B.
Streptokokken, Enterokokken, Pneumokokken, Staphylokokken usw., auszeichnen, so dass sie auch vorteilhaft als Desinfektionsmittel jeder Art zu innern und äussern Zweeken benutzt werden können. Durch ihre gute Vertraglichkeit eignen sie sich u. a. zur Desinfektion der llundhohle.
Gegenstand des vorliegenden Patentes ist nun ein Verfahren zur Herstellung soleher Bisguanylhydrazone, das dadurch-gekenn- zeichnet ist, dass man Dioxoverbindungen der Formel

Wesentlich für die Wirkung ist einerseits das Vorhandensein zweier Phenylkerne, die direkt oder durch eine Br cke miteinander verbunden sind und anderseits die Gegenwart von zwei mit zwei Molekülen Aminoguanidin kondensierten Oxogruppen.
Zur erfindungsgemässen Kondensation kön- nen neben dem Aminoguanidin selbst z. B.

können verwendet werden. Die erwähnten Aminoguanidine sind teilweise in der Literatur bekannt oder können naeh bekannten Methoden leicht hergestellt werden. Erwähnt seien hier zum Beispiel die Reduktion von Nitroguanidinen, die Umsetzung von Isothioharnstoffsalzen mit Hydrazin, die Umsetzung von S-Alkylthiosemicarbazidverbindungen mit Aminen, die Anlagerung von Hydrazin an Cyanamide oder die Entschwefelung von Thioharnstoffen in Gegenwart von Hydrazin.
Die Darstellung der zur Kondensation verwendeten Dialdehyde bzw. Diketone kann auf verschiedene Weise erfolgen, je nach der gewiinschten Art der Brücke. So können z. B.
Diphenyldialdehyd, Diphenylätherdialdehyd, Diphenylthioätherdialdehyd, Diphenylsulfondialdehyd oder Diphenylmethandialdehyd usw. aus den entsprechenden Dimethylverbindungen durch Oxydation bzw. über die co-Tetrabromverbindungen nach der Literatur erhalten werden. Besonders leicht lassen sich Dialdehyde bzw. Diketone mit Methylengrup- pen, die teils durch Heteroatome, wie Sauerstoff, Schwefel oder die Iminogruppe ersetzt sind, herstellen, indem man Oxybenzaldehyde und Oxybenzoketone bzw. die entsprechenden Thio-oder Aminoverbindungen mit Alkylen- dihalogeniden in Gegenwart von Alkali kondensiert.
Die erfindungsgemässe Umsetzung erfolgt am besten in alkoholischer, wässrigalkoholiseher Lösung oder in einem Gemisch von wendet werden, ferner solehe, bei denen die Alkylgruppen durch Heteroatome unterbrochen oder weiter substituiert sind. Auch solche Aminoguanidine, bei welchen zwei Stiekstoffatome der Guanylgrnppe in einen hetero- cyclischen Ring eingebant sind, wie z. B.
Athylen-aminoguanidin, Propylenaminoguani- din,

Wasser und einem wasserlösliehen Lösungs- mittel, wie z. B. Dimethylformamid, Dioxan usw. Besonders gut erfolgt die Umsetzung mit den Salzen, zweckmässig in Gegenwart von etwas übersehüssiger Säure. Jedoeh kann auch in alkalischem Medium kondensiert werden.
Die Salze der neuen Verbindungen mit starken Säuren sind meist in Wasser in der Kälte schwer, in der Hitze leicht loslieh. Aus ihnen können die freien Basen leieht mit Alkalien, besonders auch mit Ammoniak, dargestellt werden. Es sind farblose, schon kristallisierte Verbindungen, die mit organischen Säuren, wie z. B. mit Essigsäure, Milchsäure usw., leicht l¯sliche Salze bilden.
