Verfahren zur Herstellung von 1,2-Diamino-cycloalkanen
Die vorliegende Erfindung bezieht sich auf ein neues Verfahren zur Herstellung von 1, 2- Diamino-cycloalkanen, welche beispielsweise wertvolle Ausgangsstoffe für Heilmittel für Tiere und Pflanzen, Schädlingsbekämpfungs- mittel und Textilhilfsmittel darstellen.
Es wurde überraschenderweise gefunden, dass man 1,2-Diamino-cycloalkane mit 6 bis 8 Ringgliedern entsprechend der Formel

Als Halogenwasserstoffacceptor kann beispielsweise ein Uberschuss an Harnstoff bzw. substituiertem Harnstoff von mindestens der halben molaren Menge dienen. Zweckmässig verwendet man in diesem Falle etwa zwei Mol oder mehr Harnstoff auf ein Mol Halogenketon und verfolgt den Reaktionsverlauf anhand der Kohlendioxydentwicklung. Andere in Frage kommende Halogenwasserstoffacceptoren sind beispielsweise Magnesiumoxyd, Calciumearbonat, Dimethylanilin und Kaliumcarbonat.
Die Reaktion zwischen Halogencycloalkanon und Harnstoff setzt meist zwischen 50-150 ein und geht unter Wärmeentwicklung vor sich, so dass meist die Verwendung eines organischen Losungs-und Verdünnungsmittels zur Beherrschung des Reaktionsverlaufes notig ist.
Als geeignete Verdünnungsmittel haben sich Diäthylenglykoläther, wie z. B. der Diäthylen- glykoldiäthyläther, erwiesen.
Als Ausgangsstoffe der Formel II kommen zum Beispiel :
2-Chlor-cyclohexanon,
2-Brom-cyclohexanon, 3-Chlor-l-methyl-cyelohexanon- (2), 4-Chlor- : l-methyl-cyclohexanon- (3),
3-Chlor-1-methyl-cyclohexanon- (4),
2-Chlor-und 2-Brom-cye. loheptanon sowie auch anellierte Halogencycloalkanone, wie z. B. das 2-Chlor-und das 2-Brom-tetralon-(I) in Frage.
Als Ausgangsstoffe der Formel III kön- nen neben dem unsubstituierten Harnstoff beispielsweise der N-Methyl-, N-Äthyl-, N-cyclo- hexyl-oder N-Phenyl-harnstoff verwendet werden.
Geeignete Katalysatoren für die Hydrierung der Polymethylenimidazolone der Formel IV sind z. B. Nickelkatalysatoren wie Raney-Nickel oder Nickel auf Kieselgur.
Als Losungsmittel kommen bei der Hydrierung zum Beispiel Alkohole, wie Athanol, Propanol usw. in Betracht. Der Wasserstoff wird mit Vorteil unter Druck bei Temperaturen von 100-200 einwirken gelassen, bis die theoretische Menge aufgenommen ist.
Die Hydrolyse der Cycloalkylenharnstoffe zu den gewünschten Endstoffen kann mittels Mineralsäure, z. B. 60 /oiger Schwefelsäure mit guter Ausbeute durchgeführt werden.
Die erfindungsgemäss erhältlichen 1,2-Di amino-cycloalkane lassen sich im Gegensatz zu den Produkten aus andern Verfahren quantitativ in einheitliche kristallisierte Derivate überführen. Nach ihrem Verhalten gegen über Verbindungen mit zwei reaktionsfähigen Gruppen kann vermutet werden, dass die beiden Aminogruppen zueinander in cis-Stellung stehen, während sie in den Hauptprodukten anderer technischer Verfahren zueinander in trans-Stellung stehen. Das erfindungsgemässe Verfahren ermöglieht somit, Verbindungen in technischem Massstab mit guter Ausbeute her- zustellen, welche bisher nur sehwer zugängliel waren.
In den nachfolgenden Beispielen bedeuten Teile Gewiehtsteile, diese verhalten sich zu Volumteilen wie g zu emS. Die Temperature sind in Celsiusgraden angegeben.
Beispiel 1
Eine Misehung von 66 Teilen Harnstoff und 100 Volumteilen Diäthylenglykol-diäthyS- äther werden zusammen auf 140 erhitzt, bis der Harnstoff geschmolzen ist. Hierauf lässt man unter energischem Rühren innert 20 bis 30 Minuten 66 Teile friseh destilliertes 2-Chlor- eyelohexanon zutropfen, wobei die Temperatur trotz Entfernen der Heizung bis gegen 150 ansteigt und gleichzeitig Kohlendioxyd entweieht. In der Flüssigkeit bemerkt man bald eine fortsehreitende Ausscheidung weisser Kristalle. Naeh beendetem Zutropfen rührt man noch etwa eine halbe Stunde bei 140 , bis die Kohlendioxydabspaltung abgeklungen ist.
