Verfahren zur Herstellung neuer moschusartiger Geruchs-und Aromastoffe
Aus Arbeiten von M. Stoll [Manufacturing Perfumer 1, 107 (1937)] ist bekannt, dass makrocyclische Verbindungen mit 14-19 Ringgliedern, wie Lactone und Ketone, einen starken reinen Moschusgeruch besitzen. Infolgedessen finden Verbindungen diesen Typs ausgedehnte Anwendung als Parfums.
Eine andere Gruppe von Verbindungen, die auf Grund ihres Moschusgeruchs von grosser Bedeutung ist, ist die der aromatischen Nitroverbindungen (vgl.
T. F. West, H. J. Strausz und D. R. H. Barton, Synthetic. Perfumes 1949, Seite 297). Die wichtigsten Vertreter dieser Gruppe sind 2,4,6-Trinitro-1,3dimethyl-5-tert.-butyl-benzol (Xylolmoschus), 3,5-Dinitro-2,6-dimethyl-4-tert.-butyl-acetophenon (Ketonmoschus) und 3-Methoxy-2,6-dinitro-4-tert.-butyltoluol (Ambrettemoschus).
Die makrocyclischen Verbindungen besitzen den Nachteil, dass die meisten Vertreter nur auf kompliziertem Wege erhältlich sind, wodurch ihr Preis im allgemeinen sehr hoch liegt. Sie besitzen jedoch einen sehr feinen Geruch und ausgezeichnete Haftfähig- keit und sind ausserordentlich licht-und alkalibeständig.
Die Nitroverbindungen dagegen können zu niederem Preis hergestellt werden, besitzen jedoch einen viel gewöhnlicheren Geruch als die makrocyclischen Verbindungen und ihre Neigung zur Verfärbung, insbesondere wenn sie nicht ausserordentlich hochgradig gereinigt sind, ist als ein Nachteil anzusehen.
Es ist deshalb wünschenswert, Verbindungen zu erhalten, die einerseits den feinen reinen Moschusgeruch und dessen Aroma, die Haftfähigkeiten und die Stabilität der makrocyclischen Verbindungen besitzen und anderseits auf einfache Weise hergestellt werden können.
Zwei Veröffentlichungen lassen den Schluss zu, dass in dieser Richtung nach stickstofffreien aromatischen Verbindungen mit den gewünschten Eigen- schaften gesucht wurde.
M. S. Carpenter, W. M. Easter und T. F. Wood [Journal of Organic Chemistry 16,586 (1951)] beschreiben eine stickstofffreie aromatische Verbindung mit Moschusgeruch, nämlich den 2,4-Di-tert.-butyl5-methoxy-benzaldehyd.
In der niederländischen Patentschrift Nr. 74524 ist eine Anzahl von Verbindungen mit Moschusgeruch beschrieben, die durch Einführung einer Alkylgruppe mit 4-6 Kohlenstoffatomen und einer Acetylgruppe in meta-oder para-Cymol erhalten wurden.
Die vorliegende Erfindung betrifft ein Verfahren zur Herstellung neuer moschusartiger Geruchs-und Aromastoffe der Formel

Beispiel 1
4-Acetyl-6-tert.-butyl-1, 1-dimethyl-indan
Einer Mischung aus 1423 g konzentrierter Schwefelsäure und 76,5 g Eis werden in 10 Minuten bei 15 730 g 1, 1-Dimethyl-indan und hierauf eine Mischung aus 185 g tert.-Butanol und 30 g 1, 1-Di- methyl-indan in 11/4 Stunden bei 10 zugesetzt. Man rührt noch 1142 Stunden bei 10-17 und arbeitet auf. Es wird im Vakuum mittels einer geeigneten Kolonne fraktioniert. Das tert.-Butylierungsprodukt vom Kp 2 5 = 90-97 , n D = 1,5080-1,5100 wird in einer Ausbeute von 91,2 /o der Theorie erhalten.
