Verfahren zur Herstellung von 11,21-Dioxy-4,17(20)-pregnadien-3-on Vorliegende Erfindung bezieht sieh auf ein neues Verfahren zur Herstellung von 11, 21-Dioxy-4,17(20)-pregnadien-3-on, das da- dureh gekennzeichnet ist, dass man einen 3,11 Diketo - 4,17 (20) - pregnadieri- 21- earbonsäure- ester oder einen 3-Keto-11.-oxy-4,17(20)
-pre- gnadien-21-carbonsäureester mit einem sekun dären Amin zum entsprechenden N-substituier- ten 43,5,17(20)-3-Amino-steroid umsetzt, die ses mit. einem Reduktionsmittel behandelt und das erhaltene N-substituierte 3-Amino-11,21- dioxv-3,.5,17(20)-pregnatrien zum. 11,21-Di- oxy-4,17(20)-pregnadien-3-on hydrolysiert.
Für die Stufe I des erfindungsgemässen Verfahrens verwendet man am besten Alkyl- ester der in Frage stehenden Säuren und als sekundäres Amin ein zyklisches Alkylenimin, wobei man vorzugsweise in Gegenwart eines sauren Katalysators arbeitet.
Die zweite Stufe des Verfahrens, die Reduktion des Enamin- esters, führt man am besten mit Lithium-Alu- miniiunhydrid in einem organischen Lösun gs- mittel mit anschliessender Zersetzung von etwa vorhandenem Organometallkomplex und über schüssigem Lithium-Aluminiumhydrid durch.
Man kann die einzelnen Verfahrensstufen ge trennt durchführen; doch eignet sich das Ver fahren speziell zur Durchführung in einem Arbeitsgang, ohne Isolierung der Zwischen produkte, wie aus den Beispielen ersichtlich wird. Schematisch kann der Reaktionsverlauf im Falleder Verwendung eines Alkyl.enimins wie folgt dargestellt werden:

wobei 1t1 eine a- oder p-usygruppe oder eine Ketogruppe, -COOR.z ein Carbonsäureester- rest, in dem R2 z. B. ein Kohlenwasserstoffrest mit 1-8 Kohlenstoffatomen darstellt, und R3 eine Alkylengiuppe, die mit dem Stickstoff atom z.
B. einen 5-6gliedrigen Ring bildet und vorzugsweise weniger als 9 Kohlenstoff atome enthält., bedeuten. Die 11-Oxygruppe kann die a- oder ss-KonfigLiration aufweisen. Das Radikal R2 ist wie gesagt vorzugsweise ein Alkylrest, insbesondere ein niedriger Alkyl- rest.
Es wurde gefunden, dass Stufe I (Bildung des Enamins) bei geeigneter Wahl der Bedin gungen selektiv verläuft, wobei man die Selek tivität weitgehend beeinflussen kann. Es wurde festgestellt, dass die 21-Ca:
rbonsätire- esterfunktion mit dem Amin nicht merklich reagiert, bevor dasselbe mit der 3-Ketofunk- tion praktisch vollständig reagiert hat. Dies ist im Hinblick auf die Leichtigkeit, mit der Py rroliddn mit Estern unter Bildtang von Pyrrol.ididen reagiert, überraschend (siehe E.
I. du Pont de Nemours & Co., New Pro- ducts, Bulletin No. 28, revidiert 26. Mai 1950). Ausserdem wurde gefunden, dass man den Grad der Umsetzung der Amine mit. der 21- Carbonsäurefunktiondurch das Verhältnis der Reaktionsteilnehmer, Katalysatoren,' Reak tionszeit, Temperatur und Wahl der Amine beeinflussen kann. So kann man ohne weiteres ein Produkt. erhalten, das praktisch frei ist von Amid oder doch nur Spuren davon ent hält.
Unter gewissen Bedingungen würde man praktisch vollständig aus. Amid bestehende Produkte erhalten, was natürlich vermieden werden muss. Die 3-Ketogrnppe ist aber jeden falls stets praktiseh vollkommen in die En- am:ingrippe übergeführt.
Die Enaminfunktion in 3-Stellung des Steroidkernes und die zji7(=o)-21.-Carbonsäure- esterseitenkette der Verbindungen II beein- flussen das gesamte Steroidmolekül in über raschender, tiefgreifender und nicht voraus sehbarer Weise.
