Verfahren zur Herstellung des Chlorids des 2-[N4-(a-N-Pyridinium-propionyl)- sulfanilamidol-pyrimidins. Es wurde gefunden, dass die bisher nicht beschriebenen wasserlöslichen Sulfanilamid- derivate, die einen heterocyclischen Ring in der N'-Stellung des Sulfanilamidmoleküls und einen aliphatischen Acylre-st.in der N4 Stellung, der eine quaternäre Stickstoff gruppe besitzt, aufweisen, und der allgemei nen Formel:

worin R Wasserstoff oder Alkyl, R' einen quaternären Stickstoffrest und R" einen heterocyclischen Ring bedeuten, entsprechen, wertvolle Darmantiseptica sind. In der Praxis haben sich diese Verbindungen gegen die Bakterienflora des Darmes als ausgesprochen wirksam erwiesen, wobei sie in dieser Hin sicht den bekannten Sulfanilamidderivaten überlegen sind.- Die neuen Verbindungen sind in Wasser sehr leicht löslich.
Trotz ihrer Löslichkeit wurde jedoch überraschender- weise festgestellt, dass sie nur sehr langsam und unvollständig vom Darmtrakt resorbiert werden und dass demnach nur eehr niedrige Blutspiegel nach deren oralen Verabreichung beobachtet werden. Diese Eigenschaften .sind sehr wünschenswert für Verbindungen, wel che als Darmantiseptica Verwendung finden sollen,. da vermöge der unvollständigen Re sorption das Eintreten von renalen Kompli- kationen, die bei den bekannten Sulfanil- amidderivaten als Begleiterscheinungen auf treten, verhindert wird.
Die neuen Verbin dungen stellen somit eine Gruppe von Darm- antiseptica von geringer oraler Giftigkeit dar.
Als Ausgangsverbindungen für die Her stellung der neuen Verbindungen kommen in Betracht: N4-Halogenacyl-derivate von Sulfa- verbindungen, welche in N'-Stellung des Sulfanilamidmoleküls einen heteracyclischen Ring enthalten, wie beispielsweise 2-(N4- Chloracetyl-sulfanilamido)-pyrimidin, 2-[N4 (a-.iChlor-propionyl)-sulfanilamido]-pyrimi- din, 2 - [N4 - (a - Brom - propionyl)
- sulfanil- amido]-pyrimidin, 2-(N4-Chloracetyl-sulf- anilamido)-thiazol, 2-[N4-(a-Brom-propio- nyl)-sulfanilamido]-thiazol, 2-(N4-Chlorace- tyl-sulfanilamido)-pyridin und ähnliche Ver bindungen.
An Stelle der N4-Halogenacyl- derivate von Sulfaverbindungen können auch die ähnlich reagierenden Sulfite, Sulfate, Al- lyl-sulfonate oder Aryl-sulfonate der ent sprechenden N'-Oxyacyl-derivate verwendet werden.
Diese Ausgangsmaterialien sind ent weder bekannte Verbindungen oder können leicht dadurch erhalten werden, dass. man Sulfanil-amidderivate, welche in N'-Stellung einen heterocyclischen Ring und in NO-Stel lung eine freie Aminogruppe enthalten, in üblicher Weise mit. Anhydriden oder Säure halogeniden von aliphatischen Halogensäuren zur Umsetzung.
bringt. _ Werden diese Ausgangsverbindungen mit tertiären Aminen, beispielsweise mit Pyridin oder Trimethylamin, zur Reaktion gebracht, so erhält man die der obigen allgemeinen Formel entsprechenden, neuen, wasserlösli- chen Sulfanilamidderivate in guter Ausbeute.
Die .Umsetzung. kann in der Kälte oder bei erhöhter Temperatur und in Gegenwart oder Abwesenheit eines Lösungsmittels bzw. Ver- dünnungsmittels durchgeführt werden. Ver wendet man ein wässriges Reaktionsgemisch, so wird man, statt direkt zu den wasserlösli chen .Salzen der neuen quaternären ,Stick- stoffderivate zu gelangen, die entsprechenden freien Basen erhalten.
In- gewissen Fällen werden ihre intramolekularen Anhydride, die Betaine; als Zwischenprodukte anfallen. Die selben können leicht durch Behandeln mit einer ;Säure in die entsprechenden löslichen Salze umgewandelt werden.
