Verfahren zur Herstellung des Br omids des 2-iNi-(a-N-Pyridinium-propionv1)- sulfanilamidol-pyrimidins. Es wurde gefunden, dass die bisher nicht beschriebenen wasserlöslichen Sulfanilamid- derivate, die einen heterocyclischen Ring in der NL-Stellung des Sulfanilamidmoleküls und einen aliphatischen Acylrest,
der in der NO-Stellung eine quaternäre Stickstoffgruppe besitzt, aufweisen und der allgemeinen Formel:

worin R Wasserstoff oder Alkyl, R' einen quaternären Stickstoffrest und R" einen heterocyclischen Ring bedeuten, entsprechen, wertvolle Da-rmantiseptica sind.
In der Pia xis haben sich diese Verbindungen gegen die Bakterienflora des Darmes als ausgesprochen wirksam erwiesen, wobei sie in dieser Hin sicht den bekannten Sulfanilamidderivaten überlegen sind. Die neuen Verbindungen sind in Wasser sehr leicht löslich. Trotz ihrer Löslichkeit wurde jedoch überraschender weise festgestellt, dass sie nur sehr langsam und unvollständig vom Darmtrakt resorbiert werden und dass demnach nur sehr niedrige Blutspiegel nach deren oralen Verabreichung beobachtet werden.
Diese Eigenschaften sind sehr wünschenswert für Verbindungen, welche als Darmantiseptica Verwendung finden sol len, da vermöge der unvollständigen Resorp tion das Eintreten von renalen Komplikatio- nen, die bei den bekannten Sulfanilamidderi- vaten als Begleiterscheinungen auftreten, ver hindert wird. Die neuen Verbindungen stellen somit eine Gruppe von Darmantiseptica von geringer oraler Giftigkeit dar.
Als Ausgangsverbindungen für die Her stellung der neuen Verbindungen kommen in Betracht: N4-Halogenacyl-derivate von Sulfa- verbindungen, welche in N'-Stellung des Sulfanilamidmoleküls einen heteroevclisehen Ring enthalten, wie beispielsweise 2-(N'- Chlora.cetyl-sulfanilamido)-pyrimidin, 2-[N4- (a- Chlor-propionyl) - sulfanilamido]-pyrimi- din,
2 - [N4 - (a -Brom - propionyl) - sulfanil- amido] -pyrimidin, 2 - (N4- Chloracetyl-sulf- anilamido)-thiazol, 2-[N4-(a-Brom-propio- nyl) -sulfanilamido] -@thiazol, 2-,(N4- Chlor- acetyl-sulfanilamido)-pyridin und ähnliche Verbindungen.
An Stelle der N4-Halogen- acyl-derivate von Sulfaverbindungen können auch die ähnlich reagierenden Sulfite, Sul fats, Alkyl-sulfonate oder Aryl-sulfonate der entsprechenden N4-Oxyacyl-derivate verwen det werden.
Diese Ausgangsmaterialien sind entweder bekannte Verbindungen oder kön nen leicht dadurch erhalten werden, dass man Sulfanilamidderivate, welche in N'-Stellung einen heterocyclischen Ring und in NO-Stel lung eine freie Aminogruppe enthalten, in üblicher 'Weise mit Anhydriden oder Säure halogeniden von aliphatischen Halogensäuren zur Umsetzung bringt.
Werden diese Ausgangsverbindungen mit tertiären Aminen, beispielsweise mit Pyridin oder Trimethylamin, zur Reaktion gebracht, so erhält man die der obigen allgemeinen Formel entsprechenden,
neuen wasserlös- lichen Sulfanilamidderivate in guter Aus- beute. Die Umsetzung kann in der gälte oder bei erhöhter Temperatur und in Gegen wart oder Abwesenheit eines Lösungsmittels bezw. Verdünnungsmittels durchgeführt wer den.
Verwendet man ein wässriges Reaktions gemisch, so wird mann, statt direkt zu den wasserlöslichen Salzen der neuen quaternären Stickstoffderivate zu gelangen, die entspre chenden freien Basen erhalten. In gewissen Fällen werden ihre intramolekularen An hydride, die Betaine, als Zwischenprodukte anfallen. Dieselben können leicht durch Be handeln mit einer Säure in die .entsprechen- den löslichen Salze umgewandelt werden.
Gegenstand des vorliegenden Patentes ist ein Verfahren zur Herstellung des Bromids des 2-[1V@-(a-N-Pyridinium-propionyl)-sulf- anilamidol-pyrimidins, welches dadurch ge kennzeichnet ist, dass 2-Sulfanilamido-pyr- imidin mit a-Brom-propionylbromid umge setzt wird und das entstandene 2-[N-(a- Brom 4-propionyl)
-sulfanilamidol - pyrimidin mit Pyridin erwärmt wird.