Einer Mischung aus 4780 g Tetrachlorkohlenstoff, 565 g (7,2 Mol) Acetylchlorid und 1212 g Dimethyl-tert.-butyl-indan (in der oben erwähnten Weise hergestellt) werden bei 0-5 in 4 Stunden 881 g Aluminiumchlorid (6,6 Mol) zugesetzt. Man rührt weitere 2 Stunden bei der gleichen Temperatur und arbeitet auf. Nach Fraktionieren und Kristallisation wird erhalten :
1. ein reines kristallisiertes Hauptprodukt vom F. = 77,2-77,7 in einer Ausbeute von etwa 55 /o der Theorie, welches einen reinen starken Moschusgeruch vom Pentadecanolidtyp zeigt ; es handelt sich um 4-Acetyl-1, 1-dimethyl-6-tert.-butyl-indan ;
2. eine sehr geringe Menge eines Isomeren, wahrscheinlich 6-Acetyl-1, 1-dimethyl-4-tert.-butyl- indan vom F. = 105,2-106,0 , das ebenfalls einen starken pentadecanolidähnlichen Geruch besitzt.
Beispiel 2 4-Propionyl-6-tert.-butyl-1, 1-dimethyl-indan
5000 g Tetrachlorkohlenstoff, 610,5 g (6,6 Mol) Propionylchlorid und 1212 g (6 Mol) 1, 1-Dimethyl- tert.-butyl-indan werden in einen 6-Liter-Reaktionskolben eingebracht. 881 g Aluminiumchlorid werden unter Rühren bei 3-5 in etwa 4 Stunden zugesetzt.
Man rührt weitere 2 Stunden, giesst auf eine Mischung von Eis und Salzsäure und arbeitet auf.
Das Reaktionsprodukt wird im Vakuum mit einer 16bödigen Kolonne fraktioniert und die kristallisierenden Fraktionen durch Umkristallisation gereinigt.
272 g eines flüssigen Vorlaufes mit praktisch vollständig konstantem Siedepunkt (103-104 bei 0,2 mm) und einem konstanten Brechungsindex (nid = 1,5340) werden erhalten. Diese Verbindung hat einen Carbonylgehalt von 4,84 m Aquiv./g und ist wahrscheinlich Propionyl-dimethyl-indan. Ausbeute 22,4"/ & der Theorie.
Als Hauptprodukt werden weiterhin 52,4 /o der theoretisch möglichen Menge an Propionyl-dimethyltert.-butyl-indan vom F. = 45,1-45, 9 erhalten.
Diese Verbindung besitzt einen starken Moschusgeruch.
Schliesslich wird noch eine kleine Menge einer isomeren Verbindung vom F. = 59-60 , die ebenfalls Moschusgeruch besitzt, erhalten.
Beispiel 3 4-Acetyl-l, l-dimethyl-6-tert.-butyl-indan 38 g Natrium werden in 400 g absolutem Methanol gelöst und 512 g Diäthyl-malonat in 10 Minuten bei 40 zugesetzt. Man kühlt auf 24 ab und fügt unter Rühren in 11, ; Stunden 292 g 4-tert.-Butyl- benzylchlorid zu. Dann lässt man die Mischung bei Raumtemperatur stehen, bis die Reaktion beendet ist.
Die Verseifung wird durch Abdestillieren des Athanols und allmählicher Zugabe von 960 g 33/oiger Natronlauge und so viel Wasser, als notwendig ist, um die Masse in rührfähigem Zustand zu halten, ausgeführt.
Man rührt noch 2 Stunden in der Siedehitze und säuert mit verdünnter Schwefelsäure in der Hitze an.
Dann trennt man die obere Schicht und decarboxyliert in bekannter Weise. Die rohe 4-tert.-Butyl phenyl-propionsäure wird durch Destillation im Vakuum und Umkristallisieren aus Petroläther gereinigt. F. = 110-112 ; Ausbeute=74 /o derTheorie.
217 g 4-tert.-Butyl-phenylpropionsäure werden in eine Mischung aus 640 g Methanol und 40 g Schwefelsäure eingebracht und 5 Stunden gekocht.
