Es wurde nun gefunden, dass Vitamin A A.cetat dadurch erhalten werden kann, da.ss man 3-Oxy-3-methyl-hexen-(1)-in-(5) mit 2 Mol Alkylmagnesiumbromid umsetzt, die erhaltene Grignard-Verbindung mit f-Ionon kondensiert, das Kondensationsprodukt unter schonenden Bedingungen hydrolysiert, an die Dreifa.chbindung des erhaltenen Reaktions produktes 1 Hol Wasserstoff anlagert, das so gebildete 3,7-Dioxy-3,7-dimethyl-9-(2',6',
6'- trimethyl - cyclohexen - [1'] - y1) - nonatrien- (1,5,8) durch Behandeln mit mindestens 22 Mol eines Acetylierungsmittels und Erwär men diacetyliert und der Ally lumlagerung unterwirft und Essigsäure aus 4,5-Stellung abspaltet.
Die Synthese dürfte nach folgendem Formelschema verlaufen:

Die Synthese des Vitamin A-Acetats ge lingt erfindungsgemäss durch Zusammen fugen von zwei Teilstücken I und II mit 13 re_p. 7 Kohlenstoffatomen zu einem Konden sationsprodukt III mit 20 Kohlenstoffato- n.en. Dieses Kondensationsprodukt enthält eine Dreifachbindung und drei Doppelbin dungen, von denen nur zwei zueinander kon jugiert sind.
Daraus wird das empfindliche S3-stein von den 5 zueinander konjugierten Doppelbindungen des Vitamin A erst in der letzten Reaktionsstufe nach vorangehender partieller Hydrierung der Dreifachbindung dureh zwei Ally lumlagerungen und durch Abspaltung von 1 Mol Essigsäure gebildet.
Das als Ausgangsmaterial verwendete 3- Oxy-3-methyl-hexen-(1)-in-(7), das mit Ionon kondensiert wird, war bisher noch nicht bekannt. Seine Darstellung gelingt bei spielsweise aus, Propargylbromid und IVlethyl- vinyllieton:
1 Mol Propa_rgylbromid und 2 bis 4 Mol trockenes l@lethylvinylketon werden in benzo- lischer Lösung unter den üblichen Bedingun gen der Reformatsky-Reaktion in der Siede hitze mit 1 bis 3 11o1 Zinkspänen konden siert. Das entstandene Zinksalz dieses Kon densationsproduktes wird mit Säuren zer setzt und das Lösungsmittel abgedampft..
Aus dem viskosen Rückstand kann das 3 Oxy-3-methyl-hexen-(1)-in-(5) in guter Aus beute herausdestilliert werden.

Die eiste Stufe des vorliegenden Verfah rens ist eine Grignard-Reaktion, bei welcher 3-Oxy-3-methyl-hexen-(1)-in-(5) zuerst mit 2 Äquivalenten Alkylmagnesiumhalogenid (z. B. Äthylmagnesiumbromid) umgesetzt wird. Die entstehende Di-3lagnesiumbromid- verbindung wird dann mit ss-Ionon konden siert. Die Umsetzung gelingt in den gebräuch lichen Lösungsmitteln, wie beispielsweise Äthyläther. Das Reaktionsprodukt wird un ter schonenden Bedingungen hydrolysiert. z.
B. mit: einer Ammoniumsalzlösung in der Kälte. Dabei werden die Reaktionsbedingun gen zweckmässig so gewählt, da.ss keine uner wünschte Wasserabspaltung erfolgt. Unver ändertes 3-Oxy-3-methyl-hexen-(1)-in-(5) lässt sich im Vakuum abdampfen. Das zu rückbleibende Rohprodukt kann direkt wei terverwendet werden. Zur Darstellung des reinen 3,7-Dioxy-3,7-dimethyl-4-(2',6',6'-tri- methyl-eyclohexen-.[l']-yl)-nona.dien- (1,8)- in-(5) wird das nicht umgesetzte ss-Ionon vor teilhaft entweder in Form eines kristallisie renden Derivates (z.
B. Semicarbazon) oder mittels eines Durchlaufchromatogrammes, z. B. durch eine wenig aktivierte Aluminium oxydsäule, abgetrennt. Das so gewonnene Kondensationsprodukt III, ein gelbes 01, zeigt im Ultraviolettspektrum keine oder nur sehr geringe Absorption über 260 mp.. Bei der Zerewitinoff-Bestimmung ergeben sich Werte für 2 aktive Wasserstoffatome.
3,7 - Dioxy - 3,7 - dimethyl"- 9"- (2',6',6'-tri- methyl-cyclohexen- [l'] -y1) -nona.dien-(1,8) -in- (5) wird nun der partiellen Hydrierung der Dreifachbindung unterworfen.
