Verfahren zur Herstellung von Pregnanderivaten Vorliegende Erfindung bezieht sich auf ein Verfahren zur Herstellunug der neuen 3-Oxy- oder Acyloxy-11.',177a-dioxy-pregnan- 20@-one.
Die nach dem Verfahren erhältlichen neuen Verbindungen lassen sich durch fol gende .Strukturformel darstellen:

in der R. Wasserstoff oder den Acylrest einer organisehen Monoearbonsäure, insbesondere einer solchen mit einem bis einschliesslich acht Kohlenstoffatomen bedeutet.
Solehe Säuren sind Ameisen-, Essig-, Pro- pion-, Butter-, Valerian-, Capron-,@ Heptyl-, Caprylsäure, Cyclopentancarbonsäure, Cyclo- hexancarbonsäure, Cyclopentylpropion-, Ben- zoe-, Toluy lsäure und dergleiehen.
Die neuen Verbindungen sind wertvolle Produkte für die Synthese physiologisch ak- tiver :Steroide, wie Cortison und Kendalls Ver bindung F, sowie für die Synthese von 11ss- Oxy-steroiden. Ausserdem besitzen sie an sich schon eine gewisse physiologische Wirkung.
So kann man zum Beispiel das Acetat von Kendalls Verbindung F aus dem neuen 3,11ss,17a-Trioxy-pregnan-20'-on herstellen, in dem man es mit Brom in Chloroform bromiert und in das so erhaltene 2'1-Brom-derivat mit Kaliumacetat in Essigsäure die 21-Aeetoxy-, gruppe einführt.
Das so erhaltene 3.,11ss,l#7a- Trioxy-21-acetoxy-pregnan-2,0-on wird nach dem Verfahren von Oppenaur in 3@Stellung selektiv oxydiert und die erhaltene 3--Keto- verbindung mit Brom in Essigsäure in das. 3=Keto-4-bromderivat übergeführt, das durch Behandlung mit.
Semicarbazidchlorhydrat un ter Abspaltung von Bromwasserstoff und Aus bildung einer Doppelbindung zwischen den Kohlenstoffatomen 4 und bin das 3=Semi- carbazon übergeht. Durch Entfernung der Semicarbazongruppe mit Brenztraubensäure erhält man das Acetat von Kendalls Verbin- d-ung F.
Die 3-Acyloxy-11ss,17.a-dioxy-pregnan- 2ss-one können in gleicher Weise in das Acetat ; von Kendalls Verbindung F übergeführt wer den, indem man- zuerst die 3-Acyloxygruppe mit verdünnter Natronlauge ztu '3-Oxygruppe verseift.
Die im erfindungsgemässen Verfahren als i Ausgangsstoffe verwendeten- neuen 3-Oxy- oder Acyloxy-17a-oxy-pregnan-11,20-dion-20- ketale entsprechen folgender Formel:

3a,17a- Dioxy-pregnan-11,20Ldion und dessen. 3-Ester können erhalten werden nach Kritchevsky, Garmaise und Gallagher, J.
Am. Chem. Soc., 74, 483 (19522). ,3.,1'71a#-Dlioxy-pregnan-11,20- dion und dessen 3-Ester werden aus dem be- kannten 3ss-Oxy-pregnan-11,20-dion [von Euw, Lardon und Reichstein, Helv. Chim. Acta, 27, & 21 (19@)], wie*in der Präparation 1, A bis D,
beschrieben, erhalten.
Nach dem Verfahren der vorliegenden Er findung stellt mann 3-Oxy- oder Acyloxy- 11fl,17,a - dioxy - pregnan-20-one dadurch her, dass mann auf ein 17a-Oxy-pregnan-11,20-dion- 20-ketal, das in 3-Stellung eine Oxy- oder Acyl- oxygruppe aufweist, deren Acylradikal von einer organischen Monocarbonsäure abgeleitet ist, ein Reduktionsmittel,
wie zum Beispiel ein gemischtes Metallhydrid oder Wasserstoff in Gegenwart eines Katalysators einwirken lässt, das entstandene Produkt hydrolisiert und das entstandene 3@-Oxy- oder Acyloxy-11f 17a- dioxy-pregnan-20-on isoliert.
