Dans le brevet principal, on a décrit un procédé de préparation d'un 1-nitrophényl-2- acylamido - propanediol -1,3, c'est-à-dire du pseudo -1. p -nitrophényl-2-dichloracétamido- propanediol-1,3, dont l'isomère optique (1) ayant un pouvoir rotatoire négatif dans l'acé tate d'éthyle est bien connu sous le nom de chloramphénicol.
Le présent brevet a pour objet le procédé de préparation d'un autre composé 1-nitro- phényl-2-amino-propanediol-1,3, c'est-à-dire du pseudo-1-p-nitrophényl-2-amino-propanediol- 1,3 qui est -une substance connue et dont toutes les formes optiques (d, 1 et dl) sont des pro duits intermédiaires importants pour la pré paration de substances à activité antibiotique.
Le procédé selon l'invention pour la pré paration du pseudo-1-p-nitrophényl-2-amino- propanediol-1,3 est caractérisé en ce que l'on soumet à une réduction sélective un pseudo-1- p-nitrophényl-amino-propane de formule

où R' est de l'hydrogène ou un radical alcoyle, pour obtenir le pseudo-1-p-pitrophényl-2- amino-propanediol-1,3. Dans le corps de dé part, R' est de préférence un radical alcoyle inférieur tel que méthyle ou éthyle.
La réduction sélective doit être effectuée avec un agent réducteur n'agissant pas sur le groupe nitro, mais affectant uniquement le groupe carboxyle estérifié ou non. De préfé- rence, on utilise, pour la réduction sélective, l'hydrure de lithium et d'aluminium LiAlH4 dans un solvant organique anhydre exempt de groupes hydroxyle. On peut citer comme solvants appropriés l'éther diéthylique, l'éther dibutylique, le dioxane, le tétrahydrofurane, l'éther diméthylique de l'éthylène-glycol,
l'éther diéthylique de l'éthylène-glycol, etc.
La quantité d'hydrure de lithium et d'alu minium utilisée pour effectuer la réduction sélective du corps de départ dépend de la nature des groupes présents. On utilise suffi samment de LiAIH4 pour réagir avec les groupes amino et hydroxyle et avec le groupe carboxyle estérifié ou non.
D'ordinaire, on obtient les meilleurs résultats en utilisant entre 10 % de plus et 10 % de moins que la quantité théorique de LiAlH4 nécessaire. On peut,
si on le désire, utiliser=. jusqu'à 35 % d'excès de Li 1.H4, mais, comme on l'a indi qué plus haut, on obtient les meilleurs résul- tats avec une quantité moindre.
La quantité théorique de LiAlH4 à utiliser pour une mole du corps de départ peut être calculée très facilement au moyen du tableau suivant:

Pour effectuer la réduction, on ajoute l'hydrure de lithium et d'aluminium au corps de départ, de manière à éviter la présence d'un excès initial d'agent réducteur dans le mélange réactionnel. On obtient de meilleurs résultats en effectuant l'addition lentement et d'une manière telle qu'elle ne dépasse pas la vitesse de réaction. La température au cours de la réduction n'est pas particulièrement critique. C'est ainsi qu'on peut utiliser des températures atteignant 50 Ç ou des tempé ratures dans le voisinage de 0 C. La tempé rature optimum est de l'ordre de 15 à 35 C.
Après achèvement de la première phase de la réduction, on décompose le complexe métallique formé au moyen d'eau. La quan tité d'eau utilisée dans ce but est d'au moins quatre moles par mole d'hydrure de lithium et d'aluminium utilisée dans la réduction. Dans la pratique,. on utilise ordinairement plusieurs fois la quantité minimum d'eau, et la limite supérieure de cette quantité n'est fixée que par des raisons de commodité. On peut faciliter l'isolement du produit final en ajoutant à l'eau un acide minéral, de manière à dissoudre les hydroxydes de lithium et d'aluminium formés par la décomposition du complexe métallique.
Les produits de départ utilisables sont des substances connues.
L'invention est illustrée par les exemples suivants <I>Exemple 1:</I> On ajoute goutte à goutte une solution de 5,13 g d'hydrure de lithium et d'aluminium dans 200 -cms d'éther anhydre, en agitant, à une solution de 25,4 g de 1-y-p-nitrophényl- sérinate d'éthyle dans 3 litres d'éther anhydre, en l'espace de six heures et à la température ordinaire.
L'addition terminée, on agite le mé lange réactionnel, pendant encore six heures et on le traite ensuite avec 200 cm3 d'eau froide pour décomposer le complexe métalli que insoluble. On évapore ensuite le mélange réactionnel à siccité dans le vide et on extrait le résidu complètement à l'aide d'alcool absolu froid. On évapore les extraits réunis et on dissout le résidu dans 300 cm3 d'une solution aqueuse de NaOH 0,1N et on laisse reposer le mélange à la température ambiante pendant plusieurs heures.
Ensuite, on évapore la solu tion dans le vide pour éliminer l'acétone et on extrait le résidu aqueux avec de l'acétate d'éthyle. On sèche les extraits réunis et on distille l'acétate d'éthyle pour obtenir le 1-y-1- p-nitrophényl-2-amino-propanediol-1,3 désiré de formule

Celui-ci peut être purifié par reeristallisation d'ans de l'eau; point de fusion: 162-163 C; (a) D = - 23 dans du méthanol.

