4-Methyl-4-hydroxy-hexanon- (3), 4-Athyl-4-hydroxy-hexanon- (3),
4-Methyl-4-hydroxy-heptanon- (3),
4,5-Dimethyl-4-hydroxy-hexanon- (3) sowie die 2-Cycloalkyl-2-hydroxy-butanone- (3) und 3-Cycloalkyl-3-hydroxy-pentanone- (4) mit 3-7 C-Atomen im Cycloalkylrest genannt.
Zu a-Ketolen der Formel (II), worin Ri und R2 zusammen einen der genannten Alkylenreste bedeuten und worin R3 durch die Methylgruppe verkörpert ist, gelangt man beispielsweise durch Kondensation von entsprechenden cycloaliphatischen Ketonen mit Alkalimetallacetyliden und Wasseranlagerung an die so erhaltenen Athinyl-cycloalkanole oder durch die (oben erwähnte) Umsetzung von entsprechenden K-Hydroxy-carbonsäuren mit Methyllithium ;
Process for the preparation of bis-tertiary glycols Bis-tertiary glycols of the formula (I)
EMI1.1
where R1 is an alkyl radical with 1-3 carbon atoms or a cycloalkyl radical with 3-7 carbon atoms, R2 is the methyl or ethyl group or R, and R2 together is also an alkylene radical with 4-5 carbon atoms and 4 carbon atoms between the two free valences and Rs is an alkyl radical with 1-4 carbon atoms, where R3 has at least 2 carbon atoms when Ri stands for an unbranched alkyl radical with 1-3 carbon atoms, have not yet become known.
As has now been found, these compounds have excellent hypnotic, narcotic, sedative and anticonvulsant activity. They can also be used as intermediate products for the production of other pharmacologically valuable substances.
The present patent now relates to a process for the preparation of the glycols defined above, which is characterized in that an α-hydroxyketone (α-ketol) of the formula (II)
EMI1.2
condensed with a mono-organometallic acetylene compound. Monometal acetylides can be used as such acetylene compounds and in this case the condensation can be carried out, for example, in liquid, dry ammonia or in anhydrous, organic solvents such as methylal, diethyl ether or a mixture thereof, formamide, dimethyl sulfoxide or dimethylformamide. Of the monometal acetylides, in particular monoalkali metal acetylides can be used which, for.
B. be formed in situ either before the reaction by reacting acetylene with the metals dissolved in the ammonia or during the reaction. In the latter case, for example, acetylene is used with the a-ketol in an organic solvent in the presence of a finely suspended alkali metal hydroxide under normal or slightly elevated pressure, e.g. At 1-1.33 atmospheres.
An acetylene monomagnesium halide can also be used as a mono-organometallic acetylene compound and this can be mixed with an α-hydroxy-ketone of the formula (II) defined above, e.g. B. in an ether as a solvent, such as tetrahydrofuran. A suspension of the Grignard component in tetrahydrofuran is z. B. obtained by adding portions of a solution of ethyl magnesium bromide in tetrahydrofuran to a saturated solution of acetylene in tetrahydrofuran while continuously passing in acetylene (E. R. H. Jones, L. Skatteböl and M. C.
Whiting, J. Chem. Soc. 1956, 4765-68). In this case, the further addition of magnesium compound is best carried out only after the evolution of ethane has ended and the reaction mixture has been saturated again with acetylene, so that the latter is always present in excess.
The presence of a free hydroxyl group in the ketone component naturally means that both z. B. mono-metal acetylide, such as. B. acetylene monomagnesium halide, must be used in at least a molar excess.
The a-ketols of the formula (II), in which R2 and Rg are embodied by the methyl group, are obtained, for example, by condensation of suitable methyl ketones of the formula R, -CO-CH3 with alkali metal acetylides and addition of water, e.g. . B. in the presence of mercury and sulfuric acid, to the thus obtained Athinyl-R, -methyl-carbinole.
Furthermore, the (von Billimoria et al., J. Chem. Soc.
