Description
Process for the preparation of 1-ascorbic acid from 2-keto-l-gulonic acid esters According to previous experience, esters of 2-keto-l-gulonic acid are converted in 1-ascorbic acid under the action of either acids or alkaline reactants Additives. Good yields were only obtained when the acids were in large Excess were used that this neutralizing all of the forming Ascorbic acid maintains an alkaline environment until it is terminated the implementation guaranteed. Even those under the influence of certain metal hydroxides Conversion of the esters which takes place requires at least one of these to be added equivalent amount.
It has now been found that neutral amine salts are also weaker Acids in alcoholic solution are able to convert esters of ketogulonic acid and that, surprisingly, much smaller amounts are required here. It it turned out that you can use small amounts, down to a tenth Equivalent, can achieve the same yields that would otherwise only be possible with one material higher expenditure on the additives which make the processing of the ascorbic acid difficult could be achieved.
It was further found that it is not even necessary to use foreign acids bring into the reaction mixture, but that this also includes amine salts Ascorbic acid itself are suitable.
The advance over known methods is that the Reaction mixture after the reaction only small amounts of amine or, if amine salts of ascorbic acid itself are used, no foreign constituents at all contains more.
It is known that keto-gulonic acid esters with equimolecular Quantities of alkali salts of weak acids in alcoholic solution with good yield allow to rearrange, but then all of the ascorbic acid falls in the form of its alkali salt at. It must therefore be set free from this retrospectively, which is the case with the Solubility properties of ascorbic acid is cumbersome to carry out.
Es bedeutet daher einen großen Vorteil, daß bei vorliegendem Verfahren etwa go °/o der anfallenden As= corbinsäure unmittelbar als freie Säure gewonnen werden können. It therefore means a great advantage that in the present method about 20% of the As = corbic acid obtained was obtained directly as free acid can be.
It has also been found to be particularly advantageous in which Method of using salts of secondary amines.
Beispiel i 2,o8 Teile 2-Keto-l-gulonsäuremethylester werden mit einer o,2i Teile (o,i Äquivalente) Triäthanolaminacetat enthaltenden alkoholischen Lösung in einem geschlossenen Gefäß 7 Stunden auf 8o bis go° erwärmt und das Reaktionsgemisch in üblicher Weise aufgearbeitet. Man erhält 1,25 Teile 1-Ascorbinsäure, entsprechend 71 °/o der Theorie.Example i 2, o8 parts of 2-keto-l-gulonic acid methyl ester are mixed with a 0.2i parts (0.1 equivalents) of triethanolamine acetate-containing alcoholic solution in a closed vessel for 7 hours to 8o to go ° heated and the reaction mixture worked up in the usual way. 1.25 parts of 1-ascorbic acid are obtained accordingly 71 per cent of theory.
Example 2 By heating a solution of 2.08 parts of methyl ketogulonate with 0.25 parts (0.1 equivalents) of diethylamine ascorbic acid in methyl alcohol, 1.62 parts of 1-ascorbic acid are obtained under the same conditions. The yield is 82% of the amount to be expected from the ester.
Hierbei entstehen 1,52 Teile (86 °/a der Theorie) 1-Ascorbinsäure.Example 3 2, o8 parts of keto-gulonic acid ester are in alcoholic Solution with 0.4 parts (0.3 equivalents) of diethylamine acetate for 7 hours to 8o to go ° warmed up. This produces 1.52 parts (86% of theory) of 1-ascorbic acid.
Beispiel 4 Aus 2,o8 Teilen Keto-gulonsäureester und o,2 Teilen (0,i Äquivalente) Diäthylaminbenzoat erhält man durch 6stündiges Erwärmen auf 8o bis go° 1,44 Teile (82 °/a der Theorie) 1-Ascorbinsäure.Example 4 From 2.08 parts of keto-gulonic acid ester and 0.2 parts (0.2 parts) Equivalents) diethylamine benzoate is obtained by heating to 8o for 6 hours go ° 1.44 parts (82 ° / a of theory) 1-ascorbic acid.
