Die neue Verbindung wird erhalten, wenn man 2-Phenyl-pyridazon-(3), das in 6-Stellung einen austauschfähigen Substituenten auf weist, mit Ammoniak umsetzt.
Ein austausch fähiger Substituent ist zum Beispiel ein Ha logen-, insbesondere ein Bromatom, oder auch eine substituierte Oxy- wie Pheno-xy-, oder Mercapto- wie 112ethylmercaptogruppe.
Die Reaktion kann in An- oder Abwesen heit von Verdünnungsmitteln und/oder Kon densationsmitteln und/oder Katalysatoren im offenen oder geschlossenen Gefäss unter Druck durchgeführt werden. So kann man zum Beispiel 2-Phenyl-6-halogen-pyridazon- (3), insbesondere 2-Phenyl-6-brom-pyridazon- (3), in Gegenwart von Kupfer in einem Auto klav bei höherer Temperatur mit einer wäs serigen Ammoniaklösung umsetzen.
Das Verfahrensprodukt, das 6-Amino-2- phenyl-pyridazon-(3) vom F. = 149-l.51 , ist neu. Es zeigt eine antipyretische Wirkung und soll als Heilmittel oder als Zwischenpro dukt Verwendung finden.
Die Erfindung wird im nachfolgenden Bei spiel näher beschrieben. Zwischen Gewichtsteil und Volumteil besteht die gleiche Beziehung wie zwischen Gramm und Kubikzentimeter. <I>Beispiel</I> 50 Gewichtsteile 2 - Phenyl - 6 - brom-pyrid- azon-(3) werden in einem Autoklav mit 250 Volumteilen einer konz. wässerigen Ammo- niaklösumg, 250 Voltunteilen Wasser und 1 Gewichtsteil Kupferbronze 10 Stunden auf 155-165 erhitzt.
Die Kupferbronze wird ge trennt und die Lösung mehrmals mit Chloro form ausgeschüttelt. Die vereinigten Chloro- formextrakte werden eingedampft und der Rückstand mit Äther verrieben. Das so erhal tene 6-Amino-2-phenyl-pyridazon-(3) schmilzt bei 149-151 . Durch Behandeln mit äthyl.- alkoholischer Salzsäure kann man die freie Base in das Hydrochlorid vom F. = 170 bis 171 verwandeln.
Das als Ausgangsstoff verwendete 2-Phe- nyl-6-brom-pyrida.zon-(3) kann wie folgt her gestellt werden: 5 Gewichtsteile 2-Phenyl-6-oxy-pyridazon- (3) werden mit 12 Gewichtsteilen Phosphor- pentabromid vermischt und im Ölbad auf. 125 bis 130 erhitzt. Das Reaktionsgemisch wird auf 50 Volumteile Wasser gegossen, wobei sich der #berschuss des Phosphorpentabromids zer- setzt.
Nach 15stündigem Stehen wird ge- nutseht und das erhaltene graubraune Pro dukt mit. Wasser gewaschen und im Vakuum getrocknet. Zur Reinigung wird es in Benzol heiss gelöst. Man versetzt die Lösung mit. Pe- troläther, bis ein flockiges Nebenprodukt aus fällt, und bringt aus der filtrierten und mit Tierkohle gereinigten Benzol-Petrolä.ther- lösung durch weitere Zugabe von Petroläther das 2-Phenyl-6-brom-pyridazon-(3) zur Kri stallisation. Es schmilzt bei 122-124 .
