Verfahren zur Herstellung von Dehydroverbindungen der Steroidreihe Es ist bereits bekannt, dass man in Verbindun gen der Pregnanreihe einen Abbau der Seitenkette auf mikrobiologischem Wege herbeiführen kann; es entstehen dabei Androstanverbindungen und gleich zeitig wird in 1,2- und gegebenenfalls in 4,5-Stel- lung eine Doppelbindung eingeführt; bei genügend langer Inkubation wird ausserdem der D -Ring auf gespalten.
Auf diesem Wege lässt sich zum Beispiel das Progesteron, wie auch d5-3f-Oxy-20-oxo-pre- gnen, 11-Desoxy-corticosteron oder 3,20-Dioxo-allo- pregnan in einer Verfahrensstufe in di,4-3,17 Dioxo- androstadien bzw. 1,2-Dehydro-testololacton über führen.
Es wurde nun gefunden, dass in Stereoiden, ins besondere der Pregnanreihe, die 1,2-Doppelbindung ohne Seitenkettenabbau oder Ringaufspaltung auf biochemischem Wege eingeführt werden kann, wenn man in 1,2-Stellung gesättigte Steroide der aeroben Einwirkung von Enzymen von Calonectria decora, Altemaria passiflorae, Ophiobolus heterostrophus, oder Ophiobolus Miyabeanus unterwirft.
Als Ausgangsstoffe können zum Beispiel ver wendet werden: Abkömmlinge des Spirostans, Allo- spirostans, Furostans, Allofurostans, Cholans, Allo- cholans, Pregnans, Allopregnans, Androstans und Testans. Vorzugsweise weisen die Ausgangsstoffe in 3-Stellung eine freie oder funktionell abgewandelte Oxy- oder Oxogruppe auf.
Sie können gesättigt sein oder Doppelbindungen, ausser in 1-Stellung, enthal ten, z. B. in 4-, 5-, 6-, 7-, 8-, 9: 11-, 14- oder 16- Stellung, oder zusätzliche Substituenten besitzen, wie freie oder abgewandelte Oxy-, Oxo- oder Carboxyl- gruppen, ferner Epoxygruppen oder Halogenatome, beispielsweise in 6-, 7-, 8-, 9-, 11-, 12-, 14-, 15-, 16-, 17-, 18-, 20- oder 21-Stellung oder Methyl- gruppen,
zum Beispiel in 17a- oder 17f-Stellung. Die oben genannten Ausgangsstoffe können von beliebiger sterischer Konfiguration sein und können auch als Racemate vorliegen. Auch Steroide der sogenannten Nor- und/oder Homo-Reihen, insbe sondere 19-Nor- und D-Homo-Verbindungen lassen sich als Ausgangsstoffe verwenden. Besonders wich tige Ausgangsstoffe sind Verbindungen der Pregnan- reihe, die in 3- und 20-Stellung freie oder funktionell abgewandelte Oxy- oder Oxogruppen enthalten, z. B.
Progesteron, 11-Dehydro-progesteron, 11-, 12-, 14-, 15-, 16-, 17- oder 18-Oxy-progesterone, 45- oder 44- Pregnen-3-ol-20-one, 45-Pregnen-3,20-diole, 11 Des- oxy-corticosteron, Cortison, Hydrocortison, 11-Epi- hydrocortison, Aldosteron, 18-Oxy-corticosteron, 11- Epi-18-oxy-corticosteron, 18-Oxo-corticosteron,

die entspre chenden 21-Oxo-verbindungen und bzw. oder 9a- Fluor- und Chlorderivate, beispielsweise 9a Fluor cortison, 9a-Fluor-hydrocortison, 9a-Fluor-aldosteron und 9a-Fluor-18-oxy-corticosteron, ferner in 1,2- Stellung gesättigte Androstanverbindungen, zum Bei spiel 44- oder d5-Androsten-3,17-dion, Testosteron, d5-Androsten-3-ol-17-on, Adrenosteron, Androstan- 3,17-dion,
Ätiansäuren, zum Beispiel 44- oder d5 Ätiensäuren, Alloätiansäuren, die insbesondere in 3-, 11- und bzw. oder 18-Stellung Oxy- oder Oxo- gruppen aufweisen, bzw.
