Verfahren zur Herstellung organischer Verbindungen Gegenstand der Erfindung ist. ein Verfah ren zur Herstellung einer neuen Säure sowie deren Salzen.
Es ist durch unsere Untersuchungen be kannt, da.ss sieh aus Rauwolfia serpentina Benth. ein Alkaloid mit sedativer Wirkung in reiner Form gewinnen lässt, das Reserpin be- minnt wurde [Experientia, Vol. VIII, Seite 338 (1952 ].
Reserpin zeigt auch eine ausgeprägte blutdrucksenkende Wirkung und besitzt als hypotensives Mittel grosse therapeutische Be- deutun-. Es lässt sieh aus Pflanzenmaterial der Rauwolfiaarten isolieren, wie z. B. nach dem am Schluss des Beispiels 1 angegebenen Verfahren. über die Konstitution des Reser- pirrs ist bis jetzt nichts bekannt geworden.
Es wurde nun gefunden, dass man uner- warteterweisedurch Behandlung von Reserpin mit bestimmten, unten näher bezeichneten Mit teln zu einer neuen Carbonsäure gelangen kann. Sie soll den Namen Reserpsäure tragen.
Reserpsäure besitzt, wie sieh aus unsern Unter- suehungen ergeben hat, neben der freien Ca.rb- oyylgruppe eine freie I-Iydroxy lgruppe und kann durch folgende Formel repräsentiert wer- den:

worin Res den in der Reserpsäure an die freie IIy droxy 1- und Carboxylgruppe gebundenen, zweiwertigen organischen Rest bedeutet.
Reserpsäure ist. durch folgende physikali sche Daten gekennzeichnet: F. 239-245 . Ultraviolett-Spektrum (in Äthanol) : Maxima bei 2 = 224 m,u (a = 31000); 270 m, < c <I>(a =</I> 5040) ; 249 my (a = 6520).
Mi nima bei 7. = 250 m/4 (a = 3440); 280 mit (a = 4210). Infrarot-Spektiuun (in Nujol) Absorptionsbanden bei 3520-3480 (Schul ter), 3240 (breite Bande), 2900, 2850 (breite Bande), 1625, 1605-1585 (breite Bande), <B>1</B>.570, 1505, 1465, 1395, 1378, 1365, 1317, 1280, 1242, 1221 (flach), 1201, 1163, 1140, 1108, 1078, 1028, 973, 950, 905, 804, 750, 720 cm-1. Die Analyse ergibt. folgende Werte: C = 65,66; H = 7,33; N = 6,98%; die Bruttoformel ist. C22H2805N2.
Reserpsäure und deren Salze sind neu und können als Zwischenprodukte zur Her stellung von Heilmitteln mit reserpinähnlieher Wirkung dienen.
Das erfindungsgemässe Verfahren zur Herstellung der Reserpsäure sowie deren Sal zen ist. dadurch gekennzeichnet, dass man Re- serpin oder ein Salz davon mit einem, vorteil- haft stärkeren, alkalischen verseifenden Mittel behandelt.
Dabei werden beide im Reserpin enthal tenen Estergruppen gespalten. Das Verfahren kann man mit den verschiedensten alkalischen verseifenden Mitteln, z. B. der Lösung eines Alkalihydroxyds, wie Kaliumhydroxyd, in einem Alkohol, wie Methanol, oder unter ver schiedenen Bedingungen durchführen, wie in Gegenwart oder Abwesenheit von Wasser, bei tieferen oder höheren Temperaturen oder während kürzerer oder längerer Zeitdauer.
.1e nach der Arbeitsweise erhält man die Reserpsäure in freier Form oder als Salz. Da die Reserpsäure neben der Carboxylgruppe eine Basisehe Gruppe aufweist, kann sie sowohl Salze mit Basen als auch mit. Säuren bilden. So ist es möglich, von Reserpsäure z. B. durch Umsetzung mit Metallhydroxyden, Metallsalze, z. B. Alkalimetallsalze, zu gewinnen.
Ander seits lässt sieh Reserpsäure beispielsweise durch Behandeln mit anorganischen oder organischen Säuren, wie Halogenwasserstoffsäuren, Sehwe- felsäure, Phosphorsäure, Salpetersäure, Oxy- äthansulfonsäure, Toluolsulfonsäure, Essig säure, Weinsäure, Zitronensäure, in ihre Salze mit Säuren überführen. Aus den Salzen kann Reserpsäure in freier Form gewonnen werden. So gewinnt man z.