Auch in die ser Stufe wird man darauf achten, dass keine unerwünschte'Vasserabspaltung eintritt. Für die selektive Aufnahme von 1 Mol Wasser stoff hat sich die katalytische Hydrierung besonders bewährt. Als Katalysatoren eignen sich beispielsweise Pa,lladium-Caleiumcarbo- nat, Palladium-Bariumsulfat und Palladium kohle, an die vor Gebrauch Chinolin adsor- biert wird.
Wird das rohe Kondensationspro dukt der partiellen Hydrierung unterworfen, so wird mit einer dem ss-Ionongehalte entspre chenden, geringeren Wasserstoffmenge hy driert. Nach Aufnahme der berechneten lenge Wasserstoff wird die Wasserstoff <B>T</B> zufuhr unterbrochen. Das hydrierte Produkt, <B>3,7</B> - Dioxy-3, 7;-dimethyl-9- (2', 6',6'-trimethyl- cyclohexen-[l']-yl)-nonatrien-(1,5,8), das nicht unbedingt gereinigt werden muss, ist ein gelbliches, viskoses 01 vom nD = 1,503.
Es zeigt im Ultraviolettabsorptionsspektruni keine oder fast keine Absorption über 260 mu. Bei der Zerewitinoff-Bestimmung ergeben sich Werte für 2 aktive Wasserstoffatome. Das durch partielle Hydrierung gewon nene Kondensationsprodukt wird nun durch Behandeln mit mindestens 2 Mol eines Ace- tylierungsmittels diacetyliert und der Allyl- umlagerung
(Hückel, Theoretische Grund lagen der Organischen Chemie, 4. Aufl., Bd. I, S: 297/98 ff., Leipzig 1943) unterwor fen; feiner wird Essigsäure aus 4,5-Stellung abgespalten.
Dabei treten an einem Molekül zwei Allylumlagerungen ein. Die dreistän dige Hydroxylgruppe der Verbindung IV wandert unter gleichzeitiger Substituierung in die 1-Stellung und die benachbarte Dop pelbindung aus der 1- in die 2-Stellung. Zu dem wandert in entsprechender Weise die siebenständige Hydroxylgruppe in die 5-Stel- lung und die Doppelbindung von der 5- in die 6-Stellung. Man arbeitet zweckmässig der art,
dass möglichst wenig unerwünschte Ne benreaktionen (Polymerisation, Cyclssierung und dergleichen) eintreten; besonders zweck mässig ist es, darauf zu achten, dass eine un erwünschte Säureabspaltung vorgängig der Umlagerung möglichst vermieden wird. Zu diesem Zwecke wird zum Beispiel das- 3,7- Dioxy -<B>3,7</B> - dim:
ethyl -9-(2', 6', 6'-trimethyl-ey- clohexen-[1'1-yl)-nonatrien-(1,5,8) in Gegen wart eines Verdünnungsmittels mit dem Ace- tylierungsmittel behandelt.
Als Acetylie- rungsmittel eignet sich sowohl Acetanhydrid als auch Acetylehlorid. Das Zwischenprodukt dürfte der Formel V entsprechen. Seine Iso lierung ist nicht notwendig:
Durch die Essigsäureabspaltung erfolgt die Einführung einer weiteren Doppelbin dung, und zwar in der Weise, dass die in die 5-Stellung gewanderte veresterte Hydroxyl- gruppe mit einem Wasserstoffatom des be nachbarten vierständigen Kohlen.stoffatoms als Essigsäure austritt. Je nach den Bedin gungen bei der Allylumlagerung kann die Essigsäureabspaltung in der gleichen Reak
tionsstufe vor sich gehen wie die Umlage rung (z. B. bei Umlagerung bei hoher Tem peratur), oder die Abspaltung kann einen gesonderten Verfahrensschritt darstellen. So bildet sieh beim Kochen der Verbindung IV mit Essigsäureanhydrid, zweckmässig in Ge- genwart von Natriumacetat, unmittelbar die gesuchte Pentaenverbindung VI. Wird die Allylumlagerung zwar bei höherer, jedoch unterhalb der Siedetemperatur durchgeführt, z.
B. durch Einwirken von Acetylierungs- mitteln unter 100 C, so wird aus dem gebil deten Produkt der Formel V anschliessend. z. B. durch Erhitzen, zweckmässig in Gegen wart von basischen Mitteln, wie organischen Basen und basische Salze, Essigsäure abge spalten. Hiezu eignet sieh beispielsweise das Erhitzen in Chinolin auf 160 bis 170 C.
Nach dem vorstehend erläuterten Verfah ren erhält man synthetisches Vitamin A-Ace- tat, das in seinen biologischen Eigenschaften dem Vitamin A und seinen Estern. aus na türlichen Quellen entspricht.