Zum Verständnis der Wir kung dieser beiden Substituenten des Moleküls auf die Anordnung des Steroidkernes und die auf diesen ausgeübte Spannung sind folgende Beobachtungen von Interesse. Reduziert man das 3,20-bis-(Äthyleng1ykolketal) des 11-Keto- progesterons mit Li-Al-Hydrid,
so erhält man weniger als 511/0 des l l.a-Oxy isomeren. Redu ziert man unter gleichen Bedingungen das 3-Ätliy lenglykolketal des 3,11-Diketo-4,17 (20)- pregnadien-21-carbonsätiremethylesters, der sieh vom 3,20-Ät.hylenglykoldiketa.l des 11- Keto-progesterons nur in der Seitenkette un terscheidet,
so erhält man etwa 251/o des 11a- Oxyisomeren. Die 917(=e)=?l-Carbonsäiireester- seitenkette übt also auf den Rest. des Steroid- kernes, insbesondere auf die 11-Ket.ogruppe, eine ungewöhnliche Wirkung aus.
Wird Je doch die 3-Ketalgruppe im Ausgangsstoff durch die 3-Pyri,olidylgrttppe ersetzt., so wird' die Wirkung der Seitenkette wiederum be- triiehtlieh verändert. Reduziert man den 3- I'yrroliclyl-11-heto-3,5,1 7 (20)-pre;natrien-21- carbonsäureinethylester 1I, z.
B. mit Li -M- llydrid wieder unter den gleichen Bedingun- gen, so erhält man nur Spuren des 11a-Iso- meren. Der kombinierte und unerwartete Ein fluss der gleichzeitigen Anwesenheit :
sowohl der f17("0)-21-Carbonsäureesterseitenl@ette als auch der 3-Aminogruppe auf die gesamte Steroid- verbindun- äussert sieh deutlich durch die Wirkung auf die Sauerstoffunktion in 1.1-Stel- lung, wie sieh aus den oben beschriebenen anormalen Resultaten bei er Reduktion er geben.
Diese überraschende Eigenschaft ist jedoch sehr vorteilhaft und wird in der zwei ten Stufe des vorliegenden Verfahrens nutz bar gemacht, da zur überführung in aktive Steroidhormone die f-Oxpfoi-m besonders er wünscht ist.
Die zweite Stufe des Verfahrens, die Re duktion des N-substituierten 3-Amino-11-oxy- (oder keto)-3,5,17(20)-pregnatrieü21-carbon- Säureesters 1I erfolgt, wie gesagt.
am besten mit Li-Al-Hydrid in einem organischen Lösungs mittel mit anschliessender lIydroli-se von über- schüssigem Li A1-Ilydrid und organischen 1Ie tallkomplexen. In dieser Stufe wird der 21 Ester zu einem 21 -Alkohol reduziert, ohne da.ss die 3-Enainingiaippe angegriffen wird, wobei das N-substituierte 3-Amiiro-11,21-dioxy-3,5, 17 (20)-pregnatrien III entsteht.
Die Reduk- tion erfolgt auch ohne Angriff auf die 17(20)- Doppelbindung und das konjugierte Dien system des Steroidkernes. Wenn der Ausgangs- stoff in 11-Stellung eine Ketogruppe trägt, so wird diese in die 11ss-Ox-ygruppe umgewan delt, während eine Hydroxylgruppe unver ändert bleibt.
Die dritte Stufe des Verfahrens, nämlich die Hydrolyse des 3-Amino-11,21-dioxy-3,5, 17(20)-pregnatriens, kann praktisch neutral, sauer oder alkalisch durchgeführt werden.
Wenn man für die Stufe II Li-Al-Hydrid als Reduktionsmittel verwendet, so reagiert die durch wässrige Zersetzung von Organo- metallkomplexen und Li-Al-Hydrid erhaltene Mischung ohnehin alkalisch; in diesem Fall kann bereits einverlängerter Kontakt, mit dem 3-Amino-11,21-:
dioxy-3,5,17 (20 )-pregnatrien zu einer Hydrolyse der 3-Aminogruppe unter Bildung von 11,21-Dioxy-4,17(20)-pregnadien- 3-on führen. Die Verbindungen IV oder Ester der selben bilden bei Behandlung mit Osmium tetioxyd 17,20-Osmiatester, die durch Behand lung mit. einem Agens wie Chlorsäure, Per chlorsätire, H,02, Perbenzoe- oder Peressig- säure oder dergleichen in ein 11,17a,21-Trioxy- 4-pregnen-2,20-@dion. oder :dessen Ester über gehen.
Wenn also die Verbindung IV eine 11ss-Oxygruppe aufweist, so ist die bei der Reaktion entstehende Verbindung Kendalls Verbindung F (17-Oxy-eortieosteron) oder ein Ester derselben. Wenn die 11-Oxygruppe die a-Konfiguration besitzt, so kann man die durch die oben beschriebenen Reaktionen er haltene Verbindung nach Blockierung der 21- Oxygruppe durch Veresterung mit Chrom säure oxydieren, wobei ein Ester des Cortisons erhalten wird.
