Chemistry and Indus.try, 58 (l.939), S. 802.] Diese letzten zwei Synthesen wurden durch keine biolo gischen Prüfungsresultate gestützt und ein dazu veröffentlichtes Ultraviolettabsorptions- spektrum zeigt kein Absorptionsmaximum bei dem für Vitamin-A-Äther charakte ristischen Wert von 328 m,ct [Journal of the American Chemica.l Society, 67 (1945), Seite 1627e.
Wie nun gefunden wurde, kann Vitamin- A-Methyläther dadurch erhalten werden, dass man ss-Jonon mit 1-Methoxy-3-methyl'-6- halogen-hexen-(2)-in-(4) mittels Zink kon densiert, das zinkhaltige Kondensationspro dukt zersetzt und das entstandene 1-Methoxy- 3, 7-dimethyl -7- oxy-9-trimethylcyclohexenyl- nonadien-(2,8)-in-(4)
an der Dreifachbindung paxtiell hydriert, im erhaltenen Tetraen die Hydroxylgruppe durch einen Säurerest er setzt und die erhaltene Verbindung durch Säureabspaltung in den Vitamin-A-Methyl- äther überführt.
Die Synthese verläuft nach folgendem Formelschema

Es handelt sich somit um die Einwirkung von ss-Jonon (I) mit 13 Kohlenstoffatomen auf den Methyläther eines substituierten Propargylhalogenids (II) mit 8 Kohlenstoff atomen in Gegenwart von. Zink.
Durch Zer setzung des zinkhaltigen Kondensations produktes erhält man eine Verbindung (III) mit 21 Kohlenstoffatomen. Dieses:
Produkt enthält eine Dreifachbindung und drei Dop pelbindungen, von denen je nur zwei zu einander konjugiert sind. Daraus wird das System der fünf zueinander konjugierten Dop pelbindungen des Vitamin-A-Methyläthers durch partielle Hydrierung der Dreifach bindung, Ersatz der Hydroxyl!gruppe durch einen Säurerest und Säureabspaltung zwecks Einfügung einer weiteren Doppelbindung gebildet.
Das zur Kondensation mi ss-Jonon ver wendete 1-Methoxy - 3 - methyl - 6 - halogen- hexen-(2)-in-K4) isst noch nicht bekannt.
Seine Darstellung gelingt aus 3-Oxy-3-methyl- penten - (1) - in - (4) auf folgendem Wege: 3-Oxy-3-methyl-penten-(1)-in-(4) wird mit Phosphortribromid halogeniert und das ent stehende primäre Bromid mit Alkalimethyl'at veräthert. Aus 1-Methoxy-3-methyl-penLen- (2) -in- (4) (Emil Barell-Festschrift 1946, Seite 40)
und 33prozentigem wässerigem Formaldehyd gewinnt man in Analogie. z. Schweiz. Patent Nr. 22$202 in Gegenwart von Kupferkatalysatoren bei 120 C im Druckgefäss in 70proz. Ausbeute 1-Methoxy- 3-methyl-6-oxy-hexen-(2)-in-(4) vom Sdp. 127-129 C/15 mm;
n\ = 1,496. 1-Meth- oxy-3-methyl-6-oxy-hexen-(2)-in-(4) wird, zweckmässig unmittelbar vor der Zinkkonden sation, in Ätherlösung, durch Einwirkung von Phosphortrihal'ogenid in Gegenwart von etwas Pyridin in 1-Methoxy-3-methyl-6- halogen-hexen-(2)-in-(4) übergeführt. Es ist eine leicht bewegliche Flüssigkeit, welche die Schleimhäute reizt. Das 1-Methoxy-3-methyl- (j-broin-hexen-(?)-in-(4) ist ein wasserklares t)1 vom nv - 1,524. .
Die erste Stufe des vorliegenden Ver fahrens ist eine Zinkkondensation, bei wel- elier 1-Methoxy-3-methyl-6-halbgen-hexen- (?)-in-(4) und ss-Jonon ähnlich einer Refor- ma,tskischen Synthese miteinander verbunden erden.
Die Umsetzung gelingt in einem inerten Lösungsmittel, wobei trockener .@.thylä.th.er besonders vorteilhaft ist. Zweck- mässig wendet man einen Überschuss des Halogenids an und lässt die Lösung der R.e""a.ILtionskomponenten unter kräftigem Rüh ren zum vorgewärmten Kondensationsmittel, z. B. Zinkgriess, zufliessen. Das entstehende zinkhaltige Kondensationsprodukt kann in g;ebrttuchlieher Weise, z.
B. mit Iprozentiger Schwefelsäure, hydrolysiert werden. Nicht umgesetztes ss-Jonon kann in Form :eines kri- sta.llisierenden Derivates (z. B. als Semi- ea.rbazon) abgetrennt werden.
