Verfahren zur Isolierung von d',4-3-Keto-steroiden Es hatte sich gezeigt, dass die Einführung einer Doppelbindung in die 1,2-Stellung von Cortison und Cortisol (und ihren Estern) die therapeutische Wirk samkeit dieser Verbindungen erhöht. Nach Bekannt werden dieser Entdeckung wurden eingehende Unter suchungen zur Herstellung von Cortison-Analogen und andern Steroiden mit ähnlicher therapeutischer Wirksamkeit durchgeführt, welche Stoffe alle eine Doppelbindung in der 1,2-Stellung besitzen.
Ausser Cortison und seinen Analogen sind auch noch an dere Al.4-3-Keto-steroide von Interesse.
Es wurden verschiedene Methoden für die Syn these von JI.4-3-Keto-steroiden vorgeschlagen. Eine dieser Methoden besteht darin, aus 2,4-Dibrom-3-keto- steroiden Bromwasserstoff abzuspalten, z. B. mittels einer tertiären Base, wie Collidin. Eine geeignete und Erfolg versprechende Methode dieser Art wäre von grossem Wert, insbesondere in den Allo-Reihen, da 2,4-Dibrom-3-keto-5a-steroide nach Methoden, wie sie für die Synthese von Cortison verwendet werden, leicht herzustellen sind.
Es wurde jedoch gefunden, dass eine solche Bromwasserstoffabspaltung immer zu Gemischen führt, welche das gewünschte 41,4-3- Keto-steroid in mehr oder weniger grossen Mengen zusammen mit JI-, d4- und/oder J4,6-3-Keto-steroiden als Verunreinigungen enthalten, je nach dem Erfolg der Bromwasserstoffabspaltung.
Es wurden nun Versuche unternommen, solche Gemische mit den üblichen Methoden, wie Chroma- tographie und fraktionierte Kristallisation, zu tren nen, aber es wurden keine Erfolge damit erzielt.
Andere Methoden für die Herstellung von d1,4-3- Keto-steroiden ergeben ebenfalls Gemische, welche diese Steroide zusammen mit JI-, 44- und/oder J4.6-3_Keto-steroiden enthalten.
Es wurde nun gefunden, dass gesättigte 3-Keto- steroide, :A'- und J4-3-Keto-steroide sowie auch A4,6_3_Keto-steroide mit den Girard-Reagenzien T und P Derivate ergeben, wogegen die 4l.4-3-Keto- steroide keine solchen Derivate geben, sofern sie ausser der Ketogruppe in 3-Stellung keine weiteren Carbonylgruppen enthalten oder nur solche, welche mit Girard-Reagens T oder P nicht reagieren. Es ist z.
B. wohlbekannt, dass eine Ketogruppe in der 11- Stellung des Steroidringsystems mit Girard-Reagenzien nicht reagiert, was auch für eine Ketogruppe in der 20-Stellung gilt, wenn diese sterisch gehindert ist; die Ketogruppe in der 20-Stellung des Cortisons ist reak tionsfähig, während sie bei 21-Estern des Cortisons. nicht reaktionsfähig ist.
Diese Entdeckung gestattet nun, Gemische von dl.4-3-Keto-steroiden mit gesät tigten 3-Keto-steroiden und den genannten ungesät tigten 3-Keto-steroiden leicht zu trennen. Es soll in diesem Zusammenhang erwähnt sein, dass diese Ent deckung sehr überraschend ist, da der Anmelderin kein anderes 3-Keto-steroid, das kein Girard-Derivat gibt, bekannt ist, und es muss deshalb vermutet wer den, dass dies eine Folge des Einflusses der beiden benachbarten Doppelbindungen ist.
Gegenstand der Erfindung ist somit ein Verfahren zur Isolierung von d1,4-3_Keto-steroiden, welche ausser der Ketogruppe in 3-Stellung entweder keine weiteren reaktionsfähigen Carbonylgruppen enthalten oder nur solche, welche mit Girard-Reagens T oder P nicht reagieren, aus Gemischen, welche solche d1,4-3-Keto-steroide zusammen mit 3-Keto-, J'-3- Keto-, J4-3-Keto- und/oder J4.6-3-Keto-steroiden enthalten.
