Gewisse der erfindungsgemäss erhältlichen neuen Steroide sind Vorläufer bekannter Steroide, insbe sondere der physiologisch aktiven Nebennieren rindenhormone, llss,17a,21-Trioxy-4-pregnen-3,20- dion (Kendalls Verbindung F) und 17a,21-Dioxy-4- pregnen-3,11,20-trion (Kendalls Verbindung E) so wie der Ester dieser Verbindungen, die durch eine Reihe von Reaktionen gemäss den in der Zeichnung dargestellten Formeln erhältlich sind.
Dieses Formelschema zeigt anhand von Beispie len wie die Ausgangsstoffe für das vorliegende Ver fahren (1I und II') zugänglich sind und nach dem erfindungsgemässen Verfahren in die 21,21-Dihalo- gen-steroide (11I und III') übergeführt werden kön nen, sowie deren weitere Umwandlung in die be kannten Verbindungen F oder E von Kendall. In diesen Formeln bedeuten:
M = H, Alkalimetall X = Halogen R = H, -COO-Kohlenwasserstoff R', R" = H, Kohlenwasserstoff R" = H, Acylrest n = 1 oder 2 Ac = Acylrest So ergibt die Weiterverarbeitung eines 11-Keto- 21,21-dihalogen-21-alkoxyoxalyl-progesterons (III', R = COO-Alkyl) oder eines 11-Keto-21,21-dihalo- gen-21-formyl-progesterons (III', R = H) durch Be handlung mit einem Alkalimetallalkoholat, z.
B. Na- triummethylat, einen 3,11-Diketo-4,17(20)-pregna- dien-21-säure-alkylester (IV', R' = Alkyl). Die Be handlung einer dieser Verbindungen nach Blockie rung der 3-Ketogruppe durch Überführung in eine Ketalgruppe, z.
B. eine Athylenglykolketalgruppe (V', R' = Alkyl, R" - H, n = 1), oder in eine Alkylenoläthergruppe oder dergleichen, mit einem zur Reduktion eines Carbonsäureesters und eines Ketonsauerstoffes befähigten Reduktionsmittel, z. B.
Li-Al-Hydrid oder andern Alkalimetall-Al-Hydriden, und anschliessende Hydrolyse des Reaktionspro duktes (VI) ergibt llss,21-Dioxy-4,17(20)-pregna- dien-3-on (VII). Diese Verbindung oder deren 21- Ester lassen sich leicht in 11ss,17a,21-Trioxy-4- pregnen-3,20-dion (IX, R' = H) (Kendalls Ver- bindung F) überführen, indem man mit Osmium- tetroxyd den 11ss,
17a,20,21-Tetraoxy-4-pregnen-3- on-osmiatester herstellt und anschliessend z. B. mit Perchlorsäure, deren Salzen oder andern äquivalen ten Oxydationsmitteln, wie H202, Dialkyloxyden, organischen Persäuren, wie Peressig- oder Perbenzoe- säure, in einem Lösungsmittel, wie einem Äther oder Alkohol, z.
B. tert. Butylalkohol oder Di- äthyläther, nach bekannten Methoden [Prins und Reichstein, Helv. Chim. Acta, 25, 300 (1942); Ruzicka und Müller, Helv. Chim. Acta 22, 755 (l939)] oxydiert.
In gleicher Weise erhält man ausgehend von einem 11-Oxy-21,21-dihalogen-21-alkoxyoxalyl- progesteron (III, R = COO-Alkyl) oder 11-Oxy- 21,21-dihalogen-21-formyl-progesteron (III, R = H) nach den gleichen Massnahmen wie oben, jedoch unter Oxydation der 11-Oxygruppe zur 11-Keto- gruppe, z.
B. mit Chromsäure, vorgängig der Hy- droxylierung mit Osmiumtetroxyd und der anschlie ssenden Oxydation 17a,21-Dioxy-4-pregnen-3,11,20- trion (X, R = H) (Kendalls Verbindung E).