Die, Reinigung des gebildeten 1-1VIethoxy-3,7-dimethyl-7- oxy - 9 -trimietrhylcyclohexenyl-nonadien-(2,8)- in-(4) gelingt darauf durch Trennen zwi- #chen Lösungsmitteln, wie leicht siedendem Petrol-ä.ther und wässerigem Methylalkohol. Unter geeigneten Bedingungen ist die Va- hianmdest.illation vorteilhaft.
Das gereinigte Kondensationsprodukt ist ein viskoses, gelbes C)1, das keine Ultra- --iolc,ttstr,ihlen von grösserer Wellenlänge als ?60 m,y absorbiert. Es gibt bei der Zerewiti- iioffbes@immung Werte für 1 aktives Wasser stoffatom und spaltet beim Erhitzen leicht Wasser ab.
Das Kondensationsprodukt III wird nun, zu-Peltinässi.g nach Reinigung, der partiellen Hydrierung der Dreifachbindung unterwor fen. Hierzu sind die bei Polyenverbindungen gebräuchlichen Mittel, wie Behandeln mit Zink und Eisessig, mit Natriumamalgam und einem basischen Mittel oder mit Natrium in flüssigem Ammoniak anwendbar.
Besonders bewährt hat sich die katalytische Hydrierung unter Verwendung von Palladium-Katalysa, goren. Bei der katalytischen Teilhydrierung wird die Wasserstoffzufuhr nach Aufnahme der berechneten Wasserstoffmenge unterbro chen. Das hydrierte Produkt [1-Methoxy-3,7- dimethyl - 7 - oxy - 9 - trimethylcyclohexenyl- nonatrien-(2,4,8)1 muss nicht. isoliert und ge reinigt werden. Es ist ein viskoses, gelbliches G1.
Die Einführung einer weiteren Doppel Bindung in Verbindung IV wird dadurch be werkstelligt, da.ss man die Hydroxylgruppe durch einen Säurerest ersetzt und an schliessend Säure abspaltet. So kann man die Verbindung IV mit einem organischen Acylierungsmittel verestern und aus dem ge bildeten Ester durch Erhitzen, zweckmässig in Gegenwart von alkalischen .Mitteln, Säure abspalten.
Ferner kann man auch die Hy. droxylgruppe unter schonenden Bedingungen durch Halogen ersetzen und darauf durch Be- handeln mit alkalischen Mitteln serstoff abspalten. Hierzu eignet sich beispielsweise die Umsetzung mit Phosphor- trichlorid in Gegenwart von Pyridin und an schliessendes Erwärmen mit methylalkoho- lischem Alkalihydroxyd auf 60-70 C.
Man kann auch das Tetraen in Gegenwart von Kaliumhydroxyd mit Schwefelkohlenstoff umsetzen und den erhaltenen Xanthogen- säureester mit einer ätherischen Lösung von M ethyliodid kochen.
Das bekannte Verfahrensprodukt (Vit- amin-A-Methyläther) kann nach den. gleichen Methoden -erei.nigt werden wie aus natür lichen Materialien gewonnene, Hochkonzen trate von Vitamin A und seinen Derivaten (Trennen zwischen Lösungsmitteln, chromato- graphische Adsorption, schonende Destilla tion usw.). Das Produkt ist, genau wie na türliches Vitamin A, vor dem zerstörenden Einfluss von Licht, Luft und Hitze zu ,schützen. Es: empfiehlt sich die Zugabe von Antioxydantien, welche auch während des gesamten Ablaufes der Synthese anwesend sein können;
als Antioxydantien eignen sich besonders Tocopherole.
Der Vitamin-A-Dlethyläther soll als Arzneimittel verwendet werden. <I>Beispiel</I> .T Eine Lösung von 5,6 Gewichtsteilen 1-Methoxy-3-methyl-6-b.rom-hexen-(2)-in-(4)
und 3 Gewichtsteilen ss-Jonon in 20 Raum teilen Äther wird unter kräftigem Rühren im Verlaufe einer halben. Stunde in Stick- stoffatmosphäre zu 4 Gewichtsteilen vorge- wärmtem Zinkgriess zulaufen gelassen,
wo bei das Lösungsmittel durch die Reaktions wärme fortlaufend unter Rückfluss sieden mini. Nach dem Abklingen der Reaktion wird noch kurz unter Rückfluss erhitzt, dann abgekühlt und mit Eis und verdünnter Schwefelsäure zersetzt. Man nimmt in Äther auf, wäscht mit lproz. Schwefelsäure und Wasser,
trocknet und verdampft das Lö- sungsmittel. Der Rückstand wird in Methyl alkohol gelöst, mit Semicarbazid versetzt und 12 Stunden stehen gelassen. Man filtriert, fügt die doppelte Menge Wasser zu, nimmt in Petroläther vom Sdp. 30-70 C auf und schüttelt .die Petrolätherlösung mehrmals mit 90proz. Methylalkohol aus.
