sedativer Wirkung in reiner Form gewinnen lässt, das Reserpin benannt wurde [Experientia, Vol. VIII, Seite 338 (1952)]. Reserpin zeigt auch eine blutdrueksenkeiide Wirkung und besitzt als hypotensiv es Mittel grosse thera peutische Bedeutung. Es lässt sich aus Pflan zenmaterial der Rauwolfiaarten isolieren, wie z.
B. nach dem am Schlusse des Beispiels 1 angegebenen Verfahren. Über die Konstitution des R.eserpins isst bis jetzt nicht bekanntge worden.
Es wurde gefunden, dass man unerwarte- terweise durch Behandlung von Reserpin mit starken alkalischen verseifenden Mitteln zu einer neuen Carbonsä.ure gelangen kann.
Sie soll den 'amen Reserpsäure tragen. Reserp- säure besitzt, wie sieh aus unseren Z"ntersu- eluingen ergeben bat, neben einer freien Carboxylgruppe eine freie Hvdroxylgruppe und kann durch folgende Formel repräsen tiert werden:

worin Res den im Reserpin an die veresterte Hydroxyl- und Carboxylgruppe gebundene, zweiwertigen organischen Rest bedeutet.
Es wurde nun gefunden, dass man durch Behandlung von Reserpin mit alkalisch ver- seifenden Mitteln auch nur die veresterte Oxygruppe des Reserpins freisetzen und so zu einem Reserpsäureester mit freier Hy- droxylgrüppe gelangen kann.
Reserpsäureester, in denen die Carboxyl- gruppe verestert ist, Salze der genannten Ver bindungen sowie die quaternären Ammonium verbindungen der Reserpsäureester sind neu.
Die erfindungsgemäss erhältlichen Re- serpsäureester können als Zwischenprodukte zur Herstellung von Heilmitteln mit reserpin- ähnlicherWirkung dienen.
Das Verfahren zur Herstellung eines Re serpsäureesters mit. freier Hydroxvlgruppe oder eines Salzes eines solchen Esters ist da durch gekennzeichnet, dass man die veresterte Oxygruppe des Reserpins durch Behandlung von Reserpin oder eines Reserpinsalzes mit einem alkalisch verseifenden Mittel freisetzt..
Dabei kann man mit alkalisch verseifen- den Mitteln in Gegenwart oder Abwesenheit von Wasser, bei tieferen oder höheren Tem peraturen oder während kürzerer oder länge rer Zeitdauer arbeiten. So wird durch kur zes Erhitzen des Reserpins mit der Iösung eines Alkalihydroxydes, wie Kaliumhy- droxyd, in einem Alkohol, wie Methanol, nur die veresterte Oxygruppe gespalten.
Man be handelt Reserpin jedoch vorzugsweise mit einem solchen alkalisch verseifenden Mittel, das aus einer veresterten Hydroxylgruppe die Hydroxylgruppe unter- Bildung eines Esters, das heisst durch Alkoholyse, in Frei heit setzt, wobei je nach den angewandten Bedingungen zusätzlich eine Umsetzung der Carbomethoxygruppe des Reserpins eintritt.
So arbeitet man vorzugsweise in wasserfreien Alkoholen in Gegenwart von Alkoholaten, wie Alkalimetall- oder Aluminiumalkoholaten, oder andern Alkoholyse bewirkenden Mitteln, wie Natriumcarbonat oder Piperidin. In absolutem Methanol in Anwesenheit z.
B. eines Alkalimetallmethylats, wie von Natrium- methyjat oder Aluminium-tert.-butylat, Pi- peridin oder Natriumearbonat, entsteht Re- serpsäuremethylester. Wird die Alkoholyse in andern absoluten Alkoholen, wie Äthanol, Propanol oder Butanol, in Anwesenheit z. B. der entsprechenden All:oholate, wie z.