entsprechende Verbindun gen mit funktionell abgewandelten Oxy- oder Oxo- gruppen. In den Ausgangsstoffen kann die funktionell abgewandelte Oxygruppe zum Beispiel eine mit einer aliphatischen, aromatischen oder heterocyclischen Carbonsäure, beispielsweise Essigsäure, Propionsäure, Benzoesäure oder Furancarbonsäure,
veresterte oder eine verätherte Oxygruppe darstellen, zum Beispiel die Tetrahydropyranyloxy-, Benzyloxy- oder Tri- phenylmethoxygruppe. Die funktionell abgewandelte Oxogruppe ist vorteilhaft eine ketalisierte Oxogruppe, abgeleitet insbesondere von einem zweiwertigen Alko hol, wie die Äthylendioxygruppe.
Die genannten Ausgangsstoffe werden verfah rensgemäss mit Enzymen der oben genannten Pilz arten in Reaktion gebracht. Für die Gewinnung dieser Enzyme eignen sich die an sich hierfür bekannten Medien, zum Beispiel solche mit Zuckern, wie Glucose oder Lactose, mit Peptonen, Maisquellwas ser, Sojaprodukten und dergleichen, sowie mit Mine ralsalzen, oder aber synthetische Nährlösungen. Man arbeitet unter aeroben Bedingungen, beispiels weise in Schüttelkultur oder bei untergetauchtem Wachstum unter Rühren und Luftzufuhr. Die ge nannten Pilze unterscheiden sich von andern Mikro organismen, zum Beispiel den Bakterien, durch gutes Wachstum unter verhältnismässig einfachen Zucht bedingungen.
Die verfahrensgemässe Reaktion kann in den beschriebenen Pilzkulturen stattfinden. Man kann gegebenenfalls auch angereicherte oder abge trennte Enzyme verwenden. Im einfachsten Fall wird man die Dehydrierung in einer Aufschwemmung des abgetrennten Pilzmycels, des homogenisierten Pilz- mycels oder in Filtraten bzw. wasserhaltigen Extrak ten davon durchführen.
Die Isolierung der Verfahrensprodukte lässt sich nach bekannten Methoden vornehmen. Ihre Abtren nung kann zum Beispiel durch Extraktion des Re aktionsgemisches mit einem organischen Lösungs mittel, zum Beispiel Methylenchlorid oder Essig ester, erfolgen. Für die weitere Reinigung des so erhaltenen Extraktes eignen sich besonders Chro- matographie, zum Beispiel an Aluminiumoxyd oder Silicagel, Anwendung von Verteilungsmethoden, zum Beispiel des Gegenstromverfahrens, oder die Tren nung mittels Girard-Reagentien,
wie Trimethyl- ammonium- oder Pyridinium-essigsäurehydrazid. An schliessend an die Reinigung oder an ihrer Stelle kann schliesslich, vorzugsweise aus organischen oder wäs serig - organischen Lösungsmitteln, umkristallisiert werden.
Durch Einführung der 1,2-Doppelbindung ge langt man zu wertvollen Heilmitteln der Steroid-, insbesondere der Pregnanreihe, die sich, verglichen mit den in 1,2-Stellung gesättigten, therapeutisch wirksamen Verbindungen durch eine gesteigerte Wirksamkeit auszeichnen.

Sofern die Verfahrensprodukte nicht die Konfiguration und die Substituenten von therapeutisch verwendbaren Steroiden aufweisen, können sie als Zwischenprodukte zu deren Herstel lung, zum Beispiel der obengenannten Verbindungen, dienen.
Verfahrensprodukte mit freien Oxy- und/oder Oxogruppen lassen sich in an sich bekannter Weise in ihre funktionellen Derivate, beispielsweise Ester, Äther, Enolester, Enoläther, Ketale, Thioäther und Thioketale, ferner Hydrazone, Oxime und Enamine überführen. Vorhandene freie Oxy- und/oder Oxo- gruppen lassen sich vollständig oder teilweise funk tionell abwandeln.
