Verfahren zur Herstellung von neuen heterocyclischen Stickstoffverbindungen
Die vorliegende Erfindung betrifft ein Verfahren zur Herstellung neuer heterocyclischer Stickstoffverbindungen, die sich als wertvolle Schutzmittel gegen Mikroorganismen erwiesen haben.
In den schweizerischen Patentschriften Nummern 350301 und 357402 ist die Herstellung und Verwendung neuer 2-Imino- 1,3 -di-N-heterocyclen der Formel

beschrieben. In dieser Formel stellen R1 einen lipophilen, aliphatischen Rest und R2 Wasserstoff oder einen niedermolekularen Alkylrest vor. A hat die Bedeutung eines den heterocyclischen Ring ergänzenden gesättigten Kohlenwasserstoffrestes, in welchem a, oder a,y-Kohlenwasserstoffatome mit den beiden Stickstoffatomen verbunden sind. Dieser zweiwertige Rest kann sowohl der aliphatischen wie der cycloaliphatischen Reihe angehören und ist vorwiegend eine acyclische oder monocyclische Alkylen- gruppe.
Mit dem Ausdruck lipophile Reste werden solche Gruppen verstanden, welche die Fett- oder Öllöslichkeit erhöhen, das heisst in erster Linie gesättigte Kohlenwasserstoffreste mit mindestens 10 Kohlenstoffatomen.
Diese neuen 2-Imino-1,3-di-N-heterocyclen sind wertvolle biocide Mittel, die sich durch ihre grosse Wirkungsbreite, besonders gute bakterizide, aber auch durch fungizide bzw. fungistatische Wirkung auszeichnen. Sie sind ebenfalls algicid gut wirksam.
Die Mittel können für sich allein als Pulver, in Lösung oder in Dispersion oder in Mischung mit andern Stoffen, wie inerten Träger- oder Füllstoffen, Reinigungsmitteln, Salbengrundlagen, Cremen, andern bakterizid und fungizid wirksamen Substanzen usw. gebraucht werden und in den verschiedensten Applikationsgebieten als desinfizierende und antiseptische Mittel Verwendung finden.
Bei der Weiterverarbeitung des Erfindungsgegenstandes wurde nun gefunden, dass 2-Imino-1 ,3-di-N- heterocyclen der Formel

