Verfahren zur Herstellung von 17-Oxy-steroiden Es ist bereites bekannt, mittels Enzymen aus Nebennieren, insbesondere mit deren Ilomogenaten oder auch bei ihrer Durchströ- mung, Hydroxylgruppen, u. a. in 17a-Stellung von Steroiden einzuführen.
So interessant diese Funktion tierischer Enzyme vom theo retischen Standpunkt aus ist, so vermag sie zur präparativen und speziell industriellen Gewinnung von 17a-Oxy-Steroiden nicht zu befriedigen. Mit mikrobiologisehen Methoden, die sich auch in grösstem Stile anwenden las sen, ist die genannte Reaktion bisher nicht durchführbar gewesen.
Es wurde nun gefunden, dass 17-Oxy- steroide hergestellt werden können, wenn man auf Verbindungen der Pregnanreihe, die in 17-Stellung ein Wasserstoffatom aufweisen, durch aerobe Kulturen von Pilzen der Ord nung ivIoniliales, vorzugsweise der Art. Tri- ehothecium roseum, produzierte Enzyme ein wirken lässt.
Ausgangsstoffe für das neue Ver fahren sind allgemein Verbindungen der Pregnanreihe, wie Pregnane, Allopregnane, Isopregnane, wie 14- oder 17-Isopregnane, so wie Homo-. und Nor-pregnane, z.
B. A-Nor-, D-Homo- und 19-N or-pregnane, die in 17- Stellung ein Wasserstoffatom aufweisen. Die Verbindungen können gesättigt oder unge sättigt, sein und beliebige Substituenten auf weisen. Doppelbindungen befinden sich z. B.
in 1-, 4-, 5-, 6-, 7-, 9-, 14-, 15- und/oder 20- Stellung. Die Konfiguration der Ausgangs- stoffe ist vorzugsweise diejenige des Pregnans, 5a-Pregnans, 17a-Pregnans oder entsprechen der Racemate, wie sie bei der Totalsynthese erhalten werden.
Als Substituenten kommen besonders. in Frage freie bzw. funktionell ab gewandelte Hydroxyl=, Oxo- oder Carboxyl- grup:pen, wie Ester-, Äther-, Thioester-, Thio- äther-, Thiol- und Thronester-, Acetal-, Mer- captal-, Ketal-, Hydrazon-, Semicarbazon- und Enolgruppen, z.
B. in 3-, 7-, 11-, 12-, 16-, 18-, 1.9-, 20- und 21-Stellung. Ausgangsstoffe sind u. a. Cortexon, 9,11- oder 11,12-Dehydro-cor- texon, 16a-Oxy-cortexon, 18-Oxo- und 18-Oxy- cortexon, Corticosteron, 11-Epi-corticost.eron, 11-Dehydro-corticosteron,18-Oxy-corticosteron, Progesteron, 17a-Progesteron, 11-Keto-Pro- gesteron,
11a- sowie 11ss-Oxy Progesteron, 9,11- oder 11.,12-Dehydro-progesteron, 19-Oxo-pro- gesteron, 19-Nor-progesteron, Pregnenolon, 21- Oxy-pregnenolon, Aldosteron bzw. ihre funk tionellen Derivate.
Die verwendeten Enzyme sind mikrobio logischer Herkunft und durch aerobe Kul turen von Pilzender Ordnung Moniliales, vor- zugsweise d-er Art Trichot.hecium roseum, pro duziert. Zur Einführung von Sauerstoff in kubiert man meist direkt die Ausgangsstoffe mit den unter an sich bekannten aeroben Be dingungen submers wachsenden Kulturen der genannten Pilze.
Diese werden zweckmässig bewegt., d. h. geschüttelt oder gerührt, und enthalten assimilierbaren Kohlenstoff, insbe- sondere Kohlenhydrate sowie gegebenenfalls Wuchsstoffe, beispielsweise Maisquellwasser oder Bierwürze, und anorganische Salze. Es sind somit natürliche, synthetische oder halb synthetische Nährlösungen brauchbar.