Zur Herstellung von Estern und Enolestern sind beliebige organische und anorganische Säuren, wie aliphatische, alicyclische, araliphatische, aromatische oder heterocyclische Carbon-, Thioncarbon-, Thiol- carbon- oder Sulfonsäuren verwendbar, vorzugsweise Ameisensäure, Essigsäure, Chloressigsäuren, Tri- fluoressigsäure, Propionsäure, Buttersäuren,
Valerian- säuren, Diäthylessigsäure, Capronsäuren, Önanth- säuren, Caprylsäuren, Palmitinsäuren, Crotonsäure, Undecänsäure, Undecylensäure, Oxalsäure, Bernstein säure, Pimelinsäure, Maleinsäure, Milchsäure, Carb- aminsäuren, Alkoxycarbonsäuren,
ss-Cyclopentyl-pro- pionsäure, Hexahydrobenzoesäure, Benzoesäure, Phenylessigsäure, Cyclohexylessigsäure, Phenylpro- pionsäuren, Trimethylgallussäure, Phthalsäure, Fu- ran-2-carbonsäure, Isonicotinsäure, Methansulfon- säure, Toluolsulfonsäure, Schwefelsäuren,
Halogen wasserstoffsäuren oder Phosphorsäuren.
Verfahrensprodukte mit funktionell abgewandel ten Oxy- und/oder Oxogruppen lassen sich in Ver bindungen mit freien Oxy- oder Oxogruppen über führen. Auf diese Weise können insbesondere in polysubstituierten Derivaten die funktionell abgewan delten Gruppen auch teilweise in Freiheit gesetzt werden.
Dies erfolgt zum Beispiel durch chemische oder enzymatische Hydrolyse, beispielsweise unter Verwendung saurer oder basischer Mittel, durch Umesterung oder Umacetalisierung. Aus den auf diese Weise oder auch direkt erhaltenen, nur par tiell abgewandelten, wie veresterten bzw. veräther- ten Derivaten lassen sich durch anschliessende funk tionelle Abwandlung, zum Beispiel Veresterung oder Verätherung, polysubstituierte Derivate, insbeson dere auch gemischte Ester oder Äther bzw.
Ester- Äther, herstellen.
In den folgenden Beispielen sind die Tempera turen in Celsiusgraden angegeben. <I>Beispiel 1</I> 4 Liter 70prozentige Bierwürze werden in einem Schüttelgefäss sterilisiert (PH 5,1) und mit 150 em3 einer 2 Tage alten Schüttelkultur von Calonectria decora in 70prozentiger Bierwürze beimpft. Das Ge fäss lässt man 24 Stunden bei 270 schütteln, wobei sich die Kultur gut entwickelt. Dann gibt man eine Lösung von 1 g Cortexon in 25 cm3 Aceton unter sterilen Bedingungen zu und lässt weiter bei 270 schütteln. Nach 2 Tagen wird das Mycel abgetrennt und gut mit Wasser und Essigester gewaschen.
Nach erschöpfender Extraktion der vereinigten Filtrate mit insgesamt 4 Liter Essigester werden die Extrakte mit Wasser gewaschen, getrocknet und im Vakuum eingedampft. Der so gewonnene rohe Extrakt (1,1 g) wird an einer Säule von 30 g Silicagel nach der Durchlaufmethode chromatographiert, wobei man mit Chloroform und mit Chloroform-Aceton-Ge- mischen steigenden Acetongehaltes eluiert. Die ein zelnen Fraktionen (je 100 cm3)
untersucht man pa- pierchromatographisch. Die mit Chloroform eluierten Anteile enthalten nur Verunreinigungen, während sich in den Chloroform - Aceton<B>(9:</B> 1)-Fraktionen eine neue Substanz befindet, die im Papierchromato- gramm (System Propylenglykol-Toluol) etwas weni ger weit als Cortexon wandert. Die Fraktionen mit höherem Acetongehalt enthalten nur wenig hoch polares Material. Die die erwähnte neue Substanz enthaltenden Fraktionen vereinigt man und dampft sie im Vakuum ein.
