Verfahren zur Herstellung der optisch aktiven Formen des -Methyl-/3- (3, 4-dihydroxyphenyl)-alanins und deren N, 0, 0-Triacetylderivate

Es ist bekannt, dass nur die L-enantiomorphe Form des < x-Methyl-8- (3, 4-dihydroxyphenyl)-ala- nins die blutdrucksenkende Wirkung aufweist, während das D-Isomere keine solche Wirkung, aber die gleiche Toxizität wie das L-Isomere zeigt ; aus diesem Grund ist die Zerlegung des Racemats in seine optischen Antipoden und die mit guter Ausbeute erfolgende Gewinnung der L-Form von grosser praktischer Wichtigkeit.
Es wurden schon verschiedene chemische, physikalische und biochemische Verfahren zur Gewinnung der optisch aktiven Formen dieser Verbindung vorgeschlagen. Praktisch werden die chemischen Methoden bevorzugt ; so wird z. B. nach der brit. Patentschrift Nr. 936,074 bzw. den österr. Patentschriften Nr. 234666 und Nr. 234674 das N-acetylierte racemische a-Methyl-8- (3, 4-dimethoxyphe- nyl)-alanin in einem organischen Lösungsmittel, z.B. in Methanol, mit 1-α-Phenyläthylamin oder das N, 0, O-triacetylierte racemische α-Methyl-ss-(3,4-dihydroxyphenyl)-alanin in Aceton mit Chinin um- gesetzt und das Salz der L-Verbindung durch fraktionierte Kristallisierung abgetrennt.
Nach der nicht vorveröffentlichten österr. Patentschrift Nr. 250936 wird der racemische n-Butyl-

Im Laufe unserer diesbezüglichen Forschungsarbeit haben wir festgestellt, dass man optisch reine Produkte und wesentlich bessere Ausbeuten erzielen kann, wennmandasracemischeN-Acetyl-a-me- tlyl- (3, 4-dimethoxyphenyl)-alanin in wässerigem Medium mit l-a-Phenyläthylamin umsetzt, das aus der wässerigen Lösung auskristallisierende L-of-Phenyläthylaminsalz des L-N-Acetyl-a-methyl- - B- (3, 4-dimethoxyphenyl)-alanins abtrennt und die erhaltene Mutterlauge, welche im wesentlichen das entsprechende Salz der D-Verbindung enthält, als Lösungsmittel bei den weiteren Zerlegungsoperationen anwendet.
Aus solchen Losungen scheidet sich das kristalline 1-α-Phenyläthylaminsalz der L-Verbindung mit um 10-15'% grösserer, fast quantitativer Ausbeute aus und die Reinheit der auf diese Weise aus einer, die D-Verbindung in viel grösserer Konzentration enthaltenden Lösung erhaltenen L-Verbindung ist in überraschender Weise ebenso gut wie bei der ersten, in rein wässerigem Medium durchgeführten Abscheidung.
Die Mutterlauge der nacheinander durchgeführten Zerlegungen kann so lange immer wieder zur weiteren Zerlegung angewendet werden bis die Gesamtkonzentration der in ihr gelösten diastereomeren Salze einen Wert von etwa 350 bis 400 g/l erreicht. Bei einer solchen Konzentration kann durch

Abkühlen der Lösung eine grosse Menge von mit der L-Form ein wenig verunreinigten D-N-Acetyl- -α-methyl-(3,4-dimethoxyphenyl)-alanin -1-α-phenyläthylaminsalze auskristallisiert werden, nach dessen Abtrennen die Mutterlauge wieder zu weiteren Spaltungen verwendet werden kann ; aus dem derart auskristallisierten Salz der D-Verbindung kann das 1-a-Phenyläthylamin in bekannter Weise regeneriert werden.
Beider weiteren Entwicklung dieser Methode haben wir nun gefunden, dass das 1-ct-Phenyläthylamin bei dem oben beschriebenen Verfahren vorteilhaft auch in Form von mit anorganischen oder organischen Säuren gebildeten Salzen zur Zerlegung angewendet werden kann unddassdasl- < x-Phenyläthyl- amin bzw. seine Salze auch in kleineren Mengen als die stöchiometrische Menge eingesetzt werden können, wenn man die Spaltung in Anwesenheit einer anorganischen bzw. optisch inaktiven organi-

