Verfahren zur Herstellung eines neuen Antibiotikums Die vorliegende Erfindung betrifft ein Verfahren zur Herstellung eines neuen, wasserlöslichen Anti biotikums, das im folgenden mit A 9578 bezeichnet wird.
Das Antibiotikum A 9578 entsteht bei der Kultur eines neuen Actinomyceten-Stammes, der aus einer bei Boston, Mass., USA, gesammelten Erdprobe isoliert worden ist und der in unseren Laboratorien, sowie in der Eidg. Technischen Hochschule, Institut für spezielle Botanik unter der Bezeichnung A 9578 aufbewahrt wird.
Streptomyces A 9578 gehört der Art Strepto- myces griseoflavus (Krainsky) Waksman et Henrici an, unterscheidet sich aber von andern Vertretern dieser Art durch die Bildung des neuen wasser löslichen Antibiotikums. Es ist bis jetzt nur ein Stamm von Streptomyces griseoflavus bekannt, der ein Antibiotikum produziert. Dabei handelt es sich um das sogenannte Griseoflavin [Y. Waga, J.
Anti biotics Japan, A 6, 66 (1953)], das sich aber vom neuen Antibiotikum A 9578 schon durch seine Lös lichkeit in organischen Lösungsmitteln unterscheidet. Das Luftmycel des Streptomyces A 9578 ist aschgrau. Die Sporenträger sind verzweigt und bilden reichlich Spiralen mit meist 2-5 Windungen. Die Sporen selbst haben eine Grösse von 1,0-l,3 @t X 0,7-0,9 ,u und tragen an ihrer Oberfläche etwa 0,2 y lange, spitz zulaufende und an der Basis nur wenig verbreiterte Stacheln.
Das Wachstum ist relativ wenig temperatur abhängig, .sowohl bei 18 als auch bei 400 entwickelt sich der Pilz gut, doch liegt das Optimum zwischen 25 und 32 .
Zur weiteren Charakterisierung wird im folgenden das Wachstum von Streptomyces A 9578 auf ver schiedenen Nährmedien beschrieben. Die Nährmedien 1-7 sowie 10 wurden nach W. Lindenbein, Arch. Mikrobiol. 17, 361 (1952), hergestellt.

Zur Herstellung des Antibiotikums A 9578 kann man auch Varianten der Streptomyces-Art A 9578 verwenden, wie sie z. B. durch Selektionierung oder Mutation, insbesondere unter der Einwirkung von Ultraviolett- oder Röntgenstrahlen oder von Stick- stoff-Senfölen gewonnen werden. Ein derartiger Streptomyceten-Stamm wird, z. B. in wässeriger, Kohlenhydrate, stickstoffhaltige Verbindungen sowie anorganische Salze enthaltender Nährlösung, aerob gezüchtet, bis diese eine wesentliche antibakterielle Wirkung zeigt, und das Antibiotikum A 9578 hierauf isoliert.
Als assimilierbare Kohlenhydrate kommen z. B. Glucose, Saccharose, Lactose, Mannit, Stärke sowie Glycerin in Frage. Geeignete stickstoffhaltige Nähr stoffe und gegebenenfalls wachstumsfördernde Stoffe sind:
Aminosäuren, Peptide und Proteine, sowie deren Abbauprodukte, wie Pepton oder Trypton, ferner Fleischextrakte, wasserlösliche Anteile von Getreide körnern, wie Mais und Weizen, von Destillations- rückständen der Alkoholherstellung, von Hefe, Boh nen, insbesondere der Sojapflanze, von Samen, bei spielsweise der Baumwollpflanze usw., aber auch Ammoniumsalze und Nitrate. Von andern anorga nischen Salzen kann die Nährlösung beispielsweise Chloride, Carbonate, Sulfate von Alkalien, Erd alkalien, Magnesium, Eisen, Zink und Mangan ent halten.
Die Züchtung erfolgt aerob, also beispielsweise in ruhender Oberflächenkultur oder vorzugsweise sub- mers unter Schütteln oder Rühren mit Luft oder Sauerstoff in Schüttelflaschen oder den bekannten Fermentern. Als Temperatur eignet sich eine solche zwischen 18 und 40 . Eine wesentliche antibakterielle Wirkung zeigt die Nährlösung dabei im allgemeinen nach 11/2-5 Tagen.
Zur Isolierung des Antibiotikums A 9578 kann man z. B. wie folgt vorgehen: Man trennt das Mycel vom Kulturfiltrat ab, wonach die Hauptmenge des Antibiotikums im Kulturfiltrat gefunden wird. Es blei ben aber trotzdem namhafte Mengen des Antibioti kums am Mycel adsorbiert. Es ist daher vorteilhaft, letzteres gut auszuwaschen. Dazu eignen sich Wasser und wässrige organische Lösungsmittel, wie Alkohole, z. B. wässriges Methanol. Um das Antibiotikum dem Kulturfiltrat zu ent ziehen und zu reinigen, sind verschiedene Methoden geeignet, die einzeln oder in Kombination miteinander angewandt werden können.
Es hat sich als vorteilhaft erwiesen, die Kulturlösung während dieser Operatio nen auf einem pH zwischen 3 und 5 zu halten.
Man kann Adsorptionsmittel verwenden, z. B. Aktivkohlen, wie Norit (Markenprodukt), aktivierte Erden, wie Fullererde oder Floridin , oder Harz- adsorber, wie Asmit (beides Markenprodukte). Die Elution der Adsorbate erfolgt zweckmässig mit Ge mischen von mit Wasser mischbaren organischen Lö sungsmitteln mit Wasser oder wässrigen Säuren, z. B.
mit Wasser-Methanol-, Wasser-Pyridin-, verdünnte Essigsäure-Methanol- oder Wasser-Methanol-Eisessig- Butanol-Gemischen. Als besonders vorteilhaft für die Elution eines Noritadsorbates hat sich das Gemisch von Wasser (2 Volumteile), Methanol (1 Volumteil), Eisessig (1 Volumteil) und Butanol (2 Volumteile) erwiesen.
Als Adsorptionsmittel für das Antibiotikum kann man auch Kationenaustauscher verwenden; besonders geeignet sind Säuregruppen enthaltende Harze, wie Amberlite IRC-50 (Markenprodukt). Letzteres kann sowohl in der Säureform als auch in der Na triumform verwendet werden, doch hat sich ein Ge misch dieser beiden Formen im Volumenverhältnis 1 : 2 besonders bewährt. Die Elution geschieht zweck mässig mit verdünnten Säuren, z. B. mit methano- lischer Salzsäure.
Weiter kann das basische Antibiotikum auch direkt aus dem Kulturfiltrat ausgefällt werden, zum Beispiel mittels einer organischen Säure, zum Beispiel Pikrinsäure. Wenn man diese Fällung mit Salzen orga nischer Basen, z. B. mit Triäthylammoniumsulfat, oder mit verdünnten Säuren behandelt, wird das Anti biotikum in Form des entsprechenden Salzes erhalten. Man kann dabei sowohl in wässrigem Medium als auch in einem mit Wasser mischbaren Lösungsmittel, wie Methanol oder Aceton, arbeiten.