Durch Umkristallisieren des Rückstandes aus Aceton-Petroläther-Gemischen er hält man das reine 1-Dehydro-cortexon vom F. = 185 bis .1920 in beinahe quantitativer Ausbeute. Diese Verbindung reduziert alkalische Silberdiammin- Lösung rasch und stark.
Die Analyse stimmt auf die Bruttoformel C21H,803 (Ber. C 76,79 H 8,59 %, Gef. C 76,67 H 8,84 0/0). Das UV-Spektrum der freien Verbindung zeigt eine starke Bande mit dem Maximum bei<I>244</I> m,y (E <I>=</I> 14 200), dasjenige des Dinitrophenylhydrazons eine solche bei 388 mu (a = 39 000).
Die entsprechenden Banden des Cor- texons liegen bei 240 m/c (s = 16 200) resp. 378 mit (a = 39 000).
<I>Beispiel 2</I> Verwendet man zur Dehydrierung des Cortexons statt der in Beispiel 1 beschriebenen Kultur von Calonectria decora in analoger Weise Kulturen von Alternaria passiflorae, Ophiobolus heterostrophus oder Ophiobolus Miyabeanus, so erhält man nach der in Beispiel 1 beschriebenen Aufarbeitung das 1-Dehydrocortexon in hoher Ausbeute.
<I>Beispiel 3</I> Man züchtet, wie in Beispiel 1 beschrieben, in einem Schüttelgefäss 4 Liter einer Kultur von Calonectria decora und gibt unter sterilen Bedingun gen eine Lösung von 1 g Cortison.in 25 cm3 Me thanol zu. Das Gefäss wird darauf bei 270 geschüt telt. Nach 4 Tagen trennt man das Mycel ab und extrahiert das Kulturfiltrat erschöpfend mit insge samt 3 Liter Essigester. Die vereinigten Extrakte wer den mit Wasser gewaschen, getrocknet und im Va kuum eingedampft.
Der so erhaltene rohe Rückstand wird, wie in Beispiel 1 beschrieben, an 30 g Silicagel nach der Durchlaufmethode chromatographiert. Die mit Chloroform-Aceton (6 : 4)-Gemischen eluierten Anteile bestehen zur Hauptsache aus einer Substanz, die etwas polarer ist als das Ausgangsmaterial. Diese Fraktionen werden vereinigt und im Vakuum einge dampft. Aus Aceton-Äther-Gemischen wird das 1- Dehydro-cortison kristallisiert. F. = 231 bis 2340.
Die Substanz reduziert alkalische Süberdiammin-Lö- sung rasch und stark und zeigt im UV-Spektrum eine starke Absorption bei 240 mu (E = 15 800). Durch Acetylierung mit Pyridin-Acetanhydrid erhält man das entsprechende 21-Acetat vom F. = 224 bis 2300. In ähnlicher Weise lässt sich zum Beispiel das Önan- that oder Undecylenat herstellen.
<I>Beispiel 4</I> Zu 4 Liter einer gut entwickelten Kultur von Calonectria decora gibt man unter sterilen Bedin gungen eine Lösung von 1 g Hydrocortison in 25 cm3 Methanol. Nach 4tägigem Schütteln bei 270 wird, wie in Beispiel 1 beschrieben, extrahiert und chro- matographiert. Die mit Chloroform-Aceton (1 : 1)-Ge- mischen eluierten Anteile enthalten eine Substanz, die etwas polarer als Hydrocortison ist.
Diese Frak tionen vereinigt man und dampft sie ein. Das 1-De- hydro-hydrocortison wird aus Methanol oder Aceton Äther-Gemischen umkristallisiert. F. =<B>238</B> bis 2410. Es reduziert alkalische Silberdiammin-Lösung rasch und stark und zeigt im UV-Spektrum eine starke Absorption bei 244 m,ec (s = 15 100).
<I>Beispiel S</I> In einem Erlenmeyerkolben von 500 cm3 Fas sungsvermögen werden 150 cm3 70prozentige Bier würze sterilisiert und mit Calonectria decora be- impft. Man lässt 2 Tage bei 270 schütteln und gibt zu der nun gut entwickelten Kultur unter sterilen Bedingungen eine Lösung von 30 mg Aldosteron in 1,5 cm3 Aceton zu. Dann schüttelt man bei 270 weiter und filtriert nach 3 8 Stunden das Mycel ab.