B. von I'#Tatriumäthylat, -propylat bzw. -butylat oder andern Alkoholyse bewirkenden Mitteln durch geführt, so erhält man dabei durch Ume-sterung die entsprechenden Reserpsäureester, wie den Reserpsäur eäthyl-, -propy 1- bzw. -butylester. .
Je nach der Arbeitsweise erhält man den Reserpsäureester in freier Form oder als Salz. Da Reserpsäureester eine basische Gruppe aufweisen, können sie Salze mit. Säuren bilden.
So lässt sich der erhaltene Reserp- säureester beispielsweise durch Behandeln mit anorganischen oder organischen Säuren, wie Halogenwasserstoffsäuren, Schwefelsäure, Ph(Y.sphorääure, Salpetersäure, Oxyäthansul- fonsäure, Toluolsulfonsäure, Essigsäure, Wein säure, Zitronensäure, in Salze mit Säuren über führen. Alls den Salzen- kann der Reserp- säureester in freier Form gewonnen werden.
Das Reserpin kann auch in Form der ge nannten Salze verwendet werden. An Stelle von Reserpin kann auch Reserpin enthaltendes Material, wie Extrakte aus Pflanzenmaterial von Rauwolfiaarten, wie von Rauwolfia ser- pentina Benth., als Ausgangsstoff dienen.
In den nachfolgenden Beispielen. besteht zwischen Gewichtsteil und Volumteil die gleiche Beziehung wie zwisehen Gramm und Kubikzentimeter. Die Temperaturen sind in Celsiusgraden angegeben.
<I>Beispiel 1</I> Man kocht 1 Gewichtsteil Reserpin wäh rend 2 Stunden mit. 20 Volumteilen abs. Me thanol, 0,5 Gewichtsteilen Kalilauge und einer Spur Natriumbisulfit in Stickstoffatmosphäre. Dabei bildet sich eine gelbbraune Lösung, die mit konzentrierter Salzsäure auf den 1);i- Wert 2 eingestellt wird.
Man filtriert vom aus gefallenen Kaliumchlorid ab, dampft. das Lö sungsmittel unter vermindertem Druck ein, mischt den Rückstand mit. Wasser und trennt nach kurzer Zeit die auskristallisierte Tri- methoxybenzoesäure dureh Filtrieren ab.
Das Filtrat wird mit Natriumearbonat alkalisch gestellt, mit Essigsäureäthylester extrahiert, dann im Vakuum eingeengt. und mit Salz säure stark angesäuert, wobei 0,6 Gewiehts- teile Reserpsäurehy drochlorid auskristalli sieren. Der Essigesterextrakt wird über -Na triumsulfat getrocknet, dann unter vermin dertem Druck eingedampft.
Der spontan kri stallisierte Rückstand besteht. aus 0,38 CTe- wichtsteilen Methy lreserpat, das nach Um- kristallisieren aiis Äther bei 2-10,5-241,5 (Zers.) schmilzt. Es kommt ihm die Brutto formel C23H3005N2 zu und seine Analyse ergibt die folgenden Werte in Prozenten: C = 66,6'8; H = 7,34; N = 7,06; 0 = 18,92 (als Differenz). [a] D = -101 _t- 3 (in Chlo roform).
Der Ester ist unlöslieh in Wasser und lös lich in Methanol, Äthanol und Chloroform. Sein Ultraviolett-Spektrum in Äthanol zeigt Maxima bei 226 mJ,u. (E = 33830), 2-70 my (a = 5090) und 298 mcc (a = 6080) und Mi nima bei 252-254 mu (E = -1110) und 282 mti 4070).
Im Infrarot-Teil des Spektrums zeigt die in einem Kohlenwasserstoff (Nujol) aufgeschlämmte Verbindung charakteristische Absorptionsbanden bei den folgenden in re ziproken Zentimetern angegebenen Frequen zen: 3510, 3365, 2850-2950 (breite Bande), 1724, 1632, 1578, 1500, 1465, 1380, 1362, 1355, 1310, 1332, 1312, 1298, 1268, 1215, 1225, 1202, 1155, 1088, 1068, 1055, 1040, 1030, 1020, 1008, 970, 940 912, 890, 860, 848, 835, 785, 770, 753, 720, 710, 655 und 625.