Beispiel 1
14, 2 g p, p'-Dialdehydo-1, 3-diphenoxy-pro- pan (F. 126 , dargestellt aus p-Oxy-benzaldehyd und Trimethylenbromid in Gegenwart von Alkali) werden in 380 cm Alkohol warm gelöst und mit einer Lösung von 11 g Amino guanidinchlorhydrat in 15 ex3 tasser unter Zusatz von 5 cm3 konzentrierter Salzsäure versetzt. Es wird bei 60-80 zwei bis drei Stunden stehengelassen, wobei bald Kristallisa- tion einsetzt. Nach Abkühlen werden die Kristalle des Diehlorhydrates vom F. 2400 isoliert.
Durch Auflösung in heissem Wasser und Fällen mit Ammoniak wird das 4, 4'-Bisguanyl- hydrazon des obigen Aldehyds als Base erhalten. Es wird noeh einmal mit etwas Essig- ester aufgekocht und in gelbliehweissen Kristallen vom F. 226¯ erhalten.

31,2 g p,p' - Dialdehydo - 1,5 - diphenoxypentan (F. ; dargestellt aus p-Oxy-benz- aldelyd und Pentamethylenbromid in Gegen wart von Alkali) werden in 250 em3 Alkohol warm gelost und mit 22 g Aminoguanidinchlorhydrat in 25 cm3 Wasser unter Zusatz von 10 em3 konzentrierter Salzsäure versetzt.
Nach der heftigen Reaktion beginnt nach einigem Stehen bei mässiger Wärme wie in Beispiel 1 bald die Kristallisation des Dichlor- hydrates vom Schmp. 226 . Nach Isolieren der Kristalle kann daraus die Base durch Auflosen in etwa 2, 5 1 warmem Wasser und Fällen mit Ammoniak dargestellt werden. Aus ver dünntem Methanol erhält man das Bisguanyl- ho-dralon des 4, 4'-Dialdehydo-1, 5-diphenoxy- pentans in sehönen weissen Kristallen vom Zer setzungspunkt 240 .
Beispiel 3
14,2 g p, p'-Dialdehydo-1,3-diphenoxy-propan werden in 380 em3 Alkohol warm gelost und mit 19,7 g Isopropylaminoguanidinbrom liydrat (F. 112 ; erhalten aus S-Äthylisopro py]lthioharnstoffbromhvdrat und Hvdrazin) in 20 em3 Wasser unter Zusatz von etwas konzentrierter Salpetersäure versetzt. Naeh längerem Stehen bei gelinder Wärme wird auf 1, 5 1 Wasser unter Zusatz von Ammoniak gegossen. Die ausgefallene Base wird abge sangt, in Methanol gelöst und mit Salzsäure das Diehlorhydrat des Bisguanylisopropyl- hydrazons des obigen Aldehyds ölig gefällt.
Nach Dekantieren und Waschen mit Wasser und Behandeln mit Essigester kristallisiert das Salz.
Beispiel 4
14,2 g p,p'-Dialdehydo-1,3-diphenoxypropan werden in 380 cm3 Alkohol warm gelöst und mit 18, 1 g ¯thylenaminoguanidinbromhydrat (F. 186¯ ; erhalten aus S-¯thylÏthylen thioharnstoffbromhydrat und Hydrazin) in 18 em3 Wasser unter Zusatz von 5 cm3 Bromwasserstoffsäure versetzt. Nach einstündigem Stehen auf dem Wasserbad kristallisiert das Dibromhydrat aus, das naeh dem Isolieren bei 302 schmilzt. Durch Losen in Wasser und Behandeln mit Ammoniak-Wasser bekommt man die Bisäthylenguanylhydrazonbase des obigen Aldehyds, die nach Umkristallisieren aus Äthylalkohol in weissen Kristallen von F. 226 anfällt.
Beispiel 5
15, 6 g p, p'-Dialdehydo-1, 5-diphenoxy- pentan werden in 100 emS Alkohol warm gelöst und mit 18, 1 g Äthylenaminoguanidin- bromhydrat in 18 em3 Wasser unter Zusatz von 5 cm3 Bromwasserstoffsäure versetzt.