Das Kulturfiltrat wird mit Essigester erschöpfend extrahiert, die Extrakte mit Wasser gewaschen, ge trocknet und eingedampft. Der Rückstand wird, wie in Beispiel 1 beschrieben, an 1 g Silicagel chromato- graphiert, wobei die einzelnen Fraktionen (je 20 cm3) papierchromatographisch untersucht werden. In den mit Chloroform-Aceton (1: 1) -Gemischen eluierten Anteilen befindet sich eine Substanz, die etwas po larer ist als Aldosteron und alkalische Silberdiammin- Lösung reduziert.
Sie wird aus Aceton-Äther-Ge- mischen kristallisiert und stellt das 1-Dehydro-aldo- steron dar. Im UV-Spektrum zeigt sie starke Absorp tion bei 244 m@t.
<I>Beispiel 6</I> In 14 Erlenmeyerkolben von 200 cm3 Fassungs vermögen werden je 50 cm3 70prozentige Bierwürze sterilisiert und mit Calonectria decora beimpft. Nach 2tägigem Schütteln bei 270 haben sich die Kulturen gut entwickelt.
Unter sterilen Bedingungen gibt man zu den 14 Kulturen je eine der nachstehen den 14 Verbindungen (je 10 mg in 0,5 cm3 Me thanol gelöst): Cortexon, Cortison, Hydrocortison, 17a-Oxy- cortexon, Corticosteron, 11-Epi-corticosteron, 11- Epi-hydrocortison, Aldosteron, 17a - Oxy-aldosteron, Progesteron, Pregnenolon, 21-Oxy-pregnenolon,
An- drostan-3,17-dion und Androsten-3,17-dion. Man lässt 2 Tage lang bei 270 weiter schütteln und extra hiert dann die Kulturen mit Essigester. Die papier- chromatographische Untersuchung der einzelnen Ex trakte zeigt, dass alle Ausgangssubstanzen in die ent sprechenden 1 Dehydro-Verbindungen übergeführt worden sind,
die gegenüber den Ausgangsstoffen. ganz leicht erhöhte Polarität besitzen und sich in ihren Spektren charakteristisch von diesen unter scheiden.<I>Beispiel 7</I> Verwendet man zur Dehydrierung der in Bei spiel 6 genannten Substanzen statt der dort beschrie benen Kulturen von Calonectria decora Kulturen von Alternaria passiflorae Ophiobolus heterostrophus oder Ophiobolus Miyabeanus,
. so zeigt die papierchromato- graphische Untersuchung der Extrakte die gleichen Umsetzungsprodukte, wie sie in Beispiel 6 beschrie ben sind.<I>Beispiel 8</I> Zu 4 Liter einer gut entwickelten Kultur von Calonectria decora gibt man unter sterilen Bedin gungen eine Lösung von 1 g 3,11,20-Triketo-17a,21- dioxy-allopregnan in 25 cm- Methanol. Man lässt 3 Tage bei 270 schütteln und trennt dann das Mycel ab.
Die Extraktion des Kulturfiltrates und die Chro- matographie des Rohextraktes wird in gleicher Weise wie in Beispiel 1 beschrieben durchgeführt. Die mit Chloroform-Aceton (1: 1)-Gemischen eluierten An teile enthalten das 1 Dehydrocortison, das aus Ace- ton-Äther-Gemischen kristallisiert erhalten wird, F. = 230 bis 2330. Es besitzt die in Beispiel 3 beschriebenen Eigenschaften.
Wird an Stelle der Lösung von 3,11,20-Tri- keto - 17a,21 - dioxy - allopregnan eine Lösung von 3,11,20-Triketo-17a,21-dioxy - pregnan in 25 cm3 Methanol einer analogen Kultur von Calonectria decora zugesetzt und in der beschriebenen Weise aufgearbeitet, so erhält man ebenfalls das 1-Dehydro- cortison in guter Ausbeute.