Das Verfahren ist dadurch gekennzeich net, dass man das Gemisch in einem Lösungsmit tel mit Girard-Reagens T oder P umsetzt, wobei sich Girard-Derivate der genannten andern Steroide bil den, und dass man das Jl,4_3-Keto-steroid unter Aus nützung seiner Schwerlöslichkeit in Wasser aus dem Reaktionsgemisch entfernt.
Das erfindungsgemässe Verfahren ist besonders geeignet für die Isolierung von JI,4-3-Keto-steroiden der Formel

worin R1 eine Keto- oder Hydroxylgruppe, R.= Wasserstoff oder Halogen, insbesondere Fluor, R, eine Acylgruppe und R4 Wasserstoff oder eine Acyl- gruppe bedeutet. Bevorzugte Acylgruppen sind die jenigen, welche sich von Karbonsäuren ableiten, z. B.
die Acetyl-, Propionyl-, Benzoyl- oder Phenylacetyl- Gruppe. Beispiele von bevorzugten Verbindungen der vorstehenden Formel sind Ester von 1,2-Dehydro- cortison und 1,2-Dehydro-cortisol, insbesondere die 2l Acetate und 17,21-Diacetate davon.
Das erfindungsgemässe Verfahren wird in der Regel so durchgeführt, dass man das Ausgangs gemisch der Steroide mit Girard-Reagens P oder T in einem geeigneten Lösungsmittel erhitzt, z. B. in einer Mischung von Äthanol und Essigsäure, und darauf die Abtrennung des JI,4_3-Keto-steroids in irgendeiner gewünschten Weise vornimmt. Da die gebildeten Girard-Derivate wasserlöslich sind, kön nen sie leicht aus dem resultierenden Gemisch extra hiert werden, weil JI>4-3-Keto-steroide in Wasser un löslich sind.
Wenn das für die Herstellung der Deri vate verwendete Lösungsmittel mit Wasser nicht mischbar ist, verbleiben die dI,4-3-Keto-steroide nach der Zugabe von Wasser in der organischen Phase und können daraus leicht gewonnen werden. Wenn jedoch das für die Bereitung der Girard- Derivate verwendete Lösungsmittel mit Wasser mischbar ist, ist es in der Regel notwendig, die JI.4_3_Keto-steroide mittels eines mit Wasser nicht mischbaren Lösungsmittels nach der Zugabe von Wasser zu extrahieren. In der Praxis ist es zweck mässig, nach der Bildung der Girard-Derivate dem Reaktionsgemisch eine verdünnte wässrige Lösung eines schwachen Alkalis zuzusetzen, wie z.
B. eine wässrige Natriumbicarbonatlösung, und dann das A'4_3_Keto-steroids zu extrahieren.
Geeignete mit Wasser nicht mischbare Lösungs mittel für die Extraktion sind z. B. Ester, wie Äthyl- acetat, sowie Methylenchlorid.
Das J1.4-3-Keto-steroid kann aus dem Extrakt in irgendeiner üblichen Weise abgetrennt werden, z. B. durch Destillation des Lösungsmittels. Es kann anschliessend durch Umkristallisieren gereinigt wer den, wenn nötig nach vorangehender Behandlung mit Holzkohle. Die Erfindung ist insbesondere anwendbar auf die Trennung von Steroidgemischen, wie sie bei. der Bromwasserstoffabspaltung von 2,4-Dibrom-3-keto- steroiden, insbesondere 2,4 - Dibrom - 3 - keto - 5a - steroiden, entstehen.
Eine besonders vorteilhafte Methode für die Bromwasserstoffabspaltung aus 2,4-Dibrom-3-keto- 5a-steroiden besteht im Erhitzen dieser Steroide mit Semicarbazid oder seinen Salzen in substituierten Amiden der allgemeinen Formel W-CO-NR"RW", worin R' ein Wasserstoffatom oder eine aliphatische Gruppe mit bis zu 6 C-Atomen bedeutet,
während R" und R"' Alkylgruppen mit 1 bis 4 C-Atomen oder zusammen Bestandteil eines alicyclischen oder heterocyclischen Ringes sind.
Als primäres Reaktionsprodukt entsteht dabei das Semicarbazon des J1.4-3-Keto-steroids. Letzteres kann nach irgendeiner bekannten Methode zum freien Keton zersetzt werden, z. B. durch Erhitzen während 11/2 Stunden in einer wässrigen Mischung von Essigsäure und Brenztraubensäure. Auf diese Weise erfolgt die Umwandlung in das entsprechende J1,4-3-Keto-steroid glatt und mit guter Ausbeute.
