Verfahren zur Herstellung von Verbindungen mit blutdrucksenkender Wirkung
Die vorliegende Erfindung betrifft ein Verfahren zur Herstellung von Verbindungen mit blutdrucksenkender Wirkung der Formel

worin n eine ganze Zahl von 4 bis 6, R, Rl, R2, R3 und R4 Alkylgruppen und X ein Anion bedeuten und deren Gesamtzahl an Kohlenstoffatomen im Kation 11 bis 16 beträgt, dadurch gekennzeichnet, dass ein entsprechendes di-N-substituiertes Alkylthioalkylamin oder ein quaternäres Ammoniumsalz eines solchen mit einem Überschuss eines quaterni- sierenden Mittels umgesetzt wird. Verbindungen dieser Formel haben eine ausgesprochen blutdrucksenkende Wirkung (wie sich im Tierversuch ergab).
Hierbei zeigte sich, dass die blutdrucksenkende Wirkung insbesondere denjenigen Verbindungen zukommt, in denen n 4 oder 5 ist.
Die Alkylgruppen R, Ri, R2, R3 und R4 können gleich oder verschieden sein.
Unter den Verbindungen der Formel I haben sich diejenigen mit 13 bis 15 Kohlenstoffatomen im Kation als besonders wirksam erwiesen.
Im nachfolgenden bedeuten die Abkürzungen Me, Et, Prll, Pr'und Bun die Methyl-, Äthyl-, n-Propyl-, Isopropyl-und n-Butylgruppe.
Die Verbindung, in welcher n = 4 ist und sämt- liche Reste R Methyl sind, welche nach Scheider und Kaufmann (Annalen., 1912, 392, 10) hergestellt werden kann, indem man das Amin CH,. S- (CH,),. NH, mit Methyliodid in Gegenwart von Natriummethoxyd vollständig methyliert, hat nur geringe blutdrucksenkende Wirkung. Demgegenüber erweist sich die Verbindung der Formel I, in welcher n = 4 ist und die Reste R bis R4 das Äthylradikal bedeuten und deren Herstellung in Beispiel I beschrieben ist, als ausserordentlich stark blutdrucksenkend.
Die Art des Anions X ist ohne besondere Bedeutung. Es kann irgendeines der normalerweise für pharmazeutische Zwecke verwendeten Anionen verwendet werden, sofern die betr. Verbindung geeignete physikalische Eigenschaften (Stabilität, Löslichkeit usw.) aufweist. Selbstverständlich sind hierbei toxisch wirkende Anionen, wie z. B. das Oxalation, zu vermeiden. Die Herstellung der Salze organischer Säuren kann vorgenommen werden, indem man die Halogensalze mit Silbersalzen der geeigneten Säuren zur Reaktion bringt, oder indem geeignete Säuren in Gegenwart von Äthylenoxyd reagieren gelassen werden (Sackur, Bull. Soc. chim. 1952, 796).
Vorzugsweise stellt man nach dem erfindungsgemässen Verfahren die Salze der folgenden Verbin- dungen her :
1-Diäthylsulfonium-4-triäthylammoniumbutan, 1-Äthylmethylsulfonium-4-triäthylammonium- butan, 1-Athylmethylsulfonium-4-äthyl-di-n-propyl- ammoniumbutan,
1-Dimethylsulfonium-4-methyl-di-isopropyl ammoniumbutan, 1-Athylmethylsulfonium-4-methyl-di-isopropyl- ammoniumbutan,
1-Methylisopropylsulfonium-4-methyl-di isopropylammoniumbutan, 1-Dimethylsulfonium-4-methyl-di-n- propylammoniumbutan, 1-Diäthylsulfonium-5-triäthylammoniumpentan, und 1-Athylmethylsulfonium-5-äthyl-di-n-propyl- ammoniumpentan.
Der Ausdruck quaternisierendes Mittel, wie er im vorstehenden verwendet worden ist, bezieht sich auf solche Reagenzien, wie z. B. Methylhalogenide, welche befähigt sind, alkylierte Stickstoffatome in die quaternäre Ammoniumbase überzuführen bzw. alkylierte Schwefelatome in die tertiäre Sulfoniumbase zu verwandeln.
Die di-N-substituierten Alkylthioalkylamine, in welchen die Gesamtzahl der Kohlenstoffatome 7 bis 14, vorzugsweise 11-13 ist, sind neue Verbindungen und können durch Reaktion geeigneter Halogenalkylamine in Form ihrer halogensauren Salze (ge wöhnlich werden die Chloralkylamine in Form ihrer chlorwasserstoffsauren Salze verwendet) hergestellt werden mit dem Metallsalz eines Alkan-thiols (meist im Überschuss verwendet) in einem Lösungsmittel, z. B. Athanol (Brighton und Reid, J. Amer. Chem.
Soc., 1943, 65, 458 ; Andrews, Bergel und Morrison, J. C. S., 1953, 2998 und Kirchner, Soria und Cavallito, J. Chem. Soc. 1955, 77, 4599).
Ein anderer Weg zur Herstellung von di-N-substituierten Alkylthioalkylaminen ist die Umwandlung geeigneter di-N-substituierter Hydroxyalkylamine in die Thioalkylamine durch Einwirkung von Bromwasserstoffsäure und Thioharnstoff (Frank und Smith, J. Amer. Chem. Soc., 1946, 68, 2103 ; Plant, Tarbell und Whiteman, ibid, 1955, 77, 1572), worauf das Reaktionsgemisch alkalisch gemacht wird und die Alkylierung der auf diese Weise gebildeten Thiolgruppe mit üblichen Methoden durchgeführt wird.
Die folgenden Reaktionsschemen verbildlichen, wie die genannten Verbindungen hergestellt werden können.

