Verfahren zur Herstellung von neuen Phenazinen
Die vorliegende Erfindung betrifft ein Verfahren zur Herstellung neuer Phenazine mit wertvollen chemotherapeutischen und insbesondere tuberkulostatischen Eigenschaften.
Es ist bekannt, dass das durch Oxydation von N-Phenyl-o-phenylendiamin-hydrochlorid mit Ferrichlorid erhältliche 2-Anilino-3-imino-5-phenyl-3 5- dihydro-phenazin (Anilinoaposafranin) tuberkulostatische Wirksamkeit besitzt, vgl. Nature 162, 622 bis 623 (1948).
Überraschenderweise wurde nun gefunden, dass Phenazine der Formel

worin Ar und Ar' gleiche oder verschiedene gegebenenfalls durch Halogenatome oder niedermolekulare Alkyl- oder Alkoxygruppen substituierte Phenylreste bedeuten, wobei jedoch Ar' keinen unsubstituierten Phenylrest bedeuten darf, wenn beide Ar Phenyl- oder p-Tolylreste sind, eine wesentlich stärkere tuberkulostatische Wirkung als die bisher beschriebenen Phenazinderivate ausüben. Von bekannten Tuberkulostaticis unterscheiden sie sich im Tierversuch überdies dadurch, dass sie z. B. mit Tuberkelbazillen infizierte Mäuse weit über die Behandlungsdauer hinaus am Leben zu erhalten vermögen.
Die Verbindungen der Formel I werden erfindungsgemäss hergestellt, indem man auf ein Gemisch von Brenzcatechin und einem Amin der Formel NH2-Ar II vorzugsweise im molaren Verhältnis von 1 : 2 und in homogener Phase, ein Alkalijodat einwirken lässt, und das so erhaltene Reaktionsprodukt, welches einer der beiden tautomeren Formeln