Diese Bromwasserstoffabspaltungsmethode ist insbesondere auf Steroide mit einer empfindlichen Seitenkette am Cl7-Atom anwendbar und ist deshalb sehr geeignet für die Umwandlung von 2,4-Dibrom- 4,5a-dihydro-cortisonestern in die 1,2-Dehydro- cortisonester.
Als Reaktionsmedium für die Bromwasserstoff- abspaltung sind insbesondere folgende substituierten Amide geeignet: Dimethylacetamid, N,N-Dimethyl-formamid, N,N-Dimethyl-valeriansäureamid, N,N-Diätyhl-formamid, N,N-Dipropyl-formamid, N,N-Dibutyl-formamid, N-Formyl-piperidin und N-Formyl-morpholin. Das Semicarbazid kann in Form der freien Base oder in Form eines seiner Salze verwendet werden, z. B. als Hydrochlorid, Sulfat oder Acetat.
<I>Beispiel 1</I> 5,25 g eines Gemisches von 3-Keto-steroidell, 2 g Girard-Reagens P, 50 cm- Äthanol und 5 cm33 Essigsäure wurden während 15 Minuten am Rück fluss erhitzt, abgekühlt und in wässrige Natrium- bicarbonatlösung gegossen.
Nach Extraktion mit Äthylacetat und Entfernung des Lösungsmittels unter verringertem Druck erhielt man 3,66 g eines Pro duktes mit

Dieses Produkt wurde in Pyridin gelöst und wäh rend 16 Stunden bei Zimmertemperatur mit Acet- anhydrid acetyliert. Hierauf wurde Wasser zugege ben und das Produkt mit Methylenchlorid extrahiert, gewaschen, getrocknet und unter vermindertem Druck eingedampft, wobei 5,25 g rohes 1,2-Dehydro- cortison-acetat erhalten wurden.
<I>Beispiel 2</I> Ein rohes Gemisch, welches erhalten wurde durch Bromwasserstoffabspaltung aus 2,4-Dibrom-4,5a- dihydro-cortison-acetat (bereitet aus 25 g 4,5a-Di- hydro-cortison-acetat) und welches aus 1,2-Dehydro- cortison-acetat, Cortisonacetat, 4,5a-Dihydro-cortison- acetat nebst Verunreinigungen bestand, wurde wäh rend 30 Minuten mit 11,2 g Girard-Reagens T in 375 cm-- Alkohol und 37,
5 cm3 Eisessig am Rück fluss erhitzt. Die abgekühlte Lösung wurde in 2 Liter einer 4 o/oigen wässrigen Natriumbicarbonatlösung und 750 cm3 Äthylacetat gegossen. Dann wurde die wässrige Schicht dreimal mit 250 cm- Äthylacetat extrahiert.
Die ersten drei Extrakte wurden vereinigt und mit 500 cmi, 4 o/o-igem wässrigem Natrium- bicarbonat und 500 cm3 Wasser gewaschen und der vierte Extrakt dazu verwendet, diese Lösungen noch mals zu extrahieren. Die vereinigten Extrakte wurden über Magnesiumsulfat getrocknet und gleichzeitig während 1 Stunde mit 7,5g Holzkohle behandelt. Die filtrierte Lösung wurde zur Trockne eingedampft.
Der Rückstand wurde mit 25 cm-' siedendem Me thanol ausgelaugt, über Nacht bei 0 C gekühlt, fil triert und mit Äther gewaschen, wobei 9,4 g 1,2-De- hydro-cortison-acetat erhalten wurden (entsprechend 37,8 0/0, bezogen auf 4,5a Dihydro-cortison-acetat), Smp. = 235-238 C, HE, = -E-192 in Chloroform.
<I>Beispiel 3</I> Ein durch Bromwasserstoffabspaltung aus 2,4-Di- brom-4,5a-dihydro-cortison-17,21-diacetat erhaltenes rohes Gemisch (hergestellt aus 25 g 4,5a-Dihydro- cortison-acetat), welches die analogen A'-3-Keto-, d4-3-Keto- und 3-Keto-5a-steroide enthielt, wurde während einer halben Stunde mit 12,5 g Girard- Reagens P in 375 cm3 Alkohol und 37,5 cm3 Eis essig unter Rückfluss erhitzt.