Beispiel 1 1-DiÏthylsulfonium-4-triÏthylammoniumbutan-di-iodid a) Zu einer L¯sung von 4-¯thyl-thio-butyldiÏthylamin (1, 9 g) in trockenem Aceton (20 ml) wurden Athyliodid (4, 9 ml) zugegeben und die erhaltene Mischung am Rückfluss auf dem Wasserbad 8 Stunden lang gerührt. Nach Entfernen des ersten Anteils eines kristallinen Produktes (2, 2 g) Smp. 143 C (u. Z.) wurde die Mutterlauge weitere 14 Stunden lang am Rückfluss gekocht, wobei ein zweiter Anteil des Reaktionsproduktes (1, 1 g) vom Smp. 139-140¯C (Z.) anfiel. Die vereinigte Ausbeute (3, 3 g) wurde aus Athanol (10 ml) umkristallisiert, wobei farblose Prismen erhalten wurden (2, 9 g, 59 /o) Smp. =142¯C (u.
Z.) (Gefunden : C, 33, 4 ; H, 6, 5 ; I, 50, 3 0/o, berechnet für C 14H33 NSI,, : C, 33, 5 ; H, 6, 6 ; I, 50, 6 /o).
Zur Herstellung des Dihydrogentartrates wurde eine wässrige Lösung des Diodids (1 Mol) mit einer Aufschlemmung von neutralem Silbertartrat (1 Mol) in wässeriger Weinsäurelösung (1 Mol) geschüttelt.
Das ausgefallene Silberjodid wurde abfiltriert, das Filtrat im Vakuum (Wasserstrahlpumpe) eingeengt und die halbfeste, weisse kristalline Masse im Vakuum (ohne Verwendung eines Trocknungsmittels) getrocknet, wobei ein weisses, kristallines Pulver (97 /o) vom Smp. 103-104¯C anfiel. Beim Umkristallisieren aus 95 /o igem Athanol ergaben sich Büschel farbloser Nadeln des Dihydrats vom Smp. 103-105¯C.