Die Ausgangsstoffe des erfindungsgemä ssen Verfahrens, die Ester der 3-Keto-lla- (oder ss)-oxy-4,17(20)-pregnadien-21-carbon- säure bzw. der 3,11-Diketo-4,17(20)-pregnadien- 21-earbonsäure (I), können hergestellt werden durch Umsetzung eines 17.-Oxy-(oder ket:
o)-21- alkoxyoxalyl-21,21-dihalogeii-progesterons der Formel

in der R eine a- oder fl-OxygiLippe bzw. eine Keta"grruppe und X Chlor oder Brom bedeuten, mit einem Alkalimetallalkoholat in Gegenwart eines Alkohols, z.
B. mit Natriummethylat in Methanol. Diese Dihalogenverbindungen er hält man durch Umsetzung von 11a- oder ss Oxy- oder 1.1-Keto-progesteron mit einem Di- alkylester der Oxa@lsäure und etwa einem Mol- äquivalent einer Alkalimetallbase, z.
B. Na- tritunmethy lat, und anschliessende Halogenie- rqing der erhaltenen Verbindungen mit etwa zwei Maläquivalenten Chlor oder Brom, wie in den folgenden Präparationen näher erläu tert wird.
Für die Stufe I des Verfahrens geeignete Amine sind: Pyrrolidin, Piperidin, C-alkyl- substituierte Pyrrolidine und C-alky1substi- tuierte Piperidine, z.
B. 2,4-Dimethyl-pyrro- lidin, 3-Prop.#-l-piperidin, 2-Methyl-pyrrolidin, 3,4-Dimethyl-py rrolidin, 3-Ätlry 1-py r r olidin, 3- fsopropyl-pyrrolidin, 3,3-Dimethylpyrralidin und andere mit.
niedrigen Alky lgruppen C- substituierte Pyrrolidine und Piperidine. Von diesen Aminen werden Pyrrolidin und Piperi- din bevorzugt. Pyrrolidin scheint im vorlie genden Prozess das reaktionsfähigste Amin zu seit und gibt stets ausgezeichnete Ergeb nisse, so dass es besonders bevorzugt wird.
Das Amin wird in der Regel im überschuss verwendet, wobei man optimale Ausbeuten an Enaminverbindung erhält. Man erzielt auch mit grossen Überschüssen an Amin gute Ausbeuten an 3-Enanün ohne Bildung einer 2l-Amidgruppe;
ein Verhältnis von Amin zum Ausgangssteroid zwischen 1,1-7 Mol Amin pro Mol Steroid, insbesondere zwischen 1,1 und 2, ergibt aber die besten Resultate.
Die Enaminbildung (Stufe I) kann ohne Katalysator dürchgeführt werden; zweekiiiä- ssig verwendet man jedoch einen Katalysator, z. B. einen sauren Katalysator, wie p-Toluol- sulfosäure, Benzolstilfosäure, Stilfoessigsäure, wasserfreien Chlorwasserstoff, konzentrierte Schwefelsäure, andere organische und anorga nische Säuren und dergleichen.
Da diese Kata lysatoren mit dem im Reakt.ionsgemiseh vor handenen Amin in sitze ein Salz bilden, kann man auch Aminsalze wie Anilinsulfat, Pyri- dinchlorhydrat, Pyridin-p-toluolstilfonat und dergleichen als Katalysatoren verwenden. Die Wahl des Kataly Bators scheint. nicht kritisch zu sein, da. die Reaktion auch in Abwesen heit eines solchen vor sieh geht.
Zur Errei chung einer praktisch vollständigen und ra schen Reaktion wird jedoch die Verwendung eine-, Katalysators, insbesondere einer Sulfo- säure, z. B. p-Toluolsirlfosäure, bevorzugt.
Die Enaminbildung wird durch Anwesen heit von Feuchtigkeit im Reaktionsgemisch nachteilig beeinflusst, geht. jedoch auch ohne Entfernung des Wassers bis zu einem gewissen Grad vor sieh. Um optimale Ausbeuten an Enamin zu erzielen, sollte das Reaktions- wasser jedenfalls entfernt. werden.
Dies er folgt vorteilhaft, indem man die Reaktion in einem mit Wasser nicht. mischbaren Lösungs mittel durchführt und das Wasser mit letzte- rem nach Massgabe seiner Bildung abclestil- liert.Man kann das Wasser z. B. auch mittels Trocknungsmitteln entfernen, und diese Me thode ist in gewissen Fällen, z. B. wenn ein kleiner Aminüberschuss verwendet wird, sehr zweckmässig.
