<B>Procédé de préparation</B> d'un <B>dérivé nitré d'un</B> acylamidodiol aromatique. La présente invention concerne la prépa ration de certains dérivés nitrés d'aminodiols aromatiques N-acylés.
On a trouvé que des acylamidodiols nitrés ayant. la formule générale suivante:

oü Ri et R2 sont identiques ou différents et- représentent de l'hydrogène, un atome d'halo gène, des radicaux alcoyle inférieurs ou des radicaux alcoxy inférieurs, sont. des subs tances importantes soit par leurs propriétés thérapeutiques et antibiotiques, soit comme produits intermédiaires pour la préparation d'autres substances organiques possédant des propriétés antibiotiques.
On sait que les composés représentés par la formule précédente ainsi que leurs produits de départ et les produits intermédiaires pour leur synthèse peuvent exister sous forme d'iso mères de structure (formes diastéréo-isoméri- que5) et sous forme d'isomères optiques. Par isomères de structure , on entend les dérivés eis et trans, c'est-à-dire les dérivés qui se dis tinguent par la position stérique, relativement à. un même plan, des groupes polaires fixés sur les deux atomes de carbone asymétriques.
Pour différencier ces deux différentes formes diastéréo-isométriques, on dénommera par la suite réguliers (rég.) les composés cis , et pseudo (y) les composés trans . Les compo sés Pis ou rég. peuvent aussi être désignés par le préfixe érythro ou allo, et les composés trans ou pseudo par le préfixe thréo. Les composés ayant la configuration cis (ou rég.)
sont ceux dont les deux groupes les plus fortement po laires sont du même côté du plan dans lequel se trouvent les deux atomes de carbone asy métriques ou dont, dans le cas de composés intermédiaires oxazoline, le groupe -CH20H (ou un groupe équivalent) et le groupe phé nyle sont disposés du même côté du plan du noyau oxazoline. Réciproquement,
les compo sés trans ou pseudo sont ceux dont les deux groupes mentionnés ci-dessus sont de part et d'autre du plan des atomes de carbone asymé triques ou du plan du noyau oxazoline.
Chacune des deux formes régulière et pseudo peut exister elle-même sous trois formes optiques: la forme racémique (dl) et deux isomères optiquement actifs: un isomère\' dextrogyre (d) et un isomère lévogyre (1).
Par suite de la difficulté qu'il y a à repré senter ces différences de configuration par des formules graphiques, on utilise ici les for mules - développées ordinaires, une notation placée au-dessous ou à côté de la. formule dé signant la. structure particulière et la confi guration optique du composé. Quand la for mule représente le mélange des isomères géométriques et optiques, on utilise la nota tion mélange .
Il est cependant entendu que si la formule n'est accompagnée d'aucune no tation, cette formule doit être interprétée dans son sens le plus général, c'est-à-dire *qu'elle représente les isomères (1)-y, (d)-v, (1)-rég. et (d)-rég. sous forme séparée, aussi bien que les formes racémiques (dl) -y et (dl) -rég., ainsi que les mélanges des isomères géomé triques et optiques.

L'opération finale d'hydrolyse partielle peut être effectuée en une étape à l'aide d'un agent hydrolysant acide, ou bien en deux étapes en passant par la yr2-m-nitrophényl-4- hydroxyméthyl-5- p - nitr ophényl -J2 - oxazoline comme produit intermédiaire, étant donné que l'hydrolyse du groupe acyle se fait plus faci lement que la scission hydrolytique du noyau oxazoline.. Les corps de départ du procédé selon l'invention peuvent aussi être mis en réaction sous forme d'un de leurs sels,
par exemple sous forme de leurs chlorhydrates.
La nouvelle substance obtenue par le pré sent procédé est cristallisée et sa. forme racé mique fond à l43-144 C.