Aus dem Meta..nol- auszug gewinnt man 3-3,5 Gewiehtsteile 1 Methoxy-3,7 - dimethyl'- 7 -oxy-9-trimethyl- cyclohexenyl - nonadien -<B>(2,8)</B> - in - (4) als viskoses, gelbliche:. Öl vom n" = 1,515. Ausbeute, auf ss-Jonon bezogen, 65-70%.
10 Gewichtsteile -dieses Kondensations- produktes werden in 100 Raumteilen Methyl alkohol gelöst und unter Verwendung von 0,5 Gewichtsteilen 4proz. Pal'ladium-Calcium- carbonat-gatalysator bei Raumtemperatur hydriert. Nach Aufnahme von 1 Mol Wasserstoff wird die Hydrierung unterbro chen und der Katalysator abgetrennt.
Das Filtrat wird zur Entfernung von etwas Wasserabspaltungsprodukt mit 6 Gewichts teilen Wasser versetzt und mit wenig Petrol- äther vom Sdp. 30-60 C ausgeschüttelt. Die Methylalkohollösung wird darauf mit 200 Teilen Wasser verdünnt, mit Petroläther ausgeschüttelt, und die Petrolätherauszüge werden getrocknet und eingeengt.
Dabei ge winnt man 8-9 Gewichtsteile 1-Methoxy- 3,7 -.dimethyl-7-oxy-9-trimethylcyclohexenyl- nonatrien-(2,4,8) als gelbliches, viskoses Öl vom n" = 1,512. Die Zerewitinoffbestim- müng zeigt 1 aktives urasserstoff an.
Die Ultmviolettabsorptionsmessung gibt keine Absorption über 260 mu. Beim Verfüttern der Verbindung an Vitamin-A-Mangel'ratten zeigt sich eine deutliche Wachstumswirkung.
4 Gewichtsteile des Produktes der Teil- hydrierung in 50 Raumteilen Petroläther vom Sdp. 30-60 C werden in Gegenwart von 1 Raumteil Pyridin im Verlaufe einer Stunde mit einer Lösung von 0,7 Gewichts teilen Phosphortribromid in 30 Raumteilen Petroläther vom Sdp. 30-60 C versetzt.
Man rührt nach der Zugabe eine Stunde bei 0 C, giesst die Reaktionslösung von einer geringen braunen Ausscheidung ab, versetzt mit Eis, wäscht mit Wasser, trocknet mit Natriumsulfat und verdampft das Lösungs mittel. Der Rückstand wird in 40 Raum- teilen Pyridin gelöst und mit 2 Gewichts- teilen pulverisiertem galiumhydroxyd eine Stunde unter Rühren auf 60-70 C erhitzt.
Nach dem Erkalten fügt man Petroläther vom Sdp. 30-60 C zu, wäscht mit 90proz. Methylalkohol und Wasser, trocknet mit Na triumsulfat und verdampft das Lösungsmit- tel, zuletzt unter Anwendung von Vakuum.
Die Reinigung des Vitamin-A-Methyl- äthers gelingt im Durchl'aufchromatogramm durch Säulen mit wenig aktiviertem Alumi niumoxyd mittels Petroläther vom Sdp. 60 bis 80 C.
Dabei haften Zwisehenprodukte mit einer freien Oxygruppe am Aluminium oxyd, während Nebenprodukte, die bei 280 bis 310 m,u ultraviolettes Licht absorbieren, weniger adsorbiert werden und sich in den Vorläufen anreichern.
Der Vitamin-A- Methyläther selbst wandert langsam durch die Säule, wobei seine: Lage durch die inten siv gelbgrüne Fluoreszenz im Ultraviolett licht festgestellt werden kann. -Die gute Fraktion ist gekennzeichnet durch die Be ständigkeit der Carr-Price-Farbreaktion. Man verdampft den Petroläther und destilliert den so gereinigten Vitamin-A-Methyläther im Molekularvakuum. Gelbes Ö'1 vom Sdp. 90 bis 95 C/10-5 mm $g mit dem charakteristi schen <RTI

iet (315-335 nlp.) zeigst; und im Wachstumstest an der Vita.min- A-llangelrat.te hoch wirksam isst.

As has now been found, vitamin A methyl ether can be obtained by condensing ss-ionone with 1-methoxy-3-methyl'-6-halo-hexen-(2) -yne- (4) using zinc, the zinc-containing condensation product decomposes and the resulting 1-methoxy-3, 7-dimethyl-7-oxy-9-trimethylcyclohexenyl nonadiene- (2,8) -in- (4)
paxtiell hydrogenated at the triple bond, the hydroxyl group in the tetraene obtained is replaced by an acid residue and the compound obtained is converted into vitamin A methyl ether by splitting off acid.