Geeignete Trocknungsmittel sind Ka.lziumkarbid, wa:sserfreiesKalziumsulfat und wasserfreies Ka:liumkarbonat. Eine geeignete Methode zur Entfernung des Reaktionswassers besteht. darin, das am R.ückfluss destillierende Lösungsmittel entweder in Dampfform oder als Kondensat oder in beiden Formen durch ein Troekntingsmittel der oben genannten Art zii leiten.
Wenn, auf das Ausgangssteroid be rechnet, etwa ein Moläquivalent Wasser ent- fernt ist, kann die Reaktion als praktisch be endet betrachtet werden.
-Man kann die Reaktion manchmal bei Zim mertemperatur oder darunter durehführen; doeh empfiehlt, es sich, oberhalb Zimmertem peratur, d. h. oberhalb etwa 25 C, beispiels weise zwischen etwa 25 und 150 C, am besten beim Siedepunkt des Reaktionsgemisches, zu arbeiten.
Falls die 11-Sauerstoffunktion eine ss-Oxygruppe ist, arbeitet. man vorzugsweise unter etwa 100 C, da bei höheren Tempera turen die fl-Oxygruppe in Form von Wasser abgespalten werden kann. Die zur Erzielung optimaler Ausbeuten an dem gewünschten Produkt erforderliche Reaktionszeit ist in der Regel umgekehrt proportional zur Reaktions temperatur.
Die Reaktionszeit. kann zwischen wenigen @Iinuten und mehreren Tagen liegen, je nach dem Lösungsmittel., dem Verhältnis der Re agenzien, dem Amin, der Entfernung des Wassers und dem Reaktionsprodukt. Setzt man einen 3-Keto-1.1-oxy-(oder keto)-4,17(20)- pregnadien-21-carbonsäureester in Gegenwart von p-Toluolsulfosäure mit einem grossen l.\berschuss an Pyrrolidin in Benzol am Rück fluss um,
wobei das entstehende Wasser fort laufend entfernt wird, so ist die Bildung des 3-Enamins in weniger als 30 Minuten prak- tiselr beendigt. Verwendet man nur 1-1,1 11-Ioläquiva.lente Pyrrolidin, so benötigt die Re aktion unter sonst. gleichen Bedingungen etwa 11i2 Stunden zur Beendigung.
Als Lösungsmittel verwendet man mit Vor teil die wasserninlöslichen aromatischen Koh- lenwasserstoffe, wie z. B. Benzol, Toluol, Xylo1, Chlorbenzol und dergleichen; doch können aueli andere Lösungsmittel, wie z.
B. Pentan, Ilexan, Chloroform, Methylenchlorid, Tetra ehlorkohlenstoff, sowie mit Wasser mischbare hö.5ungsmittel, wie Methanol, Äthanol, tert. Butanol, Tetrahydrofuran, Dioxan und an dere Verwendung finden. Bei Verwendung von mit Wasser mischbaren Lösungsmitteln erfoll t die Entfernung des Reaktionswassers zweckmässig mittels eines Trocknungsmittels.
Zweckmässig geht man von Verbindungen der Formel<B>1</B> aus, worin R" Methyl oder Äthyl ist. Für die Reduktion der erhaltenen 3 Am-ino-11-oxy-(oder keto)-3,5,17(20)-pregna- trien-21-carbonsäureester mit Li-AI-Hydrid oder äquivalenten Reduktionsmitteln werden als organische Lösungsmittel vorzugsweise Äther, Dioxan, Tetrahydrofuran, Benzol, Hexan und Mischungen davon verwendet.
Die Reaktion erfordert für je 4 llol des Steroids 3 Mol Li-Al-Hydrid; doch verwendet man letzteres gewöhnlich in beträchtlichem Über schuss, um optimale Ausbeuten an dem ge wünschten Produkt zu erhalten.
Wenn das Steroid und das Li-AI-Hydrid, vorzugsweise unter Kühlung, gründlich miteinander ver mischt sind, ist die Reaktion praktisch be- endigt. Manchmal wird zur Sicherstellung des vollständigen Reaktionsverlaufen noch weiter gerührt und/oder erwärmt, doch ist ein Er wärmen in der Regel nicht erforderlich und manchmal sogar schädlich.