Das praktisch einfachste Verfahren ist im folgen den geschildert: Man züchtet die Organismen in Apparaturen und unter ähnlichen Bedin gungen wie sie bei der Antibiotika-Fabxika- tion als sog. T.ieftankverfahren bekannt sind. Nach Entwicklung der Kulturen gibt man die genannten Ausgangsstoffe in feiner Disper sion oder Lösung, beispielsweise in Methanol, Aceton oder Äthylenglykal zu und inkubiert weiter.
Schliesslich trennt man vom Mycel ab, extrahiert das Filtrat und/oder die Myeel- masse und isoliert aus dem Extrakt die Re- aktionsprodukte in an sich bekannter Weise, z.
B. durch Entmischungsverfahren, Adsorp- tion, Chromatographie, Kristallisation, Über führung in funktionelle Derivate, wie Girard- Verbindungen und dergleichen.
Dieselben Um setzungen lassen sich aber auch durchführen, indem man die wirksamen Enzyme aus ent sprechenden aeroben Kulturen von Pilzen der Ordnung Moniliales, wie der Art Tricho- thecium roseum, zuerst abtrennt und unter Ausschluss der wachsenden Kulturen verwen det.
Die Verfahrensprodukte können als Heil mittel Verwendung finden oder als Zwischen produkte zu ihrer Herstellung dienen. Von grösstem praktischem Interesse sind z. B. Cor- tison., Hydrocortison, 1,2-Dehydrocortison, 1,2- Dehydro-hydrocortison, Reichsteins Substanz S, 1.7a-Oxy-aldosteron sowie die z.
B. aus Pro gesteron., Pregnenolon und 11-Ketoprogesteron entstehenden polyoxygenierten Derivate.
In den nachfolgenden Beispielen besteht zwischen Gewichtsteil und Volumteil die gleiche Beziehung wie zwischen Gramm und Kubikzentimeter. Die Temperaturen sind in Celsiusgraden angegeben. <I>Beispiel 1</I> 2000 Volumteile einer sterilisierten Czapek- Dox-Nährlösung werden mit einer Kultur von Trichothecium roseum beimpft und 5 Tage bei 2711 geschüttelt.
Dann gibt man zur gut entwickelten Kultur unter sterilen Bedingun gen eine Lösung von 0,5 Gewichtsteilen Cor- texon in 15 Volumteilen Aceton zu und schüt telt bei 27 weiter. Nach 4 Tagen nutseht man ab, wäscht den Rückstand einige Male mit Wasser und extrahiert das Kulturfiltrat 3mal mit je 1200 Volumteilen Essigester.
Die Essig-! esterlösungen werden mit ln-Salzsäure, Was ser, 1n-Natriumbicarbonatlösung und Wasser gewaschen, getrocknet und bei 40-50 im Vakuiun eingedampft. Der 0,62 Gewichtsteile betragende Rückstand wird an 10 Gewichts-, teilen Silicagel chromatographiert, wobei zi erst mit Chloroform allein, dann mit Chloro- form-Aceton-Gemischen mit steigendem Ace- ton:gehalt eluiert, wird.
Mit Chloroform und Chloroform-Aceton <B>(95:5)</B> werden wenig Cor- texon enthaltende Verunreinigungen eluiert. Die ersten Chloroformfraktionen mit 1011/o Aceton geben beim Eindampfen einen Rück stand, der mit. konz. Schwefelsäure die für Substanz S (17a-Oxy-cortexon) charakteri stische karminrote Färbung gibt..
Dieser Rück stand wird aus Aceton-Äther-Gemischen um- kristallisiert, wobei reine Substanz S vom F. = 202-213 und der spezifischen Drehung [a]D = -I-132 (in Äthanol) erhalten wird. Zur Acetylierung werden 0,02 Gewichtsteile der reinen Substanz mit 0,2 Volumteilen Pyri- din und 0,4 Volumteilen Acetanhydrid 15 Stunden bei 20 stehengelassen.
