Verfahren zur Herstellung von 17-Alkyl-19-nortestosteronen Die vorliegende Erfindung bezieht sich auf ein Verfahren zur Herstellung einer neuen Gruppe von blutdrucksenkenden, den Aufbaustoffwechsel beein flussenden Mitteln mit geringer androgener Wirksam-. keit, nämlich von 17 Alkyl-19-nortestosteronen, deren Alkylgruppe 2 bis 8 Kohlenstoffatome auf weist.

Die erfindungsgemäss erhältlichen Verbindungen sind wertvolle, anabolische Mittel, indem sie die Re tention von Stickstoff begünstigen. Diese Wirkung tritt bei Dosierungen ein, die einen nur sehr geringen. Grad androgener Wirksamkeit verursachen.
Es ist bekannt, dass Testosteronpropionat ein wirksames, den Aufbaustoffwechsel beeinflussendes Mittel dar- stellt. Seine klinische Verwendbarkeit für diesen Zweck ist jedoch weitgehend begrenzt, da bei vielen Patienten die androgenen Wirkungen unerwünscht sind.
Bei den erfindungsgemäss erhältlichen Verbin dungen sind die wirksamen, den Aufbaustoffwechsel beeinflussenden Dosierungen so gering, dass auch bei langdauernden Verabreichungen bisher keine uner wünschten Nebeneffekte festgestellt wurden. Eine weitere wichtige Eigenschaft dieser Verbin dungen ist ihre blutdrucksenkende Wirkung, eine Wirkung, die das 19 Nortestosteron oder sein 17-Me- thyld'erivat nicht aufweist.
Die erfindungsgemäss er hältlichen Verbindungen sind besonders wirksam bei der Überwindung des hohen Blutdruckes, der durch das Mineralocorticoidhormon Desoxycortico- steron erzeugt wird.
Das erfindungsgemässe Verfahren zur Herstellung der 17 Alkyl-19-nortestosterone besteht in der Hy-, drierung eines 19-Nortestosterons, das in. 17a-Stel- lung eine ungesättigte aliphatische Kohlenwasser stoffgruppe mit 2 bis 8 Kohlenstoffatomen enthält. Solche Gruppen können ungesättigte Bindungen des Äthylen- oder Acetylentyps enthalten.
Zu ihnen gehören beispielsweise Athinyl-, Propinyl-, Butinyl-, Pentinyl-, Hexinyl-, Heptinyl- und Octinylgruppen sowie Vinyl-, Propenyl-, Allylgruppen und deren höhere Homologen.
Die Hydrierung wird beispiels weise folgendermassen durchgeführt: 17-Äthinyl-19- nortestosteron wird in einer Lösung oder Suspen sion in einer Wasserstoffatmosphäre in Gegenwart eines Katalysators, beispielsweise eines Edelmetall- katalysators, wie Palladium auf Holzkohle, geschüt telt. Diese Verbindung wird stufenweise zu 17-Vinyl- 19-nortestosteron und dann zu 17 Äthyl.-19-nortesto- steron reduziert.
Unter den hier beschriebenen Reak- tionsbedingungen tritt eine Hydrierung der im Kern befindlichen Doppelbindung nur in verhältnismässig geringfügigem Masse ein. Eine geeignete andere Ar beitsweise zur Herstellung von 17-Äthyl-19-nortesto- steron ist die katalytische Reduktion von 17-Vinyl- 19-nortestosteron. Die Ausgangsstoffe können durch Anlagerung von Acetylen,
einem Homologen des Acetylens oder einer entsprechenden Organo-Metall- verbindung an ein 17-Keto-steroid erhalten werden. Beispielsweise kann 17-Äthinyl-19-nortestosteron durch Anlagerung von Acetylen an 3-Methoxy- 2,5(10)-östradien-17-on und anschliessende, durch Säure katalysierte Hydrolyse und Isomerisierung des erhaltenen Produktes hergestellt werden.