Die als Ausgangsstoffe zu verwendenden 20-Keto- 21-acyl-steroide erhält man durch Umsetzung ent sprechender in 21-Stellung unsubstituierter 20-Keto- steroide mit einem Ester der Oxalsäure oder Amei sensäure in Gegenwart einer Alkalimetallbase bei Temperaturen zwischen etwa 0 C und dem Siede punkt der Reaktionsmischung, vorzugsweise in einem Lösungsmittel, wie z.
B. Toluol, Benzol, Me thanol, Äthanol, Mischungen derselben usw., wobei die Alkalienolate der angegebenen Formel entstehen (M = Alkalimetall). Beim Ansäuern der wässrigen Lösungen dieser Alkalienolate erhält man die freien Enole (M = Wasserstoff).
Die Ausgangsstoffe können im Kern substituiert sein, z. B. am Kohlenstoffatom 3 und/oder 11 durch eine a- oder ss-Oxygruppe oder eine in diese durch Hydrolyse oder sonstige Massnahmen überführbare Gruppe, wie eine Acyloxy- oder Alkoxygruppe oder dergleichen oder eine Ketogruppe. Sie können im Kern gesättigt sein oder eine oder mehrere Doppel bindungen enthalten.
Verwendet man als Ausgangsmaterial eine 21- Alkoxyoxalylverbindung, so enthält die Alkoxy- gruppe vorzugsweise 1-8 Kohlenstoffatome, wobei die Methoxy- und Äthoxygruppe bevorzugt werden. Von besonderem Interesse sind die Natriumenolate des lla-Oxy- und 11-Keto-21-formylprogesterons.
Die Halogenierung erfolgt üblicherweise in einem organischen Lösungsmittel, das unter den Re aktionsbedingungen inert ist. Methanol und Äthanol haben sich als besonders wertvoll erwiesen, und wer den bevorzugt, obschon man auch in Chloroform oder Methylenchlorid, in Gegenwart von Pyridin, Essigsäure in Gegenwart von Kaliumacetat und an dern Medien arbeiten kann. Da die Anwesenheit einer Base, z.
B. des Alkalimetallsalzes, einer alipha- tischen Säure oder der Kohlensäure, wie Natrium= karbonat-, -bikarbonat oder dergleichen, die Aus- beute zu erhöhen scheint, wird die Halogenierung in der Regel in Gegenwart einer basisch reagierenden Verbindung, speziell von Kalium- oder Natrium acetat, durchgeführt.
Wenn man als Halogenierungsmittel Brom ver wendet, so wird in der Regel ohne Verdünnungs mittel gearbeitet; doch kann man auch ein organi sches Lösungsmittel, wie Chloroform, Methylen- chlorid oder dergleichen, verwenden. Chlor kann man in die Reaktionsmischung einleiten oder in einem organischen Lösungsmittel gelöst zusetzen. Jod kann man in Methylenchlorid, Chloroform, Tetrachlorkohlenstoff, Äthanol gelöst oder in sonst einer geeigneten Form zusetzen.
Man verwendet pro Mol des Ausgangssteroids, wie gesagt, etwa 2 Mol Halogen. Die Anwendung wesentlich geringerer oder grösserer Mengen Halogen würde die Ausbeute vermindern. Man arbeitet zweckmässig bei etwa Zimmertemperatur, z. B. 20 bis 30 C, doch kann man bei 0-60 C arbeiten.
Die 21,21-Dihalogen-steroide können aus dem Reaktionsgemisch entweder durch Zusatz eines grossen Volumens Wasser, wenn das Lösungsmittel wasserlöslich ist, bei Zimmertemperatur oder darun ter ausgefällt werden, oder man kann das Lösungs mittel abdestillieren, wobei man einen Rückstand erhält, der nach dem Auswaschen mit Wasser die gewünschte Dihalogenverbindung darstellt.
