Die punktierte Linie in den Formeln XIII bis XXVI bedeutet eine JI-Doppelbindung, die vorhanden sein kann oder nicht, das heisst, die Formeln stellen sowohl die -14- wie auch die A1,4-Verbindungen dar.
Die erfindungsgemäss erhältlichen Verbindungen sind hochwirksame entzündungshemmende Mittel mit verbessertem Verhältnis der therapeutischen Wirkung gegenüber den unerwünschten Nebenwirkungen, z. B. Magen-Darmstörungen, Salzrückhalt im Gewebe, 6dembildung usw., welche bei ähnlichen bekannten physiologisch wirksamen Steroiden auftreten können. Die oben erwähnten Verbindungen sind wirksam bei der Behandlung verschiedener Entzündungszustände der Haut, der Augen, der Atmungsorgane, Knochen und innerer Organe tierischer Organismen, welche auf Bakterien- oder Virusinfektionen, Kontaktderma- titis und allergische Reaktionen zurückzuführen sind,
und besitzen eine bessere therapeutische Aktivität der Entzündungsbekämpfung gegenüber unerwünsch ten Nebenwirkungen als die entsprechenden Verbin dungen ohne den Methylrest in 16-Stellung. Zu diesem Zweck können sie den verschiedenen bekann ten inerten Salben, Cremen, Lösungen, Lotionen, Suspensionen und Sprühmitteln beigemischt werden. Sie können mit jedem der bekannten Antibiotika, insbesondere Penicillin, Neomycin, Tetracyclin, Chlormycetin und Novobiocin gemischt werden.
Die Ausgangsmaterialien des vorliegenden Ver fahrens werden aus dem bekannten 11-Keto-16- dehydroprogesteron (1) durch folgende Reaktionen hergestellt: Die 3-Ketogruppe wird durch L7ber- führung in ein 3-Enamin, z. B. Pyrrolidylenamin, selektiv geschützt.
Das 11-Keto-16-dehydroproge- steron, in der 3-Stell'ung durch das 3 Enamin ge schützt (II), wird dann mit einem Methylgrignard in Gegenwart eines die 1,4-Addition fördernden Mittels, z. B. Kupfer-(I)-chlorid, versetzt. Der 3-Enarninrest wird dann hydrolysiert, wobei 11-Keto-16a-methyl- progesteron (IV) hergestellt wird.
Das 11-Keto-16a-methylprogesteron wird in den 3,11 - Diketo -16a -methyl-4,17(20) -pregnadien-21- säure-niedrigalkylester nach der im USA-Patent Nr.<B>2790814</B> beschriebenen Methode umgesetzt, das heisst das 11-Keto-16a-methylprogesteron wird mit mehr als zwei Moläquivalenten eines Oxalsäure- dialkylesters und einer Base versetzt, um das ent sprechende Alkalimetalldienolat von 2,21-Dialkoxy- oxalyl-11-keto-16a-methylprogesteron (V,
M = Al- kalimetal1) herzustellen. Diese Verbindung, oder das freie, durch Versetzen des Alkalimetalldienolats mit Essigsäure (V, M - H) hergestellte Enol wird dann mit Chlor oder Brom, vorzugsweise letzterem, tri- halogeniert, um 2,21-Dialkoxyoxalyl-2,21,21-tri- halogen-11-keto-16a-methylprogesteron (VI) zu er geben.
Diese Verbindung wird mit einer starken Base umgelagert, um den 2 -Halogen - 3,11- diketo- 16a-methyl - 4,17(20) - pregnadien-21-säurealkylester (VII) zu ergeben. Der 2-Halogenrest wird mit Zink und Essigsäure oder einem andern Halogen entfer nenden Mittel entfernt, um den 3,11-Diketo-16a- methyl-4,17(20)-pregnadien-21-säurealkylester (VIII) zu ergeben. Diese Verbindung ist ein wertvolles Zwi schenprodukt in der Herstellung einer grossen Anzahl 16-Methylsteroide.
Die Umsetzung dieser Verbindung VIII zum 11f,21-Dioxy-16a-methyl-4,17(20)-pregnadien-3-on (XI) wird nach der in den USA-Patenten Nummern 2 715 621, 2 781 343 und 2 790 814 beschriebenen Methode durchgeführt, das heisst, indem die 3-Keto- gruppe durch ein Ketal, oder vorzugsweise durch ein Enamin, z.
B. 3-Pyrrolidylenamin, geschützt wird, wie dies durch Formel IX dargestellt wird, mit nach folgender Reduktion des 11-Ketorestes und des 21-Carbonsäureesterrestes mit Lithiumaluminium- hydrid, Natriumaluminiumhydrid oder einem andern zu einer solchen Reduktion befähigten Reagenz, das die Doppelbindung nicht angreift, um 11ss,21-Dioxy- 16a-methyl-4,17(20)-pregnadien-3-on zu erhalten.
Der die 3-Ketogruppe schützende Rest wird dann durch Hydrolyse entfernt, um Ilf,21-Dioxy-16a- methyl-4,17(20)-pregnadien-3-on (X1) zu ergeben. Diese Verbindung kann dann mit Selendioxyd oder einer die Dehydrierung in 1,2-Stellung bewirkenden Pilzart in die entsprechende A',--Verbindung umge setzt werden, um 1 lss,21- Dioxy - 16a - methyl- 1,4,17(20)-pregnadien-3-on (XII) zu ergeben.
Diese zwei Verbindungen, das heisst XI und XII, werden nach folgender Methode in Verbindungen umgesetzt, welche die Seitenkette des Nebennierenrinden- hormons besitzen: Der 21-Oxyrest wird vorzugsweise verestert, um ihn während der oxydativen Hydroxylierungsreaktion zu schützen, was Verbindung XIII ergibt.
Der Ester wird dann mit Osmiumtetroxyd und einem Oxydationsmittel oxydativ hydrolysiert nach dem in den USA-Patenten _Nrn. 2 769 825 und 2 759 823 oder bei Hogg et a1., J.
Am. Chem. Soc., 77, 4436 (1955) beschriebenen Verfahren, um die entspre chende 17a - Oxy - 20 - keto -21-acyloxy-Verbindung (XIV) zu ergeben, welche mit einer Base, vorzugs weise unter Durchblasen von Stickstoff, hydrolysiert wird, um die 21-Oxy-Verbindung zu ergeben, das heisst Ilfl,17a,21-Trioxy-16a-methyl-4-pregnen-3,20- dion und llss,17a,21-Trioxy-16a-methyl-1,4-pregna- dien-3,20-dion.
Die durch Formel XIV dargestellten Verbindun gen werden zu den entsprechenden 9a-Halogen- Verbindungen durch folgende Reaktionsfolge um gesetzt: Der llss-Oxyrest wird mit einem der be kannten Dehydratationsmittel zur Dehydratation eines 11-Oxyrestes nach bekannten Methoden dehy dratisiert, um die entsprechende 9(11)-Dehydrover- bindung (XV) zu ergeben.
Diese Verbindung wird dann mit unterchloriger, unterjodiger oder unter- bromiger Säure versetzt, um die entsprechende 9a-Halogen-ll,B-oxy-Verbindung (XVI) zu ergeben.
Behandeln einer .dieser letzteren Verbindungen mit einer Base ergibt die entsprechende Epoxy-Verbin- dung (XVII). Diese Epoxy-Verbindung wird dann mit wässrigem oder wasserfreiem Fluorwasserstoff versetzt, nach bekannten Verfahren, um eine 9a-Fluor-llss-oxy-Verbindung (XVIII) zu ergeben, welche dann, gleich wie oben für die entsprechenden 9-Wasserstoff-Verbindungen beschrieben, hydroly- siert wird zu den 21-Oxy-Verbindungen,
das heisst 9a - Fluor -11 ss,17a,21-trioxy-16a-methyl-4-pregnen- 3,20-dion und 9a-Fluor-llss,17a,21-trioxy-16a-me- thyl'-1,4-pregnadien-3,20-dion.