Ein anderes Beispiel ist die Anlagerung von Allylmagnesium- bromid an 3-Methoxy-2,5(10)-östradien-17-on, wobei die anschliessende saure Hydrolyse und Isomerisie- rung 17-Allyl 19-nortestosteron ergibt.
<I>Beispiel 1</I> Einer am Rückflusskühler siedenden Lösung von 47,5 Teilen 3 - Methoxy - 13 - methyl - 17a - äthinyl 1,4,6,7,8,9,11,12,13,14,16,17-dodecahydro-15-cyclo- penta-[a]-phenanthren-17-ol in 3200 Teilen Methanol und 1000 Teilen Wasser werden 240 Teile konzen trierter Salzsäure zugesetzt. Die Rückflussbehandlung wird weitere 5 Minuten fortgesetzt, worauf die Lö sung 15 Minuten lang bei Raumtemperatur gehalten wird. Dann werden 13 000 Teile Wasser zugesetzt, und das Gemisch wird auf 0 C gekühlt.
Nach mehr stündigem Stehen bei dieser Temperatur wird das Gemisch filtriert und die Fällung getrocknet und aus Äthylacetat umkristallisiert. Das auf diese Weise erhaltene 17 Äthinyl-19-nortestosteron schmilzt bei etwa 202 bis 24011 C.
Durch ,ein Gemisch aus 11 Teilen Holzkohle, die 5 0!o Palladium enthält, und 2000 Teilen Dioxan wird 60 Minuten lang ein Wasserstoffstrom geleitet. Dann werden 86 Teile 17-Äthinyl-19-nortestosteron in 1500 Teilen Dioxan zugesetzt, und das Gemisch wird hydriert, bis 2 Mol Wasserstoff absorbiert sind'. Der Katalysator wird dann abfiltriert und das Lö sungsmittel unter Vakuum verdampft.
Der kristalline Rückstand wird in 2700 Teilen Benzol gelöst und dann auf eine chromatische Kolonne gebracht, die 5000 Teile Kieselerdegel enthält. Die Kolonne wird mit 2700 Teilen Benzol, 4500 Teilen einer 10prozentigen Lö sung von Äthylacetat in Benzol und 27 000 Teilen einer 20prozentigen Lösung von Äthylacetat in Benzol gewaschen und dann mit 30000 Teilen einer 30prozentigen Lösung von Äthylacetat in Benzol eluiert. Das erhaltene Eluat wird unter Va kuum konzentriert und der Rückstand aus Methanol umkristallisiert
und bei 75 C bis zur Erreichung eines konstanten Gewichts getrocknet. Das so er haltene 17-Äthyl-19-nortestosteron schmilzt bei etwa 140 bis 141 C. Sein Ultraviolettabsorptions- spektrum zeigt ein Maximum bei 240 my mit einem molekularen Extinktionskoeffizienten von 16 500. <I>Beispiel 2</I> Eine Spur Jod wird einem Gemisch von 1,7 Teilen Magnesium und 18 Teilen Äther zugesetzt.
Der gesamte Äther wird abdestilliert und das Magne sium mit einer frischen Menge von 70 Teilen Äther bedeckt. Eine Lösung von 9 Teilen Allylbromid in 70 Teilen Äther wird dem gerührten Reaktions gemisch langsam zugesetzt und darauf eine Lösung von 2 Teilen 3 Methoxy-2,5(10)-östradien-17-on in 70 Teilen Äther allmählich zugegeben. Das Reak- tionsgemisch wird eine weitere Stunde am Rückfluss- kühler erhitzt und dann eine Lösung von 10 Teilen Natrium-Kaliumtartrat in 100 Teilen Wasser unter gutem Rühren vorsichtig zugesetzt.
