trifluoroacetic acid.
As an alkaline agent for the inventive implementation z. B. inorganic bases, such as alkali metal hydroxides or carbonates, or organic bases, preferably tertiary amines, are used.
The process is advantageously carried out in the presence of organic solvents such as dimethylformamide, acetonitrile, dioxane, tetrahydrofuran or mixtures thereof, optionally also in the presence of water. The yield can be increased further by adding acidic catalysts, for example glacial acetic acid or sulfuric acid, to the salt of the peptide ester.
In the following example the temperatures are given in degrees Celsius.

340 mg of L-valyl-N8-tosyl-L-ornithyl-L-leucyl-D-phenylalanyl-L-proline-nitrophenyl ester-trifluoroacetate are dissolved in 6 ml of dimethylformamide, 6 drops of glacial acetic acid are added and in 5 hours with stirring at 55O in 70 ml of pyridine were added dropwise. The mixture is then stirred for a further 2 hours at this temperature, then the solvents are completely evaporated off in vacuo. The residue is dissolved in 60 ml of methanol-isopropanol-water 1: 1: 1 and filtered through a column with a strongly acidic and a basic ion exchanger. This is washed out with 200 ml of the same mixture; the combined filtrates are evaporated to dryness in vacuo. The residue is crystallized from methanol-water, 125 mg of di- <3tosyl-gramicidin S being obtained in the form of the dihydrate.
The mother liquor is evaporated to dryness and the residue is chromatographed on 2 g of aluminum oxide. 20 mg of crude product can be eluted with chloroform and with ethyl acetate; after recrystallization from methanol-water, these give a further 10 mg of pure di-tosyl-gramicidin-S-dihydrate. The total yield is accordingly 135 mg = 31 / o d. Th.
This product is converted into the gramicidin S as follows:
60 mg of the dihydrate of di-tosyl-gramicidin S are dissolved in 20 ml of liquid ammonia and mixed with 100 mg of sodium (in small pieces). After the sodium has completely dissolved, the solution remains blue. After adding a little ammonium chloride, the ammonia is evaporated and the residue is freed from the mercaptan formed in a high vacuum. The reaction product is taken up with absolute ethanol to which a few drops of a solution of hydrochloric acid in ethyl acetate are added. The solvent is evaporated after filtration.
The residue becomes 65 per cent in a few drops.
Dissolved ethanol and mixed with water at 50O until a clear cloudiness has arisen.
The dihydrochloride of gramicidin S then crystallizes in the form of fine needles. M.p. 268-2700 with decomposition (insertion of the m.p. tube at 2500).
The trifluoroacetic acid salt of L-valyl-Na-tosyl-L-ornithyl-L-leucyl-D-phenylalanyl-L-proline-p-nitrophenyl ester used as starting material can be obtained as follows:
500 mg N-trityl-L-valyl- (N5-tosyl) -L-ornithyl-L-leucyl-D-phenylalanyl-L-proline (shown e.g. according to additional patent no. 363350) and 800 mg di- (p -nitrophenyl) sulfite in 1.5 ml of abs. Pyridine left to stand for 20 hours at room temperature. The reaction mixture is then diluted with ethyl acetate, extracted three times with a citric acid solution, then extracted with water, dried over sodium sulfate and evaporated to dryness in vacuo. The remaining resin is rubbed 1: 1 to 1 with ether-petroleum ether, the nitrophenyl ester remaining solid.
The yield is 550 mg = 98/0 d. Th.
435 mg of the nitrophenyl ester obtained in this way are dissolved in 10 ml of trifluoroacetic acid and 10 ml of water are slowly added at −50. A precipitate of triphenylcarbinol forms. The solvents are completely evaporated in a high vacuum at 30O, then the residue is repeatedly washed with ether, petroleum ether 1: 1 and abs. Ether rubbed in. 340 mg of trifluoroacetate are obtained as a solid precipitate.