Nach zweistündigem Stehen bei geringer WÏrme wird filtriert und zur Kristallisation gestellt. Man erhält das Dibromhydrat des Bisäthylenguanylhydrazons des obigen Aldehyds vom Sehmp. 206 . Die daraus hergestellte Base schmilzt nach Umkristallisieren aus verdünntem Methanol bei 232 .
Beispiel 6 16, 3 g p, p'-Dialdehydo-1, 6-diphenoxyhexan werden in 120 cm3 Alkohol warm gelöst und mit 11, 1 g Aminoguanidinchlorhydrat in 11 em3 Wasser unter Zusatz von 5 em3 konzentrierter Salzsäure versetzt. Bald setzt auf dem Wasserbad Kristallisation ein. Nach zweistündigem Stehen bei gelinder Wärme wird abgekühlt und das Dichlorhydrat vom Sehmp.
230 isoliert. Daraus wird in üblicher Weise die Bisguanylhydrazon-Base des obigen Aldehyds mit dem F. 242¯ erhalten.
Beispiel 7
11, 3 g DiphenylÏther-4, 4'-dialdehyd (dargestellt aus 4, 4'-Dimethyl-diphenylÏther durch Tetrabromierung und Verseifung) in 100 cm3 Alkohol werden mit 11 g Aminoguanidinchlorhydrat in 11 em3 Wasser unter Zusatz von 5 em3 konzentrierter Salzsäure versetzt.
Nach dreistündigem Stehen auf dem Wasserbad wird im Vakuum abgedampft. Der Rückstand wird in viel Wasser gelöst und mit Ammoniak gefällt. Aus Methanol-Wasser umkristallisiert, erhält man die Bisguanylhydrazon-Base des Diphenyläther-4, 4'-dialdehyds in weissen Kristallen vom Z. 257 .
Beispiel 8
21 g Diphenyl-4, 4'-dialdehyd (dargestellt aus 4, 4'-Ditolyl durch Oxydation mit Chromsaure in Eisessig-Essigsäureanhydrid) werden in 400 cm3 Alkohol warm gelöst und mit 28 g Aminoguanidinearbonat in 53 em3 4n-SalzsÏure versetzt. Nach dreistündigem Stehen auf dem Wasserbad und Abkühlen wird das Dichlorhydrat des Bisguanylhydrazons des Diphenyl-4, 4'-dialdehyds abgesaugt und gewaschen.
Beispiel 9
18, 4 g p, p'-Dipropionyl-1, 5-diphenoxy- pentan (F. 110 ; aus p-Oxy-propiophenon und Pentamethylenbromid dargestellt) in 200 em3 Alkohol werden mit 14 g Aminoguanidinear- bonat in 43 em3 2, 44n-SalzsÏure versetzt. Nach kurzem Stehen auf dem Wasserbad wird der Alkohol grösstenteils abgedampft und die zurückbleibende Lösung in etwa einen Liter Wasser unter Zusatz von Ammoniak k gegossen. Die abgesaugte Base wird durch Aus- koehen mit absolutem Alkohol gereinigt und in weissen Kristallen vom F. 220¯ erhalten.
Aus dem Alkohol kristallisiert noch eine weitere Menge derselben Verbindung.
Beisptel 10
25, 8 g o, o'-Dialdehydo-1,5-diphenoxypentan (Fp. 79 ; dargestellt aus o-Oxy-benz- aldehyd und Pentamethylenbromid in Gegenwart von Alkali) werden in 160 cm3 warmem Alkohol gelost md mit 18, 2 g Aminoguanidinchlorhydrat in 18 em3 Wasser unter Zusatz von 9 em3 konzentrierter Salzsäure versetzt.
Nach dreistündigem Stehen bei etwa 80 wird im Vakuum abdestilliert. Der Rüekstand wird in etwa 1 1 heissem Wasser gelöst und mit Ammoniak gefällt. Aus verdünntem Alkohol erhält man das Bisguanylhydrazon des 2, 2' Dialdehydo-1, 5-diphenoxy-pentans in weissen Kristallen vom Schmelzpunkt 209 .
