Verfahren zur Herstellung von in 3-Stellung cyclisch ketalisierten Steroidenx Vorliegende Erfindung bezieht sich auf ein Ver fahren zur Herstellung von in 3-Stellung cyclisch ketalisiertem 11,21-Dioxy-4,17(20)-pregnadien-3-on, welches dadurch gekennzeichnet ist, dass man 3,11- Diketo-4,17(20)-pregnadien-21-säure oder Alkyl- ester derselben Formel:

Die neuen Ketale wie auch die freien 3-Keto- verbindungen können zur Herstellung von Cortical- hormonen verwendet werden. Die Reaktionen kön nen durch die nachfolgende Formelreihe wiedergege ben werden:

Das 11,21-Dioxy-4,17(20)-pregnadien-3-on (IV) kann nach den im Schweiz. Patent Nr. 335663 er läuterten Methoden in Cortison oder 17-Oxy-cortico- steron oder deren Ester übergeführt werden.
Setzt man 11,21-Dioxy-4,17(20)-pregnadien-3-on (IV) oder einen 21-Ester davon, in welchem die 11-Oxygruppe die ss-Stereokonfiguration hat, mit Osmium - tetroxyd um, so erhält man den 11 ss,17a,20,21 - Tetraoxy -4- pregnen - 3 -on-17,20- osmiatester oder einen 21-Ester davon;
durch an schliessende Oxydation mit Perchlorsäure oder deren Salzen oder mit einem andern gleichwertigen Oxyda tionsmittel, wie z. B. Wasserstoffperoxyd, Dialkyl- peroxyden, organischen Persäuren, wie Peressigsäure oder Perbenzoesäure, Kaliumchlorat und dergleichen, in einem Lösungsmittel wie Äther oder Alkohol, z.
B. tert. Butylalkohol oder Diäthyläther, erhält man nach bekanntem Verfahren [vgl. Prins und Reichstein, Helv. Chim. Acta, 25, 300 (1942); Ruzicka und Müller, Helv. Chim. Acta, 22, 755 (1939)] das 11ss,17a,21-Trioxy-4-pregnen-3,20-dion (Kendalls Verbindung F).
In analoger Weise erhält man, ausgehend von lla,21-Dioxy-4,17(20)-pregnadien-3-on (IV), durch die gleichen Reaktionen wie oben und selektive Oxy dation der 11-Hydroxylgruppe zur 11-Ketogruppe (z.
B. durch Veresterung der 21-Oxygruppe des lla,17a,21-Trioxy-4-pregnen-3,20-dions, das durch Osmiumtetroxyd -Behandlung und anschliessende Oxydation erhalten wurde, und Oxydation der 11 a Hydroxylgruppe zur 11-Ketogruppe mit Chrom säure) das 17a,21-Dioxy-4-pregnen-3,11,20-trion (Kendalls Verbindung E).
Die Ausgangsstoffe, näm lich die 4,17(20)-Pregnadien-3,11-dion-21-säureund ihre Ester, erhält man durch Behandlung von Ver bindungen der Formel:

in der X Chlor, Brom oder Jod, R Wasserstoff oder einen Rest der Formel -C0-0 R' und R' einen Kohlenwasserstoffrest bedeutet, mit einer Base, z. B. einem Alkalimetallhydroxyd oder -alkoholat.
Die Ketalisierung wird in der Regel in Gegen wart eines organischen Lösungsmittels bei Tempera turen zwischen Zimmertemperatur und dem Siede punkt des verwendeten Lösungsmittels durchgeführt und dauert etwa eine halbe bis etwa 18 Stunden oder mehr. Wenn man das bei der Reaktion ent stehende Wasser kontinuierlich entfernt, ist die be- vorzugte Reaktionsdauer die Zeit, in der etwa ein Moläquivalent Wasser pro Mol Steroid aus der Mi schung entfernt wird.
Unter diesen Bedingungen kann, wenn man von der freien Säure ausgeht, die Säuregruppe bis zu einem gewissen Grad durch das Alkandiol verestert werden, wobei der Glykolester gebildet wird. Durch Behandlung der Reaktions mischung mit einer wässrigen oder alkoholischen Base, vorzugsweise Alkalimetallbase, wird die Ester- gruppe wieder verseift; es ist jedoch dafür Sorge zu tragen, dass die Ketalgruppe nicht ebenfalls hydroly- siert wird. Man erhält so das Produkt Il, in dem R in 21-Stellung Wasserstoff ist, das heisst die freie Säure, praktisch rein.
Gut geeignete Alkan-a-diole und Alkan-ss-diole sind Äthylenglykol, Trimethylenglykol und alkyl- substituierte Athylen- und Trimethylenglykole, die vorzugsweise mit nicht mehr als 2 Alkylgruppen substituiert sind, wie z.