Pour effectuer la première étape (A), on maintient de préférence la température au- dessous de 50 , avantageusement entre 20 et 35 C. Eta.nt donné que les bases libres des imidoéthers sont souvent assez instables, il est préférable d'utiliser le composé imido-éther -sous forme d'un de ses sels d'un acide miné ral.
L'ester y-hydroxyaminopropionique (ester de la y-phényl-sérine) peut être employé sous forme de base libre ou sous forme d'un sel. Quand on utilise un sel, on ajoute au mélange réactionnel un équivalent d'une substance ba- sique telle que l'ammoniaque aqueuse, le bi carbonate ou l'hydroxyde de sodium pour libérer in situ l'ester yrhydroxyaminopropio- nique sous forme de base. On peut utiliser comme milieu réactionnel une grande variété de solvants tels que l'eau, les alcools aliphati ques inférieurs soit aqueux, soit. anhydres, le chloroforme, l'éther, etc.
La. réduction de la -p-4-carboalcoxy-42- oxazoline en y-4-hydroxyméthyl-42-oxazoline correspondante (étape B) peut être effectuée soit catalytiquement à l'aide d'hydrogène ga- Yeux en présence d'un catalyseur métallique d'hydrogénation, soit au moyen de l'hydrure de lithium et d'aluminium (LiA'H4). Cette dernière méthode de réduction est exécutée dans un solvant organique pratiquement an hydre et.
exempt de groupes hydroxyle, tel que ]'éther, le benzène, le dioxane, le toluène, le tétrachlorure de carbone ou le tétrahydro- furane. Après l'achèvement de la réaction, on décompose l'hydrure d'aluminium et de lithium en excès, par adjonction d'eau et on isole la y-4-hydroxyméthyl-42-oxazoline du mélange réactionnel. La quantité du LiAIH4 employée pour la réduction peut varier dans des limites assez larges. Il est toutefois préfé rable d'employer une quantité légèrement plus grande que la, quantité théoriquement né cessaire.
La transformation de la y-4-hydroxy- méthyl-42-oxazoline en y-4-acyloxyméthyl- 42-oxazoline (étape C) peut être effectuée en traitant la yy-4-hydroxyméthyl-42-oxazoline à l'aide d'un halogénure ou d'un anhydride d'un acide carboxylique. De préférence, on effec tue cette réaction dans des conditions prati quement anhydres en utilisant un excès d'agent d'acylation et un catalyseur basique tel que la pyridine, la triméthylamine, la: diméthyl- aniline, la. N-éthylpipéridine ou une amine tertiaire analogue.
L'acylation peut aussi se faire dans un milieu réactionnel aqueux en employant un catalyseur basique tel qu'un hydroxyde ou un carbonate d'un métal alca lin, ou un des catalyseurs organiques basiques susmentionnés.
La nitration des noyaux phényliques en position 2 et 5 de la. y,-4-acyloxyméthyl-42- oxa7oline (étape D) se fait de préférence en traitant ce composé avec de l'acide nitrique à 1.00 /o, ou avec de l'acide nitrique fumant. ou avec un mélange d'acide nitrique concentré et d'acide sulfurique concentré. On maintient la température pendant. la réaction au-dessous de 25 C et de préférence entre - 10 et + 10 C. On préfère utiliser de l'acide nitri que à 100% ou de l'acide nitrique fumant, étant. donné qu'on obtient avec ces agents des rendements plus élevés en y-2-m-nitrophényl- 5-p-nitrophényle que par l'utilisation d'un mé lange d'acide nitrique et d'acide sulfurique.