Nach Aufarbeiten wird der Methylester fraktioniert.
Die Ausbeute ist praktisch quantitativ. Kp3= 128 .
Erstarrungspunkt 40 .
In bekannter Weise wird eine Lösung von Methylmagnesiumbromid aus 58,3 g Magnesium hergestellt und zu dieser 222 g Methyl-4-tert.-butyl-phenyl- propionat in 3 Stunden bei 0-4 unter Rühren zugesetzt. Nach Aufarbeitung wird fraktioniert.
Kp3 = 136-138 ; n2r, = 1,5030-1,5038 ; Erstarrungspunkt etwa 40 .
207 g 3-Methyl-l- (4-tert.-butyl-phenyl)-butanol- (3) werden unter Rühren in einer Stunde bei 0-5 einer Mischung aus 540 g Schwefelsäure und 66 g Wasser zugesetzt. Man rührt noch eine Stunde bei der gleichen Temperatur weiter und anschliessend noch 11/o Stunden bei 10-15 . Nach Aufarbeiten werden 158 g 1, 1-Dimethyl-6-tert.-butyl-indan erhalten. Kp4 = 97 ; nid = 1, 5070.
Zu 119 g Aluminiumchlorid und 80 g Acetylchlorid in 600 g Tetrachlorkohlenstoff werden in 2 Stunden bei 0-5 158 g 1, 1-Dimethyl-6-tert.- butyl-indan zugesetzt. Man rührt noch eine Stunde bei der gleichen Temperatur und arbeitet in üblicher Weise auf. Das Reaktionsprodukt wird schnell im Vakuum ohne Kolonne destilliert. Es kristallisiert vollständig und wird aus Methanol umkristallisiert.
F. (korr.) = 76,7-77,2 ; Ausbeute 125 g = 65 oxo der Theorie.
Ein Mischschmelzpunkt mit dem nach Beispiel 1 hergestellten ProdUkt zeigt keine Depression. Die Verbindung besitzt einen starken Geruch, der stark an denjenigen des Pentadecanolids erinnert.
Beispiel 4 4-Acetyl-6-tert.-butyl-1, 1-dimethyl-indan
800 g Tetrachlorkohlenstoff, 94 g Acetylchlorid (1,2 Mol) und 202 g (1 Mol) 1, 1-Dimethyl-5-tert.- butyl-indan (erhalten durch Fraktionierung des in Beispiel 1 hergestellten 1, 1-Dimethyl-tert.-butyl- indans) werden in einen Reaktionskolben eingebracht.
147 g (1,1 Mol) Aluminiumchlorid werden unter Rühren bei 0 bis-5 in 3 Stunden zugesetzt. Man rührt weitere 2 Stunden, giesst auf eine Mischung von Eis und Salzsäure und arbeitet auf.
Das Reaktionsprodukt wird schnell im Vakuum destilliert und mit einer 12bödigen Kolonne frak tioniert. Man bekommt 60 g Acetyl-1, 1-dimethylindan (n20= 1,5390) und man reinigt die kristallisierenden Fraktionen durch Umkristallisation.
Ausbeute 66 g (27 /o) 4-Acetyl-l, l-dimethyl-6- tert.-butyl-indan (Schmelzpunkt : 76,6-77,0O).
Aus den Mutterlaugen bekommt man noch 26 g (Smp. 53,5-68 ) einer Mischung von isomeren Verbindungen, die ausser 4-Acetyl-1, 1-dimethyl-6-tert.butyl-indan (Hauptprodukt) auch das höherschmelzende Isomere enthält.
Beide können mit einer 16bödigen Kolonne fast vollständig getrennt werden.
Process for the production of new musky fragrances and aromas
It is known from work by M. Stoll [Manufacturing Perfumer 1, 107 (1937)] that macrocyclic compounds with 14-19 ring members, such as lactones and ketones, have a strong, pure musk odor. As a result, compounds of this type find extensive use as perfumes.
