Chacune des deux formes cis et trans existe encore sous forme d'isomères optiques; c'est- à-dire comme racémate et sous forme d'iso mères optiquement actifs dextrogyre (d) et lévogyre (Â).
En raison de la difficulté à représenter ces différences de structure dans les formules graphiques, on utilise conventionnellement les formules de structure habituelles en portant, au-dessous ou à côté de chaque formule, une. notation indiquant la structure et la configu ration optique particulières du composé. Le présent brevet a pour objet un procédé de préparation du pseudo-1-p-nitrophényl>2- dichloracétamido-propanediol-1,3, dont l'iso mère (1) est connu sous le none de chlor- amphénicol .

où R' est de l'hydrogène ou un radical alcoyle, de façon à obtenir le pseudo-1-p-nitrophényl- 2-dichloracétamido-propanediol-1,3. Ce dernier est une substance bien connue dont toutes les formes optiques (d, 1 et dl) possèdent des propriétés antibiotiques, particulièrement les formes optiques 1 et dl, et peuvent également servir de produits intermédiaires pour la pré paration d'autres composés- organiques à acti vité antibiotique. Dans le corps de départ, R' est de préférence un radical alcoyle infé rieur, tel que méthyle ou éthyle.
La réduction sélective doit être effectuée avec un agent réducteur n'agissant pas sur les groupes nitro et dichloracétamido du pro duit de départ, mais affectant uniquement le groupe carboxyle estérifié ou non. De préfé rence, on utilise pour la réduction sélective l'hydrure de lithium et d'aluminium LiA1H4 dans un solvant organique anhydre, exempt de groupes hydroxyle.
On peut citer comme solvants appropriés l'éther diéthylique, l'éther dibutylique, le dioxane, le tétrahydrofurane, l'éther diméthylique de l'éthylène-glycol, l'éther diéthylique de l'éthylène-glycol, etc.
La quantité d'hydrure de lithium et d'alu minium utilisée pour effectuer la réduction sélective du groupe carboxyle estérifié ou non du corps de départ dépend de la nature des groupes présents dans la matière première. On utilise assez d'hydrure de lithium et d'alu minium pour réagir avec le groupe hydroxyle et avec le groupe ester ou carboxyle libre.
D'ordinaire, on obtient les meilleurs résultats en utilisant entre 10 % de plus et 10 0/0 de moins que la quantité théorique de LiAlH4 nécessaire à la réaction avec les groupes ester ou carboxyle et hydroxyle présents dans le corps de départ.
On peut, si on le veut, utili- ser jusqu'à 35 % d'excès de LiAlH4, mais, comme on l'a indiqué, on obtient les meilleurs résultats avec une quantité moindre. La quan tité théorique d'hydrure de lithium et d'alu minium à utiliser pour une mole du corps de départ peut être calculée très facilement nu moyen du tableau suivant.

On obtient les meilleurs résultats en effectuant l'addition lentement et d'une manière telle qu'elle ne dépasse pas la vitesse de la réaction. La température au cours de la réduction n'est pas particulière ment critique.
C'est ainsi qu'on peut utiliser des températures atteignant 50 C ou des tem pératures dans le voisinage de 0 C. La tempé rature optimum est de l'ordre de 15 à 35 C.
Après achèvement de la. première phase de la réduction, on décompose le complexe métal lique formé au moyen d'eau. La quantité d'eau utilisée dans ce but est d'au moins quatre moles par mole d'hydrure de lithium et d'alu minium utilisé dans la réduction. Dans la pratique, on utilise ordinairement plusieurs fois la quantité minimum d'eau, et la limite supérieure de cette quantité n'est fixée que par des raisons de commodité. On peut faci liter l'isolement du produit final en ajoutant. à l'eau un acide minéral de manière à dis soudre les hydroxydes de lithium et d'alumi nium formés par la décomposition du com plexe métallique. Les produits de départ utilisables sont des substances connues.
L'invention est illustrée par les exemples suivants: Exemple <I>1:</I> On verse goutte à goutte une solution de 3,23 g d'hydrure de lithium et d'aluminium dans 100 ems d'éther anhydre, en agitant, dans une solution de 36,5 g de (1)-y-N-di- chloroacétyl - p - nitrophényl-sérinate d'éthyle dans 5,5 litres d'éther anhydre, en l'espace de six heures à la température ambiante. L'addi tion terminée, on agite le mélange réactionnel pendant encore quatre heures.
On verse alors lentement dans le mélange réactionnel 350 em3 d'acide chlorhydrique 2N pour décomposer le complexe métallique insoluble et l'on sépare la phase éthérée. On extrait la phase aqueuse au moyen de plusieurs portions d'acétate d'éthyle, on mélange les extraits, on sèche et on éva pore sous vide jusqu'à siccité. On lave la phase éthérée à l'eau, on la sèche et on l'éva pore sous vide jusqu'à siccité. On réunit les résidus provenant de la phase éthérée et de l'extrait d'acétate d'éthyle, et on les dissout dans 500 ems d'acétone.
On ajoute 500 ems d'une solution aqueuse d'hydroxyde de sodium 0,1N et on abandonne le mélange au repos à 20 C pendant une heure. On neutralise à froid la solution au moyen d'acide chlor hydrique dilué et on évapore l'acétone sous vide. On alcalinise le résidu aqueux au moyen d'une solution diluée d'hydroxyde de sodium et on épuise la solution au moyen de plu sieurs portions d'acétate d'éthyle.
On lave les extraits réunis, on les sèche et on distille l'acétate d'éthyle de manière à obtenir le (1)-y-1-p-nitrophényl-2-dichloroacétamidopro- panediol-1,3 (chloramphénicol) désiré, de for mole

L'addition terminée, on agite le mé lange réactionnel pendant cinq heures, puis on traite ce mélange au moyen de 350 em3 d'acide chlorhydrique 2 N pour décomposer le complexe métallique insoluble.
On sépare la phase organique et on épuise la phase aqueuse. au moyen d'acétate d'éthyle. On ajoute les extraits d'acétate d'éthyle à la phase organique, on lave le mélange à l'eau puis au moyen d'une solution étendue d'hydroxyde de sodium, enfin de nouveau à l'eau. On sèche la phase organique et on chasse les solvants par distillation. On recueille le résidu qui est formé de (1)-yp-1-p-nitrophényl-2-dichloro- acétamidopropanediol-1,3 (chloramphénicol) brut et on le fait recristalliser au sein d'acé tate d'éthyle; point de fusion: 150-151 C; [a] 21 :<B>-2505</B> dans l'acétate d'éthyle.
<I>Exemple 3:</I> On verse goutte à goutte une solution de 1,61 g d'hydrure d'aluminium et de lithium dans 75 em3 d'éther anhydre, en agitant, dans une solution de 18,3 g de (dl)-y.;-N-dichloro- acétyl-p-nitrophényl-sérinate d'éthyle dans 3 litres d'éther anhydre, en l'espace de six heures à la température ambiante. L'addition terminée, on agite Je mélange réactionnel pen dant cinq heures. On ajoute lentement 350 em3 d'acide chlorhydrique I N au mélange réac tionnel pour décomposer le complexe métalli que insoluble.
