vorzugsweise von Alkalialkoholaten, umsetzt. Die erhaltenen 3-Amino-thiophen-2-carbonsäureester können in üblicher Weise verseift werden.
Das Verfahren gemäss der Erfindung ermöglicht in gegenüber dem bekannten Stand der Technik sehr einfacher Weise die Herstellung von Thiophenderi- vaten. Das Gelingen der an sich eigenartigen Reak tion war nicht zu erwarten. Sie kommt nur dadurch zustande, dass die primär als Ringschlussprodukte entstehenden Ketimine durch die Ausbildung des Thiophenringsystems sofort zu den entsprechenden Aminen stabilisiert werden.
Die als Ausgangsmaterial verwendeten a,ss-Di- halogennitrile lassen sich leicht aus den entsprechen den a,ss-ungesättigten Nitrilen, wie Acrylnitril, Cro- tonnitril, Zimtnitril und ähnlichen, durch Anlage rung von Halogen gewinnen. Sie können auch aus in a-Stellung substituierten a,ss-ungesättigten Nitri- len, z.
B. a-Methylacrylnitril, hergestellt werden. Als Ausgangsstoffe seien beispielsweise erwähnt: a,ss-Dichlor-propionitril, a,ss-Dibrom-propionitril, a,ss-Dichlor-butyronitril, a,ss-Dibrom-butyronitril, a,ss-Dichlor-isobuttersäurenitril, a,ss-Dichlor-valeriansäurenitril, a,ss-Dichlor-capronsäurenitril, a,ss-Dichlor-önanthsäurenitril, a,ss-Dichlor-hydrozimtsäurenitril, a,ss-Dichlor-(p-methyl)-hydrozimtsäurenitr il, a,ss-Dichlor-(o-methoxy)
hydrozimtsäurenitril und a,ss,p-Trichlor-hydrozimtsäurenitril.
Als Ester der Thioglykolsäure kommen insbeson dere solche mit niedrigen aliphatischen Alkoholen in Frage.
Die Umsetzung führt man zweckmässig so durch, dass man den Thioglykolsäureester mit einem alkali schen Kondensationsmittel, vorzugsweise mit einem Alkalimetallalkoholat, in die entsprechende Alkali verbindung überführt und diese dann, zweckmässig unter Kühlung mit dem a,ss-Dihalogennitril umsetzt.
Unter Halogenwasserstoffabspaltung erfolgt gleich zeitig Kondensation zum 3-Amino-thiophen-2-carbon- säureester. Dieser kann nach dem Neutralisieren mit einem organischen Lösungsmittel ausgeschüttelt und durch Destillation oder Umkristallisieren gereinigt werden.
Die 3-Amino-thiophen-2-carbonsäureester be sitzen einen angenehmen Geruch, der an denjenigen der Anthranilsäureester erinnert. Sie lassen sich in wässriger Lösung diazotieren und kuppeln bzw. der Reaktion nach Sandmeyer unterwerfen. Durch Verseifung der erhaltenen 3-Aminothio- phen-2-carbonsäureester in üblicher Weise erhält man die entsprechenden freien Carbonsäuren.
Die Verfahrenserzeugnisse stellen wertvolle Zwi schenprodukte dar, die sich nach mannigfachen Re aktionen in Farbstoffe und Pharmazeutika über führen lassen.
<I>Beispiel 1</I> 3-Amino-thiophen-2-carbonsäuremethylester Eine Suspension von 29,2 g alkoholfreiem Na- triummethylat in 250 cm3 Äther wird unter Kühlung und Rühren mit einer Lösung von 31,8 g Thio- glykolsäuremethylester in 30 cm3 Äther versetzt. Dem Reaktionsgemisch tropft man im Verlauf von 11/2 Stunden 24,8 g a,ss-Dichlor-propionsäurenitril in 30 cm3 Äther zu und lässt das Rührwerk noch wei tere 30 Minuten laufen.
Das ockerfarbene Reak tionsgemisch wird mit Wasser versetzt, mit Essig säure schwach angesäuert und die ätherische Schicht abgetrennt. Die wässrige Schicht wird mehrmals mit Äther extrahiert, bis sie mit einem mit Salzsäure angefeuchteten Fichtenspan keine Orangefärbung mehr gibt. Nach dem Trocknen wird der Äther ab destilliert und der Rückstand im Vakuum fraktio niert. Man erhält den 3-Amino-thiophen-2-carbon- säuremethylester als hellgelbes Öl vom Kp<B>0,1</B> 100 bis 102 , das beim Stehen erstarrt. Aus Methanol bekommt man farblose Nadeln vom Schmelzpunkt 65,5 .