Aus 11l,17a,21-Trioxy-16a-methyl-1,4-pregna- dien - 3,20 - dion, 11/l,17a,21-Trioxy-16a-methyl-4- pregnen-3,20-dion oder den entsprechenden 9a-Fluor- oder 9a-Chlor-Verbindungen (XX) werden mit einem organischen Sulfonylhalogenid die entspre chenden 21-Sulfonsäureester (XXI) erhalten.
Die so hergestellten 21-Alkyl- oder Arylsulfonate ergeben mit Natriumjodid in Acetonlösung die entsprechen den 21-Jod-Verbindungen (XXII). Aus den 21-Jod- Verbindungen erhält man gemäss vorliegendem Ver fahren durch Einwirkung von Silberfluorid, vorzugs weise in Acetonitrillösung, die entsprechende 21- Fluor-Verbindung (XXIV). Diese kann, wenn ge wünscht, mit Chromsäureanhydrid,
N-Bromacetamid, N-Bromsuccinimid oder dergleichen oxydiert werden, um die entsprechende 11-Keto-Verbindung (XXV) zu erhalten. Erfindungsgemäss kann auch der 21 Sulfonsäureester (XXI), direkt mit Kaliumfluorid in Dimethylsulfoxyd, z. B. bei 100 C während 18 Stunden oder länger, versetzt werden, um die 21- Fluor-Verbindung direkt zu ergeben.
Die entspre chenden 11-Keto-Verbindungen werden gleicher weise aus deren 21-Sulfonsäureestern direkt zu den 21-Fluor-Verbindungen umgesetzt.
Die ll,B-Oxy- wie die 11-Keto-Verbindungen sind Diuretika, vor allem orale Diuretika, und be sitzen den Salzgehalt herabsetzende Eigenschaften, wie auch andere wertvolle corticalhormonale und pharmakologische Aktivität. Präparation <I>l:</I> 11-Keto-16a-methylprogesteron. Zu einer heissen Lösung von 4,56 g (0,014 Mol) 11-Keto-16-dehydroprogesteron in 45 ml Methanol wird 2,25 ml Pyrrolidin unter Quirlen zugesetzt.
Das Reaktionsprodukt, 3-Pyrrolidylenamin von 11-Keto- 16-dehydroprogesteron, scheidet bald als gelbliche Kristalle aus. Nach Kühlen auf 5 C wird das Ge misch filtriert, der Kuchen mit kaltem Methanol ge waschen und im Vakuum getrocknet. Man erhält 4,56 g 3-Pyrrolidyl-3,5,16-pregnatrien-11,21-dion, bei 154-169 C schmelzend.
Einer Lösung des so erhaltenen Enamins in 110 ml Tetrahydrofuran wird langsam eine ge rührte Suspension von 45 ml Handelsgrad 3 M Me- thylmagnesiumbromid in Äther, 0,90g Kupfer-(I)- chlorid und 50 ml Tetrahydrofuran zugesetzt. Das Gemisch wird auf Zimmertemperatur abgekühlt und während 3 Stunden unter Stickstoffatmosphäre ge rührt. Das überschüssige Grignard wird vorsichtig zerstört, indem 29 ml Wasser zugetropft werden.
Zu dem 3 - Pyrrolidyl - 16a - methyl-3,5-pregnadien- 11,20-dion enthaltenden Gemisch werden 18 ml Essigsäure und 72 ml Methanol zugesetzt und das Gemisch erwärmt, bis eine klare dunkelgelbe Lö- sung erhalten wird. 72 ml einer 10 % igen wässrigen Natronlaugelösung wird zugegeben, was das pH auf 8 bringt.
Das Gemisch wird am Rückfluss während .15 Minuten erhitzt. Dann werden 4,5 ml Essigsäure und 250 ml Wasser zugesetzt und das abgekühlte Gemisch dreimal mit Methylenchlorid extrahiert. Die vereinigten Extrakte werden mit wässriger Natrium- bicarbonatlösung und mit Wasser gewaschen und dann über Natriumsulfat getrocknet.
Die getrocknete Lösung wird verdampft und der im wesentlichen aus 11 - Keto -16a - methylprogesteron bestehende Rückstand in 400 ml Methylenchlorid gelöst und durch eine Säule von 200 g Magnesiumsilikat (Flo- risil) chromatographiert. Die Säule wird mit 400-ml- Portionen Lösungsmittel folgender Zusammensetzung und Folge entwickelt:
5 von Hexanen (Skellysolve B) plus 3 % Aceton; 5 von Hexanen plus 5% Aceton; 8 von Hexanen plus 7<B>',/0</B> Aceton;
5 von Hexanen plus 10% Aceton; 4 von Hexanen plus 15% Aceton und schliesslich 1 von Aceton. Die Fraktionen 9-25 (einschliesslich der Methylenchloridfraktion) werden vereinigt und aus einem Gemisch von Aceton und Wasser umkristallisiert, um 2,03 g 11-Keto-16a- methylprogesteron, bei 179-182 C schmelzend, zu ergeben.
Eine Probe, die aus einem Gemisch von Athylacetat und Hexanen umkristallisiert wurde, schmolz bei 183-185 C mit [a]D +255o (CHCl3), <B>),Alk</B> 238 mp, am 15,850 und folgende Analyse: Berechnet für C."H3003: C 77,15 H 8,83 Gefunden:
C 76,95 H 8,98 Präparation <I>2:</I> 21-Säuremethylester von 3,11-Diketo-16a- methyl-4,17(20)-pregnadien. Eine Lösung von 2,00 g (5,8 mM) 11-Keto- 16a-methylprogesteron in 30 ml trockenem tert. Butanol wird auf 50 C erwärmt und unter Stick stoff gerührt.
Zur Lösung werden 3,2 ml Äthyl- oxalat und 3,03 g 25 % ige Natriummethoxydlösung in Methanol zugesetzt. Ein gelbgrüner Niederschlag des Natriumdienolats von 2,21-Diäthoxyoxalyl-11- keto-16a-methylprogesteron zeigt sich fast augen blicklich.
Das Gemisch wird während 20 Minuten ge rührt, worauf eine gekühlte Lösung von 0,98 g wasserfreiem Natriumacetat und 0,84 ml Essigsäure in 40 ml Methanol zugesetzt werd-.n, was das freie Dienol ergibt. Die Lösung wird auf 0' C abgekühlt und dann tropfenweise mit einer kalten Lösung 2,0 g Brom in Methanol während 10 Minuten versetzt. Dies ergibt 2,21,21-Tribrom-2,21-diäthoxyoxalyl-11- keto-16a-methylprogesteron.
Das Kühlungsbad wird entfernt und zur Lösung 5,72 g einer 25 % igen Natriummethoxydlösung in Methanol zugesetzt. Das Rühren wird während 2,5 Stunden fortgesetzt, und man erhält den 21-Säure- methylester von 2-Brom-3,11-diketo-16a-methyl- 4,17(20)-pregnadien.
Zur erhaltenen Lösung werden dann 5 ml Essig säure und 1 g Zinkstaub zugesetzt und das Rühren während weiterer 30 Minuten weitergeführt. Das Gemisch wird filtriert, das Filtrat mit Wasser ver dünnt und dann gründlich mit Methylenchlorid extrahiert. Der Extrakt wird über Natriumsulfat ge trocknet und eingedampft. Der Rückstand wird in 400 ml Methylenchlorid gelöst und über eine 200-g- Säule Magnesiumsilikat (Florisil) gegossen.
Die Säule wird mit-400-ml-Portionen Lösungsmittel folgender Zusammensetzung und Folge entwickelt: 4 von Hexanen (Skellysolve B) plus 5 % Aceton; 10 von Hexanen plus 7% Aceton;
10 von Hexanen plus 10 % Aceton und schliesslich 1 von Aceton.
Die Fraktionen 14-23 (einschliesslich die Methylenchloridfraktion) enthalten das Ausgangs-ll-Keto-16a-methylprogesteron. Frak tionen 7-12 enthalten den 21-Säuremethylester von 3,11-Diketo-16a-methyl-4,17(20)-pregnadien, wel cher, nach Umkristallisieren aus Methanol und Wasser und dann aus Methanol, bei 177-184 C schmilzt und<B>2 Alk</B> 232,5 m,u, am 26,200 a]D + 137 (CHC13) und folgende Analyse hat: Berechnet für Cz3H3004: C 74,56 H 8,16 Gefunden:
C 74,58 H 8,04 Die Ausbeute des Produkts wird erhöht, wenn die Reaktion des 11-Keto-16a-methylprogesterons mit Äthyloxalat und Natriummethoxyd bei etwas höheren Temperaturen undjoder verlängerter Reak tionszeit durchgeführt wird.
