Verfahren zur Herstellung von Steroiden Vorliegende Erfindung bezieht sich auf ein Ver fahren zur Herstellung von 6-Alkyl(oder 6-Aryl)- llss-oxy(oder 11-oxo)-17a-oxy-1,4-pregnadien-3,20- dionen, insbesondere der 6-Methylderivate. Das Verfahren wird durch folgende Formeln veranschaulicht:

in denen R einen Alkyl- oder Arylrest, wie Methyl, Äthyl, Propyl, Tolyl, Naphthyl usw., und vorzugs weise Methyl, bedeutet.
Das erfindungsgemässe Verfahren ist dadurch ge kennzeichnet, dass man ein 6-Alkyl(oder 6-Aryl)- 11ss - oxy(oder 11- oxo) -17a - oxy - 21-halogen-1,4- pregnadien-3,20-dion enthalogeniert. Die Enthalo- genierung kann mit einem reduzierenden Mittel, wie Natriumthiosulfat, Natriumbisulfit,
Kaliumbisulfit oder dergleichen in wässrig-organischem Lösungsmit- telgemisch durchgeführt werden. Die auf diese Weise hergestellten 11-Oxyverbindungen lassen sich ge- wünschtenfalls mit Chromsäureanhydrid, N-Brom- acetamid, N-Bromsuccinimid oder dergleichen zum 6-Alkyl(oder 6-Aryl)-17a-oxy-1,4-pregnadien-3,11,20- trion oxydieren.
Die 6a-Epimeren stellen die bevorzugten Pro dukte des erfindungsgemässen Verfahrens dar. Die erfindungsgemäss erhältlichen Steroidverbin- dungen besitzen hohe physiologische Wirksamkeit und Wirkungsbereiche, die von den in der Natur vor kommenden Nebennierenrindenhormonen, wie Hy- drocortison und Cortison, insbesondere hinsichtlich ihrer Wirkung auf den Mineral- und Wasserstoff wechsel verschieden sind.
Diese Verbindungen be wirken einen Verlust an Salz und Wasser, die sie speziell zur Behandlung chronisch kongestiver Herz schwächen, der Lebercyrrhose, nephrytischer Syn drome sowie Eclampsia und Praeeclampsia geeignet machen.
Die erfindungsgemäss erhältlichen neuen Hormone haben auch entzündungshemmende, glucocorticoide, anästhetische, wachstumshemmende sowie adreno- corticoide Wirksamkeit. Ferner wirken sie auf Ge bärmutter, Eierstock und Nebennieren. Die ent zündungshemmende Wirkung ist besonders beach- tenswert. Die 6ss-Epimeren haben: das gleiche Wir kungsspektrum wie die 6a-Epinieren.
Die neuen Steroidhormone eignen sich zur Her stellung von Präparaten für die orale, parenterale und topische Verabreichung, vorzugsweise in Kombi nation mit Antibiotika.
Die 6 - Methyl -11 ss,17a - dioxy-1,4-pregnadien- 3,20-dione und ihre 11-Keto-analoga sind auch wert volle Ausgangsstoffe zur Herstellung anderer physio logisch aktiver Verbindungen. So zeigen z.
B. die daraus herstellbaren 9a-Halogen-analoga, insbeson dere 6a-Methyl-9a-fluor-llss,17a-dioxy-1,4-pregna- dien-3,20-dion und das entsprechende 17a-Oxy- 3,11,20-trion entzündungshemmende und gluco- corticoide Wirkung und können anstelle der halogen freien Verbindungen verwendet werden.
Die als Ausgangsstoffe für das vorliegende Ver fahren verwendbaren 6-Alkyl(oder 6-Aryl)-llss-oxy- (oder 11-oxo) -17a - oxy-21-halogen-1,4-pregnadien- 3,20-dione können gemäss Schweiz. Patentschrift Nr. 351598 und Präparationen 1 bis 5 hergestellt werden.
Bei der Durchführung des erfindungsgemässen Verfahrens wird z. B. rohes oder gereinigtes 6-Alkyl- (oder 6-Aryl)-llss,l7a-dioxy-21-jod-1,4-pregnadien- 3,20-dion mit einem Reduktionsmittel, wie Natrium- oder Kaliumthiosulfat, Natriumbisulfit oder derglei chen, behandelt.