Das Gemisch versetzt man mit. Wasser, dampft im Vakuum ein und kristallisiert den Rückstand aus Ace ton um, wobei Substanz S-acetat vom F. = 239 bis 241 erhalten wird.
Die weiteren Chl.oroformeluate mit 101/o Aceton geben beim Eindampfen eine etwas stärker polare Verbindung, die mit konz. Schwefelsäure zuerst eine rötliche, dann braungelbe Färbung mit grüner Fluoreszenz zeigt. Diese Substanz kristallisiert aus Aceton in klaren Prismen, die beim Erwärmen ab etwa 130 verwittern und bei 202-212 schmel zen und im Gemisch mit Substanz S eine starke Depression .des Schmelzpunktes er geben.
Die bei 135 im Hochvakuum getrock neten Kristalle geben Analysenwerte, die mit der Bruttoformel 021H3,04 gut übereinstim men. Da die Substanz im UV absorbiert. und alkoholische Silberdiaminlösung reduziert, handelt es sich hier um ein neues Oxy-eorte- xon, das von Substanz S verschieden ist.
An Stelle von Czapek-Dox-Nährlösung kön nen auch mit Wasser verdünnte oder unver dünnte Bierwürze oder andere Nährmedien verwendet werden, die auf je 2000 Volum- teileWasser die folgenden Zusätze enthalten:
Lösung <I>A:</I> 20 Gewichtsteile Glukose, 8 Ge wichtsteile sek. Ammoniumphosphat, 10 Ge- wichtsteile Na.triumchlorid, 4 Gewichtsteile sek.
Ka.liumphosphat, 2 Gewichtsteile krist. Magnesiumsulfat, 0,8 Gewichtsteile Caleium- chlorid, 0,04 Gewichtsteile krist. Eisen-(II)- sulfat, 0,02 Gewichtsteile krist. Mangan-(II)- sulfat.
Lösung <I>B:</I> 40 Volumteile Maisquellwasser (Cornsteepliquor), 20 Gewichtsteile Rohglu- kose, 4 Gewichtsteile sek. Kaliumphosphat.
<I>Lösung C: 20</I> Gewichtsteile Pepton, 200 Gewichtsteile Glukose, 4 Gewichtsteile prim. Ammoniumphosphat, 4 Gewichtsteile Ka,lium- nitrat, 1 Gewichtsteil krist. Magnesiumsulfat, 0,2 Gewichtsteile Cal'eiumchlorid.
<I>Lösung D:</I> 20 Gewichtsteile Pepton, 7 Ge wichtsheile Fleischextrakt, 80 Gewichtsteile Glukose, 15 Gewichtsteile Natriumchlorid.
Beispiel N 50 Volumteile einer sterilisierten Czapek- Dox-Nährlösun:g werden mit Triehothecium roseum beimpft und 5 Tage bei 27 geschüt telt.. Die gut entwickelte Kultur versetzt man unter sterilen Bedingungen mit einer Lösung von 0,01 Gewichtsteil 11-Dehydro-corticosteron (11-Keto-cortexon) in 0,5 Volumteilen Aceton und schüttelt bei 27 weiter. Nach 3 Tagen wird wie in Beispiel 1 beschrieben abgenutscht und extrahiert.
Der Extraktionsrückstand ent hält auf Grund der papierehromatographi- schen Untersuchung neben wenig Ausgangs material zur Hauptsache Cortison (11-Keto- 17a-oxy-eortexon), das sieh nach an sich be kannten Methoden isolieren und reinigen lässt. Eine analoge Umsetzung erhält man nach 6tägiger Inkubation des 11-Dehydro-cortico- sterons.