Präparation <I>3:</I> 1 lss,21-Dioxy-16a-methyl-4,17(20)-pregnadien- 3-on. Eine Lösung von 6,0 g des 21-Säuremethylesters von 3,11- Diketo -16a - methyl-4,17(20)-pregnadien, 6 ml Pyrrohdin und 180 mg p-Toluolsulfonsäure in 120 ml Benzol werden bei Rückflusstemperatur des Gemisches während einer Stunde erhitzt unter gleichzeitigem Entfernen des Reaktionswassers.
Das Lösungsmittel wird aus dem Gemisch abdestilliert und ergibt einen im wesentlichen aus dem 21-Säure- methylester von 3-Pyrrolidyl-11-keto-16a-methyl- 3,5,17(20)-pregnatrien bestehenden Rückstand.
Dieser Rückstand wird in 30 ml Benzol und 10 ml Äther gelöst, eine Suspension von 2,2 g Lithium- aluminiumhydrid in 350 ml Äther während 5 Mi nuten zugesetzt und das Gemisch bei Zimmertempe ratur während 1,5 Stunden gerührt. 20 ml Äthyl- acetat werden zugesetzt, um das überschüssige Li- thiumaluminiumhydrid zu zerstören, gefolgt von 30 ml Wasser.
Äther und Benzol werden unter ver mindertem Druck vom Gemisch abdestilliert, was einen im wesentlichen aus 3-Pyrrolidyl-llss,21- dioxy-16a-methyl-3,5,17(20)-pregnatrien und anor ganischem Material bestehenden Rückstand ergibt.
Dieser rohe Destillationsrückstand wird mit 400 ml Methanol bei 40 C gemischt, bis er völlig gelöst ist. 70 ml einer wässrigen 5%-igen Natron- laugelösung wird zugegeben und das Erwärmen bei 40 C während weiterer 10 Minuten fortgesetzt. Die Lösung wird mit Essigsäure neutralisiert und das Lösungsmittel unter vermindertem Druck abdestil- liert bei einer Temperatur unter 45 C.
Der Rück stand wird mit 100 ml Wasser, das 10 ml konzen trierte Salzsäure enthält, gemischt, filtriert und mit Wasser gewaschen, um llss,21-Dioxy-16a-methyl- 4,17(20)-pregnadien-3-on zu ergeben. Präparation <I>4:</I> l Iss,21-Dioxy-16a-methyl=1,4,17(20)- pregnatrien-3-on. Septomyxa affinis (A. T. C.
C. 6737) wird auf schrägem Malzagar, bestehend aus 50 g trockenem Malzextrakt, 5 g Edamine enzymatisch verdautem Lactalbumin und 20g Agar, bis auf 1 Liter mit Leitungswasser verdünnt, eingestellt auf ein pH von 6,5 bis 7,0, gezüchtet. Impfstoff vom Schrägagar wird in 1 Liter Erlenmeyerkolben welche 100 ml Malzextraktagar enthalten, verbracht und die Inku bation bei Zimmertemperatur während 4 bis 7 Tagen durchgeführt, um Sporen zu erzeugen.
Die Sporen werden in 100 ml steriler 1%iger Kochsalzlösung supendiert. 5 ml dieser Kochsalzlösungsporenimpfung wird dann in je fünf 250-ml-Kolben mit je 100 ml eines Mediums aus 1'% Cerelosedextrose und 20/0 Corn
steep liquor mit 600/0 Festsubstanz, welches mit Natronlauge auf ein pH von 4,8 bis 5,0 einge stellt wird, zugesetzt. Die Inkubation wird unter Schütteln bei Zimmertemperatur während 48 bis 72 Stunden durchgeführt und dann zu 6 Liter des oben beschriebenen Gärungsmediums zugesetzt. Diese Kultur wird während 24 Stunden bei Zimmertempe ratur und Durchlüftung von 1 1,\m gehalten. Diese 6 Liter Impfkultur werden dann zu 100 Liter Me dium zugesetzt und das Wachstum während 24 Stun den bei Zimmertemperatur und Schütteln und Durchlüftung von 2 1,1m weitergeführt.
Zu dieser 24 Stunden alten Kultur werden 25 g Ilss,21-Dioxy- 16a-methyl-4,17(20)-pregnadien-3-on, in 500 ml Aceton gelöst, zugesetzt. Schütteln und Durchlüftung bei 1 Ilm wird während weiterer 24 Stunden fort gesetzt.
Der Brei wird filtriert, um Mycelien und gefil tertes Bier zu erhalten. Die Mycelien werden mit 20 Liter Wasser gewaschen und das Waschwasser zum filtrierten Bier zugesetzt. Die gewaschenen Mycelien werden suspendiert und zweimal aufge schlämmt je mit 12 Liter Aceton und dann zweimal aufgeschlämmt je mit 12 Liter Methylenchlorid. Die so erhaltenen Aceton- und Methylenchloridmycelien- extrakte werden vereinigt und zu einem Extrakt zu gesetzt, der aus vier Extraktionen des gefilterten Bieres und Waschwassers erhalten wurde,
wobei jede der Extraktionen mit 24 Liter Methylenchlorid durchgeführt wird. Die Extrakte von gefiltertem Bier und Waschwasser und diejenigen der Mycelien werden vereinigt und zweimal gewaschen, je mit 12 Liter 2%iger Natriumbicarbonatlösung und dann zweimal je mit 12 Liter Wasser. Der gewaschene Extrakt wird in einem Destillationsapparat einge engt und ergibt 3 Liter Konzentrat, welches dann zur Trockne auf einem Dampfbad verdampft wird, was einen rohen, kristallinen Rückstand ergibt.
Dieser Rückstand wird sechsmal je mit 25 ml Di- äthyläther zu feinem Pulver zermahlen. Die zurück bleibenden Kristalle werden in 250 ml heissen Me thanols gelöst, filtriert und abgekühlt und ergeben Kristalle von llss,21-Dioxy-16a-methyl-1,4,17(20)- pregnatrien-3-on. Präparation <I>S:
</I> 21 -Essigsäureester von l 1ss,21 -Dioxy-16a-me- thyl-4,17(20)-pregnadien-3-on. 500 mg I 1ss,21- Dioxy -16a - methyl-4,17(20)- pregnadien-3-on, nach dem Verfahren von Präpa- ration 3 hergestellt, werden mit 5 ml Essigsäure anhydrid in 10 ml Pyridin bei 26 C während 4 Stunden versetzt.
Das erhaltene Gemisch wird mit 10 ml Wasser unter Kühlung gemischt, um das über schüssige Essiganhydrid zu hydrolysieren, und die Lösung mit Methylenchlorid extrahiert. Der Extrakt wird mit verdünnter Salzsäure, Wasser, gesättigter wässriger Natriumbicarbonatlösung und schliesslich mit Wasser gewaschen.
Das Methylenchlorid wird aus der Lösung abdestilliert und der Rückstand aus einem Gemisch von Aceton und Hexanen (Skelly- solve B) umkristallisiert, um den im wesentlichen reinen 21-Essigsäureester von llss,21-Dioxy-16a- methyl-4,17(20)-pregnadien-3-on, Smp. 171-173 C, zu ergeben.
Präparation <I>6:</I> 21-Essigsäureester von 11/;,21-Dioxy-16a- methyl-1,4,17(20)-pregnatrien-3-on. 1 g 11ss,21- Dioxy -16a - methyl -1,4,17(20)- pregnatrien-3-on wird in 20 ml Pyridin und 15 ml Essigsäureanhydrid gelöst und das Gemisch während 4 Stunden auf 40 C erhitzt. Die Lösung wird abge kühlt und dann langsam mit Wasser verdünnt.
