Verfahren zur Herstellung eines Abkömmlings des ss-Phenylserins. Die Erfindung betrifft ein Verfahren zur Herstellung eines Abkömmlings des ss Phenylserins, nämlich des L-(-f-)-threo-ss-p- Nitrophenylserin-äthylesters, welcher eine neue, als Zwischenprodukt für die Synthese des Chloramphenicols verwendbare Substanz darstellt.
Das erfindungsgemässe Verfahren ist da durch gekennzeichnet, dass man L- (-!- ) -threo- ss-Phenylserin-äthylester mit einem Alkyl- imidoäther zum entsprechenden L-(. )-threo- 2-Alkyl-4-carbäthoxy-5-phenyl-oxazolin um setzt, letzteres nitriert und dann hydroly tiseh zu L-(+)-threo-ss-p-Nitrophenylserin-äthyl- ester aufspaltet.
Der so erhaltene neue Ab kömmling des ss-Phenylserins schmilzt bei <B>1.32</B> bis 135 C und besitzt das optische Dreh- vermögen [a] D =-I-13,3 (c = 0,8 in Metha- nol)

Der für den Aufbau des Chlorampheni- cols benötigte L-(. )-threo-ss-Phenylserin- äthylester wird dadurch gewonnen, dass man DL-threo-fl-Phenylserin am Stickstoff acety- liert, das Chinolinsalz des Acetylderivates ,aus Wasser fraktioniert kristallisiert, die schwerer lösliche Komponente mit Alkali in L-(+)-threo-N-Acetyl-ss-phenylserin spaltet,
anschliessend zum L-(+)-threo-f-Phenylserin entacyliert und verestert. Gemäss vorliegen dem Verfahren wird der L-(+)-threo-f-Phe- n@-lserin-äthylester mit einem Alkylimido- äther, vorzugsweise mit Acetimidoäther, zum L - (+) -threo-2-Alkyl- [resp.-2-#vlethyl-] -4-earb- äthoxy-5-phenyloxazolin umgesetzt, letzteres nitriert und anschliessend zum L-(+)
-tlireo- fl-p-Nitrophenylserin-äthylester hydroly siert.
Der gemäss der Erfindung erhaltene, bis her unbekannte Stoff ist eine beständige, kri stallisierte Verbindung von basischem Cha rakter. Der Ester ist in den meisten orga- nisehen Lösungsmitteln leicht, in Wasser oder Petroläther dagegen schwer löslich.
Mit )lineralsäur en bildet er beständige Salze; durch Verseifung geht er in L-tlireo-f-p- NitrophenyIserin über; nach Acylierung der Aminogruppe kann die neue Verbindung mit Lithiiimborhydrid zum D-threo-l-p-Nitro- phenyl-2-acylamino-1,3-propandiol reduziert werden, woraus durch Abspaltung des Acyl- restes das bekannte D-(-)-threo-l-p-Nitro- phenyl-2-amino-1,3-propandiol gewonnen wer den kann.
<I>Beispiel:</I> Man suspendiert 135 Gewichtsteile Acet, imidoätherhydrochlorid in 460 Rahmteilen Benzol und versetzt mit einer eiskalten Lö sung von 46 Gewichtsteilen Natriumhydroxyd in 200 Raumteilen Wasser, schüttelt gut durch und trennt die benzolisehe Lösung des Acetimidoäthers ab. Nun gibt man auf ein mal 200 Gewichtsteile L-(+)-threo-fl-Phenyl- serin-äthylester-hydroehlorid zur vorigen ben- zolisehen Lösung und rührt während 12 Stun den.
Dann gibt man 200 Raumteile Wasser zur Auflösung des abgeschiedenen Ammon- chlorids hinzu, trennt die benzolische Lösung ab und wäscht sie zweimal mit je 100 Raum teilen Wasser, trocknet sie über Natrium sulfat und dampft das Benzol im Vakuum ab. Der Rückstand, das L-(+)-tlireo-2-Methyl- 4-carbäthoxy-5-phenyl-oxazolin destilliert im Hochvakuum (0,15 mm) bei 118 C; [a] = + 214 (e = 2 in Methanol). Ausbeute 170 Gewichtsteile.