B. Propan-1,2-diol, Butan- 1,2-diol, 3-Methyl-butan-1,2-diol, Octan-1,2-diol, Butan-2,3-diol, Pentan - 2,3 - diol, 5,5-Dimethyl- octan-2,3-diol, Butan - 1,3 - diol, Pentan-2,4-diol, 4-Methyl-pentan-1,3-diol, Octan-1,3-diol.
Als Katalysatoren für die Ketalisierung kommen z. B. konzentrierte Salzsäure, konzentrierte Schwefel säure, p-Toluolsulfosäure, Benzolsulfosäure, Sulfo- essigsäure in Betracht. Diese Säuren wirken schon in Spuren katalytisch.
Geeignete Lösungsmittel sind Kohlenwasser stoffe, Halogenkohlenwasserstoffe, Äther, Ester und dergleichen, wie z. B. Benzol, Toluol, Xylol, Hexan, Heptan, Chloroform, Tetrachlorkohlenstoff, Chlor benzol, Diäthyläther, Dioxan, Tetrahydrofuran und andere oder ein Überschuss des verwendeten Alkan- diols.
Eine geeignete Methode zur Durchführung der Ketalisierung besteht darin, das Ausgangssteroid in einem organischen Lösungsmittel, das vorzugsweise mit Wasser nicht mischbar ist, z. B. Benzol, Toluol, Tetrachlorkohlenstoff, aufzulösen und dann auf Rückflusstemperatur zu erhitzen, während gleichzei tig das bei der Reaktion entstehende Wasser konti nuierlich entfernt wird, bis etwa ein Moläquivalent Wasser pro Mol des Steroids entfernt ist.
Die Reak tionszeiten können bis zur Beendigung der Ketalisie- rung 30 Minuten bis zu mehreren Tagen dauern.
Die Isolierung des ketalisierten Steroids (1I) er folgt zweckmässig durch Waschen der Reaktions mischung mit einer verdünnten Base, z. B. verdünn tem wässrigem Natriumbikarbonat, Natriumkarbonat, Kaliumhydroxyd, Natriumhydroxyd in Methanol, Natriummethylat oder dergleichen, und anschliessen des Eindampfen der Mischung zur Trockne.
Wenn das Lösungsmittel stark wasserlöslich ist, kann man die Behandlung mit der Base nach der Entfernung des Lösungsmittels durchführen oder die Destillation durch die Ausfällung des Steroids mit grossen Men gen Wasser, das vorzugsweise genügend Base zur Neutralisierung des Katalysators enthält, ersetzen.
Zur Reduktion der in 3-Stellung cyclisch ketali- sierten 14,17(20)-Pregnadien-3,11-dion-21-säure oder ihrer Ester (1I) verwendet man vorzugsweise Lithium-Aluminiumhydrid.
Die Reduktion mit Li-Al-Hydrid erfolgt in der Regel durch Zugabe einer Lösung des zu reduzieren den Ketals in einem unter den Reaktionsbedingun gen inerten organischen Lösungsmittel zu einer Lö sung oder Suspension von Li-Al-Hydrid in Äther. Andere geeignete Lösungsmittel sind Dioxan, Tetra- hydrofuran usw. sowie andere bei der Li-Al-Hydrid- reduktion gewöhnlich verwendete Lösungsmittel.
Verwendet man Äther, so wird die Reaktion in der Regel zwischen Zimmertemperatur und dem Siede punkt des Lösungsmittels durchgeführt; doch kann man auch bei Temperaturen wesentlich unterhalb Zimmertemperatur arbeiten.
Das Li-Al-Hydrid wird in der Regel in einem wesentlichen überschuss verwendet, um optimale Ausbeuten zu gewährleisten. Wenn das Steroid und das Li-Al-Hydrid gut gemischt sind und sich keine Reaktionswärme mehr entwickelt, ist die Reaktion praktisch beendigt. Gewöhnlich rührt und erwärmt man jedoch noch weiter, um die Beendigung der Re aktion sicherzustellen. Das überschüssige Li-Al- Hydrid und etwa vorhandene Steroid-Li-Al-Hydrid- komplexe können durch sorgfältige Zugabe von Wasser zum Reaktionsgemisch zersetzt werden.
Wenn man die Mischung alkalisch hält, das heisst, wenn während oder nach der Zersetzung des Li-Al- Hydrids keine Mineralsäure zugesetzt wird, kann das entsprechende, in 3-Stellung cyclisch ketalisierte 11,21-Dioxy-4,17(20)-pregnadien-3-on leicht direkt aus der Mischung isoliert werden.