Diese überfüh- rung der schwerlöslichen Salze in leichtlösliche Salze des Antibiotikums wird entweder mit Mineralsäuren oder an Ionenaustauscherharzen, z. B. an Amberlite IRA 400 , durchgeführt.
Eine Anreicherung des Antibiotikums wird z. B. auch dadurch erreicht, dass man wässrige oder alko- holisch-wässrige Lösungen des Salzes mit einem über schuss von organischen, mit Wasser mischbaren Lö sungsmitteln, wie Aceton oder Dioxan, versetzt, wobei die Salze in fester Form ausgefällt werden.
Eine weitere Methode der Anreicherung des Anti biotikums besteht darin, dass man wässrige Lösungen desselben mit Lösungen von Phenol in Chloroform extrahiert, wobei sowohl das pH der wässrigen Lösung als auch der Phenolgehalt der Chloroformlösung variiert werden. So befindet sich. das Antibiotikum z.
B. bei einer Verteilung zwischen einer Lösung, die auf 100 cm3 Chloroform 100 g Phenol enthält, und einer wässrigen Phase von pH 1 bis 6 fast ausschliess lich in der organischen Phase, während es bei der Verwendung einer Lösung, die auf 100 cm3 Chloro form nur 33 g Phenol enthält, der wässrigen Phase lediglich bei einem pH zwischen 4 und 6 annähernd vollständig entzogen wird. Versteht man unter dem Verteilungskoeffizienten des Antibiotikums das Ver hältnis von Konzentration in der organischen Phase zur Konzentration in der wässrigen, so geht aus den obigen Angaben hervor,
dass der Verteilungskoeffi zient mit steigendem Phenolgehalt der organischen Phase zunimmt und mit sinkendem pH der wässrigen Phase abnimmt. Da es somit möglich ist, jeden belie bigen Verteilungskoeffizienten des Antibiotikums in diesem System einzustellen, kann man durch Kombi nation von wenigen Verteilungsoperationen einen Grossteil inaktiver Verunreinigungen abtrennen.
Eine andere Anreicherungsmethode für das Anti biotikum stellt die Chromatographie dar, wie Adsorp- tionschromatographie an verschiedenen Materialien, z. B. an Norit (Markenprodukt), Aluminiumoxyd, Magnesiumsilikaten, Silicagel, Calciumsulfat, sowie Verteilungschromatographie mit Cellulose, Stärke, Silicagel, Celite (Markenprodukt) als Trägersub stanzen,
oder aber Chromatographie an Ionenaus- tauscherharzen; z. B. an Dowex 50 , Amberlite IRC-50 (beides Markenprodukte). Beispielsweise hat sich die Verteilungschromatographie an Cellulose unter Verwendung des Lösungsmittelsystems Butanol (4 Volumteile), Eisessig (1 Volumteil) und Wasser (5 Volumteile) besonders. bewährt.
Weiter kann das Antibiotikum durch Gegenstrom verteilung nach Craig zwischen zwei nicht mischbaren Lösungsmittelphasen angereichert werden. Folgende Lösungsmittelsysteme haben sich dabei besonders be währt: a) sek. Butanol und 1/1o n Ammoniumacetat- puffer vom pH 4,68.
b) 1/"o n Ammoniumacetatpuffer vom pH 4,6 und 10 fl/o ige Lösung von Phenol in Chloroform. Der Verteilungskoeffizient des Antibiotikums im System b) und damit die Lage des Aktivitätsmaxs-mums in der Verteilung kann einerseits durch Änderung des pH der Pufferlösung und anderseits durch Variierung des Phenolgehaltes der organischen Phase beliebig ver ändert werden.
Zur Herstellung eines weitgehend reinen Präpa rates hat sich folgende Kombination der aufgeführten Anreicherungsverfahren bewährt: Aus dem Kultur filtrat wird das Antibiotikum am gepufferten Ionen- austauscher Amberlite IRC-50 adsorbiert und mit tels methanolischer Salzsäure eluiert. Die Eluate wer den bei pH 5 im Vakuum konzentriert,
wobei ein wässriges Konzentrat des Antibiotikums erhalten wird, das volumenmässig ungefähr 1/10o der Kulturlösung entspricht. Dieses wird einige Male zwischen Phenol- Chloroform-Gemischen einerseits und wässrigen Lö sungen von variierendem pH anderseits verteilt, wobei nach Gefriertrocknung ein Präparat erhalten wird, das in bezug auf das Kulturfiltrat eine um den Faktor 500-1000 erhöhte spezifische Wirksamkeit besitzt.
Durch Behandlung eines solchen Präparates mittels Verteilungschromatographie an Cellulose und Craig- scher Gegenstromverteilung kann das basische, weit gehend einheitliche Antibiotikum A 9578 vorzugs weise in Form eines Salzes erhalten werden.
Wie aus papierchromatographischen Untersuchun gen hervorgeht, besteht das Antibiotikum A 9578 aus mindestens zwei Komponenten, nämlich der Haupt komponente A und der Nebenkomponente B. Diese werden im Verlaufe der beschriebenen Anreiche rungsprozesse weitgehend voneinander getrennt, spe ziell bei der Verteilungschromatographie an Cellulose und bei der Gegenstromverteilung.
Die beiden Komponenten von A 9578 werden papierchromatographisch .definiert durcheinen direkten Vergleich ihrer Rf-Werte mit den RI- Werten einer Reihe von bekannten Antibiotika (2-l1) in den Systemen A-G (Tabelle).
Beim System H wird durchlaufend chromatographiert. Die angegebenen Zahlen dort die in cm gemessenen Laufstrecken. der Antibiotika nach 16stündiger Chromatogrammdauer. Der Nachweis der Antibiotika erfolgt autobiographisch mit Staphylococcus aureus oder Bacillus subtilis.

Die Wanderungsgeschwin digkeit ist etwa halb so gross wie diejenige des Strepto- mycins.
Die Komponente A des Antibiotikums A 9578 ist ein orangegelbes Pulver, das sich leicht in Wasser, gut in Methanol und schwer in :den meisten orga nischen Lösungsmitteln löst.
Die Base A gibt folgende Farbreaktionen: FeCl3: braunrot FeC13-K.Fe(CN)(,: blau Ninhydrin: schwach violett.
Der Sakaguchi-, Maltol- und Ehrlich-Morgantest sind negativ. Eine wässrige Lösung der Komponente A zeigt im UV-Spektrum Absorptionsmaxima bei 215 mu (E i m - 312) und bei 315 mu (E I % - 42).
Die freie Base des Antibiotikums A 9578 ist leicht aus ihren Salzen zugänglich, aus dem Sulfat z. B. durch Umsetzung in wässrigem Medium mit Barium hydroxyd, Neutralisierung des überschüssigen Baryts mit Kohlendioxyd sowie Abtrennung des Barium- carbonat- und -sulfat-Niederschlages und Isolierung der freien Base mittels Gefriertrocknung. Einfacher erfolgt die Herstellung aus den Salzen unter Verwen dung eines stark basischen Anionenaustauschers, z. B. der OH-Form des unter der Bezeichnung Dowex-2 (eingetragene Marke) im Handel befindlichen Pro duktes.