Gemäss einer bevorzugten Aus führungsform wird das 21-Halogensteroid in Essig säure suspendiert, mit einer wässrigen Lösung des Reduktionsmittels versetzt und bei Zimmertemperatur 10 Minuten bis 2 Stunden gerührt. Das Produkt wird aus der wässrigen Lösung nach üblichen Methoden, wie Filtration oder Extraktion mit wasserunlöslichen organischen Lösungsmitteln, wie Äther, Benzol, Me- thylenchlorid, Äthylenchlorid, Tetrachlorkohlenstoff, Chloroform, Hexan,
Heptan usw., und Verdampfung des Extraktes, isoliert. Die Reinigung des so erhal tenen 6 - Alkyl(oder 6 - Aryl) -11<B>ss,</B> 17a<I>-</I> dioxy - 1,4 pregnadien-3,20-dions erfolgt in. üblicher Weise, wie z. B. Umkristallisieren aus Äther, Aceton, Methanol, Äthanol, Skellysolve B-Hexan, Mischungen derselben usw., oder chromatographisch.
Das so erhaltene 6-Alkyl(oder 6-Aryl)-llss,17a- dioxy-1,4-pregnadien-3,20-dion kann gewünschten falls in üblicher Weise, wie z. B. durch Umsetzung mit Chromsäureanhydrid in Essigsäurelösung, wobei man die berechnete Menge oder einen geringen über schuss von 10 bis 30 /o an Chromsäureanhydrid ver wendet, oder mit einem Halogenamid oder Halogen imid, wie N-Bromacetamid, N-Chlorsuccinimid, N-Bromsuccinimid in Pyridin,
Dioxan oder andern Lösungsmitteln, oxydiert werden. Nach Beendigung der Oxydation wird überschüssiges Oxydationsmittel zerstört, z. B. durch Zugabe von Methyl- oder Athyl- alkohol, wenn Chromsäure verwendet wurde, oder von Alkalibisulfit bei Verwendung von Chromsäure oder N-Bromsuccinimid oder andern Halogenacyl- imiden. Dann wird das 6-Alkyl(oder 6-Aryl)-17a- oxy-1,4-pregnadien-3,
11,20-trion in üblicher Weise, z. B. durch Extraktion mit in Wasser unlöslichen Lösungsmitteln, z. B. Methylen- oder Äthylenchlorid, Chloroform, Tetrachlorkohlenstoff, Äther, Benzol, Toluol oder dergleichen, oder, falls erforderlich, chromatographisch gewonnen.
Präparation <I>1</I> Eine Lösung von 164 mg (0,437 Millimol) 1-De- hydro-6a-methylhydrocortison (siehe Schweiz. Pa tentschrift Nr. 351598) in 1 ml Pyridin wird auf 0 gekühlt und mit einer ebenfalls gekühlten Lösung von 75 mg Methansulfonylchlorid in 0,5 ml Pyridin behandelt, wonach man zwei Stunden bei 0 bis 5 stehenlässt.
Dann setzt man Eis und genügend Salz säure zu, um das Pyridin zu neutralisieren, und extrahiert dreimal mit je 25 ml Methylenchlorid. Die Extrakte werden vereinigt, mit kalter Natrium bicarbonatlösung, dann mit Wasser gewaschen, über wasserfreiem Natriumsulfat getrocknet und unter ver mindertem Druck eingedampft. Man erhält so kri stallines 6a-Methyl-llss,17a,21-trioxy-1,4-pregnadien- 3,20-dion-21-methansulfonat.
Präparation <I>2</I> Das kristalline Methansulfonat des 6a-Methyl- 11ss,17a,21-trioxy-1,4-pregnadien-3,20-dions nach Präparation 1 wird in 15 ml Aceton gelöst und mit einer Lösung von 200 mg Natriumjodid in 5 ml Aceton versetzt. Die Mischung wird unter Rühren 15 Minuten am Rückfluss erhitzt. Dann wird das Erhitzen unterbrochen und unter vermindertem Druck zur Trockne eingedampft. Man erhält 6a-Methyl- 11ss,17a-dioxy-21-jod-1,4-pregnadien-3,20-dion.