entspricht, mit einem Salz eines N-Phenyl-o-phenylendiamins der Formel

umsetzt.
Zur oxydativen Reaktion des Brenzcatechins mit einem Amin der Formel II kann man die beiden Reaktionskomponenten beispielsweise in einem mit Wasser mischbaren organischen Lösungsmittel, wie Alkohol oder Aceton, lösen und unter Rühren bei Raumtemperatur mit einer wässrigen Lösung von Natriumjodat versetzen.
Alkalijodate sind überraschenderweise weitaus die geeignetsten Oxydationsmittel und ermöglichen Ausbeuten von 90950/o, während beispielsweise Silberoxyd oder Bleioxyd als Oxydationsmittel viel weniger geeignet sind. Die Umsetzung der erhaltenen Benzochinone der Formel III bzw. IV mit einem Salz eines Diamins der Formel V kann durch gemeinsames Erwärmen auf mässig hohe Temperaturen, z. B. durch Kochen in Alkohol, erfolgen. Geeignete Ausgangsstoffe der Formel II sind z. B. Anilin, o-, m- und p-Toluidin, o-, m- und p-Anisidin, p Phenetidin, p-Isopropoxy-anilin, p-Chlor-anilin und p-Brom-anilin.
Als Ausgangsstoffe der Formel V können beispielsweise N-Phenyl-o-phenylendiamin, N-(2-Chlor-phenyl)-o-phenylendiamin, N-(3-Chlorphenyl)-, N-(4-Chlor-phenyl)-, N-(4-Brom-phenyl)-, N-(2-Methyl-phenyl)-, N-(4-Methyl-phenyl)-, N-(4 Methoxy-phenyl)-, N (4-l2ithoxy-phenyl)-oder oder N-(4 Isopropoxy-phenyl) -o-phenylendiamin verwendet werden.
In den nachfolgenden Beispielen bedeuten Teile Gewichtsteile; diese verhalten sich zu Volumteilen wie g zu cm3. Die Temperaturen sind in Celsiusgraden angegeben.
Beispiel I
2,2 Teile Brenzcatechin und 4,1 Teile p-Chloranilin werden in 100 Volumteilen Äthanol gelöst und unter Rühren mit einer Lösung von 4,0 Teilen Natriumjodat in 100 Teilen Wasser versetzt. Die Mischung wird 3 Stunden bei Raumtemperatur gerührt. Hierauf wird der glänzend rote Niederschlag abfiltriert und mit Wasser und anschliessend mit wenig Methanol gewaschen. Eine weitere Reinigung des so erhaltenen 2-Hydroxy-5-(p-chlor-anilino)-1, 4- benzochinon-4-(p-chlor-phenylimin) ist zur Weiterverarbeitung unnötig, die Ausbeute beträgt 90 bis 950/o.
Das obige Reaktionsprodukt wird zusammen mit 4,0 Teilen N-Phenylo-phenylendiamin-hydrochlorid in heissem Alkohol gelöst und 3 Stunden unter Rückfluss gekocht. Nach Alkalischstellen des Reaktionsgemisches wird das basische Kondensationsprodukt durch Versetzen mit Wasser ausgefällt, abfiltriert, mit Wasser gewaschen und getrocknet. Die weitere Reinigung erfolgt durch Chromatographieren des Rohproduktes in Benzollösung an einer Aluminiumsäule und Kristallisation aus Benzol, Benzol/Petrol äther oder Alkohol. Das so erhaltene 2-(p-Chlor anilino-3-(p-chlor - pheny1imino)-5henyl-3,5-di- hydro-phenazin schmilzt bei 241-242".
Beispiel 2
22 Teile Brenzcatechin und 37,2 Teile Anilin werden in 900 Volumteilen Alkohol gelöst und bei 20250 innerhalb 25 Minuten eine Lösung von 40 Teilen Natriumjodat in 900 Teilen Wasser unter Rühren zufliessen gelassen. Nach 6stündigem Rühren bei Raumtemperatur nutscht man das ausgefallene rote Reaktionsprodukt ab, wäscht es mit 1500 Teilen Wasser und dreimal mit je 50 Volumteilen Methanol.
Das so erhaltene 4, 5-Dianilino-1, 2-benzochinon schmilzt bei 185-190", seine Reinheit genügt für die nächste Stufe.
21,8 Teile dieses Produktes und 19,1 Teile N - (4- Chlor-phenyl)-o-phenylendiamin-hydrochlorid werden in 1600 Volumteilen heissem Alkohol suspendiert und das Gemisch 6 Stunden unter Rühren zum Rückfluss erhitzt. Dann lässt man auf 65-70" abkühlen, fügt innerhalb 3/4 Stunden 30 Teile konz.
Ammoniak zu und rührt noch eine weitere Stunde bei derselben Temperatur. Nach dem Erkalten nutscht man das rohe Kondensationsprodukt ab, wäscht es mit 500 Volumteilen Wasser, trocknet es und kristallisiert es aus einem Gemisch von Benzol und Petroläther um. Man erhält das 2-Anilino-3 phenylimino-5-(p-chjor-phenyl) -3, 5-dihydro-phenazin als dunkelrote Kristalle vom Schmelzpunkt 205 bis 206".
Process for the production of new phenazines
The present invention relates to a process for the preparation of new phenazines with valuable chemotherapeutic and in particular tuberculostatic properties.
It is known that the 2-anilino-3-imino-5-phenyl-35-dihydrophenazine (anilinoaposafranin) obtainable by oxidation of N-phenyl-o-phenylenediamine hydrochloride with ferric chloride has tuberculostatic activity, cf. Nature 162: 622-623 (1948).
Surprisingly, it has now been found that phenazines of the formula

where Ar and Ar 'mean the same or different phenyl radicals, optionally substituted by halogen atoms or low molecular weight alkyl or alkoxy groups, but Ar' must not mean an unsubstituted phenyl radical if both Ar are phenyl or p-tolyl radicals, a significantly stronger tuberculostatic effect than before exercise described phenazine derivatives. In animal experiments, they also differ from known tuberculostaticis in that they are e.g. B. are able to keep mice infected with tubercle bacilli alive well beyond the duration of the treatment.

