Pyridin oder Dimethylanilin) durchgeführt wird. Das Säurebindemittel ist entbehrlich, wenn das Phenol in der Form einer Alkalimetallverbindung verwendet wird.
Die Art des Anions der Säureadditionssalze ist nicht von wesentlicher Bedeutung, abgesehen davon, dass es selbstverständlich nicht von pharmakologisch ungünstiger Wirkung sein sollte. Geeignete Beispiele sind Halogenide, wie Chlorid oder Bromid, Bitartrat, Citrat und neutrales Sulfat.
Die Bezeichnung zu niedere mit Bezug auf Alkyl- Gruppen soll angeben, dass die Gruppe im allgemeinen nicht mehr als vier Kohlenstoffatome enthält.
Das erfindungsgemäss erhaltene Produkt lässt sich in ein quaternäres Ammoniumsalz umwandeln, indem es mit einer Verbindung der Formel RGX, worin R6 eine Alkylgruppe und X ein Anion einer Säure bedeuten, zur Reaktion gebracht wird.
Die neuen erfindungsgemäss erhaltenen Verbindungen sind in pharmakologischer Hinsicht dadurch wertvoll, dass sie örtliche Betäubungsmittel von sehr kräftiger Wirkung sind. Viele davon besitzen den besonderen Vorteil einer verlängerten Wirkung zusammen mit niedriger Toxizität. Unter den Aminoalkyläthern der Formel I werden diejenigen wegen ihrer starken Wirkung bevorzugt, bei denen R, und R2 Methylgruppen sind, R5 ein Wasserstoffatom, A die Gruppe -CH2-CH2- darstellen, sowie R4 und R5 untereinander gleich sind und eine Methyloder Äthylgruppe bedeuten.
Es handelt sich mit an dem Worten um die Verbindungen 2-(ss-Dimethyl- amino-äthoxy)- 1,3 -dimethyl-benzol und 2-(4-Diäthyl- amino-äthoxy)-1, 3-dimethyl-benzol.

kann Isomerbildung im Laufe des erfindungsgemässen Verfahrens eintreten, wodurch gleichzeitig zwei Isomere erzeugt werden. Diese können jedoch ohne weiteres getrennt werden. Zum Beispiel kann eine Mischung isomerer Ätherbasen in Hydrochloride umgewandelt werden, wonach die Hydrochloridmischung aus einem Lösungsmittel, z. B. Aceton, durch fraktionierte Kristallisation getrennt werden kann, worauf, wenn freie Ätherbasen erwünscht sind, die so erhaltenen getrennten Isomere je für sich mit Alkalilauge behandelt werden können.
Beispiel I
2,6-Dichlor-phenol (27,7 g; 0,17 Mol) wird einer Lösung von 85e/eigem Kaliumhydroxyd (13 g; 0,20 Mol) in Wasser (3 ml) und Äthanol (60 ml) zugefügt, und die sich ergebende Mischung wird eine Stunde lang auf dem Wasserbad mit ss-Dimethylamino-äthylchlorid-hydrochlorid (15,0 g; 0,1 Mol) unter Rückfluss behandelt. Die Mischung wird dann abgekühlt, das ausgefallene Kaliumchlorid durch Filtrieren entfernt, wonach das Filtrat und die Spülreste im Vakuum konzentriert werden. Das restliche öl wird in 2n-Salzsäure (50 ml) aufgenommen, und das unveränderte Dichlorphenol wird mit Ather extrahiert.
Ein Überfluss an festem Kaliumcarbonat wird der wässrigen Lösung zugesetzt, die ausgefallene Base wird mit Äther extrahiert und die Ätherlösung mit Kaliumcarbonat getrocknet. Trockener Chlorwasserstoff wird danach zugegeben, bis die Mischung eben dauernd sauer gegenüber Kongorot wird. Das ausgefällte Salz wird abfiltriert und mit Äther gewaschen, wodurch 2-(B-Dimethylamino-äthoxy) -1,3-dichlor- benzol-hydrochlorid (11,2 g) erhalten wird mit dem Schmelzpunkt 165-169" C, das nach Umkristallisierung aus einer Mischung von Alkohol, Aceton und Äther bei 1691700 C schmilzt.
Beispiel II
Natrium (1,38 g; 0,06 Mol) wird in Methanol aufgelöst, 2,6-Dichlor-phenol (9,9 g; 0,06 Mol) wird hinzugefügt, wonach die Lösung bis zur Trockenheit verdampft und der feste Rückstand in Aceton (50 ml) aufgelöst wird. Zu einer Lösung von ss-Diäthylamino äthylchlorid-hydrochlorid (10,5 g; 0,06 Mol) in Wasser (6,0 ml) wird zuerst Äther und dann ein Überschuss an festem Kaliumcarbonat (15 g) unter Kühlung zugesetzt. Die Ätherlösung und die Ätherspülreste werden danach von der steifen Paste in die obige Acetonlösung abdekantiert. Der Äther wird dann so lange abdestilliert, bis die Temperatur des Reaktionsgemisches 55" C erreicht, und die Mischung wird dann 4 Stunden lang einer Rückflussbehandlung ausgesetzt. Die Mischung wird danach abgekühlt.
Das ausgefällte Natriumchlorid wird durch Filtrierung weggeschafft, und es wird in derselben Weise wie im Beispiel I angegeben 2-(ss-Diäthylamino äthoxy)-1,3-dichlor-benzol-hydrochlorid (14,5 g), Schmp. 115-117"C, erhalten, welches nach Umkristallisierung aus Aceton-Äther bei 117-118"C schmilzt.
