l'acide perphtha- l'ique, etc.
La 2-diehloroacétylamino-4-mé- thylmercaptoacétophénone est traitée avec de la formaldéhyde en présence de bicarbonate de sodium ou d'un catalyseur de condensation alcalin équivalent;

<I>Exemple 1:</I> On dissout 7 g de y-2-dichloroacétylamino- 1-(4-méthylmercaptophényl) -1,3-propanediol (1) dans 30 mil d'acétone.
Process for the preparation of an N-acylated aromatic aminodiol with antibiotic power. The present invention relates to the preparation of novel N-acylated aromatic aminodiols, corresponding to the formula:
EMI0001.0008
where R. is a hydrocarbon radical having 1 to 7 carbon atoms and Y an aliphatic acylamino radical. R may be an aliphatic, eycloaliphatic, aryl (benzyl) radical having from 1 to 7 carbon atoms.
The acylamino radical Y may or may not be saturated, the acyl group deriving from an aliphatic carboxylic acid.
It will be noted that these compounds can exist in the form of structural isomers (or diastereomers) and in the form of optical isomers. The term structural isomer used herein refers to the relative spatial configuration of the groups attached to the two asymmetric carbon atoms.
The compounds before. the same spatial configuration as threose and pseudo-ephedrine will henceforth be designated as pseudo (zp), or threo or notches, and those having the same spatial configuration as erythrosis and ephedrine will be considered regular (reg.), or erythro or cis.
Each of the two cis and trans forms still exists as optical isomeric silks, i.e. as a racemate and as optically active dextro gyre (d ') and levorotatory (1) isomers.
Due to the difficulty of representing these differences in structure in graphic formulas, the usual structural formulas are conventionally used by carrying, below or next to each formula, a notation indicating the structure and configuration: particular optics of the compound.
The N-acylated aminodiols of the above formula are useful antibiotic agents. They have, for example, a bactericidal and bacteriostatic action on gram-positive and gram-negative bacteria and ricliettsial organisms. This action is.
particularly marked for the racemic and dextrorotatory forms, the dextrorotatory form being moreover generally more active than the racemic form; in many cases, in fact, the dextro gyre form has almost double the antibiotic activity of the racemic form.
The novel compounds involved are also useful as intermediates in the preparation of other valuable organic substances. Thus, for example, they can be hydrolyzed with strong mineral acids to produce the corresponding amino derivatives.
The present patent relates to a process for preparing one of these compounds, namely V-2-diehloroac6tyT-amino-l- (4-methyl-suHonylphéiiyl) -1,3-propanediol. This process is characterized by oxidizing y-2-dichloroacetylamino-l- (4-methyl-mercaptophenyl) -1,3-propanediol by treating it with -an peracid to produce yi-2-d ehloroacetylamino-l - (4-methylsulfonylphenyl) - 1,3-propanediol. This product is a new substance, crystallized.
Its racemic form melts at 179 9 to 180 6 C and its background form at 165 3 to 166 6 C. The optical forms d1 and d have antibiotic activity.
The reaction that takes place in this process can be represented by the following equation:

<tb> 0 Peracids which can be used to effect the oxidation include, for example, Caro's acid (H2S05), peracetic acid, perbenzoic acid,
perphthaic acid, etc. In general, it is preferable to employ as the oxidizing agent an organic peracid, for example peracetic acid or perbenzoic acid. It is advantageous to carry out the oxidation in the presence of an organic solvent which is neutral to the material. with peracid used as an oxidant.
The oxidation is most advantageously carried out at a temperature between 10 and 100 ° C., depending on the choice of the oxidant, but for optimum yield, it is preferable, if an organic peracid is used, not to leave the reaction temperature exceed 65 C.
The y) - 2 - dichloroacetylamino-1- (4-methyl-mercaptophenyl) -1,3 - propanediol used as a product. starting can. be obtained as follows:
2-Diehloroacetylamino-4-methylmercaptoacetophenone is treated with formaldehyde in the presence of sodium bicarbonate or an equivalent alkaline condensation catalyst;
thus obtaining 2-di chloroacetylamino-, 8.hydroxy-4-methylmercaptopropiophenone; this derivative is reduced by treatment with an oxidizable lower aluminum alkoxide; the desired W-2-dichloroacetyl-amino -1- (4-methylmer captophenyl) -1, 3 - pr o-panediol is obtained.

<tb> 0 It will be noted that if one uses as starting product, in the process according to the invention,
the racemic form of y-2-dichlor acetylamino-1- (4-methylmercaptophenyl) -1,3-propanedio1, will be obtained naturally. the racemic form of y-2-dichloroacetylamino-1- (4-methylsulfonylphenyl) -1,3-propanediol. On the other hand, if an optically active form of the starting material is employed, the end product will also be optically active. Optically active forms of the starting material are not very windy. easily be obtained as follows:
The racemic form of the compound is deacylized by treating it with a hot mineral acid to obtain the racemic form of y-2-amino-1- (4-methylmercaptophenyl) -1,3-propanediol which is dissolved again in the. using an optically active organic acid. The optically active forms of the amines thus obtained are then treated with an aliphatic acylating agent to obtain the desired optically active forms of the compound. The following examples illustrate the invention.
<I> Example 1: </I> 7 g of y-2-dichloroacetylamino- 1- (4-methylmercaptophenyl) -1,3-propanediol (1) are dissolved in 30 mil of acetone. To this solution is added dropwise, with stirring, 10 ml of 40% peracetic acid. The temperature is maintained between 39 and 45 C during the reaction by cooling the vessel.
The mixture was then stirred for two hours, then diluted with 100 ml of water, and the resulting solution was left for about two days in a refrigerator. The precipitate separating from the. solution is collected in a filter, washed several times with. ice water, and dried overnight at 70 C.
In this way 6.2 g of levorotatory yr2-diehloroacetylamino-l- (4-methyl-sidfo-nylphenyl) -1,3-propaEediol is obtained which melts at 1640 3 to 166 <B> 0 </B> 3 C and has a [a] D of -120 (1% solution in 95 / a ethanol).
Dextrorotatory 7p-2-dichloroacetylamino-l- (4-methyl-sitlfonylphenyl) -1,3-propanediol is analogously prepared by oxidizing y-2-dichloroacetylamino-1- (4-methylmercaptophenyl) -1.3 -propanediol dextrorotatory. The product melts at 165 2 to 166 6 C and has an [a] D = 13 (1 / o solution in 95/0 ethanol).
<I> Example </I> 2: 21 ml of 40% peracetic acid are added to a stirred solution of 15.0 g of y-2 dichloroacetylamino-1- (4-methylmercaptophenyl) -1.3- propanediol (dl) dissolved in 55 ml of acetone.
The container is cooled from the outside with ice, and the flow of peracetic acid is adjusted so that the. The temperature of the reaction mixture does not exceed 40 ° C. The reaction mixture is then stirred for a further hour and cooled to -5 ° C. The precipitate which forms is collected in a filter. In this way 14.0 g of a white precipitate is obtained which is due to γ-2-diehloroacetyl amino-1- (4-methylsulfonylphenyl) -1,3-propanediol (dl) melting. to 179 4 to 180 6 C.
