Verfahren zur Herstellung von Tetrahydrofurfuryl-ammoniumverbindungen Die Erfindung betrifft ein Verfahren zur Herstel lung von Tetrahydrofurfuryl-ammoniumverbindungen mit acetylcholinähnlicher Wirkung, welche als Sym- pathicomimetika verwendet werden können.
Das erfindungsgemässe Verfahren zur Herstellung dieser Verbindungen ist dadurch gekennzeichnet, dass man die Oxogruppe im [(2-Methyl-3-oxo-tetrahydro- furan-5-yl)-methyl]-trialkylammoniumhalogenid zur Hydroxylgruppe reduziert.
Die Reduktion der Oxogruppe zur Hydroxyl- gruppe kann beispielsweise durch katalytische Reduk tion mit Wasserstoff in Gegenwart eines Nickelkata- lysators, z. B. Raneynickel, oder durch chemische Re duktion mittels eines Metallhydrides, z. B. Lithium- aluminiumhydrid oder Natriumborhydrid, erfolgen.
Die nach dem erfindungsgemässen Verfahren her stellbaren Tetrahydrofurfuryl-ammoniumsalze besit zen die folgende Formel

in welcher Z eine quaternisierte Aminogruppe dar stellt. Besonders bevorzugte Verbindungen der obigen Formel sind solche, in denen Z eine Trialkylammo- niumgruppe, insbesondere eine Trimethylammonium- gruppe, bedeutet. Die Verfahrensendprodukte weisen mit Vorteil solche Anionen auf, die üblicherweise in pharmazeutischen Präparaten zur Anwendung kom men, z. B.
Chlor-, Brom-, Sulfat- oder Phosphationen.
Die Verfahrensprodukte stellen zum Teil hygro skopische Produkte dar, welche zwecks Isolierung oder Reinigung in die kristallisierten Chloroaurate, Reineckate oder Tetraphenylboronate überführt wer den können. Aus diesen kristallisierten Salzen kann man die gewünschten pharmazeutisch verwendbaren Salze durch Behandeln mit den entsprechenden Säu ren gewinnen.
Da die Moleküle der Verfahrensendprodukte drei asymmetrische Kohlenstoffatome enthalten, gibt es acht stereoisomere Formen derselben. In den nach dem erfindungsgemässen Verfahren erhältlichen Re aktionsprodukten kommen mehrere solche Formen vor. Sie können voneinander in üblicher Weise, z. B. durch fraktionierte Kristallisation, getrennt werden. Es ist festgestellt worden, dass eine dieser stereoiso- meren Formen mit dem wirksamen Prinzip des Flie genpilzes, dem Muscarin, identisch ist.
<I>Beispiel</I> Man löst 2 g 2-Methyl-3-oxo-tetrahydrofuran-5- carbonsäure-methylester in 4 cms Orthoameisensäure- methylester und versetzt die Lösung mit 2 Tropfen konzentrierter Schwefelsäure. Allmählich tritt leichte Erwärmung ein. . Nach 2stündigem Stehen bei Zim- mertemperatur wird das Gemisch mit einer gesättig ten Natriumbicarbonatlösung neutralisiert und das entstandene Ketal mit Äther extrahiert.
Der ätherische Auszug wird verdampft und der Rückstand im Hoch vakuum destilliert. Der gewonnene 2-Methyl-3,3-di- methoxy - tetrahydrofuran - 5 - carbonsäuremethylester stellt ein farbloses und fast geruchloses Öl vom Siede punkt 121 /1 mm dar. Ausbeute: 85 %.
2 g 2-Methyl-3,3-dimethoxy-tetrahydrofuran-5- carbonsäuremethylester werden im Bombenrohr mit 10 cm3 33 0/aiger alkoholischer Lösung von Dimethyl- amin während 12 Stunden auf 120 erhitzt. Das Lö sungsmittel wird dann abdestilliert und das leicht gelb- lich gefärbte Reaktionsprodukt im Hochvakuum de stilliert.
Das so gewonnene 2-Methyl-3,3-dimethoxy- 5-dimethylaminocarbonyl-tetrahydrofuran siedet bei 1400/0,01 mm.
8 g 2-Methyl - 3,3 - dimethoxy - 5 - dimethylamino- carbonyl-tetrahydrofuran werden in 250 cm3 absolu tem Äther mittels 4 g Lithiumaluminiumhydrid redu ziert. Nach 2stündigem Erwärmen unter Rückfluss wird das Reaktionsgemisch mit Salzsäure versetzt und die Ätherschicht nach dem Abtrennen nochmals mit verdünnter Salzsäure extrahiert.
Man lässt die ver einigten wässrigen Lösungen zwecks Verseifung des Dimethylketals noch eine Stunde bei Raumtempera tur stehen, versetzt sie dann mit konzentrierter Kali lauge bis zur stark alkalischen Reaktion, sättigt mit Kochsalz und extrahiert erschöpfend mit Äther aus. Nach Abdestillieren des Äthers wird das zurückblei bende braune<B>öl</B> im Hochvakuum destilliert.
Man ge winnt so das 2-Methyl-3-oxo-5-dimethylaminomethyl- tetrahydrofuran vom Siedepunkt 90 /0,01 mm. Das Destillat wird in der fünffachen Menge Äther mit dem gleichen Volumen Methyljodid versetzt, worauf das [(2-Methyl-3-oxo-tetrahydrofuran-5-yl)-methyl]-trime- thylammoniumjodid auskristallisiert.
Dieses Jodid bil det aus Aceton Prismen vom Schmelzpunkt 140 , das Tetraphenylboronat aus Aceton/Methanol Kristalle vom Schmelzpunkt 181 . 0,13 g dieses Ammonium- jodides werden in 3 cm3 Isopropanol gelöst und mit einer Lösung von 0,1g Natriumborhydrid in 3 cm3 Isopropanol während 12 Stunden stehengelassen.
Nach Ansäuern mit Essigsäure wird das Lösungsmit tel abgedampft, der Rückstand in Wasser gelöst und mit einer Lösung von 0,2 g Tetraphenylbornatrium in 3 cm3 Wasser versetzt, wobei das [(2-Methyl-3- hydroxy-tetrahydrofuran- 5 -yl)-methyl]-trimethylam- monium-tetraphenylboronat ausfällt, das nach Umkri stallisation aus Aceton/Methanol Kristallnadeln vom Schmelzpunkt 170 bildet.

This iodide forms prisms with a melting point of 140 from acetone, and the tetraphenylboronate from acetone / methanol crystals with a melting point of 181. 0.13 g of this ammonium iodide are dissolved in 3 cm3 of isopropanol and left to stand for 12 hours with a solution of 0.1 g of sodium borohydride in 3 cm3 of isopropanol.