Beispiel III
Es wird wie im Beispiel II vorgegangen, wobei jedoch von Natrium (3,45 g; 0,15 Mol), 2,6-Xylenol (18,3 g; 0,15 Mol) und 3-Dimethylamino-2-chlor propan-hydrochlorid (24 g; 0,15 Mol) ausgegangen wird, so dass ein rohes Hydrochlorid (31 g), Schmp.
146-170" C, erhalten wird, aus dem 2-(2'-Dimethyl amino-l'-methyl-äthoxy) - 1,3-dimethyl-benzol-hydro- chlorid, Schmp. 1591600 C, durch fraktionierte Kristallisation aus Aceton hergestellt wird.
Beispiel IV
Stimmt mit Beispiel II überein, wobei jedoch von Natrium (2,8 g; 0,122 Mol), 2,6-Xylenol (16,5 g; 0,135 Mol) und 3-Diäthylamino-2-chlor-propanhydrochlorid (22,4 g; 0,12 Mol) ausgegangen wird, aus denen eine Mischung von Hydrochloriden (33 g), Schmp. 110-149"C, erhalten wird, aus der durch fraktionierte Kristallisation aus Aceton 2-(2'-Diäthyl amino-2'-methyl-äthoxy) - 1,3 -dimethyl-benzol-hydro- chlorid (11,8 g), Schmp. 161-162"C, und 2-(2' Diäthylamino-l'-methyl-äthoxy)-1, 3-dimethyl-benzol- hydrochlorid (12,6 g), Schmp. 115-121"C, erhalten werden, wobei das letztere nach nochmaliger Kristallisation aus einer Mischung von Alkohol, Aceton und Ather bei 120-121"C schmilzt.
Beispiel V
Die folgenden Verbindungen können in ähnlicher Weise wie in den vorhergehenden Beispielen hergestellt werden: a) 2-(ss-Dimethylamino-äthoxy)-1, 3-dimethyl-ben- zol. Flüssig, Siedepunkt 124" C/10 mm.
Das Hydrobromid kristallisiert aus Methanol in Nadeln vom Schmelzpunkt 1660 C aus. b) 2-(ss-Diäthylamino-äthoxy)-1, 3-dimethyl-ben- zol. Flüssig, Siedepunkt 131" C/10 mm.
Das Hydrobromid kristallisiert aus Methanol in Nadeln vom Schmelzpunkt 151 C aus.

For example, a mixture of isomeric ether bases can be converted into hydrochlorides, after which the hydrochloride mixture can be converted from a solvent, e.g. B. acetone, can be separated by fractional crystallization, whereupon, if free ether bases are desired, the separated isomers thus obtained can be treated individually with alkali.
Example I.
2,6-dichlorophenol (27.7 g; 0.17 mol) is added to a solution of 85e / pure potassium hydroxide (13 g; 0.20 mol) in water (3 ml) and ethanol (60 ml), and the resulting mixture is refluxed for one hour on a water bath with β-dimethylamino-ethyl chloride hydrochloride (15.0 g; 0.1 mol). The mixture is then cooled, the precipitated potassium chloride is removed by filtration, after which the filtrate and the rinse residues are concentrated in vacuo. The remaining oil is taken up in 2N hydrochloric acid (50 ml), and the unchanged dichlorophenol is extracted with ether.
An excess of solid potassium carbonate is added to the aqueous solution, the precipitated base is extracted with ether and the ether solution is dried with potassium carbonate. Dry hydrogen chloride is then added until the mixture just becomes permanently acidic to Congo red. The precipitated salt is filtered off and washed with ether, whereby 2- (B-dimethylamino-ethoxy) -1,3-dichlorobenzene hydrochloride (11.2 g) with the melting point 165-169 "C, which is obtained after recrystallization from a mixture of alcohol, acetone and ether melts at 1691700 C.
Example II
Sodium (1.38 g; 0.06 mol) is dissolved in methanol, 2,6-dichlorophenol (9.9 g; 0.06 mol) is added, after which the solution is evaporated to dryness and the solid residue in Acetone (50 ml) is dissolved. To a solution of ß-diethylamino ethyl chloride hydrochloride (10.5 g; 0.06 mol) in water (6.0 ml) is first added ether and then an excess of solid potassium carbonate (15 g) with cooling. The ether solution and the ether rinsing residue are then decanted from the stiff paste into the above acetone solution. The ether is then distilled off until the temperature of the reaction mixture reaches 55 ° C., and the mixture is then refluxed for 4 hours. The mixture is then cooled.
The precipitated sodium chloride is removed by filtration, and it is indicated in the same manner as in Example I 2- (ss-diethylamino ethoxy) -1,3-dichlorobenzene hydrochloride (14.5 g), m.p. 115-117 " C, which melts at 117-118 "C after recrystallization from acetone-ether.
Example III
The procedure is as in Example II, except that sodium (3.45 g; 0.15 mol), 2,6-xylenol (18.3 g; 0.15 mol) and 3-dimethylamino-2-chloropropane hydrochloride (24 g; 0.15 mol) is assumed, so that a crude hydrochloride (31 g), m.p.
146-170 "C, is obtained from the 2- (2'-dimethylamino-l'-methyl-ethoxy) -1,3-dimethyl-benzene hydrochloride, melting point 1591600 C, by fractional crystallization from acetone will be produced.
Example IV