Eine andere Methode zur Umwandlung des di Iodids in das di-Hydrogentartrat folgt in der Arbeitsweise, nach Sackur (loc. cit.), nach welcher di-Iodid (1 Mol) und Weinsäure (3 Mol) in warmer, konzentrierter alkoholischer Lösung mit Athylenoxyd (4 Mol) behandelt wurden. Das di-Hydrogentartrat fiel aus und wurde in der oben beschriebenen Weise gereinigt. b) Eine Mischung von Athyliodid (0, 81 mol) und 4-¯thyl-thiobutyldiÏthylamin (1, 9 g) wurde 7 Tage lang bei Raumtemperatur ohne Gegenwart eines Lösungsmittels reagieren gelassen.
Die erhaltene feste Substanz wurde mit trockenem Ather digeriert, abfiltriert und gereinigt, indem sie in Aceton (5 ml) aufgelöst und mit trockenem Ather (10 ml) wieder ausgefällt wurde, wobei das 4-¯thyl-thiobutyltriÏthyl ammoniumiodid in Form farbloser Nadeln anfiel (2, 0 g, 58 /o). Smp. 95-96¯C. (Gefunden : C, 42, 1 ; H, 8, 0 ; I, 36, 8% berechnet für CI2H28NSI : C, 41, 7 ; H, 8, 1 ; I, 36, 8%.)
Zu einer Lösung von 4-Athyl-thiobutyltriäthylammoniumiodid (1, 8 g) in warmem, trockenem Aceton (10 ml) wurde Athyliodid (2, 0 ml) hinzugefügt und die erhaltene Lösung am Rückfluss auf dem Wasserbad 24 Stunden lang gekocht.
Hierauf wurde die Lösung abgekühlt, das Reaktionsprodukt abfiltriert (2, 0 g, 76 /o), und aus Athanol umkristallisiert.
Hierbei wurden farblose Prismen erhalten, Smp. 138 bis 140 C (u. Z.). (Gefunden : I, 50, 6%.)
Das bei diesem Herstellungsverfahren verwendete 4-¯thyl-thiobutyldiÏthylamin kann folgendermassen hergestellt werden :
Zu einer Lösung von 4-Hydroxybutyldiäthylamin (156 g) (vison, J. Appl. Chem. 1951, 1 469) in trockenem Chloroform (160 ml) wurde unter Rühren und bei äusserer Kühlung eine Lösung von Thionylchlorid (97 ml) in 200 ml trockenem Chloroform zugegeben. (Gekühlt wird mit einem Kohlensäurebad.) Das Reaktionsgemenge wurde auf Raumtemperatur erwärmen gelassen, eine halbe Stunde lang am R ckfluss gekocht und hierauf konzentriert, zuerst unter normalem Druck und schliesslich im Vakuum, um Spuren von Chloroform zu entfernen.
Das verbleibende Íl wurde zweimal mit trockenem Benzol in azeotroper Mischung abdestilliert und dann abgekühlt, worauf es auskristallisierte. Das erhaltene 4-Chlorbutyldiäthylaminhydrochlorid konnte im nächsten Schritt der Synthese ohne weitere Reinigung direkt verwendet werden.
Zu einer Lösung von Natrium (89 g) in Athanol (2 1) wurde unter Kühlung Athylmercaptan (290 ml) zugegeben. Zu der eben auf Rückflusstemperatur erwärmten Lösung wurde im Verlauf von einer Stunde die vorstehend erhaltene gesamte Menge von 4-Chlorbutyldiäthylaminhydrochlorid in Athanol (550 ml) zugefügt. Hierauf wurde die Mischung 2 Stunden lang am Rückfluss gekocht und schliesslich unter normalem Druck auf einem siedenden Wasserbad der Alkohol so vollständig wie möglich abdestilliert. Der Rückstand wurde abgekühlt, mit Wasser (1, 5 1) verdünnt und mit Ather (3 mal 1 1) extrahiert.
Die vereinigten Extrakte wurden mit Wasser (3 mal 250 ml) gewaschen, getrocknet (MgSO4), konzentriert und im Vakuum destilliert, wobei ein farbloses Íl erhalten wurde (172, 8 g, 87%) Sdp. 105-113 C/10 mm, nD 1, 4705. (Gefunden : C, 63, 6 ; H, 12, 3 ; S. 16, 9 /ol, berechnet für C10H23NS : C, 63, 5 ; N, 12, 2 ; S, 16, 9%.)
Das auf die vorstehend beschriebene Weise hergestellte 4-Mercaptobutyldiäthylamin (161 g) wurde einer gekühlten Lösung von Natrium (23 g) in Atha- nol (460 ml) zugegeben, hierauf Athylbromid (120 g) zugefügt und das Gemisch am Rückfluss 3 Stunden lang gekocht.
Aus der Reaktionsmischung wurde das ausgefallene Natriumbromid abfiltriert, das Filtrat eingeengt, erneut filtriert und schliesslich im Vakuum destilliert, wobei in Form eines farblosen Ols das 4-¯thylthiobutyldiÏthylamin anfiel (152 g, 80%.)
Beispiel 2 1-A'thylmethylsulf onium-4-tridthyl-ammonium- butan-di-iodid
In einer Lösung von Nitromethan (8, 4 ml) wurden Athyliodid (8, 4 ml) und 4-Methylthiobutyldiäthylamin (3, 0 g) eine Woche lang bei Raumtemperatur miteinander reagieren gelassen. Nach Zugeben von trockenem Ather (25 ml) wurde das ölige Produkt absetzen gelassen und die überstehende Nitromethanätherschicht abgezogen und durch Aceton ersetzt.
Das erhaltene Produkt wurde zum Kristallisie ren gebracht und hierauf zweimal aus n-Butanol (40 ml) bei 75 C umkristallisiert, wobei hellgelbe, zerfliessliche, feste Kristalle entstunden (3, 3 g, 40 /o), Smp. 114-1160C (unter Zersetzung). (Gefunden : C, 31, 8 ; H, 6, 9 ; S, 6, 6 ; I, 52, 0 /o. Berechnet für C13H31NSI2: C, 32, 1 ; H, 6, 4 ; S, 6, 6 ; I, 52, 1 /o.)
Das als Ausgangsprodukt verwendete Methylthio butyldiäthylamin kann folgendermassen hergestellt werden :
4-Chlorbutyldiäthylaminhydrochlorid, das aus 4-Hydroxybutyldiäthylamin (11, 9 g) und Thionylchlorid (7, 4 ml) wie in Beispiel 1, angegeben wurde, hergestellt worden war, wurde in Athanol (40 ml) gelöst und im Verlauf von ungefähr 10 Minuten zu einer äthanolischen Lösung von Natriummethanthiol, welche eben auf Rückflusstemperatur erhitzt worden war, zugegeben. Verwendet wurden hierbei 147 ml einer Lösung, deren Herstellung in ¸Organic Syntheses¯, Coll. Vol. II, Seite 345 beschrieben ist und die ein Mol in 460 ml enthält. Das Gemenge wurde 2 Stunden lang am Rückfluss gekocht und hierauf der Alkohol so weitgehend wie möglich auf einem siedenden Wasserbad unter Normaldruck abgetrieben.
Der Rückstand wurde abgekühlt, mit Wasser (200 ml) verdünnt und mit Ather extrahiert (3mal 100 ml).
Die vereinigten Extrakte wurden mit Wasser gewaschen (3mal 40 ml), getrocknet (MgSO4), eingeengt und im Vakuum destilliert, wobei das 4-Methyl thiobutyldiäthylamin in Form eines farblosen Öls anfiel. (12, 1 g, 86%) Sdp. 108-112 C/19 mm, nD = 1, 4718. Der Smp. des Styphnates beträgt 71 bis 73 C, umkristallisiert aus Athanol.

