Gegenstand der vorliegenden Erfindung ist ein Verfahren zur Herstellung der bisher nicht bekannten Ester der Karbaminsäure mit sekundären oder tertiären Acetylenalkoholen der allgemeinen Formel

Man kann also z. B. die Kohlensäuredihalogenide zuerst mit Ammoniak oder Aminen in am Stickstoff substituierte oder nicht substituierte Karbaminsäurehalogenide überführen und diese alsdann mit den tertiären oder sekundären Acetylencarbinolen zu den gewünschten Estern umsetzen. Man kann aber auch zuerst die Kohlensäuredihalogenide, wie Phosgen, mit den tertiären oder sekundären Acetylenearbinolen umsetzen und dann auf die so erhaltenen Halogenameisensäureester Ammoniak oder Amine zur Einwirkung bringen.
Ferner kann man primÏre Amine zuerst mit den Kohlensäuredihalogeniden zu den entsprechenden Isoeyanaten kondensieren und alsdann an diese die Acetylencarbinole anlagern.
Bei der bekannten Reaktionsfähigkeit der Acetylengruppe, die mit dem Auftreten un- erwünschter Nebenreaktionen, z. B. von Ring- sehliissen, rechnen liess, und bei der bekannten besonderen Empfindlichkeit tertiärer Halogen- ameisensäureester (vgl. DRP. Nr. 254471) war es einigermassen überraschend, dass das er findungsgemässe Verfahren mit technise durchaus brauchbaren Ausbeuten verläuft.
Die Verfahrensprodukte besitzen thera peutisch wertvolle Eigenschaften ; z. B. zeigen einige von ihnen eine ausgeprägte hypnotische Wirkung bei gleichzeitig geringen Nebenerscheinungen.
Beispiel 1 34 g (430 mMol) Karbaminsäureehlorid (hergestellt aus Phosgen und Ammoniak) werden in 250 em3 absolutem Ather unter Eiskiihlung langsam mit 42 g (430 mMol) 3-Me thyl-pentin- (1)-ol- (3) versetzt. Naeh Stehen. über Nacht wird mit Wasser gewasehen und in üblicher Weise aufgearbeitet. Destillation im Vakuum ergibt ein farbloses Destillat, Kp. 12 120-121 . Kristalle aus Hexan F. 56-58 .
Das Reaktionsprodukt bildet ein Silbersalz, lässt sich katalytisch zum Karbaminsäureester des Methyldiäthylkarbinols hydrieren und erweist sich damit als Karbaminsäureester des 3-Methyl-pentin-(l)-ols- (3).
Beispiel. 2
Eine Lösung von 51 cm3 (750 Mol) Phosgen in 150 em3 absolutem ither wird unter Eiskochsalzlösung zu 67 em3 (720 mMol) 3-Methyl-butin- (1)-ol- (3) in 100 em3 ab solutem Äther zugegeben ; anschliessend lässt man langsam 96 cm3 (750 mMol) Chinolin in 100 cm3 absolutem Äther zutropfen, wobei die Innentemperatur-5 nicht übersteigen soll.
Nach Stehen ber Nacht bei 0 wird vom Chinolinhydrochlorid abgesaugt und das Filtrat unter gâter Eiskochsalzkühlung zu einer ätherischen Lösung von Ammoniak gegeben, wobei weiter Ammoniakgas eingeleitet wird.
Nach der Aufarbeitung analog Beispiel 1 werden 30 g Karbaminsäureester des 3-Methyl- butin- (1)-oIs- (3), Kp. 12 111¯, F. 108 (aus Cyclohexan), erhalten.
Beispiel 3
Eine Losung von 34 em3 (500 mMol) flüss.
Phosgen in 150 cm3 abs. ¯ther wird, wie in Beispiel 2, zuerst mit 62 g (500 mMol) 1 Äthinyl-eyelohexanol-(l) und 64 cm. (500 mMol) Chinolin und dann nach Weiterver- arbeitung gemäss Beispiel 1 und 2 mit Ammoniak inÄther umgesetzt. Man erhält 45 g (53% der Theorie) Karbaminsäureester des 1-¯thinyl-cyclohexan-ol-(1); Kp.3 108-110¯ ; farblose Nadeln aus Cyclohexan, F. 94-96".