Eine der auffallendsten Eigenschaften des Anti biotikums A 9578 ist ein ausgeprägtes Stabilitäts minimum zwischen pH 7 und 11. Lösungen des Anti- biotikums verlieren bei 20 C und bei einem pH von 8 bis 10 innerhalb 48 Stunden den grössten Teil ihrer Aktivität, während sie bei der gleichen Temperatur bei einem pH von 1-5 vollständig und bei 6-7 und 11 teilweise wirksam bleiben.
Die Salze des Antibiotikums A 9578 und seiner Komponente leiten sich von anorganischen und orga nischen Säuren ab, beispielsweise von der Salzsäure, den Schwefelsäuren und Phosphorsäuren, der Essig-, Propion-, Valerien-, Palmitin- oder Ölsäure, der Bern steinsäure, Zitronensäure, Mandelsäure, Glutamin säure oder Pantothensäure. Sie stellen neutrale oder saure Salze dar. Ihre Herstellung erfolgt z. B. durch Einwirkung der entsprechenden Säuren auf die freie Base oder durch doppelte Umsetzung von Salzen, beispielsweise von A 9578-sulfat mit pantothensaurem Calcium.
Das Antibiotikum A 9578 besitzt eine sehr hohe antibiotische Wirksamkeit gegenüber verschiedenen Testorganismen. Im sogenannten Agarquerstrich-Test ist es aktiv gegen folgende Testorganismen: Micro- coccus pyogenes, var. aureus, Streptococcus viridans, Streptococcus faecalis, Corynebacterium diphteriae, Escherichia coli,
Bacillus megatherium und Bacillus subtilis.
In vivo ist das Antibiotikum A 9578 ebenfalls wirksam. Bei fünfmaliger subkutaner Verabreichung von 10<B>mg/ kg</B> an mit Staphylococcus pyogenes, var. aureus infizierte Mäuse werden 100 a/oi und bei einer Dosis von 5mal 5 mg/kg 50 o/a Überlebende beob- achtet. 50 % Überlebende erhält man,
wenn man Mäusen, die mit Escherichia coli infiziert sind, fünfmal 50 mg/kg subkutan appliziert.
Die Toxizität des Antibiotikums ist gering. So wird z. B. die subkutane Applikation von 1000 mg/kg von Mäusen ohne Schädigung ertragen. Höhere Dosen wurden noch nicht geprüft.
Das Antibiotikum A 9578, seine Komponenten, seine Salze oder Derivate können als Heilmittel Ver wendung finden. Diese enthalten die genannten Ver bindungen in Mischung mit einem für die enterale, parenterale oder lokale Applikation geeigneten phar mazeutischen organischen oder anorganischen Träger material.
In den nachfolgenden Beispielen sind die Tempe raturen in Celsiusgraden angegeben.
<I>Beispiel 1</I> Die Züchtung des Streptomyces A 9578 wird nach dem Submers.verfahren durchgeführt. Man ver wendet eine Nährlösung, die pro Liter Leitungswas ser 20 g Sojamehl und 20 g Mannit enthält. Die Nähr lösung wird in den Impfkolben oder in den Fermen- tern während 20-30 Minuten bei 1 atü sterilisiert. Die sterilisierte Nährlösung zeigt ein pH von 7,5 bis 8,0. Die Animpfung erfolgt mit bis zu 10 0/a einer teilweise sporulierenden vegetativen Kultur des Orga nismus.
Man inkubiert unter gutem Schütteln oder Rühren bei 27 , wobei Kulturen in Fermentern mit etwa 2 Vol. steriler Luft pro Vol. Lösung in: der Minute belüftet werden. Nach 48-120 Stunden Be- brütung hat die Kulturlösung den grössten Hemmwert gegenüber den Testorganismen (B. subtilis, B. mega- therium, Micrococcus pyogenes, var. aureus) erreicht.
Man unterbricht die Kultur, bringt das pH durch Zu gabe von verdünnter Schwefelsäure auf 4,5 und trennt das Mycel sowie andere feste Bestandteile von der die Hauptmenge des Antibiotikums enthaltenden Lö sung durch Filtrieren oder Zentrifugieren ab, wobei gegebenenfalls der Kulturlösung vor der Filtration etwa 1% eines Filterhilfsmittels, z. B. Hyflo Super- cel (Markenprodukt), zugesetzt wird.
Die Filterrück stände wäscht man mit Wasser und mit wässrigem Methanol und vereinigt die Waschflüssigkeiten, mit dem Kulturfiltrat.
Verwendet man anstelle der oben angegebenen Nährlösung solche, die pro Liter Leitungswasser die folgenden Nährstoffe enthalten, so erhält man nach analoger Züchtung und Aufarbeitung Kulturfiltrate von ähnlich hoher antibiotischer Wirksamkeit.

<tb> Natriumnitrat <SEP> 1 <SEP> g <I>Beispiel 2</I> 3 Liter eines gemäss Beispiel 1 erhaltenen Kultur filtrates werden durch Zugabe von verdünnter Natron lauge auf pH 7,5 gebracht und mit 50 g Aktivkohle ( Norit A ) versetzt.
Man lässt während 1 Stunde mechanisch rühren, wobei die gesamte antibiotisch wirksame Substanz von der Kohle adsorbiert wird. Letztere wird durch Filtration, vorteilhaft unter Zu gabe von etwas Filterhilfsmittel, wie z. B. Hyflo Supercel (Markenprodukt), von der vollständig in aktiven Lösung abgetrennt.
Die Kohle wird darauf in 500 cm3 einer Mischung von 4 Volumteilen Wasser und 1 Volumteil Pyridin eingetragen, die Mischung i/2 Stunde mechanisch gerührt und dann filtriert, worauf der Kohlerückstand noch einmal in gleicher Weise extrahiert wird.. Die Eluate enthalten die ge samte antibiotische Aktivität.
<I>Beispiel 3</I> 3 Liter eines gemäss Beispiel 1 erhaltenen Kultur filtrates werden durch Zugabe von verdünnter Na tronlauge auf pH 7,5 gebracht und dann sofort mit einer Durchlaufgeschwindigkeit von 0,5 Liter pro Stunde auf eine Säule von Amberlite IRC-50 (H-Form) gebracht, deren Länge 30 cm und deren Durchmesser 5 cm beträgt. Das Antibiotikum wird dabei vollständig adsorbiert. Die Säule wird mit 3 Liter Wasser gewaschen.
Zur Elution verwendet man 1 Liter 0;4 n Salzsäure. Die ersten 500 cm3 des Eluates sind antibiotisch unwirksam, während die zweiten 500 cm3 die gesamte Aktivität enthalten.
Dieses aktive Eluat wird zur Entfernung der über schüssigen Salzsäure durch eine Säule von Amber- lite IR 4B perkoiiert. Durch Gefriertrocknung des Perkolates erhält man das angereicherte Antibiotikum A 9578 in Form eines hochwirksamen amorphen Pulvers.
<I>Beispiel 4</I> 6 Liter eines gemäss Beispiel 1 erhaltenen Kultur filtrates werden durch Zugabe von verdünnter Natron lauge auf pH 7,5 gebracht und dann sofort durch eine Säule von Asmit <B>173 </B> perkoliert, die 14 cm lang ist und einen Durchmesser von 4,5 cm hat. Das Antibiotikum wird dabei vollständig adsorbiert. Man wäscht das Adsorberharz hierauf mit 3 Liter Wasser und eluiert das Antibiotikum mit 1 Liter einer Mischung von Methanol und 1 n Essigsäure (1 : 1).
