Verfahren zur Herstellung von neuen Dicarbamaten aliphatischer 1. 3-Diole
Das Dicarbamat des 2-Methyl-2-propylpropan-1, 3-diols ist ein bekannter Vertreter einer Gruppe von pharmazeutisch wirksamen Verbindungen, die in der Literatur als "Tranquilizer" bezeichnet werden. Die Wirkung solcher "Tranquilizer" setzt sich aus einer sedativen und einer hypnotischen Komponente zusammen. Bei einem echten "Tranquilizer" muss die hypnotische Wirkung soweit wie möglich zugunsten der sedativen Wirkung zurückgedrängt werden. Bei dem obigen Dicarbamat spielt die hypnotische Wirkungskomponente noch eine beachtliche Rolle, wie Beispiele in der medizinischen Literatur zeigen, wo Personen unter Einfluss dieses Mittels unüberlegte und unkontrollierte Handlungen begingen.
Die genannte Verbindung ist das Dicarbamat zweier primärer Alkoholgruppen.
Es wurde nun gefunden, dass Dicarbamate von Diolen ähnlicher Konstitution, in denen jedoch die OH-Gruppen sekundärer Natur sind, unter Beibehaltung der sedativen Wirkung einen bedeutend geringeren hypnotischen Effekt aufweisen und besonders verträglich sind. Ihre Giftigkeit beträgt etwa 1/8 der des

Die Verbindungen sind in Wasser verhältnismässig schwer löslich. Sie können erfindungsgemäss aus den Diolen nach folgenden Methoden hergestellt werden. Man kann zuerst die Diole mit Phosgen in Gegenwart einer Base in einem indifferenten organischen Lösungsmittel zu Dichlorkohlensäureestern umsetzen und diese dann mit Ammoniak in Dicarbamate verwandeln. Als organische Lösungsmittel kommen Benzol, Toluol, Chloroform, Äthylenchlorid, Aceton und andere in Frage, als Basen z. B. Dimethylanilin, Pyridin, Trimethylamin, Natronlauge, Natriumbicarbonat u. dgl. Weiter kann man die Diole mit Harnstoffchlorid in einem indifferenten organischen Lösungsmittel gegebenenfalls in Gegenwart einer Base zum Dicarbamat umsetzen. Man kann die Diole auch der Einwirkung von Cyansäure in einem indifferenten organischen Lösungsmittel unterwerfen.
Durch Verschmelzen der Diole mit Harnstoff oder Harnstoffsalzen erhält man durch Ammoniakabspaltung bzw. Ammonsalzbildung ebenfalls die Dicarbamate. Es ist auch möglich, die Diole mit Alkylurethanen in Gegenwart von Katalysatoren, wie z. B. Aluminiumisopropylat, bei höheren Temperaturen umzuestern.
Beispiel 1 : 23, 6 g (0, 2 Mol) 3-Methyl-pentandiol-2, 4 werden zusammen mit 48, 4 g Dimethyl- anilin in 200 cm3 trockenem Benzol gelöst und bei 5-10 C in eine Lösung von 39, 6 g Phosgen in 200 cm3

trockenem Benzol eingerührt. Es wird noch eine Stunde bei Zimmertemperatur und dann weitere vier Stunden bei 30 - 400C gerührt. Dann wird mit Eiswasser und etwas Salzsäure und schliesslich mit reinem Wasser im Scheidetrichter gewaschen. Die Benzolschicht trocknet man mit Natriumsulfat und leitet in die filtrierte Lösung einige Stunden Ammoniak ein. Der Kristallbrei wird abgesaugt und mit Wasser gewaschen.
Der Rückstand wird zusammen mit der weiteren Fraktion, die man aus der Benzollösung nach dem Waschen mit Wasser, Konzentrieren und Ausfällen mit Ligroin, erhält, aus Methylenchlorid-Äther umkristallisiert. Weisse Kristalle des Dicarbamats vom Fp. 183 C.
In die Lösung von 10 g 3-Methylpentandiol-2, 4 in 100 cm3 Benzol wurde bei Zimmertemperatur 2 Stunden lang gasförmige Cyansäure eingeleitet und noch etwa 5 Stunden gerührt. Der weisse Nieder schlag wurde abgesaugt und mit Wasser ausgekocht. Hiebei geht das Dicarbamat in Lösung. Von rückgebildeter Cyanursäure wurde heiss abgesaugt. Beim Erhalten kristallisierte aus dem Wasser das Dicarbamat vom Fp. 1830C aus.
10 g 3-Methyl-pentandiol-2, 4, 10 g Harnstoff, 2 g Zinkacetat wurden 15 Stunden in 100 cms Toluol am Rückflusskühler gekocht. Das Roluol wurde i. V. abdestilliert und der Rückstand mit heissem Wasser behandelt, filtriert und die wässerige Lösung konzentriert. Beim Erkalten fällt das Dicarbamat vom Fp. 1830C aus.
23,6 g 3-Methylpentandiol-2, 4,38 g Äthylurethan, 4 g Aluminiumisopropylat und 250 cm Toluol wurden an einer Kolonne so erhitzt, dass das Toluol-Alkoholgemisch innerhalb von etwa 15 Stunden abdestillierte. Der Rückstand wurde in 805obigem Isopropylalkohol aufgenommen, mit Kohle filtriert, das Lösungsmittel i. V. abdestilliert. Aus dem Rückstand wurde aus heissem Wasser das Dicarbamat vom Fp. 1830C erhalten.
23, 6 g des Diols werden mit 48,4 g Dimethylanilin in 200 cm trockenem Chloroform gelöst. Zu dieser Lösung tropft man unter Rühren bei 00C 32 g Carbaminsäurechlorid mit 50 cm Chloroform verdünnt zu. Nach mehrstündigem Rühren bei Zimmertemperatur wird mit Eiswasser-Salzsäure und dann mit Wasser gewaschen und mit ChlorcaIcium getrocknet. Nach dem Einengen der Chloroformlösung wird das Dicarbamat mit Ligroin ausgefällt und aus Methylenchlorid-Äther umkristallisiert.
Beispiel 2 : 29, 2 g (0, 2 Mol) 3-Methyl-heptandiol-2, 4 werden mit 48, 4 g Dimethylanilin in 200 cm trockenem Aceton gelöst. Unter Rühren bei 0 C werden zu dieser Lösung 32 g Carbaminsäure- chlorid zugetropft.
Man rührt noch drei Stunden bei Zimmertemperatur und giesst dann unter Rühren in ein Liter Eiswasser und 1/2 Liter Methylenchlorid. Nach Abtrennen im Scheidetrichter wird noch einmal mit Methylenchlorid ausgezogen. Die vereinigten Auszüge Methylenchlorid werden mit verdünnter Salzsäure und dann mit Wasser gewaschen. Nach Trocknen mit Chlorcalcium und Einengen der Methylenchloridlösung wird bei Zugabe von Äther das Dicarbamat des 3-Methyl-heptandiols-2, 4 in weissen Kristallen vom Fp. 1760C erhalten.