Die Zersetzung des überschüssigen Li-Al-Hydrids und etwa vorhandenen Steroid-Metallkomplexes erfolgt vorteilhaft durch vorsichtige Zugabe von Was ser zrun Reaktionsgemisch. Wenn ein mit Wasser nicht mischbares Lösungsmittel ver wendet wurde, kann das erhaltene Steroid isoliert -werden, indem man die organische Phase von der wässrigen Phase trennt.
und aus der abgetrennten Schicht das Lösungs mittel abdestilliert. Falls ein wasserlösliches Lösungsmittel verwendet wurde, setzt man zweckmässig genügend Wasser zu, um das Steroid auszufällen, wonach man dasselbe in üblicher Weise abtrennt. In beiden Fällen kann das Steroid durch Abdestia@lieren des organischen Lösungsmittels aus dem Reak- tionsgemisch gewonnen werden.
Eine bevorzugte Arbeitsweise besteht darin, ,den Ester II mit Li-Al-Hydrid in einem mit Wasser mischbaren, inerten Lösungsmittel, wie Tetrahydrofuran oder Dioxan, bei wesent lich unter Zimmertemperatur, d. h.
unter 20 C, zu behandeln und dann überschüssiges Li-Al-Hydrid und entstandene Orgarnometall- komplexe mit Wasser oder einer organischen Cärbonylverhindung zu zersetzen. Eine tiefe Reaktionstemperatur gewährleistet ein Mini- mttm an Nebenreaktionen,
und die Verwen dung eines mit Wasser mischbaren Lösungs- mittels vermeidet ein Zweiphasensystem. Für die Zersetzung des Reaktionskomplexes wird Wasser vor den Säuren bevorzugt, da die 3-Amino-11,21-d@ioxy- 3,5,17 (20) - pr egnatiiene unter den bei Verwendung von Wasser auf tretenden Bedingungen stabil sind und bei Zersetzung mit Wasser keine so grosse Reak tionswärme entwickelt wird wie bei Verwen dung von Säure.
Niedrige Zersetzungstempe raturen sind deshalb von Vorteil, weil unter Einwirkung von Säure und Wärme das Re- aktionsprodukt- etwas zersetzt wird, wobei nichtkristalline, nicht identifizierbare Pro dukte entstehen.
Zur Hydrolyse der Verbindungen III (Stufe III) kann man beliebige organische Säuren oder Mineralsäuren verwenden; aller dings scheint es, dass die Mineralsäuren keine so einheitliche Reaktion ergeben wie die orga nischen Säuren, beispielsweise gepufferte Essigsäure. Basen sind den Säuren vorzu ziehen, da die Hydrolyse mit diesen in der Regel rascher und sauberer verläuft. Man kann selbst. in neutralem Medium arbeiten, wenn die Reaktionszeit genügend lang oder die Reaktionstemperatur genügend hoch ist.
So kann man das 3-Enamin-steroid etwa 18 Stunden in 95%igem Methanol am Rückfluss kochen. Gibt man eine kleine Menge einer Base, wie Natriumhydroxyd, Kaliumkarbonat, Natriumbikarbonat oder dergleichen, zu, so ist. die Reaktion bei etwa 30 C in weniger als etwa einer Stunde beendigt.
Das entstan dene 11,21-Dioxy-4,17(20)-pregnadien-3-on kann durch Ansäuern der Mischung und Ab trennung der organischen Schicht, falls in einem mit Wasser nicht mischbaren Lösungs mittel gearbeitet wurde, oder durch Extrak tion mit Methylenchlorid, Benzol oder derglei chen, falls das verwendete Lösungsmittel mit Wasser mischbar ist, und Ahdest,illieren des Lösungsmittels isoliert. werden.
Die folgenden Präparationen zeigen, wie die Ausgangsstoffe des erfindungsgemässen Verfahrens hergestellt werden können. Prdparatioiz <I>1</I> 3,11-Diketo-4,17 (20 )-pr egnadien-21-ca.rbon- säuremethylester Na-Enolat <I>des</I> 11-Keto-2I-ä-tjz.oxyoxa.lyl- progesteron.s Zu einer Mischung von 3,4 cm3 einer 3,
4 N methanolisehen Natriummethylatlösung (0,0116 Mol), 0,45 em3 absolutem Äthanol und 20 cm3 wasserfreiem Benzol gibt man, nachdem von ihr 8 en-i,3 abdestilliert wurden, nach dein Ab kühlen 2,3 eins (0,015 11o1) Äthyloxalat und eine Lösung von 3,28 g (0,01 11o1)
11-Keto- progesteron in 38 em3 wasserfreiem Benzol. Die Lösung trübt sieh, und es entsteht, ein gelber Niederschlag. Man rührt die Mischung 90 Minuten, setzt.
