Depuis cette époque, d'autres méthodes ont été préconisées pour la préparation de ces laetones et notamment les suivantes: cr) l'oxydation des cétones cycliques (Ru- zieka et Stol1, Helvetica Chimica Acta, 1928, <B>11,</B> 1159; voir également le brevet français :V 657971);
b) la cyclisation interne d'oxyacides en milieu très dilué (brevet français N 773651 et certificat d'addition N 46623); c) la dépolymérisation des polyesters cor respondants (brevets français Nos 768207 et 796410); d) le traitement de certains acides gras halogénés en milieu alcalin et en présence de certains solvants à bas point d'ébullition (bre vet français N <B>8391.88</B> de 13unsdiecker, Er lach et Vogt);
EMI0002.0028
(III <SEP> Ri-C00-R_C00--R.2 <SEP> rR_<U>_COCh</U> <SEP> + <SEP> Rr-COO-R2
EMI0002.0051
à <SEP> la <SEP> formule <SEP> r(Cgz)is
Chim. Acta, Vol. 11, 1171 [1928] ).
Process for preparing a macrocyclic lactone. Monocyclic laetones having more than 12 members in the lactonic ring have been extensively studied by Iierschbaum. (German patent N 449217 and Ber. der dent ... Chem. Ces. 1927, volume 60, pages 902 and following), which showed that they could not. not be obtained by ordinary esterification methods, as such methods give. polymers of no interest.
The special process described by herschbaum consists in heating, under special conditions, the silver salts of balogenated fatty acids in which the halogen atom is at least in position 12 relative to the carboxyl group; three musky-odor lactones were thus prepared: pentadecanolide-7., 15, hexadecanolide-1,16 and hexadecene-7-olide-1,16, but the yields obtained are not indicated.
Since that time, other methods have been recommended for the preparation of these laetones and in particular the following: cr) the oxidation of cyclic ketones (Ruzieka and Stol1, Helvetica Chimica Acta, 1928, <B> 11, </ B> 1159; see also French patent: V 657971);
b) the internal cyclization of oxyacids in a very dilute medium (French patent N 773651 and certificate of addition N 46623); c) depolymerization of the corresponding polyesters (French patents Nos. 768207 and 796410); d) the treatment of certain halogenated fatty acids in an alkaline medium and in the presence of certain low-boiling point solvents (French patent N <B> 8391.88 </B> of 13unsdiecker, Er lach and Vogt);
e) heating an ester of an osyacid in the presence of a functional group exchange catalyst (French patent: ais N 830689 and certificate of addition N 49302), etc.
The practical implementation of these methods has various drawbacks: extreme dilution, use of a very high vacuum, need to use special devices, use of uncommon reagents, great slowness of reactions, deterioration of the products obtained, etc. It should be added that the yields achieved are not always satisfactory and that the quality of the lactones obtained often leaves. desire, which is a disadvantage. particularly serious for products intended for perfumery.
The present invention, due to 11. Crabalona, relates to a new process for the preparation of clique inacrocy lactones.
In this process, conventional washing apparatus and common reagents are used; in addition, these lactones are obtained in a very high degree of purity and in excellent yields.
This process is based on an internal double decomposition reaction of certain esters in the presence of catalysts which promote the exchange of functional groups. A double decomposition reaction of this kind for the preparation of organic esters has been provided for in US Pat. No. 1,860,092.
In this patent, it is said that an ester R4-C00-R3 can be prepared by interreaction of the two esters R'-COOR3 and R-1-C00-R "in the presence of an exchange catalyst. dicals, RI-C00 and R4-C00 being anions of carboxylic acids, R '"and Ra being alkyl radicals: the balanced reaction is as follows:
(1) RI-C00-Rs + R4-COO-R2 E Rr-C00-R2 + R4-C00-R3 It is from this equation (1) that we can best understand the mechanism of the process novel which is the subject of the present invention.
For this purpose, let us consider a diester derived from an omega-hydroxylated acid, corresponding to the following formula: Rr-COOR-COOR2 in which R is a chain of at least 11 carbon atoms.
This radical R can be formally decomposed into two parts R3 and R4, each representing chains or greens containing at least 5 carbon atoms; the above diester can therefore be written: P, 1 - COOR3-R4-COOR2 Thus represented, it presents a striking analogy with the equimolecular mixture of esters RI-C00-R3 and R4-C00 - R2 The experiment showed that under certain conditions that will be.
specified later, this body behaves exactly like such a mixture by supplying the lactone
EMI0002.0024
and the ester Rr-C00-R2 according to the balanced equation:
EMI0002.0026
that we can write, by re-establishing R = R3 + R4, in the following form:
EMI0002.0028
(III <SEP> Ri-C00-R_C00 - R.2 <SEP> rR_ <U> _COCh </U> <SEP> + <SEP> Rr-COO-R2
<tb> lactone If Rr and R "are lower alkyl groups, the ester obtained RI-C00 R2 is very volatile, it therefore escapes from the reaction medium, the equilibrium is upset and reaction (II) takes place. continues from left to right giving the desired lactone.