Die Ausbeute beträgt 72%0 der Theorie. Die Acetylaminoverbindung bildet farblose Prismen vom Schmelzpunkt 100 (Methanol).
Die Benzoylaminoverbindung bildet farblose Blättchen vom Schmelzpunkt 103 (Methanol).
3,9 g 3-Amino-thiophen-2-carbonsäuremethylester werden mit 20 cm3 4n wässriger Natronlauge zwei Stunden unter Rückfluss erhitzt. Nach dem Erkalten wird mit verdünnter Salzsäure angesäuert, wobei die 3-Amino-thiophen-2-carbonsäure als dicker farbloser Niederschlag ausfällt.
Die Verbindung kristallisiert aus Benzol in Form farbloser Nadeln, die sich ab 83 zersetzen. Die Ausbeute beträgt 3,4 g (96% der Theorie).
<I>Beispiel 2</I> 5-Methyl-3-amino-thiophen-2-carbonsäure- methylester Entsprechend der in Beispiel 1 angegebenen Vor schrift werden 38,1 g Thioglykolsäuremethylester, 29,2g Natriummethylat und 27,6 g a,ss-Dichlor- butyronitril umgesetzt und aufgearbeitet. Der nach dem Verdampfen des Äthers erhaltene Rückstand erstarrt beim Stehen. Durch Umkristallisieren aus Methanol erhält man den 5-Methyl-3-aminothiophen- 2-carbonsäuremethylester in Form farbloser Prismen vom Schmelzpunkt 72 .
Die Ausbeute beträgt 29 g (85 % der Theorie). Die Acetylaminoverbindung bildet farblose Blätt chen vom Schmelzpunkt 90 (Methanol). Die Benzoylaminoverbindung bildet farblose Na deln vom Schmelzpunkt 145,5-146,5 (Methanol).
4,3 g 5-Methyl-3-amino-thiophen-2-carbonsäure- methylester werden wie unter Beispiel 1 angegeben verseift. Man erhält 3,6 g (91 Ih. der Theorie) 5-Me- thyl-3-amino-thiophen-2-carbonsäure, die aus Benzol in Form schwach gelblicher Nadeln kristallisiert (Zersetzung ab 85 ).
<I>Beispiel 3</I> 5-Phenyl-3-amino-thiophen-2-carbonsäure- methylester Entsprechend der in Beispiel 1 angegebenen Vorschrift wird das aus 47,7 g Thioglykolsäure- methylester, 43,7 g Natriummethylat und 60 g a,ss-Dichlor-hydrozimtsäurenitril erhaltene Reak tionsgemisch umgesetzt und aufgearbeitet.
Der nach dem Zersetzen mit Essigsäure erhaltene Niederschlag wird abfiltriert, die wässrige Phase mehrmals mit Me- thylenchlorid extrahiert und der Extrakt mit der Ätherlösung vereinigt.
Der nach dem Trocknen und Verdampfen der organischen Lösungsmittel verblei bende Rückstand wird mit dem oben erhaltenen Nie- derschlag aus 50%iger Essigsäure bzw. Methanol umkristallisiert. Man erhält so den 5-Phenyl-3-amino- thiophen-2-carbonsäuremethylester in Form schwach gelblicher Nadeln vom Schmelzpunkt 150-151 .
Die Ausbeute beträgt 46 g (66% der Theorie).
Die Acetylaminoverbindung bildet schwach gelb liche Nadeln (Methanol); Schmelzpunkt 124-125 . Die Benzoylaminoverbindung bildet schwachgelb liche Nadeln (Methanol); Schmelzpunkt 165-166 . 5,8 g 5-Phenyl-3-amino-thiophen-2-carbonsäure- methylester werden mit 20 cm3 Methanol und 20 cm3 4n Natronlauge drei Stunden unter Rück fluss erhitzt. Nach dem Erkalten wird das ausgeschie dene Natriumsalz der Säure abfiltriert und in ver dünnte Salzsäure eingetragen.
Dabei fällt die 5-Phenyl-3-amino-thiophen-2-carbonsäure als dicker farbloser Niederschlag aus. Aus Chloroform erhält man schwach gelbliche Nadeln, die sich ab 94 langsam zersetzen. Die Ausbeute beträgt 5,1 g (94 /o der Theorie). <I>Beispiel 4</I> 3-Amino-thiophen-2-carbonsäureäthylester Entsprechend der in Beispiel 1 angegebenen Vor schrift wird das Reaktionsgemisch aus 30g Thio- glykolsäureäthylester, 27 g alkoholfreiem Natrium- äthylat und 24,8 g a,/3-Dichlor-propionitril umgesetzt und aufgearbeitet.