Hierauf lässt man abkühlen, rührt das breiige Reaktionsgemisch in 200 Teile Wasser ein und saugt ab. Das Filtergut wird mit Wasser gewaschen und anschliessend getroeknet. Das so erhaltene rohe 4, 5-Cyclo-tetra- methylen-imida. zol-2-on wird in etwa 200 Volumteilen abs. Alkohol bei Siedetemperatur suspendiert, dann abgekühlt und abfiltriert.
Gewünschtenfalls kann dieses vorgereinigte Produkt aus absolutem Äthanol umkristalli- siert werden, worauf es bei 290-3000 unter Zersetzung schmilzt.
Zur Hydrierung suspendiert man 28 Teile 4. 5-Cyclotetramethylenimidazol-2-on in 200 Volumteilen abs. Äthanol, fügt 10 Teile Raney- Nickel hinzu und lässt bei 135 Wasser- stoff unter 120-125 atm. Druck einwirken, bis die Wasserstoffaufnahme der theoretisch erforderlichen Menge entspricht. Hierauf wird vom Katalysator abfiltriert, das Losungsmittel abgedampft und der Rüekstand im Vakuum destilliert. Der N, N'-Cyclohexylemharnstoff geht unter 14 mm Druck bei 218-220 über (Smp. 149-150).
Zur Hydrolyse erhitzt man unter Rühren 14 Teile N, N'-Cyclohexylenharnstoff mit 30 Teilen 60''/odger Schwefelsäure. Bei 95 beginnt langsam Kohlendioxydentwieklung, welche bei 105 lebhaft wird. Diese Temperatur wird etwa 1 Stunde beibehalten, bis die Kohlen dioxydentwicklung aufhört und das Reaktions- gemiseh als klare Lösung vorliegt. Nach dem Abkühlen wird mit konz. Natronlauge unter Aussenkühlung vorsichtig alkalisch gestellt (z. B. mit 80 Teilen 31,5 /oiger Lauge) und die ausgeschiedene Base mit Äther extrahiert.
Der Ätherextrakt wird eingedampft und über 3-4 Teilen Natriumhydroxyd destilliert. Das in einer Ausbeute von 7580 lo erhaltene, steriseh einheitliehe 1,2-Diamino-eyelohexan ist ein farbloses, flüssiges, in Wasser leicht lösliehes Produkt, das unter 1. mm Druck bei 70-73 siedet, und begierig Kohlensäure anzieht.
Beispiel 2
In analoger Weise erhält man unter Verwendung von 73 Teilen 2-Chlor-eycloheptanon (kip14 96-98 ) und 66 Teilen Harnstoff in 110 Teilen Diäthylenglykol-diäthyläther das 4. 5-Cyclopentamethylen-imidazol-2-on (Smp.
310-320 aus Isopropanol unter Zersetzung), welches sich wie oben beschrieben zum N, N'- Cyeloheptylen-harnstoff vom Smp. 253 (aus Methanol) hydrieren lässt.
31 Teile N, N'-Cycloheptylen-harnstoff und 60 Teile 65'Vodge Schwefelsäure werden etwa 15 Stunden bei 135-145 Innentemperatur ge- rührt. Nach dieser Zeit ist die Kohl. endioxyd- entwieklung praktisch beendet. Man kühlt ab, verdünnt mit 40 Teilen Wasser und stellt mit konz. Natronlauge alkalisch, worauf sich das 1,2-Diamino-eyeloheptan als 61 abscheidet. Es wird mit Äther extrahiert, der Extrakt mit Natriumsulfat getroeknet, eingedampft und der Rüekstand über Natriumhydroxyd destilliert.
Das 1,2-Diamino-eyeloheptan siedet unter 23 mm Druck bei 106-107 und wird aus dem N, N'-Cycloheptylen-harnstoff in einer Ausbeute von etwa 80 /o erhalten.

An excess of urea or substituted urea of at least half the molar amount can serve as the hydrogen halide acceptor. In this case, it is expedient to use about two moles or more of urea per mole of haloketone and monitor the course of the reaction on the basis of the evolution of carbon dioxide. Other possible hydrogen halide acceptors are, for example, magnesium oxide, calcium carbonate, dimethylaniline and potassium carbonate.
The reaction between halocycloalkanone and urea usually sets in between 50-150 and develops heat, so that it is usually necessary to use an organic solvent and diluent to control the course of the reaction.
Suitable diluents have been diethylene glycol ethers, such as. B. the diethyleneglykoldiethylether proved.
The starting materials of formula II include, for example:
2-chloro-cyclohexanone,
2-bromo-cyclohexanone, 3-chloro-l-methyl-cyelohexanone- (2), 4-chloro-: l-methyl-cyclohexanone- (3),
3-chloro-1-methyl-cyclohexanone- (4),
2-chloro and 2-bromo-cye. loheptanone and also fused halogenocycloalkanones, such as. B. 2-chloro and 2-bromo-tetralone (I) are possible.