<I>Beispiel 9</I> Unter sterilen Bedingungen wird eine Lösung von 1 g 3,20-Diketo-11,B,17a,21-trioxy-allopregnan in 25 cm Methanol zu 4 Liter einer Kultur von Calonectria decora gegeben und die Mischung 4 Tage bei 270 geschüttelt. Dann trennt man vom Mycel ab und führt die Extraktion des Kulturfil trates und die chromatographische Auftrennung des Extraktes wie in Beispiel 1 beschrieben durch.
Aus den mit Chloroform-Aceton (1: 1)-Gemischen eluier- ten Anteilen wird das 1-Dehydro-hydrocortison in guter Ausbeute kristallisiert erhalten, F. = 239 bis 242 . Es besitzt die in Beispiel 4 beschriebenen Eigenschaften.
<I>Beispiel 10</I> Eine Lösung von 1 g Androstan-3,17-dion in 25 cm3 Aceton wird unter sterilen Bedingungen zu 4 Liter einer Kultur von Calonectria decora gege ben. Nach 36stündigem Schütteln bei 270 trennt man das Mycel ab und schüttelt das Kulturfiltrat er schöpfend mit Methylenchlorid aus. Der Extrakt wird je zweimal mit 0,1-n. Salzsäure und 1prozenti ger Natriumbicarbonatlösung und dreimal mit Wasser gewaschen, getrocknet und im Vakuum eingedampft.
Den Rückstand (1,2 g) chromatographiert man an 30g Aluminiumoxyd nach der Durchlaufmethode, wobei man mit Petroläther-Benzol-Gemischen, mit Benzol, mit Benzol-Äther-Gemischen und mit Äther eluiert. Die mit Petroläther-Benzol und mit Benzol ab gelösten Anteile enthalten das bekannte d1.-1Andro- stadien-3,17-dion, das aus einem Aceton-Petroläther- Gemisch in rhombischen Platten kristallisiert. F. _ 145 bis 1460; [a]D =<B>+"0"</B> (CHC13).
<I>Beispiel 11</I> Eine Lösung von 40 mg, gemäss Beispiel 1 her gestelltem, 1-Dehydro-cortexon ([a]D = -I-1200 in CHC13) in 2 cm3 Pyridin werden mit 2 cm3 Essig säureanhydrid versetzt und bei Zimmertemperatur stehengelassen. Nach 16 Stunden dampft man die Lösung im Vakuum bei 300 ein und nimmt den Rückstand in einem Chloroform-Äther-(1-4)-Ge- misch auf. Man wäscht die Lösung je dreimal mit 0,1-n.

<I> Example 11 </I> A solution of 40 mg of 1-dehydrocortexon ([a] D = -I-1200 in CHC13) prepared according to Example 1 in 2 cm3 of pyridine is mixed with 2 cm3 of acetic anhydride and left to stand at room temperature. After 16 hours, the solution is evaporated in vacuo at 300 and the residue is taken up in a chloroform-ether (1-4) mixture. The solution is washed three times with 0.1-n.
Hydrochloric acid, with 2 percent sodium hydrogen carbonate solution and with water, dries it and evaporates it in a vacuum. The extraction of the culture filtrate and the chromatography of the crude extract on silica gel is carried out as described in Example 1.
The chloroform and chloroform acetone (95: 5) fractions contain impurities and some starting material, while the chloroform acetone <B> (90: </B> 10) fractions mainly consist of one substance which in the paper chromatogram runs a little slower than the starting material. This substance is d1,4-3,11,20-triketo-21-oxy-pregnadiene, which is crystallized from acetone-ether. The crystals decompose between 200 and 220, depending on the heating speed.
The substance absorbs in the W at 240 m, u and shows in the IR the bands characteristic of the di, 4-3-keto grouping at 5.99, c4, 6.14 / t and 6.22, u.
<I> Example 13 </I> A solution of 1 g of corticosterone in 25 cm 3 of acetone is added under sterile conditions to 4 liters of a culture of Calonectria decora which is grown as described in Example 1. The mixture is shaken at 270 for 4 days and the mycelium is then filtered off. The extraction of the culture filtrate and the chromatography of the crude extract on silica gel are carried out as described in Example 1.