Zweckmässig verfährt man wie folgt: das 3-Oxy- oder Aeyloxy-17a oxy-pregnan-11,20- dion-20@ketal wird mit. einem Lösungsmittel vermischt., das unter den bei der Reaktion herrschenden Bedingungen nicht reagiert. Die Mischung wir- dann inilt einem gemischten Metallhydrid, wie zum Beispiel Lithium-Alu- miniumhydrid, Natrium-Borhydrid,
Lithiuun- Bor'hydrid und dergleichen, reduziert. Man kann auch andere chemische Reduktionsmittel oder Wasserstoff in Gegenwart von Katalysa toren, wie Platin, Palladium-Kohle, Ranev Nickel und dergleichen, verwenden. Die Art des verwendeten Lösungsmittels hängt teil weise von der Löslichkeit des Ausgangs- steroids, der Reaktionstemperatur und dem verwendeten Reduktionsmittel ab.
Bei Ver wendung von gemischten Metallhydriden eig nen sich Lösungsmittel, -wie Äther, Tetrahydro- furan, Benzol, Alkane und dergleichen, sowie manchmal Wasser. Bei Anwendung anderer chemischer Methoden oder katalytischer Me thoden eignen sich Lösungsmittel, wie Äther, Dioxan, Alkohol und dergleichen, sowie manchmal Wasser.
Gemäss einer bevorzugten Ausführungsform des Verfahrens wird das Ausgangssteroid in einem inerten Lösungs mittel, wie zum Beispiel Benzol, gelöst und mit einem gemischten Metallhydrid, vorzugs weise Lithiiim-Aluminiumhydrid, das vorher mit einem Lösungsmittel, wie zum Beispiel Äther, vermischt wurde, zusammengebracht.
Während der Reduktion hält man die Tem peratur in der Regel zwischen etwa 0 und 100 C, wobei man Temperaturen zwischen etwa Zimmertemperatur und der Siedetem peratur der Mischung bevorzugt.. Die Reak tionszeit kann 30 Minuten bis etwa 8 'Stun den und mehr betragen.
Bei Verwendung eines gemischten Metallhydrids, wie Lithium-Alu- miniumhydrid, führt man die Reduktion zweckmässig zunächst bei Temperaturen zwi schen etwa 0 und 50 C, vorzugsweise bei Zimmertemperatur;
durch, während man ge gen das Ende der Reaktion höhere Tempera- tiiren anwendet, wobei die Siedetemperatur der Mischung in der Regel die obere Grenze darstellt. Vorzugsweise mischt und rührt man die Reaktionsteilnehmer etwa eine Stunde bei Zimmertemperatur, wonach man auf die Rückflusstemperatur der Mischung aufheizt. Die Gesamtdauer der Reaktion hängt zum Teil vom Verhältnis der Reaktionsteilnehmer, zum Teil von der angewendeten Temperatur ab.
In der Regel verwendet man das gemischte Metallhydrid in einem wesentlichen über- schuss, der 1- bis l0Ofacli oder grösser sein kann, wobei ein 5- bis 5,Ofacher Überschuss be vorzugt wird. Arbeitet man mit Wasserstoff und Katalysatoren als Reduktionsmittel, so schüttelt man die Mischung der Ausgangsver bindung, Lösungsmittel und Katalysator mit Wasserstoff, bis die theoretisch erforderliche Menge Wasserstoff absorbiert ist.
In den meisten Fällen verläuft die Reduktion bei Zimmertemperatur und einem Wasserstoff druck von 1 bis 3 Atmösphären vollkommen befriedigend, doch lassen sich auch höhere Temperaturen und Drucke anwenden. Bei Verwendung von Lithium-Alluniniumhydrid als Reduktionsmittel findet ausser der RedLik- tion der 11-Ketogruppe noch die Umwand lung der Estergruppe in 3-Stellung in die IIyd'roxylgruppe statt.