Nach dem in bekannter Weise, mit einer Mineralsäure oder starken Base durchgeführten Zersetzen des auskristallisierten und getrennten optisch aktiven Salzes wird dann das entsprechende optisch aktive N-Acetyl-α-methyl-ss-(3,4-dimethoxyphenyl)-alanin erhalten, aus welchem manebenfallsin an sich bekannter Weise, z. B. durch Behandlung mit Bromwasserstoffsäure oder Aluminiumbromid, das entsprechende optisch aktive a-Methyl-ss- (3, 4-dihydroxyphenyl)-alanin und gegebenenfalls durchin an sich bekannter Weise, zweckmässig unmittelbar im Reaktionsgemisch, z. B. mit Essigsäureanhydrid, durchgeführte Acylierung das entsprechende optisch aktive N-Acetyl-a-methyl-B- (3, 4-diacetoxyphe- nyl)-alanin erhalten kann.
Zur Bildung der als Spaltungsmittel anzuwendenden a-Phenyläthylaminsalze können im erfindungsgemässen Verfahren die verschiedensten Säuren verwendet werden. Gute Ergebnisse wurden z. B. mit den folgenden Säuren erzielt : Phosphorsäure, Schwefelsäure, Weinsäure, Oxalsäure, Fumarsäure, 4-Nitrobenzoesäure, 4-Aminobenzoesäure, Äpfelsäure, Pikrinsäure, Benzoesäure.
Die neben dem optisch aktiven cx-Phenyläthylamin bzw. neben dem Salz dieser optisch aktiven Base erfindungsgemäss zu verwendende optisch inaktive organische bzw. anorganische Base kann ebenfalls aus einem weiten Bereich der wasserlöslichen optisch inaktiven Basen gewählt werden ; am ein-

Der Gegenstand der Erfindung ist also ein Verfahren zur Herstellung der optisch aktiven Formen des α-Methyl-ss-(3,4-dihydroxyphenyl)-alanins und gegebenenfalls der N, O, O-Triacetylderivate dieser optisch aktiven Verbindungen, welches dadurch gekennzeichnet ist, dass man das racemische N-Ace- tyl-α
-methyl-ss-(3,4-dimethoxyphenyl)-alanin in wässerigem Medium mit einerkleinerenals stöchio-