Trennt man die organische Phase von der wässrigen Phase des zer setzten Reaktionsgemisches und destilliert das Lö sungsmittel ab, so erhält man einen Rückstand, der im wesentlichen aus dem gewünschten Produkt be steht und, wie oben beschrieben, isoliert oder ohne Isolierung, wie nachstehend beschrieben, weiter be handelt werden kann.
Das freie 11,21-Dioxy-4,17(20)-pregnadien-3-on (IV) erhält man durch Behandlung einer Lösung des rohen oder gereinigten, in 3-Stellung cyclisch keta- lisierten 11,21-Dioxy-4,17(20)-pregnadien-3-ons in einem organischen Lösungsmittel mit verdünnter wässriger Säure, vorzugsweise einer Mineralsäure, wie z. B. Salzsäure oder Schwefelsäure, was in der Regel bei Zimmertemperatur im Verlauf von etwa 30 Minuten bis 72 Stunden erfolgt. Da die Säure nur als Katalysator für die Hydrolyse wirkt, kann man sie in äusserster Verdünnung bis zu ziemlich hohen Konzentrationen anwenden.
Wenn die 11- Oxygruppe a7.-Konfiguration aufweist, so kann man die Hydrolyse mit ziemlich starken Säuren und bei Temperaturen, die wesentlich über der Zimmer temperatur liegen, durchführen. Im Falle der llp- Oxyverbindung dagegen führt man die Hydrolyse vorzugsweise bei Zimmertemperatur in Gegenwart einer verdünnteren Säure durch, da die 11 ss-Oxy- gruppe sich in Gegenwart von Säuren leicht ab spaltet.
Die Temperatur und Dauer der Hydro lyse hängt in gewissem Grade von der 3-Ketal- gruppe des Steroids ab. Die Isolierung des freien 11,21 - Dioxy - 4,17(20) - pregnadien - 3 - ons er folgt zweckmässig durch Neutralisierung der Reak tionsmischung, Abdestillieren des Lösungsmittels oder, wenn das Lösungsmittel mit Wasser mischbar ist, durch Zusatz eines grossen Volumens Wasser und Gewinnung des ausgefällten Produktes.
Das so erhal tene 11,21-Dioxy-4,17(20)-pregnadien-3-on (IV) erfordert, wenn das Ausgangs-Ketal rein ist, in der Regel nach dem Trocknen für die Weiterverarbei tung keine weitere Reinigung.
Aus der durch Reduktion mit Li-Al-Hydrid und Zersetzen mit Wasser erhaltenen Verbindung III kann die Ketalgruppe ohne Isolierung entfernt wer den.
Die Reduktion der in 3-Stellung ketalisierten 3,11-Diketo-4,17(20)-pregnadien-21-säure oder ihrer Alkylester (1I) erfolgt, wie gesagt, am besten mit r Li-Al-Hydrid in einem mit Wasser mischbaren, inerten Lösungsmittel, z. B. Tetrahydrofuran oder Dioxan, bei einer Temperatur wesentlich unterhalb Zimmertemperatur, das heisst mindestens unterhalb 20 C, gefolgt von einer Zersetzung des so entstan denen Komplexes mit Wasser.
Die niedrige Reak tionstemperatur vermindert Nebenreaktionen, und die Verwendung eines mit Wasser mischbaren Lö sungsmittels gewährleistet die innige Berührung des Wassers mit dem Komplex und verhindert die Bil dung eines Zweiphasensystems im Lösungsmittel. Für die Zersetzung des bei der Reaktion gebildeten Komplexes wird Wasser bevorzugt, weil die Steroide in neutraler Lösung stabiler sind und die Wärme, die bei der Reaktion von Lithium- und Aluminium ; hydroxyd mit einer Säure entsteht, ausbleibt. Die 3-Ketalgruppe kann, z.
B. mit Salzsäure oder der gleichen, ohne Auswechslung der Lösungsmittel hy- drolysiert werden, weil das bei der Reduktion ver wendete, mit Wasser mischbare Lösungsmittel sich besonders gut für die Ketalhydrolyse eignet.
Nach dem Verfahren gemäss der vorliegenden Erfindung gelingt es also, die Carboxy- oder Carbalkoxygruppe in eine Carbinolgruppe unter Er haltung der 44-3-Ketogruppierung umzuwandeln. Nach den bisher bekannten Verfahren würde nur die Ketogruppe oder beide Gruppen reduziert mit wahr scheinlich gleichzeitiger Absättigung der Doppel bindung des Steroids.
