Verfahren zur Herstellung von 94 5-Epoxy-5a-steroiden
Die vorliegende Erfindung betrifft ein Verfahren zur Herstellung von 9ss: 11ss-Epoxy-5a-steroiden aus 9a-Brom-l l-oxo-5a-steroiden.
Die 9ss : l l/ ?-Epoxy-5a-steroide können als Zwischenprodukte verwendet werden bei der Herstellung von 9-Fluor-bzw. 9-Jod-11-oxo-5a-steroiden, da man diese letzteren Verbindungen nicht durch direktes Halogenieren von 11-Oxo-5a-steroiden erhalten kann.
Man kann sie aber aus den entsprechenden 9fl : llp- Epoxy-Verbindungen herstellen nach der Methode von Fried und Szabo (J. A. C. S., 1953, 73, 2275 ; Ibid., 1954, 76, 1455).
Das erfindungsgemässe Verfahren ist dadurch gekennzeichnet, dal3 man ein 9a-Brom-11-oxo-5a- steroid mit einem Metallhydrid in Gegenwart eines inerten Lösungsmittels für das Metallhydrid und für das Steroid behandelt.
Geeignete Lösungsmittel sind also solche, in welchen sowohl das Steroidausgangsmaterial als auch das verwendete Metallhydrid löslich sind. Es ist natürlich klar, dass in diesem Falle nur solche Metallhydride für das Verfahren verwendet werden können, welche in einem inerten Lösungsmittel löslich sind, in dem auch das Steroidausgangsprodukt löslich ist.
Bei Verwendung von Lithiumaluminiumhydrid und Lithiumborhydrid als Metallhydride haben sich Di äthyläther und Tetrahydrofuran als besonders geeignete Lösungsmittel erwiesen.
Vorzugsweise soll die verwendete Metallhydridmenge in nicht mehr als etwa 10 /o Dberschuss zur theoretischen Menge vorliegen, da bei grösseren Mengen eine Reduktion des Epoxyds auftritt. Die Behandlung mit dem Metallhydrid wird gewöhnlich bei einer Temperatur im Bereich zwischen 0 und 80 C durchgeführt, und vorzugsweise bei erhöhter Temperatur, z. B. bei der Siedetemperatur des verwendeten Lösungsmittels.
Wenn ausser der 9a-Brom-und 11-Oxo-Gruppe noch andere reduktionsfähige Gruppen im Ausgangsmaterial vorhanden sind, so werden letztere während des Verfahrens normalerweise reduziert ; so werden Verbindungen, welche Aldehyd-oder Ketongruppen enthalten (zusätzlich zur 11-Oxo-Gruppe) normalerweise zu den entsprechenden Hydroxylverbindungen reduziert. In diesen Fällen mu# eine grössere Menge an Metallhydrid verwendet werden, als wenn keine solchen zusätzlichen reduktionsfähigen Gruppen vorhanden sind. Die in jedem speziellen Fall zu verwendende Metallhydridmenge kann durch einen Vorversuch leicht bestimmt werden.
Beispiel
3ss-Acetoxy-9ss: 11ss-epoxy-ergostan.
Eine Lösung von 376 mg 3-Acetoxy-9a-brom- ergostan-11-on und 22 mg Lithiumaluminiumhydrid (0, 83 Mol) in 20 cm3 Ather wird während einer Stunde unter Rückfluss erhitzt. Das Produkt wird mit Ather isoliert und, da die Behandlung mit Lithiumaluminiumhydrid eine Hydrolyse der 3ss-Acetoxygruppe des Ausgangsmaterials bewirkt, wird das Produkt zwecks Erleichterung der Isolierung wieder acetyliert und dann an einer entaktivierten Aluminiumoxydkolonne (30 g Au203) chromatographiert.
Elution mit 10, 1 cm3 Petroläther ergibt 150 mg 3, B-Acetoxy-9,, 6 :-epoxy-ergostan vom Schmelzpunkt 114-115 C (umkristallisiert aus Aceton Methanol). [a] D = + 23 ; gefunden : C 78, 35 H 11, 05 berechnet für C3OH5003 : C 78, 55 H 12, 06/o
Infrarotspektrum : keine Hydroxylbande
Obwohl vorstehend nur die Herstellung einer Acetyloxy-Verbindung beschrieben worden ist, ist das erfindungsgemässe Verfahren selbstverständlich auch anwendbar auf die Herstellung von anderen Verbindungen dieser Reihe, wie z. B. die Propionyloxy-, Butyloxy-, Phenylacetyloxy-und Benzyloxy-Verbindungen.

The present invention relates to a process for the production of 9ss: 11ss-epoxy-5a-steroids from 9a-bromo-l-oxo-5a-steroids.
The 9ss: l l /? -Epoxy-5a-steroids can be used as intermediates in the production of 9-fluoro or. 9-iodo-11-oxo-5a-steroids, since these latter compounds cannot be obtained by direct halogenation of 11-oxo-5a-steroids.
However, they can be prepared from the corresponding 9fl: llp epoxy compounds by the method of Fried and Szabo (J.A. C. S., 1953, 73, 2275; Ibid., 1954, 76, 1455).
The process according to the invention is characterized in that a 9a-bromo-11-oxo-5 asteroid is treated with a metal hydride in the presence of an inert solvent for the metal hydride and for the steroid.
Suitable solvents are therefore those in which both the steroid starting material and the metal hydride used are soluble. It is of course clear that in this case only those metal hydrides can be used for the process which are soluble in an inert solvent in which the steroid starting product is also soluble.
When using lithium aluminum hydride and lithium borohydride as metal hydrides, diethyl ether and tetrahydrofuran have proven to be particularly suitable solvents.
The amount of metal hydride used should preferably not be more than about 10 / o excess over the theoretical amount, since larger amounts reduce the epoxide. The treatment with the metal hydride is usually carried out at a temperature in the range between 0 and 80 ° C, and preferably at an elevated temperature, e.g. B. at the boiling point of the solvent used.
If, besides the 9a-bromo and 11-oxo groups, other groups capable of reduction are also present in the starting material, the latter are normally reduced during the process; thus compounds containing aldehyde or ketone groups (in addition to the 11-oxo group) are normally reduced to the corresponding hydroxyl compounds. In these cases, a larger amount of metal hydride must be used than if no such additional groups capable of reduction are present. The amount of metal hydride to be used in each specific case can easily be determined by a preliminary experiment.

3ss-acetoxy-9ss: 11ss-epoxy-ergostane.
A solution of 376 mg of 3-acetoxy-9a-bromergostan-11-one and 22 mg of lithium aluminum hydride (0.83 mol) in 20 cm3 of ether is refluxed for one hour. The product is isolated with ether and, since the treatment with lithium aluminum hydride causes hydrolysis of the 3ss-acetoxy group of the starting material, the product is re-acetylated to facilitate isolation and then chromatographed on a deactivated aluminum oxide column (30 g Au 2 O 3).
Elution with 10.1 cm3 of petroleum ether gives 150 mg of 3, B-acetoxy-9 ,, 6: -epoxy-ergostane with a melting point of 114-115 ° C. (recrystallized from acetone-methanol). [a] D = + 23; found: C 78.35 H 11.05 calcd for C3OH5003: C 78.55 H 12.06 / o

Although only the preparation of one acetyloxy compound has been described above, the inventive method is of course also applicable to the preparation of other compounds of this series, such as B. the propionyloxy, butyloxy, phenylacetyloxy and benzyloxy compounds.