Das Eluat, das die gesamte Aktivität enthält, wird im Vakuum bei 35 auf 15 cm3 konzentriert. Man versetzt diese Lösung mit 75 cms 0,25 n methano- lischer Salzsäure und giesst das Gemisch in 10 Liter Aceton, wobei das Hydrochlorid des Antibiotikums ausfällt. Es wird filtriert und mit Aceton gewaschen.
Zur weiteren Reinigung löst man es in 150 cm3 Methanol und filtriert die etwas trübe Lösung unter Zugabe von Celit . Nach Eindampfen des Filtrates im. Vakuum bei 25 erhält man das Hydrochlorid des Antibiotikums A 9578 in Formeines amorphen Pulvers.
<I>Beispiel 5</I> 30 Liter eines gemäss Beispiel 1 erhaltenen Kultur filtrates werden auf pH 4,5 gebracht und dann in einem Dünnschichteneindampfer auf 2 Liter einge- engt. Das Konzentrat bringt man durch Zugabe von verdünnter Natronlauge auf pH 8 und filtriert es unter Zugabe von Celit . Das klare Filtrat wird auf pH 5 gebracht und unter Rühren mit 1,5 Liter einer heissen, 5 0/0igen wässerigen Pikrinsäurelösung versetzt.
Der entstandene Niederschlag wird nach mehrstündigem Stehen bei 0 unter Zugabe von 50 g Celit abfiltriert. Das Filtrat ist antibiotisch nur sehr schwach aktiv. Der Filterrückstand wird hierauf dreimal mit je 800 cm3 kaltem Aceton ausgerührt und filtriert. Man engt das Filtrat im Vakuum auf 80 em3 ein, wobei das Pikrat des Antibiotikums und über schüssige Pikrinsäure ausfallen. Nach Abtrennung erhält man 8,5g Trockensubstanz.
Zur Isolierung des Antibiotikums als Hydro- chlorid werden 2,5 g der erwähnten Trockensubstanz in 30 cm3 Methanol gelöst. Man gibt hierauf zuerst eine Mischung von 1 em3 konz. Salzsäure und 10 cm3 Aceton und dann 500 cm3 Äther zu, wobei das Hydrochlorid quantitativ ausgefällt wird. Durch wiederholtes Lösen des Niederschlages in salzsaurem Methanol und Ausfällen mit Äther können, die letzten Reste Pikrinsäure daraus entfernt werden.
Schliesslich wird das Hydrochlorid in möglichst wenig Methanol gelöst, filtriert und im Vakuum eingedampft. Man erhält 714 mg des Hydrochlorides des Antibiotikums A 9578, das antibiotisch sehr aktiv ist.
<I>Beispiel 6</I> 600 Liter eines nach Beispiel 1 erhaltenen Kultur- filtrates werden mit 5,5 kg Hyflo Supercel (Mar kenprodukt) verrührt, mit 2,5 Liter 2n Salzsäure auf einen pH-Wert von 4,0 eingestellt und hierauf filtriert. Den Filterrückstand wäscht man mit 100 Litern Wasser.
Das klare Filtrat wird mit 7 kg vorbehan deltem Norit (Markenprodukt) während 45 Minuten verrührt. Die Vorbehandlung des Norits erfolgt durch mehrmaliges Ausrühren mit In Salzsäure und anschliessendem Neutralwaschen mit Wasser. Der mit Antibiotikum beladene Norit wird abfiltriert. Das Filtrat enthält keine antibiotische Aktivität. Das Norit -Adsorbat wird zweimal mit 200 Litern Was ser durch Rühren und Filtrieren gewaschen.
Die Waschflüssigkeit enthält keine Aktivität. Die Elution erfolgt durch zweimaliges, I ständiges Ausrühren der Aktivkohle mit einem Gemisch von n-Butanol- Methanol-EisessigWasser im Volumenverhältnis von 2: 1 : 1 :2, wobei die Aktivkohle durch Filtration abgetrennt wird. Die vereinigten Eluate (140 Liter + 46 Liter) werden mit 96 Litern n Butylacetat .gut durchmischt. Die sich abscheidende wässrige Phase wird abgetrennt und die organische Phase mit 1,2 Li tern Wasser nachgewaschen.
Die vereinigten wässrigen Phasen (65,5 Liter) werden sukzessive mit 72 Litern eines Gemisches von n-Butanol-n-Butylacetat im Volumenverhältnis von 1 : 2, 48 Liter Essigester und schliesslich mit 24 Liter Äther durch Ausrühren gewaschen. Die organischen Phasen werden verwor fen.
Die verbleibende wässrige Phase (44 Liter), die die gesamte antibiotische Aktivität enthält, wird im Quickfitt-Verdampfer bei höchstens 30 auf ein Volu men von 5,45 Litern konzentriert. Aus diesem hoch aktiven, schwarzbraun gefärbten Konzentrat erhält man das Antibiotikum durch Gefriertrocknung in Form von 509 g eines braunen Pulvers. Dieses Material zeigt in bezug auf das Kulturfiltrat eine 30-50fache spezifische antibiotische Aktivität.
<I>Beispiel 7</I> Zur Isolierung des Antibiotikums A 9578 aus 300 Litern eines nach Beispiel 1 erhaltenen Kultur filtrates werden 1 Volumteil Amberlite IRC-50 (Markenprodukt) in der H-Form mit 2 Volumteilen Amberlite IRC-50 in der Na-Form mechanisch gemischt. 6,3 Liter dieses Gemisches werden in eine Glassäule eingefüllt. Das Höhe: Durchmesser-Ver- hältnis des Harzbettes beträgt 8 : 1.
Das Kulturfiltrat wird mit 2n Salzsäure auf pH 4,0 .eingestellt und mit einer Durchlaufgeschwindigkeit von 0,2 Liter/Min./ Liter Harz durch das Harz perkoliert, wobei sich in den oberen =/.. der Säule eine orangebraune Zone aus bildet.
Das Harz wird anschliessend mit<B>30</B> Liter Was- ser und mit 60 Liter 80 % igem Methanol gewaschen. Der Durchlauf und die Waschflüssigkeiten enthalten nur wenig antibiotische Aktivität. Die Elution erfolgt in 2 Portionen mit insgesamt 37 Litern eines Ge misches von 8 Volumteilen Methanol und 2 Volum- teilen In Salzsäure.
Beide Portionen werden mit 5n NaOH auf pH 5;0, gestellt, vereinigt und hierauf im Zirkulationsverdampfer bei höchstens 30 auf 2 Liter konzentriert.
Das wässrige Konzentrat wird auf pH 5,6 gestellt und zwecks Entfernung von wenig unlöslichem Material durch Hyflo Supercel (Mar kenprodukt) filtriert. Dieses Filtrat (2,3 Liter) enthält annähernd die gesamte antibiotische Aktivität der Kulturlösung. Es wird in 4 Portionen mit insgesamt 500 cm3 eines Phenol-Chloroformgemisches, das 100 g Phenol in 100 cm3 Chloroform enthält, extra hiert. Die wässrige Phase wird verworfen.