L'hydrolyse de la y-2-m-nitrophényl-4- acyloxyméthyl-5-p-nitrophényl-42-oxazoline en le composé y-4-hyroxyméthyle correspondant qui constitue la première étape de l'hydrolyse partielle en deux phases (étape E) est effec tuée à l'aide d'un agent hydrolytique alcalin à une température inférieure à 35 C_ On ob tient les meilleurs résultats en utilisant un milieu réactionnel constitué par ou contenant un solvant organique miscible avec l'eau,
dans le but d'augmenter la solubilité de l'oxazo- line. Quelques-uns des solvants particulière ment appropriés dans ce but sont les suivants: méthanol, éthanol, isopropanol, dioxane, acé tone, méthyléthylcétone et des mélanges aqueux de ces solvants. Comme agents hydro- lytiques alcalins, on peut utiliser les hydroxydes et les carbonates des métaux alcalins ainsi que llyd'roxyde d'ammonium et les hydroxydes des métaux alcalino-terreux.
Ces substances alcalines peuvent être employées telles quelles ou peuvent être engendrées dans le milieu réactionnel par action du solvant sur un agent hydrolytique alcalin. On peut, par exemple, ajouter de l'éthylate de sodium à un milieu réactionnel alcoolique et aqueux pour obtenir de l'hydroxyde de sodium et de l'étha nol par réaction de l'éthylate de sodium avec l'eau présente dans l'alcool aqueux.
L'étape F de l'hydrolyse peut s'effectuer par traitement de la y-2-m-nitrophényl-4- hydroxyméthyl-5-p -nitrophényl -42 -oxa.zoline avec un acide minéral dilué et par neutralisa tion du sel du 1-p-nitrophényl-1-m-nitrobenz- oxy-2-aminopropane-3-ol ainsi obtenu. Après la neutralisation dudit sel, la base libre se transforme spontanément en le y-1-p-nitro- phényl-2-m-nitrobenzamidopropane-1,3-diol dé siré.
La durée préférée du traitement est en viron 1 heure pendant laquelle le mélange ré actionnel est d'à préférence maintenu à une température au-dessous de, 35 C. Si on le dé sire, on peut effectuer la réaction en mainte nant le mélange réactionnel pendant environ 10 à 15 minutes à une température d'environ 60 C. Si on fait durer le' chauffage trop longtemps, une hydrolyse complète en y-1-p- nitrophényl-2-aminopropane-l,3-dioI a lieu, ce qu'il faut éviter. La température préférée pour l'hy drolyse partielle est comprise entre 10 et 30 C.
Si on le désire, on peut encore utiliser un solvant organique miscible à l'eau, pour augmenter la solubilité de l'oxazoline à hydro lyser.
Le procédé selon l'invention est décrit. plus en détail dans l'exemple suivant.: Exemple: 12,3 g de chlorhydrate de (dl)-yp-phényl- sérinate d'éthyle (ester éthylique de l'acide a-amino-,B-hydroxy-fl-phényl-propionique) et 9,3 g de chlorhydrate du benzimido-éthyl-éther sont agités dans 50 cm3 d'éthanol absolu con tenant 3,37 cm3 d'hydroxyde d'ammonium. Un précipité blanc commence à se former qui se dépose presque immédiatement. Après 15 mi nutes, on dilue le mélange réactionnel avec quatre volumes d'eau et on extrait. l'huile for mée à l'aide de deux portions d'éther.
On sèche l'extrait avec du sulfate de magnésium, on sépare l'extrait par décantation et on éva pore. L'huile résiduelle cristallise après repos et est purifiée par recristallisation dans du méthanol; point de fusion: 83 à 84 C. Le pro duit ainsi obtenu est la (dl)-y-2,5-diphényl-4- carbéthoxy-d2-oxazoline.
On dissout 2,95 g de cette substance dans 100 em3 d'éther sec et on ajoute cette solution au cours d'une période d'environ 3 heures à. une solution de 0,2 g de LiAIH4 dans 250 em3 d'éther sec. Ensuite, on brasse le mélange ré actionnel pendant une heure et. on le décom pose par adjonction prudente de 50 ems d'eau. On évapore le mélange réactionnel à siccité et on extrait le résidu soigneusement à l'aide d'éthanol froid absolu. Après évapora tion de l'extrait alcoolique, on obtient un corps solide blanc qui fond après recristallisation dans de l'acétate d'éthyle à 156 C.