Beispiel <I>3</I> Eine wie in Beispiel 2 bereitete Tricho- theeium-roseum-Kultur wird mit einer Lösung von 0,01 Gewichtsteil Corticosteron in 0,5 Vo- lumteilen Aceton versetzt und 3 Tage bei 27 inkubiert und analog den Angaben in Bei spiel 2 aufgearbeitet. Die papierchromatogra- phzsche Untersuchung des Extraktes zeigt, dass der Ausgangsstoff grösstenteils in Sub stanz M (17a-Oxy-cortieosteron) umgewandelt ist.
Werden ' analog den obigen Angaben an Stelle von Corticosteron, Progesteron, 11-Keto- progesteron oder Pregnenolon inkubiert, so erhält man nach der Aufarbeitung ebenfalls grösstenteils höherpolare Verbindungen.
<I>Beispiel 4</I> Eine wie in Beispiel 2 bereitete Kultur von Trichothecium roseum wird: mit einer Lösung von 0,01 Gewichtsteil d1.4-3,11,20-Tri- keto-21-oxy-pregna,dien in 0,5 Volumteilen Aceton versetzt und 3 Tage bei 27 inkubiert. Die Aufarbeitung erfolgt analog den Angaben in Beispiel 2.
Die -Untersuchung des Extraktes mittels Papierchromatographie zeigt, dass der Ausgangsstoff grösstenteils in Al,4-3,11,20-Tri- heto-17a,21-dioxy=pregnadien (1-Dehydro-cor- tison) inngewandelt worden ist.
Wird analog den obigen Angaben an Stelle von 4I.4-3,11,20-Triketo-21-oxy-pregnadien das A1,4-3,20-Diketo-11ss,21-dioxy-pregna@dien in- kubiert, so erhält man nach der Aufarbeitung zur Hauptsache das dl,4-3,20-Diketo-11fl,17u, 1-t.rioxy-pregnadien (1rDehydro-hydrocorti- son).
Das Ausgangsmaterial kann folgenderma ssen gewonnen werden: dl,4-3,11,20-Triketo-21-oxy-pregnadien 2000 V olumteile sterilisierte Bierwürze wer den mit einer Kultur von Calonectria decora geimpft und bei 27 geschüttelt.
Nach 3 Tagen werden unter sterilen Bedingungen zu der gut entwickelten Kultur 0,5 Gewichtsteile 11-De- hydro-corticosteron (Verbindung A) in 15 .Volumteilen Aceton zugegeben;
sodann wird das Gemisch bei derselben Temperatur für 3 weitere Tage geschüttelt. Das Mycel wird abfiltrzert und das Kulturfiltrat wird in Bei spiel 1 extrahiert. Der Extraktionsrückstand wird an einer Säule von 15 Gewichtsteilen S:
ilicagel chromatographiert, wobei zuerst mit Chloroform, sodann mit Chloroform-Aceton- Mischungen mit steigendem Acetongehalt eluiert wird. Jede Fraktion (50 Volumt.cile) wird papierehromatographisch untersucht.
Mit Chloroform und Chloroform-Aceton (95:5) werden einige Verunreinigungen und etwas Ausgangsmaterial eluiert. Die Chloroform- Aceton <B>(90:
</B> 10-) -Fraktionen enthalten jedoch hauptsächlich eine Substanz, welche im Pa- pierchromatogTamm etwas langsamer läuft als das Ausgangsmaterial. Es handelt sich um das d1,4-3,11,20-Triketo-21-oxy-pregn.a.dien. Es wird aus Aceton-Äther umkristallisiert und schmilzt dann bei 200-215 , je nach Heiz geschwindigkeit.
Im UV zeigt die Verbindung eine Absorptionsbande mit Maximum bei 244 mu und im IR-Spektrum Banden bei 5,99,u, 6,14 ,u und 6,23 it, welche charakte ristisch für 41,4-3-Ketone sind.
d1.4-3,20 - Diketo-11ss,21- dioxy-pregnadien Zu 2000 Volumteilen einer Kultur von Calo- nectria decora., welche wie oben dargestellt ist, werden unter sterilen Bedingungen 0,5 Ge- wichtssteile Corticosteron in 15 Volumteilen Aceton gegeben.