Der Phenol- Chloroformextrakt (500 cm3) wird mit 1 Liter Chloro form verdünnt und mit 3mal 500 cm3 1/1o n Ammo- niumacetatpuffer von pH 4,60 ausgeschüttelt, wobei der organischen Phase gefärbte, antibiotisch inaktive Verunreinigungen entzogen werden. Die Chloroform phase wird nun mit 300 + 2mal 100 cm3 1/l0 n Salz säure extrahiert.
Der tiefrot gefärbte Säureextrakt, der das Antibiotikum enthält, wird mit Kaliumbicarbonat auf pH 3,5 gebracht und mit 4mal 50 cm3-Portionen Phenol-Chloroformgemisch (100 g : 100 cm3) zurück extrahiert. Man filtriert den Phenol-Chloroformextrakt durch Celite (Markenprodukt). Das klare, rotge färbte Filtrat (200 cm3) wird unter starkem Schütteln mit 50 cm3 Wasser, 500 cm- Äther und 300 cm3 Petroläther versetzt.
Nach dem Abtrennen der wäss- rigen Phase wird die organische Phase mit 2mal 50 cm3 Wasser nachgewaschen. Die vereinigten wäss- rigen Extrakte werden 2mal mit 500 em3 Äther und 1mal mit 500 cm3 Benzol ausgeschüttelt und hierauf lyophilisiert. Man erhält 2,60 g eines antibiotisch hochwirksamen, orangebraun gefärbten Pulvers. Dieses Material zeigt in bezug auf das Ausgangs material eine 500-1000fache spezifische antibiotische Aktivität (Aktivität pro Gewichtseinheit Trocken substanz).
Auf Whatman Nr. 1-Papier zeigt dieses Material im System n Butanol : n Butylacetat : Eis essig: Wasser =<B>10:</B> 3 : 1,3 :14,3 bei der autobio graphischen Entwicklung mit Staphylococcus aureus 2 Substanzflecken. Die langsamer wandernde anti- biotische Substanz wird als Base A, die 2,5mal schnel- ler wandernde Substanz als Base B bezeichnet.
Die Base A gibt auf dem Papier beim Besprühen mit FeC13 + K.FeCNE eine blaue Farbreaktion.
<I>Beispiel 8</I> 338 g eines nach Beispiel 6 erhaltenen Anti- biotikumpräparates werden in 1,5 Liter Wasser gelöst. Man setzt 150 g kristallines Ammoniumsulfat zu und extrahiert die Lösung mit 1 Liter + 2mal 500 cm3 eines Phenol-Chloroformgemisches, das 100 g Phenol in 100 cm', Chloroform enthält. Die vereinigten Phenol-Chloroformextrakte werden mit 750 cm3 In Salzsäure ausgeschüttelt und hierauf durch eine Schicht von Celite filtriert.
Das klare rotbraune Filtrat wird unter Rühren mit 600 cm3 Wasser, 4 Liter Äther und schliesslich mit 4 Liter Petroläther versetzt. Die wässrige Phase wird abgetrennt und die organische Phase mit 2mal 200 cm3 Wasser nach extrahiert. Die vereinigten wässrigen Phasen (1 Liter) werden 2mal mit 1 Liter Äther gewaschen und hierauf lyophilisiert. Man erhält 136 g eines braunen Pulvers, das in bezug auf das Ausgangsmaterial eine um den Faktor 2 erhöhte spezifische antibiotische Aktivität aufweist.
<I>Beispiel 9</I> 550 mg eines nach Beispiel 11 erhaltenen, stark aktiven Antibiotikumpräparates (Base A) werden in 55 ml eines 1(1o nAmmoniumacetatpuffers von pH 4,60 gelöst und mit 4mal 20 ml eines Phenol-Chloroform- gemisches, das 100 g Phenol in 400 ml Chloroform enthält, extrahiert. Die organischen Extrakte werden mit 2mal 15 ml Pufferlösung zurückgewaschen.
Der das Antibiotikum enthaltende organische Extrakt (80 ml) wird mit 40 ml Chloroform verdünnt, noch einmal mit 60 ml Pufferlösung gewaschen und hier auf durch ein doppeltes Faltenfilter filtriert. Das tief rot gefärbte Filtrat extrahiert man mit 30+20+10m1 0,2n Salzsäure. Die das Antibiotikum enthaltende Säurelösung wird mit 50 ml Wasser verdünnt und mit einem Phenol-Chloroformgemisch, das 100 g Phenol in 100 ml Chloroform enthält, erschöpfend extrahiert (2 X 20+ 10 ml).
Die vereinigten Phenol-Chloroform- extrakte werden durch ein doppeltes Faltenfilter fil triert und mit 20 ml Wasser, 200 ml Äther und 100 ml Petroläther geschüttelt. Nach dem Abtrennen der wässrigen Phase wird die organische Phase mit 2mal 15 ml Wasser nachextrahiert. Die vereinigten wässrigen Extrakte werden 2mal mit 50 ml Äther und 1mal mit 50 ml Benzol gewaschen und hierauf lyophilisiert. Man erhält 117 mg eines braunroten Pulvers, das in bezug auf das Ausgangsmaterial eine annähernd 5fache Anreicherung der antibiotischen Aktivität aufweist.
<I>Beispiel 10</I> 700 mg eines nach Beispiel 7 erhaltenen Anti- biotikumpräparates werden an 127g Whatman Nr. 1 Cellulosepulver chromatographiert. Als Elutionsmittel verwendet man die obere Phase eines Gemisches von 4 Teilen Butanol, 1 Teil Eisessig und 5 Teilen Was ser. Der abgetrennten oberen Phase werden 10 Vol:o/o reines Butanol zugesetzt.
Die Substanz wird mit der 10fachen Menge Cellulosepulver verrieben und als Pulver auf die Säule aufgetragen. Die Durchlauf- geschwindigkeit beträgt 15-20 ml/Stunde. Es wer den Fraktionen zu 40 ml aufgefangen. Die einzelnen Fraktionen werden mit 50 ml Petroläther geschüttelt. Die ausgeschiedene wässrige Phase wird mit Benzol gewaschen und lyophilisiert. Die Hauptmenge der antibiotischen Aktivität befindet sich in den Frak tionen 7-8 (121 mg) und 10-13 (142 mg).
Gemäss papierchromatographischen Untersuchungen enthalten die Fraktionen 7-8 vorwiegend Base B, die Frak tionen 10-13 vorwiegend Base A.
<I>Beispiel l l</I> 3 g eines nach Beispiel 8 erhaltenen Antibiotikum präparates werden in einer Craigschen Verteilungs apparatur in System sek. Butanol-O,ln Ammonium acetatpuffer pH 4,68 über 100 Stufen verteilt. Jede Einheit enthält je 100 ml obere und untere Phase. Die Substanz wird in die Einheit Nr. 3 eingefüllt. Die Auf arbeitung erfolgt durch Versetzen des Gemisches der beiden Phasen mit dem doppelten Volumen Petrol- äther und Lyophilisation der wässrigen: Phase. Die dunkel gefärbten Fraktionen 3-11 enthalten nur wenig antibiotische Aktivität.