Das ausgefällte Steroid wird mittels Filtrieren entfernt, mit Wasser gewaschen und getrocknet, um den 21- Essigsäureester von l 1ss,21 - Dioxy - 16a - methyl- 1,4,17(20)-pregnatrien-3-on zu ergeben.
Präparation <I>7:</I> 21-Essigsäureester von llp,17a,21-Trioxy-16a- methyl-1,4-pregnadien-3,20-dion. Zu einer Lösung von 100 mg 21-Essigsäureester von 11ss,21-Dioxy-16a-methyl-1,4,17(20)-pregna- trien-3-on in 5 ml trockenem tert. Butanol werden nacheinander 0,065 ml Pyridin, 0,43 ml einer Lö sung von N-Methylmorpholinoxydperoxyd in tert. Butanol mit einem Titer von 44 ml 0,
1 N-Natrium- thiosuifat pro ml hergestellt durch -die Reaktion von N-Methylmorpholin mit zwei rnolaren Äquivalenten Wasserstoffperoxyd in tert. Butanol und 0,2 mg Osmiumtetroxyd in 0,043 ml tert. Butanol. Nach 24 Stunden bei Zimmertemperatur wird die Lösung mit Wasser verdünnt und mit Methylenchlorid extra hiert.
Die Extrakte werden mit verdünnter Salz säure, verdünntem Natriumbicarbonat und schliess lich mit Wasser gewaschen und dann getrocknet. Das Lösungsmittel wird abdestilliert und der Rück stand über eine Säule von 10 g Magnesiumsilikat (Florisil) chromatographiert. Die Säule wird mit Hexanen (SkellysolVe B), welche steigende Portionen Aceton enthalten, entwickelt.
So wird der 21-Essig- säureester von 11ss,17a,21-Trioxy-16a-methyl-1,4- pregnadien-3,20-dion eluiert, der durch Verdampfen vom Lösungsmittel befreit wird.
Präparation <I>8:</I> 21-Essigsäureester von 16a-Methyl-17a,21-dioxy- 1,4,9(11)-pregnatrien-3,20-dion. Zu einer Lösung von 8,5 g 21-Essigsäureester von 11f,17a,21-Trioxy-16a-methyl-1,4-pregnadien- 3,20-dion in 42,5 ml Pyridin werden 5,63g N Brom- acetamid zugesetzt.
Nach Stehen bei Zimmertempe ratur während 15 Minuten wird die Reaktionslösung auf 5-10 C abgekühlt, und unter Schütteln wird Schwefeldioxydgas über die Oberfläche geführt, bis die Lösung keine Farbe auf einem angesäuerten Stärke-Jodpapierstreifen anzeigt. Während der Zu gabe des Schwefeldioxyds wird das Reaktionsgemisch warm. Durch äusseres Abkühlen wird die Temperatur unter 30 C gehalten, wie auch durch Variieren der Schwefeldioxydzufuhr. Dem Reaktionsgemisch wer den dann 400 ml Eiswasser zugesetzt und der ent stehende Niederschlag durch Filtrieren gesammelt.
Dieses Material wird aus Aceton und Hexanen (Skellysolve B) umkristallisiert, um 21-Essigsäure- ester von 16a-Methyl-17a,21-dioxy-1,4,9(11)-pregna- trien-3,20-dion zu ergeben.
Präpararion <I>9:</I> 21-Essigsäureester von 9a-Brom-1 1ss,17a,21- trioxy-16a-methyl-1,4-pregnadien-3,20-dion. Zu einer Lösung von 5,68 g 21-Essigsäureester von 16a-Methyl-17a,21-dioxy-1,4,9(11)-pregnatrien- 3,20-dion in 100 ml Methylenchlorid und 250 ml tert. Butanol werden eine Lösung von 14 ml 72a/o,!iger Perchlorsäure in 100 ml Wasser, gefolgt von einer Lösung 2,
34g N-Bromacetamid in 60 ml tert. Butanol, zugesetzt. Nach Rühren des Reaktions gemisches während 15 Minuten wird eine Lösung von 2,8g Natriumsulfit in 140 ml Wasser zugegeben und das Reaktionsgemisch unter vermindertem Druck, bei etwa 50 C auf ein Volumen von etwa 500 ml eingeengt. Hier setzt Kristallisation ein. Das gerührte Konzentrat wird im Eisbad gekühlt und 500 ml Wasser zugesetzt.
Nach Rühren während einer Stunde wird das kristalline Produkt durch Fil trieren isoliert, die Kristalle mit Wasser gewaschen und luftgetrocknet, um 21-Essigsäureester von 9a- Brom-11 ss,17ai,21-trioxy-16a-methyl-1,4-pregnadien- 3,20-dion zu ergeben.
Präparation <I>10:</I> 21-Essigsäureester von 9ss,llss-Epoxy-16a-me- thyl-17a,21-dioxy-1,4-pregnadien-3,20-dion. Zu einer Lösung von 6.78 g 21-Essigsäureester von 9a -Brom - l lss,17a,21- trioxy-16a-methyl-1,4- pregnadien-3,20-dion in 175 ml Aceton werden 6,78 g Kaliumacetat zugesetzt und die erhaltene Suspension unter Rückfluss während 17 Stunden ge kocht.
Das Gemisch wird dann auf etwa 60 ml Vo lumen unter vermindertem Druck auf dem Dampf bad eingeengt und dann mit Wasser verdünnt und mit Methylenchlorid extrahiert. Die Methylenchlorid- extrakte werden vereinigt, mit Wasser gewaschen, über wasserfreiem Natriumsulfat getrocknet und ein gedampft.
Der Rückstand wird in Methylenchlorid wieder gelöst und über 200 g Magnesiumsilikat (Florisil) chromatographiert. Die Säule wird mit 1- Liter-Portionen Hexanen (Skellysolve B), welche steigernde Portionen Aceton enthalten, eluiert. So wird 21-Essigsäureester von 9ss,11ss-Epoxy-16a-me- thyl-17,a.-,21-dioxy-1,4-pregnadien-3,20-dion eluiert,
welcher durch Verdampfen vom Lösungsmittel be freit wird. Präparation <I>11:</I> 21-Essigsäureester von 9a-Fluor-llss,17a,21- trioxy-16a-methyl-1,4-pregnadien-3,20-dion. Zu ungefähr 1,3g Fluorwasserstoff in einer Poly- äthylenflasche, welcher bei -60 C gehalten wird, werden 2,3 ml Tetrahydrofuran und dann eine Lö sung von 500 mg (0,0012 Mol) 21-Essigsäüreester von 9ss,11ss - Epoxy - 17a,
21- dioxy-16a-methyl-1,4- pregnadien-3,20-dion in 2 ml Methylenchlorid zuge setzt. Die Steroidlösung wird mit einem zusätzlichen Milliliter Methylenchlorid gespült. Die hellrot ge färbte Lösung wird während 1 Stunde bei etwa -30 C und während 2 Stunden bei -10 C gehalten. Dann wird sie mit einem überschuss kalter Natrium- bicarbonatlösung gemischt und das organische Ma terial mit Hilfe von zusätzlichem Methylenchlorid extrahiert.
Die vereinigten Extrakte werden mit Wasser gewaschen, über wasserfreiem Natriumsulfat getrocknet und auf etwa 35 ml Volumen eingeengt. Diese Lösung wird über 40 ml Magnesiumsilikat (Florisil) chromatographiert. Die Säule wird mit 500-ml-Portionen Hexanen (Skellysolve B), welche ansteigende Portionen Aceton enthalten, entwickelt.
So wird 21-Essigsäureester von 9a=Fluor-llss,17a,21- trioxy-16a-methyl-1,4-pregnadien-3,20-dion eluiert, welcher durch Verdampfen der Eluatfraktionen vom Lösungsmittel befreit wird.
Präparation <I>12:</I> 9a-Fluor-1 lss,17a,21-trioxy-16a-methyl-1,4- pregnadien-3,20-dion. 3,25 g 21 - Essigsäureester von 9a - Fluor- 1 lss,17a,21-trioxy-l6a-methyl-1,4-pregnadien-3,20- dion werd--n in 325 ml Methanol gelöst, welches vorher von Luft-Sauerstoff befreit wird, indem wäh rend 10 Minuten Stickstoff durchgeblasen werden. Zu dieser Lösung wird eine Lösung von 1,63 g Kaliumbicarbonat in 30 ml Wasser, welches ebenso von Sauerstoff befreit ist, zugesetzt.