Der 3-Amino-thiophen-2-carbon- säureäthylester geht bei Kp 0.3 109-112 als schwach gelbes Öl über, das beim Abkühlen erstarrt. Aus Äthanol erhält man schwach gelbliche würfelförmige Kristalle vom Schmelzpunkt 42-43 . Die Ausbeute beträgt 15,2 g (44% der Theorie).
Durch Verseifung mit 4n Natronlauge, wie in Beispiel 1 angegeben, erhält man in 90"/0iger Aus beute die dort beschriebene 3-Amino-thiophen-2- carbonsäure. <I>Beispiel 5</I> 5-Methyl-3-amino-thiophen-2-carbonsäure- äthylester Entsprechend der in Beispiel 1 angegebenen Vor schrift wird das Reaktionsgemisch aus 22,3g Thio- glykolsäureäthylester, 20 g Natriumäthylat und 20,5 g a,ss-Dichlor-buttersäurenitril umgesetzt und aufge arbeitet.
Der erhaltene 5-Methyl-3-amino-thiophen- 2-carbonsäureäthylester geht bei Kp<B>0,3</B> 120-124 als schwach gelbes Öl über, das sofort erstarrt. Aus Äthanol erhält man farblose Nadeln vom Schmelz- punkt 59-60 . Die Ausbeute beträgt 41% der Theorie.
Bei der Verseifung, wie unter Beispiel 1 ange- geb; n, entsteht in 91 o/oiger Ausbeute die in Beispiel 2 beschriebene 5-Methyl-3-aminothiophen-2-carbon- säure.
<I>Beispiel 6</I> 5-Phenyl-3-amino-thiophen-2-carbonsäureäthyl- ester Entsprechend der in Beispiel 1 angegebenen Vor schrift wird das Reaktionsgemisch aus 30 g Thio- glykolsäureäthylester, 27 g alkoholfreiem Natrium- äthylat und 40 g a,/3-Dichlor-hydrozimtsäurenitril in 300 cm- Äther umgesetzt und aufgearbeitet. Der 5-Phenyl - 3 - amino-thiophen-2-carbonsäureäthylester kristallisiert aus Äthanol in farblosen verfilzten Na deln vom Schmelzpunkt 104-105 .
Die Ausbeute be- trägt 27 g (55 % der Theorie). <I>Beispiel 7</I> 3-Amino-4-methyl-thiophen-2-carbonsäure- methylester 29,2 Gewichtsteile alkoholfreies Natriummethylat, in 300 Gewichtsteilen absolutem Äther suspendiert, werden in der in Beispiel 1 beschriebenen Weise mit 26,5 Gewichtsteilen Thioglykolsäuremethylester unter Kühlen umgesetzt.
Bei einer Innentemperatur von 10-15 lässt man zu dieser Mischung eine Lö sung von 27,6 Gewichtsteilen a,/3-Dichlor-isobutter- säurenitril in 30 cm3 Äther zutropfen und rührt noch eine Stunde weiter. Dann werden 8 em3 Eis essig und anschliessend 200 cm., Wasser zugegeben, die wässrige Schicht abgetrennt und mehrmals mit Äther ausgeschüttelt. Die vereinigten ätherischen Lö sungen werden mit Sodalösung und Wasser ge- waschen und getrocknet.
Beim Einleiten von trocke nem Chlorwasserstoffgas kristallisiert das Hydro- chlorid des 3-Amino-4-methyl-thiophen-2-carbon- säuremethylesters aus. Schmelzpunkt 78 , Ausbeute 6311/o der Theorie.
Durch Behandeln mit warmer Soda- oder Na triumacetatlösung erhält man den freien Ester. Er bildet aus Methanol umkristallisiert farblose Nadeln vom Schmelzpunkt 84-85 .
Acetylverbindung: farblose Nadeln (aus Me thanol) Schmelzpunkt 118 .
Benzoylverbindung: farblose Spiesse (aus Me thanol) Schmelzpunkt 105 .
Durch Verseifung in der in Beispiel 1 angege benen Art erhält man in 839/oIger Ausbeute die 3-Amino-4-methyl-thiophencarbonsäure in Form farbloser Nadeln vom Schmelzpunkt 123 (Essig ester).

preferably of alkali alcoholates. The 3-amino-thiophene-2-carboxylic acid esters obtained can be saponified in a customary manner.
The process according to the invention enables thiophene derivatives to be prepared in a very simple manner compared to the known prior art. The success of the reaction, which is peculiar in itself, was not to be expected. It only comes about because the ketimines, which are primarily formed as ring closure products, are immediately stabilized to the corresponding amines through the formation of the thiophene ring system.