Die ätherische Schicht wird abgetrennt, mit Wasser gewaschen, über Natriumsulfat getrocknet und filtriert. Der durch Konzentration des Filtrats erhaltene Rück stand wird aus einem Gemisch von Äther und Me thanol umkristallisiert, wobei ein bei etwa 88 bis 96 C schmelzendes Produkt erhalten wird, das vor wiegend aus 17a-Allyl-3-methoxy-2,5(10)-östradien- 17-ol besteht. Ein Gemisch aus 1,5 Teilen dieses Produktes, 32 Teilen Methanol, 5 Teilen Wasser und 1,8 Teilen konzentrierter Salzsäure wird 5 Mi nuten lang unter Rückfluss erhitzt.
Das durch Ver dünnung des Reaktionsgemisches mit Wasser erhal tene gummiartige Produkt wird mehrere Male mit Wasser durch Dekantierung gewaschen und ge trocknet. Es wird durch Kristallisieren aus einem Gemisch von Aceton und Petroleumäther oder durch chromatographisches Fraktionieren auf Kieselerde gel gereinigt. Auf diese Weise wird 17-Allyl-19- nortestosteron erhalten, das bei etwa 95,5 bis 96,5 C schmilzt.
Ein Gemisch aus 5 Teilen 17-Allyl-19-nortesto- steron, 100 Teilen Dioxan und 1,5 Teilen eines aus 54/o Palladium auf Holzkohle bestehenden Kataly- sators wird in Kontakt mit einer Wasserstoffatmo sphäre geschüttelt, bis etwa 1 Mol-Äquivalent Wasserstoff verbraucht worden ist.
Der Katalysator wird abfiltriert und das Filtrat unter reduziertem Druck konzentriert. Wenn der durch Verdampfung des Lösungsmittels erhaltene Rückstand wiederholt aus einem Gemisch von Aceton und Petroläther umkristallisiert wird, erhält man 17-Propyl-19-nor- testosteron, das bei etwa 120-122 C schmilzt.
<I>Beispiel 3</I> Ein Gemisch aus 53,7 Teilen 17-Äthinyl-19-nor- testosteron, 1500 Teilen Dioxan, 1000 Teilen Pyri- din und 30 Teilen eines aus 501o Palladium auf Calciumcarbonat bestehenden Katalysators wird in Kontakt mit einer Wasserstoffatmosphäre geschüttelt, bis etwa 1 Mol-Äquivalent Wasserstoff verbraucht worden ist.
An diesem Punkt wird die Hydrierung unterbrochen und der Katalysator abfiltriert. Das Filtrat wird unter Vakuum auf etwa 500 Teile kon zentriert, mit 3000 Teilen Äther verdünnt und mit normaler Salzsäure gewaschen, bis eine Probe mit Kongorot eine saure Reaktion ergibt. Die Lösung wird nacheinander mit Wasser, einer 5prozentigen Natriumbicarbonatlösung, wiederum mit Wasser und mit gesättigter Natriumchloridlösung gewaschen. Die ätherische Phase wird über Natriumsulfat getrock net, auf einem Dampfbad auf etwa 500 Teile kon zentriert und mit 800 Teilen Petroläther verdünnt.
Nach 16stündigem Stehen bei 0 C wird das Pro dukt auf einem Filter gesammelt, getrocknet und aus einem Gemisch von Äthylacetat und Petroläther umkristallisiert. Auf diese Weise wird 17-Vinyl-19- nortestosteron gewonnen, das bei etwa 159 bis 171 C schmilzt.
Ein Gemisch aus 10 Teilen 17-Vinyl-19-nortesto- steron, 80 Teilen Dioxan und 3 Teilen eines aus 511/o Palladium auf Holzkohle bestehenden Katalysators wird in einer Wasserstoffatmosphäre geschüttelt, bis etwa 1 Mol-Äquivalent Wasserstoff verbraucht wor den ist. Der Katalysator wird abfiltriert und das Filtrat unter reduziertem Druck zur Trockne kon- zentiert. Durch Umkristallisieren des Rückstandes aus Methanol erhält man 17 Äthyl-19-nortestosteron, das bei etwa 140 bis 14111 C schmilzt.
