Verfahren zur Herstellung von Äthern der (-)-Morphinane Die Erfindung betrifft ein Verfahren zur Herstellung von Äthern des (-)-3-Hydroxy- inorphinans, welche ani Stickstoff einen gesät tigten oder ungesättigten Alk@=l-Substituenten von mindestens 3 Kohlenstoffatomen tragen.
Diese Verbindungen besitzen wertvolle biolo- gisclie Eigenschaften; sie vermögen in klei nen Dosen die unerwünschten Nebenwirkun- gen. z. B. die atemlähmende und diuresehein- wende Wirkung des Morphins und ähnlich wirkender Pharmaka, rasch aufzuheben und sind daher als Arzneimittel von Interesse.
Nach dein erfindungsgemässen Verfahren erhält man die neuen Verbindungen, indem man ein am Stickstoffatom durch einen ge- siitt.igten oder ungesättigten, mindestens 3 Kolilenstoffatome enthaltenden Alkvlrest substituiertes (-)-3-Hydroxy-niorphinan ver- iitliert. Das am Stickstoffatom substituierte (--) - 3 - Hydroxy - morphinan, z. B.
(-) - 3- Ilydroxy-N -allyl-, -N-propargy1-, -N-methall y 1-, -N-crot;@-1-, -N-propyl- oder N-isobutv1-inorphi- nan wird mit einem zur Einführung eines gesättigten oder ungesättigten Alkylrestes am Sauerstoff geeigneten 11lkylierungsmittel, z. B.
PhenjI- trimethyl-, Phenyl - triäthi-1- oder Plienyl-cliätliylallyl.-ammoniumliydroxy d. Pro pyl-elilorid oder Diazomethan, behandelt.
Auf diese Weise erhält man die Methyl-, Äthyl-, Propyl- bzw. Allyl-tither der (-)-3-Hydroxy- N-allyl-, -propargyl-, -methallyl-, -croty 1-, pro pyl- bzw. isobutyl-morphinane. Die Ausgangsmaterialien sind teils bekannt (für (-)-3-Hydroxy-N-all3Tl-morphinan s.
Helv. 304, 2217 [1951]) oder können in analoger Weise wie die bekannte Verbindung aus (-)-3-Hydroxy-morphinan durch Erwärmen mit dem entsprechenden gesättigten oder un gesättigten Alkyl-halogenid dargestellt wer den; die N-Propyl- und N-Isobutyl-Verbin- dungen können auch aus der NT-Allyl- bzw. N-Methallyl-Verbindung durch katalytische Hydrierung gewonnen werden.
<I>Beispiel. 1</I> 25,8 g Phenyl-trimethyl-ammoniumchlorid werden in 35 cm-' absolutem Methanol gelöst und mit 26,6 g 31,5 lhiger methylalkoholischer Kalilauge bei 25 C versetzt, 30 eins absolutes Toluol werden zugegeben und vom abgeschie denen Kaliumchlorid filtriert.
Diese Lösung wird zu einer Lösung von 28,3 g (-)-3- lIydroxy-N-allyl-morphinan in 550 eins Toluol zugegeben und langsam so erwärmt, dass zuerst Methanol, dann ein Gemisch von Methanol Toluol abdestilliert. Wenn die Temperatur auf 110-111 C gestiegen ist und nur noch Toluol destilliert, ist. die Reaktion beendet, Die Reaktionslösung wird nach Abkühlen mit.
Wasser, verdünnter Natronlauge und wieder mit Wasser gewaschen; anschliessend wird das Toluol im Vakuum abdestilliert. Der Rück stand wird bei 11 mm vom Dimethylanilin (Siedepunkt. 70-90 C) befreit und dann im Hochvakuum destilliert.
Die (-)-3-1t!lethox-.#r- -\T-allyl-morphinan-Base siedet bei l60-164 Q 0,01 mm Ilg. Das Hydrobromid, aus Alkohol- Äther kristallisiert, schmilzt bei 224-226 C und zeigt eine Drehung von [a] = - 60,3 D (e = 1,9 in Methanol).
<I>Beispiel<B>29</B></I> 28,1 g (-)-3-Hydroxy-N-propargyl-mor- phinan werden, wie in Beispiel 1 beschrieben, methyliert. Die erhaltene (-)-3-Methoxy-N- pr opar gyl-Base siedet bei 160 C j 0,005 mm Hg [a]D = - 95,27 (c = 2,078 in Methanol).

<I>Beispiel 3</I> 28,5 g (-)-3-Hydroxy-N-propyl-morphinan werden, wie in Beispiel 1 beschrieben, methy- liert. Das erhaltene (-)-3-Methoxy-N-propyl- morphinan siedet bei 168 C/0,01 mm Hg. Des sen Hydrobromid schmilzt bei 278-279 C (aus Methanol-Äther) und zeigt eine Drehung von [a]2 =-35,0 (e = 2,35 in Feinsprit).