Die Züchtung, Extraktion und Chromatogra-phie an Silicagel werden wie oben angegeben ausgeführt. Die Chloroform und Chloroformaceton (90 :10)-Fraktionen enthalten Verunreinigungen und etwas Aus- gangsmaterial. Die Hauptverbindung, welche in den Chlioroformaceton(90, :
10)- und (80 20)-Fraktionen enthalten ist, läuft im Papier- chromatogramrn etwas langsamer als das Aus- ga-ngsmaterial. Es handelt sich um 41,4-3,20- Diketo-11ss,21-dioxy-pregnadien, das aus Ace ton umkristallisiert wird.
Es schmilzt bei 216 bis 220 (unter Zersetzung) und besitzt die optische Drehung -f- 158 (Äthanol) ; im UV- Absorptionsspektrum weist es ein Maximum bei 244 mu (e 15300) und im IR-Absorptions- spektrum Banden bei 2,76 ,u, 2,86,u, 5,84,u, 5,99,u, 6,14,u, 6,22,u, 7,44 1t, 8,73,u, 9,22,u,
9,38,u und 9,63 ,cc auf.
Process for the production of 17-oxy-steroids It is already known to use enzymes from adrenal glands, in particular with their ilomogenates or even when flowing through them, hydroxyl groups and the like. a. to introduce in 17a position of steroids.
As interesting as this function of animal enzymes is from a theoretical point of view, it cannot be satisfactory for the preparative and especially industrial production of 17α-oxy-steroids. With microbiological methods that can also be applied on a large scale, the aforementioned reaction has not yet been feasible.
It has now been found that 17-oxy steroids can be prepared if compounds of the pregnane series, which have a hydrogen atom in the 17-position, are obtained by aerobic cultures of fungi of the order ivIoniliales, preferably of the species Triethecium roseum, produced enzymes.
Starting materials for the new process are generally compounds of the pregnane series, such as pregnanes, allopregnanes, isopregnanes, such as 14- or 17-isopregnanes, such as homo-. and nor-pregnane, e.g.
B. A-nor-, D-homo- and 19-N or-pregnanes, which have a hydrogen atom in the 17-position. The compounds can be saturated or unsaturated, and have any substituents. Double bonds are z. B.

11a- and 11ss-oxy progesterone, 9,11- or 11th, 12-dehydro-progesterone, 19-oxo-progesterone, 19-nor-progesterone, pregnenolone, 21-oxy-pregnenolone, aldosterone or their functional ones Derivatives.
The enzymes used are of microbiological origin and produced by aerobic cultures of fungi of the order Moniliales, preferably of the species Trichot.hecium roseum. To introduce oxygen, the starting materials are usually directly incubated with the cultures of the fungi mentioned that grow submerged under aerobic conditions known per se.
These are expediently moved, d. H. shaken or stirred, and contain assimilable carbon, in particular carbohydrates and possibly growth substances, for example corn steep liquor or beer wort, and inorganic salts. Natural, synthetic or semi-synthetic nutrient solutions can therefore be used.
The simplest procedure in practice is described below: The organisms are grown in apparatus and under conditions similar to those known in antibiotic fabrication as the so-called deep tank process. After the cultures have developed, the starting materials mentioned are added in a fine dispersion or solution, for example in methanol, acetone or ethylene glycol, and the incubation continues.
Finally, the mycelium is separated off, the filtrate and / or the myelial mass are extracted and the reaction products are isolated from the extract in a manner known per se, e.g.
B. by segregation processes, adsorption, chromatography, crystallization, transfer into functional derivatives such as Girard compounds and the like.