Beispiel 4
Eine Lösung von 34 cm3 (500 mMol) flüss.
Phosgen in 150 cm3 abs. ¯ther wird wie in Beispiel 2 mit 49 g (500 mMol) 3-Methyl-pentin- (l)-ol- (3) und 64 em3 Chinolin umgesetzt und nach Abtrennung des Chinolin-hydrochlorids bei-10 in eine Lösung von etwa 40 g Me- thylamin in 100 em3 abs. Äther eingetropft.
Man lässt das Gemiseh im Lauf einiger Stun- den auf Zimmertemperatur kommen, wäscht es dann alkaliseh und sauer ans und erhält schliesslich dureh Vakuumdestillation den N-Methyl-karbaminsäureester des Methyl- pentinols in 45% iger Ausbeute, Kp.14 111 bis 112 , F. 53-54 , farblose Nadeln aus Hexan.
Beispiel. 5
Zu einer Lösung von 62 g 1-¯thinyl-cyclo hexanol-(1) in 200 cm3 abs. Toluol und 50 bis 52 g Phosgen werden unter Rühren in etwa 11/2 Std. 61 g Kollidin in 70 cm3 Toluol zugetropft, wobei die Temperatur durch Eiskühlung auf + 5 bis + 10¯ gehalten wird.
Nach 1-2st ndigem Weiterr hren lϯt man über Nacht bei etwa + 10¯ stehen. Das s ausgefallene Kollidinhydrochlorid wird abgesaugt und mit Toluol nachgewaschen, die Filtrate werden vereinigt. Durch 250 em3 vor gelegtes Toluol wird ein ständiger NH3 Strom geleitet und obiges Toluolfiltrat unter Eiskühlung'so langsam zugegeben, dass die Temperatur + 10 nicht übersteigt. Anschlie- ssend rührt man noch etwa 15 Minuten weiter und arbeitet dann wie in den vorhergehenden Beispielen auf. Die getrocknete Toluollösung des Karbaminsäureesters wird auf-5 bis 0 abgekühlt, wobei die Hauptmenge kristallisiert. Naeh dem Absaugen kann das Filtrat eingeengt und erneut zur Kristallisation angesetzt oder der Vakuumdestillation unterworfen werden.
Ausbeute insgesamt 50, 8 g -60, 7% der Theorie.
Beispiel 6
6, 2 g Äthinyleyelohexanol werden in 100 cm3 abs. Pyridin gelöst, mit 5, 5 cm3 Phe nylisoeyanat, hergestellt aus Phosgen und Ani lin, versetzt und 17 Stunden unter Rückfluss gekocht. Die Aufarbeitung über Äther liefert neben Diphenylharnstoff 4, 2 g N-Phenyl carbaminsäure-1-äthinyl-cyelohexylester-Kri- stalle, aus Hexan. F. 94-96¯.
Beispiet 7
Zu 23 g (0, 3 Mol) Karbaminsaurechlorid (hergestellt aus Phosgen und Ammoniak) in 150 cm3 trockenem Äther werden unter R hren bei etwa 0 32 g 1-Propinyl-cyelohexanol- (1) in 150 cm3 trockenem ¯ther zugetropft ; nach Stehen über Nacht wird mit Wasser auf gearbeitet, die Losung getrocknet und stark eingeengt und tiefgekühlt, wobei 10 g (25% der Theorie) farblose Kristalle vom F. 137 bis 139 erhalten werden, die den Karbaminsäurecster des Propinyl-eyelohexanols darstellen.
Analog entsteht aus Phenät. hinyleyclo- hexanol der entsprechende Karbaminsäureester, F. 110-111¯, Kristalle aus Hexan.