Die Ausgangsstoffe I können im einzelnen wie folgt hergestellt werden: a) Zu einer Mischung von 3,4 cm3 einer 3,4 normalen Lösung von methanolischem Natrium- methylat, 0,45 cm3 absolutem Äthanol und 20 cm3 trockenem Benzol, aus der zunächst 8 cm3 abdestil- liert wurden, gibt man nach dem Abkühlen 2,3 cm3 Äthyloxalat und eine Lösung von 3,
28 g 11-Keto- progesteron in 38 cm3 wasserfreiem Benzol. Die Lösung wird trüb, und es bildet sich ein gelber Nie derschlag. Die Mischung wird 90 Minuten gerührt. Darauf gibt man 55 cm3 Äther zu und rührt weitere 60 Minuten, wonach man 130 cm3 Äther zufügt. Das so ausgefällte gelbe Natriumenolat des 11-Keto- 21-äthoxyoxalyl-progesterons wird filtriert, mehr mals mit je 50 cm3 Äther gewaschen und wiegt nach dem Trocknen 3,65 g.
Die Ätherwaschlaugen ent halten 0,54 g nicht umgesetztes 11-Keto-progesteron. Die Ausbeute ist 81% der Theorie oder, bezogen auf das umgesetzte I1-Keto-progesteron, praktisch quantitativ. Durch Ansäuern der wässrigen Lösung des so entstandenen Natriumenolats erhält man 11-Keto-21- äthoxyoxalyl-progesteron, das durch Filtrieren ge wonnen werden kann.
b) Zu einer Lösung von 4,5 g (0,01 Mol) Na- triumenolat des 11-Keto-21-äthoxyoxalyl-progeste- rons und 2 g Kaliumacetat in 70 em3 Eisessig wird unter Rühren 3,09 g (1 cm3; 0,0193 Mol) Brom tropfenweise bei Zimmertemperatur hinzugegeben, wonach man die Reaktionsmischung mit einem grossen Volumen Wasser vermischt.
Man dekantiert die wässrige Schicht vom ausgeschiedenen gelben viskosen Produkt ab, das darauf nach dem Lösen in Alkohol und Zutropfen von Wasser als weisser fester Stoff ausgefällt wird. Die Ausbeute an so entstan denen 11 - Keto - 21,21 - dibrom - 21 äthoxyoxalyl- progesteron beträgt nach dem Filtrieren und Trock nen 4 g, was 70,9/o der Theorie entspricht.
c) Zu einer Lösung von 5,9g (0,01 Mol) 11- Keto - 21,21 - dibrom-21-äthoxyoxalyl-progesteron, nach b) hergestellt, in 150 cm-' Methanol wird 3,24 g (0,06 Mol) Natriummethylat (Handelsprodukt) zuge geben.
Man hält die verbleibende Mischung 3 Stun den bei etwa 25 C, verdünnt das Ganze mit Wasser und extrahiert zweimal mit Methylenchlorid. Die Methylenchloridextrakte werden über wasserfreiem Natriumsulfat getrocknet und das Lösungsmittel bei normalem Druck abdestilliert. Man erhält in quanti tativer Ausbeute 3,6 g 3,11-Diketo-4,17(20)-pregna- dien-21-säuremethylester als Öl.
Das Öl wird in 50 cm3 Benzol gelöst und über einer Säule mit 170 g Florisil (Markenprodukt, ein synthetisches Magnesiumsilikat) chromatographiert. Die Säule wird mit 400 cm3 folgender Lösungsmittel in der ange führten Reihenfolge entwickelt: 3 Portionen Methy- lenchlorid, 5 Portionen Methylenchlorid plus 511/o Aceton und 1 Portion Aceton.
Die mit Methylen- chlorid plus 5% Aceton erhaltenen Eluate werden vereinigt und das Lösungsmittel entfernt, wodurch 1,5 g kristallinisches 3,11-Diketo-4,17(20)-pregna- dien-21-on-säuremethylester zurückbleibt, der nach Umkristallisieren aus Aceton und Skellsolve B (Hexankohlenwasserstoffe) bei 213-214 C schmilzt.

d) In genau gleicher Weise wie unter e) beschrie ben, wird 3,11-Diketo-4,17(20)-pregnadien-21-säure aus dem Natriumenolat des 11-Keto-21-äthoxy- oxalyl-progesterons hergestellt unter Verwendung von 3,4 g (0,06 Mol) Kaliumhydroxyd in 10 cm3 Wasser anstelle des Alkoholats, wobei das Kalium salz der gewünschten Säure entsteht.