55 cm3 Äther zu und rührt, weitere 60 Minuten, wonach man 130 eni3 Äther zusetzt. Der entstandene gelbe Nieder schlag des Natriumenolat.s des 11-Keto-21- äthoxyoxalyl-p.rogesterons wird abfiltriert., mehrmals mit 50 cm3 Portionen Äther gewa schen. Nach dem Trocknen wiegt er 3,65 g.
Der Waschäther enthält. 0,54 g nicht iunge- setztes 1l-Keto-progesteron. Die Ausbeute an Nat,riumenolat ist 81% der Theorie oder, be- rechnet auf das inngesetzte 11-Keto-progeste- ron, praktisch quantitativ.
Das Vorhandensein eines Natriumenolats wird durch die ausser- ordentliehe Wasserlösliehkeit des Produktes und durch die positive Ferriehlorid-probe auf Enole bestätigt, welch letztere in alkoholischen und wässrigen Yerrichloridlösungen eine hell rote Färbung gibt..
11-Keto-2I,?1-dibr'orn-21-IIthoxyoxalyl- progestero-n Zu einer Lösung von 4,5 g (0,01 Mol) des Natriumenolats des 11-Keto-21-äthoxyoxalyl- progesterons und 2 g Kaliumacetat in 70 cm3 Eisessig gibt man unter Rühren 3,09 g (l.,00 cnis, 0,0193 Mal) Brom tropfenweise bei Zimmertemperatur zu.
Nach beendeter Zu gabe wird die Reaktionsmisehnng mit einem grossen Volumen Wasser versetzt, die wässrige Schicht. vom ausgefallenen viskosen gelben Produkt dekantiert, letzteres in Alkohol auf genommen und durch tropfenweise Zugabe

Man erhält 0,99 g 86% der theoretischen Ausbeute an gelben Kristallen vom Schmelzpunkt 168 bis 171 C, Erweiehungspunkt 161 C. Nach zweimaligem Umkristallisieren aus Äthylacetat schmelzen die Kristalle des Methylesters der 3-Py rrolidyl-11-keto-3,5,17(20)-pregnat.rien-21- carbonsäure bei 176-179 C.
Die Analyse ergibt- folgende Werte: Analyse: berechnet für C26H35r'TO3: C 76,2-1; H 8,61; N 3,.12 gefunden: C 76,34; H 8,31; N 3,64. B. Eine Lösung von 3 g 3,11-Diketo-4, 1-7 (20)-pregnadien-21-earbonsäuremethylester, 90 mg p-Toluolsulfosäure und 3 em3 Pyrroli- din in 60 em3 Benzol wird eine Stunde am Rückfluss erhitzt, wobei das gebildete Wasser mit dem siedenden Benzol weggeht.
Man ent fernt Benzol und, Pyrroli,din unter verminder tem Druck, löst den Rückstand in 500 em3 Methylenehlorid und wäscht die Lösung mit Eiswasser, eiskalter verdünnter Säure und schliesslich mit Wasser. Die so erhaltene Lö- sLing enthält. in praktisch quantitativer Aus beute 3-Pyrrolidyl-11-keto-3,5,17(20)-preä na- t-rien-21-earbonsäuremethylester, der durch Abdestillieren des Lösungsmittels isoliert wer den kann.
C. Die folgenden Versuche zeigen die Bil- dungsgesehwindigkeit der 3-Pyr rolidyl-enamin- gtuppe und der 21-Pyrrolididgruppe sowie die günstigste Reaktionszeit für die Bildung des 3-Enamins ohne Amidbildung in 21-Stellung unter speziellen Arbeitsbedingungen. Die Er gebnisse wurden durch Infrarotspekt.ralana- lyse erhalten.
Die Versuche wurden unter Verwendung von 1,2 g Steroid, etwa 50 mg p-Tohio,]snlfo- säure als Katalysator und 40 em3 Benzol bei Rüekflusstemperat-Llr der Mischung durchge führt. sowie andere Alkyl-py rro- lidyl- oder Alkylpiperidylrad.ikale in beispiels- weise die Methyl-, Äthyl-, Propyl-, Isopropyl-, Butyl-, Amyl-, Hexyl-, Phenyläthyl-, Benzyl-, Hexyl-, Heptyl-, Octylester.
<I>Beispiel 3</I> Nach der Methode des Beispiels 1 wird der durch Umsetzung von 11a-Oxy-21,21-di- brom-21-ät.hoxyoxalyl-progesteron mit. Na triumäthylat in Äthanol erhältliche 3-Keto- 1 la-oxy-4,17 (20) -prebma,dien-21- carbonsäure- äthylester, mit- etwa 7 Moläquiva.lenten Pipe- ridin in Gegenwart von etwa 0,
05 14loläqiü- valenten p-Toluolsulfosäure in siedendem Ben zol umgesetzt, wobei man 3-Piperidyl-lla-oxy- 3, 5,1.7 (\Z0) - p,regnatrien - 21- carbonsäureäthyl- ester enthält.