Beispiel 8
28 g Diäthylkarbaminsäurechlorid (hergestellt aus Phosgen und DiÏthylamin), 31 g 1-¯thinyl-cyclohexanol- (1) und 31 g 2, 4, 6 Kollidin werden in 150 cm3 Toluol gelöst und 5 Stunden unter Rückfluss gekocht. Vom ausgeschiedenen Kollidinhydrochlorid wird abgetrennt, die Toluollosung mit verdünnter Säure und Lauge und Wasser gewaschen, getrocknet und eingeengt und im Vakuum destilliert. Hauptfraktion Kp2 94-96 , F. 30 bis 33¯. Ausbeute 15 g (27% der Theorie) Di äthylcarbaminsäureester des Äthinyleyelo- hexanols.
Beispiel 9
Eine gekühlte Lösung von 49, 6 g 1-Äthinyl- eyelohexanol-(1) (1 Mol) in 164 cm3 Toluol (getrocknet ber Natrium) wird unter Rühren bei etwa 0 mit einer gleichfalls gekühlten Lösung von 51, 58 g Phosgen (1, 3 Mol) in 157, 6 cm3 Toluol vereinigt. Hierzu gibt man unter Rühren bei + 20 bis + 40 aus einem Tropftrichter eine Losung von 27, 2 g Trimethylamin (1, 15 Mol) in 180 cm3 Toluol im Lauf von 60-70 Minuten hinzu. Nach 1/2stündigem Nachrühren bei etwa 0 wird das ausgeschiedene Trimethylaminhydrochlorid auf der Nutsche abgesaugt und mit viermal so viel Toluol nachgewaschen, dass das Salz nach jedesmaligem Durcharbeiten gerade mit Lösungsmitteln bedeekt ist.
Das Salz wird bei 60 im Trockenschrank getrocknet. Man erhält 41, 45 g entsprechend 94, 5% vom eingesetzten Trimethylamin. Filtrat und Waschtoluol werden in den Kolben zurückgegeben und durch Einleiten von Ammoniakgas bei etwa + 3 amidiert. Nach Beendigung der Amidierung (Dauer etwa = 11/2 Stunde) wird die Reaktionsmischung mit 100 cm3 Wasser in einen Schütteltrichter gebracht und mit 2 Liter ¯ther verdünnt. Man lässt die Ammonchloridlösung ab, wäscht die Ätherschicht nacheinander mit Wasser, mehrmals mit 10% iger Schwefelsäure und dann wieder mit Wasser. Diese Waschwässer äthert man zu sammen mit der Ammonehloridlösung aus.
Die Schwefelsäurewaschwässer werden ebenfalls mehrmals mit Äther ausgeschüttelt und dieser mit der Hauptmenge vereinigt. Nach dem Trocknen ber Natriumsulfat wird die gesamte Ätherlösung (zum Schluss im Vakuum) eingedampft. Sobald Kristallisation einsetzt, f gt man 100 cm3 Benzin (50 bis 60 ) hinzu und lässt zweeks Vervollständigung der Kristallisation bei 0 mehrere Stunden stehen. Man saugt auf der Nutsehe ab und wäscht mehrmals mit Benzin. Naeh dem Trocknen bei 50 erhält man 54, 45 g Äthinyl- cyclohexylurethan vom F. 93-95 , entsprechend 81, 5% der Theorie.
Aus den Benzinlaugen gewinnt man nach dem Eindampfen und Destillation im Vakuum noch etwas Aus- gangsmaterial zurüek.
Wiedergewinnung des Trimethylamins :
In einem Destillationskolben gibt man zu 95 g (1 Mol) des erhaltenen Trimethylamin hydrochlorides aus einem Tropftrichter eine 50%ige Natronlauge mit 45 g NaOH hinzu, wobei das entwiekelte Trimethylamin in einer tiefgekiihlten Vorlage aufgefangen wird.
Dureh Erwärmung der Reaktionsmischung auf etwa 80 werden die letzten Anteile Trimethylamin übergetrieben. Man gewinnt so 58 g bereits weitgehend trockenes Trimethylamin, das man am besten nach Verdünnung mit Toluol mit Ätzkali und Bariumoxyd vollständig trocknet. Die Ausbeute beträgt 98 bis 99% der Theorie. Die Toluollösung kann direkt f r weitere Umsetzungen verwendet werden.
