Verfahren zur Herstellung eines optisch aktiven 3-Oxy-N-methyl-morphinans. Bisher sind optisch aktive 3-Oxy-N-methyl- morphinane dadurch erhalten worden, dass man racemisches 3-Oxy-N-methyl-morphinan mittels Weinsäure in die optischen Antipoden zerlegte (0. Schnider und A. Grüssner, Helve- tiea Chimica Acta, Band 34 [1951], Seite 2211).
Nach diesem Verfahren wird das rechtsdrehende 3-Oxy-N-metbyl-morphinan aus der leichter löslichen Tartratfraktion ge wonnen, welche naturgemäss nicht so leicht optisch rein anfällt, wie das schwerer lösliche Tartrat seines Antipoden.
(+)-3-Oxy-N-methyl-morphinan, seine Äther und Ester haben wegen ihrer husten stillenden und antirheumatischen Wirkung Interesse, wogegen der Antipode, das (-)-3- Oxy-N-methyl-morphinan, als Analgeticum Verwendung findet. Die therapeutische Appli kation von (+)-3-Oxy-N-methyl-morplünan bedingt daher einen hohen optischen Rein heitsgrad dieses Antipoden.
Die vorliegende Erfindung betrifft nun ein Verfahren, welches die Herstellung von <B>(+)-</B> und (-) -3-Oxy-N-methyl-morphinan erlaubt, wobei in erster Linie die (+) -Ver bindung in optischer Reinheit gewonnen wer den kann.
Dieses Verfahren besteht darin, dass racemisches 1-(p-Methoxy-benzyl) L3-me- thyl-1,2,3,4,5,6;7,8-octahydro-isochinolin mit Hilfe einer optisch aktiven. Säure, vorzugs weise D-(+)-Weinsäure, in die optischen Antipoden zerlegt, die erhaltenen optisch aktiven 1- (p - Methoxy - benzyl) - 2 - methyl- 1,2,3,4,5,6,7,
8-octahydro-isochinoline zu den entsprechenden optisch aktiven 3-Oxy-N-me- thyl-morphinanen cyclisiert und über die Tartrate isoliert werden.
Zur Spaltung des racemischen 1-(p-Meth oxy-benzyl)-2-methyl-1,2,3,4,5;6,7;8-octahydro- isochinolins (0. Schnider und J. Hellerbaeh, Helvetica Chimca Acta, Band 33 [1950], Seite 1437) wird eine leicht zugängliche optisch aktive ,Säure, vorzugsweise Wein säure, verwendet, wobei die fraktionierte Kri- stallisation aus Wasser oder besser aus Me thanol vorgenommen wird.
Dabei kristallisiert als schwerlösliche Fraktion vorerst das Tar- trat des in Äther linksdrehenden 1-(p-Meth oxy-benzyl)-2-methyl-12,3,4,5,6,7,8-octahydro- isochinolins, welches durch Umkristallisieren vom diastereomeren Tartrat des in Äther rechtsdrehenden 1-(p-Methoxy-benzyl)-2-me- thyl-1,2,3,4,5,6,7,8-octahydro-isochi!nolins ab getrennt werden kann.
Durch Cyclisierung mit Phosphorsäure erhält man daraus das rechtsdrehende 3-Oxy-N-methyl-morphinari, welches mit Vorteil aus dem Reaktionsgemisch isoliert wird. Aus dem Tartrat kann die freie Base in an sich bekannter Weise erhalten werden.
Die methanolische Mutterlauge der Race- matspaltung wird zwecks Gewinnung des in Äther rechtsdrehenden 1-(p-Methoxy-benzyl)- 2-methyl-1,2,3,4,5,6,7,8-octahydro-isochinolins vom Methanol befreit und mehrmals aus Ace ton umkristallisiert. Auf diese Weise ist es möglich, .das Tartrat des in Äther rechts drehenden 1-(p-Methoxy-benzyl)-2-methyl- 1,2,3,4,5,6,7,8-octahydro-isochinolins rein zu gewinnen, woraus .durch Cyclisierung mit der freigesetzten Base das (-)-3-Oxy-N- methyl-morphinan erhalten wird.
