Verfahren zur Herstellung eines neuen Esters
Gegenstand der vorliegenden Erfindung ist ein Verfahren zur Herstellung eines neuen lokalanästhetisch wirksamen Esters der p-n Butylaminobenzoesäure mit Nonaäthylenglykolmonomethyläther der Formel

und seiner Salze.
Der neue Ester zeigt wertvolle pharmako- logische Eigenschaften, So besitzt er einen ausgeprägten lokalanästhetischen Effekt, insbesondere wirkt er selektiv auf die Dehnungsrezeptoren der Lunge. Er soll als Heilmittel, vorzugsweise als Hustenmittel, verwendet wer.den. Bezüglich dieser Verwendung weist die neue Verbindung wesentliche Vorteile auf gegenüber bekannten, vergleichbaren Estern.
Das erfindungsgemässe Verfahren ist dadurch gekennzeichnet, dass man den Ester aus p-AminobenzoesÏure und NonaÏthylenglykol monomethyläther mit die n-Butylgruppe einführenden Alkylierungsmitteln behandelt. Solche Mittel sind zum Beispiel reaktionsfähige Ester des n-Butanols oder n-Butyraldehyds in Gegenwart von Reduktionsmitteln, wie Zink- staub.
Die Reaktionen können in An-oder Ab wesenheit von Verdünnungs-und bzw. oder Kondensationsmitteln oder Katalysatoren bei gewöhnlicher oder erhöhter Temperatur in offenen oder geschlossenen Gefϯen unter Druck durchgeführt werden.
Je nach der Arbeitsweise erhält man den neuen Ester als Base oder als Salze. Aus den Salzen kann in an sich bekannter Weise die freie Base hergestellt werden, von welcher sieh durch Umsetzung mit SÏuren, die zur Herstellung von therapeutisch verwendbaren Salzen geeignet sind, die Salze gewinnen lassen.
Bemerkenswert ist, dass der neue Ester eine beträchtliche Löslichkeit in Wasser aufweist. Eine solche Losung besitzt das Charakteristikum, sich beim Erwärmen bei einer be stimmten, von der Konzentration abhängigen Temperatur zu trüben. Dieser an einer 10 /oigen Losung bestimmte Temperaturwert wird als Trübungspunkt bezeichnet und stellt eine physikalische Konstante dar. Er liegt bei 45 . Die Tr bung ist reversibel ;'beim Abkühlen klÏrt sich die Losung wieder auf.
Die als Ausgangsstoffe genannten Verbin dungen sind bekannt oder lassen sich nach an sieh bekannten Methoden herstellen.
Die Erfindung wird im nachstehenden Beispiel beschrieben. Zwischen Gewichtsteil und Volumteil besteht die gleiche Beziehung wie zwischen Gramm und Kubikzentimeter.

Beispiel
3, 5 Gewichtsteile p-Aminobenzoesäureester des Nonaäthylcnglykol-monomethyläthers werden in 20 Volumteilen Benzol gnelöst und mit 1, 65 Gewichtsteilen Zinkstaub versetzt. Die lZischung wird zum R ckflu¯ erhitzt und mit 1,5 Gewichtsteilen Eisessig versetzt, worauf man unmittelbar eine Losung von 0, 71 Vo lumteilen Butyraldehyd in 5 Volumteilen Benzol zutropfen, lässt. Nach 25 Minuten ist die Zugabe beendet. Man erhitzt noch 3 Stunden am R ckflu¯, filtriert heiss und wäscht mit Benzol nach. Das Filtrat wird unter Küh llmg mit 50 Volumteilen Wasser und 10 Vo lumteilen konz.
Ammoniak versetzt und die wässrige Schiclit abgetrennt. Die benzolische Eösung wird hierauf mit Natriumearbonat losung und dann mit Wasser gewaschen, wobei die wässrigen Anteile nochmals mit Benzol ausgezogen werden. Aus den vereinigten ben zolischen Losungen erhält, man durch Ein- dampfen den Ester von p-n-Butylaminobenzoesäure mit NonaÏthylenglykol-monomethyl Ïther der Formel

