Verfahren zur Herstellung von 17-Oxy-20-keto-steroiden Es ist bereits bekannt, 16-Brom-d4-pregnen- 17,21-diol-3,20-dion-21-acetat durch Anlage rung von. Bromwasserstoff an die entspre chende 16,17-Oxydo-verbindung herzustellen und das Bromhydrin durch Reduktion in d4 Pregnen-17,21-diol-3,20-dion-21-acetat (Acetat der Substanz S) überzuführen. Dieses Verfahren besitzt jedoch verschie dene Nachteile, so (ass es nicht allgemein angewandt werden kann. So lässt sich beispiebs- weige die Reaktion nicht auf solche Steroide anwenden, die Doppelbindungen enthalten, an die sich Halogenwasserstoff anlagern kann. Wie wir festgestellt haben, entsteht zum Bei spiel aus 16,
17-Oxydo-d5-pregnen-3ss-ol-20-on- acetat bei der Einwirkung von Bromwasser stoff in glatter Reaktion eine Dibromverbin> dung vom Schmelzpunkt 158-161 (unter Zersetzung) gemäss folgendem Formelachema:

Auch beim Vorhandensein freier Hydroxyl- gruppen finden bei der Einwirkung von Brom- wasserstoff in Eisessig unter. den a. a.. 0. ge nannten Bedingungen Nebenreaktionen, wie teilweise Acetyliarung;
statt, so dass man Ge mische von Verbindungen erhält, die erst ge trennt werden müssen. Ferner gelingt der Austausch .des Broms in 16-Stellung durch Wasserstoff nur mit mässigen Ausbeuten. Ausserdem werden bei dieser Reaktion grosse Anforderungen an die Art und: Qualität deu Reduktionsmittels ge stellt.
Es wurde nun gefunden, dass man in guten. Ausbeuten zu 17-Oxy-20-keto-steroiden gelangt, wenn man 16,17-Oxydo-20-l#2eto- steroide in Form einer Lösung mit Salzen des Jodwasserstoffes und aus diesen Salzen Jod- wasserstoff freisetzenden organischen Säuren
umsetzt und die erhaltenen Jodhydrine redu@ ziert. Dies ist insofern, überraschend, als es aus der Literatur bekannt ist, da.ss bei der Ein- wirkung von Jodwasserstoff auf a,ss-Oxyd'o- ketone nicht die entsprechenden Jodhydxine erhalten werden,,

Als organische, Jodwasserstoff aus seinen Salzen, wenn. auch nur in geringer Konzen tration oder in Spuren, freisetzende Säuren kommen beispielsweise Carbonsäuren oder Sul- fonsäuren in.
Betracht. Insbesondere eignen sich niedere aliphatische Carbonsäuren, wie Ameisensäure, Essigsäure. Propionsäure u. ä.. Als Sulfonsäure sei beispielsweise genannt p-Toluolsulfosäure.
Alls Salze des Jodwasserstoffes können die- jenigen. eingesetzt werden, die in den verwen- deten organischen Säuren oder den allenfalls zugesetzten Lösungsmitteln genügend löslich sind.
Dies sind in erster Linie die Alkali- und Erdalkalijodide, wie Natnutmjodid, Kalium jodid, Caleiumjodid. Als Lösungsmittel können die verwendeten Säuren selbst dienen.
Man kann ferner Lö- sungsmittel, die imstande sind, die verwende ten Jodide, wenn auch in geringem Masse, zu lösen, wie niedermolekulare Alkohole, Dioxan oder Ketone, wie Aceton, auch im Gemisch mit Wasser, zusetzen.

Das Reaktionsgemisch wird in. 200 cm.3 Wasser gegossen und. der Niederschlag abgesaugt. Der Niederschlagwird in wenig Methylenchlorid aufgenommen und die Lösmig nach dem Trocknen mit Natrium sulfat bis auf 2 -m3 eingeengt. Nach Hinzu- fügen von 10 ,
cm3 Äther kristallisieren 0.97 g 16-Jod-17a-oxy-progesteron vom Schmelzpunkt 1880 C (auf der Koller-Heizbank) aus. Es wird durch. Reduktion mit Raney-Nickel, wie in Beispiel 1 beschrieben, in 17a-Oxy-proge- steron übergeführt.