The <B> '</B> * experiment also showed that reaction (II) could be carried out in the same way if the carboxylic anion R1-C00 were replaced by a mineral anion AO-, susceptible of give a very volatile ester with the alcohol R2-OH; the equation becomes. in that case
EMI0002.0045
or, which amounts to the same:
EMI0002.0046
AO-R-C00-R2 <SEP> @ -R-C00 - @ <SEP> + <SEP> AO-R2
<tb> (lactone) The present patent relates to a process for preparing one of these ma.ero-cyclic lactones, hexadecanolide-1,16, corresponding
EMI0002.0051
at <SEP> the <SEP> formula <SEP> r (Cgz) is
<tb> LCO-0] already described by Ruzicka and Stoll, Helv. Chim. Acta (1928), 11, <B> 1170 </B> and by Stoll and Rouve, Helv. Chim. Acta (1935), 18, 1105.
This process is. characterized by the decomposition, in the presence of a radical exchange catalyst, of a diester, derived from an omega-hydroxy acid, of general formula: XO- (CHz) 15-CO-OR in which R denotes a alkyl group and X a residue of an acid X-OH, with the formation, on the one hand, of the desired lactone
EMI0003.0008
and, on the other hand, of the X-OR ester, which is volatile under the reaction conditions.
The X radical can be either organic or inorganic. As radicals of organic acids, those of formic, acetic, propionic, ete. can be used.
The mineral acid radicals used will preferably be those of boric, carbonic or sulfuric acids.
R is preferably a lower alkyl radical, such as methyl, ethyl, propyl, butyl.
In any case, the choice of the acid and the alcohol must be made such that the ester obtained X-OR is volatile under the reaction conditions.
The decomposition of the above diester takes place under the influence of heat in the presence of a conventional radical exchange catalyst, and preferably under reduced pressure, in order to remove the volatile X-OR ester formed. , so that the reaction (II) can progress rapidly from left to right and thus provide, with the best yield and minimum heating, the desired monomeric lactone.
As catalysts, in this reaction of exchange of functional groups which is the basis of the claimed process, a body known for its properties of radical exchange catalyst will be employed; Without this list being able to be considered as limiting, the following two families are more particularly recommended, on the one hand, sodium derivatives of alcohols or of mono- or polyvalent phenols; on the other hand, more or less ionized acid reactants, such as sulfuric or phosphoric acids, organic acids, arylsulfonic acids, & c.
Of course, the proportion of the catalyst is not taken into account and will be determined in each case according to the activity of the catalyst and the reaction rate that is to be obtained; the present invention is therefore not linked to the use of a specific catalyst.
As for the temperature and the pressure, they will be chosen at the discretion of the manipulator according to the nature of the constituents of the equilibrium and their volatility.
Two examples of the implementation of this process are given below <I> Example 1: </I> 21 g of methyl acetyloxy-16-hexadecanoate are heated to 220-230 (temperature of the heating bath) , under a pressure of 3 mm of mercury, in the presence of sodium methoxide. In an hour and a half, <B> 11.6 </B> g of distillate are collected in the form of a colorless oil which crystallizes in the receiving vessel.
Yield: 71.3% of the theoretical yield of lactone. This distillate has the following characteristics: fully distils between 127 and 129 under a pressure of 2.5 mm of mercury; solidification point: + 38 5; density d47 = 0.9324; refractive index nD = 1.4662; saponification index: 221.5.
Except for experimental error, these constants are those of pure hexadecanolide-1,16, indicated in the literature (Helv. Chim. Acta, Vol. 11, 1171 [1928]).
<I> Example 2: </I> 25.7 g of butyl hexadecanol-16-oate are converted into boric ester by means of 1.56 g of boric acid. The water formed being driven off, the diester is heated under a pressure of 3 mm of mercury in the presence of sodium methylate. The heating bath being at 180-190, a colorless liquid, very mobile, distils. We collect it; its weight is 5.05 g. It is butyl borate, because it completely distils at 122-124 under a pressure of 24 mm of mercury and at 97-99 under a pressure of 6 mm; refractive index: 1117 = 1.1120; water hydrolyzes it to butanol and boric acid.
Heating is continued under vacuum and, when the temperature of the heating bath reaches 230-240, 13.1 g of distillate are collected in two hours, the constants of which are practically identical to those of the laetone obtained according to example 1.
Yield: 64.81% of the theoretical yield.