Beispiel 10
Eine Lösung von 70 g Butin- (l)-ol- (3) und 99 g Phosgen in 400 cm3 abs. Äther wurde bei etwa-10 in 4 Stunden unter Rühren mit 129 g Chinolin in abs.Äther versetzt. Nach längerem Stehen bei-6 wurde vom Chino linhydrochlorid abgesaugt und die ätherische Lösung bei-5 unter Rühren und Durchleiten von Ammoniak in vorgelegten Ather getropft. Nach Beendigung der Reaktion wurde in blicher Weise aufgearbeitet und das Butin- (1)-ol- (3)-urethan durch Destillation gereinigt. Kp. 12 108-109¯, F. 48-50", Kristalle aus Hexan. Ausbeute 50 g = 49% der Theorie.
Beispiel 11
Eine Lösung von 50 g Athinyl-eyelo- hexenyl-carbinol und 40 g Phosgen in 200 cm3 Toluol wird langsam bei + 10¯ unter Rühren mit 56 em3 2, 4, 6-Kollidin in 50 em3 Toluol versetzt, anschliessend weitere 2 Stunden gerührt und dann wie in Beispiel 1 weiter verarbeitet. Nach Einengen der Losung erhält man 25, 8 g (40% der Theorie) des entsprechenden Karbaminsäureesters ; nach 2facher Umkristallisation aus Äther F. 117-118 .
Beispiel 12
Zu 25 g Karbaminsäurechlorid (hergestellt aus Phosgen und Ammoniak) in 50 em3 troekenem Äther werden bei etwa 0 hunter Rühren 39 g (Endomethyleneyclohexenyl)- äthinyl-carbinol in 100 cm3 trockenem ¯ther zugetropft ; nach Stehen ber Nacht wird mit Wasser aufgearbeitet, die Losung stark eingeengt und tiefgek hlt. 28 g = 53% der Theorie. F. 114-116 .

B. first convert the carbonic acid dihalides with ammonia or amines into nitrogen-substituted or unsubstituted carbamic acid halides and then convert these with the tertiary or secondary acetylene carbinols to give the desired esters. But you can also first react the carbonic acid dihalides, such as phosgene, with the tertiary or secondary acetylenearbinols and then bring ammonia or amines to act on the haloformic acid esters thus obtained.
Furthermore, primary amines can first be condensed with the carbonic acid dihalides to form the corresponding isocyanates and then the acetylene carbinols can be added to these.
Given the known reactivity of the acetylene group, which is associated with the occurrence of undesired side reactions, e.g. B. von Ringsehliissen, and with the known particular sensitivity of tertiary haloformic acid esters (cf. DRP. No. 254471) it was somewhat surprising that the process according to the invention proceeds with technically quite useful yields.
The process products have therapeutically valuable properties; z. B. some of them show a pronounced hypnotic effect with at the same time few side effects.
Example 1 34 g (430 mmol) of carbamic acid chloride (prepared from phosgene and ammonia) are slowly added to 42 g (430 mmol) of 3-methylpentine- (1) -ol- (3) in 250 cubic meters of absolute ether while cooling with ice. Near standing. It is washed with water overnight and worked up in the usual way. Distillation in vacuo gives a colorless distillate, bp 12 120-121. Crystals from hexane F. 56-58.
The reaction product forms a silver salt, can be catalytically hydrogenated to the carbamic acid ester of methyl diethyl carbinol and thus proves to be the carbamic acid ester of 3-methylpentyn- (1) -ols- (3).
Example. 2
A solution of 51 cm3 (750 mol) of phosgene in 150 cubic meters of absolute ether is added to 67 cubic meters (720 mmol) of 3-methyl-butyn- (1) -ol- (3) in 100 cubic meters of absolute ether under ice saline; then 96 cm3 (750 mmol) of quinoline in 100 cm3 of absolute ether are slowly added dropwise, the internal temperature should not exceed -5.