worin R einen Cyclohexylrest darstellt und R1 und A die bereits weiter oben genannten Bedeutungen zukommen, ebenfalls wertvolle biocide Mittel sind.
Diese Verbindungen werden erfindungsgemäss hergestellt aus Diaminen der Formel R-NH-A-NH-R1 (11) worin R, R1 und A die vorgenannten Bedeutungen haben, durch Einwirkung von Halogencyan unter solchen Bedingungen, dass Ringschluss zum 2-Imino 1 ,3-di-N-heteroring eintritt.
Als Verbindungen der Formel II kommen durch einen Cyclohexylrest und einen lipophilen aliphatischen Rest substituierte a,P- und a,y-Diamino- alkane und -cycloalkane in Betracht, beispielsweise
N-Cyclohexyl-N'-decyl-1, 2-diaminoäthan,
N-Cyclohexyl-N'-dodecyl- 1 ,2-diaminoäthan,
N-Cyclohexyi-N'-tetradecyl- 1 ,2-diaminoäthan, N-Cyclohexyl-N'-dodecyli 1,3-diaminopropan und
N-Cyclohexyl-N'-dodecyl- 1 ,2-diaminocyclohexan.
Durch Behandlung mit Halogencyan erhält man daraus die entsprechenden 2-Imino-imidazolidine und 2-Iminohexahydropyrimidine.
Verbindungen der Formel II erhält man beispielsweise aus den entsprechenden primär-sekundären Diaminen durch Behandlung mit lipophile Reste einführenden Mitteln. Als solche kommen in erster Linie höhermolekulare Alkylhalogenide in Frage, beispielsweise Alkylchloride oder Alkylbromide mit wenigstens 10 und vorzugsweise 12 bis 14 Kohlenstoffatomen im Alkylrest. Auch die Alkalisalze entsprechender Fettalkoholmonosulfate können verwendet werden. Es ist jedoch auch möglich, den Cyclohexylrest in zweiter Stelle einzuführen, indem man cs, bzw. a,y-Aminoalkohole, in welchen die Aminogruppe durch einen lipophilen aliphatischen Rest substituiert ist, mit Säuren, beispielsweise Salzsäure oder Schwefelsäure, verestert und mittels Cyclohexylamin die Estergruppe gegen die Cyclohexylaminogruppe austauscht.
Die Umsetzung der Diaminoalkan- bzw. -cyclo- alkanverbindungen mit Halogencyan, als welches vor allem Chlorcyan und Bromcyan in Betracht kommt, erfolgt zweckmässig unter Bedingungen, wie sie bereits im Hauptpatent und im Schweizer Patent Nr. 350301 beschrieben sind. Desgleichen kann die Aufarbeitung der Reaktionsprodukte nach den dort gemachten Angaben erfolgen.
Die erfindungsgemäss hergestellten 2-Imino-1,3- di-N-heterocyclen sind farblose bis schwach ge färbte, kristallisierte bis wachsartige, lichtbeständige Substanzen, die sich in Form ihrer Salze, z. B. als halogenwasserstoffsaure Salze, in Wasser sehr leicht lösen. Die Verbindungen finden Verwendung in den verschiedensten Applikationsgebieten, z. B. in der Human- und Veterinärmedizin als desinfizierende und antiseptische Mittel, da sie gute bis sehr gute Wirksamkeit gegen Staphylokokken, Coli-, Typhus-, Paratyphus- und Enteritisbazillen, sowie gegen pathogene Pilze, wie z. B. Ctenomyces interdigitalis, zeigen. Dank der guten Wasserlöslichkeit können die neuen Verbindungen mit Vorteil auch zur desinfizie renden und antiseptischen Ausrüstung von Textilien verwendet werden.
Sie können ferner zur Raumdesinfektion dienen, beispielsweise in Form von Sprays. Ferner sind sie zur Desinfektion von Geräten und Gebrauchsgegenständen verwendbar und schliesslich können sie auch in der Kosmetik Verwendung finden, wozu sie z. B. zu Salben oder Cremen verarbeitet werden.
In den folgenden Beispielen sind die Teile als Gewichtsteile verstanden, und die Temperaturen sind in Celsiusgraden ausgedrückt. Gewichtsteile verhalten sich zu Volumteilen wie Kilogramm zu Liter.
Beispiel 1 1- Cyclohexyl - 2 - imino -3- dodecyl-imidazolidin- chlorhydrat.

73 Teile N-Cyclohexyl-N'-dodecyl- 1 ,2-diamino- äthan vom Kp. 228-233" werden in 500 Teilen Benzol gelöst. In diese Lösung leitet man bei Zimmertemperatur 14,5 Teile Chlorcyan ein. Das l-Cy ciohexyl -2-imino-3-dodecyi-irnidazolidinchlorhydrat scheidet sich als weisses Pulver aus. Es wird abgesaugt und aus Essigester umkristallisiert. Das gereinigte Produkt ist leicht wasserlöslich und schmilzt bei 15e-1520.

Gef.: 11,120/0 N, 9, 98 /o C1
Auf ähnliche Art lässt sich aus dem N-Cyclo hexyl-N'-tetradecyl-1 ,2-diaminoäthan vom Kp. 0.08 178-182" mit Bromcyan das l-Cyclohexyl-2-imino3-tetradecyl-imidazolidin-bromhydrat vom Smp. 148 bis 151" herstellen.
Die Ausgangsprodukte N-Cyclohexyl-N'-dodecyl bzw. -tetradecyl-1 ,2-diaminoäthane werden z. B. aus N-(fl-Hydroxy-äthyl)-dodecyl- bzw. -tetradecyl-amin durch Verestern mit Schwefelsäure und Kondensation mit Cyclohexylamin erhalten.
Die beiden Imidazolidinderivate zeigen sehr gute bakterizide und fungizide Wirkung und können daher als Wirkstoffe in Desinfektionsmitteln verwendet werden. Sie sind z. B. wirksam gegen die Bakterien Staphylokokkus aureus, Staphylokokkus albus, Escheria coli und den pathogenen Pilz C. interdigitalis.
Beispiel 2 1-Cyclohexyl-2-imino-3-dodecyl-hexahydropyri- midin-bromhydrat.