Die folgenden Präparationen zeigen wie die Aus gangsprodukte erhalten werden können. Präparation <I>1</I> A. Zu einer Mischung von 3,4 cm-' 3,4n-metha- nolischer Natriummethylatlösung 20 cm- absolutem Äthanol und 20 cm?- trockenem Benzol, aus der zuerst 8 cm3 abdestilliert wurden, werden nach dem Abkühlen 2,3 cm-' Athyloxalat und eine Lösung von 3,28 g 11-Keto-progesteron in 38 cm- wasserfreiem Benzol gegeben. Die Lösung wird trüb, und es ent steht ein gelber Niederschlag.
Die Mischung wird 90 Minuten gerührt mit 55 cm3 Äther versetzt, wei tere 60 Minuten gerührt, und mit 130 cms Äther versetzt. Der so entstandene gelbe Niederschlag des Natriumenolats des 11 - Keto - 21 - äthoxyoxalyl- progesterons wird abfiltriert, mehrmals mit je 50 cm3 Äther gewaschen und wiegt nach dem Trocknen 3,65 g. Der Waschäther enthält 0,54g nicht um gesetztes 11-Keto-progesteron.
Die Ausbeute an Natriumenolat des 11-Keto-21- äthoxyoxalyl-progesterons beträgt 811/6 der Theorie und entspricht, berechnet auf das umgesetzte 11-Keto-progesteron der theoretischen. Das Vorhan densein des Natriumenolats wird durch die ausser ordentliche Wasserlöslichkeit und die positive Ferri- chloridprobe für Enole bestätigt.
Beim Ansäuern einer wässrigen Lösung des so erhaltenen Natriumenolats des 11-Keto-21-äthoxy- oxalyl-progesterons erhält man das freie Enol des 11-Keto-21-äthoxyoxalyl-progesterons, das durch Filtration isoliert werden kann. B.
Man arbeitet praktisch gleich wie unter Prä paration 1 beschrieben und erhält durch Umsetzung von 11-Keto-progesteron mit Äthyloxalat in Benzol und Kalium in tert. Butylalkohol das Kaliumenolat des 11-Keto-21-äthoxyoxalyl-progesterons.
In gleicher Weise wie in den Präparationen 1 <I>A</I> und<I>B</I> werden folgende Verbindungen durch Re aktion von 11-Keto-progesteron mit dem entspre chenden Alkyloxalat und Natrium- oder Kalium- alkoholat in Benzol, Toluol, Äthanol oder andern Lösungsmitteln erhalten:
Na-enolat des 11-Keto-21-methoxyoxalyl- progesterons, Na-enolat des 11-Keto-21-propoxyoxalyl- progesterons, Na-enolat des 11-Keto-21-butoxyoxalyl- progesterons, Na-enolat des 11-Keto-21-isobutoxyoxalyl- progesteron s, Na-enolat des 11-Keto-21-amyloxyoxalyl- progesterons,
Na-enolat des 11-Keto-21-hexyloxyoxalyl- progesterons, Na-enolat des 11-Keto-21-heptyloxyoxalyl- progesterons, Na-enolat des 11-Keto-21-octyloxyoxalyl- progesterons, und die Kaliumanalogen.
Durch Ansäuern der wässrigen Lösung eines der vorerwähnten Alkalimetallenolats erhält man einen Niederschlag des freien Enols der 21-Alkoxyoxalyl- verbindung, der durch Filtration isoliert werden kann.
Präparation <I>2</I> A. Zu einer Mischung von 3,4 cm3 einer 3,4n- Natriummethylatlösung in Methanol, 045 cm?, ab solutem Äthanol und 20 cm3 trockenem Benzol gibt man nach dem Abdestillieren von 8 cms und Erkal ten 2 cm3 Athylformiat und eine Lösung von 3,28 a- (0,01 Mol) 11-Keto-progesteron in 38 cm3 trocke nem Benzol.
Die Lösung wird trüb, und es entsteht ein gelber Niederschlag. Die Reaktionsmischung wird bei Zimmertemperatur eine Stunde gerührt, mit 60 cm3 Äther versetzt, weitere 90 Minuten gerührt und nochmals 60 cm3 Äther zugegeben. Der gelbe Niederschlag des Natriumenolats des 11-Keto-21- formyl-progesterons wird auf dem Filter gesammelt und mehrmals mit je 50 cm3 Äther ausgewaschen. Nach dem Trocknen wiegt er 2,35 g.