Das Verfah rensprodukt ist besonders gekennzeichnet durch das imUltraviolettabsorptionsspektrurn bei 325 bis 328 mu auftretende Absorptions- maximum. Der erhaltene Ester kann in übli cher Weise zum Vitamin A-Alkohol verseift werden.
Das rohe synthetische Vitamin A-Acetat kann nach den gleichen Methoden gereinigt werden wie aus natürlichen Quellen gewon nene Hochkonzentrate von Vitamin A und Vitamin A-Estern. (Trennung zwischen Lö sungsmitteln, fraktionierte Fällung aus alko holischer Lösung, schonende Destillation, chromatographische Adsorption und Kristal lisation des freien Alkohols bzw. geeigneter Derivate.) Das Produkt wird zweckmässig wie das bekannte, aus Vitamin A natürlicher Her kunft gewonnene Acetat, vor dem zerstören den Einfluss von Licht, Luft und Hitze ge schützt.
Es empfiehlt sich die Zugabe von Antioxydantien, welche auch während des gesamten Ablaufes der Synthese anwesend sein können; Tocopherole sind hiezu beson ders gedignet. Beispiel <I>1:</I> 22 Gewichtsteile 3-Oxy-3-methyl-hexen- (1)-in-(5) in 100 Raumteilen Äther werden im Verlauf von 1/2 Stunde in Stickstoff- atmospliäre bei 0 unter Rühren zu einer =ltli -;
lniagnesiumbromidlösung zugefügt, die ca 9,7 Gewichtsteilen llagnesiumspänen und 2>3 Ge -,viehtsteilen Athy lbromid in 140 Raurn- teilen Äther dargestellt wurde. Anschliessend k:,@ht man 4 Stunden unter Rückfloss.
Dabei bilden sich zwei Schichten, wobei die untere, (pinkle Schicht die Di-llagnesiumbroinidver- bindung des 3-Oxy-3-methyl-hexenins ent hält.
Unter Rühren und Kühlung auf 0 fügt man nun im Verlaufe von 1 Stunde eine Lö- -ung von 36,4 Gewichtsteilen /3-Ionon in 100 Raumteilen Äther zu und vervollständigt die Kondensation durch zweistündiges Rühren bei Raumtemperatur. Das gebildete Magne- siumsalz wird durch Aufgiessen auf 200 Ge- @;
ieli tsteile Eia und 40 Gewichtsteile Ammo- niumchlorid zersetzt. Man wäscht die Äther lösung reit verdünnter Essigsäure, Natrium- bicarbonatlösung und Wasser, trocknet mit Natriumsillfat und verdampft das Lösungs mittel sowie unverändertes 3-Oxy-3-methyl- licxenin-(5), wobei man zuletzt im Wa,sser- strahlvakuum auf 60 C erhitzt.
Der Rück- ,tand besteht aus 3, 7-Dioxy-3,7-dimethyl-9- ?', 6', 6' - trimethyl - cy clohexen -. [ 1'@ -. y1) -nona- dien-(1,8)-in-(5) und etwas unverändertem /3-Ionon. Ausbeute: 52,4 Gewichtsteile oder 91,5 ö der Theorie.
Analysenreines Kondensationsprodukt er hält man durch chromatographische Adsorp- tion an die 20fache Menge Aluminiumoxyd, das mit 12 % Wasser desahtiviert wurde. Da bei wird das gesuchte Kondensationsprodukt ;in das Aluminiumoxyd adsorbiert, während (las J.,-Ionon und eventuell durch Wasser abspaltung entstandene Verunreinigungen finit Petrolüther ausgewaschen werden kön nen.
Das so gereinigte Produkt ist ein gelb liches, vi3koses. <B>01,</B> das keine. Ultraviolett strahlen von grösserer Wellenlänge als 2<B>1<I>0,</I></B> mu a bsorbiert. 10 Gewichtsteile reines 3,7-Dioxy-3, 7-di- niethyl - 9 - (9',6',6'-trimethyl-cyclohexen- [1'] - s i-1) -nonadien- (1,8) -in-(5)
werden in 150 R < ..uinteilen Methylalkohol gelöst und unter Verwendung von 0,2 Gewichtsteilen 4 % igem Palladium-Calciumcarbonat-gatalysator bei Raumtemperatur hydriert, wobei man bei der Hydrierung zweckmässig 0,05 Gewichtsteile Tocopherol als Antioxydans zusetzt.
Nach Aufnahme von 850 Raumteilen W.aeserstoff (740 mm Hg, 20 ) wird die Hydrierung un terbrochen, der Katalysator abgenutscht und das Filtrat im Vakuum eingeengt. Man er hält in 95%iger Ausbeute reines 3,7-Dioxy- 3,7-dimethyl-9 -(2',6',6'-trimethyl-cyclohexen- [1']-yl)nonatrien-(1,5,8) als gelbliches Öl, das keine Ultraviolettstrahlen von grösserer Wellenlänge als 270 mu absorbiert.