Das neue Bromid des 2-[N4-(a-N-Pyridi- nium!-propionyl) -sulfanilamido]-pyrimidins schmilzt bei 167-173 . Es löst sich leicht in Wasser, schwerer in Methanol, in den meisten organischen Lösungsmitteln ist es wenig löslich.
Es soll als Arzneimittel verwendet werden.
Beispiel: In einem mit einem Rührer, einem Tropf trichter und einem Thermometer ausgerüste ten Dreihalskolben trägt man 24 g 2-Sulf- anilamido-pyrimidin und 400 cm' Wasser ein. Man versetzt mit 60 cm' konzentrierter Salzsäure, um das Sulfanilamido-pyrimidin aufzulösen.
Das Reaktionsgemisch wird her nach auf unterhalb 10 gekühlt und bei dieser Temperatur tropfenweise derart mit 50 g a-Brom-propionylbromid, gelöst in 150 cm' Benzol, versetzt, dass die Reaktionstempe- ratur unterhalb 10 C bleibt.
Die Zugabe dauert 75 Minuten. Das Reaktionsgemisch wird hierauf während weiteren 2 Stunden bei Zimmertemperatur gerührt.
Das während dem Reaktionsverlauf ausfallende N4-(a- Brom -propionyl) - sulfanilamido - pyrimidin wird abfiltriert und mehrmals aus wässrigem Alkohol unter Zugabe von aktivierter Kohle umkristallisiert. Smp. 212-218 C unter Zersetzung. Ausbeute 17 g. 8 g der Aus gangsverbindung können aus dem Filtrat des Reaktionsgemisches nach dem Neutralisieren zurückgewonnen werden.
Die Umsetzung von 2-Sulfanilarnido-pyr- imidin mit a-Brom-propionylbromid kann auch -wie folgt durchgeführt werden: In einem mit einem Rührer, einem Tropf trichter und einem Thermometer versehenen Dreihalskolben werden 600 g 2-Sulfanil- amido-pyrimidin, 6 Liter Aceton und 280 cm' Pyridin .eingetragen. Die Suspension wird auf 5 C gekühlt.
Alsdann versetzt mann tropfenweise mit 571,6 g a-Brom-prGpionyl- bromid, wobei man kräftig rührt und die Temperatur nicht auf höher als 15 C an steigen lässt. Die Zugabe dauert etwa 4 Stun den.
Das Reaktionsgemisch wird während weiteren 5 Stunden gerührt und hierauf während 48 Stunden bei Zimmertemperatur stehengelassen. Das Reaktionsgemiseh wird hernach abgesaugt und mehrmals mit 2 Liter 2n-Salzsäure behandelt, um unver- ändertes 2-Sulfanilamido-pyrimidin zu be seitigen. Auf diese Weise können aus dem sauren Filtrat nach dem Neutralisieren mit Ammoniak 87 g 2-Sulfanilamido-pyrimidin zurückgewonnen werden.
Der in Säure un lösliche Anteil wird mit Wasser so lange gewaschen, bis das Filtrat neutral ist; hier auf wird mit Alkohol und Äther gewaschen. Nach dem Trocknen bei 60 C im Vakuum bis zum Erreichen von Gewichtskonstanz erhält mann 632 g 2-[N4-(a-Brom-propionyl)- sulfanilamido]-pyrimidin vom Smp. 214 bis 2L5 C.
120 cm; absolutes Pyridin werden tropfenweise unter Rühren einer unter Rückfluss erhitzten Suspension von 59 g <I>i-</I> [N4- (a- Brom - propionyl - sulfanilamido] <I>2</I> py rimidin in 1500 cm' absolutem Alkohol zugesetzt. Nach 2 Stunden wird das Gemisch klar. Man erhitzt während weiteren 51/2 Stun den unter Rückfluss. Das Reaktionsgemisch wird alsdann über Nacht bei Zimmertempe ratur stehengelassen.
Nach dem Eindampfen zur Trockne im Vakuum wird der ölige Rückstand mit Wasser behandelt und von wenigem unlöslichem Ausgangsmaterial erb- filtriert. Das wässrige Filtrat wird hernach mit aktivierter Kohle behandelt, filtriert und im Vakuum zur Trockne eingedampft. Der Rückstand wird nun in möglichst wenig siedendem Methanol gelöst, erneut mit akti vierter Kohle behandelt, die Lösung filtriert und etwas eingeengt. Nach dem Kühlen und Reiben kristallisiert das Reaktionsprodukt aus.
Es schmilzt nach zweimaligem Umkri- stallisieren aus Methanol bei 167-173 C. Die Ausbeute beträgt 50 g.

Despite their solubility, however, it was surprisingly found that they are only very slowly and incompletely absorbed from the intestinal tract and that accordingly only very low blood levels are observed after their oral administration.
These properties are very desirable for compounds which are to be used as intestinal antiseptics, since the incomplete absorption prevents the occurrence of renal complications, which occur as side effects with the known sulfanilamide derivatives. The new compounds thus represent a group of intestinal antiseptics of low oral toxicity.