Another group of compounds that is of great importance due to their musky odor is that of the aromatic nitro compounds (cf.
T. F. West, H. J. Strausz, and D. R. H. Barton, Synthetic. Perfumes 1949, p. 297). The most important representatives of this group are 2,4,6-trinitro-1,3dimethyl-5-tert-butyl-benzene (xylene musk), 3,5-dinitro-2,6-dimethyl-4-tert-butyl-acetophenone (Ketone musk) and 3-methoxy-2,6-dinitro-4-tert.-butyltoluene (ambrette musk).
The macrocyclic compounds have the disadvantage that most representatives can only be obtained in a complicated way, which means that their price is generally very high. However, they have a very fine odor and excellent adhesiveness and are extremely light and alkali-resistant.
The nitro compounds, on the other hand, can be produced at a lower price, but have a much more common odor than the macrocyclic compounds and their tendency to discolour, especially if they are not extremely highly purified, is to be regarded as a disadvantage.
It is therefore desirable to obtain compounds which, on the one hand, have the fine, pure musk odor and its aroma, the adhesiveness and stability of the macrocyclic compounds and, on the other hand, can be produced in a simple manner.
Two publications allow the conclusion that nitrogen-free aromatic compounds with the desired properties were sought in this direction.
M. S. Carpenter, W. M. Easter and T. F. Wood [Journal of Organic Chemistry 16,586 (1951)] describe a nitrogen-free aromatic compound with a musky odor, namely 2,4-di-tert-butyl5-methoxy-benzaldehyde.

4-acetyl-6-tert-butyl-1,1-dimethyl-indane
A mixture of 1423 g of concentrated sulfuric acid and 76.5 g of ice are added in 10 minutes at 15,730 g of 1,1-dimethylindane and then a mixture of 185 g of tert-butanol and 30 g of 1,1-dimethyl indan added in 11/4 hours at 10. The mixture is stirred for a further 1142 hours at 10-17 and worked up. It is fractionated in vacuo using a suitable column. The tert-butylation product with a boiling point of 2.5 = 90-97, n D = 1.5080-1.5100 is obtained in a yield of 91.2 / o of theory.
A mixture of 4780 g of carbon tetrachloride, 565 g (7.2 mol) of acetyl chloride and 1212 g of dimethyl-tert-butyl-indane (prepared in the above-mentioned manner) gives 881 g of aluminum chloride (6.6 Mol) added. The mixture is stirred for a further 2 hours at the same temperature and worked up. After fractionation and crystallization the following is obtained:
1. a pure crystallized main product of m.s. = 77.2-77.7 in a yield of about 55 / o of theory, which shows a pure strong musk odor of the pentadecanolide type; it is 4-acetyl-1, 1-dimethyl-6-tert-butyl-indane;
2. a very small amount of an isomer, probably 6-acetyl-1,1-dimethyl-4-tert.-butylindane with a melting point of 105.2-106.0, which also has a strong pentadecanolide-like odor.
Example 2 4-Propionyl-6-tert-butyl-1,1-dimethyl-indane
5000 g of carbon tetrachloride, 610.5 g (6.6 mol) of propionyl chloride and 1212 g (6 mol) of 1,1-dimethyl-tert-butyl-indane are placed in a 6-liter reaction flask. 881 g of aluminum chloride are added with stirring at 3-5 in about 4 hours.
The mixture is stirred for a further 2 hours, poured onto a mixture of ice and hydrochloric acid and worked up.
The reaction product is fractionated in vacuo with a 16-tray column and the crystallizing fractions are purified by recrystallization.
272 g of a liquid forerun with a practically completely constant boiling point (103-104 at 0.2 mm) and a constant refractive index (nid = 1.5340) are obtained. This compound has a carbonyl content of 4.84 m equiv / g and is probably propionyl-dimethyl-indane. Yield 22.4 "/ & of theory.
The main product still obtained is 52.4 / o of the theoretically possible amount of propionyl-dimethyltert-butyl-indane with a melting point of 45.1-45.9.