Le pro duit ainsi obtenu est la (dl)-y-2,5-diphényl- 4-hydroxy méthyl-d2-oxazoline.
On laisse au repos pendant une nuit, à la température ambiante, un mélange composé de 5 g de cette substance, de 35 cm3 de pyridine sèche et de 8 cms d'anhydride acéti que. On traite ensuite le mélange réactionnel avec de l'eau froide et. on brasse l'huile qui se sépare jusqu'à ce qu'elle cristallise. Le pro duit cristallisé ainsi obtenu est la (dl)-zp-2,5- diphényl-4-acétoxy méthy l-d2-oxazoline.
Le produit brut peut être purifié si on le désire, par recristallisation dans un mélange d'eau et de méthanol.
On ajoute sous agitation 7,5 g de ladite acétoxyméthyl-oxazoline en petites portions à 35 cm-' d'acide nitrique à 100 % en mainte- nant la température en dessous de 10 C.
Après que toute l'oxazoline a été ajoutée, on brasse le mélange réactionnel pendant une demi-heure et on le verse sur de la glace. La <B>(dl)</B> -y-2-m-nitrophény 1-4- acétoxy méthyl - 5-p- nitrophényl-d2-oxazoline insoluble et brute ainsi obtenue est, recueillie et ensuite. purifiée par recristallisation dans de l'éthanol.
On dissout 13 g de ce produit. dans de l'acétone et on ajoute la. solution résultante à 185 em3 d'une solution 0,2N de NaOH et on ajoute encore de l'acétone jusqu'à ce que la solution devienne claire. On maintient le mé lange réactionnel pendant une heure à une température d'environ 20 C et. on neutralise ensuite exactement. à l'aide d'acide chlorhydri que. On élimine l'acétone par distillation dans le vide et on recueille la (dl)-zy-2-m-nitro- phényl-4-hydroxyméthy 1-5-p - nitrophény 1- d2 oxa.zoline cristallisée.
Si on le désire, on peut encore purifier ce produit par recristallisa- tion dans de l'alcool.
On ajoute 5 g de ladite substance à 100 em3 d'acide chlorhydrique 3 N en présence d'une quantité suffisante d'alcool pour per mettre la dissolution. On maintient le mélange réactionnel pendant une heure à une tempé rature de 35 C. On élimine l'alcool par distil lation dans le vide, on filtre le résidu aqueux et on rend alcalin le filtrat à l'aide d'ammo niaque. On recueille le (dl)-y-1-p-nitrophé- nyl-2-m-nitrobenzamidopropane-1,3-diol ainsi précipité et on le purifie par recristallisation dans de l'alcool. Cette substance fond à 143 à 144 C.

where R 1 and R 2 are the same or different and represent hydrogen, a halogen atom, lower alkyl groups or lower alkoxy groups, are. substances which are important either by their therapeutic and antibiotic properties, or as intermediate products for the preparation of other organic substances having antibiotic properties.
It is known that the compounds represented by the preceding formula as well as their starting products and the intermediates for their synthesis can exist in the form of structural isomers (diastereoisomeric forms5) and in the form of optical isomers. By structural isomers is meant the cis and trans derivatives, that is to say the derivatives which are distinguished by the steric position, relative to. the same plane, polar groups attached to the two asymmetric carbon atoms.
To differentiate these two different diastereoisometric forms, the cis compounds will be called regular (reg.), And pseudo (y) the trans compounds. Pis or reg. may also be designated by the prefix erythro or allo, and the trans or pseudo compounds by the prefix threo. Compounds having the cis (or reg.) Configuration
are those in which the two most strongly polar groups are on the same side of the plane in which the two asymmetric carbon atoms are found or of which, in the case of oxazoline intermediates, the group -CH20H (or an equivalent group) and the phenyl group are arranged on the same side of the plane of the oxazoline ring. Reciprocally,
the trans or pseudo compounds are those in which the two groups mentioned above are on either side of the plane of the asymmetric carbon atoms or of the plane of the oxazoline ring.