Man isoliert die 3,11-Diketo-4,17(20)-pregnadien-21-säure durch Waschen der Reaktionsmischung mit Methylen- chlorid, Ansäuern mit verdünnter Salzsäure und Extrahieren des so entstandenen öligen Niederschlags mit Benzol. Das Benzolextrakt wird mit Wasser ge waschen, getrocknet und dann das Benzol unter ver mindertem Druck abdestilliert. Die verbleibende 3,11-Diketo-4,17(20)-pregnadien-21-säure schmilzt nach mehrmaligem Umkristallisieren bei 255 bis 260 C.
In den folgenden Beispielen werden jeweils nur einzelne Stufen des Verfahrens beschrieben: <I>Beispiel 1</I> Zu einer Lösung von 1,5 g (0,0042 Mol) 3,11- Diketo-4,17 (20)-pregnadien - 21-on-21-säure-methyl- ester in 150 cm3 Benzol gibt man 7,5 cm3 Äthylen- glykol und 0,
15 g p-Toluolsulfosäure und erhitzt das Ganze unter Rühren 51/2 Stunden am Rückfluss. Die abgekühlte Reaktionsmischung wird mit 100 cm3 1 @ /oiger wässriger Natriumbicarbonatlösung gewa schen.
Darauf giesst man die Benzolschicht durch eine Säule aus 150 g Florisil . Die Säule wird mit 100 cm-' Portionen der folgenden Lösungsmittel in der angegebenen Reihenfolge entwickelt: 8 Portionen Methylenchlorid und 3 Portionen Methylenchlorid plus 4% Aceton. Die Methylenchlorid-Eluate ent- halten 1,08 g 3-Äthylenglykolketal des 3,11-Diketo- 4,17(20)-pregnadien-21-säuremethylesters,
der nach Umkristallisieren aus einer Mischung von Äthyl- acetat und Skellysolve B bei 188-190 C schmilzt und die nachstehende Analyse zeigt.
Die mit Methy- lenchlorid plus 4% Aceton erhaltenen Eluate ent- halten 0,39 g reinen 3,11-Diketo-4,17(20)-pregna- dien-21-säure-methylester. Die Ausbeute beträgt 87% der Theorie,
bezogen auf die Menge des um- gesetzten Ausgangssteroids.

B. des Äthyl-, Propyl-, Butyl-, Amyl-, Hexyl-, Heptyl-, Octylesters, durch Umsetzung der betreffenden Alkylester dieser Säure mit Äthylenglykol in Gegenwart einer Säure als Katalysator, z. B. konzentrierter Salzsäure, Benzol- sulfosäure, p-Toluolsulfosäure und dergleichen, her gestellt.
<I>Beispiel 2</I> In genau gleicher Weise wie in Beispiel 1 be schrieben, lässt man 0,75 g (0,0021 Mol) 3,11-Di- keto - 4,17(20) - pregnadien - 21 - säure-methylester 7 Stunden mit 4 cm3 Äthylenglykol in 100 cm3 Benzol in Gegenwart von 0,075 g p-Toluolsulfosäure unter gleichzeitiger Entfernung des bei der Reaktion ent stehenden Wassers reagieren.
Man wäscht die Mi- schung nach dem Abkühlen mit kalter 2%iger Na- triumbikarbonatlösung und mit Wasser,
extrahiert die Waschflüssigkeiten mit Benzol und vereinigt die Extrakte mit der Benzolschicht. Die vereinigten Benzollösungen trocknet man über Natriumsulfat und giesst sie durch eine 75-g-Säule von Florisil . Die Säule wird mit Skellysolve B plus 5% Aceton entwickelt.
Man eluiert mit 100-cm3-Fraktionen, wobei die Fraktionen 5, 6 und 7 261, 408 bzw. 118 mg festes kristallines Produkt enthalten (die gesamte Ausbeute beträgt 93,511/o der Theorie). Die vereinigten festen Produkte werden aus einer Mi schung von 25 cm3 Skellysolve B und 8 cm3 Äthyl- acetat und 2 Tropfen Pyridin umkristallisiert.
Die erste Kristallfraktion des 3-Äthylen,lykolketals des 3,11- Diketo - 4,17 (20) -,pregnadien-21-säure-methyl- esters wiegt 0,580 g und schmilzt bei 177-179 C; [a] D in Aceton = @- 9 . Die zweite Fraktion hat einen Schmelzpunkt von 165-177 C.
<I>Beispiel 3</I> In gleicher Weise wie in Beispiel 1 stellt man aus 3,11-Diketo-4,17(20)-pregnadien-21-säure-me- thylester durch Umsetzung mit Trimethylenglykol in Gegenwart einer Säure als Katalysator das 3-Tri- methylenglykolketal des 3,11 - Diketo - 4,17 (20)- pregnadien-21-säure-methylesters her.
In analoger Weise erhält man unter Verwendung anderer Alkandiole und anderer Alkylester die ent sprechenden 3-Ketale.