40 Teile N-Cyclohexyl-N'-dodecyl- 1,3-diaminopropan vom Kp. 0.08 l82l860 werden in 250 Teilen Chlorbenzol zum Sieden erhitzt. Bei Siedetemperatur wird dann während drei Stunden eine Lösung von 13 Teilen Bromcyan in 85 Teilen Chlorbenzol zugetropft. Nach dem Zutropfen wird unter Rühren noch eine halbe Stunde bei Siedetemperatur gehalten, hierauf das Reaktionsgemisch von wenig Unlöslichem geklärt und das Chlorbenzol im Vakuum abdestilliert. Der braune harzige Rückstand wird in 100 Teilen warmem Benzol gelöst und die Lösung mit Petroläther versetzt. Das l-Cyclohexyl-2-imino-3dodecyl-hexahydro-imidazolidin-bromhydrat scheidet sich dabei ab, wird dann abfiltriert und aus 150 Teilen Aceton umkristallisiert. Der Schmelzpunkt des reinen Produktes beträgt 8890o.

Gef.: 9,550/MN, 19,00/0 Br
Auf ähnliche Art lässt sich aus N-Cyclohexyl-N'dodecyl-l,2-diaminocyclohexan vom Kp. 13 256 bis 2620 und Chlorcyan das l-Cyclohexyl-2-imino-3 dodecyl -4,5 - tetramethylenimdazolidin -chlorhydrat herstellen. Smp. 174-175".
Die Produkte sind klar wasserlöslich und können dank ihrer guten bakteriziden und algiziden Eigenschaften als Wirkstoffe in antiseptischen Mitteln verwendet werden.

described. In this formula, R1 represents a lipophilic, aliphatic radical and R2 represents hydrogen or a low molecular weight alkyl radical. A has the meaning of a saturated hydrocarbon radical supplementing the heterocyclic ring, in which a, or a, y-hydrocarbon atoms are bonded to the two nitrogen atoms. This divalent radical can belong to both the aliphatic and the cycloaliphatic series and is predominantly an acyclic or monocyclic alkylene group.
The expression lipophilic radicals are understood to mean those groups which increase the solubility in fat or oil, that is to say primarily saturated hydrocarbon radicals with at least 10 carbon atoms.
These new 2-imino-1,3-di-N-heterocycles are valuable biocidal agents which are characterized by their wide range of action, particularly good bactericidal, but also by fungicidal or fungistatic effects. They are also effective as algicides.
The agents can be used on their own as a powder, in solution or in dispersion or in a mixture with other substances, such as inert carriers or fillers, cleaning agents, ointment bases, creams, other bactericidal and fungicidal substances, etc., and in a wide variety of fields of application as Disinfectants and antiseptic agents are used.
In further processing of the subject matter of the invention, it has now been found that 2-imino-1,3-di-N-heterocycles of the formula