This compound has a strong musky odor.
Finally, a small amount of an isomeric compound of F. = 59-60, which also has a musky odor, is obtained.
Example 3 4-Acetyl-1,1-dimethyl-6-tert-butyl-indane 38 g of sodium are dissolved in 400 g of absolute methanol and 512 g of diethyl malonate are added at 40 in 10 minutes. The mixture is cooled to 24 and, while stirring, 11,; Hours 292 g of 4-tert-butylbenzyl chloride. The mixture is then left to stand at room temperature until the reaction has ended.
The saponification is carried out by distilling off the ethanol and gradually adding 960 g 33% sodium hydroxide solution and as much water as is necessary to keep the mass in a stirrable state.
The mixture is stirred for a further 2 hours at the boiling point and acidified with dilute sulfuric acid in the heat.
The upper layer is then separated and decarboxylated in a known manner. The crude 4-tert-butyl phenyl propionic acid is purified by distillation in vacuo and recrystallization from petroleum ether. F. = 110-112; Yield = 74 / o of theory.
217 g of 4-tert-butyl-phenylpropionic acid are introduced into a mixture of 640 g of methanol and 40 g of sulfuric acid and boiled for 5 hours.
After working up, the methyl ester is fractionated.
The yield is practically quantitative. Kp3 = 128.
Freezing point 40.
In a known manner, a solution of methyl magnesium bromide is prepared from 58.3 g of magnesium and 222 g of methyl 4-tert-butyl-phenyl-propionate are added to this in 3 hours at 0-4 with stirring. After working up, it is fractionated.
Bp3 = 136-138; n2r, = 1.5030-1.5038; Freezing point about 40.
207 g of 3-methyl-1- (4-tert-butyl-phenyl) -butanol- (3) are added to a mixture of 540 g of sulfuric acid and 66 g of water over the course of one hour at 0-5 with stirring. The mixture is stirred for a further hour at the same temperature and then for 11 / o hours at 10-15. After working up, 158 g of 1,1-dimethyl-6-tert-butyl-indane are obtained. Bp4 = 97; nid = 1,5070.
To 119 g of aluminum chloride and 80 g of acetyl chloride in 600 g of carbon tetrachloride, 158 g of 1,1-dimethyl-6-tert-butyl-indane are added over 2 hours at 0-5. The mixture is stirred for another hour at the same temperature and worked up in the usual way. The reaction product is quickly distilled in vacuo without a column. It crystallizes completely and is recrystallized from methanol.
F. (corr.) = 76.7-77.2; Yield 125 g = 65 oxo of theory.
A mixed melting point with the product prepared according to Example 1 shows no depression. The compound has a strong odor that is strongly reminiscent of that of pentadecanolide.
Example 4 4-Acetyl-6-tert-butyl-1,1-dimethyl-indane
800 g of carbon tetrachloride, 94 g of acetyl chloride (1.2 mol) and 202 g (1 mol) of 1,1-dimethyl-5-tert-butyl-indane (obtained by fractionating the 1,1-dimethyl-tert prepared in Example 1) .-Butyl-indane) are placed in a reaction flask.
147 g (1.1 mol) of aluminum chloride are added with stirring at 0 to -5 in 3 hours. The mixture is stirred for a further 2 hours, poured onto a mixture of ice and hydrochloric acid and worked up.
The reaction product is quickly distilled in vacuo and fractionated with a 12-tray column. 60 g of acetyl-1,1-dimethylindane (n20 = 1.5390) are obtained and the fractions which crystallize are purified by recrystallization.
Yield 66 g (27 / o) of 4-acetyl-1,1-dimethyl-6-tert-butyl-indane (melting point: 76.6-77.0O).
The mother liquors still give 26 g (melting point 53.5-68) of a mixture of isomeric compounds which, in addition to 4-acetyl-1,1-dimethyl-6-tert-butylindane (main product), also contains the higher melting isomer .
Both can be almost completely separated with a 16-tray column.