1951, 3067, described) implementation of corre sponding a-hydroxy-carboxylic acids with methyllithium mentioned. α-KetoIe of the formula (II) can also be obtained from ketones of the formula R1COR2.
These are first converted into corresponding cyanohydrins, the hydroxyl group of which is, for example, acylated or reacted with dihydropyran or a vinyl ether for closure. The a-acyloxy-, a-tetrahydropyranyloxy- or a- (a'-alkoxy-ethoxy) -alkanecarboxonitriles obtained in this way can be converted with alkylmagnesium halides of the formula R3-Mg-halogen to imines, from which the a-hydroxyketones of the formula are converted by hydrolysis (II) are available.
Examples of such compounds are 4-methyl-4-hydroxy-pentanone- (3),
4-methyl-4-hydroxy-hexanone- (3), 4-ethyl-4-hydroxy-hexanone- (3),
4-methyl-4-hydroxy-heptanone- (3),
4,5-dimethyl-4-hydroxy-hexanone- (3) and the 2-cycloalkyl-2-hydroxy-butanones- (3) and 3-cycloalkyl-3-hydroxy-pentanones- (4) with 3-7 C- Atoms in the cycloalkyl radical called.
A-ketols of the formula (II), in which Ri and R2 together represent one of the alkylene radicals mentioned and in which R3 is represented by the methyl group, are obtained, for example, by condensation of corresponding cycloaliphatic ketones with alkali metal acetylides and addition of water to the ethynyl-cycloalkanols thus obtained or by the (above-mentioned) reaction of the corresponding K-hydroxycarboxylic acids with methyl lithium;
or, if R3 represents an alkyl radical with 2-4 carbon atoms, by reacting cyanohydrins of suitable cycloaliphatic ketones (after closing the hydroxyl group as described above) with corresponding low molecular weight alkyl magnesium halides to form imines, from which the corresponding α-hydroxy ketones are hydrolyzed of formula (II) are available. Examples of such compounds are 1-acetyl-, 1-propionyl-, 1-butyryl- and 1-isobutyryl-cyclopentanol and 1-acetyl-2-methyl-cyclopentanol.
In the following examples, the temperatures are given in degrees Celsius.
example 1
50 g of finely powdered potassium hydroxide are added to 50 ml of distilled methylal and 50 ml of dry diethyl ether in a stirred flask with a capacity of 350 ml, equipped with a condenser, stirrer, toluene thermometer, gas inlet tube and dropping funnel, and the suspension is passed to -10 to 0 while stirring and cooling Indoor temperature dry acetylene to saturation. Then 13.0 g of 3,4-dimethyl-3-hydroxy-pentanone- (2) are added dropwise under a stronger stream of acetylane over the course of 10 minutes, and the mixture is then stirred at the same temperature for a further two hours with further introduction of acetylene.
The reaction mixture is then made with acetic acid lacquer wall, the ether-methylal layer is separated off and the acidic, aqueous solution is extracted with ether. The ether extracts and the ether-methylal layer are washed neutral, alone or in combination, dried over sodium sulfate and freed from the solvents. During vacuum distillation of the residue, the 3,4,5-trimethyl-3,4-dihydroxy-hexyne- (1) passes over under 13 mm pressure at 98-99.
Example 2 a) 11.3 g of n-propyl-methyl-ketone-cyanohydrin are mixed with 7.2 g of vinyl ethyl ether. Then 3-5 drops of ethereal hydrogen chloride solution are added, whereupon the temperature increases significantly. After standing for one hour, the reaction solution is neutralized with anhydrous sodium carbonate and the inorganic salts are filtered off.
b) In a stirred flask of 350 ml capacity equipped with a condenser, thermometer, dropping funnel and nitrogen transfer tube, the ethereal solution of a) obtained according to a) is converted into ethylmagnesium bromide from 2.9 g of magnesium and 15 g of ethyl bromide in 50 ml of ether in a nitrogen atmosphere and with stirring a-Methyl-a- (cct-ethoxy-ethoxy) -valeronitri was added dropwise at a weak Sither reflux and the reaction mixture was then stirred for five hours at the boiling point of the ether. Then it is decomposed with 50 ml of water while cooling with ice and acidified to the Congo with 50 ml sulfuric acid.