Falls die Estergruppe in 3-,Stellung erhalten bleiben soll, muss man andere Reduktionsmittel, wie zum Beispiel Na trium - Borhydrid, Lithiiun - Borhydrid oder Wasserstoff mit Katalysatoren verwenden. Es ist auch möglich, das Produkt nach der Reduktion wieder zu acylieren.
Wenn die Reduktion praktisch beendet ist, wird etwa vorhandenes überschüssiges Reduk tionsmittel zerstört. Falls ein gemischtes Me- tallhydrid oder ein anderes chemisches Reduk tionsmittel verwendet wurde, zum Beispiel Lithiiun-Aluminiiunhydrid', wird letzteres zer setzt, indem man in der Regel tropfenweise -unter Rühren, ein Reagens, wie Wasser, was serhaltigen Äther, Alkohol, Alkohol - Äther mischungen, wässerigen Alkohol,
verdünnte wässerige Salzsäure und .dergleichen, zusetzt, wobei das Reaktionsprodakt in der organi schen Phase gelöst bleibt. Wenn katalytisch reduziert wurde, so wird der Katalysator durch Filtration entfernt. Gewünschtenfalls kann man das durch Reduktion erhaltene 3- Oxy- oder Acyloxy-11.,
1.7a-dioxy-pregnan-920- ketal nach irgendeiner bekannten -Methode aus der organischen Lösung isolieren, indem man beispielsweise das Lösungsmittel durch Destillation entfernt. Oft ist es vorzuziehen, das Reduktionsprodukt nicht zu isolieren, da die aus dem Reduktionsprozess anfallende Lö sung genügend rein ist zur Verwendung in der zweiten Stufe des erfindungsgemässen Ver fahrens.
Eine Lösung des 3-Oxy oder Acyloxy- 11ss,17a-dioxy - pregnan - 20 - an - 20 - ketals in einem organischen Lösungsmittel, das dasselbe sein kann, wie das aus der vorherigen Reduk tionsstufe aus der der Katalysator oder das überschüssige Reduktionsmittel entfernt wurde, wird mit einem hydrolysierenden Mit tel vermischt. In der Regel verwendet man einen überschuss einer wässerigen Mineral- säurelösiung, wie Schwefelsäure, Salzsäure und dergleichen.
Man kann in gewissen Fällen auch Essigsäure, andere organische Säuren oder andere sauer reagierende .Stoffe verwen den. Der Überschuss an hydrolysierendem Mit tel kann eins bis hundertfach sein, wobei ein fünf- bis zehnfacher Überschuss bevorzugt wird. Man rührt das Reaktionsgemisch in der Regel etwa 24.Stunden bei Zimmertemperatur. Die Reaktionszeit kann länger sein oder auch nur eine Stunde betragen. Sie ist abhängig vom Ketal, der Reaktionstemperatur und der Konzentration des hydrolysierenden Mittels.
Höhere Konzentrationen und Arbeitstempera turen setzen in der Regel die Reaktionszeit herab. Man kann bei T emperaturen zwischen etwa 0 C und dem Siedepunkt der Mischung arbeiten. Wenn die Mischung aus mehr als einer Phase besteht, empfiehlt es sich, zu rühren.
Nach Beendigung der Hydrolyse wird das Produkt nach irgendeiner der üblichen Me thoden isoliert. Wenn die Mischung zum Bei spiel aus einer organischen Schicht und einer wässerigen ;Schicht besteht, trennt man die organische Schicht ab, extrahiert die wässe rige Schicht mit einem organischen Lösungs- mittel und vereinigt die Extrakte mit der organischen ischieht. Die organische Phase wird dann mit Wasser gewaschen und über einem Trocknungsmittel,
wie wasserfreiem Natriumsulfat getrocknet. Das Trocknungs- mittel wird durch Filtration entfernt -Lind das Lösungsmittel verdampft. Der Rückstand kann dann durch Umkristallisieren aus einem ge eigneten Lösungsmittel gereinigt werden.