Auf analoge Weise kann man ähnliche Ester dieser Säuren, insbesondere Alkylester und vorzugsweise niedrige Alkylest.er, herstel len, wobei insbesondere der Methyl- und Ä.thylester bevorzugt werden. Ersetzt man das Piperidin durch ein C-alkyisubstituiertes Pipe- ridin, so erhält man das entsprechende C- a:
lkylsubstituierte Piperidyl-enaminsteroid.
<I>Beispiel 4</I> In gleicher Weise wie im Beispiel 1 setzt man den Met.hylester der 3-Keto-llfl-oxy-4, 17(20) pregnadien-21-carbonsäure mit Pyrro- lidin zum 3-Pyrrolidyl-11fl'-oxy-3,5,17(20)-pre- gnatrien-21-carbonsäuremethylester mn.
In analoger Weise lassen sich andere 3- Aminoderivate der Ester dieser Steroid-21- carbonsäure herstellen, wie z. B. 3-Pyrrolidyl- 1.1 Proxy-3,5,17(20)-pregnatrien-21-carbonsäure- äthyjester und 3-Piperid@=1-11ss-oxy-2,4,17(20)- preg-natrien-21-carbonsäuremethylester.
<I>Beispiel 5</I> Das wie in Beispiel 1 beschrieben durch 3stündige Umsetzung von 2 g 3,11-Diket.o-4, 17 (20) - pregnadien -2.1.- carbonsäuremethylester zeit 2 em3 Pyrrolidin in 150 cm3 Benzol er haltene Reaktionsgemisch wird bei 10 C mit 2 g Li-AL-Hydrid in 100 cm3 Äther tropfen weise versetzt und die Mischung eine halbe Stunde bei etwa. 10 C gehalten. Dann gibt.
man tropfenweise 50 cm3 Wasser zu, trennt die organische Schicht ab und entfernt das Lösungsmittel. Der Rückstand wird mit Äthyl- a.eetat verrieben, wobei man 0,88g Kristalle (Ausbeute 41% der Theorie) erhält. Das 3- Pyrrolidyl-11,f,21-dioxy-3,5,17(20)-pregnatrien schmilzt bei 211-219 C.
Die Mutterlauge liefert weitere 0,45 g eines Öls mit gleicher Infrarotabsorptionskurve. Nach dem Umkri- stalbsieren dieser Kristalle steigt der Schmelz punkt auf 228-232 C; (a)D = -88 C in Chloroform.
In gleicher Weise erhält man das Produkt durch Umsetmuig anderer Ester der 3-Pyrro- lidyl-1.1-keto-3, 5,17 (20 )-pregnatrien-21-carbon- säur e oder der freien Säure mit Li-Al- Ily droxyd in geeigneten Lösungsmitteln, wo- bei ein längerer Kontakt mit Wasser oder Basen und insbesondere 31ineralsäuren ver mieden wird, um die besten Ausbeuten an dem gewünschten Produkt zu erhalten.
Andere 3-Amino-llss,21-dioxy-3,5,17(20)- pregnatriene werden hergestellt., indem man den Methyl- oder andere Ester einer 3-Amino- 11-keto-4,17(20) -pregnadien-21-carbonsäure oder die freie Säure, in der die Aminogruppe Pyrrolidy 1, Piperidyl, C-alky>lsnbstituiertes Pyrrolidx-1 ist, mit.
Li-Al-Hydrid wie oben beschrieben umsetzt, wobei die Aminogruppe des erhaltenen Produktes derjenigen des Aus gangsmaterials entspricht.
Beispiel <I>6</I> Ersetzt man im Beispiel 5 den 3-Pyrro- lidyl-11-keto-3,5,17(20) pregnatrien-21-carbon- säüremethylester durch 3-Pyrrolidyl-lla-oxy- 3,5,17 (20 )- pre g natrien-21- earbonsäuremethy l ester oder einen andern Alkylester, so erhält man 3-Pyrrolidyl-1la,21-dioxy-3,5,17(20)-pre- gnatrien.
In gleicher Weise erhält man andere 3- Amino-11 a, 21-dioxy - 3, 5,17 (20) - pregnatriene, indem man von andern 3-Amino-21-carbon- säure.estern ausgeht, in denen die Amino- gruppe Piperidyl oder C-alkylsubstituiertes Piper idyl bzw.