Der Wasch äther enthält 1,21 g nicht umgesetztes 11-Keto- progesteron. Die Ausbeute an Natriumenolat ist 6204 der Theorie oder, bezogen auf das umgesetzte Ausgangsmaterial praktisch quantitativ. Das Vor handensein des Natriumenolats wird durch die positive Ferrichloridreaktion für Enole bestätigt.
Die Struktur wird ferner durch Umwandlung des Enolats in das freie 11-Keto-21-formyl-progesteron vom Schmelzpunkt 85-90 C bestätigt, dessen Infrarotabsorptionsspektrum dieser Struktur ent spricht. B.
Unter Anwendung der gleichen Arbeits weise wie in Präparation 2 A wird 11-Keto-progeste- ron durch Umsetzung mit Äthylformiat und Kalium in tert. Butylalkohol in das Kaliumenolat des 11- Keto-21-formyl-progesterons übergeführt.
Wie in Präparation <I>2 A</I> und<I>B</I> beschrieben, er hält man durch Umsetzung der entsprechenden 20- Keto-steroide mit Äthylformiat oder andern Alkyl- formiaten in Gegenwart einer Natriumbase, vor zugsweise Natriumalkoholat, folgende Verbindun gen:
Na-enolat des 21-Formyl-4,16-pregnadien- 3,20-dions, Na-enolat des 3a-Oxy-21-formyl-pregnan- 11,20-dions (freies Enol, Schmelzpunkt 95 bis 102 C), Na-enolat des 3a,17a-Dioxy-21-formyl-pregnan- 11,20-dions, Na-enolat des 21-Formyl-4-pregnen-3,20-dions, Na-enolat des 16(17)-Oxydo-21-formyl-4-pregnen- 3,20-dions,
Na-enolat des 3a,l1a-Dioxy-21-formyl-pregnan- 20-ons, Na-enolat des 3a,17a-Dioxy-21-formyl-pregnan- 11,20-dions, und dergleichen. Die Kaliumenolate dieser und an derer Verbindungen werden erhalten, wenn man anstelle der oben verwendeten Natriumbase tert. Ka- liumbutylat verwendet.
Die freien Enole der oben genannten Verbindungen erhält man durch Ansäuern einer wässrigalkoholischen Lösung derselben mit Salzsäure oder einer andern Säure.
Die entsprechenden 21-Alkoxyoxalyl-derivate der oben genannten Verbindungen erhält man durch Er satz des Alkylformiats durch ein entsprechendes Alkyloxalat.
Präparation <I>3</I> A. 3,3 g IIa-Oxy-progesteron, Peterson und Murray, J. Am. Chem. Soc., 74, 1871 (1952), wer den in einer Lösung von 0,25g Na in 8 cms abso lutem Äthanol gelöst und mit 1,45g Äthyloxalat versetzt. Man lässt 6 Stunden bei Zimmertempera- tur stehen, wobei die Farbe von Gelb in Braun über geht.
Das so entstandene Natriumenolat des 11a-Oxy- 21-äthoxyoxalyl-progesterons wird durch Zusatz einer grossen Menge Äther als gelber amorpher Niederschlag, der sich oberhalb 200 C zersetzt, gewonnen.
Die Struktur der Verbindung wird durch die Infrarotanalyse bestätigt, welche folgende Absorp tionsspitzen zeigt: 1. starke Absorption bei etwa 1720 cm-', was im Bereich eines a,ss-ungesättigten Esters liegt, und so die Anwesenheit einer derartigen Gruppe anzeigt und den Erfolg der Reaktion bestätigt; 2. starke Absorption bei etwa 1631 cm-' und 1465 cm-', die für Metallenolate charakteristisch ist und zeigt, dass das Natriumenolat gebildet wurde;
3. starke Absorption bei etwa 1670 cm-', die für ein a,ss-ungesättigtes Keton charakteristisch ist und anzeigt, dass die A4-3-Ketogruppe des 11 a-Oxy- progesterons nicht verändert wurde und die Mög lichkeit der Kondensation in 2-Stellung ausschliesst. Die Kondensation erfolgt daher an der andern akti vierten Stellung, nämlich der 21-Stellung.