Die Ausgangsstoffe können folgender massen hergestellt werden: Präparation <I>1:</I> A. .3ss-Oxy-pregnan -11,20- dion (8,18 g) werden in einer Mischung von 50 cm3 Essig säureanhydrid und 2 Gm3 Pyridin gelöst und 24 ,Stunden bei Zimmertemperatur stehen ge lassen..
Dann giesst man in 325 eins Wasser und lässt mehrere Stunden bei Zimmertem peratur stehen, wobei sieh das überschüssige Essigsäureanhydrid zersetzt. Das feste Pro- dukt wird abfiltriert und im Vakuum ge trocknet.
Nach zweimaligem Umkristallisieren aus wässerigem Aceton erhält man<B>2,76</B> g Produkt vom 'Schmelzpnkt <B>155</B> bis 1162 . Wie derholtes Umkristal'lisieren aus wässerigem Aceton gibt das 3,f-Acetoxy-pregnan-11,20- lion, (a)D -I- 9,9 (-Chloroform)-.

Eine Mischung von <B>2,75</B> 5 g des so her gestellten 3fl - Acetoxy - pregnan-11,20 - dions, 0,73 g p-Toluol-s-Lilfosäuremonohydrat und 90 cm3 Essigsäureanhydiid= wird zum Sieden erhitzt und drei Stunden langsam destilliert, wobei 71 eins übergehen.
Man destilliert un ter vermindertem Di#Lick weitere 15 cm3 ab, kühlt und verdünnt mit 85 cm3 Äther, wäscht mit 1prozentiger wässeriger Biearbonat- lösung und Wasser und trocknet über wasser freiem Natriumsulfat. Nach Entfernen des Natriumsulfats und Abdestillieren des Äthers verbleiben 3,5 g 3,,11,.20 - Triacetoxy - 9 (11)- 17'(20)-pregnadien als glasiger Rückstand. C.
Die 3y5 g des 3f,11,20,:Triacetoxy-9(11)- 1'7(20)=pregnadiens aus B werden in 17 cms Chloroform gelöst und die Lösung auf 0 bis 5 C abgekühlt, dann gibt man unter Rühren eine Mischung von 170 mg wasserfreiem Na- triumacetat und 7,7 em3 38prozentiger Per essigsäure zu, wobei man die Temperatur auf 0 bis 5 C hält.
Man rührt 10 Minuten bei dieser Temperatur, lässt auf Zimmertempera tur erwärmen und rührt dann weitere 90 Minuten. Dann verdünnt man mit 80 cm3 Äther, wäscht viermal mit je 15 cm3 5pro zentiger Natronlauge und dreimal mit je 15 e m3 Wasser und trocknet über wasser freiem Natriumsulfat. Nach Entfernung des Trocknungsmittels und Abdestillieren der Lö sungsmittel unter vermindertem Druck er hält man 3;
5 g 1.7 (20,)-Oxydo-3@fl,11,20-tri- acetoxy-9@(11)-pregnen als viskoses Öl. D. Das 17(20)-OxYdo-3y3,11,20-triacetoxy- 9(11)-pregnen aus Ü (3;5 g) wird in 66 em3 Alkohol gelöst, 66 eins normale wässerige Salzsäure zugegeben und die Lösung 30 Stun den bei Zimmertemperatur unter Stickstoff atmosphäre gerührt. Dann giesst man in 250 cm3 Wasser und extrahiert die Suspension viermal mit; 50 em3 Chloroform.
Nach Ab destillieren des Lösungsmittels erhält man 2,74 g rohes 3ss,17a-dioxy-priegnan-11,20-dion. Durch Acetylierung mit Essigsäureanhydrid und Pyridin bei Zimmertemperatur erhält man das 3-Acetat. E.