The portions eluted with chloroform and with chloroform acetone (95: 5) contain impurities and starting material. With chloroform-acetone (90:10) and (80:20) a substance is mainly eluted which migrates somewhat more slowly than the starting material in the paper chromatogram. It is 1-dehydrocorticosterone, which is recrystallized from acetone. F. = 216 to 2200 (decomposition), lab = +1580 (alcohol). In the UV the substance absorbs at <I> 244 </I> mu <I> (8 = </I> 15,300) and shows in the IR bands at 2.76, 2.86, 5.84, 5.99 , 6.14, 6.22, 7.44, 8.73, 9.22, 9.38 and 9.63, and the like.
The 21-acetate produced by means of pyridine acetic anhydride at 200 melts at 159 to 161; [ab = + 1510 (dioxane), UV absorption spectrum AmOX <I> 243 </I> with (e <I> = </I> 14 800).
<I> Example 14 </I> To 4 liters of a culture of Calonectria decora, which is grown as described in Example 1, a solution of 1 g of 17a-oxycortexon (Reichstein's substance S) in 25 cm3 is added under sterile conditions Methanol. It is allowed to shake for 3 days at 270 and then the mycelium is filtered off. The extraction of the culture filtrate and the chromatography of the crude extract on silica gel is carried out as described in Example 1.
The chloroform and chloroform-acetone (97.5: 2.5) fractions contain impurities, while some starting material is eluted with chloroform-acetone (95: 5). The chloroform-acetone <B> (90: </B> 10) fractions consist of a mixture which, in addition to little starting material and higher polar products, mainly contains 1-dehydro-17a-oxy-cortexon.
This is obtained pure by crystallization from acetone; F. = 227 to 2330 (decomposition); [a] D = +760 (ethanol); UV absorption spectrum: i ", aX 244 my (a = 16,200).
In the IR there are bands u. a. at 2.76, 2.85, 5.84, 6.00, 6.15, 6.23, 9.01, 9.20, 9.55 and 10.04, and the like. The 21-acetate prepared by means of pyridine acetic anhydride melts at 216 to 222, Lah = +86 (dioxane); UV absorption spectrum: AmOX 244 mu 15 400).
<I> Example 15 </I> A solution of 1 g of progesterone in 25 cm 3 of acetone is added under sterile conditions to 4 liters of a culture of Calonectria decora which is grown as described in Example 1. The mixture is shaken at 270 for 24 hours and the mycelium is then filtered off. The extraction of the culture filtrate is carried out as described in Example 1.
The crude extract is chromatographed on a column of 30 g of alkali-free aluminum oxide, the individual fractions (100 cm3 each) being examined by paper chromatography. The petroleum ether-benzene fractions contain starting material, while benzene mainly elutes 1-dehydro-progesterone.
This is crystallized from ether-petroleum ether, F. = 151 to 152; [a] D = +1200 (ethanol); UV absorption spectrum: @ m0 @ 245 m, u (a = 16 150).
In the IR there are bands u. a. at 5.86, 5.99, 6.14, 6.23, 7.23, 7.38, 8.33, 8.62, 10.55 and 12.24, and the like. <I> Example 16 </I> In a shaking vessel, 1 liter of a nutrient solution containing 15 g peptone, 10 cm3 corn spring water (Cornsteepliquor), 0.1 g raw glucose and the rest of the tap water is adjusted to p $ 6.8 and sterilized. Calonectria decora is then inoculated with a culture.
The mixture is shaken at 270 for 48 hours and then a solution of 250 mg of cortisone in 8 cm3 of methanol is added to the well-developed culture under sterile conditions. It is shaken further at 270, the mycelium is filtered off after 3 days and the culture filtrate is extracted as described in Example 1 with ethyl acetate. The paper chromatographic examination of the crude extract shows that, in addition to the slightly polar components, only 1-dehydrocortisone is present, but no starting material.
The 1-dehydrocortisone is obtained in pure form by means of crystallization from acetone-ether and from methanol.