in which R represents a cyclohexyl radical and R1 and A have the meanings already given above, are likewise valuable biocidal agents.
These compounds are prepared according to the invention from diamines of the formula R-NH-A-NH-R1 (11) in which R, R1 and A have the aforementioned meanings, by the action of cyanogen halide under conditions such that ring closure to the 2-imino 1,3 di-N heteroring occurs.
Suitable compounds of the formula II are a, P- and a, y-diaminoalkanes and cycloalkanes which are substituted by a cyclohexyl radical and a lipophilic aliphatic radical, for example
N-cyclohexyl-N'-decyl-1, 2-diaminoethane,
N-cyclohexyl-N'-dodecyl-1, 2-diaminoethane,
N-Cyclohexyi-N'-tetradecyl-1, 2-diaminoethane, N-cyclohexyl-N'-dodecyli 1,3-diaminopropane and
N-Cyclohexyl-N'-dodecyl-1,2-diaminocyclohexane.
Treatment with cyanogen halide gives the corresponding 2-imino-imidazolidines and 2-iminohexahydropyrimidines.
Compounds of the formula II are obtained, for example, from the corresponding primary-secondary diamines by treatment with agents which introduce lipophilic radicals. Higher molecular weight alkyl halides are primarily suitable as such, for example alkyl chlorides or alkyl bromides with at least 10 and preferably 12 to 14 carbon atoms in the alkyl radical. The alkali salts of corresponding fatty alcohol monosulfates can also be used. However, it is also possible to introduce the cyclohexyl radical in the second position by esterifying cs or a, y-amino alcohols in which the amino group is substituted by a lipophilic aliphatic radical with acids, for example hydrochloric acid or sulfuric acid, and using cyclohexylamine Ester group exchanged for the cyclohexylamino group.
The reaction of the diaminoalkane or cycloalkane compounds with cyanogen halide, which is primarily cyanogen chloride and cyanogen bromide, is expediently carried out under conditions as already described in the main patent and in Swiss patent no. The 2-imino-1,3-di-N-heterocycles produced according to the invention are colorless to weakly colored, crystallized to waxy, light-resistant substances that are in the form of their salts, eg. B. as hydrohalic acid salts, very easily dissolve in water. The compounds are used in a wide variety of application areas, e.g. B. in human and veterinary medicine as disinfecting and antiseptic agents, as they have good to very good effectiveness against staphylococci, coli, typhoid, paratyphoid and enteritis bacilli, and against pathogenic fungi, such as. B. Ctenomyces interdigitalis show. Thanks to their good solubility in water, the new compounds can also be used to advantage for the disinfecting and antiseptic treatment of textiles.
They can also be used to disinfect rooms, for example in the form of sprays. They can also be used for disinfecting devices and utensils, and finally they can also be used in cosmetics, for which they are used, for. B. processed into ointments or creams.
In the following examples, parts are parts by weight and temperatures are expressed in degrees Celsius. Parts by weight relate to parts by volume as the kilogram is to the liter.

73 parts of N-cyclohexyl-N'-dodecyl-1,2-diaminoethane of boiling point 228-233 "are dissolved in 500 parts of benzene. 14.5 parts of cyanogen chloride are passed into this solution at room temperature. The 1-Cy Ciohexyl -2-imino-3-dodecyi-irnidazolidine chlorohydrate separates out as a white powder. It is filtered off with suction and recrystallized from ethyl acetate. The purified product is easily soluble in water and melts at 15e-1520.

In a similar way, 1-cyclohexyl-2-imino3-tetradecyl-imidazolidine bromohydrate with a melting point of 148 can be obtained from N-cyclohexyl-N'-tetradecyl-1,2-diaminoethane of b.p. 0.08 178-182 "with cyanogen bromide up to 151 ".
The starting products N-cyclohexyl-N'-dodecyl or -tetradecyl-1, 2-diaminoethane are z. B. obtained from N- (fl-hydroxy-ethyl) -dodecyl- or -tetradecyl-amine by esterification with sulfuric acid and condensation with cyclohexylamine.
The two imidazolidine derivatives show very good bactericidal and fungicidal effects and can therefore be used as active ingredients in disinfectants. You are e.g. B. effective against the bacteria Staphylococcus aureus, Staphylococcus albus, Escheria coli and the pathogenic fungus C. interdigitalis.
Example 2 1-Cyclohexyl-2-imino-3-dodecyl-hexahydropyrimidine bromohydrate.

40 parts of N-cyclohexyl-N'-dodecyl-1,3-diaminopropane with a boiling point of 0.081821860 are heated to boiling in 250 parts of chlorobenzene. At boiling temperature, a solution of 13 parts of cyanogen bromide in 85 parts of chlorobenzene is then added dropwise over a period of three hours. After the dropwise addition, the mixture is kept at the boiling point for half an hour while stirring, then the reaction mixture is cleared of little insolubles and the chlorobenzene is distilled off in vacuo. In a similar manner, 1-cyclohexyl-2-imino-3-dodecyl-4,5-tetramethylene-imdazolidine chlorohydrate can be prepared from N-cyclohexyl-N'dodecyl-1,2-diaminocyclohexane with a bp. 13 256 to 2620 and cyanogen chloride. M.p. 174-175 ".
The products are clearly soluble in water and, thanks to their good bactericidal and algicidal properties, can be used as active ingredients in antiseptic agents.