Beispiel 3
1-¯thylmethylsulfonium-4-Ïthyldi-n propyl-ammoniumbutan-di-iodid
Zu einer Lösung von 4-Methyl-thiobutyl-di-npropylamin (2, 0 g) in trockenem Aceton (20 ml) wurde Athyliodid (4, 9 ml) zugegeben und das erhaltene Gemisch bei Raumtemperatur 2 Monate lang stehengelassen. Die überstehende Acetonschicht wurde abdekantiert, das ölige Produkt mit trockenem Aceton gewaschen (20 ml) und das Produkt mit einer weiteren Menge von trockenem Aceton (20 ml) mehrere Stunden lang stehengelassen, worauf es kristallisierte (2, 3 g, 45 /o). Smp. 101-104 C (u. Z.). Nach Umkristallisieren aus n-Propanol wurden farblose Kristalle erhalten.
Smp. 98-101 C (Zersetzung bei 112 C). (Gefunden : C, 35, 3 ; H, 7, 1 ; I, 49, 1% ; berechnet für C15H35NSI : C, 35, 0 ; H, 6, 8 ; I, 49, 3 /o).
Das als Ausgangsprodukt verwendete 4-Methylthiobutyl-di-n-propylamin kann folgendermassen hergestellt werden :
Umsetzung von B-Carbomethoxypropionylchlorid (1 Mol) mit di-n-Propylamin (2 Mol) in trockenem Ather ergab Methyl-N, N-di-n-propylsuccinamat als ein fast farbloses 01. Sdp. 96-100 C/0, 3 mm, nid 1, 4560. Aus dieser Verbindung wurde durch Reduktion mit Lithium-aluminiumhydrid (1, 5 Mol) das 4-Hydroxybutyl-di-n-propylamin als ein farbloses Öl erhalten. Sdp. 124 C/14 mm, nid = 1, 4489. Aus dem erhaltenen Produkt wurde durch Reaktion mit Thionylchlorid, wie in Beispiel 1 angegeben, das 4-Chlorbutyl-di-n-propylamin-hydrochlorid hergestellt.
Letzteres wurde mit Natriummethanthiol auf die im Beispiel 2 angegebene Weise umgesetzt, wobei 4-Methylthiobutyl-di-n-propylamin in Form eines farblosen Öls entstund. Sdp. 120-121¯C/10 mm, nid = 1, 4678 (gefunden : C, 64, 6 ; H, 12, 6 ; N, 7, 3 /o ; berechnet für Cl, H.,, NS : C, 65, 0 ; H, 12, 3 ; N, 6, 9 /o).
Beispiel 4 I-Diynethylsulf onium-4-methyl-di-isopropyl- aminoniumbutan-di-iodid
Zu einer Lösung von 4-Methylthiobutyl-di-isopropylamin (2, 0 g) in trockenem Aceton (20 ml) wurde Methyliodid (2, 5 ml) zugegeben und die erhaltene Mischung bei Raumtemperatur 8 Tage lang stehengelassen. Das erhaltene Produkt (4, 2 g) wurde abfiltriert und zweimal umkristallisiert, wobei beim erstenmal Isopropanol (33 ml), beim Sdp. verwendet wurde und hierauf Isopropanol (180 ml) bei 70 C. Es wurde ein gelbes, kristallines Pulver erhalten (1, 9 g, 39 /o). Smp. 100-104 C (Zersetzung).

Das als Ausgangsstoff verwendete 4-Methylthiobutyl-di-isopropylamin wurde folgendermassen hergestellt :
Umsetzung des -Carbomethoxypropionylehlorids (1 Mol) mit di-Isopropylamin (2 Mol) in trockenem Ather ergab Methyl-N, N-di-isopropylsuccinamat als fast farbloses Öl vom Sdp. 112-116 C/0, 05 mm.
Brechungsindex nD = 1, 4548. Aus dieser Verbindung wurde vermittels Reduktion mit Lithium-aluminiumhydrid (2 Mol) das 4-Hydroxybutyl-di-isopropylamin hergestellt. Es war ein farbloses öl vom Sdp.
110¯C/10mm, n19D = 1,4541. Aus 4-Hydroxybutyl-diisopropylamin und Thionylchlorid wurde nach der in Beispiel 1 angegebenen Arbeitsweise 4-Chlorbutyl-di isopropylamin-hydrochlorid hergestellt. Die letztere Verbindung wurde mit Natriummethanthiol, wie in Beispiel 2 beschrieben, umgesetzt, wobei 4-Methylthiobutyl-di-isopropylamin in Form eines farblosen Íls erhalten wurde. Sdp. 123 ¯C/13 mm, n22D = 1,4683.

Beispiel 5
1-¯thylmethylsulfonium-4-methyl-di-isopropyl ammoniumbutan-di-iodid
Zu einer Lösung von 4-Athylthiobutyl-di-isopropylamin (2, 2 g) in trockenem Aceton (20 ml) wurde Methyliodid (2, 5 ml) zugegeben und die erhaltene Mischung bei Raumtemperatur 24 Stunden lang stehengelassen. Das Reaktionsprodukt wurde abfil- triert (4, 5 g) zweimal als Isopropanol (10 ml) umkristallisiert und weiter gereinigt, indem es in Nitro methan (3 ml) gelöst und mit trockenem Aceton (10 ml) ausgefällt wurde. Die letztere Reinigungsoperation wurde dreimal ausgeführt und ergab hierauf zerfliessliche, farblose Mikroprismen (1, 0 g, 20"/o).
Smp. 140-141 C (Zersetzung). (Gefunden : C, 33, 8 ; H, 6, 8 ; S, 6, 1 ; I, 50, 5"/o ; berechnet für CggH33NSI2 C, 33, 5 ; H, 6, 6 ; S, 6, 4 ; I, 50, 6%.)
Das als Ausgangsprodukt verwendete 4-Äthylthiobutyl-di-isopropylamin wurde folgendermassen hergestellt :
4-Chlorbutyl-di-isopropylamin-hydrochlorid (siehe Beispiel 4) wurde mit Natriumäthanthiol in der in Beispiel 1 angegebenen Weise umgesetzt, wobei das 4-Athylthiobutyl-di-isopropylamin in Form eines farblosen Öls erhalten wurde. Sdp. 136-137 C/16 mm, nid = 1, 4657.