After standing overnight at 0, the quinoline hydrochloride is filtered off with suction and the filtrate is added to an ethereal solution of ammonia while cooling with ice salt, with further ammonia gas being introduced.
After working up as in Example 1, 30 g of carbamic acid ester of 3-methylbutyn- (1) -oIs- (3), boiling point 12 111¯, melting point 108 (from cyclohexane), are obtained.
Example 3
A solution of 34 em3 (500 mmol) liquid.
Phosgene in 150 cm3 abs. ¯ther is, as in Example 2, first with 62 g (500 mmol) 1 ethynyl-eyelohexanol- (l) and 64 cm. (500 mmol) quinoline and then, after further processing according to Examples 1 and 2, reacted with ammonia in ether. 45 g (53% of theory) of carbamic acid ester of 1-thynyl-cyclohexan-ol- (1) are obtained; Bp 3 108-110¯; colorless needles made of cyclohexane, F. 94-96 ".
Example 4
A solution of 34 cm3 (500 mmol) liquid.
Phosgene in 150 cm3 abs. As in Example 2, it is reacted with 49 g (500 mmol) of 3-methylpentyn- (1) -ol- (3) and 64 cubic meters of quinoline and, after the quinoline hydrochloride has been separated off, at -10 in a solution of about 40 g methylamine in 100 em3 abs. Ether dripped in.
The mixture is allowed to come to room temperature in the course of a few hours, it is then washed alkaline and acidic and finally, by vacuum distillation, the N-methylcarbamic acid ester of methylpentynol is obtained in 45% yield, bp 53-54, colorless hexane needles.
Example. 5
To a solution of 62 g of 1-thynyl-cyclo hexanol- (1) in 200 cm3 of abs. Toluene and 50 to 52 g of phosgene are added dropwise with stirring in about 11/2 hours. 61 g of collidine in 70 cm3 of toluene, the temperature being kept at +5 to +10¯ by cooling with ice.
After stirring for 1-2 hours, let it stand at about +10¯ overnight. The precipitated collidine hydrochloride is filtered off with suction and washed with toluene, and the filtrates are combined. A steady stream of NH3 is passed through 250 cubic meters of toluene and the above toluene filtrate is added slowly with ice cooling so that the temperature does not exceed + 10. The mixture is then stirred for a further 15 minutes and then worked up as in the previous examples. The dried toluene solution of the carbamic acid ester is cooled to -5 to 0, the majority of which crystallizes. After suction, the filtrate can be concentrated and made up again for crystallization or subjected to vacuum distillation.
Total yield 50.8 g -60.7% of theory.
Example 6
6, 2 g Äthinyleyelohexanol are in 100 cm3 abs. Dissolved pyridine, mixed with 5.5 cm 3 of phenyl isoeyanate, prepared from phosgene and aniline, and refluxed for 17 hours. Working up via ether gives, in addition to diphenylurea, 4.2 g of N-phenylcarbamic acid 1-ethinylcyelohexyl ester crystals from hexane. F. 94-96¯.
Example 7
To 23 g (0.3 mol) of carbamic acid chloride (made from phosgene and ammonia) in 150 cm3 of dry ether, 32 g of 1-propynyl-cyelohexanol- (1) in 150 cm3 of dry ether are added dropwise with stirring at about 0; After standing overnight, the mixture is worked up with water, the solution is dried, concentrated and deep-frozen, 10 g (25% of theory) of colorless crystals having a melting point of 137 to 139 being obtained, which represent the carbamic acid ester of propynyl-eyelohexanol.
Phenate is formed analogously. hinyleyclohexanol the corresponding carbamic acid ester, F. 110-111¯, crystals from hexane.
Example 8
28 g of diethyl carbamic acid chloride (made from phosgene and diethylamine), 31 g of 1-thynyl-cyclohexanol- (1) and 31 g of 2, 4, 6 collidine are dissolved in 150 cm3 of toluene and refluxed for 5 hours. The precipitated collidine hydrochloride is separated off, the toluene solution is washed with dilute acid and alkali and water, dried and concentrated and distilled in vacuo. Main fraction Kp2 94-96, F. 30 to 33¯. Yield 15 g (27% of theory) diethylcarbamic acid ester of Äthinyleyelo- hexanols.