Each of the two regular and pseudo forms can exist itself in three optical forms: the racemic form (dl) and two optically active isomers: a dextrorotatory isomer (d) and a levorotatory isomer (1).
Because of the difficulty in representing these differences in configuration by graphical formulas, we use here the ordinary expanded formulas, a notation placed below or beside the. formula de signing the. particular structure and optical confi guration of the compound. When the formula represents the mixture of geometric and optical isomers, the notation mixture is used.
It is understood, however, that if the formula is not accompanied by any notation, this formula must be interpreted in its most general sense, that is to say * that it represents the (1) -y isomers, (d) -v, (1) -reg. and (d) -reg. in separate form, as well as the racemic (dl) -y and (dl) -reg. forms, as well as mixtures of the geometric and optical isomers.

The process of the present patent is characterized by reacting an ester of ip - fi - phenyl - fi-hydroxy- α-amino-propionic acid with a benzimido-alkyl-ether to obtain a V-2. , 5-diphenyl-4-carboalkoxy-d2-oxazoline of the formula

in that the ester group of said oxazoline is reduced to form y-2,5-diphenyl-4-hydroxymethyl-d2-oxazoline,
in that this compound is subjected to an acylation of its hydroxyl group to obtain the corresponding acylated compound, in that the latter is treated with a nitration agent under conditions resulting in the production of y @ -2 -m-nitrophenyl-4-acyloxymethyl-5-p-nitrophenyl-d2-oxazoline, and in that this last compound is partially hydrolyzed so as to obtain y-1-p-nit.ro-phenyl-2- m-nitrobenzamido-propane-1,3-diol.
The final partial hydrolysis operation can be carried out in one step using an acidic hydrolyzing agent, or in two steps through yr2-m-nitrophenyl-4-hydroxymethyl-5- p - nitr ophenyl -J2 - oxazoline as an intermediate, since the hydrolysis of the acyl group takes place more easily than the hydrolytic cleavage of the oxazoline ring. The starting substances of the process according to the invention can also be reacted in the form of 'one of their salts,
for example in the form of their hydrochlorides.
The new substance obtained by the present process is crystallized and its. racial form melts at 143-144 C.

In order to carry out the first step (A), the temperature is preferably kept below 50, advantageously between 20 and 35 C. Since the free bases of the imidoethers are often quite unstable, it is preferable to use the imido-ether compound -in the form of one of its salts of a mineral acid.
The γ-hydroxyaminopropionic ester (γ-phenyl-serine ester) can be used as the free base or as a salt. When a salt is used, an equivalent of a basic substance such as aqueous ammonia, bicarbonate or sodium hydroxide is added to the reaction mixture to liberate the hydroxyaminopropionic ester in situ as the base. . A wide variety of solvents can be used as reaction medium such as water, lower aliphatic alcohols, either aqueous or. anhydrous, chloroform, ether, etc.
The reduction of the -p-4-carboalkoxy-42-oxazoline to the corresponding γ-4-hydroxymethyl-42-oxazoline (step B) can be carried out either catalytically using hydrogen gas in the presence of a. metal hydrogenation catalyst, either by means of lithium aluminum hydride (LiA'H4). This latter method of reduction is carried out in an organic solvent which is practically anhydrous and.
free from hydroxyl groups, such as ether, benzene, dioxane, toluene, carbon tetrachloride or tetrahydrofuran. After completion of the reaction, the excess lithium aluminum hydride is decomposed by addition of water and the γ-4-hydroxymethyl-42-oxazoline is isolated from the reaction mixture. The amount of LiAIH4 used for the reduction can vary within fairly wide limits. It is, however, preferable to use an amount slightly greater than the amount theoretically required.