<I>Beispiel 4</I> 80 cm3 der überstehenden Flüssigkeit einer Lö sung von 1 g Li-Al-Hydrid in 100 cm3 absolutem Äther wird in einem Kolben im Eisbad gekühlt und 10 Minuten mit 1 g des 3-Äthylenglykolketals von 3,11-Diketo - 4,17(20) - pregnadien-21-säure-methyl- ester in 50 cm3 trockenem Benzol verrührt.
Man zersetzt das Reaktionsgemisch darauf mit einer ge sättigten wässrigen Natriumkaliumtartratlösung, die man vorsichtig zugibt, dekantiert die Lösungsmittel schicht von der Mischung ab, wäscht zweimal mit je 50 cm3 Benzol und vereinigt dann mit der Lö sungsmittelschicht. Die vereinigten Lösungen werden darauf über Natriumsulfat getrocknet und auf 75 g Florisil gegossen.
Man entwickelt mit 250 cms Portionen folgender Lösungsmittel in der angeführ ten Reihenfolge: 4 Portionen Skellysolve B plus 101% Aceton, 6 Portionen Skellysolve B plus 1511/o Aceton und schliesslich mehrere Portionen Skelly- solve B plus 25% Aceton. Die ersten
zwei mit Skellysolve B plus 101/o Aceton erhaltenen Eluate vereinigt man, destilliert das Lösungsmittel ab, kristallisiert das darin enthaltene, 488 mg wiegende feste Produkt aus Aceton plus Skellysolve B um und erhält das 3-Äthylenglykolketal des Ilss,21-Dioxy- 4,17(20)-pregnadien-3-ons vom Schmelzpunkt 183 bis 187 C; [a] D = - 15 in Aceton.

- pregnadien - 21 - säure- methylesters mit Li-Al-Hydrid in Tetrahydrofuran erhält man das 3-Trimethylenglykolketal des 11a,21- und 11ss,21-Dioxy-4,17(20)-pregnadien-3-ons, das auch durch Umsetzung der andern Alkylester des 3-Trimethylenglykolketals von 3,11-Diketo-4,17(20)- pregnadien-21-säure hergestellt werden kann.
Andere cyclische 3-Ketale des 11,21-Dioxy- 4,17(20)-pregnadien-3-ons, in welchem die 11-Oxy- gruppe die a- oder fl-Konfiguration besitzt, kann man herstellen durch Umsetzung der entsprechend ketalisierten 3,11-Diketo-4,17(20)-pregnadien-21- säure oder ihrer Alkylester mit einem Alkalimetall- Aluminiumhydrid oder dergleichen.
<I>Beispiel 6</I> Zu einer Lösung von 2,4 g (0,0642 Mol) 3-Äthylenglykolketal des 11a,21-Dioxy-4,17(20)- pregnadien-3-ons in 160 cm3 Aceton gibt man 4 Tropfen konzentrierte Schwefelsäure in 40 cm3 Wasser und erhitzt die Mischung 2 Stunden am Rückfluss. Die abgekühlte Lösung neutralisiert man durch Zugabe einer verdünnten wässrigen Natrium bikarbonatlösung und man destilliert das Aceton unter vermindertem Druck ab.
Man extrahiert das ausgefällte Produkt mit Methylenchlorid und trock net nach Entfernung der wässrigen Schicht über Na triumsulfat. Darauf destilliert man die trockene Me- thylenchloridlösung bei vermindertem Druck bis zur Trockne, und erhält 1,96 g eines Produktes, das im wesentlichen aus 11a,21-Dioxy-4,17(20)-pregnadien- 3-on besteht. Die Ausbeute beträgt 93% der Theo- rie.
<I>Beispiel 7</I> Eine Lösung von 0,572 g (0,0015 Mol) des 3-Äthylenglykolketals des llss,21-Dioxy-4,17(20)- pregnadien-3-ons in 40 cm3 Aceton verdünnt man mit Wasser auf ein Volumen von 50 cms und gibt 8 Tropfen konzentrierte Schwefelsäure zu, wonach man die Reaktionsmischung 24 Stunden bei Zimmer temperatur hält und dann durch Zugabe einer ge- sättigten wässrigen Natriumbikarbonatlösung alka lisch macht.
Man verdampft das Aceton und gibt Methylenchlorid und noch Wasser zu, entfernt die Methylenchloridschicht und destilliert das Lösungs mittel daraus ab. Der Rückstand wiegt nach dem Trocknen im Vakuum 0,518 g und besteht aus 11 ,B,21-Dioxy-4,17(20)-pregnadien-3-on.