Präparation <I>3</I> In gleicher Weise wie in Präparation 1 erhält man durch Behandlung von 1-Dehydro-6a-äthyl- hydrocortison, in Pyridin gelöst, mit Toluolsulfonyl- chlorid das 6a-Äthyl-llss,17a,21-trioxy-1,4-pregna- dien-3,20-dion-21-toluolsulfonat.
Präparation <I>4</I> In gleicher Weise wie in Präparation 2 erhält man durch Kochen von 6a-Äthyl-llss,17a,21-trioxy- 1,4-pregnadien-3,20-dion-21-toluolsulfonat mit Ka- triumjodid in Aceton am Rückfluss das 6a-Äthyl- l lss,17a-dioxy-21-jod-1,4-pregnadien-3,20-dion.
Präparation <I>5</I> In gleicher Weise wie in Präparation 1 erhält man durch Behandlung von 1-Dehydro-6a-methyl- cortison mit Methansulfonylchlorid in Pyridinlösung 6a - Methyl -17a,21- dioxy-1,4 - pregnadien-3,11,20- trion-21-methansulfonat,
das durch Kochen mit Ka- liumjodid in Aceton am Rückfluss 6a-Methyl-17a- oxy-21-jod-1,4-pregnadien-3,11,20-trion liefert.
<I>Beispiel 1</I> Das nach Präparation 2 erhaltene rohe Produkt wird in 5 ml Essigsäure aufgeschlämmt und 45 Mi- nuten gerührt. Dann gibt man eine wässrige Lösung von Natriumthiosulfat-pentahydrat (250 mg) zu, worauf die Jodfarbe verschwindet. Nach Zugabe von 50 ml Wasser wird dreimal mit je 25 ml Methylen- chlorid extrahiert. Die Methylenchloridextrakte wer den vereinigt und mit Wasser und kalter Natrium- bicarbonatlösung gewaschen, bis alle Essigsäure neu tralisiert ist.
Nachdem über wasserfreiem Natrium sulfat getrocknet wurde, wird auf etwa 15 ml ein geengt und über 10 g Florisil (synthetisches Magne- siumsilikat) chromatographiert, wobei man folgende Fraktionen zu je 50 ml auffängt:
9 Fraktionen Aceton-Skellysolve B-Hexane mit 101/o Aceton, 4 Fraktionen Aceton-Skellysolve B-Hexane mit 15 % Aceton und eine Fraktion 100'"/o iges Aceton.
Die Fraktionen 6 bis einschliesslich 11 werden vereinigt und eingedampft, wobei man<B>136</B> mg Kri stalle erhält, die nach Umkristallisieren aus Aceton- Skellysolve B 115 mg (72% Gesamtausbeute) 6a- Methyl-llss,17a-dioxy-1,4-pregnadien-3,20-dion vom Smp. 222 bis<B>2310</B> ergeben.
Analyse: berechnet für C22H3004. C: 73,71 H: 8,44 gefunden: C: 73,30 H: 8,44 Die Oxydation zum 11-Keto-steroid kann folgen dermassen erfolgen: Eine Mischung von 0,3 g 6a-Methyl-11ss,17a- dioxy-1,4-pregnadien-3,20-dion, 100 mg Chrom säureanhydrid, 10 ml Eisessig und 0,5 ml Wasser wird gerührt und dann 8 Stunden bei Zimmertempe ratur gehalten. Dann wird in 50 ml Eiswasser ge gossen, mit verdünnter Natronlauge neutralisiert, der Niederschlag auf einem Filter gesammelt und dreimal aus Äthylacetat und Skellysolve B-Hexanen um kristallisiert.
Man erhält 6a-Methyl-17a-oxy-1,4- pregnadien-3,11,20-trion.
<I>Beispiel 2</I> Zu einer Lösung von 6a-Äthyl-1 lss,17a-dioxy-21- jod-1,4-pregnadien-3,20-dion in 50o/oiger wässriger Essigsäure gibt man überschüssiges Natriumbisulfit und rührt eine Stunde bei Zimmertemperatur. Dann wird in überschüssiges Wasser gegossen und mit Me- thylenchlorid extrahiert.