Pyrrolidyl ist und der Alkyl- ester ein Methyl-, Äthyl-, Propyl-, Phenyl-, Benzyl-, Phenyläthyl-, Butyl-, Amyl-, Hexyl-, Heptyl-, Octyl- oder dergleichen Rest auf weist.
Beispiel. t1. Die organische Schicht des Reaktions- produktes von 2,28 g 3-Pyrrolidyl-11-keto-3,5, 17 (20) - preg nat.rien-21.-carb onsäuremethylester in 10 em3 Benzol mit 0,68g Li-Al-Hydrid in 100 cms Äther, das anschliessend mit 50 em3 Wasser hydrolysiert wurde, wird durch Destil lation unter vermindertem Druck vom Lö sungsmittel befreit.
Das verbleibende rohe 3- Pyrrolid\yl-llss,21-dioxy-3,5,17(20)-pregnatrien wird in 1:i0 cm3 'Methanol suspendiert, mit. 25 cms 5o/oiger wässriger Natronlauge ver setzt und 15 Minuten bei 42-46 C gehalten, worauf man die Mischung mit Essigsäure neu- tralisiert. Das Methanol wird abdestilliert und der Rückstand mit. 15 em3 Wasser verrührt.
Die Sispension wird dreimal mit je 50 cm3 Äther extrahiert, die Extrakte vereinigt. und ; eingedampft.
Man erhält in 88 % iger Ausbeute 1,67 g llss,21-Dioxy-4,17(20)-pregnadien-3-on, das nach Umkristallisieren aus Äthylaceta.t bei 151-153 C schmilzt und 1,15 g wiegt.; Ans- ; beute 61%.
B. Eine Suspension von 410 mg 3-Pyrro- lidyl-11A,21-dioxy-3,5,17(20)-pregnatrien in 55 em3 Methanol. wird bis zur vollständigen Lösung mit. 3 em3 5o/oiger wässriger Natron- i lauge auf 35-40 C erwärmt, was weniger als 10 Minuten erfordert. Die Lösung wird abgekühlt, mit Essigsäure neutralisiert und das Lösungsmittel unter vermindertem Druck abdestilliert. Der Rüelustand wird mit.
Wasser vermischt und wiederholt mit Äther extra hiert. Nach Verdampfen des Äthers aus den Extrakten erhält man in 94,4o/oiger Ausbeute 343 mg 11ss,21-Dioxy-4,1.7(20)-pregnadien-3-on vom Schmelzpunkt 150-l53 C, das nach dem, Umkristallisieren aus Äthylacetat bei 155 bis 157,5 C schmilzt und 225 mg wiegt.
<I>Beispiel 8</I> Eine Lösung von 6,0 g 3,11-Diketo-4,17(20)- pregnadien-21-carbonsäuremethylester, 6 em3 Pyrrolidin und 180 mg p-Toluolsulfosäure in 120 cm3 Benzol wird eine Stunde auf Riick- flusstemperatur erhitzt., wobei man gleichzeitig das bei der Reaktion gebildete Nasser ent fernt.
Man destilliert das Lösungsmittel ab, o und es verbleibt. ein R.üel"stand, der im wesentlichen aus 3-Pyrrolidyl-3,5,17(20)-pre# gnatiien-21-carbonsäuremethylester besteht.
Dieser Rückstand wird in 30 cm3 Benzol und 10 cm3 Äther gelöst, mit einer Suspension i von 2,2<B><U>c</U></B> Li-Al-Hvdricl in 350 em3 Äther versetzt, wozu man 5 Minuten benötigt., und dann 90 Minuten bei Zimmertemperatur ge rührt. Zur Zerstörung des überschüssigen Re duktionsmittels versetzt. man mit. 20 em3, Äthylacetat und anschliessend mit 30 cm3 Was ser.
Man destilliert die Lösungsmittel im Vakuum ab und erhält. einen im wesentlichen ans 3-Pyrrolidy111,d,21-,dioxy-3,5,17(20)-pre- nnatrien bestehenden Rückstand.
Dieser rohe Rückstand wird mit 400 cm3 Methanol bei 40 C in Lösung gebracht, mit 70 crn3 wä.ssriger 5%iger Natronlauge versetzt und weigere 10 Minuten auf 40 C gehalten.
Die Lösung wird mit Essigsäure neutralisiert und das Lösungsmittel bei vermindertem Druck und unterhalb 40 C abdestilliert. Der Rückstand wird mit 100 cm3 Wasser, das 10 em3 konz. HCl enthält, gemischt, filtriert und mit Wasser gewaschen.