Der als Ausgangsstoff verwendete Este@ der p-Aminobenzoesäure mit Nonaäthylengly kol-monomethylÏther kann wie folgt herge stellt werden:
8, 6 Gewichtsteile Nonaäthylenglykol-mono methyläther werden mit 20 Volumteilen ab b, solutem Pyridin gemischt und mit 2, 2 (Te- wichtsteilen p-Nitrobenzoylchlorid versetzt. E; entsteht unter Erwärmung eine lare, hell gelbe Lösung.
Man erwärmt während 20 Mi nuten unter Ausschluss von Feuchtigkeit au ; dem Dampfbad und lässt dannn während 2 bis 3 Stunden bei Raumtemperatur stehen Dann verdünnt man mit 100 Volumteilei Benzol und wäscht mehrmals mit Salzsäure dann mit AVasser, wobei alle wässrigen Teille mit 50 Volumteilen, frischem Benzol nach gewaschen werden.
Aus den vereinigten und getrocknetei Benzolsehichten erhält man-durch Eindamp fen 5, 5 Gewichtsteile des p-Nitrobenzoesäure esters von Nbnaäthylengiykol-monomethyl äther in Form eines Öls vom Breehungsinde Nd22 = 1,4960 und vom Tr bungspunkt 63 (10%ige wϯrige L¯sung).
5, 4 Gewichtsteile dieses Rüekstandes aver- den in 50 Volnmteilen Äthanol gelost und ir Gegenwart von 0, 5 Gewichtsteilen methanoli scher Raneynickelaufschlemmung bei 20 atü H2 hydriert. Die Wasserstoffaufnahme ist schon nach etwa 1/2 Stunde fast beendet. Man lässt noch während 21/2 Stunden bei Raumtemperatur und 20 at H1 weiterrühren und filtriert dann den Katalysator ab. Beim Ein- dampfen des Filtrates erhält man 5, 03 Gewiehtsteile p-Aminobenzoesäureester von Nona äthylenglykol-monomethyläther in Form eines hellgelben, dickfl ssigen Ols.

The new ester shows valuable pharmacological properties. It has a pronounced local anesthetic effect, in particular it acts selectively on the stretch receptors in the lungs. It should be used as a remedy, preferably as a cough suppressant. With regard to this use, the new compound has significant advantages over known, comparable esters.
The process according to the invention is characterized in that the ester of p-aminobenzoic acid and non-ethylene glycol monomethyl ether is treated with alkylating agents which introduce the n-butyl group. Such agents are, for example, reactive esters of n-butanol or n-butyraldehyde in the presence of reducing agents such as zinc dust.
The reactions can be carried out in the presence or absence of diluents and / or condensation agents or catalysts at normal or elevated temperature in open or closed vessels under pressure.
Depending on the procedure, the new ester is obtained as a base or as a salt. The free base can be prepared from the salts in a manner known per se, from which the salts can be obtained by reaction with acids which are suitable for the preparation of therapeutically useful salts.
It is noteworthy that the new ester has considerable solubility in water. Such a solution has the characteristic of becoming cloudy when heated at a certain temperature that depends on the concentration. This temperature value, determined on a 10% solution, is called the cloud point and represents a physical constant. It is 45. The turbidity is reversible; the solution clears up again on cooling.
The compounds mentioned as starting materials are known or can be prepared by methods known per se.
The invention is described in the following example. The relationship between part by weight and part by volume is the same as that between grams and cubic centimeters.

3.5 parts by weight of p-aminobenzoic acid ester of Nonaäthylcnglykol-monomethyläthers are dissolved in 20 parts by volume of benzene and mixed with 1.65 parts by weight of zinc dust. The mixture is heated to reflux and mixed with 1.5 parts by weight of glacial acetic acid, whereupon a solution of 0.71 parts by volume of butyraldehyde in 5 parts by volume of benzene is immediately added dropwise. The addition is complete after 25 minutes. The mixture is refluxed for a further 3 hours, filtered hot and washed with benzene. The filtrate is concentrated while cooling with 50 parts by volume of water and 10 parts by volume.
Ammonia is added and the aqueous schiclite is separated off. The benzene solution is then solution with sodium carbonate and then washed with water, the aqueous components being extracted again with benzene. The mixture is heated for 20 minutes with exclusion of moisture; the steam bath and then left to stand for 2 to 3 hours at room temperature. Then it is diluted with 100 parts by volume of benzene and washed several times with hydrochloric acid then with water, all aqueous parts are washed with 50 parts by volume of fresh benzene.

5.4 parts by weight of this residue were dissolved in 50 parts by volume of ethanol and hydrogenated in the presence of 0.5 parts by weight of methanolic Raney nickel slurry at 20 atm.