B. aus Reserpsäurehydro- ehlorid durch Umsetzung mit Silbercarbonat freie Reserpsäure.
Die Ausgangsstoffe können auch in Form der Salze mit den genannten Säuren verwen det werden. An Stelle von Reserpin kann auch Reserpin enthaltendes Material, wie Extrakte aus Pflanzenmaterial von Rauwolfiaarten, wie von Rauwolfia serpentina Benth., als Aus gangsstoff dienen.
In den nachfolgenden Beispielen besteht zwischen Gewichtsteil und Volumteil die glei che Beziehung wie zwischen Gramm und Ku bikzentimeter. Die Temperaturen sind in Cel siusgraden angegeben. Beispiel <I>1</I> 1 Gewichtsteil Reserpin wird mit 40 Vo- lumteilen methanolischer n-Kalilauge in Stiek- stoff-Atmosphäre während 1-11/2 Stunden unter Rückfluss gekocht.
Dann kühlt man die Lösung, stellt sie mit 1 :1-Salzsäure (6-n- Salzsäure) auf ein pH von 1-2 ein und ent fernt das entstandene Kaliumchlorid durch Filtrieren, worauf man das Filtrat beinahe zur Trockne eindampft. Der Rückstand wird zweimal mit. 25 V olumteilen Äther angerührt und dann teilweise in 25 Volumteilen Metha nol gelöst. Man dampft. beinahe zur Trockne ein und zieht. den Rückstand erneut mit 2mal 25 Volumteilen Äthers aus.
Dann löst man in 50 Volumteilen Methanol, stellt den px-Wert durch Zugabe von methanoliseher n-Kalilauge auf ungefähr 6 ein und dampft. beinahe zur Trockne ein. Auf Zu--abe von 50 Volumteilen Chloroform löst sich der grösste Anteil des Rückstandes.
Nach dem Abfiltrieren vorn ent standenen Kaliumehlorid dampft man den Chloroformextrakt ein. 7 Der Rückstand bildet rohe Reserpsäure, welche durch Zusatz von Methanol und Wärmen auf dein fVassei-bacl kristallisiert. Äther wird dann tropfenweise zur Vervollständigung der Kristallisation zu gegeben, worauf die Kristalle abfiltriert und mit Äther gewaschen werden.
Die so in Form ihres Hydroehlorids erhaltene Reserpsäure schmilzt bei 9255--J25811. Sie entspricht der einpirisehen Formel Cz2Ilz805N., - IICI. Das 1lydroehlorid ist sehr gut. löslich in Wasser, beinahe unlöslich in trockenem Chloroform und mässig löslich in einem Gemiseh von Chloroform und Methanol. [a]D = -75 (1% Wasser) und<I>[ ]n</I> = -80 3 (Chloro form). Bei der Analyse werden folgende Pro zentzahlen gefunden: C = 59,59;H = 7,06; N = 6,2; Cl = 8,12 <B>0</B> = 19,03% (als Differenz).
Im Infrarot-Spektrum zeigt die in einem Kohlenwasserstoff (Nujoh aufgeschlämmte feste Verbindung die folgenden eliarakteristi- sehen Absorptionsbanden (in en1-1: 3450 (breite Bande), 3225, 2915 (breite Bande), 2850-2880 (flache Bande), 2585 breite Bande), 1685, 1630, 1605, 1578, 1511. 1-182, 1.165, 1-150, 1.105, 1370, 1350, 1335, 1310, 1290, 1265, 1250, 1230, 1205, 1160, 1145, 1090, 1075, 1055, 1020, 980, 950, 900, 870, 840, 820, 780, 755, 712, 675 und 625.
In Äthanol zeigt Reserpsäurehydroehlorid ein Ultraviolett-Absorptionsspektrum mit den Maxima bei 222 m < i. (E = 33330), 268 inp (L = 5150), 294 ni . @(± = 6776) und den Mi nima bei 248 inP. (F V 2942) und 278 iny = 4146).