Die orangegelb gefärb ten Fraktionen 12-20 (631 mg) enthalten den Haupt teil der antibiotischen Aktivität (vorwiegend Base A). Die gelb gefärbten Fraktionen 21-40 sind schwächer aktiv und enthalten ein Gemisch von Base A und B, in dem die letztere überwiegt.
<I>Beispiel 12</I> 200 mg eines nach Beispiel 10 erhaltenen Anti- biotikumpräparates (Base A) werden in einer Craib scheu Verteilungsapparatur im System 1/zon Ammo- niumacetatpuffer pH 4,58 - 10 %, Phenol in Chloro- form über 29 Stufen verteilt. Jede Stufe enthält je 10 ml obere und untere Phase.
Die Hauptmenge der antibiotischen Aktivität befindet sich in den Fraktio nen 6-15. Diese werden vereinigt (etwa 200 ml), mit 400 ml Äther und 300 ml Petroläther versetzt und geschüttelt. Die abgetrennte, orangerot gefärbte wäss- rige Phase wird mit Chloroform gewaschen und mit 20 + 3mal 10 ml eines Gemisches von 100 g Phenol in 100 ml Chloroform extrahiert.
Den Phenol-Chloro- formextrakt versetzt man unter Schütteln mit 20 ml Wasser, 300 ml Äther und 200 ml Petroläther. Die rotgefärbte wässrige Phase wird abgetrennt, mit viel Äther und Benzol gewaschen und lyophilisiert. Man erhält 86,4 mg der Base A in Form eines gelben wasserlöslichen Pulvers.
UV-Spektrum in Wasser: Amax: 215 m@c (E i <B>%</B> = 312), 315 m,u (Ei m = 42). Farbreaktionen: FeC13: braunrot; FeC13 + K.Fe(CN),: blau; Ninhydrin: schwach positiv.

Process for the Production of a New Antibiotic The present invention relates to a process for the production of a new, water-soluble antibiotic, which is referred to below with A 9578.
The antibiotic A 9578 arises from the culture of a new actinomycete strain, which was isolated from a soil sample collected near Boston, Mass., USA and which is in our laboratories and in the Federal Institute of Technology, Institute for Special Botany under the Designation A 9578 is kept.
Streptomyces A 9578 belongs to the species Streptomyces griseoflavus (Krainsky) Waksman et Henrici, but differs from other representatives of this species in the formation of the new water-soluble antibiotic. So far, only one strain of Streptomyces griseoflavus is known to produce an antibiotic. This is the so-called griseoflavin [Y. Waga, J.
Anti biotics Japan, A 6, 66 (1953)], which differs from the new antibiotic A 9578 by its solubility in organic solvents. The aerial mycelium of Streptomyces A 9578 is ash gray. The spore carriers are branched and form copious spirals with mostly 2-5 turns. The spores themselves have a size of 1.0-1.3 x 0.7-0.9, u and have spines about 0.2 y long, tapering to a point and only slightly broadened at the base.
Growth is relatively little dependent on temperature, the fungus develops well at both 18 and 400, but the optimum is between 25 and 32.
For further characterization, the growth of Streptomyces A 9578 on various nutrient media is described below. The nutrient media 1-7 and 10 were according to W. Lindenbein, Arch. Mikrobiol. 17, 361 (1952).

Such a Streptomycete strain is e.g. B. in aqueous, carbohydrates, nitrogenous compounds and inorganic salts containing nutrient solution, grown aerobically until it shows a significant antibacterial effect, and the antibiotic A 9578 is isolated on it.
As assimilable carbohydrates z. B. glucose, sucrose, lactose, mannitol, starch and glycerol in question. Suitable nitrogenous nutrients and, if necessary, growth-promoting substances are:
Amino acids, peptides and proteins, as well as their breakdown products such as peptone or tryptone, also meat extracts, water-soluble parts of cereal grains such as maize and wheat, of distillation residues from alcohol production, of yeast, beans, in particular of the soy plant, of seeds for example the cotton plant etc., but also ammonium salts and nitrates. The nutrient solution can contain other inorganic salts, for example chlorides, carbonates, sulfates of alkalis, alkaline earths, magnesium, iron, zinc and manganese.
The cultivation takes place aerobically, for example in resting surface culture or preferably submerged with shaking or stirring with air or oxygen in shaker bottles or the known fermenters. A suitable temperature is between 18 and 40. The nutrient solution generally shows a significant antibacterial effect after 11 / 2-5 days.
To isolate the antibiotic A 9578 you can, for. B. proceed as follows: The mycelium is separated from the culture filtrate, after which the main amount of the antibiotic is found in the culture filtrate. Nevertheless, significant amounts of the antibiotic remain adsorbed on the mycelium. It is therefore advantageous to wash out the latter well. Water and aqueous organic solvents such as alcohols, e.g. B. aqueous methanol. Various methods that can be used individually or in combination with one another are suitable for removing the antibiotic from the culture filtrate and purifying it.
It has proven advantageous to keep the culture solution at a pH between 3 and 5 during these operations.
One can use adsorbents, e.g. B. activated carbons such as Norit (branded product), activated earths such as fuller earth or floridin, or resin adsorbers such as Asmit (both branded products). The elution of the adsorbates is conveniently carried out with Ge mixing of water-miscible organic solvents with water or aqueous acids, eg. B.
with water-methanol, water-pyridine, dilute acetic acid-methanol or water-methanol-glacial acetic acid-butanol mixtures. The mixture of water (2 parts by volume), methanol (1 part by volume), glacial acetic acid (1 part by volume) and butanol (2 parts by volume) has proven to be particularly advantageous for the elution of a norit adsorbate.
Cation exchangers can also be used as adsorbents for the antibiotic; Resins containing acid groups, such as Amberlite IRC-50 (branded product), are particularly suitable. The latter can be used both in the acid form and in the sodium form, but a mixture of these two forms in a volume ratio of 1: 2 has proven particularly useful. The elution is conveniently done with dilute acids such. B. with methanolic hydrochloric acid.
Furthermore, the basic antibiotic can also be precipitated directly from the culture filtrate, for example by means of an organic acid, for example picric acid. Enrichment of the antibiotic is z. B. also achieved by adding an excess of organic, water-miscible solvents, such as acetone or dioxane, to aqueous or alcoholic-aqueous solutions of the salt, the salts being precipitated in solid form.
Another method of enriching the antibiotic consists in extracting aqueous solutions of the same with solutions of phenol in chloroform, with both the pH of the aqueous solution and the phenol content of the chloroform solution being varied. So is. the antibiotic z.
B. with a distribution between a solution that contains 100 g of phenol per 100 cm3 of chloroform and an aqueous phase of pH 1 to 6 almost exclusively in the organic phase, while when using a solution that contains 100 cm3 of chloroform contains only 33 g of phenol, is almost completely removed from the aqueous phase only at a pH between 4 and 6. If one understands by the distribution coefficient of the antibiotic the ratio of the concentration in the organic phase to the concentration in the aqueous phase, then from the above it follows that

Another enrichment method for the antibiotic is chromatography, such as adsorption chromatography on various materials, e.g. B. Norit (branded product), aluminum oxide, magnesium silicates, silica gel, calcium sulfate, as well as distribution chromatography with cellulose, starch, silica gel, Celite (branded product) as carrier substances,
or else chromatography on ion exchange resins; z. B. Dowex 50, Amberlite IRC-50 (both branded products). For example, partition chromatography on cellulose using the solvent system butanol (4 parts by volume), glacial acetic acid (1 part by volume) and water (5 parts by volume) has proven particularly successful. proven.