Organic fractions are then distilled off up to a boiling point of 60 and the remaining solution is stirred for 10-15 hours in a bath of 80. After cooling, it is etherified, the ether extract is washed neutral, dried over sodium sulfate and the solvent is evaporated off. The residue is distilled in vacuo.
After rectifying twice, 4-methyl-4-hydroxy heptanone- (3) from Kpl2 7071 is obtained. c) 50 g of finely powdered potassium hydroxide are added to 50 ml of distilled methylal and 50 ml of dry diethyl ether in a stirred flask of 350 ml content equipped with a condenser, stirrer, thermometer, gas inlet tube and dropping funnel, and the suspension is passed at -10 with stirring and cooling up to 0 internal temperature, add dry acetylene to saturation. Then 13.0 g of 4-methyl-4-hydroxy-heptanone- (3) (Kp. 70-71) are added dropwise under a stronger stream of acetylene in the course of 10 minutes, and the mixture is then continued at the same temperature for two hours pass acetylene stirred.
The reaction mixture is then made with acetic acid lacquer wall, the ether-methylal layer is separated off and the acidic aqueous solution is extracted with ether. The ether extracts and the Sither-l \ tethylal layer are combined and washed neutral, dried over sodium sulfate and freed from the solvents. In the vacuum distillation of the residue, the 3-ethyl-4-methyl-3, 4-dihydroxy-heptin- (1) passes over under 12 mm pressure at 105-106.
In an analogous manner, starting from acetone cyanohydrin, 3-ethyl-4-methyl-3, 4-dihydroxy-pentyn- (l) from Kpt5 8889O is obtained.
Example 3 a) 9.9 g of methyl ethyl ketone cyanohydrin are heated to about 50-60 with 20 ml of distilled acetic anhydride and 0.5 ml of acetyl chloride in a water bath and then left to stand for half an hour. The excess acetic anhydride and the acetyl chloride are then distilled off in vacuo and the residue is rectified. The α-methyl-α-acetoxy-butyronitrile liberates at 74-76 under 12 mm pressure. b) and c) An ethereal solution of 14.1 parts of this compound is used in the Grignard reaction given in Example 2 under b) instead of the solution of the a-methyl-a- (a'-ethoxy-ethoxy) -valeronitrile.
Analogously to Example 2, b) and c), 3-ethyl-4-methyl-3, 4-dihydroxyhexyne- (I) of Kpl2 97980, melting point 33-35, is obtained therefrom.
Example 4 a) 11.3 g of diethyl ketone cyanohydrin and 8.4 g of dihydropyran are admixed with 5 drops of ethereal hydrogen chloride solution, with heating occurring. After 10-15 hours of standing at room temperature, the reaction solution is diluted with ether, shaken with bicarbonate solution and, after drying over sodium sulfate, freed from ether. The vacuum distillation of the residue gives the a-ethyl-a-tetrahydropyranyloxy-butyronitrile of Bp3 1101142. b) and c) An ethereal solution of 19.7 g of this compound is used in the Grignard reaction of Example 2 under b) and the reaction product then further processed according to c).
In this way, the 3,4-diethyl-3,4-dihydroxy hexyne- (l) of boiling point 12 106-107, melting point 36-38 is obtained.
Example 5
100 ml of dry ammonia are placed in a 350 ml stirred flask equipped with a condenser, stirrer, toluene thermometer, gas inlet tube and dropping funnel, which is cooled to below -40 in a carbon dioxide-acetone bath. At -40 to -50 initially 0.1 g of crystalline Ferric nitrate was added and then 0.4 g of sodium. After the initially appearing deep blue color has turned gray, a further 4.6 g of sodium are added in fine pieces. When the deep blue color has turned gray again, stirring is continued for half an hour. About 5 liters of dry acetylene gas are then passed in and then 12.8 g of 1-acetyl-cyclopentanol in 20 ml of absolute ether are added dropwise over the course of 10 minutes, with further passing in of acetylene.