Process for the preparation of new dicarbamates of aliphatic 1,3-diols
The dicarbamate of 2-methyl-2-propylpropane-1,3-diol is a known representative of a group of pharmaceutically active compounds which are referred to in the literature as "tranquilizers". The effect of such "tranquilizers" consists of a sedative and a hypnotic component. In a real "tranquilizer", the hypnotic effect must be suppressed as far as possible in favor of the sedative effect. In the case of the above dicarbamate, the hypnotic component of the effect still plays a considerable role, as examples in the medical literature show where people committed rash and uncontrolled acts under the influence of this agent.
The compound mentioned is the dicarbamate of two primary alcohol groups.

The compounds are relatively sparingly soluble in water. According to the invention, they can be prepared from the diols by the following methods. The diols can first be reacted with phosgene in the presence of a base in an inert organic solvent to give dichlorocarbonic acid esters and these can then be converted into dicarbamates with ammonia. As organic solvents benzene, toluene, chloroform, ethylene chloride, acetone and others come into question, as bases z. B. dimethylaniline, pyridine, trimethylamine, caustic soda, sodium bicarbonate u. The like. The diols can also be reacted with urea chloride in an inert organic solvent, if appropriate in the presence of a base, to give the dicarbamate. The diols can also be subjected to the action of cyanic acid in an inert organic solvent.
By fusing the diols with urea or urea salts, the dicarbamates are likewise obtained through the elimination of ammonia or ammonium salt formation. It is also possible to use the diols with alkyl urethanes in the presence of catalysts, such as. B. aluminum isopropoxide, transesterify at higher temperatures.

The benzene layer is dried with sodium sulfate and ammonia is passed into the filtered solution for a few hours. The crystal slurry is filtered off with suction and washed with water.
The residue is recrystallized from methylene chloride ether together with the further fraction obtained from the benzene solution after washing with water, concentration and precipitation with ligroin. White crystals of dicarbamate from mp. 183 C.
Gaseous cyanic acid was passed into the solution of 10 g of 3-methylpentanediol-2,4 in 100 cm3 of benzene at room temperature for 2 hours and stirring was continued for about 5 hours. The white precipitate was sucked off and boiled with water. The dicarbamate goes into solution. The cyanuric acid which had formed was sucked off hot. When obtained, the dicarbamate crystallized from the water with a melting point of 1830C.
10 g of 3-methylpentanediol-2, 4, 10 g of urea and 2 g of zinc acetate were refluxed in 100 cms of toluene for 15 hours. The Roluol was i. V. distilled off and the residue treated with hot water, filtered and the aqueous solution concentrated. On cooling, the dicarbamate precipitates with a melting point of 1830C.
23.6 g of 3-methylpentanediol-2, 4.38 g of ethyl urethane, 4 g of aluminum isopropylate and 250 cm of toluene were heated in a column in such a way that the toluene-alcohol mixture distilled off within about 15 hours. The residue was taken up in isopropyl alcohol above, filtered with charcoal, the solvent i. V. distilled off. The dicarbamate with a melting point of 1830C was obtained from the residue from hot water.
23.6 g of the diol are dissolved with 48.4 g of dimethylaniline in 200 cm of dry chloroform. 32 g of carbamic acid chloride, diluted with 50 cm of chloroform, are added dropwise to this solution while stirring at 00C. After several hours of stirring at room temperature, it is washed with ice-water-hydrochloric acid and then with water and dried with chlorocarbon. After the chloroform solution has been concentrated, the dicarbamate is precipitated with ligroin and recrystallized from methylene chloride-ether.
Example 2: 29.2 g (0.2 mol) of 3-methyl-heptanediol-2,4 are dissolved with 48.4 g of dimethylaniline in 200 cm of dry acetone. 32 g of carbamic acid chloride are added dropwise to this solution with stirring at 0 C.
The mixture is stirred for a further three hours at room temperature and then poured into one liter of ice water and 1/2 liter of methylene chloride with stirring. After separation in a separating funnel, it is extracted again with methylene chloride. The combined extracts of methylene chloride are washed with dilute hydrochloric acid and then with water. After drying with calcium chloride and concentration of the methylene chloride solution, the dicarbamate of 3-methyl-heptanediol-2,4 is obtained in white crystals with a melting point of 1760C upon addition of ether.
