Verfahren zur Herstellung eines Dibenzopyran-derivates. Gegenstand des vorliegenden Patentes ist ein Verfahren zur Herstellung von 6"-Oxy-2,2,5'- trimet.hyl-4"-n-amyl-dibenzopyran, dadurch gekennzeichnet, dass man, ausgehend von 5-Acyloxy-5'-methyl-7-n-amyl-3,4-cyclohexeno- cumarin, einerseits ein Methylmagnesium- halogenid einwirken lässt -Lind das Reaktions produkt hydrolysiert und dass man ander seits dehydriert.
Bei der Einwirkung des Methy lmagnesium- halogenids und der nachfolgenden Hydrolyse wird der Carbony lsauerstoff in 2-Stellung des Cumarins durch zwei Methylgruppen ersetzt und ferner die 5-Acyloxygruppe in eine freie Oxygruppe umgewandelt.

Die Ausgangsverbindung, nämlich das 5-Acyloxy-5'-methyl-7-n-amyl-3,4-cyclohexeno- cumarin kann gewonnen werden durch Kon densation von Olivetol-monohydrat mit Me- thylcyclohexan - 3 - an -4- carbonsäureäthylester in Gegenwart von konzentrierter Schwefel säure und anschliessende Acylierung. Vorteil hafterweise wird die Acetylverbindung als Ausgangssubstanz
verwendet.
Das Verfahrenserzeugnis erwies sich als identisch mit Cannabinol. Auch das als Zwi schenprodukt erhaltene Tetrahydrocannabinol (6"- Oxy-2,2,5'-,trimethyl-4"-n-amyl-3',4',5',6'- tetrahydro-dibenzopyran) zeigt starke physio logische Wirkungen, wie sie für Haschisch charakteristisch sind.
Das Verfahrenserzeug nis soll als Arzneimittel Verwendung finden. BeispieZ'1: 5-Oxy-5'-methyl-7-n-amyl-3,4-cyclohexeno- cumarin wird mit Essigsäureanhydrid in Gegenwart von Pyridin am Rückflusskühler erhitzt. Das erhaltene 5-Acetoxy-5'-methyl-7- n-amyl-3,4-cyclohexeno-cumarin bildet farb lose Prismen vom Schmelzpunkt 82 bis 830 C.
Eine Lösung von 44 Gewichtsteilen der ge nannten Verbindung in 600 Raumteilen Ani- sol wird bei Zimmertemperatur langsam einer Lösung von Methylmagnesitunjodid (aus 100 Gewichtsteilen Methyljodid und 17 Gewichts teilen Magnesium) in Anisol zugesetzt.
Nach Beendigung der Zugabe wird die Mischung während 8 Stunden auf dem Dampfbade er hitzt; es entsteht der Magnesiumjodidkomplex des 6"-Oxy-2,2,5'-trimethyl-4"-n-amyl-3',4',5', 6'-tetrahydro-dibenzopyrans. Die Mischung wird gekühlt und zur Hydrolyse des Kom plexes mit Eis und verdünnter Schwefelsäure versetzt, worauf das Anisol durch Dampf destillation entfernt wird.
Der Destillations- rückstand wird mit Äther ausgezogen und der Extrakt mit Natriumbicarbonat, Natriumbi- sulfit und schliesslich mit Wasser gut ge waschen, über wasserfreiem Natriumsulfat getrocknet und eingedampft. Man erhält 6" Oxy-2,2,5'-trimethyl-4"-n-amyl-3',4',5',6'-tetra- hydro-dibenzopyran als dickes, purpurfarbe nes Harz, welches unter einem Druck von 0,02 mm bei einer Badtemperatur von 165 bis 1750 C destilliert.
In einer Dosis von etwa 1 mg pro Kilo gramm zeigt diese Verbindung im Gayer-Test am Kaninchen (Arch. für exp. Pathologie und Pharmakologie, 128 [1928], Seite 312), die physiologische Wirkung des Haschisch.
6"- Oxy-2,2,5'-trimethyl-4"-n-amyl-3',4',5', 6'-tetrahydro-dibenzopyran wird mittels Essig säureanhydrid in Pyridin acetyliert. Das Ace tat wird in Form eines gelblichen Harzes er halten, welches ziemlich konstant bei etwa 140 bis 1450 C unter einem Druck von 10-3 mm destilliert.
0,5 Gewichtsteil dieses Erzeugnis ses wird während 30 Minuten bei 300 bis 3100C mit 0,25 Gewichtsteil Palladiumtier- kohle erhitzt, das Reaktionsprodukt wird mit Äther extrahiert und mit 5 % iger alkoholi scher Kalilauge hydrolysiert. Die Mischung wird in üblicher Weise zu einem bräunlichen Harz aufgearbeitet. Bei der Destillation des selben geht 0,3 Gewichtsteil der Hauptfrak tion, ein schwach gelbes Harz, unter einem Druck von 10-Imm bei l60 bis<B>1650C</B> über.