After the end of the dropwise addition, stirring is continued for one hour while constantly passing in acetylene. The temperature is then allowed to rise to about 20 within 10-15 hours, the mass being kept stirrable by adding absolute ether.
The reaction mixture is then stirred for two hours with warming under reflux of ether, then 50 ml of saturated ammonium chloride solution is added at 0 and hydrolyzed and then made Congo acidic with 50% sulfuric acid. The reaction product is taken up in ether and the ethereal solution is concentrated after washing neutral and drying over magnesium sulphate. The vacuum distillation of the residue gives 3-methyl-4,4-tetramethylene-3,4-dihydroxy-butyne (1) of Kpl4 122-124, melting point 6667.
Example 6
100 ml of dry ammonia are placed in a 350 ml flask equipped with a stirrer, condenser, toluene thermometer, gas inlet tube and dropping funnel, which is cooled to below -40 in a carbon dioxide-acetone bath. The subsequent reactions are carried out at internal temperatures between -40 and -50. While stirring, 0.1 g of crystalline first. Ferric nitrate, then 0.4 g sodium added. After the initially appearing deep blue color has turned gray, another 4.6 g of sodium are added in fine pieces. After the color has changed from blue to gray again, stirring is continued for half an hour.
About 5 liters of acetone-free, dry acetylene are then passed in and then, with further passing in of acetylene, 12.8 g of 2-cyclopropyl-2-hydroxy-butanone- (3), prepared from cyclopropyl-methyl-ethinyl-carbinol by the usual addition of water, are passed in within 15 minutes in the presence of mercury salts and sulfuric acid (boiling point ii: 60), added dropwise in 20 ml of absolute ether. When the addition of ketol has ended, stirring is continued for one hour while constantly passing in acetylene.
The reaction mixture is now mixed with 20 g of solid ammonium chloride in five portions and the temperature is allowed to rise to about 20 within 10-15 hours, the mixture being kept stirrable by adding ether. Then the inorganic salts are dissolved by adding the necessary amount of water and the organic layer is concentrated after washing until neutral and drying over sodium sulfate. The residue is distilled in a water jet vacuum and the fraction of bp 11 97-104 rectified with the help of a good column, the 3-methyl-4-cyclopropyl-3, 4 dihydroxy-pentin- (l) from Kplo 10311, n26o: 1.4822 received.
Example 7
A solution of ethylmagnesium bromide in tetrahydrofuran is made in the usual way from 7.0 g of magnesium turnings, 32.0 g of distilled water. Ethyl bromide and 200 ml of dry tetrahydrofuran prepared under nitrogen. The still warm Grignard solution is decanted in an inert atmosphere into a dropping funnel of a dry stirred flask with a volume of 11/2 l (equipped with condenser, stirrer, thermometer and acetylene inlet tube). 120 ml of dry tetrahydrofuran are placed in the flask and acetone-free acetylene gas is passed in with thorough stirring until it is saturated. With further introduction of acetylene, the Grignard solution is allowed to flow in in about 5 ml portions, each time vigorous foaming occurs.
After the addition has ended, the reaction mixture is saturated with acetylene at 25 ° C, whereupon an easily stirrable suspension of the ethynylmagnesium bromide in tetrahydrofuran is formed on cooling with the aid of an ice bath.
Then the solution of 14.4 g of 3-methyl-3-hydroxy-heptanone- (4) vom Kpl2 70-71 in 15 ml of abs. Tetrahydrofuran was added dropwise within 1 hour. The introduction of acetylene is stopped and the contents of the flask are stirred for about 12-15 hours at room temperature (about 20 "). Ammonium chloride is now decomposed in the usual way, and 3-methyl-4-ethinyl is obtained after vacuum distillation of the residue freed from tetrahydrofuran -3, 4-dihydroxy-heptane of bp 1 105-108.
In an analogous manner, 3-methyl-4-ethinyl-3, 4-dihydroxy-octane from Kip 12 117 is obtained from 3-methyl-3-hydroxy-octanone- (4) of bp 2 85860 after one of the ethynylation described above 120.