Das im vorliegenden Beispiel als Aus gangsstoff verwendete Reserpin kann nach folgender Methode isoliert werden: 7000 Gewichtsteile gepulverter Wurzelrinde von Rauwolfia Serpentina Benth. werden mit 35000 Volumteilen Methanol percoliert. Nach dem Eindampfen des methanolischen Extrak tes erhält man 1050 Volumteile eines stark gefärbten Pulvers.
Dieses wird wiederholt mit Wasser, dann fünfmal mit 1500 Volumteilen 10 o/oiger wässriger Essigsäure behandelt und der ölige, nicht gelöste Anteil durch Zentrifu gieren abgetrennt. Die erhaltene braune essig saure Lösung wird hierauf entweder bei tiefer Temperatur eingedickt oder mit der Hälfte ihres Volumens Wasser verdünnt und besitzt dann einen pH-Wert von ungefähr 3,9. Diese Lösung extrahiert man in 3 bis, 4 Malen mit im ganzen 3-100--4000 Volumteilen Chloroform.
Dann wäscht man die Chloroformextrakte ein mal mit Kaliumkarbonatlösung, zweimal mit Wasser, trocknet mit Natriumsulfat und dampft im Vakuum zur Trockne ein. Die 70-80 Gewiehtsteile des so erhaltenen grün braun gefärbten Pulvers werden in Benzol ge löst, auf eine Säule von 1000-1200 Gewichts teilen neutralem Aluminiumoxyd (Wirksam keit. 2-3 nach Brockmann) gegossen und mit Benzol entwickelt.
Zuerst erhält man eine kleine Menge eines gelben Öls, dann 0,9 Ge- wiehtsteile eines physiologisch unwirksamen kristallinen Materials vom F. 238-239 und dann folgen die sedativ wirksamen Komponen ten. Sobald der Hauptanteil der aktiven Kom ponenten eluiert ist, wäscht man die Kolonne mit einer Mischung von 2 Volumteilen Benzol und 1 Volumteil Aceton, wodurch der Rest des sedativen Wirkstoffes und ein physiolo- giseh unwirksamer Anteil, der bei 141-143 schmilzt, herausgelöst werden.
Man dampft die Fraktionen, welche die sedativen Wirk stoffe enthalten, zur Trockne ein, kristallisiert den Rückstand aus heissem Aceton und einer Mischung von Chloroform-Äther um und er- hält so<B>6,5-7</B> Gewichtsteile Reserpin als beinahe farblose Kristalle (F. -262-263 -Zer setzung) ; j ] aD = -117 (Chloroform).
<I>Beispiel 2</I> 0,3 Gewichtsteile Reserpin werden mit 4 Volumteilen einer normalen Lösung Natrium hydroxyd in Methanol überschichtet und dann unter Rüekfluss gekocht. Sobald die ganze Substanz gelöst ist, ungefähr nach 1/,1 Stunde, gibt man 15 Volumteile Wasser und 0,25 Ge- wiehtateile Ammoniumchlorid zu und de stilliert. das Methanol unter vermindertem Druck ab. Aus dem wässrigen Rückstand kri stallisiert Methylreserpat aus.
<I>Beispiel 3</I> Zu 50 Volumteilen wasserfreiem Metha nol gibt man 0,1 Gewichtsteil metallisches INTatrium und fügt nach Beendigung der ein setzenden Reaktion 1,0 Gewichtsteil Reserpin zu. Die Mischung wird während 3 Stunden am Rückfluss gekocht, wobei die festen Anteile in Lösung gehen.
Man dampft dann bei 40-50 im Vakuum auf ungefähr 1'5 Volumteile ein, gibt 50 Volumteile Wasser zu, stellt die Lö sung durch Zufügen von 10d/oiger Schwefel säure auf ein PH von 4,5-5 ein, extrahiert dreimal mit je 50 Volumteilen Äther, macht die wässrige Phase mit konzentriertem Ammo niak alkalisch und nimmt das ausfallende Material in Chloroform auf.