Beispiel 6
1-Methylisopropylsulfonium-4-methyl-di-isopropyl ammoniumbutan-di-iodid
Methyliodid (2, 5 ml) und 4-Isopropylthiobutyldi-isopropylamin (2, 3 g) wurden in Nitromethan (10 ml) 4 Tage lang miteinander reagieren gelassen.
Das Reaktionsprodukt wurde abfiltriert (3, 0 g) und zuerst als Nitromethan (4, 5 ml) auf einem siedenden Wasserbad und hierauf auf 96%igem Isopropanol (16, 5 ml) umkristallisiert, wobei farblose Stengel erhalten wurden (0, 9 g, 17 lo). Smp. 163-164¯C (Zersetzung). (Gefunden : C, 34, 6 ; H, 7, 1 ; S, 6, 1 ; I, 49, 7 /o ; berechnet für C15H33NSI2 : C, 34, 9 ; H, 6, 8 ; S, 6, 2 ; I, 49, 3 /0.)
Das als Ausgangsprodukt verwendete 4-Isopropylthiobutyl-di-isopropylamin wurde folgendermassen hergestellt :
4-Chlorbutyl-di-isopropylamin-hydrochlorid (siehe Beispiel 4) wurde mit Natriumisopropanthiol zur Reaktion gebracht, wobei 4-Isopropylthiobutyl-di-isopropylamin in Form eines farblosen Ols erhalten wurde.
Sdp. 133-136 C/10 mm, nD = 1, 4662.

1-DiÏthylsulfonium-5-triÏthylammoniumpentan di-iodid
Zu einer Lösung von
5-¯thylthioamyldiÏthylamin (10, 0 g) in trockenem Aceton (100 ml) wurde Athyliodid (420, 3 ml) zugegeben und die erhaltene Lösung nach Kochen auf dem Wasserbad während 8 Stunden unter Rühren abkühlen gelassen. Das erhaltene Produkt wurde abfiltriert und aus Athanol (168 ml) umkristallisiert, wobei farblose Plättchen erhalten wurden, (17, 5 g, 68 /o), Smp. 160-161¯C (Zersetzung).
(Gefunden : C, 35, 0 ; H, 6, 8 ; I, 49, 6% ; berechnet für C15H35NSI2 : C, 35, 0 ; H, 6, 8 ; I, 49, 3"/..)
Das als Ausgangsprodukt verwendete 5-Athylthioamyldiäthylamin wurde folgendermassen hergestellt :
Aus 5-Hydroxyamyldiäthylamin (vison, loc. cit.) wurde 5-Chloramyldiäthylamin-hydrochlorid (Chem.
Abs., 1934, 28, 1170) hergestellt und mit Natrium äthanthiol in der in Beispiel 1 angegebenen Weise zur Reaktion gebracht, wobei 5-¯thylthioamyldiÏthylamin als schwach strohgelbes 61, Sdp. 113 C/5 mm, nl3 = 1, 4702, erhalten wurde. (Gefunden : C, 65, 0 ; H, 12, 5 ; N, 7, 2% ; berechnet für CH25NS : C, 65, 0 ; H, 12, 3 ; N, 6, 9 o/0.)
Beispiel 16
Zu einer Lösung von 5-Methylthioamyl-di-npropylamin (2, 2 g) in trockenem Aceton wurde Athyliodid zugegeben und die erhaltene Lösung nach Kochen auf dem Wasserbad während 8 Stunden unter Rühren abkühlen gelassen.
Das erhaltene Produkt wurde abfiltriert und aus n-Propanol bei 90-95 C umkristallisiert, wobei farblose Plättchen erhalten wurden (3, 6 g, 68"/.) Smp. 126-127 C (Zersetzung).
(Gefunden : C, 36, 2 ; H, 7, 4 ; I, 48, 3 ; berechnet für C16H37NSI2 : C, 36, 2 ; H, 7, 0 ; I, 48, 0 11/o.) 5-Methylthioamyl-di-n-propylamin wurde folgendermassen erhalten :
Die Reaktion von γ-Carbomethoxybutyryl-chlorid (1 Mol) mit di-n-Propylamin (1 Mol) in Gegenwart von Triäthylamin (1 Mol) in getrocknetem Ather ergab Methyl-N, N-di-n-propylglutaramat als farbloses Íl, Sdp. 125 /0, 7 mm, nrz2 = 1, 4580.
Die Reduktion des letztern mit Lithium-aluminiumhydrid (1, 5 Mol) ergab 5-Hydroxyamyl-di-n-propylamin als farbloses 01, Sdp. 122-126 C/7 mm n15D = 1, 4545.
Diese Verbindung wurde über 5-Chloramyl-di-n- propylamin-hydrochlorid, wie in Beispiel 2 in 5-Methylthioamyl-di-n-propylamin, welches ein farbloses 01 darstellt, Sdp. 141¯C/13 mm, bergef hrt, nD = 1, 4645.

Process for the preparation of compounds having an antihypertensive effect
The present invention relates to a process for the preparation of compounds having an antihypertensive effect of the formula