Diese Verbindung zeigt die gleichen Farb reaktionen wie Cannabinol, welches aus Ha schisch isoliert wurde. Zwecks Identifizierung werden 0,16 Gewichtsteil desselben in Pyri- dinlösung mit 0,23 Gewichtsteil p Nitro-ben- zoylchlorid während 3 Stunden unter Rück fluss erhitzt. Das p-Nitrobenzoat kristallisiert aus Alkohol in schwach gelben Nadeln vom Schmelzpunkt 163 bis 1640 C; der Misch schmelzpunkt mit Cannabinol-p-nitrobenzoat zeigt keine Depression.
<I>Beispiel 2:</I> 0,7 Gewichtsteil 5-Acetoxy-5'-methyl-7-n- amyl - 3,4 - cyclohexeno - cumarin wird mit 0,4 Gewichtsteil Palladiumtierkohle während 30 Minuten auf 30.0 bis 3100 C erhitzt, wobei 100 Raumteile Gas entwickelt wer den (berechnet 98 Raumteile).
Das gebildete 5 -Acetoxy-5'-methyl-7-n-amyl-3,4-benzo-cuma- rin wird mit Äther extrahiert und mit alko holischer Kalilauge hydrolysiert. Das Hy- drolysat kristallisiert aus leicht siedendem Petroläther in farblosen Nadeln vom Sehmelz- punkt 187 C. Das Acetat wird durch Er hitzen mit Essigsäureanhydrid in Pyridin gewonnen und besteht nach Kristallisation aus Alkohol aus farblosen Nadeln vom Schmelz punkt 98 C.
Eine Lösung von 0,5 Gewichtsteil dieses Acetats in 20 Raumteilen trockenem Anisol wird langsam einer Lösung von Methylma- gnesiumjodid (aus 4 Gewichtsteilen Methyl- jodid und 0,5 Gewichtsteil Magnesium) in Anisol zugesetzt.
Nach Beendigung der Zu gabe wird das Reaktionsgemisch 4 Stunden lang auf dem Dampfbad erhitzt; es entsteht der Magnesiumjodidkomplex des 6"-Oxy-2,2,5'- trimethyl-4"-n-amyl - dibenzopyran. Die Mi schung wird gekühlt, zur Hydrolyse des Kom plexes mit Eis und verdünnter Schwefelsäure versetzt, worauf das Anisol durch Dampf destillation entfernt wird. Der Rückstand wird mit Äther extrahiert, mit Natriumbicarbonat, Natriumbisulfit und dann mit Wasser ge waschen, über Natriumsulfat getrocknet und eingedampft.
Das Erzeugnis destilliert unter einem Druck von l0-3 mm bei 140 bis 1450 C als beinahe farbloses Harz (0,4 Gewichtsteil), welches die Eigenschaften von Cannabinol be sitzt.
In alkoholischer Lösung zeigt das syn thetische Material ein Absorptionsmaximum bei 2840 A (E=17 000) und ein Minimum bei 2510 A, während natürliches Cannabinol ein Maximum bei 2850 A (E=16 790) und ein Minimum bei 2500 A aufweist (siehe Jacob & Todd, J. C. S. [1940] Seite 649). Bei der Acetylierung der synthetischen Verbindung mit Essigsäureanhydrid in Pyridinlösung er hält man das Acetat, welches aus Alkohol in farblosen Nadeln vom Schmelzpunkt 75 bis 760 C kristallisiert.
Die Eigenschaften des Acetats können. nicht von denjenigen des Cannabinolacetats (Schmelzpunkt 750 C) aus Haschisch unterschieden werden, und der Mischschmelzpunkt zeigt keine Depression.
Process for the production of a dibenzopyran derivative. The subject of the present patent is a process for the preparation of 6 "-Oxy-2,2,5'-trimet.hyl-4" -n-amyl-dibenzopyran, characterized in that, starting from 5-acyloxy-5'- methyl-7-n-amyl-3,4-cyclohexenocoumarin, on the one hand a methylmagnesium halide, allows the reaction product to act, and on the other hand it is dehydrated.

The starting compound, namely 5-acyloxy-5'-methyl-7-n-amyl-3,4-cyclohexeno-coumarin, can be obtained by condensation of olivetol monohydrate with methylcyclohexane - 3 - an ethyl -4-carboxylate in Presence of concentrated sulfuric acid and subsequent acylation. The acetyl compound is advantageously used as the starting substance
used.
The process product turned out to be identical to cannabinol. The tetrahydrocannabinol obtained as an intermediate product (6 "- oxy-2,2,5 '-, trimethyl-4" -n-amyl-3', 4 ', 5', 6'-tetrahydro-dibenzopyran) shows strong physiological Effects that are characteristic of hashish.