whereby an aqueous concentrate of the antibiotic is obtained which corresponds in volume to approximately 1 / 10o of the culture solution. This is distributed a few times between phenol-chloroform mixtures on the one hand and aqueous solutions of varying pH on the other hand, a preparation being obtained after freeze-drying which has a specific effectiveness increased by a factor of 500-1000 with respect to the culture filtrate.
By treating such a preparation by means of partition chromatography on cellulose and Craig's countercurrent distribution, the basic, largely uniform antibiotic A 9578 can preferably be obtained in the form of a salt.
As can be seen from paper chromatographic investigations, the antibiotic A 9578 consists of at least two components, namely the main component A and the secondary component B. These are largely separated from one another in the course of the enrichment processes described, especially in partition chromatography on cellulose and in countercurrent distribution .
The two components of A 9578 are defined by paper chromatography by a direct comparison of their Rf values with the RI values of a number of known antibiotics (2-11) in systems A-G (table).
System H is continuously chromatographed. The numbers given are the running distances measured in cm. of antibiotics after 16 hours of chromatogram. The antibiotics are detected autobiographically with Staphylococcus aureus or Bacillus subtilis.

A nutrient solution is used which contains 20 g of soy flour and 20 g of mannitol per liter of tap water. The nutrient solution is sterilized in the inoculation flask or in the fermenters for 20-30 minutes at 1 atm. The sterilized nutrient solution shows a pH of 7.5 to 8.0. The inoculation takes place with up to 10 0 / a of a partially sporulating vegetative culture of the organism.
Incubate at 27 with good shaking or stirring, aerating cultures in fermenters with about 2 vol. Sterile air per vol. Solution every minute. The antibiotic is completely adsorbed. The column is washed with 3 liters of water.
1 liter of 0.4N hydrochloric acid is used for the elution. The first 500 cm3 of the eluate are antibiotic ineffective, while the second 500 cm3 contain the entire activity.
This active eluate is percolated through a column of Amberlite IR 4B to remove the excess hydrochloric acid. The enriched antibiotic A 9578 is obtained in the form of a highly effective amorphous powder by freeze drying the percolate.
<I> Example 4 </I> 6 liters of a culture filtrate obtained according to Example 1 are brought to pH 7.5 by adding dilute sodium hydroxide solution and then immediately percolated through a column of Asmit 173, which 14 cm long and 4.5 cm in diameter. The antibiotic is completely adsorbed. The adsorber resin is then washed with 3 liters of water and the antibiotic is eluted with 1 liter of a mixture of methanol and 1N acetic acid (1: 1).
The eluate, which contains the entire activity, is concentrated in vacuo at 35 to 15 cm3. This solution is mixed with 75 cms of 0.25N methanolic hydrochloric acid and the mixture is poured into 10 liters of acetone, the hydrochloride of the antibiotic precipitating out. It is filtered and washed with acetone.
For further purification, it is dissolved in 150 cm3 of methanol and the somewhat cloudy solution is filtered with the addition of Celite. After evaporation of the filtrate in. At 25 vacuum, the hydrochloride of antibiotic A 9578 is obtained in the form of an amorphous powder.
<I> Example 5 </I> 30 liters of a culture filtrate obtained according to Example 1 are brought to pH 4.5 and then concentrated to 2 liters in a thin-layer evaporator. The concentrate is brought to pH 8 by adding dilute sodium hydroxide solution and filtered while adding Celite. The clear filtrate is brought to pH 5 and, while stirring, 1.5 liters of a hot, 50/0 aqueous picric acid solution are added.
After standing at 0 for several hours, the precipitate formed is filtered off with the addition of 50 g of Celite. The filtrate has only very weak antibiotic activity. The filter residue is then stirred three times with 800 cm3 of cold acetone each time and filtered. To isolate the antibiotic as the hydrochloride, 2.5 g of the dry substance mentioned are dissolved in 30 cm3 of methanol. A mixture of 1 em3 conc. Hydrochloric acid and 10 cm3 acetone and then 500 cm3 ether are added, the hydrochloride being precipitated quantitatively. By repeatedly dissolving the precipitate in hydrochloric acid methanol and precipitating it with ether, the last remains of picric acid can be removed from it.
Finally, the hydrochloride is dissolved in as little methanol as possible, filtered and evaporated in vacuo. 714 mg of the hydrochloride of the antibiotic A 9578, which is very antibiotically active, are obtained.
<I> Example 6 </I> 600 liters of a culture filtrate obtained according to Example 1 are stirred with 5.5 kg of Hyflo Supercel (branded product) and adjusted to a pH of 4.0 with 2.5 liters of 2N hydrochloric acid and then filtered. The filter residue is washed with 100 liters of water.
The clear filtrate is stirred with 7 kg of pre-treated Norit (branded product) for 45 minutes. The Norit is pretreated by stirring it several times with In hydrochloric acid and then washing it with neutral water. The norit laden with antibiotic is filtered off. The filtrate does not contain any antibiotic activity. The Norit adsorbate is washed twice with 200 liters of water by stirring and filtering.
The washing liquid contains no activity. Elution takes place by stirring the activated carbon twice continuously with a mixture of n-butanol-methanol-glacial acetic acid / water in a volume ratio of 2: 1: 1: 2, the activated carbon being separated off by filtration. The combined eluates (140 liters + 46 liters) are mixed well with 96 liters of n butyl acetate. The aqueous phase which separates out is separated off and the organic phase is washed with 1.2 liters of water.
The combined aqueous phases (65.5 liters) are washed successively with 72 liters of a mixture of n-butanol-n-butyl acetate in a volume ratio of 1: 2, 48 liters of ethyl acetate and finally with 24 liters of ether by stirring. The organic phases are discarded.
The remaining aqueous phase (44 liters), which contains all of the antibiotic activity, is concentrated in the Quickfitt evaporator at a maximum of 30 to a volume of 5.45 liters. From this highly active, black-brown colored concentrate, the antibiotic is obtained by freeze-drying in the form of 509 g of a brown powder. This material shows a 30-50-fold specific antibiotic activity with respect to the culture filtrate.
<I> Example 7 </I> To isolate the antibiotic A 9578 from 300 liters of a culture filtrate obtained according to Example 1, 1 part by volume of Amberlite IRC-50 (branded product) in the H form with 2 parts by volume of Amberlite IRC-50 in the Na -Form mechanically mixed. 6.3 liters of this mixture are poured into a glass column. The height: diameter ratio of the resin bed is 8: 1.
The culture filtrate is adjusted to pH 4.0 with 2N hydrochloric acid and percolated through the resin at a flow rate of 0.2 liters / min. / Liter of resin, an orange-brown zone forming in the upper = / .. of the column.
The resin is then washed with <B> 30 </B> liters of water and with 60 liters of 80% methanol. The run-through and wash liquids contain little antibiotic activity. Elution takes place in 2 portions with a total of 37 liters of a mixture of 8 parts by volume of methanol and 2 parts by volume of hydrochloric acid.