wherein n is an integer from 4 to 6, R, Rl, R2, R3 and R4 are alkyl groups and X is an anion and the total number of carbon atoms in the cation is 11 to 16, characterized in that a corresponding di-N-substituted alkylthioalkylamine or a quaternary ammonium salt of such is reacted with an excess of a quaternizing agent. Compounds of this formula have a pronounced antihypertensive effect (as shown in animal experiments).
It was found here that the blood pressure-lowering effect is particularly attributable to those compounds in which n is 4 or 5.
The alkyl groups R, Ri, R2, R3 and R4 can be identical or different.
Among the compounds of the formula I, those with 13 to 15 carbon atoms in the cation have proven to be particularly effective.
In the following, the abbreviations Me, Et, Prll, Pr 'and Bun mean the methyl, ethyl, n-propyl, isopropyl and n-butyl groups.
The compound in which n = 4 and all radicals R are methyl, which according to Scheider and Kaufmann (Annalen., 1912, 392, 10) can be prepared by adding the amine CH ,. S- (CH,) ,. NH, completely methylated with methyl iodide in the presence of sodium methoxide, has only a slight antihypertensive effect. In contrast, the compound of the formula I, in which n = 4 and the radicals R to R4 denote the ethyl radical and the preparation of which is described in Example I, proves to be extremely effective in lowering blood pressure.
The nature of the anion X is of no particular importance. Any of the anions normally used for pharmaceutical purposes can be used as long as the compound in question has suitable physical properties (stability, solubility, etc.). Of course, toxic anions such as B. to avoid oxalation. The salts of organic acids can be prepared by reacting the halogen salts with silver salts of the suitable acids, or by allowing suitable acids to react in the presence of ethylene oxide (Sackur, Bull. Soc. Chim. 1952, 796).

1-methylisopropylsulfonium-4-methyl-diisopropylammonium butane, 1-dimethylsulfonium-4-methyl-di-n-propylammonium butane, 1-diethylsulfonium-5-triethylammonium pentane, and 1-ethylmethylsulfonium-5-ammonium-ethyl-di-n-pentanyl.
The term quaternizing agent as used in the foregoing refers to such reagents as e.g. B. methyl halides, which are capable of converting alkylated nitrogen atoms into the quaternary ammonium base or converting alkylated sulfur atoms into the tertiary sulfonium base.
The di-N-substituted alkylthioalkylamines, in which the total number of carbon atoms is 7 to 14, preferably 11-13, are new compounds and can be prepared by reacting suitable haloalkylamines in the form of their halo acid salts (usually the chloroalkylamines are used in the form of their hydrochloric acid salts ) are produced with the metal salt of an alkane thiol (usually used in excess) in a solvent, e.g. B. Ethanol (Brighton and Reid, J. Amer. Chem.
Soc., 1943, 65, 458; Andrews, Bergel and Morrison, J.C.S., 1953, 2998 and Kirchner, Soria and Cavallito, J. Chem. Soc. 1955, 77, 4599).
Another way to prepare di-N-substituted alkylthioalkylamines is to convert suitable di-N-substituted hydroxyalkylamines to the thioalkylamines by the action of hydrobromic acid and thiourea (Frank and Smith, J. Amer. Chem. Soc., 1946, 68, 2103 ; Plant, Tarbell and Whiteman, ibid, 1955, 77, 1572), whereupon the reaction mixture is made alkaline and the alkylation of the thiol group formed in this way is carried out using conventional methods.
The following reaction schemes illustrate how the named compounds can be prepared.

Example 1 1-DiÏthylsulfonium-4-triÏthylammoniumbutane-di-iodide a) Ethyl iodide (4, 9 ml) were added to a solution of 4-ethyl-thio-butyldiethylamine (1.9 g) in dry acetone (20 ml) added and the resulting mixture stirred under reflux on a water bath for 8 hours. After the first portion of a crystalline product (2.2 g) had been removed, mp 143 ° C. (u. Z.), the mother liquor was refluxed for a further 14 hours, a second portion of the reaction product (1.1 g) of mp. 139-140¯C (Z.). The combined yield (3.3 g) was recrystallized from ethanol (10 ml), whereby colorless prisms were obtained (2.9 g, 59 / o) mp = 142¯C (u.
Z.) (Found: C, 33, 4; H, 6, 5; I, 50, 3 0 / o, calcd for C 14 H 33 NSI ,,: C, 33, 5; H, 6, 6; I, 50 , 6 / o).
To prepare the dihydrogen tartrate, an aqueous solution of the diodide (1 mol) was shaken with a slurry of neutral silver tartrate (1 mol) in aqueous tartaric acid solution (1 mol).
The precipitated silver iodide was filtered off, the filtrate was concentrated in vacuo (water jet pump) and the semi-solid, white crystalline mass was dried in vacuo (without the use of a drying agent), a white crystalline powder (97 / o) having a melting point of 103-104 ° C accrued. Recrystallization from 95% ethanol resulted in clusters of colorless dihydrate needles with a melting point of 103-105 ° C.