B. Unter Verwendung der gleichen Arbeitsweise wie unter Präparation 1 A beschrieben, wird lla- Oxy-progesteron in das Kaliumenolat des 11a, Oxy-21-äthoxyoxalyl-progesterons durch Umsetzung mit Äthyloxalat und Kalium in tert. Butylalkohol er halten.
In analoger Weise erhält man aus 11 a-Oxy- progesteron und dem entsprechenden Alkyloxalat mit Natrium- oder Kaliumalkoholat in Benzol die folgenden Verbindungen:
Na-enolat des lla-Oxy-21-methoxyoxalyl- progesterons, Na-enolat des 11 a-Oxy-21-propoxyoxalyl- progesterons, Na-enolat des lla-Oxy-21-butoxyoxalyl- progesterons, N_ a-enolat des lla-Oxy-21-amyloxyoxalyl- progesterons, Na-enolat des 11 a-Oxy-21-hexyloxyoxalyl- progesterons,
Na-enolat des lla-Oxy-21-heptyloxyoxalyl- progesterons, Na-enolat des lla-Oxy-21-octyloxyoxalyl- progesterons, und die Kaliumanaloga.
Durch Ansäuern einer wässrigen Lösung der oben genannten Verbindungen erhält man einen Nie derschlag aus dem freien Enol des lla-Oxy-21- alkoxyoxalyl-progesterons.
Präparation <I>4</I> A. Zu einer Mischung von 3,4 cm3 einer 3,4n- Natriummethylatlösung in Methanol, 0,45 cm3 ab solutem Äthanol und 20 cm3 trockenem Benzol, aus der vorher 8 cms abdestilliert wurden, gibt man nach dem Erkalten 2 cm3 Äthylformiat und eine Lösung von 3,26 g (0,01 Mol) lla-Oxy-progesteron in 38 cms trockenem Benzol.
Die Lösung wird trüb, und es bildet sich ein gelber Niederschlag. Man rührt eine Stunde bei Zimmertemperatur, gibt dann 60 cm3 Äther hinzu, rührt weitere 90 Minuten und versetzt mit 60 cm3 Äther. Der gelbe Niederschlag des Na- triumenolats des 11 a-Oxy-21-formyl-progesterons wird abfiltriert und mehrmals mit 50 cm3 Äther ge waschen.
Der Waschäther enthält nicht umgesetztes lla-Oxy-progesteron. Das Vorhandensein des Na- triumenolats ergibt sich aus der ausserordentlichen Löslichkeit des Produktes in Wasser und der posi tiven Ferrichloridprobe auf Enole. Die Struktur des Produktes wird ferner durch Umwandlung des Enolats in das freie llcc Oxy-21-formyl-progesteron bestätigt, dessen Infrarotabsorptionsspektrum mit der angenommenen Struktur übereinstimmt. B.
Nach der gleichen Arbeitsweise wird lla- Oxy-progesteron in das Kaliumenolat des lla-Oxy- 21-formyl-progesterons übergeführt, indem man es mit Äthylformiat und Kalium in tert. Butylalkohol umsetzt.
Säuert man eine wässrige Lösung dieses Kalium- enolats an, so fällt das freie Enol des lla-Oxy-21- formyl-progesterons aus. Präparation <I>5</I> A. 144 mg (6,25 Millimol) Natrium werden in 5 cm3 absolutem Äthanol unter Stickstoffatmosphäre gelöst, dieser Lösung 8 cm3 Benzol und 0,8 cm3 (0,9 g; 6,15 Millimol) Äthyloxalat zugesetzt.