Die abgekühlte Lösung wurde in 2 Liter einer 411/oigen wässrigen Natrium- bicarbonatlösung und 750 cm?, Athylacetat gegossen. Die wässrige Schicht wurde dreimal mit 250 cm3 Äthylacetat extrahiert.
Die ersten drei Extrakte wur den vereinigt und mit 500 em3- 4 o/ciger wässriger Na- triumbicarbonatlösung und dann mit 500 cm3 Wasser gewaschen und der vierte Extrakt dazu ver wendet, diese Waschlösungen nochmals zu extra hieren. Die vereinigten Extrakte wurden bis zur Trockne eingedampft, wobei man 20,1 g rohes 1,2-Dehydro-cortison-17,21-diacetat erhält, welches in 400 cm3 siedendem Äthylacetat aufgelöst und mit 4 g Holzkohle behandelt, dann filtriert und zur Trockne eingedampft wird.
Der Rückstand wurde mit 20 cm-' siedendem Methanol ausgelaugt, über Nacht bei 0 C gekühlt, filtriert und mit Äther ge waschen, was zusammen mit einem zweiten, durch Verdampfen der Mutterlaugen erhaltenen Anteil 14,7 g schwach gelbbraun gefärbte Kristalle ergab. Diese wurden nochmals in siedendem Methanol ge löst und mit 3 g Holzkohle behandelt. Die filtrierte Lösung wurde auf eine kleine Menge (etwa 50 cm3) eingedampft, auf 0 C gekühlt und die erhaltenen weissen Kristalle von reinem 1,2-Dehydro-cortison- 17,21-diacetat abfiltriert, mit Äther gewaschen und getrocknet.

<I>Beispiel 6</I> 4 g 21-Acetoxy-17a-oxy-2,4-dibrom-5a-pregnan- trion-3,11,20 und 2,2 g Semicarbazid wurden in 40 cm3 Dimethylacetamid während 40 Minuten auf 100 C erhitzt. Die Hauptmenge des Lösungsmittels wurde dann im Vakuum entfernt und 30 cm3 Wasser, 6 cm3 90 0hige Brenztraubensäure und 60 cm?, Essi- säure zugesetzt und die erhaltene Lösung während' 11/2 Stunden auf 100 C erhitzt.
Verdünnen mit Wasser und Extraktion mit Methylenchlorid ergab einen teilweise kristallisierten festen Stoff (1,92 g), welcher mit 20 cm3 Äthanol, 2 cm3 Essigsäure und 1 g Girard-Reagens P während 10 Minuten unter Rückfluss erhitzt wird.
Die Lösung wurde abgekühlt, in wässrige Natriumbicarbonatlösung gegossen und mit Methylenchlorid extrahiert. Der Extrakt wurde nacheinander mit wässriger Natriumbicarbonatlösung und Wasser gewaschen, über Magnesiumsulfat ge trocknet und unter vermindertem Druck bis zur Trockne eingedampft.
Um ein analytisch reines Pro dukt zu erhalten, wurde der Rückstand (1,08 g) in Benzol gelöst und an einer Aluminiumoxydkolonne (40 g) absorbiert. Nach Elution mit Äther-Chloro- form erhielt man 0,31 g 1,2-Dehydro-cortison-acetat, welches an Hand seines Infrarot-Spektrums identifi ziert wurde.
<I>Beispiel 7</I> Eine Lösung von 4,0 g 2,4-Dibrom-4,5ardihydro- cortison-21-acetat und 3,2 g Semicarbazid-hydro- chlorid in 40 cm3 10% Wasser enthaltendem Di- methylacetamid wurde während 10 Minuten auf einem Dampfbad erhitzt.
Nachdem die Temperatur auf 90 C gestiegen war, wurde die Hauptmasse des Lösungsmittels bei 50-60 C unter vermindertem Druck abgedampft. Dann gab man 6 cm3 90%ige Brenztraubensäure, 60 cm3 Eisessig und 30 cm3 Wasser zu und erhitzte die Lösung während 11/2 Stunden auf einem Dampfbad.
Das Reaktionsgemisch wurde gekühlt, mit Wasser verdünnt, mit Methylen- chlorid extrahiert, der Extrakt nacheinander mit ge sättigter Natriumbicarbonatlösung und Wasser ge waschen und über Magnesiumsulfat getrocknet. Das Lösungsmittel wurde unter vermindertem Druck ent fernt, dann wurden 20 cm3 Pyridin und 10 cm Essigsäureanhydrid zugegeben und die Lösung über Nacht bei Zimmertemperatur stehengelassen.