30 Gewichtsteile L-(-i-)-threo Z-Methyl- 4-carbäthoxy-5-pheny 1-oxazolin werden in 90 Raumteilen Chloroform gelöst und auf -15 C gekühlt. Unter Rühren wird ein Ge misch von 80 Raumteilen HNO3 (100 % ig) und 50 Raumteilen Chloroform so zugetropft, da.ss die Temperatur von -10 C nicht über schritten wird. Nun wird bei -10 C noch 6 Stunden weitergerührt.
Das Nitriergemisch wird darnach langsam in ein Gemisch von 200 Raumteilen Ammoniak (251/aig) und etwa 400 Gewichtsteilen Eis gegossen, die Chloroformlösung abgetrennt und die wäss- rige Phase mit 100 Rahmteilen Chloroform extrahiert. Die Chloroformauszüge werden wit 200 Raumteilen Wasser gewaschen und im Vakuum eingedampft.
Man erhält etwa 35 Gewiehtsteile rohes Nitrierprodukt. Dieses wird in 200 Raumteilen äthylalkoholischer Salzsäure und ?. Raumteilen Wasser gelöst, 3 ,Stunden unter Rüekflusskühlung gekocht und anschliessend im Vakuum eingedampft. Das Chlorhydrat des rohen Nitrophenylserin- esters wird in 200 Raumteilen Chloroform durch Erwärmen gelöst, abgekühlt, worauf man so lange gasförmigen Ammoniak einlei tet, bis die Lösung schwach phenolphthalein- alkalisch reagiert.
Nun wird wieder erwärmt und heiss vom Ammonehlorid abfiltriert. Beim Stehen bei 0 C scheidet sich der L - (+) - threo-ss-p - Nitrophenylserin - äthylester in Nadeln aus; nach Abnutschen, Auswaschen und Trocknen erhält. man so als erste Portion 1.0 bis 12 Gewiehtsteile vom Schmelzpunkt 132 bis 13j5 C; [a-120 =+133o (c=0,8 in CH3011).
Process for the preparation of a derivative of ss-phenylserine. The substance obtained according to the invention, which was previously unknown, is a stable, crystallized compound of basic character. The ester is easily soluble in most organic solvents, but sparingly soluble in water or petroleum ether.
With lineral acids it forms stable salts; by saponification it changes into L-tlireo-f-p-nitrophenylserine; after acylation of the amino group, the new compound can be reduced with lithium imborohydride to give D-threo-lp-nitrophenyl-2-acylamino-1,3-propanediol, from which the well-known D - (-) - threo- lp-nitrophenyl-2-amino-1,3-propanediol can be obtained.
<I> Example: </I> 135 parts by weight of acet, imido ether hydrochloride are suspended in 460 parts of cream of benzene and an ice-cold solution of 46 parts by weight of sodium hydroxide in 200 parts by volume of water is added, shaken well and the benzene solution of the acetimido ether is separated off. Now add 200 parts by weight of L - (+) - threo-fl-phenyl-serine-ethyl ester-hydrochloride to the previous benzene solution and stir for 12 hours.
Then 200 parts by volume of water are added to dissolve the separated ammonium chloride, the benzene solution is separated off and washed twice with 100 parts by volume of water each time, dried over sodium sulfate and the benzene is evaporated off in vacuo. 30 parts by weight of L - (- i -) - threo Z-methyl-4-carbethoxy-5-pheny-1-oxazoline are dissolved in 90 parts by volume of chloroform and cooled to -15.degree. Now it is heated again and the ammonium chloride is filtered off while hot. When standing at 0 C, the L - (+) - threo-ss-p - nitrophenylserine - ethyl ester separates out in needles; obtained after suction filtering, washing out and drying. The first portion is 1.0 to 12 parts by weight with a melting point of 132 to 13.5 C; [a-120 = + 133o (c = 0.8 in CH3011).