Example 9
A cooled solution of 49.6 g of 1-ethynyleyelohexanol- (1) (1 mol) in 164 cm3 of toluene (dried over sodium) is mixed with a likewise cooled solution of 51.58 g of phosgene (1, 3 mol) combined in 157.6 cm3 toluene. To this end, a solution of 27.2 g of trimethylamine (1.15 mol) in 180 cm3 of toluene is added from a dropping funnel with stirring at + 20 to + 40 in the course of 60-70 minutes. After stirring for 1/2 hour at about 0, the precipitated trimethylamine hydrochloride is sucked off on the suction filter and washed with four times as much toluene that the salt is just covered with solvents after each work through.
The salt is dried at 60 in the drying cabinet. 41.45 g, corresponding to 94.5% of the trimethylamine used, are obtained. The filtrate and washing toluene are returned to the flask and amidated by bubbling in ammonia gas at about +3. After the amidation has ended (duration approx. 11/2 hours) the reaction mixture is placed in a separating funnel with 100 cm3 of water and diluted with 2 liters of ether. The ammonium chloride solution is drained off and the ether layer is washed successively with water, several times with 10% sulfuric acid and then again with water. This washing water is aethered out together with the ammonium chloride solution.
The sulfuric acid wash water is also shaken out several times with ether and this is combined with the main amount. After drying over sodium sulfate, the entire ethereal solution is evaporated (finally in a vacuum). As soon as crystallization begins, 100 cm3 of gasoline (50 to 60) are added and allowed to stand at 0 for several hours to complete the crystallization. The groove is vacuumed and washed several times with gasoline. After drying at 50, 54.45 g of ethynyl cyclohexyl urethane with a melting point of 93-95 are obtained, corresponding to 81.5% of theory.
After evaporation and distillation in vacuo, some starting material is recovered from the gasoline liquors.
Recovery of the trimethylamine:
In a distillation flask, a 50% sodium hydroxide solution with 45 g of NaOH is added from a dropping funnel to 95 g (1 mol) of the trimethylamine hydrochloride obtained, the developed trimethylamine being collected in a deeply cooled receiver.
By heating the reaction mixture to about 80, the last portions of trimethylamine are driven over. In this way 58 g of already largely dry trimethylamine are obtained, which is best dried completely after dilution with toluene with caustic potash and barium oxide. The yield is 98 to 99% of theory. The toluene solution can be used directly for further reactions.
Example 10
A solution of 70 g of butyn- (1) -ol- (3) and 99 g of phosgene in 400 cm3 of abs. At about -10, 129 g of quinoline in absolute ether were added to ether in 4 hours while stirring. After standing at -6 for a long time, the quinoline hydrochloride was sucked off and the ethereal solution was added dropwise to ether at -5 while stirring and passing ammonia through. After the reaction had ended, it was worked up in the usual manner and the butyne- (1) -ol- (3) -urethane was purified by distillation. Bp 12 108-109¯, m.p. 48-50 ", crystals from hexane. Yield 50 g = 49% of theory.
Example 11
A solution of 50 g of ethynyl-eyelohexenyl-carbinol and 40 g of phosgene in 200 cm3 of toluene is slowly mixed with 56 em3 of 2, 4, 6-collidine in 50 em3 of toluene at + 10¯ with stirring, then stirred for a further 2 hours and then further processed as in example 1. After concentrating the solution, 25.8 g (40% of theory) of the corresponding carbamic acid ester are obtained; after 2 recrystallization from ether F. 117-118.
Example 12
To 25 g of carbamic acid chloride (made from phosgene and ammonia) in 50 cubic meters of dry ether, 39 g (endomethyleneyclohexenyl) -ethinyl-carbinol in 100 cm3 of dry ether are added dropwise while stirring; After standing overnight, it is worked up with water, the solution is strongly concentrated and deep-frozen. 28 g = 53% of theory. F. 114-116.