However, the same implementations can also be carried out by first separating the active enzymes from corresponding aerobic cultures of fungi of the order Moniliales, such as the species Trichothecium roseum, and using them to the exclusion of the growing cultures.
The products of the process can be used as medicinal products or as intermediate products for their manufacture. Of greatest practical interest are z. B. Cortison., Hydrocortisone, 1,2-dehydrocortisone, 1,2-dehydro-hydrocortisone, Reichstein's substance S, 1.7a-oxy-aldosterone and the z.
B. from Pro gesteron., Pregnenolone and 11-ketoprogesterone resulting polyoxygenated derivatives.
In the examples below, the relationship between part by weight and part by volume is the same as that between grams and cubic centimeters. The temperatures are given in degrees Celsius. <I> Example 1 </I> 2000 parts by volume of a sterilized Czapek Dox nutrient solution are inoculated with a culture of Trichothecium roseum and shaken at 2711 for 5 days.
A solution of 0.5 parts by weight of Cortexon in 15 parts by volume of acetone is then added to the well-developed culture under sterile conditions and the mixture is shaken at 27. After 4 days it is removed, the residue is washed a few times with water and the culture filtrate is extracted 3 times with 1200 parts by volume of ethyl acetate each time.
The vinegar! Ester solutions are washed with 1N hydrochloric acid, water, 1N sodium bicarbonate solution and water, dried and evaporated at 40-50 in a vacuum. The residue, amounting to 0.62 parts by weight, is chromatographed on 10 parts by weight of silica gel, eluting first with chloroform alone, then with chloroform-acetone mixtures with increasing acetone content.
Impurities containing a small amount of cortexon are eluted with chloroform and chloroform-acetone <B> (95: 5) </B>. The first chloroform fractions with 1011 / o acetone give a residue on evaporation, which was with. conc. Sulfuric acid which gives substance S (17a-Oxy-cortexon) the characteristic carmine-red color.
This residue is recrystallized from acetone-ether mixtures, pure substance S of F. = 202-213 and the specific rotation [a] D = -I-132 (in ethanol) being obtained. For acetylation, 0.02 parts by weight of the pure substance with 0.2 parts by volume of pyridine and 0.4 parts by volume of acetic anhydride are left to stand at 20 for 15 hours.
The mixture is mixed with. Water, evaporated in vacuo and the residue crystallized from Ace ton, substance S-acetate of F. = 239 to 241 is obtained.
The other chloroform eluates with 101 / o acetone give a somewhat more polar compound on evaporation, which with conc. Sulfuric acid first shows a reddish, then brownish-yellow color with green fluorescence. This substance crystallizes from acetone in clear prisms, which weather when heated from about 130 and melt at 202-212 and when mixed with substance S a strong depression of the melting point gives it.
The crystals dried at 135 in a high vacuum give analytical values which agree well with the gross formula 021H3.04. Because the substance absorbs in the UV. and alcoholic silver diamine solution, this is a new oxy-eortexon that is different from substance S.
Instead of Czapek-Dox nutrient solution, wort diluted or undiluted with water or other nutrient media that contain the following additives for every 2000 parts by volume of water can be used:

Solution <I> B: </I> 40 parts by volume of corn steep liquor (Cornsteepliquor), 20 parts by weight of raw glucose, 4 parts by weight of sec. Potassium phosphate.
<I> Solution C: 20 </I> parts by weight peptone, 200 parts by weight glucose, 4 parts by weight prim. Ammonium phosphate, 4 parts by weight of potassium nitrate, 1 part by weight of crystalline. Magnesium sulfate, 0.2 part by weight of calcium chloride.
<I> Solution D: </I> 20 parts by weight of peptone, 7 parts by weight of meat extract, 80 parts by weight of glucose, 15 parts by weight of sodium chloride.