Das Gemisch wird bei Zimmertemperatur während 5 Stunden unter Stickstoffatmosphäre stehengelassen und dann mit verdünnter Essigsäure neutralisiert. Das Gemisch wird unter vermindertem Druck auf einem 60' C Dampfbad auf etwa ein Drittel des Volumens einge engt, 250 ml Wasser zugefügt und das Gemisch eis gekühlt. Das kristalline Produkt wird auf einem Filter gesammelt, mit Wasser gewaschen und ge trocknet, um 9a-Fluor-llss,17a,21-trioxy-16a-methyl- 1,4-pregnadien-3,20-dion zu ergeben.
Präparation <I>13:</I> 21-Methansulfonsäureester von 11/3,17a,21- trioxy-16a-methyl-4-pregnen-3,20-dion. Es wird eine Lösung hergestellt aus 1 g (2,4 mM) l lss,17a,21-Trioxy-16a-methyl-4-pregnen-3,20-dion in 7 ml Pyridin. Diese Lösung wird auf 0' C ge kühlt und mit 0,3 ml Methansulfonylchlorid versetzt. Dann wird die Lösung bei 0-5 C während 2 Stun den stehengelassen und nachher mit Wasser verdünnt und mit drei 25-ml-Portionen Methylenchlorid extra hiert.
Die Extrakte werden vereinigt, mit kalter ver dünnter Salzsäure gewaschen, bis die wässrige Schicht ein pH von 2-3 aufweist, dann mit kalter Natrium- bicarbonatlösung und mit Wasser gewaschen und schliesslich über wasserfreiem Natriumsulfat getrock net. Verdampfen des Methylenchloridextraktes unter vermindertem Druck hinterlässt einen Rückstand von 21-Methansulfonsäureester von llss,17a,21-Trioxy- 16a-methyl-4-pregnen-3,20-dion.
Unter Verwendung von llss,17a,21-Trioxy-16a- methyl-1,4-pregnadien-3,20-dion als Ausgangssteroid erhält man nach obiger Vorschrift 21-Methansulfon- säureester von llss,17a,21-Trioxy-16a-methyl-1,4- pregnadien-3,20-dion. Präparation <I>14:
</I> 1 ss,17a-Dioxy-16a-methyl-21-jod-4-pregnen- 3,20-dion. Der rohe, in Präparation 13 beschriebene 21- Methansulfonsäureester von llss,17a,21-Trioxy-16a- methyl-4-pregnen-3,20-dion wird in 15 ml Aceton gelöst und mit einer Lösung von 1 g Natriumjodid in 10 ml Aceton versetzt. Das Gemisch wird unter Rückfluss und Rühren während 15 Minuten erhitzt, die Wärme reduziert lind das Gemisch unter vermin dertem Druck auf ein Drittel Volumen eingeengt.
Eis und Wasser werden zugesetzt und das ausgefällte Produkt auf einem Filter gesammelt, mit Wasser ge waschen und getrocknet, um 11ss,17a-Dioxy-16a- methyl-21 jod-4-pregnen-3,20-dion zu ergeben.
Unter Verwendung des 21-Methansulfonsäure- esters von l lss,17a,21 - Trioxy - 16a - methyl - 1,4- pregnadien-3,20-dion als Ausgangssteroid erhält man nach obiger Vorschrift llss,l7a-Dioxy-16a-methyl- 21-jod-1,4-pregnadien-3,20-dion. <I>Beispiel 1</I> 11ss,17a-Dioxy-16a-methyl-21-fluor-4-pregnen- 3,20-dion 1 g llss,17a-Dioxy-16a-methy121 jod-4-pregnen- 3,
20-dion wird in 150 ml kochendem wcetonitril ge löst. Nach Abkühlen auf 40 C wird die Lösung vor Licht geschützt und 0,8 ml einer 50#lloigen wässrigen Lösung Silberfluorid werden unter Rühren zugesetzt. Das Rühren wird während einer Stunde bei etwa 401 C fortgesetzt, dann werden 0,7 ml Süberfluorid- lösung zugegeben. Nach einer weiteren Stunde Rühren werden weitere 0,7 ml wässrige Silberfluorid, lösung zugegeben.
Erwärmen und Rühren wird dann während 2 Stunden weitergeführt. Das braune Ge misch wird über Celite -Kieselgur filtriert und das Filtrat unter vermindertem Druck bei einer Bad temperatur von 50 C verdampft. Der braune Rück stand wird mit zwei 100-ml-Portionen warmen Me- thylenchlorids gründlich extrahiert, die vereinigten Extrakte mit Wasser gewaschen und über wasser freiem Natriumsulfat getrocknet.
Die getrocknete Lösung wird auf etwa 100 ml eingeengt und dann über 50g Magnesiumsilikat ( Florisil ) chromato- graphiert. Die Säule wird mit Hexangemisch, welches steigende Mengen Aceton enthält, eluiert. Man er hält so in Lösung 1lss17a-Dioxy-16a-methyl-21- fluor-4-pregnen-3,20-dion und vorgängig als Neben produkt 11 fl- Oxy - 16a - methyl -17a,
21 - epoxy-4- pregnen-3,20-dion, welche durch Eindampfen vom Lösungsmittel befreit werden. <I>Beispiel 2</I> l lss,17co-Dioxy-16a-methyl-21-fluor-1,4- pregnadien-3,20-dion Nach dem Verfahren von Beispiel 1 unter Ver wendung von llss,17a-Dioxy-16a-methyl-21-jod-1,4- pregnadien 3,20-dion als Ausgangssteroid erhält man 1 lss,17.a-Dioxy-16a-methyl-21-fluor-1,4-pregnadien- 3,

Gleicherweise werden 9a-Chlor-11 ss,17a-dioxy-16a-methyl-21-jod- 4-pregnen-3,20-dion in 9a-Chlor-l lss,17a-dioxy-16a-methyl-21-fluor- 4-pregnen-3,20-dion und 9a-Chlor-11 ss-oxy-16a-methyl-17a,21-epoxy- 4-pregnen-3,20-dion umgesetzt.
<I>Beispiel 5</I> 11 ss,17 a-Dioxy-16a-methyl-21-fluor-4- pregnen-3,20-dion Eine Lösung von 1 g llss,17a,21-Trioxy-16a- methyl-4-pregnen-3,20-dion-21-methansulfonat und 0,50 g wasserfreiem Kaliumfluorid in 10 ml Di- methylsulfoxyd wird während 12 Stunden bei 100 C gekocht. Die abgekühlte Lösung wird mit Wasser verdünnt und dann mit Methylenchlorid extrahiert.
Der Extrakt wird getrocknet und nach der im Bei spiel 1 beschriebenen Weise chromatographiert, um im wesentlichen reines llss,l7a-Dioxy-16a-methyl- 21-fluor-4-pregnen-3,20-dion und llss-Oxy-16a-me- thyl-17a,21-epoxy-4-pregnen-3,20-dion zu ergeben.
Das gemäss Beispiel 1 oder 5 erhältliche Produkt kann wie folgt zum 16a-Methyl-17a-oxy-21-fluor-4- pregnen-3,11,20-trion oxydiert werden: Es wird eine Lösung bereitet aus 0,5 g 16a-Me- thyl -1 lss,17a- dioxy - 21-fluor-4-pregnen-3,20-dion, 0,15 g Chromsäure, 10 ml Eisessig und 1,1, ml Wasser. Dieses Gemisch wird gerührt und während 8 Stunden bei Zimmertemperatur gehalten.
Dann wird das Gemisch in 50 ml Eiswasser gegossen, durch Zugabe von verdünntem Natriumhydroxyd neutralisiert und der so erhaltene Niederschlag auf einem Filter gesammelt und dreimal aus einem Ge misch Äthylacetat und Hexan (Skellysolve B) um- kristallisiert, um 16a Methyl-17a-oxy-21-fluor-1,4- pregnadien-3,11,20-trion zu ergeben.