In gleicher Weise wie in folgender Präparation 2 stellt man aus dem gemäss D erhaltenen 3,,17a-Dioxy-pregnan-11,20-dion durch Umsetzung mit Äthylenglykol in Ge genwart von p=Toluolsulfosäure das 3ss',17a- Dioxy -pregnan-11,20-dion -'210 - äthylenglykol- ketal her..

<I>Präparation <SEP> 2:</I> Eine Mischung von 2i60 mg 3,a,1'7a-Dioxy- pregnan -11,20 - dion, 5 cm3 Äthylenglykol, 50 mg p-Toluol - sulfosäuremonohydrat und 100 em3 Benzol werden in einen mit Rück flusskühler versehenen Kolben gebracht. Der Kühler weist einen Wasserfänger auf, durch den die rückfliessenden kondensierten Dämpfe hindurchgehen müssen. Man erhitzt 5 Stunden am Rückfluss und rührt gleichzeitig.
Das ge bildete Wasser destilliert kontinuierlich mit dem Benzol ab und wird im Wasserfänger gesammelt. Dann kühlt man die Mischung und giesst sie in verdünnte Natriumbicarbonat- lösung. Die Benzolsahicht wird abgetrennt, mit Wasser gewaschen und zur 'Trockne ver dampft..
Der Rückstand wird über 40' g Flori- sil (synthetisches Magnesiumsilikat) chromato- graphiert, wobei man zur Eluierung je 80 cm3 Portionen Äthylendichlorid mit ziulehmendem Gehalt an Aceton verwendet. Das mit Äthylen- dichlorid-Aceton (12.:1 und 8:1) eluierte Produkt wiegt nach Entfernung der Lösungs mittel 141 mg.
Nach Umkristallisieren aus Benzol-eSkelly 'Solve B erhält man 50 mg 3a-17;ai-Dioxy - pregnan -11,20-dion-20@-äthylen- Lrlykol-ketal vom Schmelzpunkt 144 bis 146 C.

Die <SEP> folgenden <SEP> Beispiele <SEP> erläutern <SEP> das <SEP> er findungsgemässe <SEP> Verfahren <I>Beispiel 1</I> Zu einer Lösung von 2 g Lithiiim-Aluuni- niumhydrid in 2.00 cms wasserfreiem Äther gibt man tropfenweise unter Rühren 2 g 3a,17a-Dioxy-pregnan-11,20-dion-20-äthylen- glykolketal in 20 cm3 wasserfreiem Benzol ge löst zu.
Man rührt eine Stunde bei Zimmer- temperatuir und kocht eine weitere,Stunde am Rückfluss. Man kühlt unter fortgesetztem Rüh ren ab und setzt vorsichtig tropfenweise Was ser zu.

Beim Stehen scheiden sich Kristalle aus, die durch Filtration gewonnen und als das 11aIsomere (3a,11a,17a - Trioxy - pregnan-20-on) identifi ziert werden.
Bei weiterem Verdünnen des Filtrats mit Skelly .Solve B erhält Juan 5.9'0i mg .3a,11ss,17.a=Trioxy-pregnan-2@0Lon. Eine weitere Menge dergleichen-Verbindung erhält man aus der Mutterlauge durch Entfernung des Lösungsmittels unter vermindertem Druck, Auflösen des Rückstandes in möglichst wenig Äthylacetat und Verdünnen mit Skelly Solve B .
Die 590 mg des Produktes werden einmal ausÄthylacetat- Skelly Solv e B -Mischung und einmal aus Aceton- Skelly Solve B -IVlisehung umkristallisiert, und man erhält 2,75 mg ge reinigtes 3a,11fl,17a-Trioxy-pregnan-20,-on als Platten vom Schmelzpunkt 2'13 bis 2.16 C, (a:)," + 73 C (Aceton).