Da durch das eben beschriebene mehr malige Umkristallisieren des in Äther rechts drehenden 1- (p-Methoxy-benzyl) -2-methyl- 1,2,3,4,5,6,7,8-oetahydro-isochinolins aus Ace ton Verluste entstehen, kann man bei der Ge winnung von (-)-3-Oxy-N-methyl-morphinan mit Vorteil auch so vorgehen, dass für die Cyclisierung direkt das optisch nicht ganz reine, in Äther rechtsdrehende 1-(p-Methoxy- benzyl) -.2"-methyl-1,2;
3,4,5,i6,7,8-octahydro-iso- chinolin, wie es nach Isolierung des (-)-Anti poden .aus der Racematspaltung resultiert, eingesetzt wird. Das entstehende (-)-3-Oxy- N - methyl - morphinan . liefert nämlich ein schwerer lösliches Tartrat als sein Antipode (vgl. 0. Schnider und A. Grüssner, loc. cit.), welches nun in dieser Stufe durch Kristallisa tion gereinigt werden kann.
Gegenstand des Patentes ist ein Verfah ren zur Herstellung eines .optisch aktiven 3-Oxy-N-methyl-morphinans, das dadurch ge kennzeichnet ist, dass racemisches 1-(p-iMeth- o xy,- 'benzyl) -2-methyl-1,2,lss,4;
56,7, f 8-oetahydro- isochinolin durch intermediäre Salzbildung mit einer optisch aktiven Säure in die opti schen Antipoden zerlegt und das in Äther rechtsdrehende 1- (p-Methoxy-benzyl) -2-me- thyl-1,2,3,4,5,6,7,8-oetahydro-isochinolin zum (-)-3-Oxy-N-methyl-morphinan cyclisiert und über das Tartrat isoliert wird.
<I>Beispiel 1:</I> 75 Gewichtsteile racemisches 1-(p-Methoxy- benzyl) -2-methyl-1,2;3,4,5,6,7,8-oetahydro-iso- ehinolin werden mit 41,5 Gewichtsteilen D-(+)-Weinsäure auf dem Dampfbad in 425 Raumteilen Methanol gelöst. Nach 24stündi- gem Stehenlassen wird abgenutscht, einge engt, weiter stehengelassen und wieder ab- genutscht.
Die methanolische Mutterlauge, welche nach Isolierung :des schwerlöslichere Tartrates des in Äther linksdrehenden 1-(p-Methoxy benzyl);-2-methyl-1,2;3,4;5,6,7;8-octahydro-iso- chinolins zurückbleibt, wird durch Eindämp fen vom Methanol befreit, in Aceton aufge nommen und durch Stehenlassen und Impfen zur Kristallisation gebracht.
Durch Um kristallisieren aus Aceton wird das Tartrat des in Äther rechtsdrehenden 1-(p-Methoxy- benzyl) -2-methyl-1,2;3,4,5,6,7,8-octahydro-iso- chinolins rein erhalten. Schmelzpunkt 130 bis 132 C [a] D = -26 (c = 2 in Methanol).
Das äus diesem Tartrat erhaltene, in Äther rechtsdrehende 1- (p - Methoxy - benzyl) - 2 methyl-1,2,3,4,5,6,7,8-octahydro-isochinolin ist ein Öl mit einem Drehwert von [a] = -77,5 D (c = 3 in Äther).
15 Gewichtsteile dieser Base werden mit 50 Raumteilen 100 higer Phosphorsäure ver setzt und x etwa 65 Stunden bei 130 C sich selbst überlassen. Dann wird mit 50 Raum teilen Wasser versetzt und i/2 Stunde auf dem Dampfbad erwärmt. Nach Verdünnen mit 450 Raumteilen Wasser wird mit Aktivkohle kurz aufgekocht und filtriert. Nachdem mit Ammoniak auf px 9-10 eingestellt worden ist, wird die Rohbase mit Äther extrahiert, durch Verdampfen des Lösungsmittels isoliert und ins Tartrat übergeführt. Dieses wird in wässeriger Lösung mit Kohle kurz aufgekocht, filtriert und zur Kristallisation gebracht.
Das (-)-3-Oxy-N-methyl-morphinan-tartrat kri stallisiert als Dihydrat und schmilzt bei 113 bis 115 C, [a] D = -14 (c = 3 in Wasser). Das aus dem Tartrat hergestellte (-)-3-Oxy- N-methyl-morphinan besitzt einen Drehwert von [.a] = D -56 (c = 1 in Methanol) und schmilzt bei 198-199 C.