Beispiel 11
23 g o, o'-Dialdehydo-1, 4-diphenoxy-butan (Schmp. 117 ; dargestellt aus o-Oxy-benz- aldehyd und 1, 4-Dibrom-butan in Gegenwart von Alkali) werden in 350 em3 Trockensprit warm gelöst und mit 17 g Aminoguanidinchlorhydrat in 17 em3 Wasser versetzt. Naeh Zugabe von 2 cm3 konz. Salzsäure tritt Reak- tion ein. Nach etwa dreistündigem Erwärmen bei etwa 80 wird die Flüssigkeit im Vakuum abdestilliert. Der Rüekstand wird in heissem Wasser gelost und mit Ammoniak ausgefällt.
Nach Umkristallisieren aus etwa 2 1 Methanol erhält man die weissen Kristalle des Bis guanylhydrazons des 2, 2'-Dialdehydo-l, 4-di- phenoxy-butans vom Zersetzungspunkt 228¯.
Beispiel 12 7 g m, m'-Dialdehydo-1, 5-diphenoxy-pentan (dargestellt aus m-Oy-benzaldehyd und Pentamethylenbromid in Gegenwart von Alkali) werden in 25 em3 Troekensprit gelöst und mit 6 g Aminoguanidinehlorhydrat in 6 cm3 Wasser unter Zusatz von 2 em3 konz.
Salzsäure versetzt. Nach etwa dreistündigem Erwärmen bei 70-80 wird die Flüssigkeit im Vakuum abdestilliert. Der Rüekstand wird in heissem Wasser gelöst und mit Ammoniak ausgefällt. Aus verdünntem Methanol erhält man weisse Kristalle des Bisguanylhydrazons des 3, 3'-Dialdehydo-1, 5-diphenoxy-pentans vom Schmp. 165 .
Beispiel 13 24 g p, p'-Dialdehydo-1,4-diphenoxy-butan (dargestellt aus p-Oxy-benzaldehyd und 1, 1- Dibrom-butan in Gegenwart-von Alkali) werden in 400 em3 warmem Alkohol gelost und mit 18 g Aminoguanidinehlorhydrat in 18 cm3 WasserunterZusatzvon2cmkonz. SalzsÏure versetzt. Es wird 3 Stunden bei 70 bis 80 erwärmt, wobei bald das Dichlorhydrat in sehönen Kristallen in der Hitze heranskommt.
Naeh Abkühlen wird in etwa 1, 5 1 heissem Wasser gelöst und die Base mit Ammoniak @ ausgefällt. Nach Anskochen mit Methanol verbleibt im Rüekstand das Bisguanylhydra- zon des 4, 4'-Dialdehydo-1, 4-diphenoxypentans in weissen Kristallen vom Zersetzungspunkt 255 .
Beispiel 14 14 g p, p'-Dialdehydo-1, 3-diphenoxy-propa- nol- (2) (dargestellt aus p-Oxy-benzaldehyd und 1, 3-Dibrom-propanol in Gegenwart von Alkali) werden in 150 em3 heissem Alkohol gelost. Nach Zugabe von 10, 3 g Aminoguanidinchlorhydrat in 11 em3 Wasser und 5 em3 konzentrierter Salzsäure wird 3 Stunden bei 70 bis 80¯ gehalten, wobei allmählich das Diehlorhydrat vom Schmp. 250 auskristallisiert. Die Base kann durch Auflosen in heissem Wasser und FÏllen durch Ammoniak erhalten werden.
Aus verdünntem Methanol umkristallisiert erhält man so das Bisguanylhydrazon des 4, 4' Dialdehydo-1, 3-diphenoxy-propanols- (2) in weissen Kristallen vom Zersetzungspunkt 215 .
Beispiel 15
24 g Aminoguanidinehlorhydrat, gelost in 24 cm3 Wasser unter Zusatz von 8 cm3 konz.