L'addition terminée, on agite le mélange réactionnel pendant encore six heures et on le traite ensuite avec 200 em3 d'eau pour décomposer le complexe métalli que insoluble.

On sèche les extraits d'acétate d'éthyle et on distille le solvant pour obtenir le dl-V-1-p-nitrophényl-2-amino-propanediol- 1,3, lequel peut être purifié par recristallisa- tion dans du diehlorure d'éthylène; point de fusion: 140 5 C.

In the main patent, a process for preparing a 1-nitrophenyl-2-acylamido-propanediol -1.3, that is to say pseudo -1, has been described. p -nitrophenyl-2-dichloroacetamido-propanediol-1,3, the optical isomer (1) of which having negative optical rotation in ethyl acetate is well known under the name of chloramphenicol.
The present patent relates to the process for preparing another compound 1-nitro-phenyl-2-amino-propanediol-1,3, that is to say pseudo-1-p-nitrophenyl-2-amino -propanediol- 1,3 which is a known substance and of which all the optical forms (d, 1 and dl) are important intermediates for the preparation of substances with antibiotic activity.
The process according to the invention for the preparation of pseudo-1-p-nitrophenyl-2-amino-propanediol-1,3 is characterized in that a pseudo-1-p-nitrophenyl- is subjected to selective reduction. amino propane of formula

where R 'is hydrogen or an alkyl radical, to obtain pseudo-1-p-pitrophenyl-2-amino-propanediol-1,3. In the starting body, R 'is preferably a lower alkyl radical such as methyl or ethyl.
The selective reduction should be carried out with a reducing agent which does not act on the nitro group, but only affects the esterified or unesterified carboxyl group. Preferably, lithium aluminum hydride LiAlH4 in an anhydrous organic solvent free of hydroxyl groups is used for the selective reduction. As suitable solvents, mention may be made of diethyl ether, dibutyl ether, dioxane, tetrahydrofuran, dimethyl ether of ethylene glycol,
ethylene glycol diethyl ether, etc.
The amount of lithium aluminum hydride used to effect the selective reduction of the starting material depends on the nature of the groups present. Sufficient LiAIH4 is used to react with the amino and hydroxyl groups and with the esterified or unesterified carboxyl group.
Usually, best results are obtained using between 10% more and 10% less than the theoretical amount of LiAlH4 required. We can,
if desired, use =. up to 35% excess Li 1.H4, but, as stated above, the best results are obtained with less.
The theoretical amount of LiAlH4 to be used for one mole of the starting material can be calculated very easily by means of the following table:

To effect the reduction, lithium aluminum hydride is added to the starting material, so as to avoid the presence of an initial excess of reducing agent in the reaction mixture. The best results are obtained by carrying out the addition slowly and in such a way that it does not exceed the reaction rate. The temperature during the reduction is not particularly critical. It is thus possible to use temperatures reaching 50 ° C. or temperatures in the vicinity of 0 C. The optimum temperature is of the order of 15 to 35 C.
After completion of the first phase of the reduction, the metal complex formed is decomposed by means of water. The amount of water used for this purpose is at least four moles per mole of lithium aluminum hydride used in the reduction. In practice,. the minimum quantity of water is usually used several times, and the upper limit of this quantity is fixed only for reasons of convenience. The isolation of the final product can be facilitated by adding a mineral acid to water, so as to dissolve the lithium and aluminum hydroxides formed by the decomposition of the metal complex.
The starting materials which can be used are known substances.
The invention is illustrated by the following examples <I> Example 1: </I> A solution of 5.13 g of lithium aluminum hydride in 200 cms of anhydrous ether is added dropwise, in stirring, to a solution of 25.4 g of ethyl 1-yp-nitrophenylserinate in 3 liters of anhydrous ether, over six hours and at room temperature.
When the addition is complete, the reaction mixture is stirred for a further six hours and then treated with 200 cc of cold water to decompose the insoluble metal complex. The reaction mixture is then evaporated to dryness in vacuo and the residue is extracted completely with cold absolute alcohol. The combined extracts are evaporated and the residue dissolved in 300 cm 3 of a 0.1N aqueous NaOH solution and the mixture is left to stand at room temperature for several hours.
Then, the solution is evaporated in vacuo to remove acetone, and the aqueous residue is extracted with ethyl acetate. The combined extracts were dried and ethyl acetate distilled to afford the desired 1-y-1- p-nitrophenyl-2-amino-propanediol-1,3 of formula