Auf gleiche Weise können 9a,21-Difluor-1 lss,17a-dioxy-16a-methyl- 1,4-pregnadien-3,20-dion und 9a,21-Difluor-l lss,17a-dioxy-16a-methyl- 4-pregnen-3,20-dion oxydiert werden in 9a,21-Difluor-16a-methyl-17a-oxy- 1,4-pregnadien-3,11,20-trion und 9a,21-Difluor-16a-methyl-17a-oxy- 4-pregnen-3,11,20-trion. Die entsprechenden 9a-Chlorverbindungen können gleicherweise oxydiert werden.

and have a better therapeutic activity in combating inflammation against undesired side effects than the corresponding compounds without the methyl radical in the 16-position. For this purpose, they can be mixed with the various known inert ointments, creams, solutions, lotions, suspensions and sprays. They can be mixed with any of the known antibiotics, especially penicillin, neomycin, tetracycline, chlormycetin and novobiocin.
The starting materials of the present process are prepared from the known 11-keto-16-dehydroprogesterone (1) by the following reactions: The 3-keto group is converted into a 3-enamine, e.g. B. pyrrolidyleneamine, selectively protected.
The 11-keto-16-dehydroprogestone, protected in the 3-Stell'position by the 3 enamine (II), is then treated with a methyl grignard in the presence of an agent promoting 1,4-addition, e.g. B. copper (I) chloride, added. The 3-enarnine residue is then hydrolyzed to produce 11-keto-16a-methyl-progesterone (IV).

M = alkali metal1). This compound, or the free enol produced by adding acetic acid (V, M - H) to the alkali metal dienolate, is then trihalogenated with chlorine or bromine, preferably the latter, to form 2,21-dialkoxyoxalyl-2,21,21-tri - Halogen-11-keto-16a-methylprogesterone (VI) to be given.
This compound is rearranged with a strong base to give the 2 -halo-3,11-diketo-16a-methyl-4,17 (20) - pregnadiene-21-acid alkyl ester (VII). The 2-halogen radical is removed with zinc and acetic acid or another halogen-removing agent to give the 3,11-diketo-16a-methyl-4,17 (20) -pregnadiene-21-acid alkyl ester (VIII). This compound is a valuable intermediate in the manufacture of a large number of 16-methyl steroids.
The conversion of this compound VIII to 11f, 21-dioxy-16a-methyl-4,17 (20) -pregnadien-3-one (XI) is carried out according to the US Pat. No. 2,715,621, 2,781,343 and 2,790 814 described method carried out, that is, by the 3-keto group by a ketal, or preferably by an enamine, z.
B. 3-pyrrolidyleneamine, is protected, as shown by formula IX, with subsequent reduction of the 11-keto radical and the 21-carboxylic acid ester radical with lithium aluminum hydride, sodium aluminum hydride or another reagent capable of such a reduction which does not have the double bond attacks to obtain 11ss, 21-dioxy-16a-methyl-4,17 (20) -pregnadien-3-one.
The residue protecting the 3-keto group is then removed by hydrolysis to give Ilf, 21-dioxy-16a-methyl-4,17 (20) -pregnadien-3-one (X1). Hypochlorous, hypoiodous or hypobromous acid is then added to this compound to give the corresponding 9a-halo-II, B-oxy compound (XVI).

that means 9a-fluorine -11ss, 17a, 21-trioxy-16a-methyl-4-pregnen-3,20-dione and 9a-fluorine-llss, 17a, 21-trioxy-16a-methyl'-1, 4-pregnadiene-3,20-dione.
From 11l, 17a, 21-trioxy-16a-methyl-1,4-pregna- diene - 3,20 - dione, 11 / l, 17a, 21-trioxy-16a-methyl-4-pregnen-3,20-dione or the corresponding 9a-fluorine or 9a-chlorine compounds (XX), the corresponding 21-sulfonic acid ester (XXI) are obtained with an organic sulfonyl halide.
The 21-alkyl or aryl sulfonates prepared in this way give, with sodium iodide in acetone solution, the corresponding 21-iodine compounds (XXII). According to the present process, the 21-iodine compounds are obtained by exposure to silver fluoride, preferably in acetonitrile solution, to give the corresponding 21-fluorine compound (XXIV). This can, if desired, with chromic anhydride,
N-bromoacetamide, N-bromosuccinimide or the like can be oxidized to obtain the corresponding 11-keto compound (XXV). According to the invention, the sulfonic acid ester (XXI) can also be mixed directly with potassium fluoride in dimethyl sulfoxide, e.g. B. at 100 C for 18 hours or longer, added to give the 21-fluoro compound directly.
The corresponding 11-keto compounds are converted in the same way from their 21-sulfonic acid esters directly to the 21-fluoro compounds.
The ll, B-oxy and 11-keto compounds are diuretics, especially oral diuretics, and have salt-reducing properties, as well as other valuable cortical hormone and pharmacological activity. Preparation <I> l: </I> 11-keto-16a-methylprogesterone. 2.25 ml of pyrrolidine is added with stirring to a hot solution of 4.56 g (0.014 mol) of 11-keto-16-dehydroprogesterone in 45 ml of methanol.
The reaction product, 3-pyrrolidyleneamine of 11-keto-16-dehydroprogesterone, soon separates out as yellowish crystals. After cooling to 5 ° C., the mixture is filtered, the cake is washed with cold methanol and dried in vacuo. 4.56 g of 3-pyrrolidyl-3,5,16-pregnatriene-11,21-dione, melting at 154-169 ° C., are obtained.
A stirred suspension of 45 ml of commercial grade 3 M methylmagnesium bromide in ether, 0.90 g of copper (I) chloride and 50 ml of tetrahydrofuran is slowly added to a solution of the enamine thus obtained in 110 ml of tetrahydrofuran. The mixture is cooled to room temperature and stirred for 3 hours under a nitrogen atmosphere. The excess Grignard is carefully destroyed by adding 29 ml of water dropwise.
18 ml of acetic acid and 72 ml of methanol are added to the mixture containing 3-pyrrolidyl-16a-methyl-3,5-pregnadiene-11,20-dione, and the mixture is heated until a clear, dark yellow solution is obtained. 72 ml of a 10% aqueous sodium hydroxide solution is added, which brings the pH to 8.

5 of hexanes (Skellysolve B) plus 3% acetone; 5 of hexanes plus 5% acetone; 8 of hexanes plus 7 ', / 0 </B> acetone;
5 of hexanes plus 10% acetone; 4 of hexanes plus 15% acetone and finally 1 of acetone. Fractions 9-25 (including the methylene chloride fraction) are combined and recrystallized from a mixture of acetone and water to give 2.03 g of 11-keto-16amethylprogesterone, melting at 179-182 ° C.
A sample, which was recrystallized from a mixture of ethyl acetate and hexanes, melted at 183-185 C with [a] D + 255o (CHCl3), <B>), Alk </B> 238 mp, at 15.850 and the following analysis: Calculated for C. "H3003: C 77.15 H 8.83 Found:
C 76.95 H 8.98 Preparation <I> 2: </I> 21-acid methyl ester of 3,11-diketo-16a-methyl-4,17 (20) -pregnadiene. A solution of 2.00 g (5.8 mM) 11-keto-16a-methylprogesterone in 30 ml of dry tert. Butanol is heated to 50 C and stirred under nitrogen.
3.2 ml of ethyl oxalate and 3.03 g of 25% sodium methoxide solution in methanol are added to the solution. A yellow-green precipitate of the sodium dienolate of 2,21-diethoxyoxalyl-11-keto-16a-methylprogesterone can be seen almost immediately.
The mixture is stirred for 20 minutes, after which a cooled solution of 0.98 g of anhydrous sodium acetate and 0.84 ml of acetic acid in 40 ml of methanol is added to give the free dienol. The solution is cooled to 0 ° C. and a cold solution of 2.0 g of bromine in methanol is then added dropwise over 10 minutes. This gives 2,21,21-tribromo-2,21-diethoxyoxalyl-11-keto-16a-methylprogesterone.