Another method for converting the di-iodide into the di-hydrogen tartrate follows the procedure according to Sackur (loc. Cit.), According to which di-iodide (1 mol) and tartaric acid (3 mol) in a warm, concentrated alcoholic solution with ethylene oxide (4 moles) were treated. The di-hydrogen tartrate precipitated and was purified in the manner described above. b) A mixture of ethyl iodide (0.81 mol) and 4-¯thyl-thiobutyldiÏthylamine (1.9 g) was allowed to react for 7 days at room temperature without the presence of a solvent.
The solid substance obtained was digested with dry ether, filtered off and purified by dissolving it in acetone (5 ml) and reprecipitating it with dry ether (10 ml), the 4-ethyl-thiobutyltriethylammonium iodide being obtained in the form of colorless needles ( 2, 0 g, 58 / o). M.p. 95-96¯C. (Found: C, 42, 1; H, 8, 0; I, 36, 8% calculated for CI2H28NSI: C, 41, 7; H, 8, 1; I, 36, 8%.)
To a solution of 4-ethyl-thiobutyltriethylammonium iodide (1.8 g) in warm, dry acetone (10 ml) was added ethyl iodide (2.0 ml) and the resulting solution was refluxed on a water bath for 24 hours.
The solution was then cooled, the reaction product was filtered off (2.0 g, 76 / o) and recrystallized from ethanol.
Colorless prisms were obtained, m.p. 138 to 140 C (u. C.). (Found: I, 50, 6%.)
The 4-¯thyl-thiobutyldiÏthylamine used in this production process can be produced as follows:
A solution of thionyl chloride (97 ml) in 200 ml was added to a solution of 4-hydroxybutyldiethylamine (156 g) (vison, J. Appl. Chem. 1951, 1 469) in dry chloroform (160 ml) with stirring and with external cooling added to dry chloroform. (It is cooled with a carbonic acid bath.) The reaction mixture was allowed to warm to room temperature, refluxed for half an hour and then concentrated, first under normal pressure and finally in vacuo, in order to remove traces of chloroform.
The remaining oil was distilled off twice with dry benzene in an azeotropic mixture and then cooled, whereupon it crystallized out. The 4-chlorobutyl diethylamine hydrochloride obtained could be used directly in the next step of the synthesis without further purification.
Ethyl mercaptan (290 ml) was added to a solution of sodium (89 g) in ethanol (2 1) with cooling. The total amount of 4-chlorobutyldiethylamine hydrochloride in ethanol (550 ml) obtained above was added to the solution just heated to reflux temperature in the course of one hour. The mixture was then refluxed for 2 hours and finally the alcohol was distilled off as completely as possible under normal pressure on a boiling water bath. The residue was cooled, diluted with water (1.5 liters) and extracted with ether (3 times 1 liter).
The combined extracts were washed with water (3 times 250 ml), dried (MgSO4), concentrated and distilled in vacuo to give a colorless oil (172.8 g, 87%) b.p. 105-113 C / 10 mm, nD 1, 4705. (Found: C, 63, 6; H, 12, 3; p. 16, 9 / ol, calculated for C10H23NS: C, 63, 5; N, 12, 2; S, 16, 9% .)
The 4-mercaptobutyl diethylamine (161 g) prepared as described above was added to a chilled solution of sodium (23 g) in ethanol (460 ml), then ethyl bromide (120 g) was added and the mixture was refluxed for 3 hours .
The precipitated sodium bromide was filtered off from the reaction mixture, the filtrate was concentrated, filtered again and finally distilled in vacuo, the 4-ethylthiobutyldiethylamine being obtained in the form of a colorless oil (152 g, 80%.)
Example 2 1-Ethylmethylsulfonium-4-tridthyl-ammonium-butane-di-iodide
Ethyl iodide (8.4 ml) and 4-methylthiobutyl diethylamine (3.0 g) were allowed to react with one another in a solution of nitromethane (8.4 ml) for one week at room temperature. Dry ether (25 ml) was added and the oily product was allowed to settle and the supernatant layer of nitromethane ether was stripped off and replaced with acetone.
The product obtained was made to crystallize and then recrystallized twice from n-butanol (40 ml) at 75 ° C., resulting in pale yellow, deliquescent, solid crystals (3.3 g, 40 / o), m.p. 114-1160 ° C. (below Decomposition). (Found: C, 31, 8; H, 6, 9; S, 6, 6; I, 52, 0 / o. Calculated for C13H31NSI2: C, 32, 1; H, 6, 4; S, 6, 6 ; I, 52, 1 / o.)
The methylthio butyl diethylamine used as the starting product can be prepared as follows:
4-chlorobutyl diethylamine hydrochloride, which was prepared from 4-hydroxybutyl diethylamine (11.9 g) and thionyl chloride (7.4 ml) as indicated in Example 1, was dissolved in ethanol (40 ml) and added over about 10 minutes to an ethanolic solution of sodium methanethiol which had just been heated to reflux temperature. 147 ml of a solution were used here, the preparation of which is described in “Organic Syntheses”, Coll. Vol. II, page 345 and which contains one mole in 460 ml. The mixture was refluxed for 2 hours and the alcohol was then driven off as much as possible on a boiling water bath under normal pressure.
The residue was cooled, diluted with water (200 ml) and extracted with ether (3 times 100 ml).
The combined extracts were washed with water (3 times 40 ml), dried (MgSO4), concentrated and distilled in vacuo, the 4-methylthiobutyldiethylamine being obtained in the form of a colorless oil. (12.1 g, 86%) bp. 108-112 C / 19 mm, nD = 1.4718. The m.p. of the styphnate is 71 to 73 C, recrystallized from ethanol.
(Found: C, 42, 9; H, 5, 9; N, 13, 211 / o; calculated for C15H24N4O8S: C, 42, 8; H, 5, 7; N, 13, 3 / o.)
Example 3
1-ethylmethylsulfonium-4-ethyldi-n propylammoniumbutane-di-iodide
To a solution of 4-methyl-thiobutyl-di-npropylamine (2.0 g) in dry acetone (20 ml) was added ethyl iodide (4.9 ml) and the resulting mixture was allowed to stand at room temperature for 2 months. The supernatant acetone layer was decanted off, the oily product washed with dry acetone (20 ml) and the product allowed to stand with a further amount of dry acetone (20 ml) for several hours, whereupon it crystallized (2.3 g, 45 / o). M.p. 101-104 C (el.d.). After recrystallization from n-propanol, colorless crystals were obtained.
M.p. 98-101 C (decomposes at 112 C). (Found: C, 35, 3; H, 7, 1; I, 49, 1%; calculated for C15H35NSI: C, 35.0; H, 6, 8; I, 49, 3 / o).
The 4-methylthiobutyl-di-n-propylamine used as the starting product can be prepared as follows:
Reaction of B-carbomethoxypropionyl chloride (1 mol) with di-n-propylamine (2 mol) in dry ether gave methyl N, N-di-n-propyl succinamate as an almost colorless 01 bp 96-100 C / 0.3 mm, nid 1,4560. From this compound, reduction with lithium aluminum hydride (1.5 mol) gave 4-hydroxybutyl-di-n-propylamine as a colorless oil. Bp. 124 C / 14 mm, nid = 1.4489. 4-chlorobutyl-di-n-propylamine hydrochloride was prepared from the product obtained by reaction with thionyl chloride, as indicated in Example 1.
The latter was reacted with sodium methanethiol in the manner indicated in Example 2, 4-methylthiobutyl-di-n-propylamine being formed in the form of a colorless oil. Bp. 120-121¯C / 10 mm, nid = 1.4678 (found: C, 64, 6; H, 12, 6; N, 7, 3 / o; calculated for Cl, H. ,, NS: C , 65, 0; H, 12, 3; N, 6, 9 / o).
Example 4 I-Diynethylsulfonium-4-methyl-di-isopropyl-aminoniumbutan-di-iodid
To a solution of 4-methylthiobutyl-diisopropylamine (2.0 g) in dry acetone (20 ml) was added methyl iodide (2.5 ml) and the resulting mixture was allowed to stand at room temperature for 8 days. The product obtained (4.2 g) was filtered off and recrystallized twice, the first time using isopropanol (33 ml), the boiling point and then isopropanol (180 ml) at 70 ° C. A yellow, crystalline powder was obtained (1 , 9 g, 39 / o). M.p. 100-104 C (decomposition).