Man wäscht die Chloroformlösung mit Wasser, trocknet sie und dampft sie ein. Das- entstandene Methyl- reserpat schmilzt nach Umkristallisieren aus Essigsäureäthylester bei '23e6-238 .
<I>Beispiel 4</I> Verwendet man bei dem im Beispiel 3 be schriebenen Verfahren anstelle dies trockenen Methanols trockenes Äthanol, so erhält man Äthylreserpat.
<I>Beispiel 5</I> Arbeitet man nach dem Verfahren von Beispiel 3 und braucht trockenes Butanol anstelle des verwendeten Methanols, so erhält man öliges Butylreserpat. <I>Beispiel 6</I> Eine Suspension von 1 Gewichtsteil Re- serpin in 50 Volumteilen abs. Methanol, das 6 Tropfen getrocknetes Piperidin enthält, wird während 10 Stunden unter Rückfluss gekocht,
dann die Lösung gekühlt und filtriert. Das Filtrat wird auf 15 Volumteile eingedickt, mit 50 Volumteilen Wasser verdünnt und durch Zufügen von Schwefelsäure auf einen pH-Wert von 4;5-5 eingestellt. Man extrahiert mit Äther, macht die wässrige Phase alkalisch mit Hilfe von konzentriertem Ammonium hydroxyd und extrahiert mit. Chloroform. Der Chloroformextrakt wird bis zu neu traler Reaktion gewaschen, getrocknet und eingedampft.
Das so erhaltene öl wird durch Zusatz von wenig 1Tet.hanol kristallisiert. Es bildet Methylreserpat. Beispiel <I>7</I> Man kocht eine Suspension von 1 Ge wichtsteil Reserpin und 0,1 Gewichtsteil Alu minium-t-butylat in 50 Volumteilen abs. Me thanol während 12 Stunden unter Rückfluss, kühlt und filtriert.
Das Filtrat wird auf 15 Volumteile eingedickt, mit 50 Volumteilen Wasser verdünnt, mit Schwefelsäure auf ein p$ von 4,5=5 gebracht und mit Äther extra hiert. Man stellt den wässrigen Anteil mit kon zentriertem Ammoniumhydroxyd alkalisch und extrahiert mit Chloroform. Den Extrakt wäscht man, bis er neutral reagiert., trocknet und engt ihn ein. Auf Zugabe von wenig Methanol kristallisiert Methylreserpat aus.
Beispiel <I>8</I> Eine Suspension von 1 Gewichtsteil Re- serpin und 0,1 Gewichtsteil Natriumcarbonat und 50 Volumteilen abs. Methanol werden während 6 Stunden unter Rückfluss gekocht, dann abgekühlt und filtriert..
Man engt. das Filtrat auf 15 Volumteile ein, verdünnt mit 50 Volumteilen Wasser, bringt sein p$ durch Zufügen von Schwefelsäure auf einen Wert von 4,5-5, extrahiert mit. Äther, macht die wässrige Phase mit konzentriertem Ammonium hydroxyd alkalisch und extrahiert erneut mit Chloroform. Der Chloroformextrakt wird bis zu neutraler Reaktion gewaschen, getrocknet und eingedampft. Durch Zufügen von wenig Methanol kristallisiert Methylreserpat aus.

<I> Example 5 </I> If the procedure of Example 3 is followed and dry butanol is required instead of the methanol used, then oily butyl reserpat is obtained. <I> Example 6 </I> A suspension of 1 part by weight of serpin in 50 parts by volume of abs. Methanol containing 6 drops of dried piperidine is refluxed for 10 hours,

The filtrate is thickened to 15 parts by volume, diluted with 50 parts by volume of water, brought to a p $ of 4.5 = 5 with sulfuric acid and extracted with ether.