<I>Beispiel 2:</I> Die methanolische Mutterlauge, welche nach Isolierung des schwerlöslichere Tartrates des in Äther linksdrehenden 1-(p-Methoxy- benzyl) -2-methyl-1,2,3,4,5,6,7,8-octahydro-iso- ehinolins gemäss Beispiel 1 zurückbleibt, wird durch Eindampfen vom Methanol befreit und ohne weitere Kristallisation aus Aceton daraus durch Fällen mit Ammoniak und Extraktion mit Äther das optisch unreine, in Äther .rechtsdrehende 1-(p-Methoxy-benzyl)- 2-methyl-1,2,3,4,5,6,7,
8- octahydro-isochinolin isoliert. Nach Cyclisierung dieser optisch un reinen Base, gemäss Beispiel 1, wird die erhal tene Rohbase ins Tarträt übergeführt und dieses durch Kristallisation aus Wasser gerei nigt und dadurch vom leichter löslichen Tar- trat des rechtsdrehenden 3-Oxy-N-methyl- morphinans abgetrennt. Das erhaltene Tartrat kristallisiert als Dihydrat, schmilzt bei 113 bis 115 C und besitzt einen Drehwert von [a] =: D -14 (c = 3 in Wasser).
Das daraus gewonnene (-)-3-Oxy-N-methyl-morphinan schmilzt bei 198-200 C und besitzt einen Drehwert von [a] D = -56 (c = 1 in Methanol).
Process for the preparation of an optically active 3-oxy-N-methyl-morphinan. So far, optically active 3-oxy-N-methyl-morphinans have been obtained by breaking down racemic 3-oxy-N-methyl-morphinan into the optical antipodes using tartaric acid (0. Schnider and A. Grüssner, Helvetia Chimica Acta , Volume 34 [1951], page 2211).
According to this process, the dextrorotatory 3-oxy-N-metbyl-morphinan is obtained from the more easily soluble tartrate fraction, which naturally does not appear as easily optically pure as the less soluble tartrate of its antipode.
(+) - 3-Oxy-N-methyl-morphinan, its ethers and esters are of interest because of their anti-cough and anti-rheumatic effects, whereas the antipode, (-) - 3-Oxy-N-methyl-morphinan, is used as an analgesic finds. The therapeutic appli cation of (+) - 3-oxy-N-methyl-morplünan therefore requires a high degree of optical purity of this antipode.
The present invention now relates to a process which allows the production of (+) - and (-) -3-oxy-N-methyl-morphinan, primarily the (+) compound who can be obtained in optical purity.
This method consists in that racemic 1- (p-methoxy-benzyl) L3-methyl-1,2,3,4,5,6; 7,8-octahydro-isoquinoline with the help of an optically active. Acid, preferably D - (+) - tartaric acid, broken down into the optical antipodes, the optically active 1- (p - methoxy - benzyl) - 2 - methyl 1,2,3,4,5,6,7 obtained,
8-octahydro-isoquinolines can be cyclized to the corresponding optically active 3-oxy-N-methyl-morphinanes and isolated via the tartrates.
For the cleavage of the racemic 1- (p-methoxy-benzyl) -2-methyl-1,2,3,4,5; 6,7; 8-octahydroisoquinoline (0. Schnider and J. Hellerbaeh, Helvetica Chimca Acta , Volume 33 [1950], page 1437), an easily accessible optically active acid, preferably tartaric acid, is used, the fractional crystallization being carried out from water or, better, from methanol.
The sparingly soluble fraction initially crystallizes from the tar of 1- (p-methoxy-benzyl) -2-methyl-12,3,4,5,6,7,8-octahydroisoquinoline, which is levorotatory in ether and which is recrystallized can be separated from the diastereomeric tartrate of 1- (p-methoxy-benzyl) -2-methyl-1,2,3,4,5,6,7,8-octahydro-isochinoline, which is clockwise in ether.
Cyclization with phosphoric acid gives the dextrorotatory 3-oxy-N-methyl-morphinari, which is advantageously isolated from the reaction mixture. The free base can be obtained from the tartrate in a manner known per se.
The methanolic mother liquor from the racemate cleavage is freed from methanol in order to obtain 1- (p-methoxy-benzyl) -2-methyl-1,2,3,4,5,6,7,8-octahydroisoquinoline, which is dextrorotatory in ether and recrystallized several times from Ace clay. In this way it is possible to add the tartrate of 1- (p-methoxy-benzyl) -2-methyl-1,2,3,4,5,6,7,8-octahydro-isoquinoline, which turns clockwise in ether win, from which .by cyclization with the released base the (-) - 3-oxy-N-methyl-morphinan is obtained.