Example N 50 parts by volume of a sterilized Czapek Dox nutrient solution: g are inoculated with Triehothecium roseum and shaken for 5 days at 27. The well-developed culture is treated under sterile conditions with a solution of 0.01 part by weight of 11-dehydrocorticosterone ( 11-keto-cortexon) in 0.5 parts by volume of acetone and continue shaking at 27. After 3 days, suction and extraction are carried out as described in Example 1.
Due to the paper chromatographic examination, the extraction residue contains little starting material, mainly cortisone (11-keto-17a-oxy-eortexon), which can be isolated and purified using known methods. An analogous reaction is obtained after incubating the 11-dehydrocortico-sterone for 6 days.
Example 3 A Trichotheum roseum culture prepared as in Example 2 is mixed with a solution of 0.01 part by weight of corticosterone in 0.5 part by volume of acetone and incubated for 3 days at 27 and analogously to the Details in Example 2 worked up. The paper chromatographic examination of the extract shows that the starting material has largely been converted into substance M (17a-oxy-cortieosterone).

Working up is carried out analogously to the information in Example 2.
Examination of the extract by means of paper chromatography shows that the starting material has largely been converted into Al, 4-3,11,20-trieto-17a, 21-dioxy = pregnadiene (1-dehydro-cortisone).
If the A1,4-3,20-diketo-11ss, 21-dioxy-pregna @ diene is incubated in place of 4I.4-3,11,20-triketo-21-oxy-pregnadiene analogously to the above information, then After working up, the main thing obtained is the dl, 4-3,20-diketo-11fl, 17u, 1-t.rioxy-pregnadiene (1r-dehydro-hydrocortisone).
The starting material can be obtained as follows: dl, 4-3,11,20-triketo-21-oxy-pregnadiene 2000 volumetric sterilized wort are inoculated with a culture of Calonectria decora and shaken at 27.
After 3 days, under sterile conditions, 0.5 parts by weight of 11-dehydro-corticosterone (compound A) in 15 parts by volume of acetone are added to the well-developed culture;
then the mixture is shaken at the same temperature for 3 more days. The mycelium is filtered off and the culture filtrate is extracted in Example 1. The extraction residue is placed on a column of 15 parts by weight of S:
ilicagel chromatographed, eluting first with chloroform and then with chloroform-acetone mixtures with increasing acetone content. Each fraction (50 Volumt.cile) is examined by paper chromatography.
Some impurities and some starting material are eluted with chloroform and chloroform-acetone (95: 5). The chloroform acetone <B> (90:
</B> 10-) fractions mainly contain one substance, which runs a little slower in the paper chromatograph than the starting material. It is the d1,4-3,11,20-triketo-21-oxy-pregn.a.dien. It is recrystallized from acetone-ether and then melts at 200-215, depending on the heating speed.
In the UV the compound shows an absorption band with a maximum at 244 mu and in the IR spectrum bands at 5.99, u, 6.14, u and 6.23 it, which are characteristic of 41.4-3-ketones.
d1.4-3.20 - Diketo-11ss, 21-dioxy-pregnadiene To 2000 parts by volume of a culture of Calonectria decora., which is shown as above, 0.5 parts by weight of corticosterone in 15 parts by volume of acetone are added under sterile conditions given.
The cultivation, extraction and chromatography on silica gel are carried out as indicated above. The chloroform and chloroform acetone (90:10) fractions contain impurities and some starting material. The main compound, which in the chloroformacetone (90,:
10) and (80 20) fractions runs somewhat slower in the paper chromatogram than the starting material. It is 41.4-3.20-diketo-11ss, 21-dioxy-pregnadiene, which is recrystallized from acetone.
It melts at 216 to 220 (with decomposition) and has the optical rotation -f- 158 (ethanol); in the UV absorption spectrum it has a maximum at 244 mu (e 15300) and in the IR absorption spectrum it has bands at 2.76, u, 2.86, u, 5.84, u, 5.99, u, 6, 14, u, 6.22, u, 7.44 1t, 8.73, u, 9.22, u,
9.38, u and 9.63, cc.
