Verfahren zur Herstellung eines Nicotinsäureesters. Das Hauptpatent betrifft ein Verfahren zur Herstellung von Nicotinsäure-ss-butoxy- äthylester, der durch Umsetzung von Nicotin- ;äure oder eines Nicotinsäurederivates, wie z. B. Nieotinsä,urechlorid-hydrochlorid, Nico- tinsäureanliydrid oder Nieotinsäurea.lkylestern finit Äthylen.glykolmonobutylä-ther erhalten wird.
Gegenstand des vorliegenden Patentes ist nun ein Verfahren zur Herstellung eines neuen Nieotinsäureesters, nämlich des Nieot.in- säure-co-äthoxydiä.thylenglykolesters, das da- (lureli gekennzeichnet ist, dass man Nicotin- sänre oder ein Nieotinsäurederivat mit dem llonoätlivläther von Diäthylenglvkol umsetzt.
Die so erhaltene Verbindung stellt eine ölige, gelbliebe Flüssigkeit vom Kp.12 = 179 bis 1.80 dar, die ein hyperämisierendes Mittel von langer Wirkungsdauer ist.
Wird die Umsetzung der Ausgangsstoffe in Gegenwart von organischen Lösungsmitteln vorgenommen, so können beispielsweise Ben zol, Toluol, Tetrachlorkohlenstoff u. ä. Ver wendung finden.
Man kann die Umsetzung vorteilhaft in Gegenwart von wasserentziehenden oder säure bindenden Mitteln vornehmen. Als wasserent ziehende Mittel sind unter anderem Salzsäure und Schwefelsäure geeignet. Bei Verwendung von Nieotinsäurehalogeniden als Ausgangs- inaterial kommen zur Bindung des freiwer denden Halogenwasserstoffes Pyridin, Dime- t.liylanilin u. ä. in Betracht.
<I>Beispiel 1:</I> 123g Nicotinsäure und 250 g Diäthylen- glykolinonoätliyläther werden in einen Kol ben gegeben, der mit einem weiten, langen, als Luftkühler dienenden Glasrohr, einem in die Flüssigkeit. reichenden Gaseinleitungsrohr und einem ebensolchen Thermometer versehen ist. Der Kolben befindet sich in einem Ölbad, das so geheizt wird, dass die im Kolben be findliche Flüssigkeit in stetem, kräftigem Sie den gehalten wird. Gleichzeitig leitet man durch das Gaseinleitungsrohr einen steten Stickstoffstrom. Die Temperatur der Flüssig keit steigt im Verlauf von fünf Stunden kon tinuierlich von 190 auf 205 .
Danach lässt man erkalten, saugt von der nicht umgesetzten Nicotinsäure ab, die zum nächsten Ansatz wieder verwendet. wird, und destilliert im Vakuum. Der Nicotinsäure-co-äthoxydiäthylen= glykolester wurde als ölige, gelbliche Flüssig keit vom Kp.12 = 179-180 in einer Aus beute von 11-0 g erhalten. Beispiel <I>2:</I> 123 g Nicotinsäure werden mit 400 g reinem. Thionylchlorid versetzt und 30 Mina- ten unter Rückfluss erhitzt.
Man destilliert die Hauptmenge des überschüssigen T'hionyl- chlorids bei gewöhnlichem Druck und den Rest im Vakuum, gegebenenfalls unter Zusatz von Benzol, ab. Zu dem so erhaltenen Nicotin- säurechlorid-hydrochlorid gibt man unter Rückflusskühlung und kräftigem Schütteln eine Mischung von 160 g Diäthylenglykol- monoäthyläther und 200g wasserfreiem Pyri- din, wobei starke Erwärmung eintritt.
@Tach- dem vollständige Lösung eingetreten ist, lässt man einige Stunden stehen, gibt. dann Wasser und so viel Natriumearbonat zu, bis keine Kohlensäure mehr entweicht. Die abgeschie dene ölige Flüssigkeit wird der Vakuumdestil lation unterworfen. Es wurde eine Ausbeute von 109 g einer öligen, gelblichen Flüssigkeit vom Kp.h = l79-181 erhalten.
Beispiel <I>3:</I> 100 g Nicotinsäureäthylester und 300 Diäthylenglykolmonoä.thylät'her werden zweek- mä.ssig unter Zusatz von 1 g Nicotinsäure als Katalysator und unter Durehleiten eines inerten Gasstromes 12 Stunden am Luftküh ler im Ölbad in stetem Sieden gehalten. Nach dem Abkühlen wird das Reaktionsgemisch im Vakuum destilliert.
Der zuerst übergehende Anteil an unverändertem Diäthylenglykol- monoäthylät.her wird abgetrennt und das nun folgende Gemisch aus Nicotinsäureät'hylester und Nicotiusäure-o)-äthoxydiäthylenglykol- ester aufgefangen. Nach dem Durchschütteln mit Wasser erhält mann eine ölige Schicht, die nochmals im Vakuum destilliert wird. Es wurde eine Ausbeute von 80 g einer öligen, gelblichen Flüssigkeit vom Kp.i2 = 179-181 erhalten.