As a result of the repeated recrystallization of 1- (p-methoxy-benzyl) -2-methyl-1,2,3,4,5,6,7,8-oetahydro-isoquinoline from Ace ton losses arise, when obtaining (-) - 3-oxy-N-methyl-morphinane, it is also advantageous to proceed in such a way that the optically not entirely pure, dextrorotatory 1- (p-methoxybenzyl ) -.2 "-methyl-1,2;
3,4,5,16,7,8-octahydro-isoquinoline, as it results from the resolution of the racemate after isolation of the (-) - anti poden. The resulting (-) - 3-oxy- N - methyl - morphinan. namely provides a more difficultly soluble tartrate than its antipode (cf. 0. Schnider and A. Grüssner, loc. cit.), which can now be purified in this stage by crystallization.
The subject of the patent is a process for the production of an optically active 3-oxy-N-methyl-morphinans, which is characterized in that racemic 1- (p-iMeth- o xy, - 'benzyl) -2-methyl- 1.2, lss, 4;
56.7, f 8-oetahydroisoquinoline broken down into the optical antipodes by intermediate salt formation with an optically active acid, and 1- (p-methoxy-benzyl) -2-methyl-1,2,3, which is dextrorotatory in ether , 4,5,6,7,8-oetahydroisoquinoline is cyclized to (-) - 3-oxy-N-methyl-morphinan and isolated via the tartrate.
<I> Example 1 </I> 75 parts by weight of racemic 1- (p-methoxybenzyl) -2-methyl-1,2; 3,4,5,6,7,8-oetahydroisoquinoline are with 41.5 parts by weight of D - (+) - tartaric acid dissolved in 425 parts by volume of methanol on the steam bath. After standing for 24 hours, it is sucked off, concentrated, left to stand and sucked off again.
The methanolic mother liquor, which after isolation: the less soluble tartrate of 1- (p-methoxybenzyl), 2-methyl-1,2, 3,4, 5,6,7, 8-octahydroisoquinoline, which is levorotatory in ether remains, is freed from methanol by evaporation, taken up in acetone and caused to crystallize by standing and seeding.
By recrystallizing from acetone, the tartrate of 1- (p-methoxybenzyl) -2-methyl-1,2; 3,4,5,6,7,8-octahydroisoquinoline, which is dextrorotatory in ether, is obtained in pure form. Melting point 130 to 132 C [a] D = -26 (c = 2 in methanol).
The dextrorotatory 1- (p-methoxy-benzyl) -2-methyl-1,2,3,4,5,6,7,8-octahydro-isoquinoline obtained from this tartrate in ether is an oil with a rotational value of [a ] = -77.5 D (c = 3 in ether).
15 parts by weight of this base are ver with 50 parts by volume of 100 higer phosphoric acid and x about 65 hours at 130 C left to itself. Then 50 parts of the room are mixed with water and heated for 1/2 hour on the steam bath. After dilution with 450 parts by volume of water, it is briefly boiled with activated charcoal and filtered. After adjusting to px 9-10 with ammonia, the raw base is extracted with ether, isolated by evaporation of the solvent and converted into the tartrate. This is briefly boiled in an aqueous solution with charcoal, filtered and crystallized.
The (-) - 3-oxy-N-methyl-morphinan tartrate crystallizes as a dihydrate and melts at 113 to 115 C, [a] D = -14 (c = 3 in water). The (-) - 3-oxy-N-methyl-morphinan produced from the tartrate has a rotation value of [.a] = D -56 (c = 1 in methanol) and melts at 198-199 C.
<I> Example 2: </I> The methanolic mother liquor, which after isolation of the sparingly soluble tartrate of 1- (p-methoxybenzyl) -2-methyl-1,2,3,4,5,6, which is levorotatory in ether 7,8-octahydro-iso-ehinolins according to Example 1 remains, is freed from methanol by evaporation and without further crystallization from acetone therefrom by precipitation with ammonia and extraction with ether the optically impure 1- (p-methoxy-, which turns clockwise in ether) benzyl) - 2-methyl-1,2,3,4,5,6,7,
8-octahydroisoquinoline isolated. After cyclization of this optically unpure base, according to Example 1, the crude base obtained is converted into the tartrate and this is purified by crystallization from water and thereby separated from the more soluble tartrate of the dextrorotatory 3-oxy-N-methylmorphinane. The tartrate obtained crystallizes as a dihydrate, melts at 113 to 115 C and has a rotation value of [a] =: D -14 (c = 3 in water).
The (-) - 3-oxy-N-methyl-morphinan obtained from it melts at 198-200 C and has a rotation value of [a] D = -56 (c = 1 in methanol).