For the oxidative reaction of catechol with an amine of the formula II, the two reaction components can be dissolved, for example, in a water-miscible organic solvent, such as alcohol or acetone, and an aqueous solution of sodium iodate can be added while stirring at room temperature.
Surprisingly, alkali iodates are by far the most suitable oxidizing agents and enable yields of 90950 / o, while silver oxide or lead oxide, for example, are much less suitable as oxidizing agents. The reaction of the benzoquinones of the formula III or IV obtained with a salt of a diamine of the formula V can be carried out by heating them together to moderately high temperatures, e.g. B. by boiling in alcohol. Suitable starting materials of the formula II are, for. B. aniline, o-, m- and p-toluidine, o-, m- and p-anisidine, p phenetidine, p-isopropoxy-aniline, p-chloro-aniline and p-bromo-aniline.
As starting materials of the formula V, for example, N-phenyl-o-phenylenediamine, N- (2-chlorophenyl) -o-phenylenediamine, N- (3-chlorophenyl) -, N- (4-chlorophenyl) -, N - (4-Bromo-phenyl) -, N- (2-methyl-phenyl) -, N- (4-methyl-phenyl) -, N- (4-methoxyphenyl) -, N (4-l2ithoxyphenyl) -or or N- (4 isopropoxyphenyl) -o-phenylenediamine can be used.
In the following examples, parts mean parts by weight; these are related to parts of volume as g to cm3. The temperatures are given in degrees Celsius.
Example I.
2.2 parts of catechol and 4.1 parts of p-chloroaniline are dissolved in 100 parts by volume of ethanol and a solution of 4.0 parts of sodium iodate in 100 parts of water is added while stirring. The mixture is stirred for 3 hours at room temperature. The shiny red precipitate is then filtered off and washed with water and then with a little methanol. Further purification of the 2-hydroxy-5- (p-chloro-anilino) -1,4-benzoquinone-4- (p-chloro-phenylimine) obtained in this way is unnecessary for further processing; the yield is 90 to 950%.
The above reaction product is dissolved together with 4.0 parts of N-phenylo-phenylenediamine hydrochloride in hot alcohol and refluxed for 3 hours. After the reaction mixture has been rendered alkaline, the basic condensation product is precipitated by adding water, filtered off, washed with water and dried. Further purification is carried out by chromatography of the crude product in benzene solution on an aluminum column and crystallization from benzene, benzene / petroleum ether or alcohol. The 2- (p-chloroanilino-3- (p-chloro-pheny1imino) -5henyl-3,5-dihydrophenazine obtained in this way melts at 241-242 ".
Example 2
22 parts of catechol and 37.2 parts of aniline are dissolved in 900 parts by volume of alcohol and, at 20250, a solution of 40 parts of sodium iodate in 900 parts of water is allowed to flow in while stirring. After stirring for 6 hours at room temperature, the precipitated red reaction product is filtered off with suction, washed with 1500 parts of water and three times with 50 parts by volume of methanol each time.
The 4,5-dianilino-1,2-benzoquinone thus obtained melts at 185-190 ", its purity is sufficient for the next stage.
21.8 parts of this product and 19.1 parts of N - (4-chloro-phenyl) -o-phenylenediamine hydrochloride are suspended in 1,600 parts by volume of hot alcohol and the mixture is refluxed for 6 hours with stirring. Then it is allowed to cool to 65-70 ", 30 parts of conc.
Ammonia is added and the mixture is stirred for a further hour at the same temperature. After cooling, the crude condensation product is filtered off with suction, washed with 500 parts by volume of water, dried and recrystallized from a mixture of benzene and petroleum ether. The 2-anilino-3-phenylimino-5- (p-chlorophenyl) -3, 5-dihydrophenazine is obtained as dark red crystals with a melting point of 205 to 206 ".