Die Mischung wird mit Eiswasser gekühlt, eine Lösung von 1,99 g (6,03 Millimol) lla-Oxy-progesteron in 5 cm3 absolutem Äthanol zugefügt und mit 25 cm3 trockenem Benzol unter Rühren langsam vermischt. Man rührt 21/.z Stunden bei Zimmertemperatur, ver setzt mit 100 cm3 Äther, rührt eine Stunde weiter und gibt nochmals 100 cm3 Äther zu. Der entstan dene hellgelbe Niederschlag wird abfiltriert und mit Äther gewaschen. Ausbeute 1,68 g (6204).
Das Produkt wird durch Umwandlung in das be kannte Corticosteronacetat als Natriumenolat des llss-Oxy-21-äthoxyoxalyl-pro,esterons identifiziert. Es zeigte die folgende charakteristische Infrarot absorption: 1. starke Absorption bei etwa 1716 cm-' im Bereich eines a,ss-ungesättigten Esters;
2. starke Absorption bei etwa 1634 cm-' und 1470 cm-' charakteristisch für Metallenolate; 3. starke Absorption bei 1670 cm-' charakte ristisch für a,ss-ungesättigte Ketone, so dass diese Absorptionsbanden die theoretische Struktur bele gen.
<I>B.</I> Man arbeitet gleich wie unter<I>A</I> und erhält aus 11 ss-Oxy-progesteron mit Äthylformiat und Ka lium in tert. Butylalkohol das Kaliumenolat des llss- Oxy-21-formyl-progesterons.
Säuert man die Lösung des Kalium- oder Na- triumenolats an, so erhält man das freie Enol des 1 lss-Oxy-21-formyl-progesterons. <I>Beispiel 1</I> Zu einer Lösung von 4,50 g (0,01 Mol) des Na- enolats des 11-Keto-21-äthoxyoxalyl-progesterons und 2 g Kaliumacetat in 70 cm3 Eisessig tropft man bei Zimmertemperatur unter Rühren 3,09 g (1,00 cm3, 0,0193 Mol) Brom zu. Nach Beendigung der Reaktion mischt man mit einem grossen Volu men Wasser.
Die wässrige Schicht wird vom ausge schiedenen viskosen, gelben Produkt abdekantiert, letzteres in Alkohol gelöst und durch Zutropfen von Wasser als weisser fester Niederschlag ausgefällt. Nach dem Filtrieren und Trocknen beträgt die Aus beute an 11 - Keto - 21,21 - dibrom - äthoxyoxalyl- progesteron 4,0 g, was 70 1/n der theoretischen ent spricht.
In analoger Weise erhält man andere 11-Keto- 21,21 - dihalogen - alkoxyoxalyl - progesterone, z. B. solche, in denen die Alkoxygruppe Methoxy, Äthoxy, Propoxy, Butoxy, Amyloxy, Hexyloxy, Heptyloxy oder Octyloxy ist.
<I>Beispiel 2</I> In genau gleicher Weise wie im Beispiel 1 wird ein Enolat des 11-Keto-21-formyl-progesterons mit 2 Moläquivalenten Chlor in das 11-Keto-21,21-di- chlor-21-formyl-progesteron übergeführt.
In analoger Weise erhält man unter Verwendung von etwa 2 Moläquivalenten Brom bzw. Jod die ent sprechenden Dibrom- und Dijodverbindungen.
In gleicher Weise wie in den Beispielen 1 und 2 stellt man folgende Verbindungen her: 21,21-Dibrom-21-formyl-4,16-pregnadien- 3,20-dion, 3a-Oxy-21,21-dibrom-21-formyl-pregnan-11,20-dion (Schmelzpunkt 193-200 C), 3 a,17a-Dioxy-21,21-dijod-21-formyl-pregnan- 11,20-dion, 16(17)-Oxydo-21,21-dibrom-21-formyl-4-pregnen- 3,20-dion, 21,21-Dibrom-21-formyl-4-pregnen-3,20-dion, 3a,<B>11</B> a-Dioxy-21,21-dichlor-21-formyl-pregnan- 20-on, 3a,17a-Dioxy-21,
21-dijod-21-formyl-pregnan- 11,20-dion.