10 Gewichtsteile dieses Produktes der partiellen Hydrierung werden in 50 Raum teilen Essigsäureanhydrid gelöst, mit 5 Ge wichtsteilen wasserfreiem Natriumacetat ver setzt und unter Lichtausschluss 2 Stunden unter Rückfloss erhitzt. Darauf wird das überschüssige Essigsäureanhydrid und der entstandene Eisessig im Vakuum einge dampft und der Rückstand in Petroläther vom .Siedepunkt 30 bis. 60 C und Wasser aufgenommen.
Die Petrolätherlösung wird mit Natriumbica.rbonatlösung und Wasser gewaschen, mit Natriumsulfat getrocknet und eingedampft. Das verbleibende braune Öl wird im Molekularvakuum fraktioniert. Da bei erhält man neben einem geringen Vorlauf 6 bis 8 Gewichtsteile eines gelben Öls vom Siedepunkt 80 bis 90 C (10-5 mm Hg), das im Ultraviolettspektrum ein ausgeprägtes Absorptionsmaximum bei 325 bis 328 m,u be sitzt und im Wachstumstest an der Vita min A-Mangelratte hochwirksam ist.
Das so erhaltene Vitamin A-Acetat gibt bei der Elementaranalyse Werte, die auf die Formel C=2H3202 stimmen. Die Verbindung enthält aber isomere Beimengungen, die im Ultraviolettabsorptionsspektrum bei 310 m,cc ein Absorptionsmaximum besitzen.
Die Ab trennung dieser isomeren Verbindungen er folgt durch die üblichen chromatographi- schen Methoden und durch fraktioniertes Fällen resp. Kristallisieren sowohl des durch Verseifen gewonnenen Vitamin A-Alkohols als auch seiner kristallisierenden Derivate.
<I>Beispiel 2:</I> 10 Gewichtsteile von ungereinigtem 3,7 Dioxy; ..3,7 - dimethyl-9 -(2',6',6'-trimethyl-ey- clohexen-[1']-yl)-nonadien-(1,8)-in-(5) (dar gestellt nach Beispiel 1) werden in 100 Ge wichtsteilen Methylalkohol gelöst und unter Verwendung von 0,2 Gewiehtsteilen 4%igem Palladium - Bariumsulfat - Katalysator bei Raumtemperatur hydriert.
Nach Aufnahme von 800 Raumteilen Wasserstoff (740 mm Hg, 20 ) wird die Hydrierung unterbrochen und die Hydrierlösung wie im Beispiel 1 aufge arbeitet. Die Ausbeute beträgt 90 bis 95 %. Das so gewonnene 3,7-Dioxy-3,7-dimethyl- 9-(2', 6', 6'-trimethyl-cyclohexen- [1'] -y1) -nona- trien-(1,5,8) wird nach den Angaben von Bei spiel 1 weiterbehandelt.
Beim Fraktionieren im Molekularvakuum erhält man dabei 3 bis 4 Gewichtsteile Vorlauf vom Siedepunkt 30 bis 70 C (10-5 mm Hg) und 4 bis 6 Ge- wichtsteile reines Acetat vom Siedepunkt 80 bis 90 C (10-5 mm Rg), das dem Produkt aus Beispiel 1 entspricht.
<I>Beispiel 3:</I> 1 Gewichtsteil 3,7-Dioxy-3,7-dimethyl-9- (2',6',6'-trimethyl-cyclahexen- [1']-yl)-nona- trien-(1,5,8), dargestellt nach Beispiel 1, wird in 4 Gewichtsteilen Pyridin und 2 Ge wichtsteilen Acetylchlorid in Gegenwart von 15 Raumteilen Dichloräthän als Verdün- nungsmittel gelöst und während 60 Stunden bei 25 C aufbewahrt:
Die Reaktionslösung wird in Äther aufgenommen, mit wässeriger Natriumbicarbonatlösung, verdünnter Schwe felsäure und Wasser gewaschen, mit Na- triumbicarbonat getrocknet, eingedampft und wie in Beispiel 1 aufgearbeitet.
Das Verfah rensprodukt zeigt im Ultraviolettspektrum Absorptionsmaxima bei 310 und 325 mu und gibt bei der Carr-Price-Reaktion die für Vitamin. A-Acetat .spezifischen Absorptions maxima bei 620 und 580 mcc.
Die Allylumlagerung und Wasserabspal tung gelingt in gleicher Weise beim Erwär men mit Acetylchlorid und Eisessig, wobei Äther oder Benzol als Verdünnungsmittel zugesetzt werden.