Das Reserpsäurehy droclilorid kann auf folgende Weise in die freie Säure übergeführt werden: 0,1 Gewichtsteil Hydrochlorid wird in 10 Voluniteilen Methanol gelöst und während 10 Minuten mit 0,125 Gewichtsteilen gepulv er tem Silbercarbonat -erührt. Man trennt den t\bersebuss an Silberearbonat und das gebil dete Silberchlorid durch Filtrieren ab und dampft das Filtrat zur Trockne ein.
Der er- lialterie schwach gelbe Körper gibt nach aus 1-2 Volumteilen Methanol beinahe farblose Kristalle vom F.239-245 . Die Analyse. ergibt: C = 65,66; 1I = 7,35; N = f>,89; 0 = 20,03% (als Differenz).
Reserpsäure enthält sowohl eine IIy droxyl- als auch eine Carboxy-#-ruppe.
1n Äthanol zeigt die freie Reserpsäure im h'lti-aviolett-Absorptionsspektrum Maxima bei 22-1 in u (F = 31000), 270 mu. (E = 5010), 291<B>11 111,</B> (± = 6520) und Minima bei 250 m,u (s = 3440) und 280 mp (e = 4210).
Die freie, in einem Kohlenwasserstoff (Nujol) auf-esehlämmte Reserpsäure zeigt ferner die charakteristischen Absorptionsban den ini Infrarotteil des Spektrums bei den folgenden Frequenzen, ausgedrückt in rezi proken Zentimetern: 3520-3480 (Schulter), 3240 (breite Bande.), 2900, 2850, (breite Bande), 1625, 1605-1585 (breite Bande), 1570, 1505, 1465, 1395, 1378, 1365, 1317, 1280, 1242, 1221 (flache Bande), 1201, 1163, 1140, 1108, 1078, 1028, 973, 950, 905, 829, 804, 750, 720.
Zur Überführung der Reserpsäure in ihr Ilydroehloi#id kann man folgendermassen ver fahren: 0,1(Tewiehtsteil Reserpsäure wird in 5 Vo= lumteilen Methanol aufgeschlämmt und das pH finit wenigen Tropfen 1 :1 Salzsäure auf den Wert 3-1 gebracht. Man engt dann die entstandene Lösung im Vakuum auf ein klei nes Volumen ein, wobei sieh weisse Kristalle bilden und filtriert nach einigem Stehen das so erhaltene Reserpsäui-eliydi-ochlorid ab. Es schmilzt bei 257-260 .
Aus der Mutterlauge kristallisiert beim Stehen noch mehr Hy dro- ehlorid aus. Die freie Säure kann beispielsweise wie folgt in Metallsalze, z. B. Alkalimetallsalze, übergeführt werden:
Man mischt 0,1 Gewichtsteil Reserpsäure mit 0,25 Volumteilen methanolischer n-Kali- lauge, filtriert dann die erhaltene Lösung, wäscht das Filtrat mit 1 Volumteil Methanol nach und gibt zum Filtrat 25 Volumteile Äther, worauf das Kaliiunreserpat als weisses Pulver ausfällt. Das Salz wird auf einem Fil ter gesammelt und einmal mit 5 Volumtellen Äther gewaschen und dann getrocknet. Es beginnt. bei etwas über 200 zu verkohlen, ist schwarz bei 250 und schmilzt bei 270-300 .
Erdalkalimetallsalze, wie Barium und Cal ciumsalze, stellt man auf ähnliche Weise mit tels den entsprechenden Erdalkalibasen her.
Das im vorliegenden Beispiel als Ausgangs material verwendete Reserpin kann z. B. in der folgenden Weise erhalten werden: 7000 Gewichtsteile gepulverte Wurzelrinde von Rauwolfia Serpentina Benth, werden mit 35 000 Volumteilen Methanol percoliert. Nach dem Eindampfen des methanolischen Extrak tes erhält man 1050 Voliunteile eines stark gefärbten Pulvers.
Dieses wird wiederholt mit Wasser, dann 5mal mit 1500 Volumteilen 10 % iger wässriger Essigsäure behandelt und der ölige, nicht, gelöste Anteil durch Zentrifugieren abgetrennt. Die erhaltene braune essigsaure Lösung wird hierauf entweder bei tiefer Tem peratur eingedickt oder mit. der Hälfte ihres Volumens Wasser verdünnt und besitzt dann einen pH-Wert von ungefähr 3,9. Diese Lö sung extrahiert man in 3-4 Malen mit im ganzen 3400-4000 Volumteilen Chloroform.