Das durch Abdampfen des Lösungsmittels unter vermin dertem Druck erhaltene Rohprodukt (2,93 g) wurde mit 1,5 g Girard-Reagens P in 2 cm3 trockenem Äthanol und 2 cm-' Eisessig während 10 Minuten am Rückfluss erhitzt. Die Lösung wurde abgekühlt, in wässrige Natriumbicarbonatlösung gegossen und mit Methylenchlorid extrahiert. Der Extrakt wird mit wässriger Natriumbicarbonatlösung und mit Wasser gewaschen, über Magnesiumsulfat getrocknet und das Lösungsmittel unter vermindertem Druck abge dampft.
Um ein analytisch reines Produkt zu erhal ten, wird der Rückstand (1,89 g, = 238 mcc, Ei @m = 312) in Benzol gelöst und an einer 60 g Aluminiumoxyd enthaltenden Kolonne absorbiert. Elution mit 500 cm3 Chloroform, Entfernen des Lösungsmittels unter vermindertem Druck und Um kristallisieren des Rückstandes aus Äthylacetat ergab 0,5 g 1,2-Dehydro-cortison-acetat mit dem Schmelz punkt 230-234 C,

21-di- acetat und 8 g Semicarbazid-hydrochlorid wurden in 90 cm3 Dimethylformamid während 45 Minuten auf einem Dampfbad erhitzt. Die Hauptmenge des Lö sungsmittels wurde unter vermindertem Druck ent fernt, dann wurden 70 cm3 Wasser, 160 cm3 Essig säure und 26 em3 50-600/aige Brenztraubensäure zugegeben und die Lösung während 11/2 Stunden auf 90 C erhitzt.
Das Reaktionsgemisch wurde abge kühlt, mit Wasser verdünnt, mit Methylenchlorid extrahiert, der Extrakt gewaschen und mit Magne- siumsulfat getrocknet. Nach Verdampfung des Lö sungsmittels verblieben 5,86 g eines Rohproduktes mit Amax = 263 m., Ei % = 333, welches nach Wiederacetylierung mittels Acetanhydrid und Pyridin während 15 Minuten unter Rückfluss mit 40 em3 Äthanol,
4 cm3 Essigsäure und 2 g Girard-Reagens P erhitzt wurde. Nach Abkühlen wurde die Lösung in 350 cm3 40/aige wässrige Natriumbicarbonatlösung gegossen, mit Äthylacetat extrahiert, der Extrakt mit Wasser gewaschen und über Magnesiumsulfat ge trocknet.
Nach Verdampfen des Lösungsmittels unter vermindertem Druck erhielt man 3,11 g eines Roh produktes mit A"," = 237 mu, E ' ^ %," = 311, welches zur Erzielung eines analytisch reinen Materials in Benzol gelöst und an einer Aluminiumoxydkolonne absorbiert wird. Elution mit Benzol und Umkristalli- sieren des Produktes mit Methanol -ergab 1,23 g 1,2-Dehydro-cortison-17,21-diacetat vom Schmelz punkt 212-213 C;

Besides cortisone and its analogues, other Al.4-3-keto-steroids are also of interest.
Various methods have been proposed for the synthesis of JI.4-3-keto steroids. One of these methods is to split off hydrogen bromide from 2,4-dibromo-3-keto steroids, e.g. B. by means of a tertiary base such as collidine. A suitable and promising method of this type would be of great value, especially in the Allo series, since 2,4-dibromo-3-keto-5a-steroids can be easily prepared by methods such as those used for the synthesis of cortisone .
However, it has been found that such elimination of hydrogen bromide always leads to mixtures which contain the desired 41,4-3-keto-steroid in more or less large amounts together with JI-, d4- and / or J4,6-3-keto- contain steroids as impurities, depending on the success of the splitting off of hydrogen bromide.

It has now been found that saturated 3-keto steroids,: A'- and J4-3-keto-steroids and also A4,6_3_keto-steroids with the Girard reagents T and P result in derivatives, whereas the 41.4-3 -Keto- steroids do not give such derivatives as long as they contain no further carbonyl groups apart from the keto group in the 3-position or only those which do not react with Girard's reagent T or P.