The 4-methylthiobutyl-di-isopropylamine used as the starting material was prepared as follows:
Reaction of the carbomethoxypropionyl chloride (1 mol) with di-isopropylamine (2 mol) in dry ether gave methyl N, N-di-isopropyl succinamate as an almost colorless oil with a bp. 112-116 C / 0.05 mm.
Refractive index nD = 1.4548. 4-Hydroxybutyldiisopropylamine was prepared from this compound by means of reduction with lithium aluminum hydride (2 mol). It was a colorless oil from the Sdp.
110¯C / 10mm, n19D = 1.4541. 4-chlorobutyldiisopropylamine hydrochloride was prepared from 4-hydroxybutyldiisopropylamine and thionyl chloride according to the procedure given in Example 1. The latter compound was reacted with sodium methanethiol as described in Example 2, 4-methylthiobutyl-di-isopropylamine being obtained in the form of a colorless oil. Bp. 123 ¯C / 13 mm, n22D = 1.4683.
(Found: C, 65.0; H, 12.3; N, 7.1; S, 15.6%; calculated for CllH23NS: C, 65.0; H, 12, 3; N, 6.9; S, 15, 8 / o.)
Example 5
1-ethylmethylsulfonium-4-methyl-di-isopropyl ammonium-butane-di-iodide
To a solution of 4-ethylthiobutyl-di-isopropylamine (2.2 g) in dry acetone (20 ml) was added methyl iodide (2.5 ml) and the resulting mixture was allowed to stand at room temperature for 24 hours. The reaction product was filtered off (4.5 g), recrystallized twice as isopropanol (10 ml) and further purified by dissolving it in nitro methane (3 ml) and precipitating it with dry acetone (10 ml). The latter cleaning operation was carried out three times and then resulted in deliquescent, colorless microprisms (1.0 g, 20 "/ o).

The 4-ethylthiobutyl-di-isopropylamine used as the starting product was prepared as follows:
4-chlorobutyl-di-isopropylamine hydrochloride (see Example 4) was reacted with sodium ethanethiol in the manner indicated in Example 1, the 4-ethylthiobutyl-di-isopropylamine being obtained in the form of a colorless oil. Bp. 136-137 C / 16 mm, nid = 1,4657.

Example 6
1-methylisopropylsulfonium-4-methyl-di-isopropyl ammonium butane di-iodide
Methyl iodide (2.5 ml) and 4-isopropylthiobutyldiisopropylamine (2.3 g) were allowed to react with each other in nitromethane (10 ml) for 4 days.
The reaction product was filtered off (3.0 g) and first recrystallized as nitromethane (4.5 ml) on a boiling water bath and then from 96% isopropanol (16.5 ml) to give colorless stalks (0.9 g, 17 lo). M.p. 163-164¯C (decomposition). (Found: C, 34, 6; H, 7, 1; S, 6, 1; I, 49, 7 / o; calculated for C15H33NSI2: C, 34, 9; H, 6, 8; S, 6, 2 ; I, 49, 3/0.)
The 4-isopropylthiobutyl-di-isopropylamine used as the starting product was prepared as follows:
4-chlorobutyl-di-isopropylamine hydrochloride (see Example 4) was reacted with sodium isopropanethiol, 4-isopropylthiobutyl-di-isopropylamine being obtained in the form of a colorless oil.
Bp. 133-136 C / 10 mm, nD = 1,4662.

To a solution of
5-ethylthioamyldiethylamine (10.0 g) in dry acetone (100 ml) was added to ethyl iodide (420.3 ml) and the resulting solution was allowed to cool with stirring after boiling on a water bath for 8 hours. Ethyl iodide was added to a solution of 5-methylthioamyl-di-npropylamine (2.2 g) in dry acetone and the resulting solution, after boiling on a water bath for 8 hours, allowed to cool with stirring.

This compound was converted via 5-chloramyl-di-n-propylamine hydrochloride, as in Example 2, into 5-methylthioamyl-di-n-propylamine, which is a colorless oil, bp 141 ° C / 13 mm, nD = 1,4645.