Both portions are adjusted to pH 5.0 with 5N NaOH, combined and then concentrated to 2 liters in a circulation evaporator at a maximum of 30.
The aqueous concentrate is adjusted to pH 5.6 and filtered through Hyflo Supercel (branded product) to remove little insoluble material. This filtrate (2.3 liters) contains almost all of the antibiotic activity of the culture solution. It is extracted in 4 portions with a total of 500 cm3 of a phenol-chloroform mixture containing 100 g of phenol in 100 cm3 of chloroform. The aqueous phase is discarded.
The phenol-chloroform extract (500 cm3) is diluted with 1 liter of chloroform and extracted 3 times with 500 cm3 1 / 1o N ammonium acetate buffer of pH 4.60, whereby the organic phase is removed from the colored, antibiotically inactive impurities. The chloroform phase is then extracted with 300 + 2 times 100 cm3 1/10 N hydrochloric acid.
The deep red colored acid extract, which contains the antibiotic, is brought to pH 3.5 with potassium bicarbonate and extracted back with 4 times 50 cm3 portions of phenol-chloroform mixture (100 g: 100 cm3). The phenol-chloroform extract is filtered through Celite (branded product). The clear, red-colored filtrate (200 cm3) is mixed with 50 cm3 of water, 500 cm3 of ether and 300 cm3 of petroleum ether with vigorous shaking.
After the aqueous phase has been separated off, the organic phase is washed twice with 50 cm3 of water. The combined aqueous extracts are shaken out twice with 500 cm3 of ether and once with 500 cm3 of benzene and then lyophilized. 2.60 g of a highly antibiotic, orange-brown powder are obtained. This material shows a 500-1000-fold specific antibiotic activity (activity per unit weight of dry substance) in relation to the starting material.
On Whatman No. 1 paper shows this material in the system n butanol: n butyl acetate: glacial acetic acid: water = <B> 10: </B> 3: 1.3: 14.3 during autobiographical development with Staphylococcus aureus 2 Substance stains. The antibiotic substance that migrates more slowly is referred to as base A, and the substance that migrates 2.5 times faster as base B.
Base A gives a blue color reaction on paper when sprayed with FeC13 + K.FeCNE.
<I> Example 8 </I> 338 g of an antibiotic preparation obtained according to Example 6 are dissolved in 1.5 liters of water. 150 g of crystalline ammonium sulfate are added and the solution is extracted with 1 liter + 2 times 500 cm3 of a phenol-chloroform mixture containing 100 g of phenol in 100 cm3 of chloroform. The combined phenol-chloroform extracts are shaken out with 750 cm3 of hydrochloric acid and then filtered through a layer of Celite.
The clear red-brown filtrate is mixed with 600 cm3 of water, 4 liters of ether and finally 4 liters of petroleum ether while stirring. The aqueous phase is separated off and the organic phase is extracted twice with 200 cm3 of water. The combined aqueous phases (1 liter) are washed twice with 1 liter of ether and then lyophilized. 136 g of a brown powder are obtained which, in relation to the starting material, has a specific antibiotic activity which is increased by a factor of 2.
<I> Example 9 </I> 550 mg of a strongly active antibiotic preparation (base A) obtained according to Example 11 are dissolved in 55 ml of a 1 n ammonium acetate buffer of pH 4.60 and mixed with 4 times 20 ml of a phenol-chloroform mixture containing 100 g of phenol in 400 ml of chloroform, and the organic extracts are backwashed twice with 15 ml of buffer solution.
The organic extract (80 ml) containing the antibiotic is diluted with 40 ml of chloroform, washed once more with 60 ml of buffer solution and filtered through a double folded filter. The deep red colored filtrate is extracted with 30 + 20 + 10m1 0.2N hydrochloric acid. The acid solution containing the antibiotic is diluted with 50 ml of water and extracted exhaustively with a phenol-chloroform mixture containing 100 g of phenol in 100 ml of chloroform (2 × 20+ 10 ml).
The combined phenol-chloroform extracts are filtered through a double folded filter and shaken with 20 ml of water, 200 ml of ether and 100 ml of petroleum ether. After the aqueous phase has been separated off, the organic phase is extracted twice with 15 ml of water. The combined aqueous extracts are washed twice with 50 ml of ether and once with 50 ml of benzene and then lyophilized. 117 mg of a brown-red powder are obtained which has an approximately 5-fold increase in antibiotic activity with respect to the starting material.
<I> Example 10 </I> 700 mg of an antibiotic preparation obtained according to Example 7 are chromatographed on 127 g Whatman No. 1 cellulose powder. The eluent used is the upper phase of a mixture of 4 parts of butanol, 1 part of glacial acetic acid and 5 parts of water. 10 vol: o / o pure butanol are added to the separated upper phase.
The substance is triturated with 10 times the amount of cellulose powder and applied to the column as a powder. The throughput speed is 15-20 ml / hour. The fractions of 40 ml were collected. The individual fractions are shaken with 50 ml of petroleum ether. The separated aqueous phase is washed with benzene and lyophilized. Most of the antibiotic activity is found in fractions 7-8 (121 mg) and 10-13 (142 mg).
According to paper chromatographic investigations, fractions 7-8 predominantly contain base B, and fractions 10-13 predominantly contain base A.
<I> Example l l </I> 3 g of an antibiotic preparation obtained according to Example 8 are in a Craig distribution apparatus in system sec. Butanol-O, ln ammonium acetate buffer pH 4.68 distributed over 100 levels. Each unit contains 100 ml each of the upper and lower phases. The substance is poured into unit No. 3. The work-up is carried out by adding twice the volume of petroleum ether to the mixture of the two phases and lyophilizing the aqueous phase. The dark colored fractions 3-11 contain little antibiotic activity.
The orange-yellow colored fractions 12-20 (631 mg) contain the main part of the antibiotic activity (predominantly base A). The yellow-colored fractions 21-40 are less active and contain a mixture of base A and B in which the latter predominates.
<I> Example 12 </I> 200 mg of an antibiotic preparation (base A) obtained according to Example 10 are added in a Craib scheu distribution apparatus in the system 1 / zon ammonium acetate buffer pH 4.58-10%, phenol in chloroform distributed over 29 levels. Each stage contains 10 ml each of the upper and lower phases.
The bulk of the antibiotic activity is in fractions 6-15. These are combined (about 200 ml), mixed with 400 ml of ether and 300 ml of petroleum ether and shaken. The separated, orange-red colored aqueous phase is washed with chloroform and extracted with 20 + 3 times 10 ml of a mixture of 100 g of phenol in 100 ml of chloroform.
The phenol-chloroform extract is mixed with 20 ml of water, 300 ml of ether and 200 ml of petroleum ether while shaking. The red colored aqueous phase is separated off, washed with plenty of ether and benzene and lyophilized. 86.4 mg of base A are obtained in the form of a yellow, water-soluble powder.
UV spectrum in water: Amax: 215 m @ c (E i <B>% </B> = 312), 315 m, u (Ei m = 42). Color reactions: FeC13: brown-red; FeC13 + K.Fe (CN) ,: blue; Ninhydrin: weakly positive.
Sakaguchi, Maltol, Elson-Morgan: negative.
