Verfahren zur Herstellung eines Nicotinsäureesters. Das Hauptpatent betrifft ein Verfahren zur Herstellung von Nicotinsäure-ss-b-Litoxy- äthylester, der durch Umsetzung von Nicotin- säure oder eines N icotinsäurederivates, wie z. B. N icot.insäurechlorid-hydrochlorid, Nicotin- säureanhydrill oder Nicotinsäurealkylestern mit @lthyleuglykolmonobutyläther erhalten wird.
Gegenstand des vorliegenden Patentes ist nun ein Verfahren zur Herstellung eines neuen Nieotinsäureesters, nämlich des Nicotin- säure-ss-propoxyäthylesters, das dadurch ge kennzeichnet ist, dass man Nicotinsäure oder ein Nicotinsäurederivat mit dem Monopro pyläther von Äthylenglykol umsetzt.
Die so erhaltene Verbindung stellt eine ölige, gelbliche Flüssigkeit vom Kp.17 = 165 dar, die ein hyperämisierendes Mittel von langer Wirkungsdauer ist.
Wird die Umsetzung der Ausgangsstoffe in Gegenwart von organischen Lösungsmit teln vorgenommen, so können beispielsweise Benzol, Toluol, Tetrachlorkoblenstoff u. ä. Verwendung finden.
Man kann die Umsetzung vorteilhaft. in Gegenwart von wasserentziehenden oder säurebindenden Mitteln vornehmen. Als was serentziehende Mittel sind unter anderem Salzsäure und Schwefelsäure geeignet. Bei Verwendung von Nicotinsäurehalogeniden als Ausgangsmaterial kommen zur Bindung des freiwerdenden Halogenwasserstoffes Pyridin, Dimethylanilin u. ä.. in Betracht.
Beispiel <I>1:</I> 61,5 g Nicotinsäure werden mit 260 g rei nem Thionylchlorid versetzt und 30 Minuten am Rückflusskühler erhitzt. Die Hauptmenge des Thionylehlorids wird dann bei gewöhn lichem Druck und der Rest im Vakuum bei einer 70 nicht übersteigenden Aussentem peratur abdestilliert. Zu dem Kristallkuchen von Nicotinsäurechlorid-hydrochlorid gibt man, nachdem ein Rückflüsskühler aufgesetzt wurde,
ein Gemisch von 83 g Äthylenglykol- monopropyjäther und 64 g wasserfreiem P3-ri- din zu und schüttelt kräftig, wobei starke Er wärmung eintritt, bis zur völligen Lösung. Nach mehrstündigem Stehen wird Wasser zu gegeben und so viel Natr iumcarbonat, bis sich keine Kohlensäure mehr entwickelt. Die ab geschiedene ölige Flüssigkeit wird der Va kuumdestillation unterworfen.
Man erhält den Ester als ölige, gelbliche Flüssigkeit vom KP-17 = 165 .
Beispiel <I>2:</I> Eine Mischung von 61,5g Nicotinsäure und 125 g Äthylenglykolmonopropyläther wird unter Durchleitung eines Salzsäuregasstromes 4 Stunden im Ölbad von etwa 7.40 erhitzt und wie im Beispiel 1 angegeben aufgearbeitet. Der erhaltene Xicotinsäure-ss-propoxyäthyl- ester hat einen KP-17 = 165 .