Die Methylenchloridextrakte werden vereinigt, mit Wasser, dann mit Natrium- bicarbonatlösung und wiederum mit Wasser ge waschen, über Natriumsulfat getrocknet und einge dampft. Das Rohprodukt wird aus Aceton Skelly- solve B umkristallisiert, und man erhält reines 6a Äthyl-11ss,17a-dioxy-1,4-pregnadien-3,20-dion.
Die Oxydation zum 11-Keto-steroid kann folgen dermassen erfolgen: Zu 200 mg 6a-Äthyl-llss,17a-dioxy-1,4-pregna- dien-3,20-dion in 8 ml Methanol gibt man 0,2 ml Pyridin, 0,4 ml Wasser und 150 mg N-Bromacet- amid. Die Mischung wird 20 Stunden bei Zimmer temperatur gehalten und das überschüssige N-Brom- acetamid durch Zugabe von 25 ml verdünnter Na- triumsulfitlösung zerstört, dann eingedampft,
bis kräftige Kristallisation eintritt. Man kühlt auf 0 und hält eine Stunde bei dieser Temperatur.
<I>Beispiel 3</I> Aus dem nach Präparation 5 erhältlichen 6a-Me- thyl-17a-oxy-21-jod-1,4-pregnadien-3,11,20-trion wird gemäss dem Verfahren des Beispiels 1 durch Reduk tion mit Natriumthiosulfat das 6a-Methyl-17a-oxy- 1,4-pregnadien-3,11,20-trion hergestellt.
Durch Behandlung anderer 6a-Alkyl-11ss,17a- dioxy - 21 - jod -1,4 - pregnadien-3,20-dione und 6a Alkyl-17a-oxy-21-jod-1,4-pregnadien-3,11,20-trione mit einem reduzierenden Mittel, wie Zink in Essig säure, Natriumbisulfit, Natrium- oder Kaliumthio. Sulfat gemäss den Beispielen 1 bis 3 erhält man die entsprechenden 6a-Alkyl-llss,17a-dioxy-1,4-pregna- dien-3,20-dione und 6a-Alkyl-17a-oxy-1,4-pregna- dien-3,11,20-trione,
in denen die Alkylgruppe Äthyl, Propyl, Isopropyl, Butyl, Isobutyl, Pentyl, Hexyl oder Aryl, wie Phenyl und dergleichen, ist.
Anstelle der 6a-Epimeren kann man in den vorangehenden Beispielen auch die 6ss-Epimeren verwenden; wenn die Reaktionsbedingungen beim Neutralpunkt gehalten werden, kann man die 6ss Epimeren, wie 6ss-Methyl-11ss,17a-dioxy-1,4-pregna- dien-3,20-dion und 6ss-Methyl-17a-oxy-1,4-pregna- dien-3,11,20-trion, aus der Reaktionsmischung iso lieren. Durch Behandlung der 6,B-Epimeren mit einer Säure oder Base in organischen Lösungsmitteln, z. B.
Äthanol bei Zimmertemperatur, erhält man aus ihnen die 6a-Epimeren.

The 6 - methyl-11 ss, 17a - dioxy-1,4-pregnadiene-3,20-diones and their 11-keto analogs are also valuable starting materials for the production of other physiologically active compounds. So show z.

<I> Example 1 </I> The crude product obtained according to preparation 2 is slurried in 5 ml of acetic acid and stirred for 45 minutes. An aqueous solution of sodium thiosulfate pentahydrate (250 mg) is then added, whereupon the iodine color disappears. Fractions 6 up to and including 11 are combined and evaporated, giving 136 mg of crystals which, after recrystallization from acetone Skellysolve B, 115 mg (72% total yield) 6a-methyl-llss, 17a-dioxy -1,4-pregnadiene-3,20-dione from m.p. 222 to <B> 2310 </B>.

<I> Example 2 </I> To a solution of 6a-ethyl-1 lss, 17a-dioxy-21-iodo-1,4-pregnadiene-3,20-dione in 50% aqueous acetic acid are added excess sodium bisulfite and stirs for one hour at room temperature. It is then poured into excess water and extracted with methylene chloride.
The methylene chloride extracts are combined, washed with water, then with sodium bicarbonate solution and again with water, dried over sodium sulfate and evaporated. The crude product is recrystallized from acetone Skelly-solve B, and pure 6a ethyl-11ss, 17a-dioxy-1,4-pregnadiene-3,20-dione is obtained.