The process product should be used as a drug. Example 1: 5-Oxy-5'-methyl-7-n-amyl-3,4-cyclohexeno-coumarin is refluxed with acetic anhydride in the presence of pyridine. The 5-acetoxy-5'-methyl-7-n-amyl-3,4-cyclohexeno-coumarin obtained forms colorless prisms with a melting point of 82 to 830 C.
A solution of 44 parts by weight of the compound mentioned in 600 parts by volume of anisole is slowly added to a solution of methyl magnesite iodide (made of 100 parts by weight of methyl iodide and 17 parts by weight of magnesium) in anisole at room temperature.
After the addition is complete, the mixture is heated on the steam bath for 8 hours; the magnesium iodide complex of 6 "-oxy-2,2,5'-trimethyl-4" -n-amyl-3 ', 4', 5 ', 6'-tetrahydro-dibenzopyran is formed. The mixture is cooled and, to hydrolyze the complex, ice and dilute sulfuric acid are added, whereupon the anisole is removed by steam distillation.
The distillation residue is extracted with ether and the extract washed well with sodium bicarbonate, sodium bisulfite and finally with water, dried over anhydrous sodium sulfate and evaporated. 6 "oxy-2,2,5'-trimethyl-4" -n-amyl-3 ', 4', 5 ', 6'-tetra-hydro-dibenzopyran are obtained as a thick, purple-colored resin which under pressure distilled from 0.02 mm at a bath temperature of 165 to 1750 C.
In a dose of about 1 mg per kilogram, this compound shows the physiological effect of hashish in the Gayer test on rabbits (Arch. Für exp. Pathologie und Pharmakologie, 128 [1928], page 312).
6 "- Oxy-2,2,5'-trimethyl-4" -n-amyl-3 ', 4', 5 ', 6'-tetrahydro-dibenzopyran is acetylated in pyridine using acetic anhydride. The Ace did it in the form of a yellowish resin, which distilled fairly constant at about 140 to 1450 C under a pressure of 10-3 mm.
0.5 part by weight of this product is heated for 30 minutes at 300 ° to 310 ° C. with 0.25 part by weight of palladium charcoal, the reaction product is extracted with ether and hydrolyzed with 5% alcoholic potassium hydroxide solution. The mixture is worked up in the usual manner to give a brownish resin. When the same is distilled, 0.3 part by weight of the main fraction, a pale yellow resin, passes over under a pressure of 10-Imm at 160 to 1650C.
This compound shows the same color reactions as cannabinol, which was isolated from Ha shish. For identification purposes, 0.16 part by weight of the same in pyridine solution with 0.23 part by weight of p-nitrobenzoyl chloride is heated under reflux for 3 hours. The p-nitrobenzoate crystallizes from alcohol in pale yellow needles with a melting point of 163 to 1640 C; the mixed melting point with cannabinol p-nitrobenzoate shows no depression.
<I> Example 2: </I> 0.7 part by weight of 5-acetoxy-5'-methyl-7-n-amyl - 3,4 - cyclohexeno - coumarin is mixed with 0.4 part by weight of palladium animal charcoal to 30.0 to 3100 for 30 minutes C, with 100 parts of gas being developed (calculated 98 parts of space).
The 5-acetoxy-5'-methyl-7-n-amyl-3,4-benzo-cumarine formed is extracted with ether and hydrolyzed with alcoholic potassium hydroxide solution. The hydrolyzate crystallizes from low-boiling petroleum ether in colorless needles with a melting point of 187 C. The acetate is obtained by heating with acetic anhydride in pyridine and, after crystallization from alcohol, consists of colorless needles with a melting point of 98 C.
A solution of 0.5 part by weight of this acetate in 20 parts by volume of dry anisole is slowly added to a solution of methyl magnesium iodide (composed of 4 parts by weight of methyl iodide and 0.5 part by weight of magnesium) in anisole.
After the addition is complete, the reaction mixture is heated on the steam bath for 4 hours; the magnesium iodide complex of 6 "-oxy-2,2,5'-trimethyl-4" -n-amyl - dibenzopyran is formed. The mixture is cooled, ice and dilute sulfuric acid are added to hydrolyze the complex, whereupon the anisole is removed by steam distillation. The residue is extracted with ether, washed with sodium bicarbonate, sodium bisulfite and then with water, dried over sodium sulfate and evaporated.
The product distills under a pressure of 10-3 mm at 140 to 1450 C as an almost colorless resin (0.4 part by weight), which has the properties of cannabinol.
In alcoholic solution, the synthetic material shows an absorption maximum at 2840 A (E = 17,000) and a minimum at 2510 A, while natural cannabinol has a maximum at 2850 A (E = 16 790) and a minimum at 2500 A (see Jacob & Todd, JCS [1940] page 649). When the synthetic compound is acetylated with acetic anhydride in pyridine solution, the acetate is obtained, which crystallizes from alcohol in colorless needles with a melting point of 75 to 760 C.
The properties of the acetate can. cannot be distinguished from those of cannabinol acetate (melting point 750 C) from hashish, and the mixed melting point shows no depression.