Dann wäscht man die Chloroformextrakte einmal mit Kaliumcarbonatlösung, zweimal mit Wasser, trocknet mit Natriumsulfat und dampft im Vakuum zur Trockne. ein. Die 70 bis 80 Gewichtsteile des so erhaltenen grün braun gefärbten Pulvers werden in Benzol ge löst, auf eine Säule von 1000-1200 Gewichts teilen neutralem Aluminiumoxyd (Wirksam keit 2-3 nach Broekmann) gegossen und mit Benzol entwickelt. Zuerst erhält man eine kleine Menge eines gelben Öls, dann 0,9 Ge wichtsteile eines physiologisch unwirksamen kristallinen Materials vom F:238-239 und dann folgen die sedativ wirksamen Kompo nenten.
Sobald der Hauptanteil der aktiven Komponenten eluiert ist, wäscht man die Ko lonne mit einer Mischung von 2 Volumteilen Benzol und 1 Volumteil Aceton, wodurch der Rest des sedativen Wirkstoffes und ein physio logisch unwirksamer Anteil, der bei 141-143 schmilzt, herausgelöst werden. Man dampft die Fraktionen, welche die sedativen Wirk stoffe enthalten, zur Trockne ein, kristalli siert den Rückstand aus heissem Aceton und einer Mischung von Chloroform-Äther um und erhält so 6,5-7 Gewichtsteile Reserpin als beinahe farblose Kristalle.
F. 262-263 (Zer setzung); [a]D =-117 (Chloroform). <I>Beispiel 2</I> Eine Mischung von 34,8 Gewichtsteilen Reserpin, 0,3 Gewichtsteilen Natriumhydro- stulfit und 1,392 Volumteilen methanolischer n-Kalilauge werden unter Stickstoff während 11/2 Stunden am Rückfluss gekocht.
Die schwach bernsteinfarbene Lösung wird mit 255 Volumteilen 1 :1 Salzsäure auf einen pH- Wert von 1-2 gebracht und zur Entfernung des entstandenen Kaliumchlorids filtriert. Das Filtrat dampft man auf dem Wasserbad bei einer Temperatur von 45-50 im Vakuum unter Stickstoffatmosphäre zu einer halb festen Paste ein, extrahiert diese zweimal mit 750 Volumteilen frisch destilliertem Äther, löst dann den Rückstand teilweise in 750 Vo- lumteilen 3lethanol und dampft erneut im Vakuum zu einer Paste ein.
Dann extrahiert man 3mal mit 750 Volumteilen und zweimal mit 150 Volumteilen Äther, rührt den Rück stand mit 750 Volumteilen einer Mischung von 6 Teilen Chloroform und 1 Teil Methanol an und bringt den pH-Wert mittels methanolischer n-Kalilattge auf 5-6. Die entstandene Lösung wird vom Kaliumchlorid befreit und im Va- kuum unter Stiekstoff auf dem Wasserbad bei 50 zur Trockne eingedampft.
Dann rührt man den Rückstand erneut mit 750 Volum- teilen der oben genannten Chloroform-Me- thanolmischung, filtriert von den festen An teilen ab und wäscht zweimal mit 175 Volum- teilen der Bleiehen Lösungsmittelmischung nach. Die zurüekbleibende freie Reserpsäure wird getrocknet. Das Filtrat dampft man zu einem bräunlichen viskosen Rückstand ein, welcher durch Zufügen von 5-10 Volumteilen Methanol und schwachem Erwärmen zur Kri stallisation gebracht wird.
Durch Zugabe von 250 Volumteilen Äther erhält man einen wei teren Anteil Kristalle, welche abgetrennt und gewaschen werden. Man sehlämmt 0,5 Cre- wichtsteile dieser Kristalle in 10 Volumteilen Methanol auf und filtriert die so erhaltene freie, weisse Reserpsäure ab. Beim weiteren Konzentrieren des Filtrates erhält man schwach gelbe Kristalle von Reserpsäure- hydrochlorid.
Quaternäre Ammoniumderivate der Re- serpsäure können durch Behandeln der Re- serpsäure mit reaktionsfähigen Estern von niederen Alkoholen, wie z. B. mit Alky lhalo- geniden, wie Methyljodid, erhalten werden.
Process for the preparation of organic compounds is the subject of the invention. a process for the production of a new acid and its salts.