In addition to the unsubstituted urea, N-methyl-, N-ethyl-, N-cyclohexyl- or N-phenylurea, for example, can be used as starting materials of the formula III.
Suitable catalysts for the hydrogenation of the polymethyleneimidazolones of the formula IV are, for. B. nickel catalysts such as Raney nickel or nickel on kieselguhr.
For example, alcohols such as ethanol, propanol, etc. come into consideration as solvents in the hydrogenation. The hydrogen is advantageously allowed to act under pressure at temperatures of 100-200 until the theoretical amount has been absorbed.
The hydrolysis of the cycloalkylene ureas to the desired end products can be carried out by means of mineral acids, e.g. B. 60% sulfuric acid can be carried out with good yield.
In contrast to the products from other processes, the 1,2-diamino-cycloalkanes obtainable according to the invention can be converted quantitatively into uniform crystallized derivatives. Based on their behavior towards compounds with two reactive groups, it can be assumed that the two amino groups are in the cis position to one another, whereas in the main products of other technical processes they are in the trans position to one another. The process according to the invention thus makes it possible to produce compounds on an industrial scale with good yield which were previously only difficult to access.
In the examples below, parts mean parts by weight; these relate to parts by volume as g to emS. The temperatures are given in degrees Celsius.
example 1
A mixture of 66 parts of urea and 100 parts by volume of diethylene glycol diethyl ether are heated together to 140 until the urea has melted. Then, with vigorous stirring, 66 parts of freshly distilled 2-chloroeyelohexanone are added dropwise over the course of 20 to 30 minutes, the temperature rising to about 150 despite the removal of the heater and carbon dioxide escaping at the same time. One soon notices a continuing precipitation of white crystals in the liquid. After the end of the dropwise addition, the mixture is stirred for about half an hour at 140, until the elimination of carbon dioxide has subsided.
It is then allowed to cool, the pulpy reaction mixture is stirred into 200 parts of water and filtered off with suction. The filter material is washed with water and then dried. The crude 4,5-cyclo-tetramethylene-imida obtained in this way. zol-2-one is abs in about 200 parts by volume. Suspended alcohol at boiling temperature, then cooled and filtered off.
If desired, this pre-purified product can be recrystallized from absolute ethanol, whereupon it melts at 290-3000 with decomposition.
For the hydrogenation, 28 parts of 4. 5-cyclotetramethyleneimidazol-2-one are suspended in 200 parts by volume of abs. Ethanol, add 10 parts of Raney nickel and leave at 135 hydrogen below 120-125 atm. Apply pressure until the hydrogen uptake corresponds to the theoretically required amount. The catalyst is then filtered off, the solvent is evaporated off and the residue is distilled in vacuo. The N, N'-cyclohexylemurea passes under 14 mm pressure at 218-220 (m.p. 149-150).
For hydrolysis, 14 parts of N, N'-cyclohexyleneurea are heated with 30 parts of 60 ″ / odger sulfuric acid with stirring. At 95, carbon dioxide slowly begins to develop, which becomes brisk at 105. This temperature is maintained for about 1 hour until the evolution of carbon dioxide ceases and the reaction mixture is a clear solution. After cooling, with conc. Sodium hydroxide solution was carefully made alkaline with external cooling (e.g. with 80 parts of 31.5% alkaline solution) and the base which separated out was extracted with ether.
The ether extract is evaporated and distilled over 3-4 parts of sodium hydroxide. The sterically uniform 1,2-diamino-eyelohexane obtained in a yield of 7580 lo is a colorless, liquid product which is easily soluble in water, which boils at 70-73 under 1 mm of pressure and eagerly attracts carbonic acid.
Example 2
In an analogous manner, using 73 parts of 2-chloro-cycloheptanone (kip14 96-98) and 66 parts of urea in 110 parts of diethylene glycol diethyl ether, the 4th 5-cyclopentamethylene-imidazol-2-one (m.p.
310-320 from isopropanol with decomposition), which can be hydrogenated as described above to give N, N'-cyeloheptylene urea with a melting point of 253 (from methanol).
31 parts of N, N'-cycloheptylene urea and 60 parts of 65'Vodge sulfuric acid are stirred for about 15 hours at an internal temperature of 135-145. After this time the cabbage is. endioxyd development practically ended. It is cooled, diluted with 40 parts of water and made with conc. Caustic soda alkaline, whereupon the 1,2-diamino-eyeloheptane precipitates as 61. It is extracted with ether, the extract is dried with sodium sulphate, evaporated and the residue is distilled over sodium hydroxide.
The 1,2-diamino-eyeloheptane boils at 106-107 under 23 mm pressure and is obtained from the N, N'-cycloheptylene urea in a yield of about 80%.