Salzsäure, werden zu einer alkoholischen Losung des Dialdehyds (dargestellt aus 30, 4 g Vanillin und Pentamethylenbromid in Gegenwart von Alkali) gegeben und 3 Stunden auf 70-80 erwärmt. Naeh Abkühlung wird das s Diehlorhydrat isoliert und in viel heissem Wasser gelöst und mit Ammoniak die Base geJ'äDt. Aus verdünntem Methanol erhält man n das Bisguanylhydrazon des 4, 4'-Dialdehydo- 2,2'-dimethoxy-1,5-diphenoxy-pentas in wei- ssen Kristallen vom Sehmp.160 .
Beispiel 16
15, 7 g p, p'-Dialdehydo-2, 2'-diphenoxy-di äthyläther (dargestellt aus p-Oxy-benzaldehyd und ¯,¯'-Dichlor-ÏthylÏther und Alkali) werden in 900 em3 Alkohol heiss gelöst und mit 11 g Aminoguanidinehlorhydrat in 11 cm3 Wasser unter Zusatz von 8 em3 konz. SalzsÏure versetzt. Es wird auf dem Wasserbad auf etwa 150 em3 eingeengt. Beim Abkühlen erhält man das Dichlorhydrat, das nach Umkristallisieren aus Methylalkohol/Athylalkohol in gelblichen Kristallen vom Schmp. 187 anfällt.
Das Dichlorhydrat gibt nach Auflösen in Wasser und FÏllen mit Ammoniak das Bisguanylhydrazon des p, p'-Dialdehydo-2, 2' diphenoxy-diäthyläthers. Die Base ist sehr schwer löslich und wird durch Auskochen mit Alkohol gereinigt. Wei¯e Kristalle vom Zer setznngspunkt 229 werden erhalten.

31.2 gp, p '- dialdehydo - 1,5 - diphenoxypentane (F.; prepared from p-oxy-benzaldehyde and pentamethylene bromide in the presence of alkali) are dissolved warm in 250 em3 alcohol and mixed with 22 g aminoguanidine chlorohydrate in 25 cm3 of water with the addition of 10 em3 of concentrated hydrochloric acid.
After the vigorous reaction, after standing for a while at moderate heat, as in Example 1, the dichlorohydrate soon begins to crystallize with a melting point of 226. 14.2 gp, p'-dialdehydo-1,3-diphenoxypropane are dissolved in 380 cm3 of warm alcohol and mixed with 18.1 g of ethylene aminoguanidine bromohydrate (F. 186¯; obtained from S-ethylethylene thiourea bromohydrate and hydrazine) in 18 em3 of water with the addition of 5 cm3 hydrobromic acid. After standing on the water bath for one hour, the dibromohydrate crystallizes out and, after isolation, melts at 302. By dissolving it in water and treating with ammonia-water, the bisäthylenenguanylhydrazonebase of the above aldehyde is obtained, which is obtained in white crystals of F. 226 after recrystallization from ethyl alcohol.

15.6 g of p, p'-dialdehydo-1, 5-diphenoxypentane are dissolved in 100 emS of warm alcohol and mixed with 18.1 g of ethylene aminoguanidine bromohydrate in 18 em3 of water with the addition of 5 cm3 of hydrobromic acid.
After standing for two hours at low heat, it is filtered and allowed to crystallize. The dibromohydrate of bisethylenguanylhydrazone of the above aldehyde is obtained from Sehmp. 206. The base produced therefrom melts at 232 after recrystallization from dilute methanol.
EXAMPLE 6 16.3 g of p, p'-dialdehydo-1,6-diphenoxyhexane are dissolved in 120 cm3 of warm alcohol and treated with 11.1 g of aminoguanidine chlorohydrate in 11 em3 of water with the addition of 5 em3 of concentrated hydrochloric acid. Crystallization soon sets in on the water bath. After standing for two hours at gentle heat, the mixture is cooled and the dichlorohydrate is removed from the Sehmp.