Durch Umkristallisieren dieses Produktes aus einer Mischung von Äthylacetat und Skellysolve B erhält man Kristalle, die bei 156-l58 C schmelzen und in Aceton eine optische Drehung [a]23 = + 128 haben.

The ketalization is usually carried out in the presence of an organic solvent at tempera tures between room temperature and the boiling point of the solvent used and takes about half an hour to about 18 hours or more. If the water formed in the reaction is continuously removed, the preferred reaction time is the time in which about one molar equivalent of water per mole of steroid is removed from the mixture.
Under these conditions, starting from the free acid, the acid group can be esterified to a certain extent by the alkanediol, the glycol ester being formed. By treating the reaction mixture with an aqueous or alcoholic base, preferably an alkali metal base, the ester group is saponified again; however, it must be ensured that the ketal group is not also hydrolyzed. The product II is obtained in which R is in the 21-position hydrogen, that is to say the free acid, practically pure.
Suitable alkane-a-diols and alkane-ss-diols are ethylene glycol, trimethylene glycol and alkyl-substituted ethylene and trimethylene glycols, which are preferably substituted with not more than 2 alkyl groups, such as.

A suitable method of carrying out the ketalization is to soak the starting steroid in an organic solvent, which is preferably immiscible with water, e.g. B. benzene, toluene, carbon tetrachloride, to dissolve and then to reflux temperature, while at the same time the water formed in the reaction is continuously removed until about one molar equivalent of water per mole of the steroid is removed.
The reaction times can last from 30 minutes to several days until the ketalization is complete.
The isolation of the ketalized steroid (1I) he follows conveniently by washing the reaction mixture with a dilute base, for. B. diluted system aqueous sodium bicarbonate, sodium carbonate, potassium hydroxide, sodium hydroxide in methanol, sodium methylate or the like, and then evaporating the mixture to dryness.
If the solvent is highly water-soluble, the treatment with the base can be carried out after the solvent has been removed or the distillation can be replaced by the precipitation of the steroid with large quantities of water, which preferably contains sufficient base to neutralize the catalyst.

If ether is used, the reaction is usually carried out between room temperature and the boiling point of the solvent; but you can also work at temperatures significantly below room temperature.
The Li-Al hydride is generally used in a substantial excess in order to ensure optimum yields. When the steroid and the Li-Al hydride are well mixed and the heat of reaction no longer develops, the reaction is practically complete. However, stirring and heating is usually continued to ensure that the reaction has ended. The excess Li-Al hydride and any steroid Li-Al hydride complexes that are present can be decomposed by carefully adding water to the reaction mixture.
If the mixture is kept alkaline, that is, if no mineral acid is added during or after the decomposition of the Li-Al hydride, the corresponding 11,21-dioxy-4,17 (20) - cyclically ketalised in the 3-position can be pregnadien-3-one can easily be isolated directly from the mixture.

The temperature and duration of hydrolysis depend to some extent on the 3-ketal group of the steroid. The isolation of the free 11,21 - dioxy - 4,17 (20) - pregnadien - 3 - one is expediently carried out by neutralizing the reaction mixture, distilling off the solvent or, if the solvent is miscible with water, by adding a large volume of water and recovering the precipitated product.
The 11,21-dioxy-4,17 (20) -pregnadien-3-one (IV) obtained in this way, if the starting ketal is pure, usually does not require any further purification after drying for further processing.
From the compound III obtained by reduction with Li-Al hydride and decomposition with water, the ketal group can be removed without isolation.
The reduction of the 3,11-diketo-4,17 (20) -pregnadiene-21-acid or its alkyl esters (1I) ketalized in the 3-position is, as mentioned, best carried out with r Li-Al hydride in one with water miscible, inert solvents, e.g. B. tetrahydrofuran or dioxane, at a temperature well below room temperature, that is at least below 20 C, followed by decomposition of the complex with water which resulted.
The low reaction temperature reduces side reactions, and the use of a water-miscible solvent ensures that the water comes into close contact with the complex and prevents the formation of a two-phase system in the solvent. For the decomposition of the complex formed in the reaction, water is preferred because the steroids are more stable in neutral solution and the heat generated in the reaction of lithium and aluminum; hydroxide is formed with an acid, does not occur. The starting materials I can be prepared individually as follows: a) To a mixture of 3.4 cm3 of a 3.4 normal solution of methanolic sodium methylate, 0.45 cm3 of absolute ethanol and 20 cm3 of dry benzene, from which 8 cm3 were distilled off, after cooling, 2.3 cm3 of ethyl oxalate and a solution of 3,
28 g of 11-keto-progesterone in 38 cm3 of anhydrous benzene. The solution becomes cloudy and a yellow precipitate forms. The mixture is stirred for 90 minutes. Then add 55 cm3 of ether and stir for a further 60 minutes, after which 130 cm3 of ether are added. The yellow sodium enolate of 11-keto-21-ethoxyoxalyl-progesterone precipitated in this way is filtered, washed several times with 50 cm3 of ether each time and, after drying, weighs 3.65 g.