Gegenstand des vorliegenden Patentes ist ein- Verfahren zur Herstellung des Chlorids des 2-[N4-(a-N=Pyridinium-propionyl)-sulf- anüamido]-pyrimidins, welches dadurch ge kennzeichnet ist, dass 2-Sulfanilamido-pyri- midin mit a-Chlor-propionylchlorid umge setzt und das entstandene 2-[N@-(a-Chlor- propionyl)
-sulfanilamido]-pyrimidin mit Py- ridin erwärmt wird.
Das neue Chlorid des 2-[N''-(a-N-Pyridi- nium-propionyl)-isulfänilamido] -pyrimidins bildet Kristalle, die bei 234 sintern und bei 235 bis 23.6 unter Zersetzung schmelzen. Es löst sich leicht in Wasser, schwerer in Alko hol und soll -als Arzneimittel verwendet wer den.
Beispiel: In. einen mit einem wirksamen inechani- schen Rühr-er, einem Tropftrichter und einem Thermometer versehenen Dreihalskolben wer den 789 g 2-Sulfanilamido-pyrim'din und 5.
Liter Wasser eingetragen. Hierauf versetzt man mit genügend konz. Salzsäure, um das 2-Sulfanilamido-pyrimidin zu lösen. Die Lö sung wirrt kräftig gerührt und tropfenweise mit 789 g a-Chlor-propionylchlorid versetzt, wobei man die Temperatur des Reaktions- gemischen durch- äusseres Kühlen mit einem Kochsalz-Bis-Gemisch auf 0 bis 5 C hält.
Nach Zugabe einiger Tropfen Säurechlorid beginnt sich ein Niederschlag des Reaktions produktes zu bilden, welcher nach Massgabe der Zuführung von a-Chlor-propionylchlorid zunimmt. Nach vollständiger Zugabe dieser letzteren Verbindung, was ungefähr 4 bis 5 Stunden in Anspruch nimmt, wird das Reak- tionsgemisch während weiteren 2 bis 3 Stun den gerührt. Das Reaktionsprodukt wird als dann abgesaugt.
Es wird auf dem Filter mit verdünnter ,Salzsäure und hierauf mit Was ser gründlich gewaschen, bis das Filtrat neu tral ist. Nach dem Waschen mit Alkohol und Äther wird die Verbindung im Vakuum bei 60 bis 70 C bis zur Gewichtskonstanz ge trocknet.
Man erhält 756 g rohes 2-[N4-(a-Chlor- propionyl) -sulfanilamido] -pyrimidin. Das Produkt sintert bei 218 C und schmilzt bei 220 C unter Zersetzung. Eine zweimal aus wässrigem Alkohol umkristallisierte Probe schmilzt bei 223 bis 224 C unter Zersetzung.
Beim Neutralisieren des sauren Filtrates des Reaktionsgemisches mit Ammoniak kön nen 175 g nicht umgesetztes 2-Sulfanilamido- pyrimidin zurückgewonnen werden.
Eine Suspension von 450 g trockenem 2 [N4 - (a - Chlor - pröpionyl) - sulfanilamido] - pyrimidin in 13'00 em@ absolutem Pyridinwird während 3 Stunden gelinde unter Rückfluss erwärmt, worauf das 2-[N4-(a-Chlor-propio- nyl)
-sulfanilamido]-pyrimidin unter Bildung einer gefärbten Lösung in Reaktion tritt. Nach 2 Stunden beginnt die Ausfällung des Reaktionsproduktes. Nach dem Kühlen des Reaktionsgemisches auf GO <B>C</B> wird der Nie derschlag filtriert und so gut als möglich durch Absaugen getrocknet. Er wiegt zwi schen 450 bis 470 g und wird in einem sie denden Gemisch von 7,1/2 Litern Alkohol und 650 cm' Wasser gelöst.
Diese Lösung wird hierauf während einer Stunde unter Zusatz von 25 g aktivierter Kohle unter Rückfluss erhitzt und durch einen mit Dampferhitzten Trichter- filtriert. Beim Kühlen kristallisiert das Reaktionsprodukt aus. Die Ausbeute -be trägt 445 g. _