The conversion of y-4-hydroxy-methyl-42-oxazoline to y-4-acyloxymethyl-42-oxazoline (step C) can be carried out by treating yy-4-hydroxymethyl-42-oxazoline with the aid of a halide or anhydride of a carboxylic acid. Preferably, this reaction is carried out under substantially anhydrous conditions using an excess of acylating agent and a basic catalyst such as pyridine, trimethylamine, la: dimethylaniline, la. N-ethylpiperidine or an analogous tertiary amine.
The acylation can also be carried out in an aqueous reaction medium using a basic catalyst such as an alkaline metal hydroxide or carbonate, or one of the aforementioned basic organic catalysts.
The nitration of the phenyl rings in position 2 and 5 of the. y, -4-acyloxymethyl-42-oxa7oline (step D) is preferably carried out by treating this compound with 1.00% nitric acid, or with fuming nitric acid. or with a mixture of concentrated nitric acid and concentrated sulfuric acid. The temperature is maintained for. reaction below 25 C and preferably between -10 and + 10 C. It is preferred to use nitri acid rather than 100% or fuming nitric acid, being. since higher yields of γ-2-m-nitrophenyl-5-p-nitrophenyl are obtained with these agents than with the use of a mixture of nitric acid and sulfuric acid.
The hydrolysis of y-2-m-nitrophenyl-4-acyloxymethyl-5-p-nitrophenyl-42-oxazoline to the corresponding y-4-hydroxymethyl compound which constitutes the first step of the partial hydrolysis in two phases (step E) is carried out using an alkaline hydrolytic agent at a temperature below 35 C_ The best results are obtained using a reaction medium consisting of or containing an organic solvent miscible with water,
for the purpose of increasing the solubility of oxazoline. Some of the solvents particularly suitable for this purpose are as follows: methanol, ethanol, isopropanol, dioxane, acetone, methyl ethyl ketone and aqueous mixtures of these solvents. As alkali hydrolytic agents, there can be used the hydroxides and carbonates of the alkali metals as well as ammonium hydroxide and the hydroxides of the alkaline earth metals.
These alkaline substances can be used as such or can be generated in the reaction medium by the action of the solvent on an alkaline hydrolytic agent. It is possible, for example, to add sodium ethoxide to an alcoholic and aqueous reaction medium to obtain sodium hydroxide and ethanol by reacting sodium ethoxide with the water present in it. aqueous alcohol.
Step F of the hydrolysis can be carried out by treating the y-2-m-nitrophenyl-4-hydroxymethyl-5-p -nitrophenyl -42 -oxa.zoline with a dilute mineral acid and by neutralizing the salt. 1-p-nitrophenyl-1-m-nitrobenz-oxy-2-aminopropan-3-ol thus obtained. After neutralization of said salt, the free base spontaneously transforms into the desired y-1-p-nitro-phenyl-2-m-nitrobenzamidopropane-1,3-diol.
The preferred treatment time is about 1 hour during which the reaction mixture is preferably maintained at a temperature below .35 C. If desired, the reaction can be carried out while maintaining the reaction mixture. for about 10 to 15 minutes at a temperature of about 60 C. If the heating is continued too long, complete hydrolysis to γ-1-p-nitrophenyl-2-aminopropane-1,3-dioI takes place, this that must be avoided. The preferred temperature for partial hydrolysis is between 10 and 30 C.
If desired, an organic solvent miscible with water can also be used to increase the solubility of the oxazoline to be hydrolyzed.
The method according to the invention is described. in more detail in the following example: Example: 12.3 g of ethyl (dl) -yp-phenyl-serinate hydrochloride (a-amino-, B-hydroxy-fl-phenyl acid ethyl ester -propionic) and 9.3 g of benzimido-ethyl-ether hydrochloride are stirred in 50 cm3 of absolute ethanol containing 3.37 cm3 of ammonium hydroxide.