The ether wash liquors contain 0.54 g of unreacted 11-keto-progesterone. The yield is 81% of theory or, based on the converted I1-keto-progesterone, practically quantitative. By acidifying the aqueous solution of the resulting sodium enolate, 11-keto-21-ethoxyoxalyl-progesterone is obtained, which can be obtained by filtration.
b) To a solution of 4.5 g (0.01 mol) of sodium enolate of 11-keto-21-ethoxyoxalyl-progesterone and 2 g of potassium acetate in 70 cubic meters of glacial acetic acid, 3.09 g (1 cm3; 0.0193 mol) of bromine was added dropwise at room temperature, after which the reaction mixture was mixed with a large volume of water.
The aqueous layer is decanted from the yellow viscous product which has separated out and which is then precipitated as a white solid after dissolving in alcohol and adding dropwise water. The yield of 11-keto-21.21-dibromo-21 ethoxyoxalyl progesterone produced in this way is 4 g after filtration and drying, which corresponds to 70.9% of theory.
c) To a solution of 5.9 g (0.01 mol) of 11-keto-21.21-dibromo-21-ethoxyoxalyl-progesterone, prepared according to b), in 150 cm- 'methanol, 3.24 g (0, 06 mol) of sodium methylate (commercial product) are added.
The remaining mixture is kept for 3 hours at about 25 ° C., the whole is diluted with water and extracted twice with methylene chloride. The 3,11-diketo-4,17 (20) -pregnadiene-21-acid is isolated by washing the reaction mixture with methylene chloride, acidifying with dilute hydrochloric acid and extracting the oily precipitate thus formed with benzene. The benzene extract is washed with water, dried and then the benzene is distilled off under reduced pressure. The remaining 3,11-diketo-4,17 (20) -pregnadien-21-acid melts after repeated recrystallization at 255 to 260 C.
In the following examples, only individual steps of the process are described in each case: <I> Example 1 </I> To a solution of 1.5 g (0.0042 mol) 3,11-diketo-4,17 (20) -pregnadiene - 21-on-21-acid methyl ester in 150 cm3 benzene is added 7.5 cm3 ethylene glycol and 0,
15 g of p-toluenesulfonic acid and heated the whole thing while stirring for 51/2 hours under reflux. The cooled reaction mixture is washed with 100 cm3 1% aqueous sodium bicarbonate solution.
The benzene layer is then poured through a column of 150 g of Florisil. The column is developed with 100 cm portions of the following solvents in the order given: 8 portions of methylene chloride and 3 portions of methylene chloride plus 4% acetone. The methylene chloride eluates contain 1.08 g of 3-ethylene glycol ketal of the 3,11-diketo-4,17 (20) -pregnadiene-21-acid methyl ester,
which after recrystallization from a mixture of ethyl acetate and Skellysolve B melts at 188-190 C and the analysis below shows.
The eluates obtained with methylene chloride plus 4% acetone contain 0.39 g of pure 3,11-diketo-4,17 (20) -pregnadiene-21-acid methyl ester. The yield is 87% of theory,

The cooled solution is neutralized by adding a dilute aqueous sodium bicarbonate solution and the acetone is distilled off under reduced pressure.
The precipitated product is extracted with methylene chloride and, after removal of the aqueous layer, dry over sodium sulfate. The dry methylene chloride solution is then distilled to dryness under reduced pressure, and 1.96 g of a product is obtained which consists essentially of 11a, 21-dioxy-4,17 (20) -pregnadien-3-one. The yield is 93% of theory.
<I> Example 7 </I> A solution of 0.572 g (0.0015 mol) of the 3-ethylene glycol ketal des IIss, 21-dioxy-4,17 (20) pregnadien-3-one in 40 cm 3 of acetone is diluted with Water to a volume of 50 cms and 8 drops of concentrated sulfuric acid are added, after which the reaction mixture is kept at room temperature for 24 hours and then made alkaline by adding a saturated aqueous sodium bicarbonate solution.
The acetone is evaporated off and methylene chloride and more water are added, the methylene chloride layer is removed and the solvent is distilled off from it. After drying in vacuo, the residue weighs 0.518 g and consists of 11, B, 21-dioxy-4,17 (20) -pregnadien-3-one.
By recrystallizing this product from a mixture of ethyl acetate and Skellysolve B, crystals are obtained which melt at 156-158 ° C. and have an optical rotation [a] 23 = + 128 in acetone.
