Verfahren zur Herstellung von neuen Derivaten des Benzimidazols
Die therapeutische Wirkung der Substitutionsprodukte des Benzimidazols ist wiederholt untersucht worden. Man hat festgestellt, dass die Wirkung je nach der Natur der Substituenten stark schwankt.
Bezüglich der Substituenten in der 2-Stellung des Benzimidazols ist festgestellt worden, dass Kohlenwasserstoffreste als Substituenten in der 2-Stellung zu Produkten mit sehr geringer spasmolytischer Wirkung führen (J. Am. Chem. Soc. 71, S. 2035), wäh- rend Substituenten mit basischen N-Atomen in der 2-Stellung des Benzimidazols dann zu praktisch brauchbaren Produkten führen, wenn die 1-Stellung durch einen substituierten oder nichtsubstituierten Aralkyl-, Aryl- oder heterocyclischen Rest bzw. heterocyclisch-substituierten Alkylrest besetzt ist (DBP Nr. 890644).
Es sind auch schon eine Anzahl von Derivaten des Benzimidazols für die Behandlung von Pilzinfektionen des menschlichen oder tierischen Körpers vorgeschlagen worden, jedoch haben sich diese Verbindungen nicht als völlig zufriedenstellend erwiesen.
Die vorliegende Erfindung bezieht sich nun auf ein Verfahren zur Herstellung von neuen Derivaten des Benzimidazols, welche eine überlegene therapeutische Wirkung ausüben und der Formel I entsprechen:

worin X einen gegebenenfalls durch Hydroxylgruz pen substituierten Alkylrest mit bis zu 4 Kohlenstoffatomen, Y ein Halogenatom und die Reste Rl bis % Wasserstoff, niedere Alkylreste, Halogenatome, Nitrogruppen oder gegebenenfalls substituierte Aminogruppen bedeuten. Die vorgenannten neuen Derivate werden erfindungsgemäss dadurch erhalten, dass ein Benzimidazol der Formel II

oder ein Metallderivat eines solchen Benzimidazols mit einem entsprechenden Halogenbenzylhalogenid umgesetzt wird.
Die erfindungsgemäss erhältlichen neuen Benzimidazolderivate lassen sich in an sich üblicher Weise mit Säuren in ihre Salze überführen.
Die neuen Derivate des Benzimidazols wirken sehr stark fungistatisch. Daneben haben sie sehr gute spasmolytische Eigenschaften. Die überlegene Wirkung der neuen Benzimidazolderivate ergibt sich aus Vergleichsversuchen, bei denen die Wirkung der Substanz auf den durch Acetylcholin oder Bariumchlorid hervorgerufenen Krampf des Meerschweinchendarmes mit dem des Papaverins verglichen wurde. B. im Vergleich zu dem aus dem deutschen Patent Nr. 927993 bekannten l-p-Chlorbenzyl-benzimidazol (Verbindung I) geht aus der folgenden Tabelle hervor:

Beispiel 1
26,4 g 2-Methylbenzimidazol werden in 350 cm5 Dioxan gelöst und mit 10 g Natriumamid versetzt.
Nach etwa 5 Minuten gibt man zu diesem Gemisch 41,2 g p-Chlorbenzylbromid hinzu und kocht dann 6 Stunden lang am Rückflusskühler. Dann wird das Dioxan abdestilliert, der Rückstand gut mit verdünnter Salzsäure verriihrt und die dabei erhaltene kristalline Masse nach Absaugen aus Wasser umkristallisiert. Die beim Abkühlen erhaltenen farblosen Kristalle werden in heissem Wasser gelöst und diese Lösung mit verdünnter Ammoniaklösung versetzt, bis sie schwach alkalisch reagiert. Die anfangs flüssige Base des l-p-Chlorbenzyl-2-methylbenzimidazols erstarrt unter Hydratbildung zu einer weissen Masse, die aus wässrigem Alkohol umkristallisiert werden kann und einen Schmelzpunkt von 67-68" besitzt.
Die Aufarbeitung kann auch so erfolgen, dass nach dem Abdestillieren des Dioxans der Rückstand im Vakuum destilliert wird. Dabei geht die Base des l-p-Chlorbenzyl-2-methylbenzimidazols bei 12 mm Druck bei 240-242"C als farbloses Öl über, aus dem durch Verreiben mit Wasser das Hydrat vom Schmelzpunkt 67-68" erhalten werden kann.
Nach dem gleichen Verfahren wurden folgende weitere l-p-Chlorbenzylbenzimidazolderivate erhalten:
1 -pChlorbenzyl-2-äthyIbenzimidazol (Fp. 116118O),
1 -p-ChIorbenzyl-2-propylbenzimidazol (Fp. 115116 ) und 1 -p-Chlorbenzyl-2-(i-butyl)-benzimidazol- hydrochlorid (Fp. 2400).
Beispiel 2
13,2 g 2-Methylbenzimidazol werden in 200 cm3 Toluol mit 5 g Natriumamid und 20,6 g pChlorbenzylbromid versetzt. Nach 5 stündigem Kochen am Rückflusskühler wird das Toluol abdestilliert und der Rückstand wie in Beispiel 1 aufgearbeitet.
Beispiel 3
26,4 g 2-Methylbenzimidazol werden in 200 cm5 Dimethylformamid gelöst, mit 12 g Kaliumhydroxyd versetzt und dieses Gemisch für 10-15 Minuten unter Rückfluss gekocht, wobei das Kaliumhydroxyd in Lösung geht. Zu der so erhaltenen Lösung gibt man dann eine Lösung von 20,6 g p-Cldorbenzyl- bromid in 50 cm5 Dimethylformamid und erhitzt nun für 2 Stunden am Rückfluss-Kühler zum Sieden. Der nach dem Abdestillieren des Dimethylformamids erhaltene Rückstand wird ! wie in Beispiel 1 aufge- arbeitet.
Process for the preparation of new derivatives of benzimidazole
The therapeutic effect of the substitution products of benzimidazole has been repeatedly investigated. It has been found that the effect varies widely depending on the nature of the substituents.
With regard to the substituents in the 2-position of the benzimidazole, it has been found that hydrocarbon radicals as substituents in the 2-position lead to products with a very low spasmolytic effect (J. Am. Chem. Soc. 71, p. 2035), while Substituents with basic N atoms in the 2-position of the benzimidazole lead to practically useful products if the 1-position is occupied by a substituted or unsubstituted aralkyl, aryl or heterocyclic radical or heterocyclic-substituted alkyl radical (DBP No. 890644).
A number of derivatives of benzimidazole have also been proposed for the treatment of fungal infections of the human or animal body, but these compounds have not proven entirely satisfactory.
The present invention now relates to a process for the preparation of new derivatives of benzimidazole which exert a superior therapeutic effect and correspond to formula I:

or a metal derivative of such a benzimidazole is reacted with a corresponding halobenzyl halide.
The new benzimidazole derivatives obtainable according to the invention can be converted into their salts with acids in a conventional manner.
The new derivatives of benzimidazole have a very strong fungistatic effect. They also have very good spasmolytic properties. The superior effect of the new benzimidazole derivatives results from comparative tests in which the effect of the substance on the spasm of the guinea pig intestine caused by acetylcholine or barium chloride was compared with that of papaverine. 26.4 g of 2-methylbenzimidazole are dissolved in 350 cm5 of dioxane, and 10 g of sodium amide are added.
After about 5 minutes, 41.2 g of p-chlorobenzyl bromide are added to this mixture and the mixture is then refluxed for 6 hours. The dioxane is then distilled off, the residue is mixed well with dilute hydrochloric acid and the crystalline mass obtained is recrystallized from water after suction. The colorless crystals obtained on cooling are dissolved in hot water and dilute ammonia solution is added to this solution until it has a weakly alkaline reaction. The initially liquid base of l-p-chlorobenzyl-2-methylbenzimidazole solidifies with hydrate formation to form a white mass that can be recrystallized from aqueous alcohol and has a melting point of 67-68 ".
Working up can also be carried out in such a way that, after the dioxane has been distilled off, the residue is distilled in vacuo. The base of the l-p-chlorobenzyl-2-methylbenzimidazole changes to a colorless oil at 12 mm pressure at 240-242 "C, from which the hydrate with a melting point of 67-68" can be obtained by trituration with water.
The following further l-p-chlorobenzylbenzimidazole derivatives were obtained using the same procedure:
1-p-chlorobenzyl-2-ethylbenzimidazole (mp. 116118O),
1-p -chlorobenzyl-2-propylbenzimidazole (m.p. 115116) and 1 -p-chlorobenzyl-2- (i-butyl) -benzimidazole hydrochloride (m.p. 2400).
Example 2
13.2 g of 2-methylbenzimidazole in 200 cm3 of toluene are mixed with 5 g of sodium amide and 20.6 g of p-chlorobenzyl bromide. After boiling for 5 hours on the reflux condenser, the toluene is distilled off and the residue is worked up as in Example 1.
Example 3
26.4 g of 2-methylbenzimidazole are dissolved in 200 cm5 of dimethylformamide, 12 g of potassium hydroxide are added and this mixture is refluxed for 10-15 minutes, the potassium hydroxide going into solution. A solution of 20.6 g of p-chlorobenzyl bromide in 50 cm5 of dimethylformamide is then added to the solution obtained in this way and the mixture is then heated to boiling for 2 hours on the reflux condenser. The residue obtained after distilling off the dimethylformamide is! worked up as in example 1.