Die entsprechenden Alkoxyoxalylverbindungen, deren Alkoxygruppe 1-8 Kohlenstoffatome ent hält, erhält man in analoger Weise.
Die nachfolgenden Beispiele führen zu Produk ten, die besonders wertvoll sind als Vorstufen zum 17a,21-Dioxy-4-pregnen-3,11,20-trion (Kendalls Verbindung E).
<I>Beispiel 3</I> Zu einer Lösung von 4,52 g (0,01 Mol) des Na- triumenolats des 11a-Oxy-21-äthoxyoxalyl-progeste- rons in 150 cm3 Methanol tropft man unter Rühren 1 cm3 (0,02 Mol) Brom. Ersetzt man das Chlor durch Brom oder Jod, so erhält man die entsprechenden 21,21-Dibrom- bzw. Dijodverbindung.

The yellow precipitate of the sodium enolate of 11 - keto - 21 - ethoxyoxalyl progesterone thus formed is filtered off, washed several times with 50 cm3 of ether each time and, after drying, weighs 3.65 g. The washing ether contains 0.54 g of unreacted 11-keto-progesterone.
The yield of sodium enolate of 11-keto-21-ethoxyoxalyl-progesterone is 811/6 of theory and, calculated on the converted 11-keto-progesterone, corresponds to the theoretical one. The presence of the sodium enolate is confirmed by the extraordinary water solubility and the positive ferric chloride test for enols.
Acidification of an aqueous solution of the sodium enolate of 11-keto-21-ethoxy-oxalyl-progesterone obtained in this way gives the free enol of 11-keto-21-ethoxyoxalyl-progesterone, which can be isolated by filtration. B.
The procedure is practically the same as described under preparation 1 and obtained by reacting 11-keto-progesterone with ethyl oxalate in benzene and potassium in tert. Butyl alcohol the potassium enolate of 11-keto-21-ethoxyoxalyl-progesterone.

Preparation <I> 2 </I> A. To a mixture of 3.4 cm3 of a 3.4N sodium methylate solution in methanol, 045 cm ?, from absolute ethanol and 20 cm3 of dry benzene, after distilling off 8 cm3 and Erkal th 2 cm3 of ethyl formate and a solution of 3.28 a- (0.01 mol) 11-keto-progesterone in 38 cm3 of dry benzene.
The solution becomes cloudy and a yellow precipitate forms. The reaction mixture is stirred at room temperature for one hour, 60 cm3 of ether are added, the mixture is stirred for a further 90 minutes and another 60 cm3 of ether are added. The yellow precipitate of the sodium enolate of 11-keto-21-formyl-progesterone is collected on the filter and washed out several times with 50 cm3 of ether each time. After drying, it weighs 2.35 g.
The washing ether contains 1.21 g of unreacted 11-keto progesterone. The yield of sodium enolate is 6204 of theory or, based on the converted starting material, practically quantitative. The presence of the sodium enolate is confirmed by the positive ferric chloride reaction for enols.
The structure is further confirmed by the conversion of the enolate into the free 11-keto-21-formyl-progesterone with a melting point of 85-90 ° C. whose infrared absorption spectrum corresponds to this structure. B.
Using the same procedure as in preparation 2 A, 11-keto-progesterone is converted into tert. By reaction with ethyl formate and potassium. Butyl alcohol converted into the potassium enolate of 11-keto-21-formyl-progesterone.

Preparation <I> 3 </I> A. 3.3 g IIa-oxy-progesterone, Peterson and Murray, J. Am. Chem. The solution becomes cloudy and a yellow precipitate forms. The mixture is stirred for one hour at room temperature, then 60 cm3 of ether are added, the mixture is stirred for a further 90 minutes and 60 cm3 of ether are added. The yellow precipitate of the sodium enolate of 11 a-oxy-21-formyl-progesterone is filtered off and washed several times with 50 cm3 of ether.