The cooling bath is removed and 5.72 g of a 25% strength sodium methoxide solution in methanol are added to the solution. Stirring is continued for 2.5 hours, and the 21-acid methyl ester of 2-bromo-3,11-diketo-16a-methyl-4,17 (20) -pregnadiene is obtained.
5 ml of acetic acid and 1 g of zinc dust are then added to the resulting solution and stirring is continued for a further 30 minutes. The mixture is filtered, the filtrate is diluted with water and then extracted thoroughly with methylene chloride. The extract is dried over sodium sulfate and evaporated. The residue is dissolved in 400 ml of methylene chloride and poured over a 200 g column of magnesium silicate (Florisil).
The column is developed with 400 ml portions of solvent of the following composition and sequence: 4 of hexanes (Skellysolve B) plus 5% acetone; 10 of hexanes plus 7% acetone;
10 of hexanes plus 10% acetone and finally 1 of acetone.
Fractions 14-23 (including the methylene chloride fraction) contain the starting II-keto-16a-methylprogesterone. Fractions 7-12 contain the 21-acid methyl ester of 3,11-diketo-16a-methyl-4,17 (20) -pregnadiene, wel cher, after recrystallization from methanol and water and then from methanol, melts at 177-184 C. and <B> 2 Alk </B> 232.5 m, u, am 26.200 a] D + 137 (CHC13) and has the following analysis: Calculated for Cz3H3004: C 74.56 H 8.16 Found:
C 74.58 H 8.04 The yield of the product is increased if the reaction of the 11-keto-16a-methylprogesterone with ethyl oxalate and sodium methoxide is carried out at somewhat higher temperatures and with a longer reaction time.
Preparation <I> 3: </I> 1 lss, 21-dioxy-16a-methyl-4,17 (20) -pregnadien-3-one. A solution of 6.0 g of the 21-acid methyl ester of 3,11-diketo -16a-methyl-4,17 (20) -pregnadiene, 6 ml of pyrrohdin and 180 mg of p-toluenesulfonic acid in 120 ml of benzene are at the reflux temperature of the mixture during one hour heated while removing the water of reaction.
The solvent is distilled off from the mixture and gives a residue consisting essentially of the 21-acid methyl ester of 3-pyrrolidyl-11-keto-16a-methyl-3,5,17 (20) -pregnatriene.
This residue is dissolved in 30 ml of benzene and 10 ml of ether, a suspension of 2.2 g of lithium aluminum hydride in 350 ml of ether is added over the course of 5 minutes and the mixture is stirred at room temperature for 1.5 hours. 20 ml of ethyl acetate are added to destroy the excess lithium aluminum hydride, followed by 30 ml of water.
Ether and benzene are distilled off from the mixture under reduced pressure, which results in a residue consisting essentially of 3-pyrrolidyl-llss, 21-dioxy-16a-methyl-3,5,17 (20) -pregnatriene and inorganic material.
This crude distillation residue is mixed with 400 ml of methanol at 40 ° C. until it is completely dissolved. 70 ml of an aqueous 5% sodium hydroxide solution is added and heating is continued at 40 ° C. for a further 10 minutes. The solution is neutralized with acetic acid and the solvent is distilled off under reduced pressure at a temperature below 45 C.

C. 6737) is grown on sloping malt agar, consisting of 50 g dry malt extract, 5 g Edamine enzymatically digested lactalbumin and 20 g agar, diluted to 1 liter with tap water, adjusted to a pH of 6.5 to 7.0. The vaccine from the agar slant is placed in 1 liter Erlenmeyer flasks containing 100 ml of malt extract agar and incubated at room temperature for 4 to 7 days in order to generate spores.
The spores are suspended in 100 ml of sterile 1% saline solution. 5 ml of this saline solution pore inoculation is then in each case five 250 ml flasks with 100 ml each of a medium of 1% cerelose dextrose and 20/0 corn

25 g of Ilss, 21-dioxy-16a-methyl-4,17 (20) -pregnadien-3-one, dissolved in 500 ml of acetone, are added to this 24-hour culture. Shaking and aeration at 1 ilm is continued for a further 24 hours.
The slurry is filtered to obtain mycelia and filtered beer. The mycelia are washed with 20 liters of water and the wash water is added to the filtered beer. The washed mycelia are suspended and slurried twice each with 12 liters of acetone and then slurried twice with 12 liters of methylene chloride each time. The acetone and methylene chloride mycelia extracts obtained in this way are combined and added to an extract that was obtained from four extractions of the filtered beer and wash water,
each of the extractions being carried out with 24 liters of methylene chloride. The extracts of filtered beer and wash water and those of the mycelia are combined and washed twice, each time with 12 liters of 2% sodium bicarbonate solution and then twice each with 12 liters of water. The washed extract is concentrated in a still to give 3 liters of concentrate which is then evaporated to dryness on a steam bath to give a crude, crystalline residue.
This residue is ground to a fine powder six times with 25 ml of diethyl ether each time. The remaining crystals are dissolved in 250 ml of hot methanol, filtered and cooled and give crystals of llss, 21-dioxy-16a-methyl-1,4,17 (20)-pregnatrien-3-one. Preparation <I> S:
</I> 21 -acetic acid ester of l 1ss, 21-dioxy-16a-methyl-4,17 (20) -pregnadien-3-one. 500 mg I 1ss, 21-Dioxy -16a - methyl-4,17 (20) - pregnadien-3-one, prepared by the method of Preparation 3, are mixed with 5 ml of acetic anhydride in 10 ml of pyridine at 26 ° C. during 4 hours staggered.
The mixture obtained is mixed with 10 ml of water with cooling in order to hydrolyze the excess acetic anhydride, and the solution is extracted with methylene chloride. The extract is washed with dilute hydrochloric acid, water, saturated aqueous sodium bicarbonate solution and finally with water.
The methylene chloride is distilled off from the solution and the residue is recrystallized from a mixture of acetone and hexanes (Skellysolvent B) in order to obtain the essentially pure 21-acetic acid ester of llss, 21-dioxy-16amethyl-4,17 (20) -pregnadien-3-one, m.p. 171-173 C.
Preparation <I> 6: </I> 21-acetic acid ester of 11 / ;, 21-dioxy-16a-methyl-1,4,17 (20) -pregnatrien-3-one. 1 g of 11ss, 21-dioxy-16a - methyl -1,4,17 (20) - pregnatrien-3-one is dissolved in 20 ml of pyridine and 15 ml of acetic anhydride and the mixture is heated to 40 ° C. for 4 hours. The solution is cooled and then slowly diluted with water.
The precipitated steroid is removed by filtration, washed with water and dried to give the 21-acetic acid ester of 1 1ss, 21-dioxy-16a-methyl-1,4,17 (20) -pregnatrien-3-one.
Preparation <I> 7: </I> 21-Acetic acid ester of IIp, 17a, 21-trioxy-16a-methyl-1,4-pregnadiene-3,20-dione. To a solution of 100 mg of 21-acetic acid ester of 11ss, 21-dioxy-16a-methyl-1,4,17 (20) -pregna- trien-3-one in 5 ml of dry tert. Butanol are successively 0.065 ml of pyridine, 0.43 ml of a solution of N-methylmorpholine oxide peroxide in tert. Butanol with a titer of 44 ml 0,
1 N sodium thiosulfate per ml produced by the reaction of N-methylmorpholine with two molar equivalents of hydrogen peroxide in tert. Butanol and 0.2 mg osmium tetroxide in 0.043 ml tert. Butanol. After 24 hours at room temperature, the solution is diluted with water and extracted with methylene chloride.
The extracts are washed with dilute hydrochloric acid, dilute sodium bicarbonate and finally with water and then dried. The solvent is distilled off and the residue was chromatographed on a column of 10 g of magnesium silicate (Florisil). The column is developed with hexanes (SkellysolVe B) containing increasing portions of acetone.
The 21-acetic acid ester of 11ss, 17a, 21-trioxy-16a-methyl-1,4-pregnadiene-3,20-dione is eluted, which is freed from the solvent by evaporation.