It is known from our investigations that see Rauwolfia serpentina Benth. an alkaloid with sedative effect can be obtained in pure form, the reserpine was mined [Experientia, Vol. VIII, page 338 (1952]).
Reserpine also has a pronounced antihypertensive effect and is of great therapeutic importance as a hypotensive agent. Reserpic acid and its salts are new and can serve as intermediate products in the manufacture of remedies with reserpine-like effects.
The inventive method for the production of reserp acid and its salts is zen. characterized in that serpin or a salt thereof is treated with an advantageously stronger, alkaline saponifying agent.
Both ester groups contained in reserpine are cleaved. The process can be carried out with a wide variety of alkaline saponifying agents, e.g. B. the solution of an alkali hydroxide, such as potassium hydroxide, in an alcohol such as methanol, or perform under various conditions, such as in the presence or absence of water, at lower or higher temperatures or for shorter or longer periods of time.

Reserpic acid can be obtained in free form from the salts. So you win z.

The starting materials can also be used in the form of the salts with the acids mentioned. Instead of reserpine, material containing reserpine, such as extracts from plant material from Rauwolfia species, such as from Rauwolfia serpentina Benth., Can also serve as starting material.

The residue is used twice. 25 parts by volume of ether are mixed in and then partially dissolved in 25 parts by volume of methanol. The 1lydroehlorid is very good. soluble in water, almost insoluble in dry chloroform and moderately soluble in a mixture of chloroform and methanol. The Reserpsäurehy droclilorid can be converted into the free acid in the following way: 0.1 part by weight of the hydrochloride is dissolved in 10 parts by volume of methanol and stirred for 10 minutes with 0.125 part by weight of powdered silver carbonate. Reserps acid contains both a hydroxyl and a carboxy group.

The following procedure can be used to convert the reserp acid into its Ilydroehloi # id: 0.1 (part of the reserp acid is suspended in 5 parts by volume of methanol and the pH finitely a few drops 1: 1 hydrochloric acid is brought to a value of 3-1 the resulting solution is reduced to a small volume in vacuo, white crystals forming, and after standing for a while the reserve acid eliydi-chloride thus obtained is filtered off, melting at 257-260.
Even more hydrochloride crystallizes out of the mother liquor on standing. The free acid can be converted into metal salts, e.g. B. alkali metal salts, are converted:
0.1 part by weight of reserp acid is mixed with 0.25 part by volume of methanolic n-potassium hydroxide solution, the resulting solution is then filtered, the filtrate is washed with 1 part by volume of methanol and 25 parts by volume of ether are added to the filtrate, whereupon the potassium reserve precipitates as a white powder. The reserpine used in the present example as a starting material can, for. B. obtained in the following manner: 7000 parts by weight of powdered root bark of Rauwolfia Serpentina Benth, are percolated with 35,000 parts by volume of methanol. F. 262-263 (decomposition); [a] D = -117 (chloroform). <I> Example 2 </I> A mixture of 34.8 parts by weight of reserpine, 0.3 part by weight of sodium hydrostulfite and 1.392 parts by volume of methanolic n-potassium hydroxide solution are refluxed under nitrogen for 11/2 hours.
The slightly amber-colored solution is brought to a pH of 1-2 with 255 parts by volume of 1: 1 hydrochloric acid and filtered to remove the potassium chloride formed. The resulting solution is freed from potassium chloride and evaporated to dryness in vacuo under nitrogen on a water bath at 50.
The residue is then stirred again with 750 parts by volume of the above-mentioned chloroform-methanol mixture, the solid parts are filtered off and washed twice with 175 parts by volume of the lead solvent mixture. The remaining free reserp acid is dried. The filtrate is evaporated to a brownish viscous residue which is brought to crystallization by adding 5-10 parts by volume of methanol and gentle heating.
By adding 250 parts by volume of ether, a further proportion of crystals is obtained, which are separated off and washed. 0.5 parts by weight of these crystals are lamed up in 10 parts by volume of methanol and the free, white reserp acid thus obtained is filtered off. If the filtrate is concentrated further, pale yellow crystals of reserpic acid hydrochloride are obtained.
Quaternary ammonium derivatives of reserpic acid can be prepared by treating the reserpic acid with reactive esters of lower alcohols, such as. B. with alkyl halides, such as methyl iodide, can be obtained.