The washing ether contains unreacted lla-oxy-progesterone. The presence of the sodium enolate results from the extraordinary solubility of the product in water and the positive ferric chloride test for enols. The structure of the product is further confirmed by conversion of the enolate into the free llcc oxy-21-formyl-progesterone, the infrared absorption spectrum of which agrees with the assumed structure. B.
In the same way, Ila-oxy-progesterone is converted into the potassium enolate of Ila-oxy-21-formyl-progesterone by adding ethyl formate and potassium to tert. Butyl alcohol.
If an aqueous solution of this potassium enolate is acidified, the free enol of the IIIa-oxy-21-formyl-progesterone precipitates. Preparation <I> 5 </I> A. 144 mg (6.25 millimoles) of sodium are dissolved in 5 cm3 of absolute ethanol under a nitrogen atmosphere, this solution 8 cm3 of benzene and 0.8 cm3 (0.9 g; 6.15 millimoles ) Ethyl oxalate added.
The mixture is cooled with ice water, a solution of 1.99 g (6.03 millimoles) of IIIa-oxy-progesterone in 5 cm3 of absolute ethanol is added and slowly mixed with 25 cm3 of dry benzene while stirring. The mixture is stirred for 21/2 hours at room temperature, mixed with 100 cm3 of ether, stirred for a further hour and another 100 cm3 of ether is added. The resulting pale yellow precipitate is filtered off and washed with ether. Yield 1.68 g (6204).
The product is identified by conversion into the known corticosterone acetate as sodium enolate of llss-oxy-21-ethoxyoxalyl-pro, esterone. It showed the following characteristic infrared absorption: 1. Strong absorption at about 1716 cm- 'in the region of an α, ß-unsaturated ester;
2. Strong absorption at around 1634 cm- 'and 1470 cm-' characteristic of metal enolates; 3. Strong absorption at 1670 cm- 'characteristic of a, ß-unsaturated ketones, so that these absorption bands prove the theoretical structure.
<I> B. </I> You work the same way as under <I> A </I> and you get from 11 ss-oxy-progesterone with ethyl formate and potassium in tert. Butyl alcohol the potassium enolate of llss-oxy-21-formyl-progesterone.
If the solution of the potassium or sodium enolate is acidified, the free enol of the 1 lss-oxy-21-formyl-progesterone is obtained. <I> Example 1 </I> A solution of 4.50 g (0.01 mol) of the Na enolate of 11-keto-21-ethoxyoxalyl-progesterone and 2 g of potassium acetate in 70 cm3 of glacial acetic acid is added dropwise at room temperature Stir in 3.09 g (1.00 cm3, 0.0193 mole) of bromine. In an analogous manner, the corresponding dibromine and diiodine compounds are obtained using about 2 molar equivalents of bromine or iodine.
The following compounds are prepared in the same way as in Examples 1 and 2: 21,21-dibromo-21-formyl-4,16-pregnadiene-3,20-dione, 3a-oxy-21,21-dibromo-21- formyl-pregnan-11,20-dione (melting point 193-200 C), 3 a, 17a-dioxy-21,21-diiodo-21-formyl-pregnan-11,20-dione, 16 (17) -oxydo-21 , 21-dibromo-21-formyl-4-pregnene-3,20-dione, 21,21-dibromo-21-formyl-4-pregnene-3,20-dione, 3a, <B> 11 </B> a -Dioxy-21,21-dichloro-21-formyl-pregnan- 20-one, 3a, 17a-Dioxy-21,

<I> Example 3 </I> To a solution of 4.52 g (0.01 mol) of the sodium enolate of 11a-oxy-21-ethoxyoxalyl-progesterone in 150 cm3 of methanol is added dropwise with stirring 1 cm3 ( 0.02 mol) bromine. The resulting IIIa-oxy-21,21-dibromo-21-ethoxyoxalyl-progesterone is isolated by pouring it into a large volume of water and separating off the precipitate.