Preparation <I> 8: </I> 21-acetic acid ester of 16a-methyl-17a, 21-dioxy-1,4,9 (11) -pregnatriene-3,20-dione. 5.63 g of N bromoacetamide are added to a solution of 8.5 g of 21-acetic acid ester of 11f, 17a, 21-trioxy-16a-methyl-1,4-pregnadiene-3,20-dione in 42.5 ml of pyridine .
After standing at room temperature for 15 minutes, the reaction solution is cooled to 5-10 C, and sulfur dioxide gas is passed over the surface with shaking until the solution shows no color on an acidified starch-iodine paper strip. During the addition of the sulfur dioxide, the reaction mixture becomes warm. Preparation <I> 9: </I> 21-acetic acid ester of 9a-bromo-11ss, 17a, 21-trioxy-16a-methyl-1,4-pregnadiene-3,20-dione. To a solution of 5.68 g of 21-acetic acid ester of 16a-methyl-17a, 21-dioxy-1,4,9 (11) -pregnatriene-3,20-dione in 100 ml of methylene chloride and 250 ml of tert. Butanol, a solution of 14 ml of 72% perchloric acid in 100 ml of water, followed by a solution 2,
34g N-bromoacetamide in 60 ml tert. Butanol, added. After the reaction mixture has been stirred for 15 minutes, a solution of 2.8 g of sodium sulfite in 140 ml of water is added and the reaction mixture is concentrated to a volume of about 500 ml under reduced pressure at about 50.degree. Preparation <I> 10: </I> 21-acetic acid ester of 9ss, llss-epoxy-16a-methyl-17a, 21-dioxy-1,4-pregnadiene-3,20-dione. 6.78 g of potassium acetate are added to a solution of 6.78 g of 21-acetic acid ester of 9a-bromine-lss, 17a, 21-trioxy-16a-methyl-1,4-pregnadiene-3,20-dione in 175 ml of acetone and the resulting suspension boiled under reflux for 17 hours.
The mixture is then concentrated to about 60 ml volume under reduced pressure on the steam bath and then diluted with water and extracted with methylene chloride. The methylene chloride extracts are combined, washed with water, dried over anhydrous sodium sulfate and evaporated.
The residue is redissolved in methylene chloride and chromatographed over 200 g of magnesium silicate (Florisil). The column is eluted with 1 liter portions of hexanes (Skellysolve B) containing increasing portions of acetone. Thus, 21-acetic acid ester is eluted from 9ss, 11ss-epoxy-16a-methyl-17, a .-, 21-dioxy-1,4-pregnadiene-3,20-dione,
which is freed from the solvent by evaporation. Preparation <I> 11: </I> 21-acetic acid ester of 9a-fluoro-llss, 17a, 21-trioxy-16a-methyl-1,4-pregnadiene-3,20-dione. To about 1.3 g of hydrogen fluoride in a polyethylene bottle, which is kept at -60 C, add 2.3 ml of tetrahydrofuran and then a solution of 500 mg (0.0012 mol) of 21-acetic acid ester of 9ss, 11ss - epoxy - 17a,
21-dioxy-16a-methyl-1,4-pregnadiene-3,20-dione in 2 ml of methylene chloride is added. The steroid solution is flushed with an additional milliliter of methylene chloride. The light red colored solution is kept at about -30 C for 1 hour and at -10 C for 2 hours. Then it is mixed with an excess of cold sodium bicarbonate solution and the organic material is extracted with the aid of additional methylene chloride.
The combined extracts are washed with water, dried over anhydrous sodium sulfate and concentrated to a volume of about 35 ml. This solution is chromatographed over 40 ml of magnesium silicate (Florisil). The column is developed with 500 ml portions of hexanes (Skellysolve B) containing increasing portions of acetone.
Thus, 21-acetic acid ester is eluted from 9a = fluorine-IIss, 17a, 21-trioxy-16a-methyl-1,4-pregnadiene-3,20-dione, which is freed from solvent by evaporation of the eluate fractions.
Preparation <I> 12: </I> 9a-fluoro-1 lss, 17a, 21-trioxy-16a-methyl-1,4-pregnadiene-3,20-dione. 3.25 g of 21-acetic acid ester of 9a-fluoro-1 lss, 17a, 21-trioxy-16a-methyl-1,4-pregnadiene-3,20-dione are dissolved in 325 ml of methanol, which was previously removed from air - Oxygen is released by blowing nitrogen through it for 10 minutes. A solution is prepared from 1 g (2.4 mM) lss, 17a, 21-trioxy-16a-methyl-4-pregnen-3,20-dione in 7 ml of pyridine. This solution is cooled to 0 ° C. and 0.3 ml of methanesulfonyl chloride is added. Then the solution is left to stand at 0-5 C for 2 hours and then diluted with water and extracted with three 25 ml portions of methylene chloride.
The extracts are combined, washed with cold dilute hydrochloric acid until the aqueous layer has a pH of 2-3, then washed with cold sodium bicarbonate solution and with water and finally dried over anhydrous sodium sulfate. Evaporation of the methylene chloride extract under reduced pressure leaves a residue of 21-methanesulfonic acid ester of IIIss, 17a, 21-trioxy-16a-methyl-4-pregnen-3,20-dione.
Using llss, 17a, 21-trioxy-16a-methyl-1,4-pregnadiene-3,20-dione as the starting steroid, 21-methanesulfonic acid esters of llss, 17a, 21-trioxy-16a-methyl are obtained according to the above procedure -1,4-pregnadiene-3,20-dione. Preparation <I> 14:
</I> 1 ss, 17a-Dioxy-16a-methyl-21-iodo-4-pregnen-3,20-dione. The crude 21-methanesulfonic acid ester of 11ss, 17a, 21-trioxy-16a-methyl-4-pregnen-3,20-dione described in preparation 13 is dissolved in 15 ml of acetone and with a solution of 1 g of sodium iodide in 10 ml of acetone offset. The mixture is heated under reflux with stirring for 15 minutes, the heat is reduced and the mixture is concentrated to a third volume under reduced pressure.
Ice and water are added and the precipitated product collected on a filter, washed with water and dried to give 11ss, 17a-dioxy-16a-methyl-21 iodine-4-pregnene-3,20-dione.
Using the 21-methanesulphonic acid ester of lss, 17a, 21-trioxy-16a-methyl-1,4-pregnadiene-3,20-dione as the starting steroid, one obtains llss, l7a-dioxy-16a-methyl- 21-iodine-1,4-pregnadiene-3,20-dione. <I> Example 1 </I> 11ss, 17a-Dioxy-16a-methyl-21-fluoro-4-pregnene-3,20-dione 1 g llss, 17a-Dioxy-16a-methy121 iodine-4-pregnene-3 ,
20-dione is dissolved in 150 ml of boiling wcetonitrile. After cooling to 40 ° C., the solution is protected from light and 0.8 ml of a 50% aqueous solution of silver fluoride are added with stirring. The product obtainable according to Example 1 or 5 can be oxidized to 16a-methyl-17a-oxy-21-fluoro-4-pregnen-3,11,20-trione as follows: A solution is prepared from 0.5 g of 16a- Methyl -1 lss, 17a-dioxy-21-fluoro-4-pregnen-3,20-dione, 0.15 g chromic acid, 10 ml glacial acetic acid and 1.1 ml water. This mixture is stirred and kept at room temperature for 8 hours.
The mixture is then poured into 50 ml of ice water, neutralized by adding dilute sodium hydroxide and the precipitate thus obtained is collected on a filter and recrystallized three times from a mixture of ethyl acetate and hexane (Skellysolve B) to produce 16a methyl-17a-oxy- 21-fluoro-1,4-pregnadiene-3,11,20-trione.
In the same way, 9a, 21-difluoro-1 lss, 17a-dioxy-16a-methyl-1,4-pregnadiene-3,20-dione and 9a, 21-difluoro-1 lss, 17a-dioxy-16a-methyl- 4-pregnen-3,20-dione are oxidized in 9a, 21-difluoro-16a-methyl-17a-oxy-1,4-pregnadiene-3,11,20-trione and 9a, 21-difluoro-16a-methyl- 17a-oxy-4-pregnen-3,11,20-trione. The corresponding 9a-chlorine compounds can be oxidized in the same way.