The α, β-dihalogenonitriles used as starting material can easily be obtained from the corresponding α, β-unsaturated nitriles, such as acrylonitrile, crononitrile, cinnamonitrile and the like, by adding halogen. You can also choose from α, β-unsaturated nitriles substituted in the α-position, eg.

Suitable esters of thioglycolic acid are in particular those with lower aliphatic alcohols.
The reaction is conveniently carried out by converting the thioglycolic acid ester into the corresponding alkali compound with an alkali condensation agent, preferably with an alkali metal alcoholate, and then reacting this with the α, ß-dihalogenonitrile, advantageously with cooling.
With elimination of hydrogen halide, condensation takes place at the same time to give the 3-amino-thiophene-2-carboxylic acid ester. After neutralization, this can be shaken out with an organic solvent and purified by distillation or recrystallization.
The 3-amino-thiophene-2-carboxylic acid esters have a pleasant odor that is reminiscent of that of the anthranilic acid esters. They can be diazotized and coupled in aqueous solution or subjected to the Sandmeyer reaction. <I> Example 1 </I> 3-Amino-thiophene-2-carboxylic acid methyl ester A suspension of 29.2 g of alcohol-free sodium methylate in 250 cm3 of ether is mixed with a solution of 31.8 g of thioglycolic acid methyl ester in 30 cm3 of ether added. 24.8 g of α, β-dichloropropionic acid nitrile in 30 cm3 of ether are added dropwise to the reaction mixture in the course of 11/2 hours and the stirrer is allowed to run for a further 30 minutes.
The ocher-colored reaction mixture is mixed with water, acidified weakly with acetic acid and the ethereal layer is separated. The aqueous layer is extracted several times with ether until it no longer turns orange with a spruce chip moistened with hydrochloric acid. After drying, the ether is distilled off and the residue is fractionated in vacuo. The 3-amino-thiophene-2-carboxylic acid methyl ester is obtained as a pale yellow oil with a boiling point of 0.1 100 to 102 which solidifies on standing. Colorless needles with a melting point of 65.5 are obtained from methanol.
The yield is 72% of theory. The acetylamino compound forms colorless prisms with a melting point of 100 (methanol).
The benzoylamino compound forms colorless flakes with a melting point of 103 (methanol).
3.9 g of 3-amino-thiophene-2-carboxylic acid methyl ester are heated under reflux for two hours with 20 cm3 of 4N aqueous sodium hydroxide solution. After cooling, it is acidified with dilute hydrochloric acid, the 3-amino-thiophene-2-carboxylic acid precipitating out as a thick, colorless precipitate.
The compound crystallizes from benzene in the form of colorless needles, which decompose from 83 onwards. The yield is 3.4 g (96% of theory).
<I> Example 2 </I> 5-Methyl-3-aminothiophene-2-carboxylic acid methyl ester According to the instructions given in Example 1, 38.1 g of thioglycolic acid methyl ester, 29.2 g of sodium methylate and 27.6 ga, ss -Dichlorobutyronitrile implemented and worked up. 4.3 g of 5-methyl-3-aminothiophene-2-carboxylic acid methyl ester are saponified as indicated in Example 1. 3.6 g (91 hours of theory) of 5-methyl-3-aminothiophene-2-carboxylic acid are obtained, which crystallizes from benzene in the form of pale yellowish needles (decomposition from 85).
<I> Example 3 </I> 5-Phenyl-3-aminothiophene-2-carboxylic acid methyl ester According to the procedure given in Example 1, the mixture is prepared from 47.7 g of methyl thioglycolate, 43.7 g of sodium methylate and 60 ga , ß-dichlorohydrozinnonitrile reac tion mixture obtained implemented and worked up.

The 5-methyl-3-aminothiophene-2-carboxylic acid ethyl ester obtained passes over at boiling point 0.3 120-124 as a pale yellow oil which solidifies immediately. Colorless needles with a melting point of 59-60 are obtained from ethanol. The yield is 41% of theory.
For the saponification, as stated under Example 1; n, the 5-methyl-3-aminothiophene-2-carboxylic acid described in Example 2 is obtained in 91% yield.
<I> Example 6 </I> 5-Phenyl-3-aminothiophene-2-carboxylic acid ethyl ester According to the instructions given in Example 1, the reaction mixture of 30 g of thioglycolic acid ethyl ester, 27 g of alcohol-free sodium ethylate and 40 ga, / 3-dichloro-hydrocinnamonitrile reacted in 300 cm ether and worked up. By saponification in the manner indicated in Example 1, the 3-amino-4-methyl-thiophenecarboxylic acid is obtained in 839% yield in the form of colorless needles with a melting point of 123 (ethyl acetate).