metrisch äquivalenten Menge von optisch aktivem a-Phenyläthylamin oder vorteilhaft von einem Säu- readditionssalz dieser optisch aktiven Base in der Anwesenheit einer optisch inaktiven organischen oder anorganischen Base unter solchen Mengenverhältnissen umsetzt, dass die Gesamtmenge des optisch ak- tiven a-Phenyläthylamins bzw. eines Salzes dieser optisch aktiven Base und der anwesenden optisch in- aktiven organischen bzw. anorganischen Base mit der Menge des zu zerlegenden racemischen Ausgangs- stoffes mindestens äquivalent ist, dann aus der in dieser Weise erhaltenen wässerigen Lösung des Salz- gemisches die besser kristallisierende diastereomere Form des mit dem α
-Phenyläthylamin gleicherop- tischer Aktivität gebildeten Salzes des optisch aktiven N-Acetyl-α-methyl-ss-(3,4-dimethoxyphenyl)- - alanins durch Kristallisieren abtrennt, die Mutterlauge wiederholt zum Zerlegen weiterer Mengendes racemischen N-Acetyl-α-methyl-ss-(3,4-dimethoxyphenyl)-alanins verwendet, aus dem in kristalliner Form gewonnenen optisch aktiven Salz das optisch aktive N-Acetyl-α
-methyl-ss- (3, 4-dimethoxyphenyl)- - alanin in an sich bekannter Weise freisetzt, die erhaltene optisch aktive Aminosäure durch Behandlung mit Bromwasserstoffsäure oder Aluminiumbromid in an sich bekannter Weise demethyliert unddesace- tyliert und gegebenenfalls-vorteilhaft in dem selben Reaktionsgemisch - das erhaltene optisch aktive a-Methyl-ss- (3, 4-dihydroxyphenyl)-alanin mit Essigsäureanhydrid in an sich bekannter Weise in das entsprechende Triacetylderivat überführt.
Das erfindungsgemässe Verfahren zeigt erhebliche Vorteile gegenüber den bisher bekannten Zerlegungsmethoden von diesen Phenylalaninderivaten. Zum Zerlegen der racemischen Aminosäure wird eine wesentlich kleinere Menge von optisch aktiver Base verwendet als stöchiometrisch erforderlich wäre, wodurch das Verfahren in erheblichem Mass wirtschaftlicher wird. Die Reinheitdergewonnenen optisch aktiven Produkte ist ebenfalls grösser als bei den bisher bekannten Verfahren ; da femer die Mutterlauge nach Abscheiden des gewünschten optisch aktiven Produktes wiederholt verwendet wird, kann unter Umständen auch auf die Zurückgewinnung des Spaltungsmittels aus der Mutterlauge verzichtet werden. Das optisch aktive a-Phenyli1thylamin kann sowohl in Form von freier Base als auch in Form irgendeines Salzes verwendet werden.
Die letztere Verfahrensweise, d. h. die Verwendung eines Phenyl- äthylaminsalzes ist wesentlich vorteilhafter ; die bei der Zerlegung des racemischen a-Phenyläthyl- amins bzw. bei der Rückgewinnung des a-Phenyläthylamins nach der Zersetzung des abgetrennten op- tisch aktiven N-Acetyl-α-methyl-ss-(3,4-dimethoxyphenyl)-alanin-phenyläthylaminsalzes erhaltenen optisch aktiven a-Phenyläthylaminsalze können auf diese Weise unmittelbar zum Zerlegen des racemischen N-Acetyl-α-methyl-ss-(3,4-dimethoxyphenyl)-alanins verwendet werden und somit kanndie bisher zur Isolierung der optisch aktiven Basen allgemein angewendete und erhebliche Verluste verursachende Vakuumdestillation vermieden werden.
Ein weiterer, sehr wesentlicher Vorteil des erfindungsgemässen Verfahrens ist, dass die optisch aktiven diastereomeren Salze mit erheblich holleren Ausbeuten erhalten werden können als bei den bisher bekannten Verfahren ; das erfindungsgemässe Verfahren liefert praktisch quantitative, zip der theoretischen Mengen entsprechende Ausbeuten an optisch aktiven Produkten.
Gleichzeitig ist auch die optische Reinheit der in dieser Weise mit höheren Ausbeuten erhaltenen Produkte grösser ; das spezifische Drehvermögen der erhaltenen optisch aktiven N-Acetyl-a-methyl- -ss-(3,4-dimethoxyphenyl)-alanin-α-phenyläthylaminsalze erreicht unmittelbar ¯ 76 ; mit dem bisher bekannten Verfahren wurden unter betrieblichen Verhältnissen unmittelbar optische Drehvermögen von : ! : 550 bzw. nach Umkristallisieren von : ! : 690 erhalten. Das erfindungsgemässe Verfahren ermög- licht also die unmittelbare Herstellung von optisch aktiven Produkten mit höherer optischer Reinheit, unter gleichzeitiger Erhöhung der Ausbeuten.
Das erfindungsgemässe Verfahren wird durch die nachstehenden Beispiele näher veranschaulicht.
